In view of the influence that nutritional and physiological status exert on the immunological capacity of the subject, a study was carried out for the purpose of studying the changes induced by three protein levels in the diet: (4%, 10% (control), and 20%) on total plasma proteins (TPP) and their fractions, as well as Ig G and Ig M levels in non-pregnant (NP) and pregnant (P) rats and their offspring. Effect of the diet on adult rats--In non-pregnant rats submitted to the high protein diet, Ig G levels increased while TPP decreased in P rats fed on 4% and 20% protein diets. The higher the protein level in the diet, the higher were the TPP values. Effect of pregnancy--Ig G and Ig M levels suffered an increase in rats fed the 4% and 10% protein diets, while a decrease was observed in rats submitted to the 20% protein level diet. The TPP rate diminished in rats fed on the low protein diets, and increased when the highest protein diet was administered. Effect of the diet on offspring--Ig M levels were only detected in neonates from rats fed with the low and high protein diets. Moreover, the TPP rate increased as a direct function of the dietary protein intake.  相似文献

It is well known that the dietary protein level influences both the pregnancy process and development of the offspring. Therefore, a study was carried out to determine the effect that different protein percentages: 10%, 4% and 20% in diets administered to rats during pregnancy, had on food efficiency and on the substrates partition between rat dams and their neonates. Experimentation was thus carried out over a period of 21 days (pregnancy) and comparisons were made with well-nourished rats receiving 10% protein (controls) and between one and other group. Moreover, the effect on pregnancy was observed by comparing pregnant rats with non pregnant rats within each dietary situation. Food intake, weight parameters and food efficiency ratio were recorded in adult rats. Weight parameters were also evaluated in newborns. As results revealed, the highest the protein level in the diet, the highest the food efficiency ratio, both in the pregnant group and in the non pregnant group. Weight changes were determined in rat dams and their neonates, as a consequence of the different protein intakes. These also involved alterations in the substrates partition between the mothers and their offspring. Such findings might lead to the retention of substrates by the maternal tissues and, hence, to impairment of neonatal development.  相似文献

The purpose of this research was to study the binding capacity of estrogens to the sex hormone-binding globulin (SHBG) in mothers and their intrauterine malnourished newborns. Blood samples were obtained from mothers at delivery, and from babies, of the umbilical cord. SHBG was measured according to the method of Mickelson and Petra. It was found in mothers of malnourished babies that the binding capacity of serum protein to dehydrotestosterone (DHT) was significantly decreased in comparison to the controls (10.63 +/- 1.61 vs 13.25 +/- 2.18 micrograms DHT/dl serum, respectively), whereas it was significantly increased in intrauterine malnourished newborns (1.01 +/- 0.24 vs 0.77 +/- 0.18, respectively). These results suggest that SHBG decrease in mothers of intrauterine malnourished newborns occurs due to a decrease in the production of fetal adrenal hormone precursors and may, therefore, be a compensating mechanism to increase placental flow.  相似文献

In order to compare intestinal morphology and function, diarrhea was produced in rats using laxatives in the diet. The 14 day study included two groups of rats with diarrhea (osmotic or secretory), two groups without diarrhea but with a degree of malnutrition which was similar to that seen in the rats with diarrhea (malnourished without diarrhea) and a well-nourished group (control). The inclusion of laxatives(lactose or bisoxatin acetate) cause a reduction in food intake, diarrhea an malnutrition. It also caused a reduction in dietary protein and fat digestibility which was proportional to the severity of diarrhea and more pronounced in secretory diarrhea. In the malnourished rats without diarrhea, malnutrition did not affect their absorptive function. Both in the rats with secretory and osmotic diarrhea an intestinal hypertrophy was observed. This hypertrophy was proportional to the severity of diarrhea and independent of its aetiology. In the intestines of the rats with both types of diarrhea there was inflammation, a greater number of mitotic figures but the flattening of the villi seen in the malnourished rats without diarrhea was not seen. In osmotic diarrhea there was, in addition, a patchy damage of the surface of the jejunal mucosa and an increment in the number of goblet cells, indicating a more severe intestinal deterioration. Since despite this greater deterioration, these rats absorbed more protein and fat we concluded that the alterations in intestinal morphology seen in this study was not predictive of intestinal function. The study also showed that diarrhea had a trophic effect on the intestine which did not occur in malnourished rats without diarrhea.  相似文献

The influence of feeding cholesterol to rats during pregnancy and postpartum (from the 11th day of gestation to the third day after delivery) on the serum and hepatic cholesterol levels and on the bile acid composition in the pool and in the liver in relationship to the dams and their pups was examined. The hepatic content of cholesterol in both dam and offspring increased during cholesterol feeding without any changes in serum cholesterol level. In the dams, mainly the esterified cholesterol was increased; in the pups, mainly the free cholesterol was increased. Cholesterol feeding led to a pronounced increase in the pool of β-muricholic acid and a relative decrease in the lithocholic acid concentration in pregnant rats. In fetal rats, the chenodeoxycholic acid pool was increased by cholesterol intake. The lithocholic acid pool was larger in the postpartum rats fed cholesterol than in the controls, while the concentration of α- and β-muricholic acids was decreased. The neonates of cholesterol-fed dams had a larger pool of chenodeoxycholic acid but a smaller pool of β-muricholic acid. These results suggest that the metabolism of cholesterol and of bile acids in dams and their offspring respond differently to cholesterol intake.  相似文献

Dietary supplements of olive oil (OO) or fish oil (FO) during the first (G1: day 1–60) or second half of gestation (G2: day 60 to term, day 115) were offered to pregnant sows. The proportion of fatty acids in milk and plasma were determined by gas chromatography. When supplements were given during G1, the proportions of oleic acid (OA) and arachidonic acid (AA) in the plasma were higher in the OO group than in the FO group, whereas docosahexaenoic acid (DHA) was higher in the latter group at day 56 of gestation. These differences in plasma DHA were still apparent at day 7 of lactation. Similarly, DHA was also higher in the colostrum and milk on days 3 and 21 of lactation and in the plasma of piglets from FO dams compared to the OO group, whereas AA was lower. When the FO supplement was given during G2, AA was lower and DHA higher in the plasma at day 105 of gestation and at day 7 of lactation compared with the OO group. Likewise, DHA was greater in FO than in OO animals during lactation in colostrum and in milk on days 3 and 21 of lactation, and in 3-day old suckling piglets plasma, whereas AA was lower in these animals. Thus, maternal adipose tissue plays an important role in the storage of dietary long-chain polyunsaturated fatty acids (LCPUFA) during G1. They are mobilized around parturition for milk synthesis, and an excess of dietary n-3 LCPUFA decreases the availability of AA in suckling newborns.  相似文献

The nutritional potentiality of the "Regional Basic Diet" (RBD) was assessed in albino rats. The diet, which was prepared according to data from food consumption surveys and was similar--in terms of quality and quantity--to those consumed by human populations in Northeast Brazil, was compared to a balanced diet (22% casein). The centesimal composition, aminogram, minerals content, NDpCal% and GCal%/PCal% ratio were determined. Vitamin contents were calculated from data in the literature. The RBD was shown to be imbalanced and poor in certain nutrients, mainly protein (7.8%). The growth curve of the RBD group was severely impaired; the naturally occurring sex-related differences in body weight were not detected. For fetuses and suckling pups, the weights of the liver and diaphragm paralleled body weight; brain weights were about 20% lower than those of controls, and its ratio was higher. RBD pregnant females presented, despite their higher food and energy intakes, lower weight gain as compared to the controls. During lactation food and energy intakes of RBD dams were lower than those of controls. The mortality rate among RBD pups was 24%. Our data indicate that RBD produces in the rat a type of malnutrition similar to that prevalent among children from Northeast Brazil, namely an association to nutritional dwarfism with some clinical signs of marasmus. RBD is considered to be a useful experimental model for studies on human malnutrition in countries where basic food patterns are similar to those in RBD-consuming regions.  相似文献

Cortisol, a stress hormone, plays key roles in mediating stress and anti-inflammatory responses. As abnormal cortisol levels can induce various adverse effects, screening cortisol and cortisol analogues is important for monitoring stress levels and for identifying drug candidates. A novel cell-based sensing system was adopted for rapid screening of cortisol and its functional analogues under complex cellular regulation. We used glucocorticoid receptor (GR) fused to a split intein which reconstituted with the counterpart to trigger conditional protein splicing (CPS) in the presence of targets. CPS generates functional signal peptides which promptly translocate the fluorescent cargo. The sensor cells exhibited exceptional performance in discriminating between the functional and structural analogues of cortisol with improved sensitivity. Essential oil extracts with stress relief activity were screened using the sensor cells to identify GR effectors. The sensor cells responded to peppermint oil, and L-limonene and L-menthol were identified as potential GR effectors from the major components of peppermint oil. Further analysis indicated L-limonene as a selective GR agonist (SEGRA) which is a potential anti-inflammatory agent as it attenuates proinflammatory responses without causing notable adverse effects of GR agonists.  相似文献

Bisphenol A (BPA) is largely used as a monomer in some types of plastics. It accumulates in tissues and fluids and is able to bypass the placental barrier, affecting various organs and systems. Due to huge developmental processes, children, foetuses, and neonates could be more sensitive to BPA-induced toxicity. To investigate the multi-systemic effects of chronic exposure to a low BPA dose (100 μg/L), pregnant Wistar rats were exposed to BPA in drinking water during gestation and lactation. At weaning, newborn rats received the same treatments as dams until sex maturation. Free and conjugated BPA levels were measured in plasma and adipose tissue; the size of cerebral ventricles was analysed in the brain; morpho-functional and molecular analyses were carried out in the liver with a focus on the expression of inflammatory cytokines and Sirtuin 1 (Sirt1). Higher BPA levels were found in plasma and adipose tissue from BPA treated pups (17 PND) but not in weaned animals. Lateral cerebral ventricles were significantly enlarged in lactating and weaned BPA-exposed animals. In addition, apart from microvesicular steatosis, liver morphology did not exhibit any statistically significant difference for morphological signs of inflammation, hypertrophy, or macrovesicular steatosis, but the expression of inflammatory cytokines, Sirt1, its natural antisense long non-coding RNA (Sirt1-AS LncRNA) and histone deacetylase 1 (Hdac1) were affected in exposed animals. In conclusion, chronic exposure to a low BPA dose could increase the risk for disease in adult life as a consequence of higher BPA circulating levels and accumulation in adipose tissue during the neonatal period.  相似文献

目的探讨孕妇IgG类红细胞血型不规则抗体对早期诊断Non-ABO-HDN的意义。方法筛查4200份孕妇血清中红细胞血型不规则抗体,阳性者检测抗体特异性、免疫球蛋白类型及效价;对检测出IgG类不规则抗体者,分娩时取脐血(或新生儿血)检测血清(浆)游离抗体,并进行红细胞直接抗球蛋白试验和红细胞放散试验,以诊断其是否发生HDN。结果4200份孕妇血清中检出红细胞血型不规则抗体44份(阳性率为1.05%),其中20份孕妇血清的抗体为IgG类或IgG及IgM混合抗体,脐血(或新生儿血)检测结果,10名婴儿发生了Non-ABO-HDN,1名发生了宫内死胎。致病的抗体特异性分布为:抗-D2例、抗-c1例、抗-E2例、抗-cE1例、抗-M4例(其中1例并有抗-A)。结论孕妇体内IgG类的Rh血型系统的抗体及抗-M易引起non-ABO-HDN,检测孕妇IgG类红细胞血型不规则抗体对早期诊断Non-ABO-HDN、鉴别引发HDN的抗体型别、评估HDN的严重程度及制定治疗方案均具有重要意义。  相似文献

Diarrhea increases the effects of malnutrition. Accordingly, the effect of diarrhea on two types of malnutrition (protein deficiency and protein-calorie deficiency) was studied. The experiment included 42 young Sprague Dawley rats. The rats were distributed into three groups with 14 rats per group. During the first 16 of the experiment, the first group was fed a control diet ad libitum, the second received the same diet but with food intake reduced in 50% whereas the third group was offered a protein deficient diet. Thus, at the end of this period there were well-fed rats (control), as well as protein and protein-calorie malnourished rats. Then one half of the rats in each group were given lactose to produce diarrhea and all rats continued with their previously assigned diet and feeding regime during one more week. Therefore, during this period there were control rats, protein deficient rats and protein-calorie deficient rats with and without diarrhea. The results showed that diarrhea caused a substantial reduction in food intake and growth in the well-fed rats and also in the group fed the protein deficient diet. However, the protein-calorie deficient group did not reduce its intake nor its growth rate. As a result, diarrhea caused malnutrition in the control group and increased malnutrition in the protein deficient but it did not have an additional effect in the protein-calorie deficient rats. The apparent absorption of lipids and nitrogen measured in these rats showed that the absorption reduction caused by diarrhea was more pronounced in the protein deficient group. This group also had the lowest activities of intestinal disaccharidases. These results showed that diarrhea had a more detrimental effect in protein deficient than in protein-calorie deficient rats.  相似文献

The purpose of this study was to determine whether OE treatment affects the expression of genes related to lipid metabolism under two physiological conditions: late pregnancy and mid-lactation, both characterized by lipid mobilization. Samples of periovarian and retroperitoneal adipose tissue from 21-day pregnant or 15-day lactating dams were used. The expression of LPL, FATP1, FABP4, HSL, ACC1, FAS, PEPCK, GLUT4, PDK4, SREBP1c, adiponutrin and leptin, were compared with their expression in virgin rats. In pregnant rats, FABP4, HSL, PEPCK and PDK4 were over expressed in the periovarian site compared to virgin rats, whereas adiponutrin, FAS, GLUT4 and SREBP1c were underexpressed; the retroperitoneal fat depot showed a similar pattern but ACC1 and leptin were also underexpressed. OE treatment caused a generalized decrease in gene expression in both adipose depots. In lactating dams, the gene expression profile at the periovarian depot was similar to that observed in pregnant rats. OE treatment mimicked the trend observed in pregnant rats, although the intensity of the gene expression changes was lower. After OE treatment, the retroperitoneal adipose depot showed a completely different pattern since the values were close to those of virgin rats. These results corroborate that OE effects in adipose tissue, lowering lipids and depressing their metabolism, already described under other physiological situations, can be also found in late pregnancy and lactation.  相似文献

Even though there has been an increase in the coverage of governmental services and an adequate national food availability, malnutrition continues to exist in Panamanian children. In order to geographically and administratively identify those groups more seriously affected by this problem and orient governmental actions towards them, data from the National Nutrition Survey carried out in 1980 in 14 occupational (functional) groups were classified. Social, economic and cultural characteristics of each functional group were associated with food and nutrition problems. More than half of the malnourished children fall within the functional groups who work in the agricultural/sector. Within them, more than 40% of the malnourished live in homes where two-thirds of their income is derived from work performed outside their own farms. In urban as well as in rural areas low food availability exists at the family level in 25% of the families with inadequate diets studied. The food problem in the rural area is worsened due to limited access to health services and poor environmental conditions. Therefore, the malnutrition problem in Panama is linked to low incomes that prevent acquisition of sufficient foods and other goods and services, as well as to the difficulty of accessibility to public services by an important part of the population.  相似文献

The objective of the study was to evaluate the nutritional status of the hospitalised elderly patients using the Mini Nutritional Assessment (MNA) and nutritional classic parameters, to estimate the prevalence of undernutrition among them and to value the correlation of undernutrition with epidemiological characteristics of the sample for determine the highest risk's group among the malnourished patients. A cross sectional survey was done among a sample of 200 elderly patients admitted to the USP San Carlos Hospital (Murcia, Spain) during a period of 3 months. Patients answered a questionnaire including demographic data and the MNA clinical tool. Lymphocyte concentration and albumin and transferrin serum levels were also collected. Statistical analyses were performed with the Student t and chi 2 tests. For the MNA, the Spearman's coefficient was employed. Mean age of the 200 patients was 80.72 SD 7.43 years. Mean weight was 63.41 SD 19.57 kg and mean height was 160.93 SD 8.36 cm, with a mean Body Mass Index of 24.27 SD 7.31. The serum values of albumin and transferrin and total lymphocytes in blood were 3.09 SD 0.5 g/l, 1.69 SD 0.37 mg/dl and 1412 mm3 respectively. Average score of the MNA was 15.9 SD 6.21 and, according to this scale, 50% (n=100) of the patients were malnourished. Correlation analyses resulted in significant association (p < 0.001) between values of undernutrition in the MNA and measurements under normality of the biochemical and immunological parameters. The patients who presents chronic diseases, advanced physical or mental deterioration and minor self-sufficiency are either malnourished or at high risk of being. The individualized nutritional evaluation must be performed routinely when the patient is admitted to the hospital for detect and treat early signs of malnutrition.  相似文献

The effect of maternal malnutrition on the energy metabolism of the developing rat placenta, related to fetal growth, was studied. Female virgin rats of the Wistar strain were fed restricted amounts of 25% casein diet, from puberty and throughout pregnancy. According to results, a significant decrease occurred in the activity of adenylate kinase, as well as a significant increase in the energy charge of the adenylate system (ATP, ADP), per gram of DNA of placental tissue near term, in the malnourished group. The data suggest that the significant increase of energy charge (ATP + 1/2 ADP:/ATP + ADP + AMP) of the placenta in the malnourished group, is the consequence of an inhibition of the reactions controlling ATP-consuming process, such as macromolecular synthesis pathways and active transport of substrates near term. This coincides with the simultaneous and significant decrease in fetal growth in this group.  相似文献

Rapidly growing neonatal mammals accrete relatively large quantities of long chain (≥C20) polyunsaturated fatty acids (LCP) in membrane phospholipids. We have examined accumulation of ω6 LCP in suckling neonatal rat pups during the first 14 d of life when their dams received essential fatty acids in the form of triglycerides containing linoleic acid or arachidonic acid. Dietary levels of these fatty acids were either 1 or 5% of total dietary fatty acids. The fatty acid profile of pup stomach contents (composed solely of the dams' milk) and plasma lipids, as well as liver and brain phospholipids, were determined. Stomach linoleic and arachidonic acid levels reflected the diet of the dams. Pup plasma and liver arachidonic acid levels increased progressively from the group receiving 1% linoleic acid to 5% linoleic acid and from 1% arachidonic acid to 5% arachidonic acid. Interestingly, brain phosphatidylethanolamine and phosphatidylcholine arachidonic acid levels were more stable than plasma or liver levels. These results suggest that the brain may be capable of either selective transport of ω6 LCP or chain elongation/desaturation of linoleic acid. These data indicate that care must be exercised when adding LCP to infant formula since widely divergent accretion rates of arachidonic acid may occur in various tissues.  相似文献

We aimed to assess the effects of maternal supplementation with the main fat sources used in the human Western diet (olive oil, butter, margarine) on milk FA composition and on plasma FA profile of offspring, and to determine whether it may influence body-weight-gain (BWG) and adiposity of offspring during the suckling period. Wistar rats were supplemented with the different fat sources from day 14 of gestation and throughout lactation. Olive oil-supplemented dams showed the highest proportion of oleic-acid in milk, with no changes in plasma. Their offspring also showed the highest proportion of this FA in plasma, lower BWG during the suckling period, and higher levels of UCP1 in brown adipose tissue (BAT) at weaning. Margarine-supplemented dams showed the highest percentage of PUFA in milk, and a similar tendency was found in plasma of their offspring. Butter-supplemented dams displayed higher proportion of saturated FA (SFA) in milk compared to other fat-supplemented dams, but lower than controls. Control offspring also showed higher proportion of SFA in plasma and greater BWG during the suckling period than fat-supplemented groups. Significant correlations were found between the relative content of some milk FA and BWG of offspring, in particular, oleic-acid levels correlated negatively with BWG and positively with UCP1 levels. These results show that maternal dietary source of fat affects milk FA composition and circulating FA profile, as could be expected, but also BWG and thermogenic capacity of offspring during the suckling period. An effect of oleic-acid stimulating BAT thermogenic capacity of suckling pups is proposed.  相似文献

One hundred female adolescents (13-18 y) were clinical and anthropometrically studied to select only those with adequate nutrition. Most adolescents belonged to IV socio-economic stratum families (worker class). Height, weight, age, body mass index and medial arm circumference were used as anthropometric parameters. After screening, only 41 non pregnant girls (control) and 42 pregnant girls with adequate nutrition were selected to analyze plasma amino acids. Fasting peripheral venous blood was drawn, and plasma amino acids were analyzed by HPLC. Amino acid concentrations were expressed as umol/L +/- SE. SAS/STAT program was used for statistical analysis. Amino acid values of control adolescent group were found in ranges reported by other investigators, with slight variations, mostly in diminution, presumably due to nutritional, metabolic or genetic conditions of people living in tropical regions. In pregnant healthy adolescents, distributed according to gestational age: < 32 weeks (n = 30) and > 32 weeks (n = 12), a diminution of total molar plasma amino acids was found, by comparing with control values. Ten amino acids (Pro, Gly, Gln, Arg, Ser, Orn, Tau, Leu, Thr and Val) appeared significantively diminished throughout gestation, being Gly. Gln and Arg most affected since earlier weeks. During the 2nd period. Thr and Val increased their grade of affectation; whereas some amino acids values (Orn, Pro and Tau) tended to recuperate. Several of affected amino acids are gluconegoenic, thus, they could be utilized to supply the energy required by the pregnant adolescent against her double stress: the fetus development and her own development. The plasma amino acid values reported in both, healthy non pregnant and pregnant adolescents, could be taken as regional referential profile of plasma amino acids in this poblational group for further research on adolescent and fetal--maternal malnutrition.  相似文献

The accumulation of fatty acids in retina, brain, liver, and plasma of 30-day-old rat pups consuming various levels of linoleic acid (LA, 18:2n-6) and constant α-linolenic acid (ALA, 18:3n-3) is reported. Dams were fed graded levels of LA during gestation and lactation, and the pups were maintained on the diet of their dams until the end of the brain growth spurt at 30 d of life. Milk, and pup brain, retina, liver, and plasma were analyzed quantitatively for fatty acid profile. The percentage of docosahexaenoic acid (DHA, 22:6n-3) in retina increased from an LA-deficient dietary level, peaked at the 9:1 (LA/ALA) level, then fell for the 41:1 and 69:1 levels. In contrast, the brain DHA percentage was unaffected by dietary LA levels. Retinal unsaturated fatty acid levels paralleled liver and plasma levels. The milk fatty acid composition mirrored the diets. These data show that the retinal fatty acid composition responds sensitively to dietary fatty acid composition, similar to liver and plasma, while the brain unsaturate composition is nearly independent of dietary composition.  相似文献

Serum zinc was measured in 103 Brazilian children from low income families, 3 months to 6 years of age, with the purpose of characterizing their zinc nutritional status. Serum albumin was determined in some of the children studied. They were grouped in three categories according to the adequacy of height for age and weight for height indices related to the growth curves determined for Brazilian children. There were 30 normal, 18 acutely malnourished and five chronically malnourished children. This criteria was complemented by the Gomez criteria. Thirty healthy adults of both sexes served as a reference group for the analytical procedures. In the adults, serum zinc was 107.5 +/- 14.5 micrograms/dl, similar to values observed by other investigators, and significantly higher than in all of the children studied. Serum zinc of normal children was 98.3 +/- 15.7 micrograms/dl. It decreased with the degree of malnutrition, being significant for second degree acutely malnourished (87.8 +/- 9.2 micrograms/dl; p less than 0.05) and chronically malnourished children (68.7 +/- 8.9 micrograms/dl; p less than 0.001). Serum zinc and albumin correlated significantly (r = 0.51; n = 37; p less than 0.01), particularly in the severely malnourished children (r = 0.63; n = 8; p less than 0.05).  相似文献
