利用BACTEC-960全自动分枝杆菌培养鉴定药敏仪建立动物试验模型,以观察结核分枝杆菌(M.tuberculosis,MT)MPT64质粒DNA疫苗预防结核病的效力,第一组小鼠接受H37RV攻击一个月后的肺,肝,脾培养结果:90%阳性标本培养时间为7-13天,肝脏仅75%出现阳性结果。第二组小鼠接受H37RV攻击两个月后,其肺、肝、脾培养结果:90%阳性标本培养时间为10-15天,肝脏仅60%出现阳性结果。第二组比第一组生长天数总体向后推迟,生长时间明显集中,生长曲峰强度比第一组明显减弱,第二组在肝脏中的感梁率比第一组有所下降。可能是因为小鼠轻DNA疫苗刺激后,免疫力增强,使结核分枝杆菌生产缓慢之故。 相似文献

以电液伺服三轴仪伺服轴向力的伺服系统为研究对象,采用定量反馈理论(QFT)设计的鲁棒控制器进行控制其设计,并利用AMESim软件建立液压系统的动态数学模型,代替传统的传递函数所建立模型的进行联合仿真。研究表明,QFT控制器可以有效抑制参数扰动,如弹性负载刚度变化、油液有效体积弹性模数变化,系统摩擦力变化。工程上易于实现,可以有效地改善伺服三轴仪的控制性能。 相似文献

Recent progress in single DNA manipulation with atomic force microscope has proved its potential in analysing genetic information at the molecular level instead of macro ensemble approach. However, current manipulation of DNA is mainly carried out by manual operations, which is labour-intensive and time-consuming, thus limits the further applications in other fields. In this paper, an optimized DNA manipulation method is successfully established. An image correlation technique is introduced to realize automatic thermal drift compensation. Combined with elaborately designed tip movement control for different manipulation purposes, accurate and efficient DNA manipulations such as dissection, folding and picking are realized. The efficiency has been improved for an order of magnitude compare to manual manipulations, whereas the performance is demonstrated to be the same. This newly developed method has shed light on high-efficiency nanomanipulation of small molecules with complex structures, and thus provides the possibility of deeply understanding the intrinsic properties of single biomolecules. 相似文献

Cellular DNA crosslinks are a type of DNA damage induced by toxic chemicals or high‐energy radiation. If damaged DNA is not rapidly repaired, cells will die or mutate. To evaluate the types of DNA damage and their influence on vital cell activities, it is necessary to be able to detect DNA crosslinks. To date, indirect methods such as alkaline elution, potassium chloride–sodium dodecyl sulfate assay and comet assay have been used to detect DNA damage. Direct morphological observation, on the other hand, may be a useful tool to differentiate the types of DNA damage. In this report, atomic force microscopy (AFM) has been employed to visualize the breakage and crosslinking of cellular DNA strands in cells treated with formaldehyde and hydrogen peroxide. Our results showed that toxic chemical‐induced crosslinking of cellular DNA occurred in a dose‐dependent manner. DNA conglomerates were observed with high concentrations of formaldehyde, and the AFM observations were consistent with those of a comet assay. A DNA computing algorithm (DNACA) with an electron–ion interaction potential (EIIP) decoding scheme is proposed to identify a class of transfer functions. The DNACA includes enzyme and virus operators which provide a highly modular, flexible, and accurate self-organizing structure environment. 为改进结核分支杆菌药敏试验方法,用BACTEC-960液体培养分离株300例,根据模板将菌液稀释至10-6CFU/mL,分别接种于14种不同药物改良7H10药敏培养基和改良罗氏药敏培养基上,观察耐药情况.改良7H10培养基比改良罗氏早12~15天报告结果.在两种培养基中的14种,耐药结果无显著性差异(P>0.05).说明BACTEC技术与改良7H10药敏培养基方法结合,弥补BACTEC只做四种药敏的不足,扩展药敏试验范围,并比传统改良罗氏培养和药敏提前30天报告结果,更利于实际应用. 相似文献
