Since there is no available information about the natural occurrence of ochratoxin‐producing fungi and ochratoxin A (OTA) in peanut seeds in Argentina, the aims of this work were to isolate and identify Aspergillus section Nigri and to evaluate the natural occurrence of OTA in stored peanut seeds. Likewise, the capacity to produce OTA by Aspergillus section Nigri was studied. Forty‐seven samples of peanut seeds were obtained from a storage plant in the south of Córdoba Province, Argentina. Each sample of 100 seeds was surface‐disinfected and plated onto a dichloran 18% glycerol agar (DG18). OTA was detected by HPLC. The production of OTA in strains belonging to section Nigri was tested in YES medium (2% yeast extract, 15% sucrose). Aspergillus spp., the most frequent mould, occurred in 100% of samples, followed by Penicillium spp. (87%) and Eurotium spp. (6.4%). A. flavus was isolated in 100% of the samples, followed by A. niger var. niger, A. niger var. awamori and A. carbonarius at a lower frequency, 89%, 68% and 4%, respectively. OTA was found in 32% of the peanut seed samples, with mean levels ranging from 0.5 to 170 ng g?1. The mean recovery of the method used was 85 ± 2%. Forty‐three isolates (27%) of Aspergillus section Nigri, were found to be OTA producing strains. The highest percentage of ochratoxigenic strains (57%) was found within the A. carbonarius ssp. ABSTRACT

Food decay by spoilage fungi leads to significant economic losses and hazards to consumers’ health due to the potential of mycotoxin occurrence. Ochratoxin A (OTA) is a mycotoxin known as nephrotoxic and carcinogenic to humans. Natural capsaicin was evaluated for its effectiveness against the growth of five Aspergillus section Nigri strains and accumulation of OTA in inoculated black grapes. Results showed that capsaicin was effective in inhibiting fungal growth and OTA production by new four Aspergillus section Nigri strains (ATHUM 6997, 6998, 6999, 7000) and by Aspergillus carbonarius as well. Moreover, capsaicin addition exhibited maximum inhibition of OTA produced by ATHUM 6997, 6998, 6999, and 7000 in black grapes at 28.9%, 8.6%, 68.4%, and 78.1%, respectively. Inhibition percentage of OTA production by A. carbonarius in grapes treated with capsaicin was estimated at 61.5%. These results suggest that capsaicin influences the OTA biosynthesis pathway of all Aspergillus section Nigri strains and therefore could be used as an effective natural preservative against OTA contamination of vineyards. Risk assessment revealed that when grapes are treated with capsaicin, consumers are less exposed to OTA.  相似文献

Fungi responsible for the presence of Ochratoxin A (OTA) in grapes have been identified as belonging to the black aspergilli, Aspergillus section Nigri, among which Aspergillus carbonarius is the main producer. Despite the widespread occurrence of OTA in various types of wine, there is no specific information on the ability of black aspergilli to infect berries and to produce OTA in different grape varieties. The aim of this study was to investigate the susceptibility to A carbonarius of twelve grape varieties representative of a wide range of characteristics, including geographical distribution. In all the conditions of berry status (intact and damaged) and temperature (20 and 25 °C) tested, the incidence of colonised berries and their OTA contents were always low in ‘Bianco d'Alessano’, ‘Pampanuto’ and ‘Uva di Troia’. In contrast, ‘Cabernet Sauvignon’, ‘Trebbiano’ and ‘Verdeca’ showed high fungal incidence and OTA content. The information gathered is relevant for defining risk factors for OTA presence in grapes, even if these data need to be confirmed by observations in vineyards. Aspergillus section Nigri are described as the main source of ochratoxin A (OTA) contamination in grapes and wine worldwide. The knowledge of the factors affecting grape contamination by species included in this section and OTA production is essential to be able to reduce their presence, not only to improve wine quality, but also to maintain their safety. Therefore, the aims of this study were to determine the incidence of Aspergillus section Nigri species harvested in different grape-growing regions from Argentina, their ability to produce OTA, to correlate with meteorological conditions and geographical coordinates with their prevalence and to evaluate the OTA natural occurrence in grapes and wines. The morphological identification showed that Aspergillus niger aggregate species were the most prevalent ones, followed by Aspergillus carbonarius and Aspergillus uniseriate. These populations were confirmed through using AFLP markers and sequencing and, Aspergillus tubingensis was separated from A. niger aggregate. Climatic factors, altitude, longitude and latitude have influenced on the distribution of species included in the section. A. carbonarius and A. niger were OTA producers but differed in their OTA producing ability. Temperature was the factor which influenced the most over the highest incidence of A. carbonarius in La Rioja and San Juan regions. The trellis system in vineyards and drip irrigation also influenced the species isolation. The OTA levels detected in grapes and wines were low, but grape variety was more important in susceptibility to fungal infection and OTA levels.  相似文献

The objective of this study was to characterize the mycoflora of sorghum grains commercialized in the Tunisian retail market and to identify aflatoxins (AFs), ochratoxin A (OTA) and zearalenone (ZEA) producing species. Sixty four samples of sorghum (37 samples of Tunisian sorghum and 27 samples of Egyptian sorghum) were analyzed. Dilution plating (CFU, colony forming units) was used for fungal enumeration. The isolation of mycobiota was carried out by plating of grains on PDA and malachite green medium. Aspergillus section Flavi and section Nigri and Fusarium isolates were sub-cultured in CYA to test their ability to produce AFs, OTA and ZEA, respectively. The selected Aspergillus section Flavi and section Nigri, Penicillium and Fusarium isolates were subjected to specific PCR assays using published species-specific primers. The results revealed the dominance of Fusarium (95.3%), followed by Aspergillus (87.2%) and Alternaria (81.2%) species. The fungal counts ranged from 100 to 1.3·104 CFU/g for Tunisian sorghum and from 100 to 5.7·103 CFU/g for Egyptian sorghum. Among Aspergillus section Flavi isolates identified by molecular biology, Aspergillus flavus was the most dominant (90.1%) while Aspergillus parasiticus represent 9.9% only. About Aspergillus section Nigri, results showed the dominance of Aspergillus niger aggregate species, including Aspergillus niger, Aspergillus tubingensis and other species. Among Fusarium species, Fusarium incarnatum was the most dominant in both Tunisian and Egyptian sorghum. Penicillium citrinum was the dominant Penicillium species in the studied samples. More than 890 isolates belonging to the genus Aspergillus and Fusarium were tested in order to test their capacity to produce AFs, OTA and ZEA. The percentage of mycotoxin producing isolates in Aspergillus section Flavi, A. section Nigri, and Fusarium was 30.0%, 4.6% and 11.1%, respectively.  相似文献

This study aimed to determine the changes in mould and ochratoxin A (OTA) occurrence in sultanas under three different conventional drying conditions. Five different vineyards were chosen, and the three different treatments were applied to these grapes while drying. At the end of the drying process, total mould and black aspergilli (BA) populations in the samples varied from 2.45 to 5.61 log colony-forming units (CFU) g1 and from 0 to 4.92 log CFU g1, respectively. Significant increases (p < 0.05) occurred in mould loads depending on the extending drying period. However, independent of vineyard location, all the samples treated with cold dipping solution showed the lowest fungal loads. These results indicate that dipping solution treatment was the most effective drying method to minimise fungal infection of grapes. The expected results could not be achieved by drying grapes artificially contaminated with ochratoxigenic Aspergillus carbonarius spores. Seventy-one of 96 isolates (73.95%) obtained during drying were Aspergillus spp., and the remaining (n = 25, 26.05%) belonged to other genera, such as Penicillium, Trichoderma and Cladosporium. Grape juice-based agar medium was used to determine the realistic OTA production capacities of the isolated mould strains. The highest OTA production capacities were 809.70 ± 9.19, 87.58 ± 16.89 and 45.44 ± 18.78 ng g–1 in 50% grape juice agar (GJ50), all five of which were from A. niger isolates. OTA was not present in any sample during the drying period; however, OTA was detected in two samples at 0.32 ± 0.15 and 0.52 ± 0.36 µg kg1 after the end of the drying process. The limit of detection (LOD) and limit of quantitation (LOQ) of the method used for detecting OTA in samples were 0.1 and 0.3 µg kg1, respectively.  相似文献

Aspergillus spp. and Penicillium spp. are the main producers of ochratoxin A (OTA), a mycotoxin responsible for fatal human diseases. Some authorities have established a maximum of 2 μg/L of OTA in wine. Although the incidence and occurrence of OTA in grapes and wine is highly related to climate conditions, as has been extensively documented, there is no conclusive information on the effects of cultivation systems on the presence of OTA. This study focuses on determining the effect of the trellis system, planting density and cordon height on plant microclimate and thus on Aspergillus spp. contamination and OTA production in Tannat wines in Southern Uruguay. Two experiments were conducted during the 2010–2011 growing season: (1) a strip split plot design with five replicates and two cordon heights (CH) (0.5 m and 1.0 m above the soil) were compared in two planting densities (PD) (0.8 and 1.5 m between plants); (2) a randomised complete block design, vertical shoot positioning (VSP) versus Lyra trellis systems were evaluated. The results suggest that, even the macro- and micro-climate growing conditions play an important part in Aspergillus developing on grapes. Agronomical practices also have an undoubted impact on the risk and control of OTA accumulation in wine.  相似文献

Aspergillus niger has been recently found to produce fumonisin B2 (FB2). Thirty-one strains belonging to four Aspergillus species isolated from grape were evaluated for FB2 production on agar plates. Four out of eight strains of A. niger produced FB2 (29–293 µg g?1). None of the strains of A. uvarum (n = 7), A. tubingensis (8) and A. carbonarius (8) produced detectable amounts of toxin. The capability to produce FB2 was also confirmed by some A. niger strains artificially inoculated on grape berries. Natural occurrence of FB2, at levels of 0.01 and 0.4 µg ml?1, was found in two samples of must collected in Apulian cellars in 2007. This is the first report of FB2 contamination in must. These findings suggest that there is a potential risk of exposure to FB2 in the grape–wine chain for consumers and that A. niger may represent the major fumonisin-producing species among black Aspergilli occurring on grapes.  相似文献

BACKGROUND: The presence of ochratoxin A (OTA) in grapes and grape derivatives has been reported mainly in the Mediterranean area. Consequently, great efforts are being made to prevent the growth of Aspergillus on grapes. However, the European Commission suggests that climate change may result in increased temperatures and longer drought periods in southern Europe. Therefore the aim of this study was to investigate how ochratoxigenic fungal growth and the efficiency of fungicides used at present might be affected by environmental conditions predicted with climate change. RESULTS: The effectiveness of grape field antifungals (Switch, Flint Max and Equisetum arvense extract) under two alternating temperature, photoperiod and relative humidity (RH) scenarios (current: 20/30 °C, 16 h light/8 h darkness, 80% RH; predicted: 25/37 °C, 16 h light/8 h darkness, 75% RH) on the growth and OTA production of two Aspergillus carbonarius isolates and one Aspergillus ochraceus isolate on grapes was investigated. CONCLUSION: Predicted conditions reduced A. carbonarius and limited A. ochraceus growth. Antifungals reduced fungal infection (by 40‐84%), although no correlation between climatic conditions and effectiveness of the antifungals was found. However, Switch always showed the greatest reduction and E. arvense (0.02 g mL?1 extract) the least. Higher temperatures affected OTA production by the isolates in different ways. Mycoflora, the mycotoxigenic properties of moulds, and natural contamination with mycotoxins such as aflatoxins (AFs), cyclopiazonic acid (CPA), fumonisin B1 (FB1) and ochratoxin A (OTA) were investigated in dried figs. Dry fig samples were collected from orchards during the drying stage in the Aegean Region of Turkey. Fungal isolates were identified using morphological, chemical as well as molecular methods. Mycotoxigenic characteristics of moulds were assessed by thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC). Mycotoxins except CPA (by TLC) were determined by HPLC. All the fig samples were contaminated with moulds and 94.7% contained one or more mycotoxigenic species. The most prevalent moulds present in dried figs belong to the Aspergillus section Nigri members, being 93.9% positive for the samples, followed by Fusarium spp., Aspergillus section Flavi and Penicillium spp. On the other hand, Fusarium spp. had the highest count and the number of fumonisin producing Fusarium was also high. A total of 48% of 115 dried fig samples contained OTA (range?=?0.1–15.3?ng?g?1), 74.7% of the samples had FB1 (range?=?0.05–3.65?mg?kg?1), 10.0% of the samples had aflatoxin (range?=?0.1–763.2?ng?g?1) and 24.3% of the samples were tentatively identified as being contaminated with CPA (range?=?25–187?ng?g?1). Dried fig samples were contaminated with one (33.0%), two (47.0%), three (5.2%) and four mycotoxins (3.5%). A total of 11.3% of dried fig samples were not contaminated with any of the four mycotoxins. To the best of our knowledge, CPA and fumonisin have been found for the first time in dried figs.  相似文献

BACKGROUND: Ochratoxin A (OTA) is the main mycotoxin found in grapes, wines and grape juices and is considered one of the most harmful contaminants to human health. In this study, samples of tropical wines and grape juices from different grape varieties grown in Brazil were analysed for their OTA content by high‐performance liquid chromatography. RESULTS: The detection and quantification limits for OTA were 0.01 and 0.03 µg L?1 respectively. OTA was detected in 13 (38.24%) of the samples analysed, with concentrations ranging from < 0.03 to 0.62 µg L?1. OTA was not detected in any of the grape juice samples. Most of the red wine samples proved to be contaminated with OTA (75%), while only one white wine sample was contaminated. However, the OTA levels detected in all samples were well below the maximum tolerable limit (2 µg L?1) in wine and grape juice established by the European Community and Brazilian legislature. CONCLUSION: The results of this study indicate a low risk of exposure to OTA by consumption of tropical wines and grape juices from Brazil. © 2012 Society of Chemical Industry  相似文献

In the present study, ochratoxigenic mycobiota in cocoa beans was identified at species level by digestion of the ITS products using the endonucleases HhaI, NlaIII and RsaI. Of the 132 isolates of Aspergillus section Nigri collected from cocoa beans, 89 were identified as A. tubingensis, 27 as A. niger, 10 as A. tubingensis-like and 6 as A. carbonarius. No variation was observed between RFLP patterns (C, N, T1 and T2) described previously for grape isolates and those of the cocoa isolates analysed. With respect to OTA-producing fungi, a high percentage of black aspergilli (50.7%) was able to produce OTA. Additionally, most of the OTA-producing isolates were of moderate toxigenicity, producing amounts of OTA from 10 μg g−1 to 100 μg g−1. Percentages of OTA-producing isolates in the A. niger aggregate were higher than in other substrates, ranging from 30% to 51.7%. Furthermore, the detected levels of OTA production in the A. niger aggregate, particularly in A. tubingensis species was higher than in A. carbonarius, ranging from 0.7 μg g−1 to 120 μg g−1 (mean 24.55 μg g−1). Due to the high occurrence, percentage of ocratoxigenic isolates and their ability to produce OTA, isolates belonging to the A. niger aggregate could be considered as the main cause of OTA contamination in cocoa beans used for manufacturing cocoa products.  相似文献

Relatively few data exist regarding concentrations of aflatoxins and their causative organisms in dusts within occupational environments. Here, we examined Aspergillus Section Flavi populations and aflatoxin levels in 54 samples of dusts generated by agricultural processing facilities as possible indicators of aflatoxin exposure in the Philippines. The average incidence of Aspergillus Section Flavi expressed as a percentage of total mould populations in rice dust, corn dust, feed dust and copra dust were 8, 4, 31 and 10%, respectively. Predominant aflatoxigenic fungi isolated were Aspergillus flavus and A. parasiticus with ratios of 31:1, 40:5, 16:4 and 1:1 in rice dust, corn dust, feed dust and copra dust, respectively. Aflatoxins produced by selected isolates in in vitro rice culture ranged from 100 µg kg?1 to 100.5 mg kg?1. Toxigenicity of isolates based on the average aflatoxin concentrations produced by positive isolates were in the order of copra dust > corn dust > rice dust > feed dust. Average natural concentrations of aflatoxins in rice dust, corn dust, feed dust, and copra dust were 25, 6, 15 and 10 µg kg?1, respectively. Estimates of the amount of inhaled aflatoxins by workers in an 8‐h work shift ranged from 0.06 to 114 ng, the average of which is higher than the amount of aflatoxins ingested by Filipinos due to the consumption of polished rice. The presence of highly toxigenic Aspergillus Section Flavi and aflatoxins in agricultural dust is a critical health risk for workers, considering the frequency of exposure and the possibility of inhalation and subsequent absorption of aflatoxins in the respiratory tract. ABSTRACT

Occurrence of aflatoxins (AFs) AFB1, AFB2, AFG1, AFG2 and ochra toxin A (OTA) was studied in 65 samples of stored and freshly harvested wheat, barley and maize collected in Tunisia. The mycotoxins were simultaneously extracted and quantified by high performance liquid chromatography. Determination of AF-producing (section Flavi) and OTA-producing Aspergillus species (sections Nigri and Circumdati) was conducted in these samples by species-specific polymerase chain reaction (PCR). Results showed that most of maize samples were contaminated with AFs, data after storage showing lower values than those collected at harvest. All contaminated maize samples contained AFG1 and AFG2, among which 27.78% also had AFB1 and AFB2. This AFs pattern was consistent with the A. parasiticus toxin profile. A. flavus however showed the highest frequency in maize but was also found in barley and wheat where no AFs were detected. In contrast, OTA was neither found in maize nor in barley and only one wheat sample contained OTA. A. niger was the only OTA-producing species detected.  相似文献

Aspergillus species from section Flavi were isolated from soil samples in three peanut‐growing regions of Córdoba Province, Argentina. The samples were collected during the planting and harvest periods. Both total fungal population and Aspergillus species from section Flavi showed no significant differences between planting and harvest time in two of the regions evaluated. Only in one region were there significant differences in cfu g?1 of total fungal population and Aspergillus species from section Flavi. A flavus was the dominant species isolated in all three localities during the planting and harvest periods. There were significant differences (p < 0.05) in the ratio of toxigenic and atoxigenic strains dependent on the period and the region evaluated. In one region, higher frequencies of toxigenic A flavus and A parasiticus in soil were found and a high contamination level of aflatoxins was detected in peanut seeds. Fungi associated with ochratoxin A (OTA)-contaminated liquorice root and their capabilities for OTA production were investigated. Medicinal materials of mouldy liquorice root were collected from herbal markets located in Jiangxi, Zhejiang, Henan provinces and Beijing, China, respectively. Sixteen fungal species belonging to Penicillium, Aspergillus, Eurotium, Fusarium, Mucor and Scopulariopsis were isolated; the fungal composition was different in each liquorice root sample. Penicillium polonicum was predominant, comprising 54% of the total isolates in the liquorice root sample from Jiangxi province, which was contaminated with OTA at the highest level. In other samples with lower OTA contents, species of Aspergillus and Eurotium were predominant. OTA production of representative strains on rice media was detected by LC-MS/MS; all Penicillium polonicum isolates and a P. chrysogenum were ochratoxigenic; OTA concentrations ranged from 6.94 to 217.37?ng?g?1. This is the first study to report P. polonicum as an OTA-producing fungus. OTA contamination of mouldy liquorice root constitutes a major health hazard in consumption. A survey on the occurrence of ochratoxin A (OTA) in 240 grape‐based beverages was carried out. Red and white wines from four different Spanish Designations of Origin (n = 160), musts (n = 20), grape juices (n = 10), ordinary wines (n = 20), special wines (Malaga, muscatel, sherry, vermouth, etc) (n = 20) and sparkling wines (n = 10) were assayed for OTA content using immunoaffinity column clean‐up and high‐performance liquid chromatography with fluorimetric detection (detection limit 0.05 µg l?1). Forty‐three (17.9%) of the samples tested contained detectable levels of OTA. The overall mean OTA concentration in red and white wines of Designations of Origin was 0.30 and 0.18 µg l?1 respectively (ranges 0.05–3.19 and 0.05–1.13 µg l?1 respectively). The percentage of wine samples with detectable amounts of OTA was higher for red (18.3%) than for white (10%) wines. OTA was also found in two of 10 red ordinary wines (0.68 and 4.24 µg l?1), whereas none of 10 white ordinary wines contained OTA. The mean OTA amount detected in sparkling wines was 0.44 µg l?1 (range 0.14–0.71 µg l?1). Two of 20 must samples contained OTA at low levels (0.08 and 0.18 µg l?1), while none of 10 grape juice samples contained OTA. This study was conducted to assess major Aspergillus species and aflatoxins associated with groundnut seeds and cake in Eastern Ethiopia and evaluate growers’ management practices. A total of 160 groundnut seed samples from farmers’ stores and 50 groundnut cake samples from cafe and restaurants were collected. Fungal isolation was done from groundnut seed samples. Aspergillus flavus was the dominant species followed by Aspergillus parasiticus. Aflatoxin analyses of groundnut seed samples were performed using ultra performance liquid chromatography; 22.5% and 41.3% of samples were positive, with total aflatoxin concentrations of 786 and 3135 ng g?1 from 2013/2014 and 2014/2015 samples, respectively. The level of specific aflatoxin concentration varied between 0.1 and 2526 ng g?1 for B2 and B1, respectively. Among contaminated samples of groundnut cake, 68% exhibited aflatoxin concentration below 20 ng g?1, while as high as 158 ng g?1 aflatoxin B1 was recorded. The study confirms high contamination of groundnut products in East Ethiopia.  相似文献

Grape and wine production in South America represents about 6.6% and 10% respectively of the world grape and wine production. The available information on the ochratoxigenic mycoflora and ochratoxin A (OTA) presence in wine grapes, wines, grape juices and dried vine fruits is limited. Surveys have been carried out in Argentina and Brazil which showed that Aspergillus niger aggregate are predominant in the Argentinean varieties while from the Brazilian varieties the species A. niger, Aspergillus ochraceus and Aspergillus carbonarius were isolated. A mycobiota survey from wine grapes in Argentina showed that while Alternaria alternata was predominant, Aspergillus section Nigri species were isolated from 60% of samples. About 41% of black Aspergilli isolates produced OTA with levels ranging from 2 to 24.5 ng mL(-1). In another study, about 83% of A. carbonarius isolates from dried vine fruits produced OTA, with levels ranging from 2 to 5200 ng mL(-1). A survey of grape juices and wines of Brazilian, Argentinean and Chilean origin were found to contain very low levels of OTA. Studies are in progress in Latin America on the ecophysiology of ochratoxigenic fungi and OTA occurrence to reduce the impact of this toxin in the food chain.  相似文献

Ochratoxin A (OTA) is a mycotoxin mainly produced by mould species of the genera Aspergillus and Penicillium, which grow on a variety of agricultural products. OTA-contaminated foodstuffs pose a major health hazard to consumers, including human and animal. In Côte d’Ivoire, numerous studies are being carried out to find the best way of preventing OTA contamination of cocoa raw material. The objectives of this investigation were to assess the impact of industrial treatment on OTA content in cocoa-derived products. Samples of cocoa pods were prepared under specific conditions promoting fungal proliferation on cocoa beans before processing. The beans underwent the usual industrial treatments – roasting, shelling, crushing, pressing and additive addition – and samples were taken at each stage. OTA was extracted with a methanol/3% sodium hydrogen carbonate solution and purified using an immunoaffinity column prior to HPLC analysis with fluorescence detection. OTA was detected in artificially contaminated cocoa beans at levels ranging from 3.4 to 44.7 µg kg?1 with a mean value of 22.9 ± 3.6 µg kg?1. OTA was mainly concentrated in the shell (93%). Roasting, shelling and additive addition significantly decreased levels of OTA by 24–40, 76 and 52%, respectively, with an overall reduction of ~91%.
