The purpose of this study was to evaluate the effect of breed, stage of lactation, and health status of the udder on the plasmin-plasminogen system in ovine milk. A total of 38 ewes were used from 3 breeds [Boutsiko (n = 12), Chios (n = 12), and a synthetic breed (50% Boutsiko, 25% Arta, and 25% Chios, n = 14)] with major differences in their genetic potential with respect to milk yield. Milk samples were collected every 2 wk throughout the lactation period and were analyzed for fat, protein, lactose, and somatic cell count (SCC). In addition, milk plasmin (PL), plasminogen (PG), and plasminogen activator (PA) activities were determined. The Chios breed had the greatest average daily milk yield, the synthetic breed had an intermediate milk yield, and ewes of the Boutsiko breed had the lowest milk yield. Milk samples obtained from the Boutsiko breed had similar PL and PA activities, compared with those obtained from the other 2 breeds. The ratio of PG:PL was less in milk samples from the Boutsiko breed compared with the other 2 breeds, indicative of an increased rate of conversion of PG to PL for this breed. There was no correlation between PL activity and daily milk yield in ewes from all 3 breeds. Activities of PL, PG, and PA were greater in ovine milk with elevated SCC (>300,000/mL) compared with activities in milk with low SCC (<300,000/mL). The ratio of PG:PL was less in the high-SCC group compared with the low-SCC group, which indicates an increased rate of conversion of PG to PL for the high-SCC group. There was a decrease in PG and PA activities as well as in the PG:PL ratio in late lactation milk (mo 5 to 6) when compared with early or mid lactation milk (mo 1 to 4). Thus, the PL-PG system is affected by breed, stage of lactation, and the health status of the udder. No relationship was found between PL activity and daily milk yield in the 3 Greek dairy sheep breeds. Plasmin is not a marker for gradual involution in the Greek sheep breeds studied.  相似文献

Thirty-five multiparous Holstein cows were used to determine the role of mitochondrial carnitine palmitoyltransferase I (CPT I) in liver on peripartal adaptations of fatty acid metabolism. From dry-off to parturition, cows were fed a diet at either ad libitum (n = 17) or restricted intake (RI, 80% of calculated requirements for net energy; n = 18). After parturition, all cows were fed a lactation diet. At 4 d in milk (DIM), cows underwent a physical examination and were classified as healthy (n = 15) or having at least one periparturient disorder (PD; n = 17). Cows in the healthy group were assigned to either a control (n = 6) group or a ketosis induction (KI; n = 9) group. Cows with periparturient disorders were assigned to a third (PDC; n = 17) group. Cows in control and PDC groups were fed for ad libitum intake. Cows in KI were fed at 50% of their respective intake at d 4 postpartum starting from 5 DIM and continuing to signs of clinical ketosis or until 14 DIM; cows then were returned to ad libitum intake. Liver was biopsied at −30 d, 1 d, at signs of clinical ketosis or 14 d, and 28 d relative to parturition. Mitochondria were isolated by differential centrifugation. Activity of CPT I was 5.4 and 7.6 nmol of palmitoylcarnitine formed per min/mg of protein for ad libitum and RI, respectively, at −30 DIM. Sensitivity of CPT I to its inhibitor, malonyl CoA, did not differ between ad libitum and RI cows. Differences in CPT I activity between ad libitum and RI were no longer significant at 1 DIM. Postpartum CPT I activity and malonyl CoA sensitivity at 1 DIM, onset of clinical ketosis or 14 DIM, and 28 DIM were not affected by prepartum intake (ad libitum vs. RI), postpartum health status (healthy vs. PD), or ketosis induction status (control vs. KI vs. PDC). Activity of CPT I was positively correlated with liver total lipid, liver triglyceride, liver triglyceride to glycogen ratio, and serum nonesterified fatty acids. Activity of CPT I and dry matter intake were not correlated. Prepartum intake affected prepartum CPT I activity but not malonyl CoA sensitivity. Neither induction of primary ketosis nor periparturient disorders greatly affected CPT I activity or sensitivity, which indicates that alterations of CPT I may not be a major factor in the etiology of primary ketosis or other periparturient disorders.  相似文献

The objective of this study was to determine the effect of partial replacement of forage neutral detergent fiber (NDF) with by-product NDF in close-up diets of dairy cattle on periparturient metabolism and performance. Holstein cows (n = 45) and heifers (n = 19) were fed corn silage-based diets containing 1) 30% oat hay, or 2) 15% oat hay and 15% beet pulp from d −21 relative to expected parturition until parturition. After parturition, all animals received the same lactation diet. Animals were group-fed from d −21 to −10 relative to expected parturition and fed individually from d −10 until 14 d in milk. Animals were required to have at least 5 d of prepartum dry matter intake (DMI) data to remain on the study. Data were analyzed as a randomized design and subjected to ANOVA using the MIXED procedure of SAS. Close-up diet did not affect DMI, total tract nutrient digestibility, energy balance, or serum content of nonesterified fatty acids and β-hydroxybutyrate during the last 5 d prepartum. Prepartum body weight and body condition score were similar between treatments. There was no carryover effect of close-up diet on DMI, energy balance, milk yield, body weight, body condition score, or serum content of nonesterified fatty acids and β-hydroxybutyrate during the first 14 d in milk. In summary, partial replacement of forage NDF (oat hay) with by-product NDF (beet pulp) did not affect periparturient metabolism or performance.  相似文献

The effects of milk yield at dry-off (DO), different calving intervals (CI; 12 and 15 mo) and breed on metabolism and udder health were studied in 56 primiparous and multiparous cows of the Swedish Red and White (SRB) and Swedish Holstein (SH) breeds. The cows were dried off 55 ± 5 d prior to expected parturition. They were fed 4 kg of DM as silage and wheat straw ad libitum for 5 d, and were milked in the morning of d 2 and 5. Depending on their daily milk yield, the cows were divided into 3 numerically equal groups on 2 d during the week prior to DO: low (LY; 5.0 to 11.4 kg of milk/d, n = 19), medium (MY; 11.5 to 17.7 kg of milk/d, n = 19), and high (HY; 17.8 to 29.5 kg of milk/d, n = 18). The plasma cortisol concentration increased during DO only in MY and HY cows. Plasma nonesterified fatty acids increased during DO in all groups, but the maximum nonesterified fatty acid concentration was related to the milk yield prior to DO. The plasma glucose level during the DO period was not significantly affected by yield, but the insulin concentration decreased after DO, with a more pronounced drop in the HY group. The CI 15-mo group had a higher glucose level and tended to have a higher insulin level in plasma than the CI 12-mo group before DO. They also had a higher body condition than the CI 12-mo group. The results indicate that the CI 15-mo cows had a more positive nutrient balance. There were no effects of CI on milk production or composition during DO. The SRB and SH breeds did not differ in any of the measured plasma parameters or milk production. However, the lower somatic cell counts in SRB than in SH observed before and during DO, as well as after parturition, were attributed to being an effect of breed. The proportion of cows with intramammary infections (IMI) was significantly lower just after calving in the LY group than in the other yield groups. At 2 and 3 wk after DO, significantly fewer cows in the LY group had open teat canals compared with the HY and MY groups, respectively, but teat-end condition did not differ between yield groups. The yield before DO did not significantly influence the somatic cell counts during the first 4 wk after parturition or the presence of IMI 4 wk after parturition. We concluded that in the present study, higher milk yield prior to DO gave rise to a more pronounced metabolic response and a higher risk of contracting IMI during the dry period, at calving, or both, but yield at DO did not have any long-term effects on udder health. A prolonged CI did not facilitate a rapid decrease in milk production. The SRB and SH breeds responded equally in decreasing the milk production during DO, but the SRB breed had lower somatic cell counts.  相似文献

Effect of breed on carcass measurements and meat quality characteristics were investigated by using 46 lambs from Turkish Merino, Ramlic, Kivircik, Chios and Imroz breeds. Chios and Imroz carcasses had smaller values for carcass quality characteristics. Breed had no significant effect on pH at 45 min and 24 h post-mortem, water holding capacity and cooking loss. Kivircik and Imroz lambs had lower Warner Bratzler shear force values than those of Ramlic and Turkish Merino lambs (P < 0.01). Meat samples from Kivircik lambs had the highest redness value. Differences among breeds for sensory characteristics, except tenderness were not significant. Tenderness scores given to meat samples of Kivircik lambs were significantly higher (P < 0.01) than those of Turkish Merino, Ramlic and Imroz lambs. Indigenous Kivircik breed, which had high carcass quality as those of improved breeds, might be considered for production of better quality meat in Marmara Region of Turkey.  相似文献

Data from multiparous Holstein cows (n = 43) were used to determine whether supplementation of anions to low-potassium (K) prepartum diets would improve periparturient energy and macromineral status and affect performance during the postpartum period. Beginning 21 d before expected parturition, cows were fed a control diet (1.29% K; +10 mEq/100 g; n = 21) or a low dietary cation-anion difference (DCAD) diet (1.29% K; −15 mEq/100 g; n = 22) with anions provided through a combination of sulfate from calcium sulfate dihydrate (0.40% S total ration) and chloride (1.17% Cl total ration) from SoyChlor 16-7 (West Central, Ralston, IA). All cows were fed the same postpartum diet from parturition through 63 d postpartum. Feeding anions decreased overall urine pH (8.17 vs. 6.70) during the prepartum period. Overall, peripartum concentrations of plasma Ca, P, and Mg were similar between treatments; however, concentrations of plasma Ca tended to be increased during the first 24 h postcalving in cows fed the low DCAD diet. Overall, concentrations of plasma P tended to be increased by feeding the anionic diet prepartum; this effect was more pronounced during the immediate peripartal period. Anionic supplementation did not affect incidence of clinical (<5 mg/dL) and subclinical (5 to 8 mg/dL) hypocalcemia, clinical hypophosphatemia (<2 mg/dL), or clinical (<1.1 mg/dL) and subclinical (1.1 to 1.8 mg/dL) hypomagnesemia. Nevertheless, subclinical hypophosphatemia (2 to 4 mg/dL) tended to be decreased at 16 h postcalving and was decreased at d 2 postpartum for cows fed the anionic diet prepartum. Anion supplementation decreased prepartum dry matter intake (15.6 vs. 14.4 kg/d), but did not affect postpartum dry matter intake (22.4 vs. 23.0 kg/d), milk yield (46.5 vs. 46.1 kg/d), or content and yield of milk fat and true protein. Plasma concentrations of energy-related metabolites (glucose, nonesterified fatty acids, β-hydroxybutyrate) were similar for both groups during the prepartum and postpartum periods. Glucose rate of appearance was determined by continuous infusion of 6,6-dideuterated glucose in a subset of cows between 6 and 10 d prepartum (control, n = 12; low DCAD, n = 9) and 7 and 10 d postpartum (control, n = 9; low DCAD, n = 8) periods. Glucose rate of appearance was not affected by treatment during the prepartum or postpartum periods. Overall, anion supplementation of low K diets improved P status during the early postpartum period, but did not affect aspects of energy metabolism or periparturient performance.  相似文献

A total of 25,160 milk test-day records from 2,516 cows in first lactation of 3 dairy cattle breeds [Simmental (n = 1,900), Brown Swiss (n = 444), and Tyrol Grey (n = 172)] in Kosovo were analyzed using nested repeatability and random regression test-day models with varying (co)variance structures. The different models were compared based on likelihood-based criteria. The best model was a second-order random regression model, with heterogeneous cow variance per breed and heterogeneous residual variance per lactation month and breed, which was used for further analysis. The highest milk production was found in Brown Swiss, followed by Simmental and Tyrol Grey. Substantial breed differences were found for the trajectories of cow and residual variances by month of lactation, with the highest variances found for Brown Swiss, followed by Simmental and Tyrol Grey. High cow and residual variances indicated a high degree of environmental sensitivity on the macro- and microenvironmental levels, respectively. Thus, these results indicate increased environmental sensitivity for breeds with higher genetic potential for milk production. These results support the conclusion that dairy cattle production under the current environmental conditions of Kosovo should be based on a breed with moderate production that is robust to the diet offered (e.g., Tyrol Grey).  相似文献

Dairy cows often experience decreased immune function around the time of calving, typified by impaired polymorphonuclear neutrophil (PMN) function and a transient neutropenia. This is associated with increased disease incidence, including mastitis, retained placenta, and metritis. In an attempt to improve PMN functional capacity during the periparturient period, we injected cows with recombinant bovine granulocyte colony-stimulating factor covalently bound to polyethylene glycol (PEG rbG-CSF) twice subcutaneously, about 6 d before calving and within 24 h after calving. Twenty-one cows in their second pregnancy were enrolled in this study and divided into 2 groups: PEG rbG-CSF treated (n = 11) and saline-treated controls (n = 10). The PMN numbers quickly and dramatically increased after PEG rbG-CSF administration and remained elevated through the end of the experiment (13 d after calving). Exocytosis of myeloperoxidase by stimulated PMN, which is generally decreased in periparturient cows, was markedly increased by PEG rbG-CSF after injection. Higher myeloperoxidase exocytosis persisted for at least 10 d after calving. The PMN superoxide anion release and phagocytosis activity did not differ between groups. Injection of PEG rbG-CSF was safe for cows, with no significant negative effects observed. The greatest single effect of PEG rbG-CSF administration was a dramatic increase in circulating numbers of PMN. The increased numbers of PMN ready to move to a site of infection early in the course of an infection may improve the ability of the cow to ward off clinical disease in the periparturient period.  相似文献

Multiparous Holstein cows (n = 61) were used to determine the effects of chromium propionate (Cr-Pro) supplementation during the periparturient period and early lactation on metabolism, performance, and the incidence of cytological endometritis (CE). After a 1-wk preliminary period, cows were assigned randomly to 1 of 2 treatments from 21 d before expected calving through 63 d postpartum: (1) control (n = 31) and (2) Cr-Pro (n = 30) administered by daily topdress at a rate of 8 mg/d of Cr. A tendency was detected for increased dry matter intake (DMI) during the prepartum period for cows fed Cr-Pro. Moreover, cows fed Cr-Pro tended to have lower plasma concentrations of nonesterified fatty acids during the prepartum period. However, effects of Cr-Pro supplementation on postpartum DMI and milk yield were not significant. Cows fed Cr-Pro tended to have higher urea N concentrations in milk. An interaction of treatment and day existed during the postpartum period, such that cows fed Cr-Pro had lower plasma glucose concentrations within the first day postpartum compared with controls. Plasma haptoglobin concentration was not affected by treatment during the postpartum period. Blood neutrophil glycogen concentrations were not affected by treatment when sampled at either 7 d postpartum or on one day between 40 and 60 d (48 d ± 0.44 standard error) postpartum. Evaluation of endometrial cytology by low volume lavage at 7 d postpartum (first lavage) and on one day between 40 and 60 d (second lavage) postpartum revealed that cows fed Cr-Pro tended to have a higher percentage of neutrophils at first lavage and decreased incidence of CE as assessed at second lavage. In conclusion, supplementation with Cr-Pro resulted in trends for increased DMI and lower plasma nonesterified fatty acids prepartum. Postpartum production and energy metabolism were not affected by treatment; however, Cr-Pro supplementation tended to affect the postpartum influx of neutrophils into the uterus and decreased the incidence of CE, suggesting positive effects of Cr-Pro supplementation on uterine health.  相似文献

Heat stress (HT) and photoperiod affect milk production and immune status of dairy cows. The objective was to evaluate the effects of HT abatement prepartum under controlled photoperiod on hepatic metabolic gene expression and cellular immune function of periparturient Holstein cows (n = 21). Cows were dried off 46 d before expected calving date and assigned to treatments by mature equivalent milk production. The treatments were 1) HT and 2) cooling (CL), both imposed during a photoperiod of 14L:10D. Rectal temperature was measured twice daily, whereas respiration rate was measured 3 times/wk at 1500 h during the entire dry period. After calving, cows were housed in a freestall barn with cooling, and milk yield was recorded daily up to 140 d in milk. Liver samples were taken at dry off, −20, 2, and 20 d relative to calving by biopsy. Under a similar schedule, neutrophil function was determined in blood of cows on HT (n = 12) and CL (n = 9). Blood samples were taken on −46, −32, −18, 0, 14, 28, and 42 d relative to calving for measurement of metabolites and were collected twice daily from −7 to 2 d relative to calving for prolactin (PRL) analysis. The HT cows had greater concentrations of PRL at 0 d relative to calving (150 vs. 93; SEM = 11 ng/mL) and had higher afternoon rectal temperatures (39.4 vs. 39.0; SEM = 0.04°C) and elevated respiration rates (78 vs. 56; SEM = 2 breaths/min) during the prepartum period compared with CL cows. Relative to HT cows, CL cows had greater hepatic expression of PRL-R, SOCS-3, and CAV-1 mRNA. Neutrophil oxidative burst was greater in CL cows relative to HT cows at 2 d (61 vs. 42; SEM = 6%) and at 20 d (62 vs. 49; SEM = 5%) relative to calving, and phagocytosis was greater in CL cows at 20 d (47 vs. 33; SEM = 4%) relative to calving compared with HT cows. Humoral response, as measured by IgG secretion against ovalbumin challenge, was greater for CL cows at −32 d (0.44 vs. 0.33; SEM = 0.05 OD) and −21 d (0.60 vs. 0.50 ± 0.04 OD) relative to calving compared with HT cows. These results suggest that HT abatement during the dry period improved innate and acquired immune status as measured by neutrophil function and immunoglobulin secretion against ovalbumin challenge, and altered hepatic gene expression related to PRL signaling in the periparturient period or subsequent lactation.  相似文献

The objective of the study was to evaluate the relationships between body condition and lymphocyte functions in periparturient dairy cows. Thirty days before expected calving, 21 Holstein cows were categorized as thin (n = 6), medium (n = 8), or overconditioned (n = 7) based on body condition score (BCS). Blood samples were collected on 21, 14, 7, and 3 d before calving and on d 3, 7, 14, 21, 28, and 35 after parturition. An aliquot of blood was used to determine plasma nonesterified fatty acids (NEFA) and glucose. At 14 and 7 d before, and 14 and 35 d after calving, a second aliquot of blood was used to assess peripheral blood mononuclear cell (PBMC) functions: DNA synthesis, immunoglobulin (Ig) M, and interferon-gamma (IFN-γ) secretion after mitogen stimulation. During the experiment, all 21 cows showed a decline in BCS. Overconditioned cows lost significantly more BCS than thin cows. After calving, overconditioned cows had higher plasma NEFA compared with thin and medium cows. Conversely, plasma glucose never differed between the 3 categories of cows. Regardless of BCS, DNA synthesis and IgM secretions were significantly lower in PBMC isolated on 7 d before calving compared with those recorded 14 and 35 d after parturition. Conversely, PBMC from the 21 cows did not show any change of IFN-γ secretion during the experimental period. Taking into consideration the BCS categories, PBMC isolated from overconditioned cows presented lower IgM secretion compared with thin cows on d 14 and 35 after calving. Furthermore, PBMC isolated from overconditioned cows secreted less IFN-γ compared with thin and medium cows on d 7 before calving. The DNA synthesis of PBMC stimulated with the 3 mitogens did not differ between the 3 categories of cows. In conclusion, immunodepression occurring in cows around calving would be particularly evident in overconditioned cows.  相似文献

The objectives were to evaluate the effects of differential supplementation of Ca salts (CS) of fatty acids (FA) on plasma acute phase proteins and both FA composition and function (i.e., activity and cytokine production) of neutrophils, during the peripartum and breeding periods. Holstein cows were assigned randomly to receive either CS of palm (PO) or safflower (SO) oils from 30 d prepartum until 35 d postpartum (dpp) and CS of PO or fish oil (FO) from 35 to 160 dpp. Supplementation of CS of FA was at 1.5% of dietary dry matter. Cows (n = 32) were sampled three times weekly from parturition to 35 dpp for analyses of plasma concentrations of haptoglobin and fibrinogen. Cows (n = 47) were sampled for neutrophil phagocytic and oxidative burst activities toward Escherichia coli and Staphylococcus aureus, and neutrophil abundances of L-selectin and β2-integrin assessed by flow cytometry at 32 d prepartum, within 7 h after parturition, and 4 and 7 dpp. Profiles of FA in neutrophils and cytokine production (i.e., tumor necrosis factor alpha, TNF-α, and IL-1β) were assessed prepartum (n = 14), 35 (PO vs. SO; n = 26) and 85 dpp (PO vs. FO; n = 28). Plasma concentrations of haptoglobin and fibrinogen were greater for cows fed SO compared with PO. The percentage of neutrophils with phagocytic and oxidative burst activities was not affected by transition diets, but activities per neutrophil were greater in SO compared with PO diets at 4 (phagocytosis and oxidative burst) and 7 dpp (oxidative burst). Neutrophil abundance of L-selectin, but not β2-integrin, was greater in SO compared with PO at 4 and 7 dpp. Neutrophil productions of TNF-α and IL-1β were increased at 35 dpp in SO compared with PO diets, but production of TNF-α was attenuated in FO compared with PO at 85 dpp. Neutrophil ratios of n-6:n-3 FA were greater at 35 dpp in the SO diet and less at 85 dpp in FO compared with PO diets. In conclusion, cows supplemented with CS of SO had improved innate immunity (i.e., acute phase response and neutrophil function) to better cope with the bacterial challenges in the postpartum period. Conversely, CS of FO attenuated neutrophil cytokine production.  相似文献

Thiazolidinediones (TZD) are potent, synthetic ligands for peroxisome proliferator activated receptor-gamma (PPAR-γ) that reduce plasma nonesterified fatty acids (NEFA) and potentiate the action of insulin in peripheral tissues of several species. Holstein cows (n = 9) entering their second or greater lactation were used to determine whether late prepartum administration of TZD would affect periparturient metabolism and milk production. Cows were limit-fed a total mixed ration (TMR) during the prepartum period to provide no more than 130% of predicted energy requirements. During the postpartum period cows were fed a common TMR for ad libitum intake. Cows were administered either 2,4-TZD (4.0 mg/kg of body weight) or saline (control) by intrajugular infusion once daily from 25 d before expected parturition until parturition. Plasma samples were collected daily from 26 d before expected parturition through 7 d postpartum. Plasma NEFA concentrations decreased during the prepartum period (d −21 to −1; 70 vs. 83 ± 4 μEq/L) and tended to be decreased during the peripartum period (d −7 to d +7; 113 vs. 205 ± 32 μEq/L) due to prepartum TZD administration. Plasma concentrations of glucose were not affected by treatment; however, plasma β-hydroxybutyrate concentrations decreased in TZD-treated cows (8.6 vs. 10.7 ± 1.7 mg/dL) as parturition approached, and plasma insulin concentrations increased during the peripartum period (0.65 vs. 0.38 ± 0.07 ng/mL). Postpartum liver triglyceride and glycogen content was not affected by treatment. Prepartum TZD administration tended to increase dry matter intake during the peripartum and postpartum periods (16.6 vs. 14.6 ± 0.8 kg/d and 20.0 vs. 17.2 ± 1.2 kg/d, respectively). Milk yield for the first 30 d postpartum and milk composition measured on d 8 postpartum were not affected by treatment. There was no effect of prepartum TZD administration on insulin-dependent glucose utilization assessed using insulin challenge during either the prepartum or postpartum periods. These results suggest that administration of TZD during the late prepartum period has the potential to improve metabolic health and DMI of periparturient dairy cows and warrants further investigation.  相似文献

Mature Holstein cows were drenched daily with either 500 mL of water (control; n = 28) or propylene glycol (PPG; n = 28) from d 10 before parturition until d 25 postpartum. Follicular development was monitored thrice weekly by transrectal ultrasound. Blood samples were collected every 30 min from a subset of 10 cows per treatment on d −10, 2, and 25 to assess glucose and insulin response to treatments, and on d 10 postpartum, blood was collected every 10 min for 12 h to determine LH pulse profiles. Both insulin and glucose were elevated on d 2 and 25 following PPG administration, but only insulin was elevated on d −10. On d 10 postpartum, the number of LH pulses, mean LH, and pulse amplitude were not different between control and PPG cows. The proportion of first postpartum dominant follicles that became ovulatory, atretic, or cystic was not different between control and PPG cows. Despite evidence of improved metabolic status, PPG failed to increase LH pulse frequency, and failed to increase the proportion of first postpartum follicle waves resulting in ovulation. The dominant follicle of each cow was retrospectively categorized as being ovulatory (n = 17), nonovulatory high estradiol (n = 6), nonovulatory low estradiol (n = 24), or cystic (n = 8). Differences in dry matter intake and energy balance among cows in the different follicle categories were apparent as early as 3 wk before parturition. Neutrophil function is reduced in periparturient dairy cows. Possible factors that reduce neutrophil function include endocrine changes associated with parturition and metabolic stresses associated with lactogenesis. In this study, mastectomized and intact cows were studied to specifically examine the effects of lactogenesis on neutrophil function in periparturient cows. Expression of adhesion molecules on neutrophils (L-selectin, mediating capture and rolling adhesion, and beta 2-integrin, mediating tight adhesion vital to egress) and neutrophil myeloperoxidase activity (an index of bactericidal activity) were assessed in mastectomized and intact cows. Expression of L-selectin decreased at parturition followed by rapid recovery to prepartum values in both intact and mastectomized cows. Expression of beta 2-integrins increased in intact cows at parturition but not in mastectomized cows. Expression of beta 2-integrins was greater in intact cows than in mastectomized cows throughout the study. Neutrophil myeloperoxidase activity decreased from baseline prepartum values as parturition approached in both intact and mastectomized cows, which suggests the endocrine changes associated with the act of parturition are predominant factors causing loss of neutrophil function. Myeloperoxidase activity recovered to prepartum values within a week of parturition in mastectomized cows; however, myeloperoxidase activity remained depressed in neutrophils obtained from intact cows throughout the first 20 d of lactation. The presence of the mammary gland and its attendant metabolic stresses slowed recovery of neutrophil function, which suggests that the metabolic stress of lactation exacerbated periparturient immunosuppression.  相似文献

The objectives of this study were: 1) to determine the effect of providing additional prepartum concentrate on the occurrence and severity of ruminal acidosis (RA) and lactational performance during the periparturient period in primiparous cows; and 2) to characterize the occurrence and severity of RA during the periparturient period. We hypothesized that providing additional concentrate prepartum would reduce postpartum RA. Fourteen ruminally cannulated Holstein heifers were paired by expected calving date and body condition score. The heifers were assigned to 1 of 2 prepartum feeding regimens: 1) a control treatment consisting of a far-off diet (forage:concentrate, F:C = 80:20) fed from d −60 to d −25 and a close-up diet (F:C = 54:46) fed from d −24 until parturition; or 2) a high-concentrate (HC) feeding program consisting of 4 prepartum diets, HC-1 (F:C = 68:32) fed from d −60 to d −43, HC-2 (F:C = 60:40) fed from d −42 to d −25, HC-3 (F:C = 52:48) fed from d −24 to d −13, and HC-4 (F:C = 46:54) fed from d −12 until parturition. All cows received the same lactation diet postpartum. Ruminal pH was measured continuously from d −5 to d +5, and for 3 consecutive days starting on d +17 ± 1.2, d +37 ± 1.4, and d +58 ± 1.5 relative to parturition using an indwelling ruminal pH system. Ruminal acidosis was considered to occur when ruminal pH was <5.8 (total RA). Ruminal acidosis was further partitioned into: 1) mild RA (5.8 > ruminal pH > 5.5), 2) moderate RA (5.5 > ruminal pH > 5.2), and 3) acute RA (ruminal pH < 5.2). Feeding additional concentrate prepartum did not reduce postpartum RA. In fact, cows fed the HC treatment had more daily episodes of acute RA than cows fed the control treatment. Day relative to parturition affected the occurrence and severity of RA; RA increased following parturition and was sustained thereafter. The DM intake during the last 5 d of gestation was lower for cows fed the HC treatment compared with cows fed the control treatment, but lactational performance was not affected. We conclude that, under the conditions imposed, feeding additional concentrate prepartum does not reduce postpartum RA. Furthermore, the incidence and severity of RA increases immediately postpartum, emphasizing the need to develop and implement feeding strategies that reduce this risk.  相似文献

The objectives of the current study were to investigate the influence of denaverine hydrochloride and carbetocin on softening and dilatation of the birth canal, the need for assistance during parturition, calf mortality, retention of fetal membranes, endometritis, and subsequent fertility. Altogether 200 animals (100 cows and 100 heifers) of the Simmental breed were divided into 2 groups: treatment (n = 100) and control (n = 100). Animals in the treatment group received denaverine hydrochloride and carbetocin (a maximum of twice for each, depending on the progression of labor) during delivery over a maximum of 4 waiting periods (30 min each), whereas control animals experienced the same waiting periods but received no treatment. The treatment protocol had a positive influence on the ease of calving and postpartum reproductive health. The treatment increased the number of animals with the birth canal dilated by more than 25 cm, and halved the need for any assistance at parturition. In addition, treatment decreased the occurrence of difficult calving, the need for episiotomy, the appearance of birth canal lesions, and clinical endometritis. The treatment protocol had an effect throughout the entire puerperal period, as treated animals conceived with fewer artificial inseminations (1.3 vs. 1.6 artificial inseminations/pregnancy) and sooner (67 vs. 78 d open) compared with control animals. Denaverine hydrochloride and carbetocin administered in combination during parturition affected the progression and ease of calving, and thus the welfare of cows in labor and subsequently. However, further studies are needed to confirm the findings and to establish best practices.  相似文献

Acid whey filtrates from the bulk milk of different indigenous greek ovine and caprine breeds were investigated for the quantitative and qualitative characteristics of α-lactalbumin (α-LA) and β-lactoglobulin (β-LG). For comparison reasons acid wheys from bovine milk and from Saanen and Alpine caprine breeds were included. The main characteristic of ovine acid wheys was the low α-LA percentage. The β-LG/α-LA ratio of ovine acid wheys ranged from 3.91 in Chios breed to 6.65 in Boutsiko breed. It was higher than the estimate for bovine acid wheys which ranged from 3.09 to 3.37. The chromatographic and isoelectric focusing profiles of ovine β-LG and α-LA from the different breeds were also variable. The β-LG percentage of caprine acid wheys was lower compared to ovine and bovine acid wheys. Their β-LG/α-LA ratios ranged from 2.02 in Saanen breed to 3.04 in the indigenous breed Skopelos.  相似文献

Bull calves (n = 49), born at the University of Minnesota West Central Research and Outreach Center (Morris) between March and May 2011, were used to compare growth measurements and profitability of conventional and organic dairy steers. Calves were assigned to 1 of 3 replicated groups at birth: conventional (CONV; n = 16), organic (pasture and concentrate; ORG; n = 16), or organic grass only (GRS; n = 17), and analysis of variables was on a pen basis. Breed groups of calves were Holstein (HO; n = 9); Holsteins (n = 11) maintained at 1964 breed average level; crossbreds (n = 19) including combinations of HO, Montbéliarde, and Swedish Red; and crossbreds (n = 10) including combinations of HO, Jersey, Swedish Red, and Normande. The CONV steers were fed a diet of 80% concentrate and 20% forage. The ORG steers were fed a diet of organic corn, organic corn silage, and at least 30% of their diet consisted of organic pasture during the grazing season. The GRS steers grazed pasture during the grazing season and were fed high-quality hay or hay silage during the nongrazing season. Intakes of a total mixed ration were recorded daily with herd management software. A profit function was defined to include revenues and expenses for beef value, feed intake, pasture intake, health cost, and yardage. The GRS (358.6 kg) steers had lesser total gains from birth to slaughter than ORG (429.6 kg) and CONV (534.5 kg) steers. Furthermore, the GRS (0.61 kg/d) steers had lesser average daily gain from birth compared with ORG (0.81 kg/d) and CONV (1.1 kg/d) steers. The GRS and ORG steers had smaller rib eye area (49.5 and 65.8 cm2, respectively) compared with CONV (75.4 cm2) steers. For profitability, GRS steers had 43% greater profit than CONV steers due to organic beef price premiums and lower feed costs. On the other hand, ORG steers had substantially less profit than CONV steers. The higher cost of production for the ORG steers is due to the extreme high value of organic corn. The results of the current study illustrate the economic potential of alternative strategies for growing and marketing male offspring of organic dairy cattle in the Midwest.  相似文献
