The occurrence of eight pharmaceuticals (beta-blockers: acebutolol, atenolol, metoprolol and sotalol; antiepileptic: carbamazepine; fluoroquinolone antibiotics: ciprofloxacin, norfloxacin, ofloxacin) were assessed in the raw and treated sewage of 12 sewage treatment plants (STPs) in Finland. The average concentrations in the raw and treated sewage ranged from 100 to 1060 ng L(-1) and from <24 to 755 ng L(-1), respectively. The average daily loads ranged from 36 to 405 mg/1000 inh and from 2 to 302 mg/1000 inh, respectively. In the treatment plants, fluoroquinolones were eliminated by >80%. Carbamazepine was not eliminated during the treatment and in fact even higher concentrations were frequently found in the treated than in the raw sewages. The increase in concentration was shown to be most likely due to enzymatic cleavage of the glucuronic conjugate of carbamazepine and release of the parent compound in the treatment plant. The beta-blockers were eliminated in average by less than 65% and the elimination varied greatly between the treatment plants. Especially the dilution of raw sewage by rainwater and a consequent decrease in the hydraulic retention time of a treatment plant was found to deteriorate the elimination of the beta-blockers. The work shows that especially carbamazepine and the beta-blockers may reach the recipient waters and there is a need to enhance their elimination in the sewage treatment plants. In this attempt, a denitrifying biofilter as a tertiary treatment could be of minor importance since in this study it did not result in further elimination of the target compounds. 相似文献

Sorption of 75 active pharmaceutical ingredients (APIs) to three different types of sludge (primary sludge, secondary sludge with short and long sludge age respectively) were investigated. To obtain the sorption isotherms batch studies with the APIs mixture were performed in four nominal concentrations to water containing 1 g of sludge. The range of APIs concentrations was between ng L−1 to μg L−1 which are found in the wastewater effluents. Isotherms were obtained for approximately 45 of the APIs, providing distribution coefficients for linear (Kd), Freundlich (Kf) and Langmuir (KL) isotherms. Kd, Kf and KL ranging between 7.1 × 104 and 3.8 × 107, 1.1 × 10−2 and 6.1 × 104 and 9.2 × 10−3 and 1.1 L kg−1, respectively. The obtained coefficients were applied to estimate the fraction of APIs in the water phase (see Abstract Graphic). For 37 of the 75 APIs, the predicted presence in the liquid phase was estimated to >80%. 24 APIs were estimated to be present in the liquid phase between 20 and 80%, and 14 APIs were found to have <20% presence in the liquid phase, i.e. high affinity towards sludge. Furthermore, the effect of pH at values 6, 7 and 8 was evaluated using one way ANOVA-test. A significant difference in Kds due to pH changes were found for 6 of the APIs (variation 10-20%). 相似文献

The degradation of the beta-blockers atenolol, metoprolol tartrate and propranolol hydrochloride was studied by electro-Fenton (EF) and solar photoelectro-Fenton (SPEF). Solutions of 10 L of 100 mg L−1 of total organic carbon of each drug in 0.1 M Na2SO4 with 0.5 mM Fe2+ of pH 3.0 were treated in a recirculation flow plant with an electrochemical reactor coupled with a solar compound parabolic collector. Single Pt/carbon felt (CF) and boron-doped diamond (BDD)/air-diffusion electrode (ADE) cells and combined Pt/ADE-Pt/CF and BDD/ADE-Pt/CF cells were used. SPEF treatments were more potent with the latter cell, yielding 95-97% mineralization with 100% of maximum current efficiency and energy consumptions of about 0.250 kWh g TOC−1. However, the Pt/ADE-Pt/CF cell gave much lower energy consumptions of about 0.080 kWh g TOC−1 with slightly lower mineralization of 88-93%, then being more useful for its possible application at industrial level. The EF method led to a poorer mineralization and was more potent using the combined cells by the additional production of hydroxyl radicals (•OH) from Fenton’s reaction from the fast Fe2+ regeneration at the CF cathode. Organics were also more rapidly destroyed at BDD than at Pt anode. The decay kinetics of beta-blockers always followed a pseudo first-order reaction, although in SPEF, it was accelerated by the additional production of •OH from the action of UV light of solar irradiation. Aromatic intermediates were also destroyed by hydroxyl radicals. Ultimate carboxylic acids like oxalic and oxamic remained in the treated solutions by EF, but their Fe(III) complexes were photolyzed by solar irradiation in SPEF, thus explaining its higher oxidation power. NO3− was the predominant inorganic ion lost in EF, whereas the SPEF process favored the production of NH4+ ion and volatile N-derivatives. 相似文献

The presence of nitrosamines in wastewater might pose a risk to water resources even in countries where chlorination or chloramination are hardly used for water disinfection. We studied the variation of concentrations and removal efficiencies of eight N-nitrosamines among 21 full-scale sewage treatment plants (STPs) in Switzerland and temporal variations at one of these plants. N-nitrosodimethylamine (NDMA) was the predominant compound in STP primary effluents with median concentrations in the range of 5-20 ng/L, but peak concentrations up to 1 μg/L. N-nitrosomorpholine (NMOR) was abundant in all plants at concentrations of 5-30 ng/L, other nitrosamines occurred at a lower number of plants at similar levels. From concentrations in urine samples and domestic wastewater we estimated that human excretion accounted for levels of <5 ng/L of NDMA and <1 ng/L of the other nitrosamines in municipal wastewater, additional domestic sources for <5 ng/L of NMOR. Levels above this domestic background are probably caused by industrial or commercial discharges, which results in highly variable concentrations in sewage. Aqueous removal efficiencies in activated sludge treatment were in general above 40% for NMOR and above 60% for the other nitrosamines, but could be lower if concentrations were below 8-15 ng/L in primary effluent. We hypothesize that substrate competition in the cometabolic degradation explains the occurrence of such threshold concentrations. An additional sand filtration step resulted in a further removal of nitrosamines from secondary effluents even at low concentrations. Concentrations released to surface waters were largely below 10 ng/L, suggesting a low impact on Swiss water resources and drinking water generation considering the generally high environmental dilution and possible degradation. However, local impacts in case a larger fraction of wastewater is present cannot be ruled out. 相似文献

We investigated the survival of Escherichia coli in two STPs utilising UV irradiation (STP-A) or chlorination (STP-B) for disinfection. In all, 370 E. coli strains isolated from raw influent sewage (IS), secondary treated effluent (STE) and effluent after the disinfection processes of both STPs were typed using a high resolution biochemical fingerprinting method and were grouped into common (C-) and single (S-) biochemical phenotypes (BPTs). In STP-A, 83 BPTs comprising 123 isolates were found in IS and STE, of which 7 BPTs survived UV irradiation. Isolates tested from the same sites of STP-B (n = 220) comprised 122 BPTs, however, only two BPTs were found post-chlorination. A representative isolate from each BPT from both STPs was tested for the presence of 11 virulence genes (VGs) associated with uropathogenic (UPEC) or intestinal pathogenic (IPEC) E. coli strains. Strains surviving UV irradiation were distributed among seven phylogenetic groups with five BPTs carrying VGs associated with either UPEC (4 BPTs) or IPEC (1 BPT). In contrast, E. coli strains found in STP-B carried no VGs. Strains from both STPs were resistant to up to 12 out of the 21 antibiotics tested but there was no significant difference between the numbers of antibiotics to which surviving strains were resistant to in these STPs. Our data suggests that some E. coli strains have a better ability to survive STPs utilising chlorination and UV irradiation for disinfection. However, strains that survive UV irradiation are more diverse and may carry more VGs than those surviving SPTs using chlorination. 相似文献

The occurrence and removal of salmonellae and faecal indicators in four conventional municipal wastewater treatment plants (MWTP) were investigated. In addition, we tested the efficiency of a semi-technical scale biological nutrient removal process and three pilot-scale tertiary filtration units in microbial removal. All influent samples collected from MWTPs contained salmonellae from 93 to 11,000 MPN/100 ml and indicator bacteria from about 10(7) to 10(8) CFU/100 ml. The reductions in salmonella numbers achieved in full-scale biological-chemical wastewater treatment and semi-technical scale biological nutrient removal processes were usually between 94% and virtually 100% (99.9%) and indicator bacteria reductions between 2 and 3 log units. Microbial numbers in MWTP effluents could be modelled as a function of effluent residual organic matter, suspended solids and total phosphorus concentrations. Pilot-scale tertiary treatment by rapid sand contact filter, chemical contact filter and biological-chemical contact filter reduced salmonella numbers below the detection limit and faecal coliform numbers on average by 99%, 39% and 71%, respectively. A total of 32 Salmonella serovars were identified among 197 Salmonella isolates from municipal wastewaters. Of the isolates, 32% were resistant to nalidixic acid, indicating reduced sensitivity to ciprofloxacin, the drug of choice in the treatment of salmonellosis. In addition, 18% of the isolates were multiresistant. Our results, especially antibiotic resistant Salmonella strains, indicate that conventional municipal wastewater treatment without efficient tertiary treatment, like filtration or disinfection, may constitute a risk for public health. 相似文献

The occurrence and fate of five acidic drugs (Mefenamic acid, Ibuprofen, Ketoprofen, Diclofenac and Clofibric acid) were analysed in three sewage treatment plants (STP) over 4-7 consecutive days. The results point out that the five substances were persistent in wastewater effluents after municipal wastewater treatment. At the most, half of Mefenamic acid was eliminated. Ibuprofen was well removed (80%) by one sewage treatment plant. The removal of Ibuprofen is dependent on the residence time of wastewater in the STPs. A long raining period induce an important decrease of removal of Ibuprofen and Ketoprofen. Removal rates showed a great variability according to sewage treatment plants and types of treatments (e.g. biological, physico-chemical). The concentrations of Ibuprofen, Mefenamic acid and Diclofenac were relatively high in the effluents (150-2000 ng/l), showing a potential contamination of surface water. An environmental risk assessment is presented. Mefenamic acid seems to present a risk for the aquatic environment, with a ratio PEC/PNEC higher than one. 相似文献

In a German sewage treatment plant that processes 200000 m(3) wastewater per day, the concentrations of 2,4,4'-trichloro, 2'-hydroxy-phenylether (triclosan) in the in-flowing ( approximately 1000 ng x l(-1)) as well as in the out-flowing water ( approximately 50 ng x l(-1)) are compared to the concentrations measured in sludge (1200 ng x g(-1)). Considering the mass flow of water and sludge in the respective plant, balances including water and sludge are calculated. Thirty percent of the triclosan is sorbed with weak bonds to the sludge, while some amounts are sorbed as bound residues in the sludge. About 5% is dissolved in the out-flowing water. Thus most of the in-flowing material is not recovered as the parent compound but it is likely that it is transformed to other metabolites or unrecovered bound residues. These data are compared to the monitoring of sewage sludge of 20 different plants in this region, most of which are smaller, though. The concentrations of airborne culturable microorganisms were determined in wastewater and sludge treatment processes of seven sewage treatment plants. Two types of coliphages, Salmonella and total viable bacteria were sampled by the BioSampler and the numbers of faecal coliforms and enterococci were obtained from the Andersen 6-stage impactor. The BioSampler recovered higher numbers of airborne coliphage viruses than has been measured with other liquid samplers in previous studies, suggesting that this sampler has improved efficiency for sampling airborne coliphages. Airborne coliphages were detected in many stages of the wastewater or sludge treatment process. The highest microbiological air contaminations were found in pre-treatment and aerated grit separation stages of the operation. This was attributed to aerosolisation of microorganisms by mechanical handling or forced aeration. Aeration and settling processes located outdoors caused low microbial concentrations, but the brush aerator released more microorganisms into the air. Our results emphasize the necessity for controlling the exposure of sewage workers to airborne microorganisms, especially in process areas that involve mechanical agitation or forced aeration of wastewater. 相似文献

This study surveyed the presence of bacterial pathogens in eight Swedish sewage treatment plants (STPs), with four different treatment methods, focusing on detection of zoonotic bacteria in raw and treated sludge. Salmonella spp., Listeria monocytogenes, Campylobacter coli and jejuni, Escherichia coli O157 and indicator bacteria were investigated. Samplings were performed from July 2000 to June 2002, resulting in 64 raw sludge samples and 69 treated sludge samples. The samples from raw sludge (67%) and treated sludge (55%) were positive for Salmonella; 49 different serotypes were detected. Restriction enzyme analysis and pulsed field gel electrophoresis of Salmonella serotypes indicated that Salmonella persists in STPs and that there is a continuous supply of new strains. There are differences in treatment methods concerning the reduction of pathogens and indicator bacteria. If spread on arable land, sludge increases the environmental load of pathogens; this increases the risk for spreading diseases to people and animals. 相似文献

During 8 sampling campaigns carried out over a period of two years, 72 samples, including influent and effluent wastewater, and sludge samples from three conventional wastewater treatment plants (WWTPs), were analyzed to assess the occurrence and fate of 43 pharmaceutical compounds. The selected pharmaceuticals belong to different therapeutic classes, i.e. non-steroidal anti-inflammatory drugs, lipid modifying agents (fibrates and statins), psychiatric drugs (benzodiazepine derivative drugs and antiepileptics), histamine H2-receptor antagonists, antibacterials for systemic use, beta blocking agents, beta-agonists, diuretics, angiotensin converting enzyme (ACE) inhibitors and anti-diabetics. The obtained results showed the presence of 32 target compounds in wastewater influent and 29 in effluent, in concentrations ranging from low ng/L to a few μg/L (e.g. NSAIDs). The analysis of sludge samples showed that 21 pharmaceuticals accumulated in sewage sludge from all three WWTPs in concentrations up to 100 ng/g. This indicates that even good removal rates obtained in aqueous phase (i.e. comparison of influent and effluent wastewater concentrations) do not imply degradation to the same extent. For this reason, the overall removal was estimated as a sum of all the losses of a parent compound produces by different mechanisms of chemical and physical transformation, biodegradation and sorption to solid matter. The target compounds showed very different removal rates and no logical pattern in behaviour even if they belong to the same therapeutic groups. What is clear is that the elimination of most of the substances is incomplete and improvements of the wastewater treatment and subsequent treatments of the produced sludge are required to prevent the introduction of these micro-pollutants in the environment. 相似文献

Occurrence and fate of quinolone and fluoroquinolone antibiotics in a municipal sewage treatment plant 总被引:8,自引:0,他引:8
This study developed a method for analysis of nineteen quinolone and fluoroquinolone antibiotics (FQs) in sludge samples, and investigated the occurrence and fate of the FQs in a municipal sewage treatment plant (STP) with anaerobic, anoxic, and aerobic treatment processes. Eleven compounds, including pipemidic acid, fleroxacin, ofloxacin, norfloxacin, ciprofloxacin, enrofloxacin, lomefloxacin, sparfloxacin, gatifloxacin, moxifloxacin, and sarafloxacin (only in sludge), were detected in the STP. The predominance of ofloxacin and norfloxacin, followed by lomefloxacin, ciprofloxacin, gatifloxacin, and moxifloxacin, were found in wastewater, suspended solids, and sludge. The total concentrations of FQs were 2573 ± 241 ng/L, 1013 ± 218 ng/L, and 18.4 ± 0.9 mg/kg in raw sewage, secondary effluent, and sludge, respectively. Extremely low mass change percentages were observed for FQs in anaerobic, anoxic, and aerobic treatment units, suggesting biodegradation to be of minor importance in the removal of FQs in STPs. 50-87% of the initial FQs loadings (except for pipemidic acid (36%)) were ultimately found in the dewatered sludge. Mean removal efficiencies of FQs in the STP were 56-75%, except for new generation drugs such as moxifloxacin (40 ± 5%) and gatifloxacin (43 ± 13%). A significant positive correlation was found between removal efficiencies and Kd of FQs. The major factor in the removal of FQs in the STP was sorption to sludge, which was not governed by hydrophobic interactions. The long-term cycling and persistence of FQs in the STP has made activated sludge as a huge reservoir of FQ antibiotics. 相似文献

In this paper we report on the performances of full-scale conventional activated sludge (CAS) treatment and two pilot-scale membrane bioreactors (MBRs) in eliminating various pharmaceutically active compounds (PhACs) belonging to different therapeutic groups and with diverse physico-chemical properties. Both aqueous and solid phases were analysed for the presence of 31 pharmaceuticals included in the analytical method. The most ubiquitous contaminants in the sewage water were analgesics and anti-inflammatory drugs ibuprofen (14.6-31.3 μg/L) and acetaminophen (7.1-11.4 μg/L), antibiotic ofloxacin (0.89-31.7 μg/L), lipid regulators gemfibrozil (2.0-5.9 μg/L) and bezafibrate (1.9-29.8 μg/L), β-blocker atenolol (0.84-2.8 μg/L), hypoglycaemic agent glibenclamide (0.12-15.9 μg/L) and a diuretic hydrochlorothiazide (2.3-4.8 μg/L). Also, several pharmaceuticals such as ibuprofen, ketoprofen, diclofenac, ofloxacin and azithromycin were detected in sewage sludge at concentrations up to 741.1, 336.3, 380.7, 454.7 and 299.6 ng/g dry weight. Two pilot-scale MBRs exhibited enhanced elimination of several pharmaceutical residues poorly removed by the CAS treatment (e.g., mefenamic acid, indomethacin, diclofenac, propyphenazone, pravastatin, gemfibrozil), whereas in some cases more stable operation of one of the MBR reactors at prolonged SRT proved to be detrimental for the elimination of some compounds (e.g., β-blockers, ranitidine, famotidine, erythromycin). Moreover, the anti-epileptic drug carbamazepine and diuretic hydrochlorothiazide by-passed all three treatments investigated.Furthermore, sorption to sewage sludge in the MBRs as well as in the entire treatment line of a full-scale WWTP is discussed for the encountered analytes. Among the pharmaceuticals encountered in sewage sludge, sorption to sludge could be a relevant removal pathway only for several compounds (i.e., mefenamic acid, propranolol, and loratidine). Especially in the case of loratidine the experimentally determined sorption coefficients (Kds) were in the range 2214-3321 L/kg (mean). The results obtained for the solid phase indicated that MBR wastewater treatment yielding higher biodegradation rate could reduce the load of pollutants in the sludge. Also, the overall output load in the aqueous and solid phase of the investigated WWTP was calculated, indicating that none of the residual pharmaceuticals initially detected in the sewage sludge were degraded during the anaerobic digestion. Out of the 26 pharmaceutical residues passing through the WWTP, 20 were ultimately detected in the treated sludge that is further applied on farmland. 相似文献

In order to better understand the mechanisms of N(2)O emissions from nitrifying activated sludge of urban WWTPs, sludge from the Valenton plant (Paris conurbation) are subjected to lab-scale batch experiments under various conditions of oxygenation. The results show that the highest N(2)O emissions (7.1 microgN-N(2)OgSS(-1) h(-1) in average) occur at a dissolved oxygen (DO) concentration of around 1mgO(2)L(-1). These high emissions at low oxygenation (from 0.1 to 2 mg O(2)L(-1)) are due to two processes: autotrophic nitrifier denitrification and heterotrophic denitrification. Nitrifier denitrification always dominates, representing from 58% to 83% of the N(2)O production. This N(2)O production originating from nitrifying activated sludge becomes 8 times higher when nitrite is added at a DO of 1 mg O(2)L(-1); a decrease is observed both at higher and lower oxygenation. Heterotrophic denitrification represents less than 50% of the N(2)O production, decreasing from 42% to 17% when oxygenation increases from 0.1 to 2 mg O(2) L(-1). We show that ammonium oxidizing bacteria (AOB) can shift to nitrifier denitrification when oxygen is depleted in the environments including in the WWTPs, nitrite then plays the role of oxygen as the final electron acceptor. As opposed to what happens in nitrification, the end products of nitrifier denitrification are gaseous forms of nitrogen, where N(2)O is not negligible compared to N(2). Overall, N(2)O emissions represent 0.1-0.4% of oxidized NH(4)(+), depending on the oxygenation level. N(2)O emissions would range from 0.11 to 0.42 TN-N(2)O day(-1) for a tertiary treatment of the Paris wastewater effluents, consisting exclusively of activated sludge nitrification. 相似文献

Fate of nonylphenol polyethoxylates and their metabolites in four Beijing wastewater treatment plants 总被引:6,自引:0,他引:6
Four Beijing wastewater treatment plants (WWTPs) were selected to investigate behaviours of nonylphenol polyethoxylates and their metabolites in different wastewater treatment processes. The results showed that the total concentrations of nonylphenolic compounds in the influents of the four WWTPs ranged from 0.115 to 0.347 μmol/L, as well as their removal efficiencies ranging from 75.7% to 90.8%. Both influent concentrations and removal efficiencies of nonylphenol polyethoxylates were correlated to seasons as follows: higher in the summer than in the winter, and influent concentrations were lower during the rain weather. The analysis revealed that 21.8-47.6% of nonylphenol polyethoxylates and their metabolites entering WWTPs were released via effluents and excess sludge, leaving a great part of them for biodegradation. Nonylphenol and short-chain nonylphenol polyethoxylates were disposed to the environment mainly via sewage sludge, while carboxylated nonylphenol polyethoxylates were the most abundant group of nonylphenol polyethoxylates in effluents. 相似文献

The concentrations of heavy metals in the leaves of two aquatic plants Potamogeton pectinatus L. and Potamogeton malaianus Miq., and the corresponding water and sediment samples from the Donghe River in Jishou City of Hunan Province, China were studied to investigate metal contamination from the intensive industrial activities in the surrounding area. Results showed that the concentrations of heavy metals in the sediments, especially Cd, Mn and Pb, were much higher than the eco-toxic threshold values developed by the U. S. Environmental Protection Agency. Between the two plant species, P. pectinatus showed the higher capacity in metal accumulation. The highest concentrations of Cd, Pb, Cu, Zn and Mn were found in the leaves of P. pectinatus, reaching 596, 318, 62.4, 6590 and 16,000 mg kg(-1) (DW), respectively. Significantly positive relationships were observed among the concentrations of Zn, Cu and Mn in the leaves of both aquatic plants and those in water, indicating the potential use of the two plants for pollution monitoring of these metals. In addition, a laboratory experiment was conducted to investigate the ability of P. pectinatus and P. malaianus to remove heavy metals from contaminated river water. The average removal efficiencies by P. pectinatus and P. malaianus for Cd, Pb, Mn, Zn and Cu from the spiked Donghe River water were 92%, 79%, 86%, 67% and 70%, respectively. The results indicated that P. pectinatus and P. malaianus had high capabilities to remove heavy metals directly from the contaminated water. The potential use of these plants in wastewater treatment is worth further exploration. 相似文献

Effect of the aeration system on the levels of airborne microorganisms generated at wastewater treatment plants 总被引:2,自引:0,他引:2
Six different wastewater treatment plants were monitored in order to identify the main bioaerosol sources and to evaluate the effect of the aeration system used in the biological treatment (air diffusion, horizontal rotors and surface turbine aerators) on the airborne microorganism levels to which workers may be exposed. Air samples were collected by using a single stage impactor. Total count of mesophilic bacteria was used as the monitoring parameter to compare the impact of the aeration system on generic bacterial bioaerosols rather than a quantitative estimation for pathogens or fecal indicator microbes. In this study, pre-treatment, biological treatment and sludge thickening were the processes that generated the highest amount of bioaerosols. Aeration systems involving mechanical agitation of the wastewater, such as horizontal rotors and surface turbines, generated a larger amount of bioaerosols (between 450 and 4580CFU/m(3)) than air diffuser aerators (between 22 and 57CFU/m(3)). The levels of airborne bacteria generated by air diffusers were very similar to those registered at the background locations (lower than 50CFU/m(3)), unaffected by the activities taking place in the wastewater treatment plant. The use of air diffusers as an aeration system for the biological treatment would significantly minimise the potential biological hazard that wastewater treatment plant workers may be exposed to. 相似文献

Antimicrobial resistance of enterococci was investigated in 42 samples of crude inflow, treated effluent and sludge collected in 14 municipal sewage treatment plants of Portugal. A total of 983 enterococci were recovered and tested, using the diffusion agar method, regarding their sensitivity to 10 different antimicrobial drugs. Multidrug resistance was present in 49.4% of the isolates. Only 3.3% and 0.6% of the investigated strains were resistant to ampicillin and vancomycin, respectively. Resistances found against rifampicin (51.5%), tetracycline (34.6%), erythromycin (24.8%) and nitrofurantoin (22.5%), are causes for substantial concern. Almost 14% of isolates were resistant to ciprofloxacin. Wastewater treatment resulted in enterococci decrease between 0.5 and 4log; nevertheless, more than 4.4 x 10(5)CFU/100ml were present in the outflow of the plants. Our data indicate that the use of antimicrobials had created a large pool of resistance genes and that sewage treatment processes are unable to avoid the dissemination of resistant enterococci into the environment. 相似文献

Listeria monocytogenes detection in wastewater can be difficult because of the large amount of background microbiota and the presence of viable but non-culturable forms in this environment. The aim of this study was to evaluate a Fluorescent In Situ Hybridization (FISH) assay combined with Direct Viable Count (DVC) method for detecting viable L. monocytogenes in wastewater samples, as an alternative to conventional culture methods. 16S rRNA sequence data were used to design a specific oligonucleotide probe. In order to assess the suitability of the method, the assays were performed on naturally (n = 87) and artificially (n = 14) contaminated samples and results were compared to those obtained with the isolation of cells on selective media and with a PCR method. The detection limit of FISH and PCR assays was 104 cells/mL without enrichment and 10 cells/mL after enrichment. A total of 47 samples, including 3 samples from effluent sites, yielded FISH positive results for L. monocytogenes. Using DVC-FISH technique, the presence of viable L. monocytogenes cells was detected in 23 out of these 47 FISH positive wastewater samples. PCR and culture methods yielded 27 and 23 positive results, respectively. According to these results, FISH technique has the potential to be used as a sensitive method for the detection and enumeration of L. monocytogenes in environmental wastewater samples. 相似文献
