Production of the bacteriocins enterocin A and enterocin B in Enterococcus faecium CTC492 was dependent on the presence of an extracellular peptide produced by the strain itself. This induction factor (EntF) was purified, and amino acid sequencing combined with DNA sequencing of the corresponding gene identified it as a peptide of 25 amino acids. The gene encodes a prepeptide of 41 amino acids, including a 16-amino-acid leader peptide of the double-glycine type. Environmental factors influenced the level of bacteriocin production in E. faecium CTC492. The optimal pH for bacteriocin production was 6.2. At pH 5.5, growth was slow, and very little bacteriocin was formed. The presence of NaCl or ethanol (EtOH) was also inhibitory to bacteriocin production, and at high concentrations of these solutes, no bacteriocin production was observed. The induction factor induced its own synthesis, and by dilution of the culture 106 times or more, nonproducing cultures were obtained. Bacteriocin production was induced in these cultures by addition of EntF. The response was linear, and low bacteriocin production could be induced by about 10(-17) M EntF. This response was attenuated by low pH or the presence of high concentrations of NaCl or EtOH, and 300 times more EntF was needed to induce detectable bacteriocin production in the presence of 6.5% NaCl. High levels of bacteriocin production in cultures grown at low pH or in the presence of high concentrations of NaCl or EtOH were obtained by addition of sufficient amounts of EntF.  相似文献

The aims of this follow-up study were: (a) to evaluate the role of ECT technology as a risk factor for several diseases; and (b) to determine if the effects of these diseases on cows' reproductive performance and as risk factors for culling are influenced by exposure to ECTs. Diseases considered were retained placenta, metritis, ketosis, cystic ovaries, silent heat, milk fever, clinical mastitis, and foot and leg problems. We used historical and contemporary controls (with control herds selected to match the experimental herds for size and location). Data consisted of 10,264 Swedish Red and White (SRB) and 5461 Swedish Friesian (SLB) lactation records in 150 herds of which 33 used cow-trainers. Logistic regression was used to estimate the effects of parity and exposure to electric cow-trainers on the risks of diseases and the effects of diseases and exposure to electric cow-trainers on risk of culling. The least-squares procedure was used to estimate the effects of diseases on reproductive performance. The dominant effects associated with use of electric cow-trainers were an increased risk for silent heat, clinical mastitis, ketosis and culling relative to cows in herds not using cow-trainers. Diseases had negative effects on reproductive performance and the effects were larger for cows in herds using cow-trainers. In herds using electric cow-trainers, the largest increase in the interval from first service to conception (58 days) was caused by the occurrences of silent heat, cystic ovaries and the combination of two or more diseases. Retained placenta, metritis, cystic ovaries, clinical mastitis and a combination of two or more diseases increased the risk of culling about two times relative to healthy primiparous cows with the increase being greater for cows in herds using cow-trainers. Silent heat did not increase risk of culling in control groups, but was the largest risk factor for culling in the exposed group. We concluded that exposure to electric cow-trainers increased the incidence risk of silent heat, clinical mastitis, and ketosis and changed silent heat from a neutral disease with respect to culling to a major risk factor. Finally, exposure to cow-trainers increased the general negative effect of diseases on the cows' reproductive performance and risk for culling.  相似文献

The effects of inbreeding on the lifetime performance of dairy cattle were examined using data for production, somatic cell score, and linear type for all Holstein cows that were scored between 1983 and 1993. The results of fixed and mixed animal models differed. Relative net income adjusted for opportunity cost for the 2,610,123 cows with an 84-mo opportunity for herdlife was depressed by $14.79 for fluid market pricing and by $12.40 for manufacturing pricing per 1% increase in inbreeding. Mixed model estimates of depression per 1% of increase in inbreeding were +0.55 d for age at first calving, -6 d for days of productive life, and -4.8 for days in milk. Inbreeding decreased the mature equivalent production of milk, fat, and protein during first lactation by 27, 0.9, and 0.8 kg and the lifetime production of milk, fat, and protein by 177, 6.0, and 5.5 kg, respectively, per 1% increase in inbreeding. Inbreeding had little effect on conformation traits. The effects of inbreeding were cumulative, and effects on lifetime profit functions were relatively larger than the effects on lactation traits. Registered cows had higher levels of inbreeding and larger standard deviations than did grade cows. Inbreeding in registered cows depressed relative net income adjusted for opportunity cost for fluid and manufacturing prices by $24.43 and $21.78, respectively; income was depressed $9.43 and $9.02, respectively, for grade cows. The difference between registered and grade cattle is likely due to the incomplete pedigree information in grade animals. Inbreeding among cows in this study was not high on average, but economic losses represented a significant cost to the producer.  相似文献

To assess the impact of Leptospira interrogans serovar hardjo infection on the reproductive performance of nine dairy herds with evidence of infection, forty years' fertility data were analysed relative to the year of first diagnosis. Fifty per cent of various fertility variables had their lowest values only in the year of diagnosis. Culling rates were highest during the year of diagnosis in five of the herds, and were above 22 per cent in five of nine (55-6 per cent) of the diagnosis years considered compared with seven (22-6 per cent) of the 31 non-diagnosis years. An assessment of the fertility status of the herds by means of a formula with incorporated the first service conception rate, the number of services per conception for cows conceiving, the calving to conception interval and the culling rate, revealed low reproductive performance during the year of diagnosis in six of the nine herds. Abortion rates were highest in four of the herds during the year of diagnosis, and these included the herds in which the fertility status was not lowest. It was concluded that L. interrogans serovar hardjo affected reproductive adversely by causing both abortions and low fertility, but that the effect was temporary.  相似文献

The retinal pigment epithelium (RPE) is clinically involved in diverse ocular inflammatory diseases. Because perturbed RPE cells produce a variety of inflammatory substances, RPE cells may play an integral part in these diseases. Interleukin-1 (IL-1) and granulocyte-macrophage colony-stimulating factor (GM-CSF) are pleiotropic cytokines with the ability to trigger numerous inflammatory responses. This report shows that cultured human RPE cells synthesize interleukin-1 beta (IL-1 beta) and GM-CSF in response to the potentially inflammatory cytokine, IL-1 alpha, but not to E. coli endotoxin. Control RPE cells made little or no mRNA or protein for either IL-1 beta or GM-CSF. Upon stimulation of the cells by IL-1 alpha, both IL-1 beta and GM-CSF mRNAs were readily apparent by 3 hours, persisted for over 24 hours, and were translated into immunologically detectable proteins. GM-CSF protein was secreted into the culture medium, whereas IL-1 beta protein remained cell associated. The IL-1 alpha-induced mRNA and protein production were inhibited by dexamethasone. These observations provide additional evidence that RPE cells are capable of playing a pivotal role during ocular inflammation.  相似文献

Lactation records (n = 2362) of 1402 crossbred cows in 22 cooperating dairy herds in southeastern Brazil were evaluated. Cows were mixtures of Zebu (Gir, Guzera, and unknown) and European breeding (mostly Holstein). Lactation milk yields were expressed as total, 3050-d, or deviated 305-d yields, either adjusted or unadjusted for days in milk (DIM). Mean DIM was 280. Arithmetic means unadjusted for DIM were 1942, 1666, and 5 kg per record. Milk yields of daughters from sires of 6/8 and 7/8 European breeds were higher than yields of daughters from sires of 5/8 European breeds when data were either adjusted or unadjusted for DIM. The differences associated with breed group of sire were only slightly reduced when records were adjusted for DIM. There was no evidence of a decline in milk yield as the fraction of European breeding of the sire increased from 6/8 to 7/8. For a given breed group of sire, whether the grandsire was purebred or crossbred had no detectable effect. These results should be useful in determining strategies for crossbreeding of dairy cows in tropical areas, particularly when crossbred sires are used.  相似文献

Individual milk production curves of 542 cows with 722 cases of mastitis were compared with control curves drawn from healthy lactating cows. First, differences were classified into patterns of milk loss, and their distribution was analyzed among the patterns with regard to breed, season, lactation number, stage of lactation, and milk production. Then, individual milk losses were estimated and analyzed according to the same factors. In early lactation, almost 7% of mastitis cases necessitated culling or drying off. For 36% of the cases of mastitis occurring in early lactation, milk production was affected for an extended period, and the milk loss induced was 911 kg on average over the entire lactation. Fifty-two percent of the mastitis cases occurring after lactation peak was not accompanied by marked modifications of the lactation curves. For 38% of cases of mastitis occurring from mid to late lactation, milk production was affected for an extended period. When milk production was the most affected (26% of total cases), the average loss was 850 kg over the entire lactation. For cases in early or mid to late lactation, the production at mastitis onset was a determining factor of the amount and pattern of milk production loss induced. The objective of this study was to evaluate different PGF2 alpha protocols against control protocols for herds with estrus detection efficiencies of 35, 55, and 75% using modeling and simulation: 1) PGF2 alpha treatments based on the presence of a corpus luteum diagnosed by rectal palpation, 2) PGF2 alpha treatments based on the presence of a corpus luteum diagnosed by an on-farm milk progesterone enzyme immunoassay, and 3) PGF2 alpha treatments based on a 14-d fixed treatment schedule without prior screening for ovarian status. After the start of each protocol, estrus detection efficiency was 75% for 7 d after treatment and 35 or 0% for the following week. For the third protocol, an additional modification at estrus detection efficiencies of 85 and 55%, respectively, in the 1st and 2nd wk after treatment was evaluated to establish a protocol for best case assumptions. All protocols improved reproductive performance relative to that of controls with estrus detection efficiencies of 35 and 55%. The mean number of days open was reduced from 124.3 d in the control herd to 95.9, 95.0, and 92.7 for the protocols based on rectal palpation, milk progesterone test, and the fixed treatment schedule, respectively. The protocols based on a fixed treatment schedule were superior to protocols based on rectal palpation and on-farm milk progesterone tests and resulted in better reproductive performance and a higher increase in net return per cow per year. Relative to a control herd with an estrus detection efficiency of 55%, it was cost effective to spend up to $10 per dose of PGF2 alpha, $9 per milk progesterone test, and $6 per rectal palpation.  相似文献

A number of factors limit the usefulness of blood or metabolic profiles. These include sampling problems, low correlations with nutrient intake, inconsistent patterns in disease, and difficulties in interpretation. Despite these limitations, profiles properly used may serve as an adjunct to more conventional technology in alleviating some dairy herd problems. Their use appears justified when feed analysis, ration evaluation, disease testing, and checks on management do not alleviate herd problems. Considerable potential for misuse of profiles exists due to the complexities of interpretation.  相似文献

Four multiparous Holstein cows fitted with rumen and duodenal cannulas were used in a 4 x 4 Latin square with 20-d periods. Four diets were formulated for high and low rumen availabilities of nonstructural carbohydrate and protein. Cows were milked and fed three times daily. Milk production averaged 39 kg/d and was unaffected by treatment. Dry matter intake and 4% FCM production were increased by 1 kg/d for cows fed the low rumen-available nonstructural carbohydrate diets. Milk protein percentage was elevated when either the high rumen-available nonstructural carbohydrate or high rumen-available protein diets were fed. Low rumen-available protein diets increased duodenal passage of total essential AA but did not increase passage of Arg, Ile, and Met. Passage of bacterial N was highest (262 g/d) when high rumen-available nonstructural carbohydrate was combined with high rumen-available protein and was lowest when high rumen-available nonstructural carbohydrate was combined with low rumen-available protein (214 g/d). Diets with low rumen availabilities of protein were not advantageous, possibly because duodenal passage of one or more of the limiting AA was not increased. Passage of bacterial N to the duodenum was highest when rumen availabilities of both nonstructural carbohydrate and protein were high.  相似文献

Samples of bulk milk were taken from all 33,636 Dutch dairy herds in November 1994 and tested for the presence of bovine herpesvirus 1 (BHV-1) antibodies with a gB-blocking ELISA. Sixteen per cent of the herds had a negative BHV-1 status in the bulk milk. Farms with only dairy cows were 1.9 times more likely to have a negative or weakly positive BHV-1 status than herds which also had beef/veal animals. Farms in areas containing less than one herd/km2 were 1.5 times more likely to have a negative or weakly positive BHV-1 status than herds in areas with more than three herds/km2. Differences in numbers of animals per unit area were not significantly associated with BHV-1 status. The probability of herds having a negative or weakly positive BHV-1 status decreased linearly with herd size by a factor of 1.2 per 10 animals. The purchase of stock was significantly associated with a negative or weakly positive BHV-1 status, but there was an interaction between farm type and purchase of stock. For farms with both dairy and beef/veal animals there was a weak association between the purchase of stock and BHV-1 status. For pure dairy herds the probability of having a negative or weakly positive BHV-1 status decreased linearly with the numbers of purchased stock by a factor of 1.3 per 10 animals purchased.  相似文献

Serological (Western blot) detection of bovine immunodeficiency-like virus (BIV) in Holstein dairy herds is reported in Costa Rica for the first time, as well as the isolation of the virus, from a seropositive bovine, by cocultivation of peripheral blood mononuclear cells (PBMC) with embryonic rabbit epithelial (EREp) cells. The isolated strain, BIVCR1, reacted similarly in Western blot as the reference strain BIV R29 and is clearly distinguishable from bovine leukaemia virus (BLV). The data suggest an association between BIV infections and BLV infections, as it has been reported elsewhere. From these results it can be concluded that BIV is present in Costa Rica and it is suggested that these viral infections will probably follow the epidemiological parameters of BLV infections in Costa Rica, reaching high infection rates in dairy herds.  相似文献

Data from 5468 Holstein dairy cows in 32 herds were used to determine the effect of the use of bovine somatotropin (bST) on culling practices over a 13-mo period. After an initial herd inventory, monthly information regarding cow entry and exit from the herds was obtained by seven participating veterinarians. Culling was coded by farmers for the following reasons: low production, reproduction, somatic cell count, mastitis, sickness, dairy purposes, lameness, death, and other. In the control herds, cows were not treated with bST during the trial. Adopter herds were defined as herds that utilized supplemental bST for > or = 25% of the cow-days during the trial. Mean herd use of bST in adopter herds was 38.6%. No difference in the number of cows culled per cow-day at risk was detected between control and adopter herds (0.09 and 0.11%, respectively). Amount of in-herd use of bST was unrelated to culling. No significant differences were determined between adopter and control herds in the percentage of cows that were culled for any of the nine possible culling reasons. The results of this study suggest that culling patterns in herds that use bST are unaffected for at least the first year after adoption.  相似文献

Regulatory foci of promotion and prevention have been shown to relate differentially to occupational safety and production. This research proposes that task complexity can help explain the differences reported between these 2 self-regulatory processes and safety and productivity performance. Results revealed that promotion is positively related to production and prevention is positively related to safety regardless of task complexity. However, when task complexity is high, promotion negatively relates to safety and prevention negatively relates production. Implications for work motivation theory and research, as well as avenues for future research, are discussed. Lactation has been negatively associated with fertility because pregnancy rates in maiden heifers exceed those obtained after first or subsequent calvings. The extent of this difference is less in pasture-fed dairy cows ( < 10%) than in American Holsteins ( > 20%) fed grain and conserved forages. The latter cows have pregnancy rates to first insemination and oestrus detection rates of only 40 to 45%. This suggests that the subsequent fertility of inherently fertile Holstein heifers may be severely compromised by high levels of milk production. International comparisons show that pasture-fed dairy cows may experience extended periods of anovulatory anoestrum but have normal fertility (60% pregnancy rate to first insemination) once cycling. The high-producing American Holstein may ovulate within 4 weeks postpartum but is more likely to continue ovulating without being detected in oestrus. Both situations are associated with negative energy balances (NEB) during early lactation. The severity and duration of this NEB may vary with body condition at calving, age or parity, ration formulation, production level and environmental factors. Relative daily milk yield is not an absolute indicator of NEB, because some lower producing cows within a herd have lower feed intakes and more severe energy deficits. NEB is not simple to measure; nonetheless, it is correlated with genetic improvement for milk yield. A positive energy balance, greater weight gain and higher body condition score have all been shown to be positively correlated with plasma progesterone concentrations in early lactation. No studies have investigated the possibility that the rapid increase in metabolic rate at this time may also alter steroid concentrations with consequent effects on oestrous behaviour and fertility. Studies to more precisely define the effects of increasing milk yields in early lactation, especially in Holsteins, may need to be completed in Australia and New Zealand. Oestrus detection rates and pregnancy rates for American Holsteins of less than 50% are accepted widely in the USA. Such low detection rates confound studies on fertility. The objective should be to increase these 2 rates to at least 80% and 60% respectively. This may involve the use of controlled breeding, especially if oestrous behaviour is less overt in high-producing Holstein cows.  相似文献

A survey of Isospora suis performed in 177 faecal samples from 30 swine farms detected thin wall type I. suis oocysts in seven samples. This type of oocyst measuring 23.9 by 20.7 microns had a retracted thin wall similar to that of the genus Sarcocystis. This type of oocysts, isolated from four different faecal samples, was inoculated in four-five-days-old piglets free of contamination in order to verify the life cycle and pathogenicity of the species. The pigs were kept in individual metal cages and fed with cow milk. Daily faecal collections and examinations were performed until the 21st day after infection. MacMaster and Sheather's methods were used for oocyst counting and identification. Infected piglets produced yellowish-pasty diarrhoea with slight dehydration. The prepatent and patent periods were respectively from 6 to 9 and 3 to 10 days after infection. Oocyst elimination was interrupted on the 10th and 11th days after infection with biphasic cycles. Thin and thick wall oocysts were detected in the same faecal samples. Thin walls were not observed in unsporulated oocytes. The observations suggest that this type of oocysts could appear in specific strains which occur in the later stages of their development. These oocysts seem to be responsible for clinical and pathogenic signs of neonatal isosporosis in pigs.  相似文献

OBJECTIVE: To assess the effect of vaccination against bovine respiratory syncytial virus on milk production, reproductive performance, and health in lactating dairy cows. DESIGN: Prospective randomized block design. ANIMALS: 385 Holstein dairy cows and heifers. PROCEDURE: Cows were grouped by lactation number, season of calving, and previous mature equivalent 305-day milk production (where appropriate). Prior to parturition, cows and heifers were randomly assigned to be vaccinated i.m. against infectious bovine rhinotracheitis, bovine viral diarrhea, and parainfluenza 3 viruses by use of a three-way vaccine, or to be vaccinated against those viruses as well as bovine respiratory syncytial virus, using a four-way vaccine. Milk production was measured daily through 305 days of lactation. Reproductive and medical records were reviewed to obtain insemination dates and record medical problems of cows in each vaccine treatment group. RESULTS: Compared with the three-way vaccine, administration of the four-way vaccine was associated with higher milk production (1.39 kg [3.06 lb] more milk/d) in first-parity cows during the first 21 weeks of lactation. Vaccination did not have any effect on milk production after the first 21 weeks of lactation in cows of any parity. Conception rates at first insemination were higher for four-way vaccinated first-parity cows than for three-way vaccinated first-parity cows (54.6 vs 32.7%). Compared with second-parity cows that received the three-way vaccine, first insemination conception rate was improved for second-parity cows vaccinated with the four-way vaccine (28.9 vs 47.8%, respectively). In cows of third or greater parity, first insemination conception rate was not different between the 2 vaccine treatment groups. CLINICAL IMPLICATIONS: Vaccination of heifers against bovine respiratory syncytial virus prior to partrition may increase milk production and first insemination conception rates.  相似文献

An extensive questionnaire was developed and used to collect data from 33 herds that were on the New York State Paratuberculosis Control Program, to study farm factors associated with the presence of Mycobacterium paratuberculosis infection in dairy herds. The results of the last whole herd paratuberculosis fecal culture were used to indicate presence of infection in a herd, with herds having one or more animals positive classified as 'infected'. The average prevalence within herds was 5.2%. Fourteen herds were uninfected and 19 herds had prevalences ranging from 0.7%-28.2%. Data on 31 continuous and 67 categorical risk factors were collected by questionnaire. Ten factors were significantly associated with prevalence risk of infection in the univariable logistic regression. These factors were: the type of farm operation (commercial/registered or both); earlier diagnosis of the disease before entering the control program; number of clinical cases in the previous year; whether clinical cases were raised or purchased animals; typical signs in clinical cases; exposure of calves 0-6 weeks of age to feces of adult cows; contact of young stock with adult animal feces from using the same equipment to clean the housing for both groups of animals; spreading feces on fields from which forage is later harvested and fed to animals of any age group; what is done with animals that are suspected of having paratuberculosis or test positive on culture; and frequency of cleaning the cow barn. Stepwise logistic regression was used to determine the significance of each risk factor while controlling simultaneously for the effect of other factors. The significant factors were the type of farm operation, clinical signs, and exposure of calves to feces of adult cows. Commercial herds, presence of clinical signs typical of paratuberculosis in animals, and exposure of calves 0-6 weeks old to feces of adult cows all indicate a higher likelihood that a herd is infected with M. paratuberculosis.  相似文献

The effect of seven diseases on culling was measured in 7523 Holstein cows in New York State. The cows were from 14 herds and had calved between January 1, 1994 and December 31, 1994; all cows were followed until September 30, 1995. Survival analysis was performed using the Cox proportional hazards model to incorporate time-dependent covariates for diseases. Different intervals representing stages of lactation were considered for effects of the diseases. Five models were fitted to test how milk yield and conception status modified the effect of diseases on culling. Covariates in the models included parity, calving season, and time-dependent covariates measuring diseases, milk yield of the current lactation, and conception status. Data were stratified by herd. The seven diseases and lactational risks under consideration were milk fever (0.9%), retained placenta (9.5%), displaced abomasum (5.3%), ketosis (5.0%), metritis (4.2%), ovarian cysts (10.6%), and mastitis (14.5%). Older cows were at a much higher risk of being culled. Calving season had no effect on culling. Higher milk yield was protective against culling. Once a cow had conceived again, her risk of culling dropped sharply. In all models, mastitis was an important risk factor throughout lactation. Milk fever, retained placenta, displaced abomasum, ketosis, and ovarian cysts also significantly affected culling at different stages of lactation. Metritis had no effect on culling. The magnitude of the effects of the diseases decreased, but remained important, when milk yield and conception status were included as covariates. These results indicated that diseases have an important impact on the actual decision to cull and the timing of culling. Parity, milk yield, and conception status are also important factors in culling decisions.  相似文献
