The hydrogen-producing strain PROH2 pertaining to the genus Clostridium was successfully isolated from a shallow submarine hydrothermal chimney (Prony Bay, New Caledonia) driven by serpentinization processes. Cell biomass and hydrogen production performances during fermentation by strain PROH2 were studied in a series of batch experiments under various conditions of pH, temperature, NaCl and glucose concentrations. The highest hydrogen yield, 2.71 mol H2/mol glucose, was observed at initial pH 9.5, 37 °C, and glucose concentration 2 g/L, and was comparable to that reported for neutrophilic clostridial species. Hydrogen production by strain PROH2 reached the maximum production rate (0.55 mM-H2/h) at the late exponential phase. Yeast extract was required for growth of strain PROH2 and improved significantly its hydrogen production performances. The isolate could utilize various energy sources including cellobiose, galactose, glucose, maltose, sucrose and trehalose to produce hydrogen. The pattern of end-products of metabolism was also affected by the type of energy sources and culture conditions used. These results indicate that Clostridium sp. strain PROH2 is a good candidate for producing hydrogen under alkaline and mesothermic conditions.  相似文献

In view of increasing attempts for the production of renewable energy, the production of biohydrogen energy by a new mesophilic bacterium Clostridium sp. YM1 was performed for the first time in the dark fermentation. Experimental results showed that the fermentative hydrogen was successfully produced by Clostridium sp. YM1 with the highest cumulative hydrogen volume of 3821 ml/L with a hydrogen yield of 1.7 mol H2/mol glucose consumed. Similar results revealed that optimum incubation temperature and pH value of culture medium were 37 °C and 6.5, respectively. The study of hydrogen production from glucose and xylose revealed that this strain was able to generate higher hydrogen from glucose compared to that from xylose. The profile of volatile fatty acids produced showed that hydrogen generation by Clostridium sp. YM1 was butyrate-type fermentation. Moreover, the findings of this study indicated that an increase in head space of fermentation culture positively enhanced hydrogen production.  相似文献

The production of hydrogen from soft-drink wastewater in two upflow anaerobic packed-bed reactors was evaluated. The results show that soft-drink wastewater is a good source for hydrogen generation. Data from both reactors indicate that the reactor without medium containing macro- and micronutrients (R2) provided a higher hydrogen yield (3.5 mol H2 mol−1 of sucrose) as compared to the reactor (R1) with a nutrient-containing medium (3.3 mol H2 mol−1 of sucrose). Reactor R2 continuously produced hydrogen, whereas reactor R1 exhibited a short period of production and produced lower amounts of hydrogen. Better hydrogen production rates and percentages of biogas were also observed for reactor R2, which produced 0.4 L h−1 L−1 and 15.8% of H2, compared to reactor R1, which produced 0.2 L h−1 L−1 and 2.6% of H2. The difference in performance between the reactors was likely due to changes in the metabolic pathway for hydrogen production and decreases in bed porosity as a result of excessive biomass growth in reactor R1. Molecular biological analyses of samples from reactors R1 and R2 indicated the presence of several microorganisms, including Clostridium (91% similarity), Enterobacter (93% similarity) and Klebsiella (97% similarity).  相似文献

The fermentative hydrogen production capability of the newly isolated Clostridium sp. 6A-5 bacterium was studied in a batch cultivation experiment. Various culture conditions (temperature, initial pH, and glucose concentration) were evaluated for their effects on cell growth and hydrogen production (including the yield and rate) of Clostridium sp. 6A-5. Optimal cell growth was observed at 40 °C, initial pH 7.5–8, and glucose concentration 16–26 g/L. The optimal hydrogen yield was obtained at 43 °C, initial pH 8, and glucose concentration 10–16 g/L. Hydrogen began to evolve when cell growth entered the mid-exponential phase and reached the maximum production rate at the late exponential and stationary phases. The maximum hydrogen yield, and rate were 2727 mL/L, and 269.3 mL H2/L h, respectively. These results indicate that Clostridium sp. 6A-5 is a good candidate for mesophilic fermentative hydrogen production.  相似文献

The present study aimed to evaluate the hydrogen production of a microbial consortium using different concentrations of sugarcane vinasse (2–12 g COD L−1) at 37 °C and 55 °C. In mesophilic tests, the increase in vinasse concentration did not significantly impact the hydrogen yield (HY) (from 1.72 to 2.23 mmol H2 g−1 CODinfluent) but had a positive effect on the hydrogen production potential (P) and hydrogen production rate (Rm). On the other hand, the increase in the substrate concentration caused a drop in HY from 2.31 to 0.44 mmol H2 g−1 CODinfluent in the tests performed at 55 °C with vinasse concentrations from 2 to 12 g COD L−1. The mesophilic community was composed of different species within the Clostridium genus, and the thermophilic community was dominated by organisms affiliated with the Thermoanaerobacter genus. Not all isolates affiliated with the Clostridium genus contributed to a high HY, as the homoacetogenic pathway can occur.  相似文献

We performed an experiment of continuous anaerobic hydrogen fermentation as a pilot-plant-scale test, in which waste from a bread factory was fermented by microflora of rice rhizosphere origin. The community structure of microflora during anaerobic hydrogen fermentation was analyzed using PCR-DGGE, FISH, and quinone profiles. The relation of those results to hydrogen generation was discussed. Results show that a suitable condition was a reactor temperature of 35 °C, with HRT 12–36 h, volume load of 30–70 kg-CODCr/m3 day, and maximum hydrogen production rate of 1.30 mol-H2/mol-hexose. Regarding characteristics of microflora during fermentation, PCR-DGGE results show specific 16S rDNA band patterns; Megasphaera elsdenii and Clostridium sp. of the hydrogen-producing bacteria were identified. M. elsdenii was detected throughout the fermentation period, while Clostridium sp. of hydrogen-producing bacteria was detected on the 46th day. Furthermore, FISH revealed large amounts of Clostridium spp. in the sample. The quinone profile showed that the dominant molecular species of quinone is MK-7. Because Clostridium spp. belong to MK-7, results suggest that the quinone profile result agrees with the results of PCR-DGGE and FISH. Microflora in the rhizosphere of rice plants can be a possible resource for effective bacteria of biohydrogen production.  相似文献

An anaerobic fermentation of food waste was conducted in a 0.5 L bioreactor incubated at a thermophilic temperature of 55 °C to evaluate the effects of different controlled pH values (5.0, 5.5 and 6.0) on biohydrogen production. Effective biohydrogen production was found at controlled pH 5.5 and 6.0 corresponding to lower lactic acid production compared to pH 5.0. It was demonstrated that biohydrogen production from food waste was pH-dependent with hydrogen yields of 79, 76 and 23 mmol H2/L-media/d for pH 5.5, 6.0 and 5.0, respectively. Specific microbial determination for Clostridium sp. and total bacteria quantification were carried out by the fluorescent in-situ hybridization (FISH) technique. The number of Clostridium sp. for acclimatized sludge, fermentation broth at pH 5.0, 5.5 and 6.0 were 2.9 × 108, 3.6 × 108, 7.8 × 108 and 5.4 × 108 cells/ml, respectively. The quantification analysis showed that 92% of the total bacteria belonged to Clostridium sp. from clusters I and XI from the sample at controlled pH 5.5. The denaturing gradient gel electrophoresis (DGGE) bands of the sample after heat-treatment, acclimatization and during fermentation indicated the presence of Bacteroidetes, Caloromator australicus sp. and Clostridium sp.  相似文献

Thermophilic hydrogen production from xylan by Thermoanaerobacterium thermosaccharolyticum KKU-ED1 isolated from elephant dung was investigated using batch fermentation. The optimum conditions for hydrogen production from xylan by the strain KKU-ED1 were an initial pH of 7.0, temperature of 55 °C and xylan concentration of 15 g/L. Under the optimum conditions, the hydrogen yield (HY), hydrogen production rate (HPR) and xylanase activity were 120.05 ± 15.07 mL H2/g xylan, 11.53 ± 0.19 mL H2/L h and 0.41 units/mL, respectively. The optimum conditions were then used to produce hydrogen from 62.5 g/L sugarcane bagasse (SCB) (equivalent to 15 g/L xylan) in which the HY and HPR of 1.39 ± 0.10 mL H2/g SCB (5.77 ± 0.41 mL H2/g xylan) and 0.66 ± 0.04 mL H2/L h, respectively, were achieved. In comparison to the other strains, the HY of the strain KKU-ED1 (120.05 ± 15.07 mL H2/g xylan) was close to that of Clostridium sp. strain X53 (125.40 mL H2/g xylan) and Clostridium butyricum CGS5 (90.70 mL H2/g xylan hydrolysate).  相似文献

In this study, production of hydrogen (H2) from glucose, xylose, galactose, mannose, arabinose and rhamnose by a strain isolated from activated sludge was investigated. The strain, named as Citrobacter sp. CMC-1, was enriched in cellobiose amended minimal media. Based on 16S rRNA sequence, the CMC-1 strain is a close relative of Citrobacter amalonaticus strain SA01 (99%). Optimal cultivation parameters for H2 production and growth such as pH and temperature were investigated. H2 yields from glucose at optimal conditions (pH 6.0 and 34 °C) were 1.82 ± 0.02 mol-H2/mol-glucose. Strain CMC-1 fermented galactose, mannose, xylose, arabinose and rhamnose. After 48 h incubation, the strain CMC-1 completely fermented all sugars tested, except arabinose. Increase in fermentation period lowered residual formate level in the media and improved H2 production for galactose, mannose and xylose (1.68 ± 0.24, 1.93 ± 0.14 and 1.63 ± 0.07 mol-H2/mol-substrate respectively).  相似文献

Thermoanaerobacterium-rich sludge was used for hydrogen production and phenol removal from palm oil mill effluent (POME) in the presence of phenol concentration of 100–1000 mg/L. Thermoanaerobacterium-rich sludge yielded the most hydrogen of 4.2 L H2/L-POME with 65% phenol removal efficiency at 400 mg/L phenol. Butyric acid and acetic acid were the main metabolites. The effects of oil palm ash, NH4NO3 and iron concentration (Fe2+) on hydrogen production and phenol removal efficiency from POME by Thermoanaerobacterium-rich sludge was investigated using response surface methodology (RSM). The RSM results indicated that the presence of 0.2 g Fe2+/L, 0.3 g/L NH4NO3 and 20 g/L oil palm ash in POME could improved phenol removal efficiency, with predicted hydrogen production and phenol removal efficiency of 3.45 L H2/L-POME and 93%, respectively. In a confirmation experiment under optimized conditions highly reproducible results were obtained, with hydrogen production and phenol removal efficiency of 3.43 ± 0.12 L H2/L-POME and 92 ± 1.5%, respectively. Simultaneous hydrogen production and phenol removal efficiency in continuous stirred tank reactor at hydraulic retention time (HRT) of 1 and 2 days were 4.0 L H2/L-POME with 85% and 4.2 L H2/L-POME with 92%, respectively. Phenol degrading Thermoanaerobacterium-rich sludge comprised of Thermoanaerobacterium thermosaccharolyticum, Thermoanaerobacterium aciditolerans, Desulfotomaculum sp., Bacillus coagulans and Clostridium uzonii. Phenol degrading Thermoanaerobacterium-rich sludge has great potential to harvest hydrogen from phenol-containing wastewater.  相似文献

Toxicity prevents the bioenergy content of certain industrial effluents from being recovered. An enriched Clostridium butyricum strain was employed to produce hydrogen by fermentation from cellobiose in the presence of phenol at 200–1500 mgl−1. The enriched Cl. butyricum yielded the most hydrogen at 2.1 mol H2 mol−1 cellobiose with 600 mgl−1 phenol. Butyrate was the main metabolite. Cell metabolism was substantially inhibited at a phenol concentration of 1500 mgl−1. Part of the phenol was co-degraded during the test, helping to eliminate the toxicity of wastewater. Both the pyruvate oxidative decarboxylation pathway and the NADH pathway contributed to biohydrogen production. Phenol toxicity more strongly inhibits soluble hydrogenase than it does membrane-bound hydrogenase. Although the NADH pathway dominated at low phenol concentration, increasing the phenol concentration shifted the biohydrogen pathway toward decarboxylation.  相似文献

Biological mycelia pellets, which are formed spontaneously in the process of Aspergillus niger Y3 fermentation, were explored as carrier for immobilization of Clostridium sp. T2 to improve hydrogen production. Batch fermentation tests showed that optimal dosage and size of mycelia pellets for hydrogen production were 0.350 g 150 ml−1 medium and 1.5 mm. Under these conditions, hydrogen production with immobilized cells on mycelia pellets was further investigated in continuous stirred-tank reactor (CSTR) with hydraulic retention time (HRT) ranging from 12 to 8 h. It obtained that the maximum hydrogen production rate reached 2.76 mmol H2 L−1 h−1 at 10 h HRT, which was 40.8% higher than the carrier-free process, but slightly lower than the counterpart immobilized in sodium alginate with the value of 3.15 mmol H2 L−1 h−1. SEM observation showed that abundant cells were closely adhered to mycelia pellets. The present results indicate the potential of using mycelia pellets as biological carrier for enhancing hydrogen production.  相似文献

This study evaluates the effect of pH (4-7) on fermentative biohydrogen production by utilizing three isolated Clostridium species. Fermentative batch experiments show that the maximum hydrogen yield for Clostridium butyricum CGS2 (1.77 mmol/mmol glucose) is achieved at pH 6, whereas a high hydrogen production with Clostridium beijerinckii L9 (1.72 mmol/mmol glucose) and Clostridium tyrobutyricum FYa102 (1.83 mmol/mmol glucose) could be achieved under uncontrolled pH conditions (initial pH of 6.4-6.6 and final pH of 4-4.2). Low hydrogen yields (0-0.6 mmol/mmol glucose) observed at pH 4 are due likely to inhibitory effects on the microbial growth, although a low pH can be thermodynamically favorable for hydrogen production. The low hydrogen yields (0.12-0.64 mmol/mmol glucose) observed at pH 7 are attributed not only to thermodynamically unfavorable, but also metabolically unfavorable for hydrogen production. The relatively high levels of lactate, propionate, or formate observed at pH 7 reflect presumably the high enzymatic activities responsible for their production, together with the low hydrogenase activity, resulting in a low hydrogen production. A correlation analysis of the data from present and previous studies on biohydrogen production with pure Clostridium cultures and mixed microflora indicates a close relation between the hydrogen yield (YH2) and the (YH2)/(2(YHAc+YHBu)) ratio, with the observed correlation coefficient (0.787) higher than that (0.175) between YH2 and the molar ratio of butyrate to acetate (B/A). Based on the (YH2)/(2(YHAc+YHBu)) ratios observed at different pHs, a control of pH at 5.5-6.8 would seem to be an effective means to enhance the fermentative biohydrogen production.  相似文献

The effects of lauric (LUA), myristic (MA), palmitic (PA), and a mixture of myristic:palmitic (MA:PA) acids on hydrogen (H2) production from glucose degradation using anaerobic mixed cultures were assessed at 37 °C with an initial pH set at 5.0 and 7.131 mM of each acid. The maximum H2 yield (2.53 ± 0.18 mol mol−1 glucose) was observed in cultures treated with PA. A principal component analysis (PCA) of the by-products and the microbial population data sets detected similarities between the controls and PA treated cultures; however, differences were observed between the controls and PA treated cultures in comparison to the MA and LUA treated cultures. The flux balance analysis (FBA) showed that PA decreased the quantity of H2 consumed via homoacetogenesis compared to the other LCFAs. The control culture was dominated by Thermoanaerovibrio acidaminovorans (60%), Geobacillus sp. and Eubacterium sp. (28%), while Clostridium sp. was less than 1%. Treatment with PA, MA, MA:PA, or LUA increased the H2 producers (Clostridium sp. and Bacillus sp.) population by approximately 48, 67, 86, and 86%, respectively.  相似文献

A limited supply of oil prompts the search for non-traditional energy sources to replace traditional ones. This makes hydrogen gas an appealing alternative source. Photosynthetic organisms capture sunlight very efficiently and convert it into organic molecules. A promising wild strain was isolated for the first time, from the rice paddies of Kazakhstan (Kyzylorda and Almaty regions), which can be considered as one of the most active hydrogen producers compared to the literature. The result showed that among the 13 isolated and collection cyanobacterial strains, Synechocystis sp. S-1 is the most active H2 producer (2.35 μmol H2 mg?1 Chl a h?1) in the light. In contrast, the wild-type cyanobacterium Anabaena variabilis A-1 had higher productivity, nitrogenase activity, and a stronger capacity to produce hydrogen in the dark (8.67 μmol H2 mg?1 Chl a h?1), which matched the maximum yield obtained in the research. The metabolic modulation performed significantly increased hydrogen production. The highest photohydrogen production rate was observed in cells incubated with 25 μmol HEPES and 50 μmol sodium bicarbonate (NaHCO3).  相似文献

Batch and continuous modes for bio-hydrogen production by co-digesting cassava starch wastewater with buffalo dung were investigated. Response surface methodology with central composite design was used to optimize the bio-hydrogen production conditions. A hydrogen production potential of 1787 mL H2/L was achieved under optimal conditions of 2.84 g/L of NaHCO3, an initial pH of 6.77 and a total chemical oxygen demand (tCOD)/total nitrogen ratio of 42.36. A continuous stirred tank reactor was operated under the optimum conditions from batch mode to investigate the effects of hydraulic retention time (HRT) of 72, 60 and 48 h on hydrogen production. The highest hydrogen content, hydrogen production rate and hydrogen yield of 33%, 839 mL H2/L.d and 16.90 mL H2/g-CODadded, respectively, were achieved at a HRT of 60 h. The predominant hydrogen producer under the optimal conditions in batch mode was Clostridium sp. while Clostridium sp., Megasphaera sp. and Chloroflexi sp. were observed in the continuous hydrogen production mode at an optimal HRT.  相似文献

The impact of different chemical microbial stressors (2-bromoethanesulfonate (BES), furfural, fish oil, lauric acid (LUA) and linoleic acid (LA)) on the inhibition of mesophilic hydrogen (H2) consumption was examined in this study. Hydrogen consumption half-life values were used to compare the extent of inhibition by the different microbial stressing agents. A statistical analysis of the percent H2 consumed using Tukey's analysis revealed the following trend: Control > fish oil = linoleic acid (LA (C18:2)) = furfural > BES > lauric acid (LUA (C12:0). The terminal restriction fragment length polymorphism (T-RLFP) results indicated that aceticlastic methanogens (Methanosaeta sp., Methanosarcina sp.) and hydrogenotrophic methanogens (Methanococcus sp.) were inhibited by the different chemical stressing agents. Cultures fed LUA and LA had a high abundance of Clostridium sp., Clostridium propionicum and Propionibacterium acnes. In comparison, BES and furfural fed cultures contained large fractions of Clostridium sp., Eubacteria sp. and Bacteroides sp. while in the fish oil fed cultures, the dominant organism detected was Eubacteria sp. This study indicated that H2 consumption was affected by the chemical stressing agent concentration.  相似文献

A unique thermophilic fermentative hydrogen-producing strain H53214 was isolated from a deep-sea hydrothermal vent environment, and identified as Caloranaerobacter azorensis based on bacterial 16S rRNA gene analysis. The optimum culture condition for hydrogen production by the bacterium, designated C. azorensis H53214, was investigated by the response surface methodology (RSM). Eight variables including the concentration of NaCl, glucose, yeast, tryptone, FeSO4 and MgSO4, initial pH and incubation temperature were screened based on the Plackett–Burman design. The results showed that initial pH, tryptone and yeast were significant variables, which were further optimized using the steepest ascent method and Box–Behnken design. The optimal culture conditions for hydrogen production were an initial pH of 7.7, 8.3 g L−1 tryptone and 7.9 g L−1 yeast. Under these conditions, the maximum cumulative hydrogen volume, hydrogen yield and maximum H2 production rate were 1.58 L H2 L−1 medium, 1.46 mol H2 mol−1 glucose and 25.7 mmol H2 g−1 cell dry weight (CDW) h−1, respectively. By comparison analysis, strain H53214 was superior to the most thermophilic hydrogen producers because of the high hydrogen production rate. In addition, the isolation of C. azorensis H53214 indicated the deep-sea hydrothermal environment might be a potential source for fermentative hydrogen-producing thermophiles.  相似文献

A mesophilic alkaline tolerant fermentative microbe was isolated from estuarine sediment samples and designated as Clostridium butyricum TM-9A, based on 16S rRNA gene sequence. Batch experiments were conducted for investigation of TM-9A strain for its growth and hydrogen productivity from glucose, in an iron containing basal solution supplemented with yeast extract as organic nitrogen source. Hydrogen production started to evolve when cell growth entered exponential phase and reached maximum production rate at late exponential phase. Maximum hydrogen production was observed at 37 °C, initial pH of 8.0 in the presence of 1% glucose. Optimization of process parameters resulted in increase in hydrogen yield from 1.64 to 2.67 mol of H2/mol glucose. Molar yield of H2 increased further from 2.67 to 3.1 mol of H2/mol of glucose with the decrease in hydrogen partial pressure, obtained by lowering the total pressure in the head space of the batch reactor. Acetate and butyrate were the measure volatile fatty acids generated during hydrogen fermentation. TM-9A strain produced hydrogen efficiently from a range of pentose and hexose sugars including di-, tri and poly-saccharides like; xylose, ribose, glucose, rhamnose, galactose, fructose, mannose, sucrose, arabinose, raffinose, cellulose, cellobiose and starch.  相似文献

Fermentative hydrogen (H2) production from a steam exploded switchgrass liquor using inhibited mixed anaerobic microbial communities was studied in upflow anaerobic sludge blanket reactors (UASBRs). Increasing the H2 yield was accomplished by treating the inoculum with linoleic acid (LA), varying the hydraulic retention time (HRT) and sparging liquid phase with nitrogen (N2). A maximum H2 yield of 2.56 ± 0.10 mol mol−1 hexose, was obtained at a 6 h HRT in LA treated cultures sparged with N2. Sparging or LA treatment alone was able to enhance the H2 yield by 46 ± 5% and 38 ± 3%, respectively, in comparison to control cultures operating at a 6 h HRT. Of the different methods employed, N2 sparging in combination with LA treatment proved to be more effective in enriching the H2 producing bacteria belonging to Clostridium sp. Species belonging to Propionibacterium, Bacteroides and Eubacterium, which were associated with H2 consumption and reduced byproducts formation, were observed in addition to Clostridium sp. in unsparged control cultures.  相似文献
