A series of non-ionic surfactants were prepared from bisphenol derived from acetone (A), acetophenone (AC) and cyclohexanone (CH). The prepared bisphenols were ethoxylated at different degrees of ethylene oxide (27, 35, 43). The ethoxylated bisphenols were non-esterified by fatty acids; decanoic, lauric, myristic, palmitic, stearic, oleic, linoloic and linolinic. Some surface active properties for these surfactants were measured and calculated such as, surface tension [γ], critical micelle concentration [CMC], minimum area per molecule [Amin], surface excess [Cmax], free energy of micellization and adsorption [ΔGmic] and [ΔGads]. At a certain temperature, the cloud point was measured for these surfactants. From the obtained data it was found that; the cloud point is very sensitive to the increase of the alkyl chain length, content of ethylene oxide and degree of unsaturation. The core of bisphenol affected the cloud point sharply and they are ranked regarding bisphenol structure as BA > BCH > BAC. By inspection of the surface active properties of these surfactants, a good relation was obtained with their cloud points. The data were discussed on the light of their chemical structures.  相似文献

In this work, three aromatic amines (p-toluidine, p-nitroaniline and p-chloroaniline) were chosen as bases for the repatriation of some nonionic polyesters. These amines were ethoxylated with different total number of ethylene oxide units 6, 12, 18. The prepared ethoxylated amine diols were polyesterified with maleic anhydride and polypropylene oxide polyethylene oxide block copolymers in polyesterification reaction. The demulsification efficiency of these demulsifiers was investigated using the bottle test. The effects of the molecular weight, concentration, asphaltene content, water content, Hydrophile Lipophile Balance (HLB) and temperature on the demulsification efficiency were investigated. The surface active properties were correlated with their demulsification efficiency. It was found that, NAE18D gave the best result in the demulsification process. The demulsification efficiency was discussed on the light of surface active properties, interfacial tension and the factors affecting the demulsification. The surface-active properties of the prepared demulsifiers were measured at 60 °C.  相似文献

This study evaluated the effectiveness of a supercritical carbon dioxide (SCCO2) system, with a gas–liquid porous metal contactor, for reducing Escherichia coli K12 in diluted buffered peptone water. 0.1% (w/v) buffered peptone water inoculated with E. coli K12 was processed using the SCCO2 system at CO2 concentrations of 3.1–9.5 wt%, outlet temperatures of 34, 38, and 42 °C, a system pressure of 7.6 MPa, and a flow rate of 1 L/min. Increased CO2 concentrations and temperatures significantly (P < 0.05) enhanced microbial reduction. A maximum reduction of 5.8-log was obtained at 8.2% CO2 and 42 °C. To achieve a 5-log reduction of E. coli K12 in 0.1% buffered peptone water, minimum CO2 concentrations of 9.5%, 5.5%, and 5.3% were needed at 34, 38, and 42 °C, respectively. Further reductions of cells were observed after storage for 7 days at 4 °C. But storage at 25 °C increased the number of viable cells to 8-log cfu/mL after 7 days. This study showed the potential of the pilot scale SCCO2 system with a gas–liquid porous metal contactor for microbial inactivation in liquid food.  相似文献

A standing wave field of ultrasound was established in the designed standing wave tube. The enhancement of ultrasound irradiation on desalting and dewatering process of LU-NING pipeline oil was studied. The results showed that the best conditions of the oil pretreatment was: adding 30 mg/L demulsifier, injecting 5% (v.) wash water, irradiating the crude oil 5 min by 10 kHz ultrasound (intensity 0.38 W/cm2, standing wave field), settling for 90 min. The final results of salt and water contents were 3.851 mg/L and 0.37%, respectively. The effect of standing wave field combined with demulsifier was better than that of the demulsifier only. High temperature (80 °C) in the process was better than low temperature. Salt balance study showed that the key of desalting was: the wash water and the oil should be mixed sufficiently to extract the salts into the water as more as possible, the water added in should be provided to be separated out. If one stage of desalting process could not achieve the norm, two stages of desalting process should be considered.  相似文献

This study was undertaken to determine the frequency of potentially pathogenic Escherichia coli (E. coli) strains among E. coli isolated from Casablanca, Morocco. The E. coli strains were isolated from ground beef (n = 140), turkey (n = 200), sausage (n = 120), seafood (n = 60), domestic water (n = 35) and well water (n = 50). The prevalence of E. coli was 48%, 45%, 35.5%, 30%, 8.3%, 0%, for well water, ground beef, turkey, sausage, sea food and domestic water, respectively. Two hundreds E. coli isolates were tested for the presence of 17 virulence genes associated with strains causing intestinal and extra-intestinal infections. The virulence genes included stx1, stx2, lt, st, hlyA, aggA, saa, astA, iucD, cnf1, eaeA, bfpA, ial, ipaH, afa, pap and sfa. PCR showed that 37% (74) of E. coli isolates carried one or more of these virulence genes. No virulence genes were found in E. coli strains isolated from sea food samples. In contrast, 10% of the ground beef samples, 18% of the turkey samples, 17.5% of sausage samples and 6% of well water contained specific factors for intestinal E. coli pathogens.  相似文献

In recent years, there has been an increased interest in food processing technologies that could lessen the thermal impact on food products. In the present study, thermosonication (TS) and pulsed electric fields (PEF), applied individually or in combination (TS/PEF), were investigated to determine their effects on inactivation and sub-lethal injury of Pseudomonas fluorescens and Escherichia coli. TS was applied at a low (L) and high (H) wave amplitude (18.6 μm and 27.9 μm, respectively), while PEF was applied at a low and high electrical field strength (29 kV cm−1 and 32 kV cm−1, respectively). In addition, the inhibitory effects of TS/PEF combined were assessed. For P. fluorescens, when applied individually, TS and PEF resulted in ≤9% and ≤47% inactivation, respectively, with 8% sub-lethal injury following PEF treatment. However, TS/PEF treatment caused ≤48% inactivation and ≤34% sub-lethal injury, respectively. For E. coli, TS caused ≤6% inactivation, and ≤2% sub-lethal injury, while PEF treatment alone caused inactivation and sub-lethal injury of 86% and 29%, respectively. TS/PEF caused a maximum of 66% inactivation, while sub-lethally injuring approximately 26% of the of E. coli population. The present study confirms the ability of TS and PEF to inactivate microorganisms, but shows that some bacteria were not killed, but sub-lethally injured.  相似文献

Lauricidin and lactic acid were evaluated for their effects on growth and survival of Listeria monocytogenes (L55), Salmonella enteritidis (S552) and Escherichia coli O157:H7 (E19) inoculated onto raw chicken breast. Fresh, raw chicken breasts were purchased immediately after slaughter and transported on ice to the laboratory within 20 min. Each chicken breast was decontaminated by briefly dipping in 70% ethanol and passed through a flame of a Bunsen burner and then allowed to cool. The decontaminated Chicken breast was dipped in TSB broth, at room temperature (25 °C) for 15 min, containing approximately log 9 CFU/ml of L. monocytogenes, S. enteritidis or E. coli O157:H7. Initial counts of L. monocytogenes, S. enteritidis or E. coli O157:H7 counts in chicken breast immediately after dipping in TSB broth were in the range of log 7–log 8 CFU/g. After inoculation, the chicken breasts were kept at room temperature for 20 min to allow attachment. Each inoculated chicken breast (25 °C) was dipped in 0 (control – sterile water), 0.5%, 1%, 1.5% or 2% of lauricidin (w/v) or lactic acid (v/v) for 10, 20 or 30 min and then individually placed in oxygen-permeable polyethylene bags. Breasts were subjected to microbiological analyses after treatment (day 0) and after storage for 2, 5, 7, 10 and 14 d at 4 °C. Initial counts of L. monocytogenes, S. enteritidis and E. coli O157:H7, in chicken breast treated with lauricidin decreased by 2.90, 1.31 and 2.27 log CFU/g, respectively. Lauricidin was more effective in reducing L. monocytogenes population than S. enteritidis and E. coli O157:H7 population. Dipping chicken breast in lauricidin for 30 min caused a significant reduction of L. monocytogenes, S. enteritidis and E. coli O157:H7 population compared to 10 and 20 min dipping. Initial L. monocytogenes, S. enteritidis and E. coli O157:H7 counts on chicken breast treated with lactic acid decreased by 1.97, 1.71 and 2.59 log CFU/g, respectively. Lactic acid caused a higher reduction in initial S. enteritidis and E. coli O157:H7 counts compared to lauricidin.  相似文献

Minimally processed baby spinach contaminated with Escherichia coli O157:H7 has been associated with multiple outbreaks of foodborne illnesses recently. Chlorinated water is widely used to wash vegetables commercially, but this washing procedure has limited efficacy and can lead to the formation of carcinogenic substances. This study was conducted to determine the effects of organic acids and hydrogen peroxide alone and in binary combinations with or without mild heat (40 and 50 °C) on the inactivation of Escherichia coli O157:H7 on baby spinach. Baby spinach leaves were dip-inoculated with E. coli O157:H7 to a level of 6 log CFU/g and stored at 4 °C for 24 h before treatment. Individual washing solutions (1% and 2% lactic acid [LA], citric acid [CA], malic acid [MA], tartaric acid [TA], acetic acid [AA], hydrogen peroxide [H2O2] as well as binary combinations of LA, CA, MA and H2O2 at final concentrations of 1% were used to decontaminate spinach leaves at 22, 40 or 50 °C for 2–5 min to test their efficacy in reducing E. coli O157:H7. Chlorinated water (200 ppm free chlorine) decreased the population of E. coli O157:H7 on baby spinach by only 1.2–1.6 log CFU/g, which was not significantly different from DI water washing. Washing with 1% LA at 40 °C for 5 min was the most effective treatment achieving a 2.7 log reduction of E. coli O157:H7 which is significantly higher than chlorine washing. Washing with LA + CA or LA + HP at 40 °C for 5 min was equally effective against E. coli O157:H7, resulting in a 2.7 log reduction of E. coli O157:H7. The application of mild heat significantly enhanced the efficacy of washing solutions on the inactivation of E. coli O157:H7. There was, however, no significant difference between treatments at 40 °C for 5 min and 50 °C for 2 min. The results suggested that the use of organic acids in combination with mild heat can be a potential intervention to control E. coli O157:H7 on spinach.  相似文献

In this research, a total of 200 dairy (raw milk, cheese) samples obtained from Ankara, were examined for the presence of Yersinia spp., total coliform and Escherichia coli. As expected, raw milk 55% (55/100) were significantly contaminated with Yersinia spp., than cheese samples 14% (14/100). Y. enterocolitica was the most commonly isolated species, and was recovered from 47.3% in raw milk 35.7% in cheese samples. The other Yersinia spp. were identified as Y. frederiksenii (31.0%, 21.4%), Y. kristensenii (12.7%), Y. intermedia (7.2%, 7.1%) and atypical Yersinia spp. (1.8%, 35.7%) in raw milk and cheese samples, respectively. All the samples of cheese examined were negative for Y. kristensenii. All Y. enterocolitica strains tested gave negative results in the autoagglutination tests and crystal violet binding test.Furthermore total coliform bacteria and E. coli were investigated in these samples. According to the analysis results, most of the samples containing Yersinia spp.were found high number of viable count cell total coliform than E. coli. Total coliform bacteria and E. coli, were detected (3.0 × 104 cfu/ml and 1.5 × 104 cfu/ml) in four milk samples containing Y. enterocolitica while, they were detected (2.5 × 104 cfu/ml and 1.0 × 104 cfu/ml) in eight milk samples containing Y. enterocolitica. In the present study, total coliform bacteria and E. coli were detected >1.1 × 105 cfu/g in six cheese samples containing Y. enterocolitica and atypical Yersinia spp. The results indicate that Yersinia spp. is more likely to be isolated from foods with a high level of coliforms than from foods with low coliform counts.  相似文献

The study was conducted to assess the impact of the changes in the milk collection system in Trinidad (from twice daily collection to once, introduction of chilling facilities to the collection centres and transportation of milk to the processing plant in insulated truck instead of in metal churns at ambient temperature) on the microbial load and antimicrobial residue quality of the milk as well as the temperature and pH of milk, using standard methods. The presence of antimicrobial residues was detected using the Delvo test kit. Of a total of 266 milk samples from churns, the mean ± sd temperature and log10 ± sd TAPC per ml was 20.36 ± 7.91 °C and 6.3 ± 1.09 respectively. For 20 milk samples from the chillers, the mean temperature and log10 ± sd TAPC per ml was 15.10 ± 2.73 °C and 7.04 ± 0.33 respectively compared with corresponding values for 36 samples collected from the truck, 11.64 ± 4.22 °C and 7.11 ± 0.62 respectively (P < 0.05; X2). The mean TAPC, staphylococcal and E. coli counts per ml of milk from churns were significantly (P < 0.05; X2) higher for milk at low temperature (0–20 °C) compared with milk at high temperature (>30 °C). Eight (4.2%) of 192 milk samples tested contained antimicrobial residues. Of 168 S. aureus isolates tested, 24 (14.3%) were enterotoxigenic while 53 (45.3%) of 117 isolates tested exhibited resistance to various antimicrobial agents while of 386 isolates of E. coli tested, 3 (0.8%) were O157 strain and 129 (64.5%) of 200 isolates exhibited resistance to antimicrobial agents. It was concluded that despite the changes, the microbial load of milk was still high suggesting poor sanitary practices at the farm level. The detection of antimicrobial residues agents coupled with toxigenic S. aureus and E. coli isolates could pose health hazards to consumers.  相似文献

In search for alternative disinfection methods to prevent the development of bacterial resistances, the effectiveness of benzalkonium chloride (BAC), nisin and ozone against planktonic cells and biofilms of Staphylococcus aureus CECT 4459 was compared. Maximum effect of each compound was determined by comparing the times of exposure at which maximum mortality was reached. A dose–response study was carried out at such exposure times and LD90 values (doses causing 90% of mortality) were determined. BAC and ozone were more effective against biofilms than against planktonic cells, presumably due to a higher accessibility of biofilm cells. Concerning the control of biofilms, BAC was highly effective, but the application of 250 IU/ml of nisin or 1 μg/g of ozonized water allowed to reach 99% inactivation in only 2 min. It is concluded that nisin and ozone could be good alternative disinfectants to prevent the development of bacterial resistances, but factors such as applicability, price and risk to employers and food should also be taken into account.  相似文献

The sanitization potency of slightly acidic electrolyzed water (SAEW) on pure cultures of Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) was evaluated. The potency was compared with that of strong acidic electrolyzed water (StAEW) and sodium hypochlorite (NaOCl) solution. SAEW (ca. pH 5.8 and 21 mg/l available chlorine concentration; ACC) resulted into >5 log10CFU/ml reduction of E. coli and S. aureus after 90 s of exposure. The relative bacterial reduction potency at each exposure time was in the order StAEW > NaOCl > SAEW and increased with exposure time, with relative effect being 90 s > 60 s > 30 s. The results indicate that SAEW with low ACC and near neutral pH can potentially sanitize E. coli and S. aureus within a short period of exposure presenting a potential replacement to NaOCl solution commonly used in the food industry.  相似文献

The aim of this work was to determine the chemical composition and microbiological quality of Mediterranean horse mackerel (Trachurus mediterraneus) muscle during the year and the consequent advantages or disadvantages in processing the raw material. A total of 180 samples were examined (15 samples per month) during two years (November 2001–November 2003). Fish were caught in the coastal waters of Chalkidiki peninsula (Northern Greece). Proximate composition of the muscle during the year resulted in: water 76.8 ± 1.39%, proteins 20.3 ± 0.68%, fat 1.3 ± 1.08% and ash 1.5 ± 0.08%. In March, April and May an increase in fat content of the muscle (2.5%, 2.8% and 2.1%, respectively) and a decrease in water content of the muscle (below 75.9%) were observed. On the contrary, fat content showed a decrease in September and October (0.4% and 0.6%, respectively), while water content of the muscle increased during these months (78.2% and 77.6%, respectively). The protein content of the muscle remained at high values, while the ash content was almost constant during the year. The mean value of muscle pH was 6.38 ± 0.16; extreme values (<6 or >7) of muscle pH were not observed. Total Viable Bacteria, Shewanella putrefaciens and Pseudomonas spp. were used as microbial indices to evaluate the microbiological quality of the muscle. Mean counts of Total Viable Bacteria, S. putrefaciens and Pseudomonas spp. during the year were: 4.01 ± 0.38 log10 cfu/g, 2.37 ± 0.43 log10 cfu/g and 3.34 ± 0.27 log10 cfu/g, respectively. The variations of the values within seasons are minimal and significant homogeneity in the chemical composition and microbiological quality of Mediterranean horse mackerel muscle is observed. This is an important advantage in processing of the raw material.  相似文献

Minimal inhibitory (MIC) and minimal bactericidal (MBC) concentrations of malic acid against Listeria monocytogenes, Salmonella Enteritidis and Escherichia coli O157:H7 inoculated in apple, pear and melon juices stored at 5, 20 and 35 °C were evaluated. MICs and MBCs against L. monocytogenes, S. Enteritidis and E. coli O157:H7 were significantly affected by storage temperature, juice characteristics and type of microorganism. Malic acid was more effective at 35 and 20 °C than at 5 °C in all studied fruit juices. E. coli O157:H7 was more resistant to malic acid than S. Enteritidis and L. monocytogenes. Apple, pear and melon juices without malic acid were inhibitory to E. coli O157:H7, S. Enteritidis and L. monocytogenes at 5 °C, whereas, MBCs of 1.5% (v/v) of malic acid in apple and pear juices, and 2% (v/v) in melon juice at 5 °C were needed to reduce E. coli O157:H7, those concentrations being higher than those required to reduce S. Enteritidis and L. monocytogenes in those fruit juices. In addition, concentrations of 2%, 2.5% and 2.5% (v/v) of malic acid added to apple, pear and melon juices, respectively, were required to inactivate the three pathogens by more than 5 log cycles after 24 h of storage at 5 °C. Transmission electron microscopy showed that malic acid produced damage in the cell cytoplasm of pathogens without apparent changes in the cell membrane.  相似文献

The survey was conducted on 50 unripened Van otlu cheese samples obtained in Van and Hakkari markets at retail level to determine the microbial characteristics with special emphasis on Staphylococcus aureus, Escherichia coli, E. coli O157:H7 and Salmonella spp. The results revealed that S. aureus and E. coli were present in extremely high numbers, with a mean 6.10 and 3.68 log CFU/g, respectively. S. aureus was found in all samples ranging from 2.48 to 7.15 log CFU/g and was present in more than 5.0 × 105 CFU/g in 54% of the samples whereas E. coli was found in 62% of the samples. None of the samples contained E. coli O157:H7; but 3 of the 50 samples had Salmonella spp. The results indicate that Van otlu cheese presents a potential hazard for public health; and the necessary precaution will have to be taken to improve the sanitary practices and cheese manufacturing technique.  相似文献

The effectiveness in the assurance of fresh vegetable microbiological quality of wash solutions containing 200 ppm free chlorine, 0.1 mg/ml benzalkonium chloride, 0.2% and 1% lactic acid was assessed on Escherichia coli O157:H7 and Yersinia enterocolitica contaminated lettuce and tomatoes. Y. enterocolitica reduction on tomatoes (5.08, 4.77 and 4.21 log after 0.2% lactic acid, 200 ppm chlorine and 0.1 mg/ml benzalkonium chloride treatments, respectively) were significantly higher than those for Y. enterocolitica on lettuce and E. coli O157:H7 on both vegetables. Antimicrobial treatment effects on bacterial counts and product quality after subsequent 7 day storage (4 °C and 22 °C) were determined. No pathogens were found in natural microflora of fresh vegetables.  相似文献

This study evaluated the effects of ozonated water (1, 3, and 5 ppm) alone with different exposure times (0.5, 1, 3, or 5 min), and combinations of 3 ppm ozone with 1% organic acids (acetic, citric, or lactic acids) during 5-min exposure for control of Escherichia coli O157:H7 and Listeria monocytogenes on enoki mushroom and to observe the regrowth of these pathogenic bacteria on treated enoki mushroom during storage for 10 days at 15 °C. Results showed that ozone treatment gave less than 1.0- and 0.5-log count reductions on E. coli O157:H7 and L. monocytogenes, respectively. Efficacy was improved with combined 3 ppm ozone and 1% citric acid treatment, resulting in 2.26- and 1.32-log count reductions, respectively. During storage at 15 °C (10 days) after combined treatment and packaging, populations of E. coli O157:H7 and L. monocytogenes increased to more than 8.0 log cfu/g, indicating that the combined treatment did not have a residual antimicrobial effect during storage. Although the storage study did not show control of these pathogens, the combined ozone–organic acid treatment was more effective than individual treatments in reducing initial population levels of these pathogens on enoki mushroom.  相似文献

Thirty-four tuna candy products sold in retail markets and supermarkets in Taiwan were purchased and tested to determine the biogenic amine, histamine-forming bacteria, and adulteration of bonito meat. The levels of pH value, water activity (Aw), water content, total volatile basic nitrogen (TVBN), aerobic plate count (APC) and total coliform (TC) in all samples ranged from 5.3 to 6.1, 0.47 to 0.65, 7.37% to 17.32%, 12.1 to 54.6 mg/100 g, <1 to 6.2 log CFU/g and <3 to 1700 MPN/g, respectively. None of these sample contained Escherichia coli. The average content of various biogenic amines in all tested samples was less than 1.0 mg/100 g. Four histamine-producing bacterial strains isolated from tested samples produced 1.02–1.74 ppm of histamine in trypticase soy broth supplemented with 1.0% l-histidine (TSBH) after incubation at 35 °C for 24 h. Assay of multiplex polymerase chain reaction (PCR) revealed that adulteration rate of bonito meat was 26.5% (9/34) in tuna candy samples. Tuna species in tuna candy samples was identified as Thunnus albacares for 29 samples (85.3%), Thunnus alalunga for four samples (11.8%) and Thunnus obesus for one sample (2.9%) by polymerase chain reaction–restriction fragment length polymorphism (PCR–RFLP).  相似文献

In this work, formations of water-in-diesel oil nano-emulsions using water/mixed nonionic surfactant/diesel oil system have been studied. The high energy emulsification method was used to form three emulsions using different water contents: 5%, 10% and 14% (v/v) namely; E1, E2 and E3, respectively. These nano-emulsions were stabilized with emulsifiers having different Hydrophilic–Lipophilic Balance (HLB) namely; span 80 (HLB = 4.3), emarol 85 (HLB = 11) and their mixture (SE) with HLB = 10. The effect of water on the droplet size formation has been investigated. The interfacial tension and thermodynamic properties of the individual and emulsifiers blends have been studied. The interfacial tension (γ) measurements at 30 °C were used to determine the critical micelle concentration (CMC) and surface active properties of these emulsifiers. The water droplet sizes were measured by dynamic light scattering (DLS). From the obtained data, it was found that, mean sizes between 19.3 and 39 nm were obtained depending on the water content and concentration of blend emulsifiers (SE). Also, the results show that, the interfacial tension (γ) gives minimum value (10.85 mN/m) for SE comparing with individual emulsifier (17.13 and 12.77 mN/m) for span 80 and emarol 85, respectively. The visual inspection by TEM showed that the obtained results support the data obtained by dynamic light scattering.  相似文献

Despite an increase in the number of food handlers receiving food hygiene training, a high number of food poisoning outbreaks still occur as a result of improper food handling practices in the retail industry. In this study, samples were collected from the hands and aprons of food handlers in the delicatessen sections of a prominent South African retail group and analysed for the presence of total viable counts (TVC), total coliforms, Escherichia coli, members of the family Enterobacteriaceae and Staphylococcus aureus in order to assess the levels of contamination and to establish possible relationships. Noteworthy TVC were present on 98% of hands and 84% of aprons sampled and conformed to the national standard of 1 × 102 cfu cm−2 without exception. Coliforms were present on 40% of food handler’s hands and on 26% of aprons and when compared to the literature which suggests a target value of <2.5 cfu cm−2, 32% of food handlers exceeded the target with regard to hands and 8% with regard to aprons. E. coli was found to only exceed the limit in the case of one food handler. Enterobacteriaceae were present on the hands of food handlers (44%) and on aprons (16%), ranging between 5 and 1.8 × 101 cfu cm−2 on hands and between 5 and 2.9 × 101 cfu cm−2 on aprons. S. aureus counts were present on 88% of hands and 48% of aprons and ranged between negligible and 9.8 × 101 cfu cm−2 for hands and up to 6.2 × 101 cfu cm−2 for aprons. No significant statistical correlation occurred between the organisms on hands and aprons, indicating that the latter were not likely to be cross-contaminated by hands.  相似文献
