Organelles, i.e., internal subcompartments of cells, are fundamental to spatially separate cellular processes, while controlled intercompartment communication is essential for signal transduction. Furthermore, dynamic remodeling of the cytoskeleton provides the mechanical basis for cell shape transformations and mobility. In a quest to develop cell‐like smart synthetic materials, exhibiting functional flexibility, a self‐assembled vesicular multicompartment system, comprised of a polymeric membrane (giant unilamellar vesicle, GUV) enveloping polymeric artificial organelles (vesicles, nanoparticles), is herein presented. Such multicompartment assemblies respond to an external stimulus that is transduced through a precise sequence. Stimuli‐triggered communication between two types of internal artificial organelles induces and localizes an enzymatic reaction and allows ion‐channel mediated release from storage vacuoles. Moreover, cytoskeleton formation in the GUVs' lumen can be triggered by addition of ionophores and ions. An additional level of control is achieved by signal‐triggered ionophore translocation from organelles to the outer membrane, triggering cytoskeleton formation. This system is further used to study the diffusion of various cytoskeletal drugs across the synthetic outer membrane, demonstrating potential applicability, e.g., anticancer drug screening. Such multicompartment assemblies represent a robust system harboring many different functionalities and are a considerable leap in the application of cell logics to reactive and smart synthetic materials.  相似文献

Layer‐by‐Layer (LbL) assembly is a simple and highly versatile method to modify surfaces and fabricate robust and highly‐ordered nanostructured coatings over almost any type of substrate. Such versatility enables the incorporation of a plethora of building blocks, including materials exhibiting switchable properties, in a single device through a multitude of complementary intermolecular interactions. Switchable materials may undergo reversible physicochemical changes in response to a variety of external triggers. Although most of the works in the literature have been focusing on stimuli‐responsive materials that are sensitive to common triggers such as pH, ionic strength, or temperature, much less has been discussed on LbL systems which are sensitive to non‐invasive and easily controlled light stimulus, despite its unique potential. This review provides a deep overview of the recent progresses achieved in the design and fabrication of light‐responsive LbL polymeric multilayer systems, their potential future challenges and opportunities, and possible applications. Many examples are given on light‐responsive polymeric multilayer assemblies built from metal nanoparticles, functional dyes, and metal oxides. Such stimuli‐responsive functional materials, and combinations among them, may lead to novel and highly promising nanostructured smart functional systems well‐suited for a wide range of research fields, including biomedicine and biotechnology.  相似文献

Gene therapy has great potential to bring tremendous improvement to cancer therapy. Recently, photochemical internalization (PCI) has provided the opportunity to overcome endo‐lysosomal sequestration, which is one of the main bottlenecks in both gene and chemotherapeutic delivery. Despite PCI having shown great potential in gene delivery systems, it still remains difficult to perform due to the photo‐oxidation of exogenous cargo genes by reactive oxygen species (ROS) generated from activated photosensitizers (PSs). In this paper, a new type of a stable light‐triggered gene delivery system is demonstrated based on endo‐lysosomal pH‐responsive polymeric PSs, which serve as shielding material for the polymer/gene complex. By taking advantage of the endo‐lysosomal pH‐sensitive de‐shielding ability of the pH‐responsive shielding material incorporated in the ternary gene complexes (pH‐TCs), a more significant photo‐triggered gene expression effect is achieved without damage to the gene from ROS. In contrast, pH‐insensitive material‐shielded nanocarriers cause photo‐oxidation of the payload and do not generate a notable transfection efficacy. Importantly, with the benefit of our newly developed gene delivery system, the deep penetration issue can be resolved. Finally, the light‐triggered gene delivery system using pH‐TCs is applied to deliver the therapeutic p53 gene in melanoma K‐1735 bearing mice, showing excellent therapeutic potential for cancer.  相似文献

The fabrication of multilayer assemblies from polymeric compounds is an important tool for precise control of film architecture on the nanoscale. In this report, a general, novel approach for the preparation of well‐defined polymeric multilayers is described. To achieve this, sulfonyl‐azide group containing polymers are first generated and deposited as thin films onto solid (organic) substrates. Upon thermal activation, the system crosslinks and binds to the substrate via C–H bond insertion. Through step‐and‐repeat procedures, multilayer assemblies are then generated where all the individual layers are linked to each other. As the assembly process does not require any specific molecular interactions, the described process represents a general strategy to generate tailor‐made multilayer surface coatings with wide range of film thickness and composition.  相似文献

4‐oxo‐4‐(pyren‐4‐ylmethoxy) butanoic acid is used as a photolabile protecting group to show the optical gating of nanofluidic devices based on synthetic ion channels. The inner surface of the channels is decorated with monolayers of photolabile hydrophobic molecules that can be removed by irradiation, which leads to the generation of hydrophilic groups. This process can be exploited in the UV‐light‐triggered permselective transport of ionic species in aqueous solution through the channels. The optical gating of a single conical nanochannel and multichannel polymeric membranes is characterised experimentally and theoretically by means of current–voltage and selective permeation measurements, respectively. It is anticipated that the integration of nanostructures into multifunctional devices is feasible and can readily find applications in light‐induced controlled release, sensing, and information processing.  相似文献

Inspired by the asymmetric structure and responsive ion transport in biological ion channels, organic/inorganic hybrid artificial nanochannels exhibiting pH‐modulated ion rectification and light‐regulated ion flux have been constructed by introducing conductive polymer into porous nanochannels. The hybrid nanochannels are achieved by partially modifying alumina (Al2O3) nanopore arrays with polypyrrole (PPy) layer using electrochemical polymerization, which results in an asymmetric component distribution. The protonation and deprotonation of Al2O3 and PPy upon pH variation break the surface charge continuity, which contributes to the pH‐tunable ion rectification. The ionic current rectification ratio is affected substantially by the pH value of electrolyte and the pore size of nanochannels. Furthermore, the holes (positive charges) in PPy layer induced by the cooperative effect of light and protons are used to regulate the ionic flux through the nanochannels, which results in a light‐responsive ion current. The magnitude of responsive ionic current could be amplified by optimizing this cooperation. This new type of stimuli‐responsive PPy/Al2O3 hybrid nanochannels features advantages of unique optical and electric properties from conducting PPy and high mechanical performance from porous Al2O3 membrane, which provide a platform for creating smart nanochannels system.  相似文献

A dual‐functional nanofluidic device is demonstrated that integrates the ionic gate and the ionic rectifier within one solid‐state nanopore. The functionalities are realized by fabricating temperature‐ and pH‐responsive poly(N‐isopropyl acrylamide‐co‐acrylic acid) brushes onto the wall of a cone‐shaped nanopore. At ca. 25 °C, the nanopore works on a low ion conducting state. When the temperature is raised to ca. 40 °C, the nanopore switches to a high ion conducting state. The closing/opening of the nanopore results from the temperature‐triggered conformational transition of the attached copolymer brushes. Independently, in neutral and basic solutions, the conical nanopore rectifies the ionic current. While in acid solutions, no ion rectifying properties can be found. The charge properties of the copolymer brushes, combined with the asymmetrical pore geometry, render the nanopore a pH‐tunable ionic rectifier. The chemical modification strategy could be applied to incorporate other stimuli‐responsive materials for designing smart multi‐functional nanofluidic systems resembling the “live” creatures in nature.  相似文献

The acquisition of multidrug resistance (MDR) is a major hurdle for the successful chemotherapy of tumors. Herein, a novel hybrid micelle with pH and near‐infrared (NIR) light dual‐responsive property is reported for reversing doxorubicin (DOX) resistance in breast cancer. The hybrid micelles are designed to integrate the pH‐ and NIR light‐responsive property of an amphiphilic diblock polymer and the high DOX loading capacity of a polymeric prodrug into one single nanocomposite. At physiological condition (i.e., pH 7.4), the micelles form compact nanostructure with particle size around 30 nm to facilitate blood circulation and passive tumor targeting. Meanwhile, the micelles are quickly dissociated in weakly acidic environment (i.e., pH ≤ 6.2) to release DOX prodrug. When exposed to NIR laser irradiation, the hybrid micelles can trigger notable tumor penetration and cytosol release of DOX payload by inducing tunable hyperthermia effect. In combination with localized NIR laser irradiation, the hybrid micelles significantly inhibit the growth of DOX‐resistant MCF‐7/ADR breast cancer in an orthotopic tumor bearing mouse model. Taken together, this pH and NIR light‐responsive micelles with hyperthermia‐triggered tumor penetration and cytoplasm drug release can be an effective nanoplatform to combat cancer MDR.  相似文献

Visually readable codes play a crucial role in anticounterfeiting measures. However, current coding approaches do not enable time‐dependent codes to be visually read, adjusted, and differentiated in bright and dark fields. Here, using a combined strategy of piezoelectric lattice selection, oxygen vacancy engineering, and activator doping, a lanthanide ion‐doped titanate is developed that integrates mechano‐, thermo‐, and photo‐responsive color change (>18 h for bright field), persistent luminescence (>6 h for dark field), and stimulus‐triggered multimodal luminescence. The feasibility of optical encoding, visual displaying, and stimulus‐responsive encrypting of time‐dependent, dual‐field information by using the developed material is demonstrated. In particular, the differentiated display of dual‐field modes is achieved by combining mechanostimulated abolition of only the persistent luminescence and thermo‐ and photostimulated reversal of both the color change and persistent luminescence. The results provide new insights for designing advanced materials and encryption technologies for photonic displays, information security, and intelligent anticounterfeiting.  相似文献

In this study, a single‐step microfluidic approach is reported for encapsulation of enzymes within microcapsules with ultrathin polymeric shell for controlled release triggered by an osmotic shock. Using a glass capillary microfluidic device, monodisperse water‐in‐oil‐in‐water double emulsion droplets are fabricated with enzymes in the core and an ultrathin middle oil layer that solidifies to produce a consolidated inert polymeric shell with a thickness of a few tens to hundreds of nanometers. Through careful design of microcapsule membranes, a high percentage of cargo release, over 90%, is achieved, which is triggered by osmotic shock when using poly(methyl methacrylate) as the shell material. Moreover, it is demonstrated that compared to free enzymes, the encapsulated enzyme activity is maintained well for as long as 47 days at room temperature. This study not only extends industrial applications of enzymes, but also offers new opportunities for encapsulation of a wide range of sensitive molecules and biomolecules that can be controllably released upon applying osmotic shock.  相似文献

Biological ion channels are known as membrane proteins which can turn on and off under environmental stimulus to regulate ion transport and energy conversion. Rapid progress made in biological ion channels provides inspiration for developing artificial nanochannels to mimic the structures and functions of ion transport systems and energy conversion in biological ion channels. Due to the advantages of abundant pore channels, metal–organic frameworks (MOFs) have become competitive materials to control the nanofluidic transport. Herein, a facile in situ synthesis method is developed to prepare hybrid nanochannels constructed by 2D MOFs and porous anodic aluminum (PAA). The introduction of asymmetries in the chemical composition and surface charge properties gives the hybrid outstanding ion current rectification properties and excellent ion selectivity. A power density of 1.6 W m?2 is achieved by integrating it into a salinity‐gradient‐driven device. With advantages of facile fabrication method and high ion selectivity, the prepared 2D MOFs/PAA hybrid membrane offers a promising candidate for power conversion and water desalination.  相似文献

In this study, we report on a novel composite membrane system for pH‐responsive controlled release, which is composed of a porous membrane with linear grafted, positively pH‐responsive polymeric gates acting as functional valves, and a crosslinked, negatively pH‐responsive hydrogel inside the reservoir working as a functional pumping element. The proposed system features a large responsive release rate that goes effectively beyond the limit of concentration‐driven diffusion due to the pumping effects of the negatively pH‐responsive hydrogel inside the reservoir. The pH‐responsive gating membranes were prepared by grafting poly(methacrylic acid) (PMAA) linear chains onto porous polyvinylidene fluoride (PVDF) membrane substrates using a plasma‐graft pore‐filling polymerization, and the crosslinked poly(N,N‐dimethylaminoethyl methacrylate) (PDM) hydrogels were synthesized by free radical polymerization. The volume phase‐transition characteristics of PMAA and PDM were opposite. The proposed system opens new doors for pH‐responsive “smart” or “intelligent” controlled‐release systems, which are highly attractive for drug‐delivery systems, chemical carriers, sensors, and so on.  相似文献

Optimal nanosized drug delivery systems (NDDS) require long blood circulation and controlled drug release at target lesions for efficient anticancer therapy. Red blood cell (RBC) membrane‐camouflaged nanoparticles (NPs) can integrate flexibility of synergetic materials and highly functionality of RBC membrane, endowed with many unique advantages for drug delivery. Here, new near‐infrared (NIR)‐responsive RBC membrane‐mimetic NPs with NIR‐activated cellular uptake and controlled drug release for treating metastatic breast cancer are reported. An NIR dye is inserted in RBC membrane shells, and the thermoresponsive lipid is employed to the paclitaxel (PTX)‐loaded polymeric cores to fabricate the RBC‐inspired NPs. The fluorescence of dye in the NPs can be used for in vivo tumor imaging with an elongated circulating halftime that is 12.3‐folder higher than that of the free dye. Under the NIR laser stimuli, the tumor cellular uptake of NPs is significantly enhanced to 2.1‐fold higher than that without irradiation. The structure of the RBC‐mimetic NPs can be destroyed by the light‐induced hyperthermia, triggered rapid PTX release (45% in 30 min). These RBC‐mimetic NPs provide a synergetic chemophotothermal therapy, completely inhibited the growth of the primary tumor, and suppress over 98% of lung metastasis in vivo, suggesting it to be an ideal NDDS to fight against metastatic breast cancer.  相似文献

Many soft natural tissues display a fascinating set of mechanical properties that remains unmatched by manmade counterparts. These unprecedented mechanical properties are achieved through an intricate interplay between the structure and locally varying the composition of these natural tissues. This level of control cannot be achieved in soft synthetic materials. To address this shortcoming, a novel 3D printing approach to fabricate strong and tough soft materials is introduced, namely double network granular hydrogels (DNGHs) made from compartmentalized reagents. This is achieved with an ink composed of microgels that are swollen in a monomer-containing solution; after the ink is additive manufactured, these monomers are converted into a percolating network, resulting in a DNGH. These DNGHs are sufficiently stiff to repetitively support tensile loads up to 1.3 MPa. Moreover, they are more than an order of magnitude tougher than each of the pure polymeric networks they are made from. It is demonstrated that this ink enables printing macroscopic, strong, and tough objects, which can optionally be rendered responsive, with high shape fidelity. The modular and robust fabrication of DNGHs opens up new possibilities to design adaptive, strong, and tough hydrogels that have the potential to advance, for example, soft robotic applications.  相似文献

Artificial cells or cell mimics have drawn significant attention in cell biology and material science in the last decade and its development will provide a powerful toolbox for studying the origin of life and pave the way for novel biomedical applications. Artificial cells and their subcompartments are typically constructed from a semipermeable membrane composed of liposomes, polymersomes, hydrogels, or simply aqueous droplets enclosing bioactive molecules to perform cellular‐mimicking activities such as compartmentalization, communication, metabolism, or reproduction. Despite the rapid progress, concerns regarding their physical stability (e.g., thermal or mechanical) and tunability in membrane permeability have significantly hindered artificial cells systems in real‐life applications. In addition, developing a facile and versatile system that can mimic multiple cellular tasks is advantageous. Here, an ultrastable, multifunctional and stimulus‐responsive artificial cell system is reported. Constructed from metal‐phenolic network membranes enclosing enzyme‐containing metal‐organic frameworks as organelles, the bionic cell system can mimic multiple cellular tasks including molecular transport regulation, cell metabolism, communication and programmed degradation, and significantly extends its stability range across various chemical and physical conditions. It is believed that the development of such responsive cell mimics will have significant potentials for studying cellular reactions and have future applications in biosensing and drug delivery.  相似文献

Smart nanocarriers are of particular interest for highly effective photodynamic therapy (PDT) in the field of precision nanomedicine. Nevertheless, a critical challenge still remains in the exploration of potent PDT treatment against hypoxic tumor. Herein, light‐triggered clustered polymeric vesicles for photoinduced hypoxic tumor ablation are demonstrated, which are able to deeply penetrate into the tumor and simultaneously afford oxygen supply upon light irradiation. Hydrogen peroxide (H2O2) and poly(amidoamine) dendrimer conjugating chlorin e6/cypate (CC‐PAMAM) are coassembled with reactive‐oxygen‐species‐responsive triblock copolymer into the polymeric vesicles. Upon 805 nm irradiation, the vesicles exhibit the light‐triggered thermal effect that is able to decompose H2O2 into O2, which distinctly ensures the alleviation of tumor hypoxia at tumor. Followed by 660 nm irradiation, the vesicles are rapidly destabilized through singlet oxygen‐mediated cleavage of copolymer under light irradiation and thus allow the release of photoactive CC‐PAMAM from the vesicular chambers, followed by their deep penetration in the poorly permeable tumor. Consequently, the light‐triggered vesicles with both self‐supplied oxygen and deep tissue penetrability achieve the total ablation of hypoxic hypopermeable pancreatic tumor through photodynamic damage. These findings represent a general and smart nanoplatform for effective photoinduced treatment against hypoxic tumor.  相似文献

A two‐layer approach is reported for the formation of a thermally triggered reversible adhesive, involving a thermally‐responsive polymer matrix coated on tannic acid‐pretreated substrates/tissues. Interfacial adhesion originates from strong molecular interactions of tannic acid with both the polymer matrix and the substrate/tissue. The reversibility is due to a temperature‐triggered phase transition of the polymer matrix, leading to cohesive failure. Depending on different gelation mechanisms, the polymer forms a highly cohesive gel or soft solid upon either warming or cooling, leading to a strong adhesion to the tissues at physiological temperatures. Detachment of the adhesive is triggered by a temperature‐induced compromise of cohesive strength of the polymer matrix, by the opposite gel‐to‐sol transition. This facile, low‐cost, and modular design offers a reversible adhesive platform which is useful for biomedical and industrial applications.  相似文献

Water repellency is often generated by taking advantage of surface textures and low surface energy coatings such as the one afforded by long perfluorinated side‐chains polymers. However, new regulations are phasing out these polymers because of their related health and safety hazard concerns. This is a particular challenge for water‐repellent fabrics as consumers expect safer products with stable performance and new functionalities. In this work, an approach is developed that allows for iCVD deposition of durable, conformal short fluorinated polymers stabilized with a crosslinking agent. As a result, high hydrophobicity and low liquid adhesion are achieved simultaneously while maintaining initial substrate breathability. It is explained why this polymeric coating—1H,1H‐perfluorooctyl methacrylate co divinylbenzene—exhibits remarkable hydrophobic properties amidst a wide range of other possible candidates. In order to further enhance the dynamic water repellency performance, the chemical treatment is combined with physical texturing—obtained through microsandblasting, a process particularly suitable for fabrics—thus making this combined approach a suitable candidate to meet the industrial needs. This work paves the way for the development of environmentally friendly, highly repellent coatings for large volume production and the application of roll‐to‐roll coating techniques, and multifunctionalization of fabrics and wearable devices.  相似文献

Crack‐free three‐dimensional (3D) colloidal silica crystals are fabricated on an elastomeric polydimethylsiloxane (PDMS) stamp via the lift‐up method. A surface relief structure is fabricated on the PDMS substrate to enable the formation of colloidal crystal assemblies that cannot be achieved on a plane PDMS substrate owing to the hydrophobic nature of its surface. Four samples of uniform silica particles having different sizes are prepared for colloidal crystal assembly on PDMS substrates with various relief patterns. This strategy not only provides a means for the assembly of crack‐free colloidal crystals on a soft hydrophobic surface via the lift‐up method but enables the transfer of the crack‐free colloidal crystals onto a curved surface.  相似文献

Beside pigment absorption and reflection by periodic photonic structures, natural species often use light scattering to achieve whiteness. Synthetic hydrogels offer opportunities in stimuli‐responsive materials and devices; however, they are not conventionally considered as ideal materials to achieve high whiteness by scattering due to the ill‐defined porosities and the low refractive index contrast between the polymer and water. Herein, a poly(N‐isopropylacrylamide) hydrogel network with percolated empty channels (ch‐PNIPAm) is demonstrated to possess switchable bright whiteness upon temperature changes, obtained by removing the physical agarose gel in a semi‐interpenetrating network of agarose and PNIPAm. The hydrogel is highly transparent at room temperature and becomes brightly white above 35 °C. Compared to conventional PNIPAm, the ch‐PNIPAm hydrogel exhibits 80% higher reflectance at 800 nm and 18 times faster phase transition kinetics. The nanoscopic channels in the ch‐PNIPAm facilitate water diffusion upon phase transition, thus enabling the formation of smaller pores and enhanced whiteness in the gel. Furthermore, fast photothermally triggered response down to tens of milliseconds can be achieved. This unique property of the ch‐PNIPAm hydrogel to efficiently scatter visible light can be potentially used for, e.g., smart windows, optical switches, and, as demonstrated in this report, thermoresponsive color displays.  相似文献
