海洋胶原肽预防SAMP8小鼠学习记忆功能下降的实验研究   总被引:1,自引:0,他引:1  
为探讨海洋胶原肽(MCP)对痴呆性学习记忆下降的影响及作用机制,选用6月龄SAMP8小鼠为模型,分别喂饲含0、0.225%、0.450%和1.350%MCP的特殊加工饲料4个月,之后进行Morris水迷宫检测以观察小鼠的空间学习记忆能力、Nissl染色观察海马神经元的状况,同时检测海马区脑源性神经营养因子(BDNF)的表达状况。结果表明,0.44%和1.32%MCP干预可明显预防SAMP8小鼠学习记忆功能的下降;防止神经元过度丢失,促进脑源性神经营养因子的表达可能是其重要的作用机制。结果显示,海洋胶原肽具有抑制学习记忆下降的功能,可成为抗衰老相关功能食品的新选择。  相似文献

为探讨海洋胶原肽(MCP)对自然衰老过程中自发肿瘤的影响,选用4周龄初断乳SD大鼠,雌雄各半,随机分为四个组,分别在空白标准饲料的基础上添加0%、2.25%、4.5%和9%MCP进行终生喂养,并对各组自发肿瘤的发生情况和生存时间进行观察。结果表明,MCP干预组雌雄SD大鼠自发肿瘤率有一定程度的下降,其中雌性9%干预组、雄性4.5%和9%干预组与对照组的差异具有显著性。各组乳腺肿瘤潜伏期及生长时间没有显著的组间差异,而2.25%与4.5%MCP雌性干预组乳腺肿瘤的体积与对照组相比显著减小;雌雄9%MCP干预组荷瘤大鼠的生存时间较对照组有显著延长;MCP干预组大鼠以肿瘤作为主要死因的比例与对照组相比呈明显下降的趋势。结果提示,MCP长期喂养对SD大鼠的自发肿瘤的发生及发展具有一定的抑制作用。  相似文献

The aim of this study was to evaluate the effect of a diet rich in yam tuber material [Dioscorea alata L. Var. purpurea (M.) Pouch.] on the learning and memory ability of 2‐month‐old male senescence accelerated mice (SAMP8). The mice were fed with three different diets for 12 weeks; a casein diet (control group) and either a casein diet supplemented with 10% lyophilized or with hot‐air dried yam. Results of passive and active shuttle avoidance tests showed the mice fed with the diet containing yam had significantly better learning and memory ability than the control group. The thiobarbituric acid‐reactive substances (TBARS) in the hippocampus of the group fed lyophilized yam were significantly lower than the control and also the group fed a diet containing hot‐air dried yam, whereas the spongy degeneration and the lipofuscin percentage tended to be lower but not significantly different. It was suggested that lyophilized yam was more effective than hot‐air dried yam in reducing the lipid peroxidation, brain pathological changes and the deterioration in the learning and memory ability in mice possibly because lyophilized yam contains more antioxidant compounds.  相似文献

The purpose of this investigation was to evaluate the effects of diets enriched with cactus pear oil (CPO) or seeds (CPS) on serum and liver lipid parameters compared to those of adult rats submitted to a standard diet. Male rats were divided into three groups, the first group represented control group, fed with standard diet, the second group was fed with control diet supplemented with CPO (2.5%, wt/wt) and the third group fed control diet supplemented with CPS (33%, wt/wt), for nine weeks. Feed intake and body weight of rats were measured every two days. Organ weights were determined at the end of treatment; cholesterol, HDL and triglycerides levels were determined by enzymatic methods. Liver and serum lipid extracts were analysed for their fatty acid composition for the three groups of rats. No differences in pancreas, kidney or liver weights were observed in the CPS diet whereas the CPO diet induced a significant increase in liver and pancreas weights. The tested diets significantly decreased the atherogenic index compared to the control diet, whereas serum cholesterol level was only reduced by the supplementation with CPO diet. No variations in serum lipids were observed among the groups, whereas liver lipids showed slight variations. Accordingly, these results indicated that the supplementation with CPO or CPS could be effective in decreasing the atherogenic risk factors in rats.  相似文献

为了检验巴马长寿特征饮食模式的抗衰老效果,本实验采用3 月龄年轻小鼠和15 月龄自然衰老小鼠为研究对象,依据团队前期研究结果,选取巴马长寿区饮食的特征营养素(膳食纤维、VA、VC、VE、β-胡萝卜素、大豆异黄酮、番茄红素、铁、锰、钴、硒元素)为调控因子,并加入益生菌,进一步优化出两种巴马长寿特征饮食模式(优食I和优食II)。以巴马长寿特征饮食模式喂养小鼠,连续8 周。采用Morris水迷宫实验检验小鼠的学习记忆能力,并对脑组织超氧化物歧化酶活力、谷胱甘肽过氧化物酶活力与丙二醛含量进行测定,最后采用苏木精-伊红染色观察脑组织海马区特征。结果表明,与衰老对照组相比,衰老优食I、II组逃避潜伏期显著缩短(P<0.05),衰老优食II组雄鼠目标象限停留时间显著延长(P<0.05);与年轻对照组相比,年轻优食I组雌鼠穿越平台次数显著提高(P<0.05),年轻优食I、II组雄鼠目标象限路程显著延长(P<0.05)。与对照组相比,年轻优食I组与衰老优食I、II组脑组织超氧化物歧化酶活力与谷胱甘肽过氧化物酶活力均显著提高(P<0.05);衰老优食I、II组丙二醛含量均显著降低(P<0.05)。与衰老对照组相比,衰老优食I、II组小鼠海马区神经元细胞变性、坏死和凋亡的现象得到有效缓解。结论:这两种优化的巴马长寿特征饮食模式均能够有效增强年轻小鼠与自然衰老小鼠学习记忆能力并起到一定的抗衰老效果,尤其在自然衰老组小鼠中表现明显,其中优食I效果更佳。这提示两种巴马长寿特征饮食模式在健康长寿指导方面效果明显,也为其进一步研究提供良好的参考。  相似文献

目的:探讨蛹虫草对D- 半乳糖模型小鼠学习记忆能力的影响。方法:使用D- 半乳糖建立小鼠衰老模型,用蛹虫草灌胃进行实验性治疗。持续6 周后,通过避暗实验、穿梭箱实验和跳台实验检测小鼠的学习记忆能力。结果:与对照组相比,模型组小鼠的学习、记忆能力下降(P < 0.05 或P < 0.01);与模型组相比,蛹虫草组的学习、记忆能力有不同程度地得到恢复(P < 0.05)。结论:蛹虫草可改善由D- 半乳糖所导致的学习、记忆障碍。  相似文献

The hyperphosphorylation of microtubule‐associated protein tau (tau) in the hippocampus can be caused by central and peripheral insulin resistance and these alterations are related to the development of tauopathies, such as Alzheimer's disease. In this study, we used a high‐fat diet to induce obesity and insulin resistance in adult Swiss mice and checked whether supplementation with Myrciaria jaboticaba berry peel for 10 weeks could improve insulin sensitivity, learning/memory performance, and prevent tau phosphorylation in the hippocampus. Furthermore, adipocytokines, inflammatory markers, and oxidative stress were assessed. Myrciaria jaboticaba peel has phenolic compounds (e.g., cyanidin, ellagic acid), dietary fiber and carotenoids, which contribute to great antioxidant capacity. Supplementation of the high‐fat diet with 4% M. jaboticaba peel prevented fat weight gain and reduced peripheral insulin resistance. The treated group also showed lower tau phosphorylation in the hippocampus corroborating better learning/memory performance in the Morris water maze test. Maintenance of neuronal viability, lower levels of hippocampal inflammatory markers, and improved brain antioxidant defenses were also related to the consumption of M. jaboticaba peel. These findings contribute to a better understanding of how a high‐fat diet supplemented with jaboticaba berry peel counteracts the impairment of cognitive functions caused by high‐fat diet intake and diet‐induced insulin resistance.  相似文献

Aleatico grapes (Vitis vinifera L. cv. Aleatico) picked at 20.4–20.6oBrix were treated with air containing 500 mg/L ethylene for 15 h, or 1 mg/L 1‐MCP (1‐methylcyclopropene) for 15 h, or 1 mg/L 1‐MCP for 15 h + 500 mg/L ethylene for further 15 h, or air (control). All treatments were imposed at 20oC and 95–100% relative humidity. Samples undergoing treatment were then held in air at 20oC and 85% relative humidity for 13 days. Ethylene production from ethylene‐treated grapes was very high (25 μL/kg.h) immediately after treatment, and declined progressively until day 6; 1‐MCP‐ethylene‐treated grapes showed lower ethylene production (10 μL/kg.h). This high ethylene production was mainly due to in‐out diffusion of ethylene absorbed during the treatment, but a potential effect of 1‐MCP is postulated, since 1‐MCP‐treated grapes did not show ethylene production for 2 days after treatment, whereas control grapes produced constant amounts (3–4 μL/kg.h) of ethylene. After day 6, no significant difference was observed among samples. No difference in respiration was revealed. Total phenols were increased significantly by ethylene treatment from 520 mg/L up to 710, while total anthocyanins were lost in air‐treated samples (from 175 mg/L to 120) and remained constant in ethylene and 1‐MCP‐treated grapes. HPLC analyses of anthocyanins confirmed this result: air‐treated samples lost anthocyanins rapidly while this loss was delayed in gas‐treated samples. Terpenols, esters, alcohols, and C6 compounds were increased by ethylene treatment. Conversely, 1‐MCP‐treated grapes showed a significant loss of terpenols and esters, although maintaining alcohols, especially hexan‐1‐ol. Berry pathology was also affected, with 1‐MCP and 1‐MCP‐ethylene treated grapes hosting a diffuse attack of grey mould. Wine analyses revealed higher concentrations of phenols and anthocyanins in wine from ethylene‐treated grapes, although aromatic compounds were lost, and thus the aroma profile of those wines was diminished.  相似文献

探讨了牡蛎酶解产物对小鼠空间学习记忆能力的影响。选用中性蛋白酶、动物蛋白酶和复合酶分别水解牡蛎蛋白得到EHO1、EHO2和EHO3,分析其基本营养成分和分子量分布;采用Morris水迷宫行为学方法考察EHO对小鼠学习记忆的改善能力。结果显示,EHO含有丰富的肽、糖原、牛磺酸,分子量在主要集中在1300~10000u之间;定位航行测试显示,相对于空白组,EHO干预后小鼠的潜伏期都明显的缩短,空间搜索策略的比例明显提高,EHO3组效果最好,潜伏期(16.77±7.74)s(p〈0.05),空间搜索的比例为85.71%;而空间探索实验中,给药组小鼠的穿越次数、中央活动时间和目标象限路程比都有不同程度的增加。实验结果表明一定剂量的EHO能够提高小鼠的空间学习和记忆能力。  相似文献

This study was designed to evaluate the effect of yam tuber material [Dioscorea alata L. var. purpurea (M.) Pouch.] on the amyloid β‐protein (Aβ) accumulation, monoamine oxidase (MAO) activity, cognitive ability and antioxidative defense system in senescence‐accelerated mice (SAMP8). Three‐month‐old male mice were fed with four different diets for 12 weeks: a casein diet (control group) and a casein diet supplemented with either 100, 200 or 400 g kg?1 lyophilized yam. The results of the active shuttle avoidance test showed that the mice fed with the yam‐containing diets had significantly better learning and memory ability than the control group. All yam‐containing diet groups had lower Aβ levels and MAO B activities than the control, whereas the MAO A activity did not differ among the four diet groups. At the 400 g kg?1 level, the yam‐containing diet group showed significantly lower concentration of thiobarbituric acid‐reactive substances (TBARS), higher total thiol level and superoxide dismutase (SOD) activity in the hippocampus than the control, but these values were not altered by the 100 and 200 g kg?1 yam‐containing diet groups. The addition of 200 or 400 g kg?1 yam also lowered the triglyceride levels but not the total cholesterol concentration. These results indicate that lyophilized yam could reduce brain Aβ accumulation, MAO B activity and cognitive deficits and promote the antioxidative defense system in SAMP8 mice. The improvements were in a dose‐dependent manner, possibly because the 400 g kg?1 yam‐containing diet might contain more antioxidative phytochemicals. 研究东北山核桃仁油(Juglans mandshurica oil,JMO)促进小鼠学习和记忆能力的影响。将雄性健康昆明种小鼠40只,随机分为4组,即对照组(Control)、山核桃仁油低、中、高剂量组(Low、Middle、High)。Low、Middle、High剂量组分别灌胃山核桃仁油5、10、20mL/kgbw.d;对照组灌胃生理盐水,连续灌胃30d。第31d开始,进行跳台实验和水迷宫实验。结果山核桃仁油组小鼠跳台被动回避的潜伏期明显延长,记忆巩固成绩中中剂量组与对照组比较差异显著(P<0.05);小鼠到达水迷宫终点的时间和错误次数明显减少,中剂量组与对照组比较差异显著(P<0.05)。研究表明,东北山核桃仁油能明显提高小鼠跳台被动回避反应能力和水迷宫空间辨别的学习记忆能力,具有明显促进小鼠学习和记忆能力的作用。  相似文献

用东莨菪碱(scopolamine,SCOP)诱导小鼠记忆障碍模型,探究饲料中添加核桃对小鼠学习记忆能力的影响。实验组喂食含有质量分数9%核桃(核桃高剂量组和高剂量预防组)或6%核桃(核桃低剂量组和低剂量预防组)的饲料,而对照组(空白组、模型组和阳性对照组)喂食普通饲料。在第8周时,通过Morris水迷宫(Morris water maze,MWM)和小鼠脑组织丙二醛(malondialdehyde,MDA)、谷胱甘肽(glutathione,GSH)含量和总超氧化物歧化酶(total superoxide dismutase,T-SOD)、乙酰胆碱酯酶(acetylcholinesterase,AChE)活力测定,评估小鼠的学习记忆表现。结果表明:与模型组相比,在MWM实验中,高剂量核桃预防组的逃避潜伏期在第4天极显著缩短(P<0.01),穿台次数显著增加(P<0.05),目标象限停留时间及路程占比分别显著延长和提高(P<0.05),同时,脑组织中AChE活力((0.94±0.08)U/mg)、MDA含量((11.64±0.58)nmol/mg)显著降低(P<0.05),GSH含量((69.53±2.11)mg/g)、T-SOD活力((93.36±4.94)U/mg)显著提高(P<0.05)。结论:饲料中添加核桃可以抑制小鼠大脑氧化损伤,增强胆碱能功能,预防SCOP诱导的小鼠记忆障碍,提高小鼠的学习记忆能力。  相似文献

为探讨昆仑雪菊水溶性黄酮提取物(water soluble flavonoids from Coreopsis tinctoria flowers,CTWF)对D-半乳糖致衰老小鼠学习记忆障碍的影响,以SPF级昆明种小鼠为实验对象,随机分为正常组、衰老模型组、VE组(50 mg/(kg·d))和CTWF低、高剂量组(150、600 mg/(kg·d))。除正常组皮下注射生理盐水外,其余各组均注射D-半乳糖(300 mg/(kg·d)),连续42 d造模,注射1 h后,药物组分别灌胃VE和不同剂量CTWF,正常组和模型组灌胃等量生理盐水。采用Morris水迷宫测试各组小鼠学习记忆能力,并测定小鼠脑组织乙酰胆碱(acetylcholine,Ach)含量,乙酰胆碱酯酶(acetylcholinesterase,AchE)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活力和丙二醛(malondialdehyde,MDA)含量。结果表明:CTWF能够显著提升衰老模型小鼠的学习记忆能力(P<0.05),并显著提高脑组织SOD、GSH-Px活力和Ach含量(P<0.05),降低AchE活力和MDA含量(P<0.05)。因此,CTWF具有明显改善衰老模型小鼠学习记忆障碍的作用,其机制可能与提高脑组织胆碱能系统功能和减少自由基损伤有关。  相似文献

The use of 1‐methylcyclopropene (1‐MCP) to extend the shelf life of cherry tomato (Lycopersicon esculentum var. cerasiforme) was investigated. Higher concentrations of 1‐MCP delayed the ethylene induced climacteric peaks in mature green (MG) and breaker (BR) fruits. Thirteen days after treatment, control fruits were 55% as firm as fruit treated with 0.11 μL L?1 1‐MCP. The initiation of fruit softening, chlorophyll degradation and accumulation of lycopene and carotenoids was delayed in treated fruits. Higher 1‐MCP concentrations inhibited the accumulation of lycopene and carotene such that the colour of the fruit did not reach that of control fruit. Treatment of MG cherry tomatoes with 1‐MCP enhanced shelf life optimally, while treatment of BR fruits was beneficial where full colour and high content of lycopene and carotenoids is important. Differentiation of the different stages of ripening before application of 1‐MCP is needed in order to succeed in accomplishing different postharvest objectives.  相似文献

目的观察邻苯二甲酸二异丁酯(DiBP)经口亚慢性暴露,对雄性小鼠空间学习记忆行为的影响。方法每组12只雄性昆明小鼠,空白对照组和阳性对照组给予玉米油溶剂灌胃,各剂量组给予相应剂量的邻苯二甲酸二异丁酯(50、250、500、1000mg/kg BW)玉米油溶液灌胃,连续8周。染毒前后分别对各组动物进行旷场试验和Morris水迷宫试验,其中阳性对照组在染毒结束后进行旷场试验和Morris水迷宫试验前经腹腔注射5mg/kg BW的氢溴酸东莨菪碱。结果试验前各组动物的学习记忆能力一致。8周实验结束后,阳性对照组和1000mg/kg BW剂量组小鼠与空白对照组小鼠相比在旷场中心区域停留时间明显延长(P0.05)。500mg/kg BW剂量组和阳性对照组与空白对照组相比Morris水迷宫训练第一天的逃避潜伏期明显延长(P0.01),1000mg/kg BW剂量组与空白对照组相比逃避潜伏期也明显延长(P0.05)。其他各项指标与空白对照组相比,差异均无统计学意义。结论在本试验条件下,DiBP会降低雄性昆明小鼠的空间学习记忆能力。  相似文献

Scope: We examined whether dietary supplementation with fish oil modulates inflammation, fibrosis and oxidative stress following obstructive renal injury. Methods and results: Three groups of Sprague–Dawley rats (n=16 per group) were fed for 4 wk on normal rat chow (oleic acid), chow containing fish oil (33 g eicosapentaenoic acid and 26 g docosahexaenoic acid per kg diet), or chow containing safflower oil (60 g linoleic acid per kg diet). All diets contained 7% fat. After 4 wk, the rats were further subdivided into four smaller groups (n=4 per group). Unilateral ureteral obstruction was induced in three groups (for 4, 7 and 14 days). The fourth group for each diet did not undergo surgery, and was sacrificed as controls at 14 days. When rats were sacrificed, plasma and portions of the kidneys were removed and frozen; other portions of kidney tissue were fixed and prepared for histology. Compared with normal chow and safflower oil, fish oil attenuated collagen deposition, macrophage infiltration, TGF‐β expression, apoptosis, and tissue levels of arachidonic acid, MIP‐1α, IL‐1β, MCP‐1 and leukotriene B4. Compared with normal chow, fish oil increased the expression of HO‐1 protein in kidney tissue. Conclusions: Fish oil intake reduced inflammation, fibrosis and oxidative stress following obstructive renal injury.  相似文献

探讨白藜芦醇对D-半乳糖致衰老小鼠脑组织抗氧化与学习记忆的影响,揭示白藜芦醇延缓脑衰老的作用。将50 只雄性ICR鼠随机分为正常对照组,模型组,白藜芦醇低、中、高剂量组。模型组及白藜芦醇治疗组连续8周于小鼠颈背部皮下注射D-半乳糖120 mg/(kg·d),给予不同剂量白藜芦醇(25、50、100 mg/(kg·d))灌胃;正常对照组注射、灌胃等量生理盐水。采用Morris水迷宫检测各组小鼠的学习记忆能力,并测定各组小鼠脑组织超氧化物歧化酶(superoxide dismutase,SOD)、总抗氧化能力(total antioxidant capacity,T-AOC)、丙二醛(malondialdehyde,MDA)的含量。白藜芦醇能够显著缩短D-半乳糖致衰老小鼠Morris水迷宫中找到隐藏平台时间(P<0.01),提高游泳速度(P<0.01)和穿越目标象限的次数(P<0.01);提高衰老模型小鼠脑组织SOD(P<0.01)、T-AOC活性(P<0.05),降低MDA含量(P<0.01)。白藜芦醇能够改善D-半乳糖致衰老模型小鼠的学习记忆能力,提高脑组织的抗氧化能力,具有一定延缓脑衰老的作用。  相似文献

用含Lactobacillus bulgaricus和Streptococcus Thermophilus活菌酸奶30%的饲料饲喂衰老小鼠3d,测定其胸腺重量、脾淋巴细胞增殖能力、IL-2(Interleukin,IL)产生量、脾T淋巴细胞中CD4^+/CD8^+细胞比例等指标,发现饲喂酸奶显著增加了胸腺重和IL-2的产生量,提高了脾T淋巴细胞的增殖能力,但未达到年轻小鼠的水平。此外还观察到酸奶处理提高了T淋巴细胞中CD4^+/CD8^+细胞的比例。这些结果表明,酸奶对于老年小鼠的免疫功能具有一定恢复作用。  相似文献

BACKGROUND: Coriandrum sativum L., commonly known as coriander and belonging to the family Apiaceae (Umbelliferae), is cultivated throughout the world for its nutritional value. The present study was undertaken to investigate the effects of fresh Coriandrum sativum leaves (CSL) on cognitive functions, total serum cholesterol levels and brain cholinesterase activity in mice. In this study, CSL (5, 10 and 15% w/w of diet) was fed orally with a specially prepared diet for 45 days consecutively to experimental animals. Elevated plus‐maze and passive avoidance apparatus served as the exteroceptive behavioral models for testing memory. Diazepam, scopolamine and ageing‐induced amnesia served as the interoceptive behavioral models. RESULTS: CSL (5, 10 and 15% w/w of diet) produced a dose‐dependent improvement in memory scores of young as well as aged mice. CSL also reversed successfully the memory deficits induced by scopolamine (0.4 mg kg?1, i.p.) and diazepam (1 mg kg?1, i.p.). Interestingly, brain cholinesterase activity and serum total cholesterol levels were considerably reduced by CSL administration in daily diets concomitantly for 45 days. CONCLUSION: CSL may be a useful remedy in the management of Alzheimer's disease on account of its multifarious effects such as, memory‐improving property, cholesterol‐lowering property and anticholinesterase activity. 本文比较研究了鱿鱼卵磷脂和蛋黄磷脂对老年痴呆小鼠学习记忆能力的影响。雄性BALB/c小鼠分为正常对照、模型、鱿鱼卵磷组(SL)和蛋黄磷脂组(EL),喂养1周后,腹腔注射东莨菪碱造模并进行行为学测试。测定脑皮层、白质、海马中乙酰胆碱酯酶(Tch E)、超氧化物歧化酶(SOD)、单胺氧化酶(MAO)活力和脑内丙二醛(MDA)含量。结果显示,SL组水迷宫潜伏期显著降低,而穿越平台次数和目标象限停留时间显著提高,行为学改善效果优于EL组。SL能显著降低脑内各组织Tch E活力、MDA水平和白质中MAO活力,显著提高脑皮层、海马和白质中SOD活力,且作用效果优于EL。综上,鱿鱼卵磷脂通过影响中枢胆碱能神经系统和脑内抗氧化系统,改善了东莨菪碱所致痴呆小鼠学习记忆能力。  相似文献
