Eight ruminally cannulated Holstein cows that were part of a larger lactation trial were blocked by days in milk and randomly assigned to replicated 4 × 4 Latin squares to quantify effects of nonprotein N (NPN) content of alfalfa silage (AS) and red clover silage (RCS) on omasal nutrient flows. Diets, fed as total mixed rations, contained 50% dry matter from control AS (CAS), ammonium tetraformate-treated AS (TAS), late maturity RCS (RCS1), or early maturity RCS (RCS2). Silages differed in NPN and acid detergent insoluble N (% of total N): 50 and 4% (CAS); 45 and 3% (TAS); 27 and 8% (RCS1); 29 and 4% (RCS2). The CAS, TAS, and RCS2 diets had 36% high-moisture shelled corn and 3% soybean meal, and the RCS1 diet had 31% high-moisture shelled corn and 9% soybean meal. All diets contained 10% corn silage, 27% neutral detergent fiber, and 17 to 18% crude protein. Compared with RCS, feeding AS increased the supply of rumen-degraded protein and omasal flows of nonammonia N and microbial protein, which may explain the improved milk yield observed in the companion lactation trial. However, omasal flow of rumen-undegraded protein was 34% greater on RCS. Except for Arg, omasal flows of individual AA, branched-chain AA, nonessential AA, essential AA, and total AA did not differ between cows fed AS vs. RCS. Within AS diets, no differences in omasal AA flows were observed. However, omasal flows of Asp, Ser, Glu, Cys, Val, Ile, Tyr, Lys, total nonessential AA, and total AA all were higher in cows fed RCS1 vs. cows fed RCS2. Effects of different protein supplements on milk production and nutrient utilization in lactating dairy cows 总被引:2,自引:0,他引:2
Sixteen (8 ruminally cannulated) multiparous and 8 primiparous lactating Holstein cows were used in 6 replicated 4 × 4 Latin squares to test the effects of feeding supplemental protein as urea, solvent soybean meal (SSBM), cottonseed meal (CSM), or canola meal (CM) on milk production, nutrient utilization, and ruminal metabolism. All diets contained (% of DM) 21% alfalfa silage and 35% corn silage plus 1) 2% urea plus 41% high-moisture shelled corn (HMSC), 2) 12% SSBM plus 31% HMSC, 3) 14% CSM plus 29% HMSC, or 4) 16% CM plus 27% HMSC. Crude protein was equal across diets, averaging 16.6%. Intake and production were substantially reduced, and milk urea, blood urea, and ruminal ammonia were increased on urea vs. the diets supplemented with true protein. Although intake was lower in cows fed SSBM compared with CM, no differences were observed for milk yield among SSBM, CSM, and CM. Yields of fat and protein both were lower on CSM than on CM, whereas SSBM was intermediate. Milk urea and milk protein contents also decreased when CSM replaced SSBM or CM. Diet did not affect ruminal volatile fatty acids except that isobutyrate concentration was lowest on urea, intermediate on CSM, and greatest on SSBM and CM. Urinary excretion of urea N and total N was greatest on urea, intermediate on SSBM and CM, and lowest on CSM. Apparent N efficiency (milk N/N intake) was lower on the CSM diet than on the SSBM diet. Overall, production and N utilization were compromised when the diets of high-yielding dairy cows were supplemented with urea rather than true protein and the value of the true proteins, from most to least effective, was in the order CM > SSBM > CSM. 相似文献

Eight ruminally cannulated multiparous Holstein cows that were part of a larger production trial were used to study the effects of varying dietary ratios of alfalfa silage (AS) to corn silage (CS) on omasal flow of nutrients and microbial protein. Cows were blocked by DIM and randomly assigned to 2 replicated 4 × 4 Latin squares (28-d periods). Diets fed contained (dry matter basis): A) 51% AS, 43% rolled high-moisture shelled corn (HMSC), and 3% solvent soybean meal (SSBM); B) 37% AS, 13% CS, 39% HMSC, and 7% SSBM; C) 24% AS, 27% CS, 35% HMSC, and 12% SSBM; or D) 10% AS, 40% CS, 31% HMSC, and 16% SSBM. Crude protein (CP) contents were 17.2, 16.9, 16.6, and 16.2% for diets A, B, C, and D. All 4 diets were high in energy, averaging 49% nonfiber carbohydrates and 24% neutral detergent fiber. Total microbial nonammonia nitrogen flow was lower on diet D (423 g/d) compared with diets A (465 g/d), B (479 g/d), and C (460 g/d). A significant quadratic effect indicated that microbial protein synthesis was maximal at 38% AS. Supply of rumen-degraded protein decreased linearly from 3,068 g/d (diet A) to 2,469 g/d (diet D). Omasal flow of rumen-undegraded protein did not differ among diets and averaged 1,528 g/d. However, when expressed as a percentage of dry matter intake, rumen-undegraded protein increased linearly from 5.59% (diet A) to 6.13% (diet D), probably because CP from SSBM was more resistant to degradation than CP from AS. Essential AA flow was lowest on diet D, and Lys flow tended to be lower on diet D, which may explain the lower milk and protein yields observed on that diet. 相似文献

Twenty-eight (8 with ruminal cannulas) lactating Holstein cows were assigned to seven 4 × 4 Latin squares in a 16-wk trial to study the effects on production and ruminal metabolism of feeding differing proportions of rumen-degraded protein (RDP) from soybean meal and urea. Diets contained [dry matter (DM) basis] 40% corn silage, 15% alfalfa silage, 28 to 30% high-moisture corn, plus varying levels of ground dry shelled corn, solvent- and lignosulfonate-treated soybean meal, and urea. Proportions of the soybean meals, urea, and dry corn were adjusted such that all diets contained 16.1% crude protein and 10.5% RDP, with urea providing 0, 1.2, 2.4, and 3.7% RDP (DM basis). As urea supplied greater proportions of RDP, there were linear decreases in DM intake, yield of milk, 3.5% fat-corrected milk, fat, protein, and solids-not-fat, and of weight gain. Milk contents of fat, protein, and solids-not-fat were not affected by source of RDP. Replacing soybean meal RDP with urea RDP resulted in several linear responses: increased excretion of urinary urea-N and concentration of milk urea-N, blood urea-N, and ruminal ammonia-N and decreased excretion of fecal N; there was also a trend for increased excretion of total urinary N. A linear increase in neutral detergent fiber (NDF) digestibility, probably due to digestion of NDF-N from lignosulfonate-treated soybean meal, was observed with greater urea intake. Omasal sampling revealed small but significant effects of N source on measured RDP supply, which averaged 11.0% (DM basis) across diets. Increasing the proportion of RDP from urea resulted in linear decrease in omasal flow of dietary nonammonia N (NAN) and microbial NAN and in microbial growth efficiency (microbial NAN/unit of organic matter truly digested in the rumen). These changes were paralleled by large linear reductions in omasal flows of essential, nonessential, and total amino acids. Overall, these results indicated that replacing soybean meal RDP with that from urea reduced yield of milk and milk components, largely because of depressed microbial protein formation in the rumen and that RDP from nonprotein-N sources was not as effective as RDP provided by true protein. 相似文献

Eight lactating cows were fed 4 diets in which dietary crude protein (CP) was increased in steps of approximately 2 percentage units from 11 to 17% of DM by replacing high-moisture corn with soybean meal supplemented with rumen-protected Met to maintain a Lys:Met ratio of 3:1 in metabolizable protein. Trial design was a replicated 4 × 4 Latin square; experimental periods lasted 28 d, with data and sample collection being performed during wk 3 and 4 of each period. Digesta samples were collected from the rumen as well as the omasum to measure metabolite concentrations and ruminal outflow of N fractions using infusion of 15N-enriched ammonia to quantify microbial nonammonia N (NAN) and nonmicrobial NAN. Data were analyzed using the MIXED procedure of SAS (SAS Institute Inc.). There were linear increases in the yields of milk and true protein and concentration of milk urea N, and a linear decrease in N efficiency, with increasing dietary CP. Apparent ruminal and total-tract N digestibility increased linearly with increasing dietary CP, but estimated true total-tract N digestibility was not affected. Apparent digestibility of the other macronutrients was not influenced by diet. Ruminal ammonia, total AA and peptides, and branched-chain VFA also increased linearly with dietary CP. The 15N enrichment of liquid- and particle-associated microbes linearly declined with increasing dietary CP due to decreasing 15N enrichment of the ammonia pool. Although no effect of dietary CP on nonmicrobial NAN flow was detected, total NAN flow increased linearly from 525 g/d at 11% CP to 637 g/d at 17% CP due to the linear increase in microbial NAN flow from 406 g/d at 11% CP to 482 g/d at 17% CP. Under the conditions of this study, when dietary CP was increased by adding soybean meal supplemented with rumen-protected Met, improved milk and protein yields were driven not by RUP supply but by increased ruminal outflow of microbial protein. 相似文献

The aim was to study the effects of rumen N balance (RNB), dietary protein source, and their interaction on feed intake, N partitioning, and rumen microbial crude protein (MCP) synthesis in lactating dairy cows. Twenty-four lactating Holstein cows were included in a replicated 4 × 4 Latin square experimental design comprising four 20-d periods, each with 12 d of adaptation to the experimental diets and 8 d of sampling. The dietary treatments followed a 2 × 2 factorial arrangement (i.e., 4 treatments) with 2 main protein sources [faba bean grain (FB) and SoyPass (SP; Beweka Kraftfutterwerk GmbH, Heilbronn, Germany)] offered at 2 dietary RNB levels each [0 g/kg of dry matter, DM (RNB0) and ?3.2 g/kg of DM (RNB?)]. The RNB was calculated as the difference between dietary crude protein (CP) intake and the rumen outflow of undegraded feed CP and MCP and divided by 6.25. Composition of concentrate mixtures was adjusted to create diets with desired RNB levels. Each of these protein sources supplied ≥35% of total dietary CP. Both diets for each protein source were isoenergetic but differed in CP concentrations. The DM intake (kg/d) was lower for RNB? than for RNB0 in diets containing FB, whereas no differences were seen between the RNB levels for SP diets. The RNB? decreased N intake and urinary N excretion but increased milk N use efficiency in both FB and SP diets, with greater differences between the RNB levels for FB diets than for SP diets. Similarly, duodenal MCP synthesis (g/kg of digestible organic matter intake) estimated from purine derivatives in the urine was lower for RNB? than for RNB0 in FB diets but similar between the RNB levels in diets containing SP. Low RNB of approximately ?65 g/d (approximately ?3.2 g/kg of DM) in diets reduced feed intake, N balance, and performance in high-yielding dairy cows with possibly more pronounced effects in diets containing rapidly degradable protein sources. 相似文献

The objective of this study was to evaluate the effects of a fermentation byproduct on rumen fermentation and microbial yield in high producing lactating dairy cattle. Eight ruminally cannulated multiparous Holstein cows averaging (mean ± standard deviation) 60 ± 10 d in milk and 637 ± 38 kg of body weight were assigned to 1 of 2 treatment sequences in a switchback design. Treatment diets contained (dry matter basis) 44% corn silage, 13% alfalfa silage, 12% ground corn, and 31% premix containing either a control mix of urea and wheat middlings (CON) or a commercial fermentation byproduct meal (Fermenten, Arm and Hammer Animal Nutrition, Princeton, NJ) at 3% diet inclusion rate (EXP). Diets were formulated to be isonitrogenous and isocaloric, with similar levels of neutral detergent fiber and starch. The trial consisted of three 28-d experimental periods, where each period consisted of 21 d of diet adaptation and 7 d of data and sample collection. Omasal nutrient flows were determined using a triple-marker technique and double-labeled 15N15N-urea. The EXP diet provided 18 g/d more nonammonia N versus the CON diet, representing 3.0% of total N intake. Energy-corrected milk yield (41.7 and 43.1 kg/d for CON and EXP, respectively), milk fat, and protein yield and content did not differ between treatments. Total dry matter intake was similar between treatments (25.5 and 26.4 kg/d for CON and EXP, respectively). Ammonia N concentration and pool size in the rumen was greater in cows fed the EXP diet. No differences were observed in rumen or total-tract dry matter, organic matter, or neutral detergent fiber digestibility. Ruminal degradation of feed N was 15% lower in cows fed EXP diets, resulting in differences in omasal N flows. Results demonstrated the fermentation byproduct meal had a sparing effect on degradable feed protein, but did not increase microbial N flow from the rumen. 相似文献

The objective of this study was to evaluate the effect of rolled barley supplementation on microbial composition and omasal flows of bacterial, protozoal, and nonmicrobial AA in cows fed fresh perennial ryegrass (Lolium perenne L.; PRG). Ten ruminally cannulated multiparous Holstein cows averaging (mean ± standard deviation) 49 ± 23 d in milk and 513 ± 36 kg of body weight were assigned to 1 of 2 treatments in a switchback design. The treatment diets were PRG only or PRG plus 3.5 kg of dry matter rolled barley (G+RB). The study consisted of three 29-d periods where each period consisted of 21 d of diet adaptation and 8 d of data and sample collection. A double-marker system was used to quantify nutrient flow entering the omasal canal along with 15N-ammonium sulfate to label and measure the microbial and nonmicrobial omasal flow of AA. Overall, rolled barley supplementation had no effect on the AA composition of the omasal liquid-associated and particle-associated bacteria. Rolled barley supplementation affected the AA concentrations of omasal protozoa; however, the differences were nutritionally minor. Particle-associated bacteria AA flow was increased for all AA, except for Trp and Pro, in cows fed the G+RB diet. Rolled barley supplementation had no effect on protozoal AA flow. On average, protozoa accounted for 23% of the microbial essential AA flow, which ranged from 17 to 28% for Trp and Lys, respectively. The flow of all AA in omasal true digesta increased in cows fed the G+RB diet compared with the PRG-only diet, resulting in a 228 g/d increase in total AA flow in cows fed the G+RB diet. This increase in total AA flow in cows fed the G+RB diet was due to an increase in microbial AA flow. Rolled barley supplementation had no effect on nonmicrobial AA flow. The nonmicrobial AA flow modestly contributed to total AA flow, accounting for 15.6% on average. These results indicated that extensive ruminal degradation of PRG AA occurred (83.5%), and we demonstrated that cows consuming PRG-based diets exhibit a large dependence on microbial AA to support metabolizable AA supply. Rolled barley supplementation can increase the omasal flow of microbial AA in cows consuming PRG-based diets. However, further research is required to elucidate if this increased AA supply can support higher milk yield under such dietary conditions. 相似文献

Sixteen multiparous lactating Holstein cows (four with rumen cannulae) were fed diets varying in the content and form of ruminally degradable carbohydrates and N to examine dietary effects on microbial protein synthesis (MPS) and whole animal N efficiency, and to evaluate the use of a model based on milk urea N (MUN) for predicting urinary N excretion and N utilization efficiency (NUE). A replicated Latin square design (consisting of diet and experimental period) was employed. The four diets consisted of two low protein diets with either 20% ground corn (diet LP) or 13.5% ground corn plus 3% sucrose (diet LP sucrose) and two high protein diets with 13.5% corn and 3% sucrose with either urea (diet HP urea) or soybean meal (diet HP SBM) as supplemental rumen-degradable protein sources. The intakes of dry matter and N were increased by increasing dietary crude protein (CP) level. However, the yields of milk and milk protein were not affected by CP level. Yield of microbial protein was reduced by sucrose and increased by CP level. There were no differences between urea and SBM supplementation on DM intake, milk yield, or MPS. Mean urinary N excretion for all cows (252 g/d) was underestimated by 55 g/d or overestimated by 25 or 33 g/d using alternative equations based on MUN. Subsequently, NUE (mean = 22.4%) was underestimated by 7.5, 3.2, or 2.9%, using a previously published set of equations. Urinary N excretion and NUE could be predicted within 10 and 14% of observed values, respectively, using a set of equations incorporating MUN. Therefore, MUN appears to be a useful tool to help assess N losses from lactating cows. 相似文献

Effect of dietary crude protein concentration on ruminal nitrogen metabolism in lactating dairy cows
Ten lactating Holstein cows fitted with ruminal cannulas that were part of a larger feeding trial were blocked by days in milk into 2 groups and then randomly assigned to 1 of 2 incomplete 5 × 5 Latin squares. Diets contained [dry matter (DM) basis] 25% alfalfa silage, 25% corn silage, and 50% concentrate. Rolled high-moisture shelled corn was replaced with solvent-extracted soybean meal to increase crude protein (CP) from 13.5% to 15.0, 16.5, 17.9, and 19.4% of DM. Each of the 4 experimental periods lasted 28 d with data and sample collection performed during the last 8 d. Digesta samples were collected from the omasum to quantify the ruminal outflow of different N fractions. Intake of DM was not affected but showed a quadratic trend with maxima of 23.9 kg/d at 16.5% CP. Ruminal outflow of total bacterial nonammonia N (NAN) was not different among diets but a significant linear effect of dietary CP was detected for this variable. Bacterial efficiency (g of total bacterial NAN flow/kg of organic matter truly digested in the rumen) and omasal flows of dietary NAN and total NAN also showed positive linear responses to dietary CP. Total NAN flow increased from 574 g/d at 13.5% CP to 688 g/d at 16.5% CP but did not increase further with the feeding of more CP. Under the conditions of this study, 16.5% of dietary CP appeared to be sufficient for maximal ruminal outflow of total bacterial NAN and total NAN. 相似文献

A study was conducted to determine the effects of including either wheat-based (W-DDGS) or corn-wheat blend (B-DDGS) dried distillers grains with solubles as the major protein source in low- or high-crude protein (CP) diets fed to dairy cows on ruminal function, microbial protein synthesis, omasal nutrient flows, urea-N recycling, and milk production. Eight lactating Holstein cows (768.5 ± 57.7 kg of body weight; 109.5 ± 40.0 d in milk) were used in a replicated 4 × 4 Latin square design with 28-d periods (18 d of dietary adaptation and 10 d of measurements) and a 2 × 2 factorial arrangement of dietary treatments. Four cows in one Latin square were ruminally cannulated for the measurement of ruminal fermentation characteristics, microbial protein synthesis, urea-N recycling kinetics, and omasal nutrient flow. The treatment factors were type of distillers co-product (W-DDGS vs. B-DDGS) and dietary CP content [15.2 vs. 17.3%; dry matter (DM) basis]. The B-DDGS was produced from a mixture of 15% wheat and 85% corn grain. All diets were formulated to contain 10% W-DDGS or B-DDGS on a DM basis. No diet effect was observed on DM intake. Yields of milk, fat, protein, and lactose, and plasma urea-N and milk urea-N concentrations were lower in cows fed the low-CP compared with those fed the high-CP diet. Although feeding B-DDGS tended to reduce ruminal ammonia-N (NH3-N) concentration compared with feeding W-DDGS (9.3 vs. 10.5 mg/dL), no differences were observed in plasma urea-N and milk urea-N concentrations. Additionally, dietary inclusion of B-DDGS compared with W-DDGS did not affect rumen-degradable protein supply, omasal flows of total N, microbial nonammonia N (NAN), rumen-undegradable protein, and total NAN, or urea-N recycling kinetics and milk production. However, cows fed the low-CP diet had lower N intake, rumen-degradable protein supply, ruminal NH3-N concentration, and omasal flows of N, microbial NAN, and total NAN compared with those fed the high-CP diet. Feeding the low-CP compared with the high-CP diet also resulted in lower endogenous urea-N production, urea-N recycled to the gastrointestinal tract, and urea-N excretion in urine. In summary, our results indicate that both W-DDGS and B-DDGS can be included as the major protein sources in dairy cow diets without compromising nutrient supply and production performance. However, feeding the low-CP diet lowered omasal flows of microbial protein and metabolizable protein, which, in turn, resulted in lower milk production compared with feeding the high-CP diet. 相似文献

We evaluated the effects of supplemental calcium ammonium nitrate (CAN) fed to dairy cows on dry matter (DM) intake, nutrient digestibility, milk quality, microbial protein synthesis, and ruminal fermentation. Six multiparous Holstein cows at 106 ± 14.8 d in milk, with 551 ± 21.8 kg of body weight were used in a replicated 3 × 3 Latin square design. Experimental period lasted 21 d, with 14 d for an adaptation phase and 7 d for sampling and data collection. Cows were randomly assigned to receive the following treatments: URE, 12 g of urea/kg of DM as a control group; CAN15, 15 g of CAN/kg of DM; and CAN30, 30 g of CAN/kg of DM. Supplemental CAN reduced DM intake (URE 19.0 vs. CAN15 18.9 vs. CAN30 16.5 kg/d). No treatment effects were observed for apparent digestibility of DM, organic matter, crude protein, ether extract, and neutral detergent fiber; however, CAN supplementation linearly increased nonfiber carbohydrate digestibility. Milk yield was not affected by treatments (average = 23.1 kg/d), whereas energy-corrected milk (ECM) and 3.5% fat-corrected milk (FCM) decreased as the levels of CAN increased. Nitrate residue in milk increased linearly (URE 0.30 vs. CAN15 0.33 vs. CAN30 0.38 mg/L); however, treatments did not affect nitrite concentration (average: 0.042 mg/L). Milk fat concentration was decreased (URE 3.39 vs. CAN15 3.35 vs. CAN30 2.94%), and the proportion of saturated fatty acids was suppressed by CAN supplementation. No treatment effects were observed on the reducing power and thiobarbituric acid reactive substances of milk, whereas conjugated dienes increased linearly (URE 47.6 vs. CAN15 52.7 vs. CAN30 63.4 mmol/g of fat) with CAN supplementation. Treatments had no effect on microbial protein synthesis; however, molar proportion of ruminal acetate and acetate-to-propionate ratio increased with CAN supplementation. Based on the results observed, supplementing CAN at 30 g/kg of DM should not be recommended as an optimal dose because it lowered DM intake along with ECM and 3.5% FCM, although no major changes were observed on milk quality and ruminal fermentation. 相似文献

We aimed to quantify the productive and metabolic responses, and digestive changes in dairy cows fed various concentrations of soybean oil (SBO) in high-concentrate, sugarcane-based diets. Eight rumen-cannulated multiparous Holstein cows in mid lactation (574 ± 19.1 kg of body weight and 122 ± 6.9 d in milk), averaging 22.5 ± 1.22 kg/d of milk were assigned to replicated 4 × 4 Latin squares. The experimental period lasted 21 d as follows: 14 d for adaptation, followed by a sampling period from d 15 to 21. The diets were formulated with increasing concentrations of SBO [% of dry matter (DM)]: control (0%), low (LSBO; 1.57%), medium (MSBO; 4.43%), and high (HSBO; 7.34%). Dry matter intake decreased quadratically in response to SBO addition. The greatest decrease in DM intake was observed in MSBO and HSBO diets. Both milk and energy-corrected milk yield were quadratically affected by the SBO inclusion, with a slight decrease up to MSBO and substantial decrease in the HSBO diet. The milk fat concentration linearly decreased from 3.78% in the control to 3.50% in the HSBO diet. The potentially digestible neutral detergent fiber digestibility in the rumen decreased from 55.7% in the control to 35.2% in the HSBO diet. The fractional rate of digestion of potentially digestible neutral detergent fiber in the rumen decreased linearly from 3.13 to 1.39%/h from the control to HSBO diet. The fractional rate of indigestible neutral detergent fiber passage in the rumen was quadratically affected, with the lowest value (2.25%/h) for the HSBO diet. Rumen pH increased from 6.42 to 6.67, and ammonia nitrogen decreased from 28.1 to 21.4 mg/dL, in the control and HSBO diets, respectively. Rumen volatile fatty acids decreased quadratically, with the greatest decrease observed in MSBO and HSBO diets. Serum concentrations of glucose, fatty acids, and β-hydroxybutyrate were unaffected by SBO inclusion. However, serum concentrations of total cholesterol and high- and low-density lipoproteins linearly increased with increasing concentrations of SBO in the diet. Inclusion of SBO at concentrations from 4.43 to 7.34% of the diet DM decreased DM intake, energy-corrected milk production, fiber digestibility, and rumen fermentation and was thus not recommended. Extrusion treated canola meal (TCM) was produced in an attempt to increase the rumen-undegraded protein fraction of canola meal (CM). The objective of this study was to evaluate the effects of replacing soybean meal (SBM) with CM or TCM on ruminal digestion, omasal nutrient flow, and performance in lactating dairy cows. To assess performance, 30 multiparous Holstein cows averaging (mean ± SD) 119 ± 23 d in milk and 44 ± 7 kg of milk/d and 15 primiparous cows averaging 121 ± 19 d in milk and 34 ± 6 kg of milk/d were blocked in a randomized complete block design with a 2-wk covariate period and 12-wk experimental period (experiment 1). Dietary ingredients differed only in protein supplements, which were SBM, CM, or TCM. All diets were formulated to contain (dry matter basis) 30% alfalfa silage, 30% corn silage, 4% soy hulls, 2.4% mineral-vitamin premix, and 16% CP. The SBM diet contained 25% high-moisture shelled corn and 8.6% SBM; the canola diets contained 22% high-moisture shelled corn and either 11.2% CM or 11.4% TCM. To assess ruminal digestion and omasal nutrient flow, 6 rumen-cannulated cows were blocked into 2 squares of 3 cows and randomly assigned within blocks to the same 3 dietary treatments as in experiment 1 in a replicated 3 × 3 Latin square design (experiment 2). Data were analyzed using the MIXED procedure of SAS (SAS Institute, Cary, NC). Orthogonal contrasts were used to compare effects of different protein supplements: SBM versus CM + TCM and CM versus TCM. In experiment 1, compared with SBM, apparent total-tract digestibilities of dry matter and nutrients were greater in cows fed both CM diets, and there was a tendency for nutrient digestibilities to be higher in cows fed CM compared with TCM. Diets did not affect milk yield and milk components; however, both canola diets decreased urinary urea N (% of total urinary N), fecal N (% of total N intake), and milk urea N concentration. In experiment 2, compared with SBM, both canola diets increased N intake and tended to increase rumen-degraded protein supply (kg/d) and N truly digested in the rumen (kg/d). Diets did not affect ruminal digestibility, efficiency of microbial protein synthesis, and rumen-undegraded protein flow among diets. Results from this experiment indicate that replacing SBM with CM or TCM in diets of lactating cows improved digestibility and may reduce environmental impact. Twenty-eight (8 with ruminal cannulas) lactating Holstein cows were assigned to 4 × 4 Latin squares and fed diets with different levels of rumen-degraded protein (RDP) to study the effect of RDP on production and N metabolism. Diets contained [dry matter (DM) basis] 37% corn silage, 13% alfalfa silage, and 50% concentrate. The concentrate contained solvent and lignosulfonate-treated soybean meal and urea, and was adjusted to provide RDP at: 13.2, 12.3, 11.7, and 10.6% of DM in diets A to D, respectively. Intake of DM and yield of milk, fat-corrected milk, and fat were not affected by treatments. Dietary RDP had positive linear effects on milk true protein content and microbial non-ammonia N (NAN) flow at the omasal canal, and a quadratic effect on true protein yield, with maximal protein production at 12.3% RDP. However, dietary RDP had a positive linear effect on total N excretion, with urinary N accounting for most of the increase, and a negative linear effect on environmental N efficiency (kg of milk produced per kg of N excreted). Therefore, a compromise between profitability and environmental quality was achieved at a dietary RDP level of 11.7% of DM. Observed microbial NAN flow and RDP supply were higher and RUP flow was lower than those predicted by the NRC (2001) model. The NRC (2001) model overpredicted production responses to RUP compared with the results in this study. Replacing default NRC degradation rates for protein supplements with rates measured in vivo resulted in similar observed and predicted values, suggesting that in situ degradation rates used by the NRC are slower than apparent rates in this study. 相似文献

Mixed model analysis of data from 32 studies (122 diets) was used to evaluate the precision and accuracy of the omasal sampling technique for quantifying ruminal-N metabolism and to assess the relationships between nonammonia-N flow at the omasal canal and milk protein yield. Data were derived from experiments in cattle fed North American diets (n = 36) based on alfalfa silage, corn silage, and corn grain and Northern European diets (n = 86) composed of grass silage and barley-based concentrates. In all studies, digesta flow was quantified using a triple-marker approach. Linear regressions were used to predict microbial-N flow to the omasum from intake of dry matter (DM), organic matter (OM), or total digestible nutrients. Efficiency of microbial-N synthesis increased with DM intake and there were trends for increased efficiency with elevated dietary concentrations of crude protein (CP) and rumen-degraded protein (RDP) but these effects were small. Regression of omasal rumen-undegraded protein (RUP) flow on CP intake indicated that an average 32% of dietary CP escaped and 68% was degraded in the rumen. The slope from regression of observed omasal flows of RUP on flows predicted by the National Research Council (2001) model indicated that NRC predicted greater RUP supply. Measured microbial-N flow was, on average, 26% greater than that predicted by the NRC model. Zero ruminal N-balance (omasal CP flow = CP intake) was obtained at dietary CP and RDP concentrations of 147 and 106 g/kg of DM, corresponding to ruminal ammonia-N and milk urea N concentrations of 7.1 and 8.3 mg/100 mL, respectively. Milk protein yield was positively related to the efficiency of microbial-N synthesis and measured RUP concentration. Improved efficiency of microbial-N synthesis and reduced ruminal CP degradability were positively associated with efficiency of capture of dietary N as milk N. In conclusion, the results of this study indicate that the omasal sampling technique yields valuable estimates of RDP, RUP, and ruminal microbial protein supply in cattle. 相似文献

Ten ruminally cannulated lactating Holstein cows that were part of a larger trial studying the effects of feeding different proteins on milk production were used in a replicated 5 x 5 Latin square to quantify flows of microbial and rumen-undegradable protein (RUP) in omasal digesta. Cows were fed total mixed rations containing (dry matter basis) 44% corn silage, 22% alfalfa silage, 2% urea, and 31% concentrate. The basal diet contained 31% high-moisture corn; equal N from one of four protein supplements was added to the other diets at the expense of corn: 9% solvent soybean meal (SSBM), 10% expeller soybean meal (ESBM), 5.5% blood meal (BM), and 7% corn gluten meal (CGM). Omasal sampling was used to quantify total AA N (TAAN) and nonammonia N (NAN) flows from the rumen. Estimates of RUP were made from differences between total and microbial N flows, including a correction for RUP in the basal diet. Modifying a spectrophotometric assay improved total purine recovery from isolated bacteria and omasal samples and gave estimates of microbial TAAN and NAN flows that were similar to a standard HPLC method. Linear programming, based on AA patterns of the diet and isolated omasal bacteria and ruminal protozoa, appeared to overestimate microbial TAAN and NAN flows compared to the purine assays. Yields of microbial TAAN and NAN determined using any method was not affected by diet and averaged 32 to 35 g NAN per kilogram of organic matter truly digested in the rumen. On average, National Research Council (NRC) equations underpredicted microbial N flows by 152 g/d (vs. HPLC), 168 g/d (vs. spectrophotometry), and 244 g/d (vs. linear programming). Estimates of RUP (means from the HPLC and spectrophotometric methods) were: SSBM, 27%, ESBM, 45%, BM, 60%, and CGM, 73%. Generation of ammonia from nitrate reduction is slower compared with urea hydrolysis and may be more efficiently incorporated into ruminal microbial protein. We hypothesized that nitrate supplementation could increase ammonia incorporation into microbial protein in the rumen compared with urea supplementation of a low-protein diet fed to lactating dairy cows. Eight multiparous Chinese Holstein dairy cows were used in a crossover design to investigate the effect of nitrate or an isonitrogenous urea inclusion in the basal low-protein diet on rumen fermentation, milk yield, and ruminal microbial community in dairy cows fed a low-protein diet in comparison with an isonitrogenous urea control. Eight lactating cows were blocked in 4 pairs according to days in milk, parity, and milk yield and allocated to urea (7.0 g urea/kg of dry matter of basal diet) or nitrate (14.6 g of NO3?/kg of dry matter of basal diet, supplemented as sodium nitrate) treatments, which were formulated on 75% of metabolizable protein requirements. Nitrate supplementation decreased ammonia concentration in the rumen liquids (?33.1%) and plasma (?30.6%) as well as methane emissions (?15.0%) and increased dissolved hydrogen concentration (102%), microbial N (22.8%), propionate molar percentage, milk yield, and 16S rRNA gene copies of Selenomonas ruminantium. Ruminal dissolved hydrogen was positively correlated with the molar proportion of propionate (r = 0.57), and negatively correlated with acetate-to-propionate ratio (r = ?0.57) and estimated net metabolic hydrogen production relative to total VFA produced (r = ?0.58). Nitrate reduction to ammonia redirected metabolic hydrogen away from methanogenesis, enhanced ammonia incorporation into rumen microbial protein, and shifted fermentation from acetate to propionate, along with increasing S. ruminantium 16S rRNA gene copies, likely leading to the increased milk yield. 相似文献

Effect of diet fermentability on efficiency of microbial N production was evaluated. Eight ruminally and duodenally cannulated Holstein cows (55 +/- 15.9 days in milk; mean +/- SD) were used in a duplicated 4 x 4 Latin square design with a 2 x 2 factorial arrangement of treatments. Experimental diets contained either ground high moisture corn (HM) or dry ground corn (DG) at two dietary starch concentrations (32 vs. 21%). All diets were formulated for 18% CP, and the sources of dietary protein were alfalfa silage (50% of forage at DM basis), soybean meal, distillers grain, and blood meal. The amount of OM truly fermented in the rumen varied from 7.7 (DG at 21% dietary starch) to 11.3 kg/d (HM at 32% dietary starch) among treatments, and was greater for high starch diets and HM treatments compared with low starch diets and DG treatments, respectively. Microbial N flow was greater for high starch diets compared with low starch diets, but was not affected by corn grain treatment. Microbial efficiency was lower for HM compared with DG treatment (39.7 vs. 48.4 g of microbial N/kg of true ruminally degraded OM), but was not affected by dietary starch concentration. Microbial efficiency was positively correlated with rate of passage for OM and starch (r = 0.77 and 0.75, respectively). Rapid passage rate may have decreased microbial turnover in the rumen, enhancing microbial efficiency. Microbial efficiency was negatively correlated with rate of starch digestion (r = -0.55), consistent with the energy spilling theory. However, energy spilling did not appear to be from lack of ammonia or low ruminal pH. Microbial efficiency was not related to ruminal ammonia concentration, daily mean ruminal pH, or minimum ruminal pH. Rate of starch availability and rates of passage for starch and OM from the rumen are important determinants of efficiency of microbial protein synthesis in vivo. 相似文献

An experiment was conducted to quantify the effects of incremental levels of heat-moisture-treated canola meal (TCM) fed to dairy cows on the relationship between ruminal nutrient digestion and milk production. Experimental diets were fed to 4 multiparous rumen-cannulated Nordic Red cows, averaging (mean ± standard deviation) 681 ± 54.8 kg of body weight, 111 ± 16 d in milk, and 29.1 ± 9.1 kg of milk/d at the start of the study, in a Latin square design with four 21-d periods. The 4 experimental dietary treatments consisted of a basal diet of grass silage and crimped barley, and 3 diets in which the crimped barley was replaced with TCM, giving 3 incremental levels of protein supplementation. Nutrient flow was quantified by the omasal sampling technique using 3 markers (Cr, Yb, and indigestible neutral detergent fiber). Continuous infusion of 15N was used to label bacterial crude protein. Additionally, ruminal sampling and evacuations and measurements of total-tract digestibility were conducted. The experimental diets provided 132, 148, 164, and 180 g of crude protein/kg of dry matter. The increased level of TCM linearly increased dry matter intake from 15.1 to 16.6 kg/d and energy-corrected milk yield from 21.0 to 25.6 kg/d. The increased proportion of TCM when substituting barley with TCM was associated with greater total-tract digestibility of neutral detergent fiber and potentially digestible neutral detergent fiber, which could be explained by increased digestion rate of potentially digestible neutral detergent fiber. Omasal flow of nonammonia N naturally increased with greater dietary TCM inclusion, but the increased intestinal supply of rumen-undegradable protein was partly offset by diminished microbial protein synthesis when feeding more TCM. This was also reflected in a decreased proportion of milk protein from ruminal bacterial protein when TCM supplementation increased. 相似文献
