The distribution of putative nitric oxide synthase (NOS)-containing cells in the opisthobranch mollusc Pleurobranchaea californica was studied histochemically via NADPH-diaphorase (NADPH-d) reduction of Nitro Blue Tetrazolium (NTB). Whole mounts and cryostat sections were prepared from the central nervous system and peripheral organs, including the buccal muscles, esophagus, salivary glands, foot, mantle, and gills. NADPH-d-positive neurons were localized predominantly to the buccal and pedal ganglia as well as to distinct areas of the cerebropleural and visceral ganglia. A variety of identified neurons were positive for NADPH-diaphorase in various central ganglia, including the metacerebral cells of the cerebropleural ganglion, putative locomotor neurons of the pedal ganglia, and buccal motoneurons. Specific staining was observed only in somata of central neurons, whereas neuropil areas remained unstained. However, NADPH-d-reactive axons were dense in buccal ganglion nerves, whereas peripheral nerves and connectives of other ganglia had few or no NADPH-d positive terminals. In the periphery, NADPH-d activity was detected only in a few neurons of the rhinophore and tentacle ganglia. NADPH-d staining was marked in the salivary glands and gills, but there was no or very little staining in the esophagus, buccal mass, and foot. Histochemical stain production required the presence of both beta-NADPH and NBT; alpha-NADPH could not substitute for beta-NADPH. The inhibitor of NOS, 2,6-dichlorophenol-indophenol, at 10(-3) M, totally abolished NADPH-d-positive staining. The apparent high activity of central NADPH-d contrasts with much lower activity in the ganglia of the related gastropod Tritonia. These data suggest a role for nitric oxide as a signal molecule in the central nervous system of Pleurobranchaea. 相似文献

Three psychological variables (self-efficacy, control and meaning, and perceived risk) were tested in a structural model predicting AIDS-preventive behavior. Results revealed a good model fit, indicating that these psychological variables did play a role in mediating AIDS-preventive behavior in college students. A multivariate analysis of variance (MANOVA) and individual ANOVAs conducted for men and women also revealed gender differences on individual items of self-efficacy, perceived risk, and AIDS-preventive behavior. We have addressed the question of the nature of Moloney murine leukemia virus (MoMuLV) repression in mouse embryos by assaying for the transient expression of MoMuLV-derived constructs microinjected into early cleavage embryos. We show that the same cis-acting DNA sequences responsible for the block in MoMuLV expression in embryonal carcinoma cell lines operate in early embryos: (i) the MoMuLV long terminal repeat is nonfunctional, and (ii) the +147 to +163 repressor binding site, or negative regulatory element, negatively regulates the expression from an active promoter. 相似文献

Findings from a study comparing partner abuse in African American women suicide attempters (n?=?148) and nonattempters (n?=?137) revealed higher rates of physical and nonphysical partner abuse among attempters than their demographically similar nonsuicidal counterparts. The partner abuse–suicidal behavior link was mediated by psychological distress, hopelessness, and drug use and moderated by social support. Results also revealed that nonphysical partner abuse accounted for unique variance in the prediction of suicide attempt status beyond that attributable to childhood maltreatment. Implications of the findings for assessing both suicidal and abused women are discussed, and recommendations for preventive interventions for women at risk for suicidal behavior are provided. The pentapeptide enterostatin (ENT) inhibits feeding after injection into the cerebral ventricles. To localize the central sites of action of ENT, the peptide (0.01 to 3.3 nM) was microinjected into several brain regions and the intake of a high fat diet was measured. The results show that ENT injection in the paraventricular nucleus (PVN) or the amygdala (AMYG) produced a bi-phasic dose related feeding response, low doses of ENT inhibited feeding while higher dose had no effect. The effective dose to inhibit feeding in the AMYG was 10 fold lower than that in the PVN. No changes in food intake were observed after ENT injection into the ventromedial hypothalamus and nucleus tractus solitarius. The data provide further support that there are targets in the CNS for ENT and suggest that central ENT function is site specific. 相似文献

Various segments of the 3'-nontranslated region of the renal glutaminase (GA) mRNA were tested for their ability to enhance turnover and pH responsiveness. The combined effects were retained in the 340-base R-2 segment. However, the combined R-1 and R-3 fragments also imparted a partial destabilization and pH responsiveness to a chimeric beta-globin mRNA. RNA electrophoretic mobility shift assays indicated that cytosolic extracts of rat renal cortex contain a protein that binds to the R-2 and R-3 RNAs. The binding observed with the R-2 RNA was mapped to a direct repeat of an 8-base AU sequence. This binding was effectively competed with an excess of the same RNA, but not by adjacent or unrelated RNAs. UV cross-linking experiments identified a 48-kDa protein that binds to the AU repeats of the R-2 RNA. The apparent binding of this protein was greatly reduced in renal cytosolic extracts prepared from acutely acidotic rats. Two related RNA sequences in the R-3 segment also exhibited specific binding. However, the latter binding was more effectively competed by R-2 RNA than by itself, indicating that the homologous sites may be weaker binding sites for the same 48-kDa protein. Thus, a single protein may bind specifically to multiple instability elements within the 3'-nontranslated region of the GA mRNA and mediate its pH-responsive stabilization. 相似文献

Testicular cell suspensions were prepared from obstructive and non-obstructive azoospermic men and were cultured in vitro for 96 h as (i) mixed cell populations and (ii) isolated homogeneous populations of primary spermatocytes, round spermatids and elongating spermatids. The cells lost their viability gradually during the first 24 h period. By 72 h almost 90% of the cells were non-viable. Isolated pure fractions showed better viability at each time interval (P < 0.0005). Throughout the culture period primary spermatocytes, elongating spermatids and other non-spermatogenic cells showed no change in their morphology, but almost 22% of round spermatids showed growth of flagella. Most of the round spermatids developed their flagella during the first 4-8 h period of culture. Isolated pure round spermatids showed better flagellar growth compared with mixed cell suspensions (P < 0.0005). The spermatogenic cells were successfully cryopreserved. However, when mixed spermatogenic cell suspensions were cryopreserved, more cells lost their viability compared with when isolated pure fractions were cryopreserved (P < 0.0005). 相似文献

Consummatory behavior and weight-regulation capacity were measured in 12 normal male Long-Evans rats and in 43 similar Ss that survived complete (C), sequential unilateral (U), anterolateral (A) , or posterior (P) neocortical ablations. Groups C and A displayed aphagia and adipsia followed by a sequence of recovery stages qualitatively identical to, but shorter than, recovery typically seen following lateral hypothalamic lesions. After recovery, Group C displayed long-term effects of finickiness and prandial drinking. These effects and a measure of recovery of body-weight-regulation capacity were significantly intercorrelated with lesion size, and body-weight set point remained significantly lower than normal. Group U was relatively unaffected by the 1st unilateral ablation and showed, relative to Group C, facilitated recovery of consummatory behaviors following the 2nd ablation, but displayed the long-term effects. Group P, though significantly affected by the lesion, did not display the pattern or intensity of effects described for the other bilaterally ablated groups. 1. A contact of the pteropod mollusk Clione limacina with its prey (small pteropod mollusk Limacina helicina) evokes a complex pattern of hunting and feeding behavior: protraction of tentacles to seize the prey, activation of buccal apparatus to swallow the prey, activation of locomotor system (speeding up of wing beating), reversal of reaction to tactile stimulation of the head, loss of normal (vertical) orientation in space, and swimming in circles. After injection of physostigmine (PhS), the acetylcholinesterase inhibitor, into the hemocoel of intact Clione, all these manifestations of the hunting and feeding behavior could be evoked by tactile stimulation of the head, or they arose spontaneously. 2. In the preparation of the isolated CNS, the effect of PhS on the neural networks controlling different aspects of the hunting and feeding behavior was studied by recording from neurons monitoring activity of different networks (< or = 4 neurons simultaneously). Tactile stimulation of the head was mimicked by a short-term electrical stimulation of the corresponding nerve. Before PhS application, the nerve stimulation evoked elements of the defense reaction, i.e., long-lasting inhibition of all main motor control systems: the locomotor network in the pedal ganglia, the tentacle control network in the cerebral ganglia, and the network controlling radula and hook movements in the buccal ganglia. However, after PhS application, the same stimulus evoked a long-lasting bout of excitation in all the three networks accompanied by activation of the heart-exciting neuron as well as by a modification of the activity of statocyst receptor cells controlling Clione's spatial orientation (the "fictive hunting bout"). Similar hunting bouts could arise spontaneously. 3. Injection of acetylcholine (ACh) into the hemocoel of intact Clione was less effective than injection of PhS. After ACh injection, reversal of reaction to head stimulation was observed in < or = 20% of the experiments (the percentage of positive results was higher if ACh was injected into the head just over the CNS). Bath application of ACh to the isolated CNS did not produce the hunting bouts. However, a short-term local application of ACh to the cerebral ganglia in the isolated CNS resulted in activation of the main motor systems controlling locomotion, protraction of tentacles, and movements of buccal mass. 4. During spontaneous PhS-induced bouts, excitation of different networks involved in hunting behavior was sometimes not quite synchronous. S. Enteritidis HY-1 isolated during quarantine from chicks imported from England was used. Laying hens at the age of 34 weeks were inoculated orally with 10(10) organisms (10 birds), intramuscularly with 10(9) (5 birds), and intravenously with 10(9) (5 birds). Egg production did not change in hens infected orally, although it was reduced in hens infected intramuscularly for 2-3 weeks post inoculation. For one month, internally infected eggs of which the shells were not contaminated were found: one out of 65 eggs in hens infected orally and three out of 36 eggs in hens infected intramuscularly. Ran 16 neonatal purebred beagles for 6.5 days in a 1-way shuttle box with cold air as the aversive stimulus. 8 Ss started at 1 day of age and 4 each started at 2 and 3 days of age. 8 Ss received escape conditioning and 8 received avoidance conditioning. Following this training, both groups were given a series of extinction trials. Both escape and avoidance conditioning and extinction were obtained. Findings are comparable to previous avoidance findings in neonatal dogs and superior to findings on neonatal mice and kittens. Hemolymph glucose increased following a meal of a commercially available dried seaweed (laver) in Aplysia californica. Glucose injected into the hemocoel did not affect meal size, bite latencies, swallowing rate, or 24-hr food intake. The authors found that injection of a homogenate of nerves containing a putative Aplysia insulin-like substance decreased hemolymph glucose. The nerve homogenate, however, did not affect feeding behavior. Injection of 2-deoxy-{D}-glucose was found to increase hemolymph glucose, an indication of gluco-privation, but instead of increasing feeding it either had no effect or, at high doses, debilitated animals and interfered with feeding. These studies suggest that glucose may be physiologically regulated in Aplysia, but it does not appear to play a role in the control of feeding behavior. Although influences of interest on learning are well documented, mediating processes have not been clarified. The authors investigated how individual and situational interest factors contribute to topic interest and text learning. Traditional self-report measures were combined with novel interactive computerized methods of recording cognitive and affective reactions to science and popular culture texts, monitoring their development in real time. Australian and Canadian students read 4 expository texts. Both individual interest variables and specific text titles influenced topic interest. Examination of processes predictive of text learning indicated that topic interest was related to affective response, affect to persistence, and persistence to learning. Neural regeneration in the escape circuit of the first-instar cockroach is described using behavioral analysis, electrophysiology, intracellular staining, and electron microscopy. Each of the two filiform hairs on each of the animal's cerci is innervated by a single sensory neuron, which specifically synapses with a set of giant interneurons (GIs) in the terminal ganglion. These trigger a directed escape run. Severing the sensory axons causes them to degenerate and perturbs escape behavior, which is restored to near normal after 4-6 days. Within this time, afferents regenerate and reestablish arborizations in the terminal ganglion. In most cases, regenerating afferents enter the cercal glomerulus and re-form most of the specific monosynaptic connections they acquired during embryogenesis, although their morphology deviates markedly from normal; these animals reestablish near normal escape behavior. In a few cases, regenerating afferents remain within the cercus or bypass the cercal glomerulus, and thereby fail to re-form synapses with GIs; these animals continue to exhibit perturbed escape behavior. We conclude that in most cases, specific synapses are reestablished and appropriate escape behavior is restored. This regeneration system therefore provides a tractable model for the establishment of synaptic specificity in a simple neuronal circuit. 相似文献

Central administration of alpha 2-receptor agonists stimulate food intake in mammalian and avian species. Recently we reported that inhibition of nitric oxide (NO) synthase (NOS) decreased food intake in chickens. In the present study, we investigated whether the increased eating induced by clonidine (Clon), an alpha 2-receptor agonist, is attenuated by NOS inhibition. In the first experiment, four levels (0, 9.4, 18.8 or 37.5 nmol/10 microliters) of Clon were administered into the right lateral ventricle of chickens, and food intake was monitored. Clon increased 30 min-food intake in a dose-dependent manner. In a co-administration study of L-NG-nitro-arginine methyl ester HCl (LNNA), a NOS inhibitor, and Clon, LNNA (0, 1.5, 3.0 or 5.9 mumol) attenuated food intake induced by Clon (37.5 nmol) in a dose-dependent manner. Our results suggest the possibility that NO interacts with adrenergic neurons in the central nervous system to modulate feeding behavior in the chicken. 相似文献

How neuronal activity changes cerebral blood flow is of biological and practical importance. The rodent whisker-barrel system has special merits as a model for studies of changes in local cerebral blood flow (LCBF). Stimulus-evoked changes in neural firing and 'intrinsic signals' recorded through a cranial window were used to define regions of interest for repeated flow measurements. Whisker-activated changes in flow were measured with intravascular markers at the pia. LCBF changes were always prompt and localized over the appropriate barrel. Stimulus-related changes in parenchymal flow monitored continuously with H2 electrodes recorded short latency flow changes initiated in middle cortical layers. Activation that increased flow to particular barrels often led to reduced flow to adjacent cortex. Dye was injected into single penetrating arterioles from the pia of the fixed brain and injected into arterioles in slices of cortex where barrels were evident without stains. Arteriolar and venular domains at the surface were not directly related to underlying barrels. Capillary tufts in layer IV were mainly coincident with barrels. The matching between a capillary plexus (a vascular module) and a barrel (a functional neuronal unit) is a spatial organization of neurons and blood vessels that optimizes local interactions between the two. The paths of communication probably include: neurons to neurons, neurons to glia, neurons to vessels, glia to vessels, vessels to vessels and vessels to brain. Matching a functional grouping of neurons with a vascular module is an elegant means of reducing the risk of embarrassment for energy-expensive neuronal activity (ion pumping) while minimizing energy spent for delivery of the energy (cardiac output). For imaging studies this organization sets biological limits to spatial, temporal and magnitude resolution. Reduced flow to nearby inactive cortex enhances local differences. 相似文献

The central nervous systems of the marine molluscs Pleurobranchaea californica (Opisthobranchia: Notaspidea) and Tritonia diomedea (Opisthobranchia: Nudibranchia) were examined for serotonin-immunoreactive (5-HT-IR) neurons and processes. Bilaterally paired clusters of 5-HT-IR neuron somata were distributed similarly in ganglia of the two species. In the cerebropleural ganglion complex, these were the metacerebral giant neurons (both species), a dorsal anterior cluster (Pleurobranchaea only), a dorsal medial cluster including identified neurons of the escape swimming network (both species), and a dorsal lateral cluster in the cerebropleural ganglion (Pleurobranchaea only). A ventral anterior cluster (both species) adjoined the metacerebral giant somata at the anterior ganglion edge. Pedal ganglia had the greatest number of 5-HT-IR somata, the majority located near the roots of the pedal commissure in both species. Most 5-HT-IR neurons were on the dorsal surface of the pedal ganglia in Pleurobranchaea and were ventral in Tritonia. Neither the buccal ganglion of both species nor the visceral ganglion of Pleurobranchaea had 5-HT-IR somata. Afew asymmetrical 5-HT-IR somata were found in cerebropleural and pedal ganglia in both species, always on the left side. The clustering of 5-HT-IR neurons, their diverse axon pathways, and the known physiologic properties of their identified members are consistent with a loosely organized arousal system of serotonergic neurons whose components can be generally or differentially active in expression of diverse behaviors. 相似文献
