The anti-inflammatory effects of the crude extract (CE) of Lotus corniculatus var. São Gabriel and its derived hexane (HEX), ethyl acetate (AcOEt), n-butanol (BuOH) and aqueous (Aq) fractions and isolated compounds kaempferitrin, oleanolic acid and β-sitosterol, in a mouse model of pleurisy induced by carrageenan were investigated. Swiss mice were used in the in vivo experiments. The crude extract of L. corniculatus and its derived fractions, and also its isolated compounds, inhibited leukocytes, exudation, and myeloperoxidase (MPO) and adenosine-deaminase (ADA) activities, as well as nitrite/nitrate concentration and interleukin-1 beta (IL-1β) level (< 0.05). L. corniculatus showed important anti-inflammatory activity by inhibition not only of leukocytes and/or exudation, but also of pro-inflammatory enzymes and mediators such as MPO, ADA, and IL-1β. The constituent’s kaempferitrin, oleanolic acid and β-sitosterol may well account for it.  相似文献

Nitrosamines are well-known carcinogenic and toxic compounds for humans and animals. The aim of this study was to investigate the possible formation of nitrosamine compounds from aqueous extracts of five different types of Catha edulis leaves using nitrite as a nitrosation agent, either in aqueous solution or under simulated normal fasting stomach conditions (at 37 °C and pH 2) for 1 h. Nitrosoephedrine was used as a reference compound in this study. Nitrosation of aqueous extracts of the different types of Catha edulis leaves with constant concentration of nitrite (14.4 mM) in aqueous solution showed total apparent nitrosamine compounds to be in the range 94–319 mg/100 g DM (dry matter) of CE (Catha edulis) leaves. In contrast, nitrosamines formed in simulated gastric fluid were much lower, in the range 23–79 mg/100 g DM of CE leaves. Based on the moderate formation of nitrosamines with the Sabri Catha edulis leaves, the latter was selected for investigating the effect of varying levels of nitrite in aqueous solution and simulated gastric juice. The nitrosation of aqueous extracts of Sabri Catha edulis with different levels of nitrite in aqueous solution yielded a dose-dependent amount of total apparent nitrosamine compounds, these being undetected at ?0.5 mM sodium nitrite in aqueous solution and ?1 mM in simulated gastric juice. This raises the question of whether the observed high incidence of esophageal and forestomach carcinomas in Yemen could be attributed to the formation of nitrosamines in vivo from the secondary amines present in Catha edulis leaves.  相似文献

The aerial parts of Salvia halophila and Salvia virgata were subjected to Soxhlet extraction with different solvents such as n-hexane, ethyl acetate, methanol, and aqueous methanol (50%). Plants were also extracted with water under reflux. The effects of the extracts were studied in p-benzoquinone-induced abdominal constriction test for the assessment of antinociceptive activity and carrageenan-induced hind paw edema and 12-O-tetradecanoyl-13-acetate (TPA)-induced ear edema models in mice for the anti-inflammatory activity. The extracts were analysed using a HPLC–PDA method. Results showed that methanol extract of S. virgata significantly inhibited carrageenan-induced paw edema and p-benzoquinone-induced abdominal constriction at 100 mg/kg dose, while it showed no effect in the TPA-induced ear edema. On the other hand, the other extracts did not show any inhibitory antinociceptive and anti-inflammatory activities in these in vivo models. Rosmarinic acid was found as main constituent in the extracts, while caffeic acid and luteolin derivatives were also detected.  相似文献

Both total and dialyzable Mn levels were determined in 108 duplicate meals during 36 consecutive days. Both mineral fractions were measured by a graphite furnace atomic absorption spectrometry (GFAAS) method previously optimized. A total mean Mn fraction of 1.03 ± 0.49 mg was found in the meals. The Mn supplied by the meals is directly and significantly (p < 0.001) correlated with macronutrient content (carbohydrates, fibre and protein). The mean Mn fraction dialyzed through the dialysis membrane was 0.23 ± 0.17 mg (22.0 ± 8.93% as bioaccessible fraction). The total and dialyzable Mn fractions found for breakfasts were significantly lower (p < 0.001). Nevertheless, the Mn bioavailabilities expressed as the percentage of dialyzable element, were not significantly different among the three primary meals (breakfast, lunch and dinner). A significant correlation between the total and the dialyzable fraction of Mn in meals was found (p < 0.001, r = 0.78, r2 = 0.61). The dialyzed element fractions present in meals were significantly correlated mainly with carbohydrates, protein and several amino acid levels (p < 0.01). Foods with higher carbohydrate and therefore energy contents, e.g. cereals, legumes, vegetables and fruits, would be primary sources of bioaccessible Mn in the diet. The bioaccessibility of Mn was only significant influenced by energy, carbohydrates and Se levels present in meals. The mean Mn daily dietary intake (DDI) was 3.05 ± 0.61 mg day−1.  相似文献

Lipid oxidation of cultured blue mussels (Mytilus edulis) during mechanical handling and storage on ice was investigated. Furthermore, the exposure of blue mussels to ascorbic acid (Asc) as an antioxidant and its effects on lipid oxidation of sample was monitored. Thiobarbituric acid reactive substances (TBARS) of stored mussels were significantly (p < 0.05) higher than those of the fresh mussels. Mechanical handling of mussels, which includes washing, sorting and packaging, for up to 1 h did not affect their oxidative status significantly (p < 0.05). Furthermore, exposing live mussels to specific concentrations of Asc retarded lipid oxidation significantly (p < 0.05) during storage on ice for only 5 days, after which the Asc became a pro-oxidant.  相似文献

The selenium contents of 66 mushroom samples, wild growing or commercially available in Portugal, fresh, canned or dried, were determined. The samples consisted of 18 mushroom species, mainly selected in accordance with their availability and consumer preference. The selenium contents varied considerably between different mushroom species. The highest selenium contents were found in the wild species Boletus aestivalis (48.5 mg/kg dry weight, DW), Boletus pinophilus (19.9 mg/kg DW), Boletus edulis (14.9 mg/kg DW), Boletus aereus (12.3 mg/kg DW), Boletus fragans and Boletus spretus (∼2 mg/kg DW). Other mushrooms having considerable selenium contents included Marasmius oreades (1.5 mg/kg DW), Agaricus bisporus “Portobello” (1.25 mg/kg DW), A. bisporus (1.0 mg/kg DW) and Russula cyanoxantha (0.72 mg/kg DW). The selenium contents of these mushrooms are sufficient to provide nutritionally significant amounts in relation to the total daily intake of selenium. Other edible mushrooms, such as Lentinula edodes, Pleurotus ostreatus, Cantharellus cibarius,Craterellus cornucopioides and Lepista nuda, contained only small amounts of selenium. The importance of these mushrooms as a source of selenium is therefore marginal.  相似文献

The aim of this study was to investigate the anti-inflammatory effects of the crude extract (CE) of Rosmarinus officinalis L. its derived fractions: hexane (HEX), ethyl acetate (AcOEt), and ethanolic (ET), and isolated compounds: carnosol, betulinic acid and ursolic acid, in the mouse pleurisy model induced by carrageenan. Swiss mice were used in the in vivo experiments. The CE and its derived fractions and isolated compounds inhibited leukocytes, exudation, interleukin-1 beta (IL-1β) and tumour necrosis factor-alpha (TNF-α) levels, myeloperoxidase activity (MPO), and nitrite/nitrate production (NOx) (p < 0.05). R. officinalis L. showed an important anti-inflammatory activity by inhibition not only of leukocytes and exudation, but also of a pro-inflammatory enzyme and mediators (MPO, NOx, IL-1β, and TNF-α). The present study showed that carnosol, betulinic acid and ursolic acid compounds could be responsible for this anti-inflammatory effect.  相似文献

This study investigated the optimum polyphenol extraction conditions for two Nigerian green leafy vegetables, Vernonia amygdalina (VA) and Gongronema latifolium (GL) that are widely used for food and medicinal purposes. Seven different solvents were used to extract the total phenolic contents of the leaves; 100% acetone, 80% acetone, 50% acetone, 100% ethanol, 70% ethanol, 100% methanol and 70% methanol and their total phenolic contents were determined. The results showed that the 80% acetone was significantly (p < 0.05) the most effective for polyphenol extraction from the leaves and generally the acetone-containing solvents were more effective than alcohol-containing solvents. The 80% acetone extract was fractionated on a silicic acid-packed column to give two main fractions: acetone eluted chlorophyll-enriched (flow-through) and ethanol eluted chlorophyll-depleted (column-bound). The ethanol fractions of VA and GL were significantly better (p < 0.05) DPPH radical scavengers than the acetone fractions. In contrast, the acetone fraction was a significantly better (p < 0.05) scavenger of hydroxyl radicals when compared to the ethanol fraction. Both acetone and ethanol fractions significantly (p < 0.05) and dose-dependently inhibited linoleic acid oxidation, though duration of inhibition was less at lower concentrations (10 and 25 ??g/ml) than at the highest concentration (50 ??g/ml).  相似文献

Chilean jack mackerel (Trachurus murphyi) is an underutilised medium-fat fish species, captured in large volumes but commercialised in the fish meal and surimi industries. This study provides a first approach including a novel technology for the commercialisation of this fish species as a chilled product for human consumption. Ice prepared from aqueous extracts of oregano (Origanum vulgare) and rosemary (Rosmarinus officinalis) leaves was applied as chilling system and compared to traditional ice. Chemical changes related to quality criteria were analysed throughout a 23-day chilling period. A marked antioxidant effect (p < 0.05) could be detected with fish kept under both plant-extract icing systems, according to peroxide and thiobarbituric acid reactive substance formation; meanwhile, the employment of such icing systems led to lower (p < 0.05) scores for pH value and total volatile amine formation. Additionally, the plant extract presence in the chilling medium provided a lower (p < 0.05) lipid hydrolysis (free fatty acid formation) development. According to the preservative effect observed for both plant extract systems, further research is envisaged concerning their optimal employment, this including the qualitative and quantitative analysis of active molecules present in the extracts.  相似文献

In vitro antioxidant activities of three selected Indian brown seaweeds – viz., Sargassum marginatum, Padina tetrastomatica and Turbinaria conoides – were investigated. Total phenolic content and reducing power of crude methanolic extract were also investigated. The activity of total methanolic extract and five different fractions (viz., petroleum ether (PE), ethyl acetate (EA), dichloromethane (DCM), butanol (BuOH) and aqueous) were studied using total antioxidant activity, DPPH radical scavenging and deoxyribose assays. EA fraction of S. marginatum exhibited higher total antioxidant activity of 39.62 mg ascorbic acid equivalent/g extract (or 0.31 mg ascorbic acid equivalent/g seaweed on dry weight basis) among the all the fractions. Among the fractions obtained from different seaweeds, EA fraction of S. marginatum showed higher DPPH scavenging activity of 23.16%; while PE fraction of T. conoides exhibited lower deoxyribose activity of 47.81%. Higher phenolic content (49.16 mg gallic acid equivalent/g extract or 0.86 mg GAE/g of seaweed on dry weight basis) was noticed in aqueous fraction of T. conoides. Reducing power of crude methanolic extract increased with increasing concentration. Reducing power of T. conoides and P. tetrastomatica were higher compared to standard antioxidant (α-tocopherol). Among the seaweeds, total methanolic extract of T. conoides had significantly higher phenol content (P < 0.05) compared to the other two species. In vitro antioxidant activity of methanolic extracts from all the three seaweeds showed an increase with increasing concentration indicating the dose dependency of these properties.  相似文献

We investigated the effects of Monascus-fermented mixed grain extracts (MFGEs) enriched with bioactive mevinolins (natural statins) and coenzyme Qs (CoQ9 + CoQ10) on the blood lipids, antioxidant status, and tissue levels of CoQs and α-tocopherol (α-Toc) in hyperlipidemic rats. The oral administration of MFGEs (300 mg/kg body weight per day) for 8 weeks resulted in a significant decrease in blood levels of total cholesterol (TC), triglycerides (TG), low-density lipoprotein cholesterol (LDL-C), and LDL-C/high-density lipoprotein cholesterol (HDL-C) ratio compared to the control and lovastatin supplement group of a dosage of 20 mg/kg per day (p < 0.05). Furthermore, a significant increase in the ratios of α-Toc/LDL-C and CoQs/LDL-C in plasma and tissues and improvement in plasma antioxidant status as measured by TBARS and TRAP were observed in hypercholesterolemic rats (p < 0.05). Regarding the effects of MFGEs on antioxidant levels of plasma and tissues, there were significant increases in the levels of α-Toc (p < 0.05) and CoQs (p < 0.01) after the 8-week MFGEs treatment. These data indicate that MFGEs supplementation not only decreases blood lipids and lipid peroxidation but also increases levels of antioxidants such as α-Toc and CoQs and may improve plasma antioxidant status as well as a hypolipidemic effect.  相似文献

Ether, methanolic and aqueous extracts of lyophilized rambutan (Nephelium lappaceum L.) peels and seeds were evaluated for phenolic contents, antioxidant and antibacterial activities. High amounts of phenolic compounds were found in the peel extracts and the highest content was in the methanolic fraction (542.2 mg/g dry extract). Several potential antioxidant activities, including reducing power, β-carotene bleaching, linoleic peroxidation and free radical scavenging activity, were evaluated. The peel extracts exhibited higher antioxidant activity than the seed extracts in all methods determined (P < 0.05). The methanolic fraction was found to be the most active antioxidant as shown by their 50% DPPH inhibition concentration, 4.94 μg/mL. The results indicated this fraction exhibited greater DPPH radical scavenging activity than BHT and ascorbic acid (0.32 g dry extract/g BHT or ascorbic acid). Antibacterial activity against eight bacterial strains was assessed by disc diffusion and broth macrodilution methods. All peel extracts exhibited antibacterial activity against five pathogenic bacteria. The most sensitive strain, Staphylococcus epidermidis, was inhibited by the methanolic extract (MIC 2.0 mg/mL).  相似文献

The hypocholesterolemic and hypotriglyceridemic activities of the aqueous and organic extracts of Ocimum basilicum were studied using Triton WR-1339-induced hyperlipemic mice as an experimental model. Hyperlipidemia was developed by intraperitoneal injection of Triton (200 mg/kg body weight “BW”). The animals were divided into eight groups of eight mice each: normolipidemic control group (NCG), hyperlipidemic control group (HCG), hyperlipidemic plus DMSO control (HDCG), crude aqueous basil extract-treated group (CETG), dichloromethane extract-treated group (DETG), ethyl acetate extract-treated group (EETG), methanol extract-treated group (METG), and aqueous fraction-treated group (AFTG). After 7 h and 24 h of treatment, the intragastric administration of all extracts caused a significant decrease of plasma total cholesterol. Triglyceride levels were also significantly lowered but not in DETG. Similar results were observed for LDL-cholesterol concentrations. Although no significant change of HDL-cholesterol was noticed after 7 h of treatments, a significant increase of this cholesterol fraction was observed in EETG and AFTG after 24 h. Furthermore, crude aqueous basil extract and all polar solvent (methanol, ethyl acetate, water)-soluble fractions showed a significant ameliorative action on elevated atherogenic index (AI) and LDL/HDL-C ratios, while these atherogenic markers were not statistically suppressed by the dichloromethane-soluble extract. This finding indicates that O. basilicum may contain polar products able to lower plasma lipid concentrations and might be beneficial in treatment of hyperlipidemia and atherosclerosis.  相似文献

The effects of taurine (Tau) in regulation of lipid metabolism and decreasing inflammation in chronic alcohol-fed rats was investigated. Rats were randomly divided into three groups: (1) isocaloric solution; (2) 3 g alcohol/kg BW/day; (3) 3 g alcohol/kg BW/day + 1 g Tau/kg BW/day for 6 weeks. Liver size and serum/liver lipids of alcohol-fed rats were decreased (p < 0.05) by Tau supplementation, but daily fecal lipid/bile acid outputs were increased (p < 0.05). Regarding de novo lipogenesis, Tau downregulated (p < 0.05) fatty-acid biosynthesis and upregulated (p < 0.05) cholesterol metabolism (CYP7A1) and energy expenditure (PPAR-α). Serum AST and ALT, and hepatic TNF-α levels and MMP-9 activity of alcohol-fed rats were decreased (p < 0.05) by Tau supplementation which may be related to the maintenance of higher (p < 0.05) antioxidant levels (lower thiobarbituric-acid-reactive-substances values and higher trolox equivalent antioxidant capacity) in serum and livers. Our study indicates that Tau downregulates lipogenesis, oxidative stress, and inflammation in chronic alcohol-fed rats.  相似文献

Carotenoids are effective antioxidants, which can quench singlet oxygen, suppress lipid peroxidation, and prevent oxidative damage. Both Pseudorabies virus (PRV) and human Herpes simplex virus (HSV) are DNA viruses, and their pathogenesis and immunobiology are similar. However, PRV does not infect humans. Therefore, PRV was used to infect murine macrophages (RAW264.7 cells), to mimic HSV-induced inflammation. Meanwhile, the influence of β-carotene on PRV-induced inflammation was also investigated. Results indicated that β-carotene inhibited (p < 0.05) NO, IL-1β, IL-6, and MCP-1 production in PRV-infected RAW264.7 cells. β-Carotene also suppressed (p < 0.05) NF-κB (p50 and p65), phosphorylation of extracellular-signal-related kinase (ERK), p38, and c-Jun N-terminal kinase (JNK) expression. It could be concluded that the anti-inflammatory effect of β-carotene is mainly through a suppression of cytokine expression in PRV-induced inflammation, which results from NF-κB inactivation. β-Carotene can be considered a potential anti-inflammatory agent for DNA-virus infection.  相似文献

Three species of dried mushrooms are commercially available in Taiwan, namely Agaricus blazei (Brazilian mushroom), Agrocybe cylindracea (black popular mushroom) and Boletus edulis (king bolete), and their non-volatile taste components were studied. All mushrooms were high in contents of carbohydrate, crude fiber and protein but low in contents of crude ash and fat. Arabitol, myo-inositol, mannitol and trehalose were detected in these three mushrooms, whereas glucose was not found in B. edulis. Contents of total soluble sugars and polyols ranged from 150.33 to 225.08 mg/g. Total free amino acid contents were low in these three mushrooms and ranged from 8.97 to 14.91 mg/g. The contents of MSG-like components ranged from 1.24 to 4.40 mg/g were in the descending order of the A. blazei, A. cylindracea and B. edulis. Total 5′-nucleotides contents of A. blazei and A. cylindracea were higher than that of B. edulis whereas flavor 5′-nucleotides content of A. blazei was higher than those of A. cylindracea and B. edulis. Equivalent umami concentrations values in three mushrooms ranged from 10.46 to 135.90 g per 100 g. Overall, these three mushrooms possessed highly umami taste.  相似文献

The effect of oil-based marinades containing grape seed extract (Vitis vinifera L.; 0.2, 0.4, 0.6 and 0.8 g/100 g) formulated in a water/oil emulsion or rosemary extract (Rosmarinus officinalis; 0.12, 0.2, 0.6, 1.0 and 1.5 g/100 g) in oil on the formation of heterocyclic amines (HAs) in fried beef patties was examined. After application of marinades and frying, four HAs MeIQx (2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline), PhIP (2-amino-1-methyl-6-phenylimidazo[4,5b]pyridine), Norharman, and Harman were found at low levels in all fried patties, MeIQx (0.3–1.0 ng/g), and PhIP (0.02–0.3 ng/g). The content of MeIQx and PhIP were significantly reduced by approx. 57% and 90% (p < 0.05), respectively, after use of marinades containing the highest extract concentration. The antioxidant capacity of grape seed was about two-times greater than that of rosemary extract. A correlation between inhibition of HAs and Trolox-equivalents (MeIQx, R2 = 0.85, p < 0.001; PhIP, R2 = 0.83, p < 0.001) was found. Sensory tests showed a high acceptance of flavour and colour for controls and samples.  相似文献

Antioxidant potential of four methanol extracts from three selected plant species, namely Salvia nubicola (Lamiaceae), Acer oblongifolium (Aceraceae) and Hedera nepalensis (Araliaceae) was measured using assays in aqueous and lipid systems. Antioxidant activities were investigated in aqueous systems by using DPPH radical-scavenging assay, ABTS radical-scavenging assay and DNA protection assay, while antioxidant activity in a lipid system was determined by using the thiobarbituric acid-reactive substances (TBARS) assay. Additionally, the Folin-Ciocalteu method was used to measure total phenolic content. Methanol extracts of leaves and flowers of S. nubicola showed the highest Trolox equivalent (TE) values in the case of the DPPH assay, 2484 ± 4.9 mmol TE/g extract, as well as total phenolic content, 139 ± 0.2 mg gallic acid equivalents/g extract. Three fractions (A-C) of the methanol extract of S. nubicola leaves and flowers were produced by semi-preparative HPLC. Fraction B was found to be the most active in the DPPH radical-scavenging assay and had the highest total phenol content. HPLC-DAD and LC-MS revealed rosmarinic acid in S. nubicola extracts and chlorogenic acid and rutin in H. nepalensis extracts as the main phenolic antioxidants.  相似文献

In this study the protective effects of cold buffer extract of Funalia trogii ATCC 200800 (FtE) and vitamin E (VitE) on oxidative stress induced with deltamethrin using oral administration in rats were investigated. Deltamethrin treatment caused an increase in liver enzyme activities of aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP) (p < 0.05); however, it caused a decrease in activities of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and glutathione reductase (GRd) when compared to control group (p < 0.05). Activities of AST, ALT, ALP enzymes and level of thiobarbituric acid reactive substances (TBARS) decreased significantly after VitE administration (p < 0.05). Both enzyme activities and TBARS levels were found similar in VitE and FtE treated rats shortly after pesticide administration (p < 0.05). In conclusion, it appears that FtE prepared in cold buffer has capability to prevent the liver damage like VitE against the toxic effect of deltamethrin.  相似文献
