Adult male and female Japanese quail were functionally castrated by exposure to a short photoperiod and then injected with either estradiole benzoate (EB) or testosterone propionate (TP). Females injected with EB (n = 24) exhibited normal sexual receptivity again but displayed no male courtship behavior. Males injected with EB (n = 10) showed both male and female copulatory patterns. Females injected with TP (n = 14) showed little sexual behavior of any sort, while males injected with TP (n = 10) displayed male sexual behavior. The male quail is thus more bisexual than the female. In contrast with mammalian species, the female is more bisexual. Songbirds exhibit seasonal changes in the volumes of song control nuclei. Birds on long, spring-like days have larger nuclei than do birds on short, winter-like days. The mechanisms mediating volumetric changes have not been determined unequivocally, but testosterone (T) is probably involved. This study examined whether testicular factors are uniquely responsible for seasonal changes in the song system, or whether photoperiod has testis-independent effects. Male American tree sparrows were exposed to one of three photoperiodic conditions: (1) Photosensitive birds were retained on short days (8L:16D). Plasma T is rarely detected in such birds. (2) Photosensitive birds were moved from short days to long days (20L:4D) and photostimulated for three weeks. Photostimulation elevates circulating T in photosensitive birds. (3) Photorefractory birds were held at least four months on 20L:4D. Such birds seldom have detectable levels of T, even though they are on long days. In each condition, there were both intact and castrated birds. Castration typically removes circulating T in tree sparrows. The volumes of the high vocal center (HVC), nucleus robustus archistriatalis (RA), and area X were measured. Song nuclei were largest in intact photostimulated birds. Other long-day birds (i.e. castrated photostimulated, and intact and castrated photorefractory groups) had larger song nuclei than did short-day intact or castrated photosensitive birds and did not differ from each other. These data indicate that photoperiod has both testis-dependent and -independent effects on the volumes of song control nuclei.  相似文献

The affinities and densities of 2-[125I]iodomelatonin binding sites in the brains and gonads of male Japanese quail following short photoperiod treatment were studied. At 6 weeks old, control quail were placed under a 14 hour light/10 hour dark photo-stimulatory cycle and experimental quail were housed under a 7 hour light/17 hour dark photo-inhibitory lighting regime. Eighteen weeks after photic manipulation, the birds were killed at mid-light. The photo-inhibited quail had very small testes. Brains and testes of control and experimental quail were collected for receptor binding studies. The maximum number of binding sites (Bmax) of 2-[125I]iodomelatonin determined by saturation studies and the number of 2-[125I]iodomelatonin binding sites determined by a one-point binding assay in the testes of short-day quail were significantly lower (p < 0.05) than those of the testes in reproductively active birds kept under long photoperiod. There was no significant difference (p > 0.05) between the testicular Kd (equilibrium dissociation constant) values of these two groups. As for the Kd and Bmax of 2-[125I]iodomelatonin binding sites in the whole brain, there were no significant differences (p > 0.05) between the two groups. The higher level of testicular 2-[125I]iodomelatonin binding sites in photostimulated birds may be related to an up-regulation of melatonin receptors by the suppressed pineal melatonin secretion under long photoperiod. The lower testicular 2-[125I]iodomelatonin binding sites under short photoperiod may be the result of down-regulation of melatonin receptors by the stimulated melatonin pattern in the photo-inhibited birds.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Contents of learning that result from CS–unconditioned stimulus/stimuli (UCS) pairings in sexual approach conditioning were explored with male Japanese quail (Coturnix coturnix japonica). Sexual motivation of Ss conditioned to approach an arbitrary stimulus in a Pavlovian sexual conditioning paradigm was reduced by exposing them to a short photoperiod. Decreased sexual motivation resulted in a decline in sexually conditioned approach behavior (Exps 1 and 2). Responding was restored when Ss were returned to a long photoperiod (Exp 1) and when exogenous testosterone was administered (Exp 2). Decreased sexual motivation did not affect food-conditioned approach behavior (Exp 3). These results suggest that sexually conditioned approach behavior is mediated by a representation of the UCS, which is activated by the CS. Experiment 1 demonstrated that the social proximity behavior of male Japanese quail declines significantly when photostimulation is restricted to 2 hr daily, but can be restored by reinstituting the normal schedule. Changes in the photoperiod produced corresponding changes in the size of the cloacal gland, an androgen-dependent organ. The low levels of social proximity behavior and cloacal gland size of males maintained on short daily exposures to light also could be reversed by sc implants of testosterone (Experiment 2), and this recovery was to some extent sensitive to testosterone dose (Experiment 3). The present studies indicate that social proximity behavior in male Japanese quail is androgen dependent and provide a behavioral assay for neurohormonal studies of sexual behavior that does not depend on brief phasic responses. The conditioned responses of male and female Japanese quail (Coturnix japonica) were compared in a Pavlovian conditioning procedure in which presentation of a brief conditioned stimulus was immediately followed by the release of a copulation partner. Male quail vigorously approached the conditioned stimulus and were much more likely to enter the compartment housing their copulation partner than were female birds (Experiment 1). In females, sexual conditioning resulted in increased squatting (Experiment 2). This response was the reflection of sexual behavior rather than more general social behavior (Experiment 3). These findings provide the first definitive evidence of sexual learning in female quail and are consistent with the interpretation that sexual conditioning increases sexual arousal or receptivity in both sexes but the increase has different behavioral manifestations in male and female quail. Tested the reproductive response of young female deer mice to either a long or short photoperiod combined with the presence of adult males, adult females, or social isolation. 266 Ss were reared on either a long photoperiod (15 hrs light, 9 hrs dark) or on short days (8 hrs light, 16 hrs dark) from birth. Beginning at weaning, females were housed with an adult male, with an adult female, or in social isolation. In Exp I, vaginas opened more slowly in Ss on short days than in those on long days. Vaginal introitus was also retarded in Ss reared with an adult female in comparison with Ss reared in isolation. When examined at 37 days of age, Ss reared with an adult male had larger uteri than those reared alone; uteri were also larger in long-day than in short-day Ss. In Exp II, Ss were killed at 30 days of age; again, uterine growth was stimulated by exposure to either long days or an adult male. As was previously demonstrated for male deer mice, sexual maturation in females is regulated by photoperiod and social cues. Heterospecific social stimuli accelerate maturation in individuals that otherwise would be inhibited by having been reared on a short photoperiod. In male Syrian hamsters, short days induce regression of the reproductive system, but eventually spontaneous recrudescence occurs ensuing from refractoriness to the inhibitory photoperiod. Although the photoperiod of 12 L:12 D (12 h light:12 h dark) may act like a short day by inducing the testicular cycle outlined above, it may fail to evoke the increase of circulating concentrations of prolactin that accompanies testicular recrudescence. This photoperiod may fail to induce photorefractoriness, as indicated by the prolonged low concentrations of prolactin in the blood. Herein, hamsters were exposed to either 8 L:16 D or 12 L:12 D from weaning and by 28 weeks exposure to either photoperiod, the hamsters had large testes following recrudescence from a photoperiod induced-regression). Transfer to 8 L:16 D from 12 L:12 D at 28 weeks resulted in a second testicular regression and recrudescence. In a second experiment, the testes of hamsters moved to 8 L:16 D from 12 L:12 D after 29 weeks exposure to the latter photoperiod similarly regressed and then regrew. Serum concentrations of prolactin decreased in these males after transfer to the shorter daylength but also decreased in hamsters kept under 12 L:12 D both groups were usually below those of hamsters moved to 14 L:10 D. These results show that exposure for 28 or 29 weeks to 12 L:12 D was insufficient to induce photorefractoriness, despite the recrudescence of the testes following involution.  相似文献

Studied social proximity in 6 experiments, using a total of 64 Japanese quail. Following opportunity to copulate with a female housed in an adjacent compartment, males spent 64% of their time near a window that provided visual access to the female (Exp I). This social proximity behavior persisted for at least 2 wks of continual exposure to the window despite lack of further physical access to the female (Exp II) and was evident at all daylight hours (Exp III). Females stimulated more proximity behavior in male conspecifics than did other males or birds (e.g., yellow ducklings, parakeets) of other species (Exps IV and V). The females did not have to be previous sexual partners, familiar, sexually experienced, or reproductively competent to stimulate proximity behavior in males (Exps IV and V). Exp VI demonstrated that the social proximity behavior was primarily a response to the visual aspects of the females. Results are discussed in relation to mate-guarding and surveillance behavior, and implications of the data for sexual classical conditioning of male Japanese quail are noted. Six adult Soay rams were housed under artificial lighting conditions of long days (16 h light:8 h darkness) for 4 months and this caused the animals to lapse into a state of reproductive quiescence with low levels of gonadotrophins in the circulation and regressed testes secreting very low amounts of testosterone. The photoperiod was changed abruptly to short days (8 h light:16 h darkness) to induce a resurgence of sexual activity, and a detailed study was made of the pituitary and testicular responses over the first 100 days. Plasma levels of LH and FSH first began to increase between days 6 and 12 of short days, and rose progressively until days 33-54 before declining again. Testicular growth of the rams began on days 19-26 and continued for most of the remaining period of study. Plasma testosterone levels rose in parallel with the growth of the testes, and were greatly increased by day 100 when gonadotrophin levels were reduced. At most stages there were short-term fluctuations in the plasma levels of FSH, LH and testosterone indicative of episodic secretion. Peaks in plasma levels of LH were especially conspicuous and from the changes in frequency and amplitude of these peaks it was possible to predict the way in which photoperiod influenced gonadotrophin secretion by its effect on hypothalamic LH-RH secretion. A slight 24 h rhythm in the plasma levels of all three hormones was observed, and the significance of this in relation to the photoperiodic response is discussed.  相似文献

Previous research by the present authors (see record 1986-16367-001) showed that after opportunity for copulation, male Japanese quail spent about 75% of their time throughout daylight hours near a window that provided visual access to a female conspecific. In Exp I of the present study, 8 female Japanese quail typically spent less than 20% of their time near a window that provided visual access to 1 of 8 male conspecifics under comparable conditions. In Exp II, using the same Ss, a small clear plastic cage was placed in a large test arena, and Ss were observed when the cage was either empty or contained a sexual partner. Males tended to remain much closer to the cage when it contained a female conspecific than when it was empty. In contrast, the presence of a male did not significantly attract females to the cage area. These findings demonstrate strong sexual dimorphism in the social proximity behavior of Japanese quail. Results are discussed in terms of implications concerning the social structure of Japanese quail and implications for studies of sexual classical conditioning. The effects of constant photoperiod on serum prolactin concentrations and sexual behavior of ovariectomized goats was investigated. Fourteen ovariectomized goats were divided into groups of seven goats each and placed in photoperiod-controlled rooms with either 8L:16D or 16L:8D. All goats received six estradiol (E2) treatments in a Latin square design. Following each treatment six 1-h observation periods were conducted. Blood was collected before E2 treatment and at each observation period for prolactin quantification. During the observation periods measures of attractivity, proceptivity, and receptivity were recorded. Goats exposed to 8L:16D displayed sexual behavior in response to doses of E2 lower than those required by goats exposed to 16L:8D. Prolactin concentrations were higher in goats exposed to 16L:8D than in those exposed to 8L:16D. It was concluded that photoperiod affects prolactin concentrations and behavioral sensitivity to E2 in goats. The effect of continued exposure to constant photoperiod on these variables was then investigated. The goats were held in constant photoperiod for 211 d and behavior tests as described for Exp. 1 were repeated. Scores for attractivity, proceptivity, and receptivity did not differ between the two groups after 211 d of exposure to constant photoperiod. Prolactin concentrations did not differ in this study between the two groups. It was concluded that after chronic exposure to 8L:16D ovariectomized goats become refractory to the stimulatory effects of short photoperiod.  相似文献

The effects of pharmacological manipulations of dopaminergic transmission on appetitive and consummatory aspects of male sexual behavior were investigated in castrated male Japanese quail treated with exogenous testosterone. Appetitive male sexual behavior was assessed by measuring a learned social proximity response and consummatory behavior was assessed by measuring copulatory behavior per se. The nonselective dopamine receptor agonist, apomorphine, inhibited in a dose-dependent manner both components of male sexual behavior. Two indirect dopamine agonists were also tested. Nomifensine, a dopamine re-uptake inhibitor, decreased appetitive sexual behavior but increased the frequency of mount attempts, a measure of consummatory sexual behavior. Amfonelic acid, a compound that enhances dopaminergic tone by a complex mechanism, increased aspects of both appetitive and consummatory behaviors. These data suggest that, in quail, as in rodents, increases in dopaminergic tone facilitate both appetitive and consummatory aspects of male sexual behavior. Apomorphine may be inhibitory in quail because it acts primarily on D2-like receptors, unlike in rats, where it stimulates sexual behavior and acts primarily on D1-like receptors at low doses but interacts with D2-like receptors at higher doses. This is supported by the observation that stereotyped pecking, a behavior stimulated selectively in quail by D2 agonists, was increased by apomorphine but not by the two indirect agonists. The observed partial dissociation between the effects of these dopaminergic agonists on appetitive and consummatory sexual behaviors suggests that these two components of male sexual behavior may be controlled by the action of dopamine through different neuronal systems.  相似文献

The role of estrogen and progesterone in the regulation of reproductive behavior was studied in female ring doves (Streptopelia risoria) under long vs. short photoperiods. The doves held under long (16 hours/day) or short (8 hours/day) photoperiods were treated for 15 days with saline, estrogen, or progesterone, alone or in combination. 7 days after the start of hormone therapy, the doves were placed with reproductively active males for 4 hours/day for 9 days. No difference was seen in egg-laying, courtship or nest-building by control birds under the 2 photo regimes. Ovarian follicles remained small in all doves receiving hormone treatment. Oviducts of doves receiving progesterone alone remained small in the short-photoperiod group but at long photoperiods the oviducts became enlarged. Estrogen stimulated oviduct growth at both photoperiods wheras the combined hormone treatment resulted in greater oviduct development than estrogen alone (p less than .02). Female soliciting and nest-building activity remained low in progesterone-treated doves under short photoperiod but increased rapidly under long photoperiod. Doves treated with estrogen and estrogen plus progesterone performed considerable soliciting and nest-building. No marked tendency was seen for doves under long-photoperiod conditions to be more active in nest-oriented behavior. In Exp I, 8 sexually deprived male Japanese quail were exposed to 1 of 7 female Japanese quail until they satisfied a 20-min sexual satiation criterion. The female was then replaced with a novel female or was allowed to remain in the test cage undisturbed. Introduction of the novel female did not reinvigorate sexual behavior above levels observed when the female was allowed to remain undisturbed. In Exp II, with 9 males and 15 females, males that were not sexually deprived were tested in the presence of 2 females. During experimental tests, 1 of the females was novel and the other was familiar. During control tests, both of the females were familiar. The presence of a novel female increased overall levels of male sexual behavior and also increased the number of males engaged in reproductive behavior. However, the enhanced sexual behavior was not differentially directed toward the novel or familiar female. Implications of these findings for the mating system of Japanese quail and for studies of the role of novelty in sexual behavior are discussed. Three experiments were conducted to identify species-specific sign stimuli sufficient to elicit copulatory behavior in male Japanese quail and to determine how learning is involved in the control of behavior by these sign stimuli. In Exp 1, sexually experienced Ss were tested for copulatory behavior with a live female quail and with a model consisting of a female quail's head and neck mounted in front of a foam pad. Comparable levels of copulatory behavior were observed in the two tests, indicating that static visual cues provided by a female quail's head and neck are sufficient to elicit copulatory behavior in this species. Exp 2 showed that male birds that previously received numerous opportunities to copulate with a live female quail in the test situation were significantly more likely to copulate with the head?+?neck model than were sexually inexperienced Ss. Exp 3 showed that prior sexual experience with live quail facilitated responding to the head?+?neck model only if the sexual experience was provided in the same place where Ss were later tested with the model. This finding suggests that sexual experience facilitates control of copulatory behavior by species-specific sign stimuli through contextual conditioning. The temporal relationship between testicular maturation and salmon gonadotropin-releasing hormone (sGnRH) mRNA expression was investigated in underyearling precocious male masu salmon, Oncorhynchus masou. Testicular maturation could be experimentally manipulated by changing the length of the light-dark photoperiod; maturation was accelerated in the short photoperiod group (8L-16D) and delayed in the long photoperiod group (16-8D). sGnRH mRNA and total silver grains in these loci in individual fish, increased with advancing testicular maturation. They were maximal in the short photoperiod group in August and in the long photoperiod group in September, when spermiation occurred. In contrast, marked changes in sGnRH synthetic activity in relation to testicular maturation were not observed in the terminal nerve ganglion or in the olfactory bulbs. sGnRH neurons in the preoptic area and the ventral telencephalon are clearly influenced by photoperiod and are involved in the control of gonadal maturation probably via gonadotropin secretion.  相似文献

Previously, we found that, unlike adults, adolescent male dark-eyed juncos (Junco hyemalis) maintained large Area X volumes despite having low plasma testosterone concentrations. Other studies indicate that photoperiod may act independently of testosterone to modulate vocal control region (VCR) volumes in adult songbirds. In the present study, we investigated the effects of testosterone and photoperiod on the volumes of four VCRs in adolescent male juncos. To test the hypothesis that VCR volumes in these males are testosterone independent, we treated birds exposed to short days with testosterone and later compared their VCR volumes with those of birds exposed to short days without testosterone. To examine whether photoperiod alone could affect VCR volumes independent of testosterone, we measured these volumes in photorefractory birds exposed to long photoperiod without testosterone. Administering testosterone induced singing, yet increased the volume of only one VCR, the robust nucleus of the anterior neostriatum (RA). In contrast, long photoperiod increased several VCR volumes (Area X, higher vocal center, and RA) despite low testosterone levels, but did not induce singing. Our results suggest a limited role for testosterone, but an important role for photoperiod, in controlling VCR volumes in adolescent male juncos. In addition, the results demonstrate that singing behavior can be induced in adolescent males without a concomitant increase in most VCR volumes.  相似文献

The medial preoptic area (mPOA) is a key site for the dopaminergic enhancement of male sexual behavior. Dopamine release increases in the rat mPOA with mating, supporting the critical stimulatory role played by preoptic dopamine on male sexual behavior. However, it has been questioned whether dopamine is specifically related to the occurrence of male sexual behavior and not simply involved in general arousal. To address this question, we asked whether dopamine release in the mPOA is linked to the production of male sexual behavior in Japanese quail (Coturnix japonica), a species that exhibits a much shorter temporal pattern of copulation than rats and does not have an intromittent organ, resulting in a very different topography of their sexual response. Extracellular samples from the mPOA of adult sexually experienced male quail were collected every 6 min before, during, and after exposure to a female using in vivo microdialysis and analyzed using high-performance liquid chromatography with electrochemical detection. Extracellular dopamine significantly increased in the presence of a female and returned to baseline after removal of the female. However, quail that failed to copulate did not display this increased release. These findings indicate that it is not solely the presence of a female that drives dopamine release in males, but how a male responds to her. Furthermore, in quail that copulated, dopamine release did not change in samples collected during periods of no copulation. Together, these findings support the hypothesis that dopamine action in the mPOA is specifically linked to sexual motivation and not only to copulatory behavior or physical arousal. Two experiments investigated the role of estrogens in the activation and sexual differentiation of appetitive sexual behavior (ASB) in Japanese quail (Coturnix japonica) as measured by a learned social proximity response. Injection of the aromatase inhibitor R767 13 in castrated, testosterone (T)-treated male quail completely suppressed ASB, confirming that, like consummatory sexual behavior, ASB is mediated by T aromatization. ASB is not observed in female quail, even if they are treated with T as adults. The role of embryonic estrogens in the sexual differentiation of ASB was tested by blocking estrogen synthesis in ovo. Control male and T-treated female quail deprived of estrogens during embryonic life learned the social proximity response used to assess ASB, whereas control female quail did not, despite the presence of high T. Thus, ASB is demasculinized by the action of embryonic estrogens during ontogeny as is consummatory behavior.  相似文献
