PurposeTo evaluate and compare the distribution of refractive power within the optic zone of different soft contact lenses and to investigate the effect of lens decentration on the power profiles.MethodsThe Nimo TR1504 instrument was used to measure the optical power across different aperture diameters (from 1.5 mm to 5.5 mm in steps of 0.5 mm) of four daily disposable contact lenses: DAILIES TOTAL1, Proclear 1-Day, SofLens daily disposable and 1-DAY ACUVUE MOIST. Measurements were performed using a wet cell. Power data were evaluated when contact lenses were in its centered position and after inducing different amounts of lens decentration (from 0.2 mm to 1.0 mm in steps of 0.2 mm).ResultsAll contact lenses showed an increase – more negative – in lens power with distance from the lens center. The amount of change varied depending on the lens. It was about 10% of lens power for DAILIES TOTAL1 (−0.29 diopters (D)), SofLens daily disposable (−0.36 D), and Proclear 1-Day (−0.32 D) whereas 1-DAY ACUVUE MOIST showed a percentage variation of 3.3% (−0.10 D). After inducing a lens decentration up to 1 mm, the power curves were shifted in the negative direction. However, the change obtained in lens power compared with well-centered position was always lower than a quarter of diopter both for all the lenses and aperture diameters.ConclusionsOur results showed a variation of the refractive power from the lens center, becoming more negative toward the periphery, with a negligible effect of the decentration for all disposable contact lenses studied.  相似文献

Hyaluronan, a member of the glycosaminoglycan family of biological polysaccharides, is a high-molecular-weight disaccharide polymer found throughout the human body, particularly in the eye. Bausch + Lomb Biotrue? multi-purpose solution contains hyaluronan as a lens conditioning agent. The retention of hyaluronan from Biotrue multi-purpose solution to a variety of hydrogel contact lenses was evaluated over time. Fluorescein-tagged hyaluronan was allowed to adhere to lenses, which were then rinsed with balanced salt solution at a rate comparable to human tear secretion. Results demonstrated that hyaluronan was released slowly throughout the rinse period. The chemistry of the lens materials appeared to contribute to the hyaluronan retention capacity for each lens type. The results suggest that a multi-purpose solution containing hyaluronan has the potential to provide lens conditioning regardless of the hydrogel contact lens used.  相似文献

PurposeTo evaluate the power profile of multifocal contact lenses (CLs) using a new technology based on quantitative deflectometry.MethodsThe Nimo TR1504 was used to analyze the power distribution of multifocal CLs. The CLs under study were: Air Optix Aqua Multifocal Low, Medium and High Addition and Focus Progressives. Three lenses of each model were considered.ResultsAll multifocal CLs showed a power profile characterized by a change toward more positive power values when aperture sizes become smaller. The near refractive addition of the lenses under study was +2.61 D, +1.44 D, +1.30 D and +0.30 D for the Focus Progressives, the Air Optix Aqua Multifocal High, Medium Add and Low Add, respectively. The refractive power of the Focus Progressives did not reach the value of the nominal distance power until a radial distance of 0.9 mm from the center of the lens. For the Air Optix Aqua Multifocal Low Add the distance nominal power was reached at a radial distance of 1.5 mm from the center of the lens, whereas this occurred at a distance of 1.8 mm for the Air Optix Aqua Multifocal Medium and High Add.ConclusionThe relation between the pupil diameter of the patients and the power profile of these CLs has a crucial implication on the final distance correction and near addition that these lenses provide to patients. Practitioners should know the power profile of these CLs and measure the pupil diameter of each patient in different situations in order to carry out a customized fitting.  相似文献

Smoking is a risk factor for the development of microbial keratitis and corneal infiltrates in contact lens (CL) wearers. It is still unknown if this risk is directly associated with the presence of nicotine in the eye and if adherence of nicotine on the CL can enhance these effects. A better understanding of the interaction between nicotine and CL materials could offer insights to explain this risk associated with smoking. The aim of this work was to compare the affinity of nicotine to different soft CL materials. CLs from FDA groups I, II, IV, and V were incubated in a 2-mM nicotine solution for 24 h and then in a 0.9% saline solution for the next 24 h. The amount of absorbed and released nicotine per CL was deduced as a function of time (t) by ultraviolet (UV) spectrophotometry and normalised to the mass of the hydrated CL. The data were described by the equation y = b –a t?1, where a and b are constants, and b represents the mass reached at the plateau after ~ 10 min of exposure. Groups IV and V displayed the highest (0.80 ± 0.09 µg) and lowest (0.27 ± 0.08 µg) nicotine absorption per mg of hydrated CL, respectively. The CL affinity for nicotine could be ascribed to the interaction between the positive charge of nicotine pyrrolidine nitrogen and the negative charges of the CLs, especially for the ionic IV group.  相似文献

PurposeTo compare the potential of different soft contact lenses to be contaminated with Acanthamoeba castellanii as a function of material parameters and cleaning procedures.MethodsDifferent unworn soft hydrogel and silicone hydrogel contact lenses were incubated with human pathogenic A. castellanii. The adhesion of the acanthamoebae was investigated on the contact lenses and put into relation to their material parameters. The efficacy of a recommended contact lens cleaning procedure in reducing A. castellanii adhesion was investigated.ResultsWe found that material parameters such as elastic modulus, silicone content, ionic properties and swelling do not influence the adhesion of acanthamoebae to soft contact lenses. A material parameter that influenced adhesion significantly was the water content of the lens. With increasing water content, the adhesion of acanthamoebae increased. By following the cleaning instructions of the manufacturer the contamination of the lenses with A. castellanii could be reduced to a minimum, as shown both on contact lenses and in control experiments.ConclusionWith this study we show that for the tested lenses, the adhesion of A. castellanii to contact lenses is independent of the silicone content of the lens, but depends nonlinearly on the water content of the lens. Furthermore, we demonstrate that applying proper lens cleaning procedures minimizes the risk of acanthamoebae adhesion to contact lenses.  相似文献

Lactoferrin (LF), an iron-binding protein with antioxidant activity, is significantly reduced in the lacrimal film of patients affected by keratoconus (KC) compared to healthy subjects, and this is supposed to be the cause for tear iron increase and consequent iron deposition and oxygen free radical accumulation in the cornea. We decided to study if LF-loaded contact lenses (LF-CLs) could exert antioxidant activity on epithelial cells incubated in tears collected from two patients affected by keratoconus (KC1 and KC2). Moreover through this model we indirectly estimated iron concentration in the tears of healthy and KC subjects.Reflex tears were collected during the first 2 min of light or onion-induced lacrimation and stored at −20 °C. After incubation in tears for 18 h, mortality of epithelial cells was investigated by trypan blue exclusion test. Successively, LF-CLs were deposed on cells incubated in KC1 tears. For the indirect determination of iron content, cells were incubated with LF 1.2 mg/mL and different FeSO4 concentrations, and for the estimation of iron from the patient’s tears, cells were incubated with free serum medium and healthy tears (1:1) and different FeSO4 concentrations.Epithelial cells incubated with reflex tears of KC patients showed increased mortality (27.7 ± 3.9%, p = 0.0003, for KC1 and 17.6 ± 0.95%, p = 0.014, for KC2) compared to epithelial cells maintained in control healthy tears (8.6 ± 1.2%). This difference in mortality was correlated with tear iron concentration, which was estimated at 4.58 μg/mL for the healthy subjects, at 56.28 μg/mL for KC1, and at 8.7 μg/mL for KC2 patient. Application of LF-CLs counteracted KC tear cytotoxicity restoring viability obtained in the presence of control tears.Therapeutic contact lenses obtained by LF loading can reduce oxidative stress induced by patients’ tears and might represent an efficient device to arrest the progression of keratoconus.  相似文献

Oil/water (o/w) microemulsions containing propranolol were studied. Isopropylmyristate was used as the oil, Tween 60 as a surfactant, butanol as a co-surfactant and a buffer of pH 6.5 for the continuous phase. The lipophilicity of propranolol was enhanced by formation of lipophilic ion-pairs to obtain a disperse phase that could act as a reservoir; octanoic acid was used as counter-ion. The diffusion rates of propranolol from microemulsions through a hydrophilic membrane decreased as the concentration of octanoic acid increased. The apparent permeability constant, determined at the beginning of the experiment, was 2.9 × 10−9 cm s−1 for propranolol alone and diminished until 3.5 × 10−10 cm s−1 in the presence of octanoic acid, at a concentration of 16.4 times that of propranolol.  相似文献

Two experimental spectroscopic methods for the determination of the content of hyaluronic acid in solution are provided based on the formation of complexes between the hyaluronic acid and cationic dyes with sensitivity down to concentrations of either 0.25 or 2.5 μg/ml. These techniques were applied to commercial hydrophilic contact lenses and allowed (i) distinguishing among contact lenses either loaded or non-loaded with hyaluronic acid, (ii) determining, for the loaded ones, the content of hyaluronic acid, (iii) evaluating the release of hyaluronic acid by the lens in solution, and (iv) determining the role of the hyaluronic acid in preventing the adsorption and the successive release by the contact lens of other components which can be present, for example, in the conservation solutions and/or in the blisters of the lenses. The results represent new physical chemistry insights for the field of contact lenses with impact on the advancement of current technology.  相似文献

PURPOSE: Polyvinyl alcohol (PVA) is a successful tear film stabiliser and is widely used in comfort drops and some soft contact lens materials. A PVA-containing lens, nelfilcon A has been modified to include additional (non-functional) PVA in order to provide improved comfort. This study aims to examine the clinical performance of this nelfilcon A lens with AquaRelease (AquaRelease). METHODS: Two contralateral, investigator masked, open label, subjective and objective evaluations were conducted. The first examined the effect of adding increased molecular weight PVA to nelfilcon A (n=5), and the second compared this AquaRelease lens to ocufilcon B (n=34). The principal measures were non-invasive break-up time (NIBUT) and subjective comfort, which were assessed at the beginning and end of a week of daily wear, and three times throughout 1 day at 8, 12 and 16 h. RESULTS: All subjects successfully completed the daily wearing schedule of 16h. On initial insertion, subjective comfort and NIBUT improved for AquaRelease than original nelfilcon A lenses (p<0.05). Initial comfort was better for AquaRelease compared to ocufilcon B lenses (p=0.01); however, NIBUT was not statistically different (11.7+/-15.6s versus 8.4+/-6.8s; p=0.26). Subjective comfort decreased with time (p<0.001), but there was no significant difference between AquaRelease and ocufilcon B lenses (p=0.16). NIBUT was not significantly affected by time (p=0.56) or between lenses (p=0.33). At the end of a weeks' wear, subjective initial, end-of-day, overall comfort and vision were rated significantly better with AquaRelease than ocufilcon B (p<0.01). CONCLUSIONS: Release of additional non-functionalised PVA from the nelfilcon A lenses appears to enhance comfortable contact lens wear.  相似文献

PurposeThe purpose of this study was to evaluate the uptake and release of radiolabelled polyhexamethylene biguanide (PHMB) on reusable daily wear contact lenses (CLs) over 7 days.MethodsThree silicone hydrogel (SH) contact lens materials (lotrafilcon B, balafilcon A, senofilcon A) and two conventional hydrogel (CH) materials (etafilcon A, omafilcon A) were examined. In experiment 1 (1-day study), CLs were soaked in 2 mL of phosphate buffered solution (PBS) containing radiolabelled 14C PHMB (1 µg/mL) for 8 h. The release kinetics of 14C PHMB from the CLs was measured at t = 0.25, 0.5, 1, 2, 4, 8, and 24 h in fresh 2 mL PBS. In experiment 2 (7-day study), the CLs were soaked in the 14C PHMB (1 µg/mL) solution for 8 h followed by a 16-hour release in 2 mL PBS. The lens cycle was repeated daily for 7 days. After both experiments, the residual amount of PHMB remaining within the lenses was extracted to determine the total uptake of PHMB.ResultsIn experiment 1, the total uptake of PHMB for etafilcon A was significantly greater than senofilcon A (p = 0.01). There were no significant differences in total uptake of PHMB between other lens materials (p > 0.05). Etafilcon A released more PHMB compared to all other lens types over a 24-hr period (p < 0.001). In experiment 2, all CL materials continued to sorb more PHMB over time (p < 0.001). By day 7, the amount of PHMB sorbed by etafilcon A was significantly greater than senofilcon A (p = 0.02). After day 2, the CH materials released significantly more PHMB than the SH materials (p < 0.01).ConclusionThe CL materials continued to sorb PHMB with no signs of saturation after 7 days. All lens materials released a consistent amount of PHMB each day. Radioactive labelling provides a sensitive method of assessing the uptake and release of PHMB from CL materials.  相似文献

Cold-set oil-loaded protein gels based on an emulsifying step followed by Ca2+-induced gelation of pre-denatured β-lactoglobulin (β-LG) have been recently developed. In vitro release and stability of a fat-soluble compound (α-tocopherol) therein were investigated in this work. Release of α-tocopherol was found to be controlled mainly by matrix erosion due to protein degradation. Compound release and matrix erosion were almost complete after incubation under gastric or intestinal conditions for 6.5 h. However, both processes were basically inhibited upon changing the dissolution medium from the gastric to the intestinal type, possibly due to β-LG partial hydrolysis products with greater emulsifying capacity anchoring to the surface of gel oil droplets. The stability of released α-tocopherol was apparently improved by binding to protein and/or hydrolysis products thereof.  相似文献

Faeces and the residues from the in vitro digestion of 13 forages were sequentially extracted with acid-pepsin, ethyl alcohol, hot water and diethyl ether and the quantity of the extracts was adjusted to a protein-free basis and expressed per 100 g of original forage organic matter. Undigested material inin vivo and in vitro experiments were respectively, 3·37 and 1·88 g (±0·12) for ethyl alcohol extracts, 1·13 and 1·10 g (±0·05) for hot water extracts, 0·15 and 0·12 g(±0·02) for diethyl ether extracts and 1·44 (±0·22) and 0g for 0·1 N -HCI extracts. The difference between values for in vivo and in vitro experiments was considered to represent endogenous material present in faeces.  相似文献

BACKGROUND: Higher plants possess several mechanisms of defense against plant pathogens. Proteins actively synthesized in response to those stresses are called defense‐related proteins which, among others, include certain protease inhibitors. It is of particular relevance to investigate plant natural defense mechanisms for pathogen control which include cystatins—specific inhibitors of cysteine proteases. RESULTS: In this study, a cysteine proteinase inhibitor (CPI), 11 kDa in size, was purified from green kiwifruit to homogeneity. Immuno‐tissue print results indicated that CPI is most abundant in the outer layer of pericarp, near the peel, and the inner most part of the pulp—sites where it could act as a natural barrier against pathogens entering the fruit. The purified protein (15 µmol L?1) showed antifungal activity against two phytopathogenic fungi (Alternaria radicina and Botrytis cinerea) by inhibiting fungal spore germination. In vivo, CPI (10 µmol L?1) was able to prevent artificial infection of apple and carrot with spore suspension of B. cinerea and A. radicina, respectively. It also exerted activity on both intracellular and fermentation fluid proteinases. CONCLUSION: Identification and characterization of plant defense molecules is the first step towards creation of improved methods for pathogen control based on naturally occurring molecules. 本研究通过离体和活体的抗菌实验,研究了利用CO2超临界流体萃取技术获得的蓼子朴花精油对果蔬采后病原真菌的抗菌活性作用。结果表明,在离体的条件下,蓼子朴花精油对供试菌的生长具有显著的抑制作用,抑菌圈直径均大于20 mm,属于极高敏,最小抑菌浓度为3.13 mg/m L。采用浸泡法和熏蒸法进一步研究了蓼子朴花精油的活体抑菌效果。结果显示,蓼子朴精油以熏蒸的方式处理葡萄,效果明显。在精油量为0.5 m L时,对贮藏期的葡萄鲜果具有显著的保鲜及延缓腐烂的作用。采用气相色谱-质谱联用技术从蓼子朴花精油中共鉴定出21种精油成分,其中香草醛的含量最高,达到30.35%,该成分与蓼子朴精油的抗菌活性作用密切相关。研究结果为蓼子朴花精油在当地食品工业的开发应用提供了科学依据。  相似文献

Several methods have been proposed to achieve an extended and controlled release of ocular therapeutics via contact lenses; however, the experimental conditions used to study the drug release vary greatly and significantly influence the release kinetics. In this paper, we examine variations in the release conditions and their effect on the release of both hydrophilic and hydrophobic drugs (ketotifen fumarate, diclofenac sodium, timolol maleate and dexamethasone) from conventional hydrogel and silicone hydrogel lenses. Drug release was studied under different conditions, varying volume, mixing rates, and temperature. Volume had the biggest effect on the release profile, which ironically is the least consistent variable throughout the literature. When a small volume (2–30 mL) was used with no forced mixing and solvent exchange every 24 h, equilibrium was reached promptly much earlier than solvent exchange, significantly damping the drug release rate and artificially extending the release duration, leading to false conclusions. Using a large volume (200–400 mL) with a 30 rpm mixing rate and no solvent exchange, the release rate and total mass released was significantly increased. In general, the release performed in small volumes with no force mixing exhibited cumulative mass release amounts of 3–12 times less than the cumulative release amounts in large volumes with mixing. Increases in mixing rate and temperature resulted in relatively small increases of 1.4 and 1.2 times, respectively in fractional mass released. These results strongly demonstrate the necessity of proper and thorough analysis of release data to assure that equilibrium is not affecting release kinetics. This is paramount for comparison of various controlled drug release methods of therapeutic contact lenses, validation of the potential of lenses as an efficient and effective means of drug delivery, as well as increasing the likelihood of only the most promising methods reaching in vivo studies.  相似文献

PurposeLipid deposition on contact lenses (CL) has traditionally been believed to reduce comfort during CL wear. The purpose of this study was to quantify lipid deposition on CL in a group of symptomatic and asymptomatic adapted CL wearers.MethodsThis was a single-masked, randomized clinical trial. Only confirmed symptomatic (comfortable lens wear time (CWT) < 8 h and a noticeable reduction in comfort over the course of the day) and asymptomatic (CWT > 10 h and minimal reduction in comfort over the course of the day) participants were recruited to participate in the study. Participants wore senofilcon A lenses in combination with a polyquaternium-based care solution (OPTI-FREE Replenish). Worn CL samples were collected on Day 14. Deposited lipid amounts from the lenses (including cholesteryl ester, cholesterol and triolein) were quantified using a liquid chromatography-mass spectrometry technique.ResultsLipid deposition was significantly higher in CL extracts of asymptomatic wearers compared to the symptomatic wearers for all lipid types quantified, including cholesteryl ester (2.1 ± 0.6 vs 1.6 ± 0.5 log μg/lens), cholesterol (1.5 ± 0.3 vs 1.1 ± 0.3 log μg/lens) and triolein (0.3 ± 0.2 vs 0.1 ± 0.1 log μg/lens) (all p < 0.002). The amount of cholesteryl ester deposited was greatest (p = 0.0001), followed by cholesterol, then triolein, for both the asymptomatic and symptomatic groups (both p = 0.0001).ConclusionThis study demonstrated that the asymptomatic group deposited a significantly greater amount of lipid on their CL. Although lipid levels measured are considered low to trigger any observable clinical deposition, they may influence other clinical outcomes, particularly comfort.  相似文献
