The primary objective of this study was to investigate the effects of supplementing a commercial colostrum replacer (CR) with gut active carbohydrates (GAC) on passive transfer of IgG in commercial dairy calves. A secondary objective was to evaluate the effect of treatment on preweaning health and growth. A total of 240 newborn Holstein dairy calves on a commercial dairy farm were enrolled in this study. Newborn heifer and bull calves were weighed and then randomly assigned to either the treated group [GAC: 30 g of GAC mixed into 1.5 doses (150 g of IgG) of commercial colostrum replacer; n = 119] or the control group [CON: 1.5 doses (150 g of IgG) of CR; n = 121]. The assigned CR treatment was fed within 3.5 h of birth using an esophageal tube feeder. Venous blood samples were collected at 0 and 24 h of age and used to measure serum IgG (mg/mL) and serum total protein (g/dL) concentrations and to estimate the apparent efficiency of absorption of IgG (%). The 129 heifers calves enrolled (CON = 60; GAC = 69) were also followed until weaning to assess the effect of GAC addition on preweaning health and growth. Multivariable linear regression showed that the addition of GAC to CR did not influence passive transfer of IgG, as measured by apparent efficiency of absorption at 24 h of age (CON = 54.0 vs. GAC = 54.3%), serum IgG (CON = 20.3 vs. GAC = 20.2 mg/mL), and serum total protein (CON = 5.69 vs. GAC = 5.68 g/dL). Although study sample sizes were not originally derived to evaluate health outcomes, treatment had no effect on weight gain or incidence of health events (diarrhea, pneumonia, mortality) for heifer calves between birth and 7 wk of age.  相似文献

The objective of this study was to describe the effects of feeding 1 or 2 doses of a commercially available colostrum-derived colostrum replacer (CR) on passive transfer of immunoglobulins (Ig) in newborn dairy calves, including IgG, IgG1, IgG2, IgA, and IgM. Newborn calves were removed from the dam before suckling and randomly assigned to 1 of 3 treatment groups: group 1 were fed 1 package (100 g of IgG) of CR product, group 2 was fed 2 packages (200 g of IgG) of the same CR product, and group 3 was fed 3.8 L of maternal colostrum. All colostrum treatments were fed using an esophageal tube feeder within 2 h of birth. Blood samples collected before colostrum feeding and at 24 h of age were tested for serum total protein and Ig concentrations. Mean 24-h serum total protein (TP) and IgG concentrations were significantly lower for calves in group 1 (n = 24; TP = 4.9 g/dL, IgG = 9.6 mg/mL) compared with calves in groups 2 or 3. There was no difference in 24-h serum TP or IgG concentrations between calves in group 2 (n = 23; TP = 5.5 g/dL, IgG = 19.0 mg/mL) and calves in group 3 (n = 22; TP = 5.7 g/dL, IgG = 20.7 mg/mL). Fifty-four, 100, and 91% of calves in groups 1, 2 and 3 achieved acceptable passive transfer (24-h serum IgG ≥10 mg/mL), respectively. Statistically significant but numerically small differences existed between calves in groups 2 and 3 for some 24-h serum Ig classes and subclasses (mean serum concentrations of IgG2, IgA, IgM) and for the relative percentages of Ig classes and subclasses (IgA, IgM, and IgG as a percentage of total Ig; IgG1 and IgG2 as a percentage of total IgG).  相似文献

The objective of this study was first to evaluate whether irradiation treatment of a commercial colostrum replacer (CR) affected acquisition of passive immunity. If the irradiation treatment negatively affected the acquisition of passive immunity, the second objective was to evaluate whether an increased total IgG mass, in a single feeding of CR derived from bovine serum fractions, could compensate for this effect. Acquisition of passive immunity was assessed by 24-h serum IgG levels, serum protein levels, apparent efficiency of absorption (AEA) of IgG, and the ability to prevent failure of passive transfer (FPT) in day-old dairy calves fed a single feeding of CR. Single-dose packs of CR were sent to a commercial irradiation facility for electron-beam irradiation at 3 to 7 kGy (low irradiation) or 15 to 20 kGy (high irradiation). Fifty-six Holstein, Jersey, or crossbred calves were randomly assigned to 1 of 5 treatments: 1) 130 g of IgG (460 g of CR), no irradiation; 2) 130 g of IgG (460 g of CR), low irradiation; 3) 160 g of IgG (518 g of CR), low irradiation; 4) 190 g of IgG (575.4 g of CR), low irradiation; and 5) 130 g of IgG (460 g of CR), high irradiation. All CR were reconstituted in water and mixed in a household blender to a constant solids concentration of 18.7%. Increasing doses of irradiation (130 g of Ig with no, low, or high irradiation) resulted in a linear decrease in 24-h serum IgG and AEA of IgG, and increased the percentage of calves with FPT. Increasing the IgG mass in the CR (130, 160, and 190 g of Ig with low irradiation) resulted in a linear increase in 24-h serum IgG and serum total protein levels, and a linear decrease in AEA of IgG. There was no effect of increasing the mass of IgG fed on the percentage of calves with FPT. The correlation between serum IgG and serum total protein at 24 h was positive; however, at 24 h the irradiation treatments reduced the serum IgG-to-serum total protein ratio. In this study, CR isolated from bovine serum, providing 130 g of IgG in the first feeding and receiving either no irradiation or a low irradiation treatment, was sufficient to prevent FPT in calves.  相似文献

A well-managed colostrum program on farms is the most important step in reducing disease in neonatal calves. In the last few years, colostrum replacers have increased in popularity and are designed to be an alternative to colostrum on farms that have poor colostrum quality, limited colostrum reserves, or to break the cycle of transmission for certain infectious diseases. However, it is important to make sure these products are effective and are capable of providing adequate serum immunoglobulin concentrations. The objective of this study was to evaluate the efficacy of a commercially available colostrum replacer product in dairy calves. Holstein calves from a single dairy were randomly assigned to 1 of 3 groups at birth. Group 1 (n = 21) calves were given 4 quarts of colostrum via esophageal feeder within 3 h of birth and served as the control group for this study. Group 2 (n = 21) received 2 packages of a colostrum replacer product, and group 3 (n = 21) received 3 packages of the colostrum replacer product within 3 h of birth. Blood samples from all calves were collected 24 h after colostrum administration and analyzed for serum total protein and IgG concentrations. Calves fed fresh colostrum had significantly higher serum total protein levels and IgG concentrations compared with calves fed the colostrum replacer product. Calves fed the colostrum replacer also had a significantly higher percentage of calves with failure of passive transfer (serum IgG <1,000 mg/dL). The colostrum replacer product evaluated in this study failed to routinely provide adequate IgG concentrations when fed according to label directions.  相似文献

Newborn Holstein bull calves (n = 32) were assigned to receive a colostrum supplement (CS) containing defibrinated bovine plasma or a colostrum replacer (CR) containing an immunoglobulin concentrate obtained by concentrating the immunoglobulin (Ig)G fraction of bovine plasma. The CS and CR contained 11.1 and 21.2% of dry matter as IgG, respectively. Each animal was fed two 454-g feedings at 1 and 8 h of age. The two feedings of CS and CR provided 95 and 187 g of IgG, respectively. Mean plasma IgG at 24 h of age was 8.0 and 13.6 g/L in calves fed CS and CR, respectively, indicating acceptable absorption of Ig from both sources. Mean apparent efficiency of IgG absorption in calves fed CS and CR were 33 and 30%, respectively, and did not differ between treatments. Mean plasma total protein at 24 h in calves fed CS and CR were 4.99 and 4.98 g/dl and did not differ between treatments. Increased plasma protein concentration from 0 to 24 h (4.5 g/L) was lower than the mean increase in plasma IgG concentration during the same period (10.3 g/L), indicating altered protein profile in the blood during the first 24 h of life. Correlation between plasma IgG and total protein at 24 h of age was significant within treatment, but the relationship between IgG and protein in plasma at 24 h varied between treatments. Predicted plasma total protein concentrations at 10 g of IgG/L of plasma at 24 h were 5.4 and 4.2 g/dl, in calves fed CS and CR, respectively. Prediction of plasma IgG concentration using total plasma protein may be inappropriate when calves are fed CS or CR.  相似文献

Commercially available colostrum replacers (CR) are commonly used when maternal colostrum (MC) is unavailable, for managerial convenience, to ensure quality consistency at first feeding, or in disease control and eradication programs. The objective of this study was to determine the efficacy of feeding First Day Formula (Accelerated Genetics, Baraboo, WI) CR versus pooled MC on immunological status, growth, and health of preweaned dairy calves. A total of 1,220 Jersey and Jersey × Holstein calves born on a California Central Valley dairy farm were assigned after birth to receive either CR or MC following a systematic allocation procedure. Calves assigned to MC were tube fed 2.8 L of MC, and calves assigned to CR were tube fed a total of 500 g of CR (150 g of immunoglobulin G; IgG) mixed into 1.9 L of water at 1 h ± 5 min after the calf was born. A subset of calves was selected for passive transfer (n = 592) and growth (n = 268) analyses. Although both coliform count and total bacteria count were low for MC and CR fed to calves during the study, the predicted probability of calves receiving contaminated liquid feed (coliform count >10,000 cfu/mL) at first feeding was reduced for calves fed CR (1.5%) compared with calves fed MC (6.1%). The mean blood concentration of IgG was lower for calves fed CR than for calves fed MC (19.6 vs. 23.4 mg/mL). However, the apparent efficiency of absorption of IgG did not differ between treatments (34.4 and 35.9% for CR and MC, respectively). Total proteins were lower in calves fed CR compared with MC at 24 h (5.16 vs. 5.84 g/dL, respectively). Calves fed CR were 1.5 kg lighter at weaning and gained 0.03 kg less per day (0.30 vs. 0.33 kg/d, respectively) than calves fed MC before weaning. Height at weaning did not differ between the 2 treatment groups. Calves fed CR tended to have a higher predicted probability of not being treated for diarrhea than calves fed MC (0.142 vs. 0.110, respectively). However, when the disease was present, CR had a higher number of treatment days compared with MC (11.6 vs. 10.8 d, respectively). The hazard ratio of dying did not differ between MC and CR; however, CR calves had a numerically higher risk (hazard ratio = 1.347) of dying compared with calves that received MC. In conclusion, IgG absorption and serum concentration of calves were adequate when calves were fed either CR or MC. The CR-fed calves had a lower probability of receiving contaminated liquid feed and performed similar in terms of health compared with calves receiving high-quality MC, although they were slightly lighter at weaning. Therefore, the CR evaluated in this study is a valid alternative to high-quality (>50 mg of IgG/mL) MC.  相似文献

Neonatal calf health is largely dependent on the ingestion and absorption of maternally derived antibodies via colostrum administration. The objective of this study was to evaluate the efficacy of a commercially available plasma-derived colostrum-replacement (CR) product as compared with bovine colostrum. Holstein calves were removed from the dam immediately after birth and randomly allocated to 1 of 3 groups. Group 1 calves (n = 22) were fed 1 package of the CR product; group 2 calves (n = 22) were fed 2 packages of the CR product; and group 3 calves (n = 22) were fed 3 L of bovine colostrum. Blood samples were collected from all calves 24 h after colostrum or CR feeding and analyzed for serum IgG and total protein concentrations. Calves fed bovine colostrum had significantly higher serum IgG and total protein concentration than calves in either group fed the CR product. Group 1 calves (1 package of CR product) had a significantly higher incidence of failure of transfer of passive immunity than calves in groups 2 or 3. The results of this study indicated that 2 packages of this CR product achieved adequate IgG concentrations in calves. However, calves fed 1 package of this CR product consistently had failure of transfer of passive immunity.  相似文献

Serum total protein (STP) refractometry is a widely used indicator of failed transfer of passive immunity (FTPI), defined as serum IgG concentrations of <10 mg/mL or STP levels <5.2 g/dL measured at 24 h of life. However, recent reports have demonstrated that refractometry could be inaccurate at estimating serum IgG concentrations and FTPI when calves are fed colostrum replacer (CR). The objective of this study was to evaluate the accuracy of STP measurements to estimate FTPI in calves fed CR compared with calves fed maternal colostrum. Blood was collected from dairy calves fed maternal colostrum (n = 927) or colostrum-derived CR (n = 1,258) and analyzed for STP and serum IgG. Serum total protein was measured with a digital refractometer, whereas radial immunodiffusion was used to determine IgG concentrations. Calves fed maternal colostrum had a mean STP of 5.80 ± 0.72 (standard deviation) g/dL and a mean IgG concentration of 22.81 ± 10.14 mg/mL, respectively, whereas calves fed CR had a mean STP and IgG concentration of 5.14 ± 0.50 g/dL and 12.78 ± 4.60 mg/mL, respectively. Rates of FTPI for calves fed maternal colostrum or CR were 4.2% and 27.26%, respectively. Calves were considered to have FTPI if their IgG postcolostrum feeding was <10 mg/mL. Logistic and linear regression analyses were performed to determine cutoff points and existent relationships between STP and IgG. Serum total protein and IgG for calves fed maternal colostrum were highly correlated. In contrast, STP and IgG for calves fed CR were lowly correlated. A receiver operator characteristic curve analysis demonstrated that an STP cutoff point that could predict FTPI when calves are fed CR would be 4.9 g/dL (sensitivity = 0.68; specificity = 0.75). This study suggests that current cutoff points used for STP inflates the number of calves estimated to have FTPI when they are fed CR.  相似文献

The objective of the present study was to compare serum IgG concentration, weight gain, and health characteristics in Irish spring-born dairy calves fed colostrum stored using a range of conditions. Immediately after birth, 75 dairy heifer calves were assigned to 1 of 5 experimental colostrum treatments: (1) fresh pasteurized colostrum, fed immediately after pasteurization; (2) fresh colostrum, fed immediately after collection but not pasteurized; (3) colostrum stored unpasteurized at 4°C in a temperature-controlled unit for 2 d before being fed to calves; (4) colostrum stored unpasteurized at 13°C in a temperature-controlled unit for 2 d before being fed to calves; and (5) colostrum stored unpasteurized at 22°C in a temperature-controlled unit for 2 d before being fed to calves. All colostrum had IgG concentrations >50 g/L and was fed to calves promptly after birth. Blood samples were obtained from calves via the jugular vein at 0 h (before colostrum feeding) and at 24 h of age to determine the rate of passive transfer of IgG; individual calf live-weights were recorded to monitor weight gain (kg/d) from birth to weaning. Colostrum stored in warmer conditions (i.e., 22°C) had >42 times more bacteria present and a pH that was 0.85 units lower and resulted in a serum IgG concentration that was almost 2 times lower compared with colostrum that was pasteurized, untreated, or stored at 4°C for 2 d. Colostrum stored at 4°C for 2 d had more bacteria present than pasteurized and fresh colostrum but did not result in reduced calf serum IgG concentrations. Average daily weight gain from birth to weaning did not differ among treatments. Even if colostrum has sufficient IgG (>50 g/L) but cannot be fed to calves when freshly collected, storage at ≤4°C for 2 d is advisable to ensure adequate passive transfer when it is consumed by the calf.  相似文献

The objective of this study was to show a new function of Se in IgG absorption from colostrum by newborn calves. The same amount and quality of colostrum with or without Se addition was fed to paired calves (n = 60) 4 times at <2, 12, 24, and 36 h after birth, respectively. Four-time feeding of colostrum containing 1.0 ppm Se significantly increased IgG amount in the blood plasma of calves 24 h after birth; however, its effect was small (about 20% increase). Although the addition of 3.0 ppm Se once at the first colostrum feeding was more effective on IgG absorption, its significant effect was a 42% increase on average. The increased IgG concentration of blood plasma continued for about 2 wk. It is known that the absorption of colostrum IgG is mediated by intestinal pinocytosis, which continues for only 24 h after birth. The addition of Se to colostrum might directly activate this physiological pinocytosis of intestinal epithelial cells because of the rapidity of the reaction. This effect is not nutritional but rather pharmacological. Supplemented Se also resulted in its increased concentration in blood plasma. Selenium is an essential mineral for animals; however, newborn calves are always deficient in Se at birth. Application of this method in calves would also provide an immediate supply of Se and might contribute to the development of the immune system of calves. This study showed that Se supplementation to colostrum increased IgG amount and Se concentration in blood plasma in newborn calves.  相似文献

Efficacy of passive transfer of immunity in young calves is commonly assessed using total serum protein (STP) or serum immunoglobulin G (IgG) concentration tested within the first few days of life. To our knowledge, no research has measured changes in these concentrations over this period to establish an appropriate age range for testing. The aim of this study was to monitor changes in STP and serum IgG concentrations from birth until 10 d of age to provide a basis for recommendations for when passive transfer of immunity in dairy calves can be measured. Concentrations of STP and IgG of 12 calves were measured at 11 time points: at approximately 30 min before colostrum feeding, at 24 h after colostrum feeding, and daily from d 2 to 10 of age. Mean (± standard deviation) STP and IgG concentrations were 4.61 ± 0.3 g/dL and 0.6 ± 0.6 mg/mL at birth, 5.83 ± 0.73 g/dL and 22.2 ± 9.6 mg/mL at 24 h after colostrum feeding, and 5.78 ± 0.52 g/dL and 16.1 ± 7.3 mg/mL at d 10 of age, respectively. The IgG concentration declined over subsequent days relative to IgG measured at 24 h at a rate of approximately 0.69 mg/mL per day, declining by 27.6 ± 6.2% (mean ± SD) on d 10. The concentration of STP did not decrease over time. Concentrations of IgG at 24 h after colostrum feeding were highly correlated with each of the measures of IgG over the 10-d period (r ≥0.97). These correlations were supported by the Bland-Altman plots of agreement between the 24-h sample and subsequent samples. Compared with the reference value at 24 h, STP concentrations were highly correlated on d 2 and 3 (r ≥0.98), highly correlated but variable from d 4 to 9 (r ≥0.88), and lower at d 10 (r = 0.76). These results indicate that calves may be reliably tested for passive transfer of immunity using IgG or STP concentrations up to 9 d of age.  相似文献

Forty-two Majorera kids (21 males and 21 females) were assigned to 3 groups, a colostrum group (C), a colostrum whey group (CW), and a colostrum whey plus milk replacer group (CWMR). All kids were fed twice on the first day and received 4 g of IgG/kg of body weight. No differences were found in serum IgG among the different treatments. Kid serum IgG concentrations on d 2 were 14.57, 17.25, and 13.32 mg/mL in the C, CW, and CWMR group, respectively. Labor time per animal was higher in the C and CW treatments than in the CWMR group (24.2 ± 2.3, 20.9 ± 3.4, and 16.1 ± 1.5 min, respectively). This new management system may decrease labor costs during the colostrum feeding period.  相似文献

Fifty-two dairy calves were blocked by birth date and, within each block, randomly assigned to 1 of 4 treatments to investigate the effects of incremental levels of sodium bicarbonate (NaHCO3) on IgG metabolism. Treatments were (1) colostrum replacer (CR) + 0 g of NaHCO3 (control); (2) CR + 15 g of NaHCO3; (3) CR + 30 g of NaHCO3; or (4) CR + 45 g of NaHCO3. Calves were fed colostrum replacer (>200 g of IgG) in one feeding within 45 min of birth (0 h) and 2 L of milk replacer at 12, 24, 36, and 48 h. Only calves born in calving pens from multiparous cows with no dystocia were used in this study. Blood samples were taken at 0, 6, 12, 24, and 48 h postpartum, and serum was analyzed for IgG using radial immunoassay and bicarbonate using spectrophotometry. Feeding increasing levels of sodium bicarbonate had negative linear effects on IgG concentration, IgG apparent efficiency of absorption, and IgG area under the curve, primarily due to the effect of the highest dose of NaHCO3 (45 g). Sodium bicarbonate treatments had no effect on serum bicarbonate concentration. However, area under the curve of serum bicarbonate increased linearly with the amount of NaHCO3 fed.  相似文献

The objective of this study was to investigate the effect of method of feeding (nipple bottle vs. esophageal tube feeder) on passive transfer of immunoglobulin (Ig) G when either a large or small volume of colostrum was fed. Newborn bull calves were removed from the dam before suckling and randomly assigned to 1 of 4 colostrum replacer (CR) treatment groups: 1.5 L (100 g of IgG) of CR fed using a nipple bottle (group 1; n = 24); 1.5 L (100 g of IgG) of CR fed using an esophageal tube feeder (group 2; n = 24); 3.0 L (200 g of IgG) of CR fed using a nipple bottle (group 3; n = 24), or 3.0 L (200 g of IgG) of CR fed using an esophageal tube feeder (group 4; n = 25). Blood samples collected at 24 h of age showed that serum IgG levels were significantly greater in calves fed large (3 L) volumes of CR compared with calves fed small (1.5 L) volumes of CR, regardless of feeding method. These differences were attributed to the larger mass of IgG ingested by calves fed 3 L of CR (200 g of IgG) compared with calves fed 1.5 L of CR (100 g of IgG). For calves fed small (1.5 L) volumes of colostrum, serum total protein (TP, g/dL), serum IgG (IgG, mg/mL), acceptable passive transfer rates (APT, %), and apparent efficiency of absorption of IgG (AEA, %) were significantly greater for calves fed with a bottle (TP = 5.3 g/dL; IgG = 12.5 mg/mL; APT = 100%; AEA = 51.1%) compared with calves fed with an esophageal tube feeder (TP = 5.0 g/dL; IgG = 9.8 mg/mL; APT = 41.7%; AEA = 40.5%). However, for calves fed large (3 L) volumes of colostrum, there was no difference in passive transfer indices for calves fed with a bottle (TP = 5.8 g/dL; IgG = 19.7 mg/mL; APT = 100%; AEA = 41.1%) compared with calves fed with an esophageal tube feeder (TP = 5.9 g/dL; IgG = 18.7 mg/mL; APT = 100%; AEA = 39.0%).  相似文献

Forty-four Holstein calves (19 male and 25 female) were used in this study of the relationships among age at first colostrum feeding, IgG intake, and apparent efficiency of IgG absorption. Time of birth was recorded for each calf and the calves were fed colostrum ad libitum after birth at either 0930 or 1630 h. Blood samples were collected immediately before and 24 h after colostrum feeding. Data from calves were then categorized into 4 groups representing time from birth to colostrum feeding: A = fed within 1 h (n = 5); B = fed from 1 to 6 h (n = 10); C = fed from 6 to 12 h (n = 21); and D = fed from 12 to 18 h (n = 8) after birth. Average total intake of colostrum was 3.6 ± 0.1 L. Over 80% of the calves consumed ≥3 L of colostrum. Apparent efficiency of IgG absorption declined remarkably 12 h after birth. Mean apparent efficiency of absorption of IgG in group D (15.8 ± 3.0%) was lower than that in groups A (30.5 ± 3.9%) and B (27.4 ± 2.8%). Serum IgG concentration in calves was positively correlated with IgG intake in all groups. The relationship between mass of IgG consumed and calf serum IgG at 24 h was different for each time of colostrum feeding, with only limited differences observed between groups A and B. We concluded that failure of transfer of passive immunity in newborn calves may be avoided if calves consume ≥3 L of colostrum with IgG concentration >40 mg/mL within 6 h after birth. These findings help define the opportunity to minimize failure of transfer of passive immunity to newborn calves under management programs similar to those used on commercial dairy farms.  相似文献

Adequate absorption of bovine colostrum correlates with improved neonatal health. The apparent efficiency of absorption (AEA) of immunoglobulins can be measured using a mathematical equation based on serum and colostral IgG concentration levels, as well as calf body weight and the volume of colostrum being fed. Although commonly measured in research projects, little information is available on the normal AEA across a large group of healthy calves on multiple farms. The purpose of this study was to observe how contributing factors (volume of feeding, birth weight, and time of feeding) can alter AEA and establish a reference range for AEA in healthy calves. Study subjects were 100 Holstein heifer calves from 5 different dairies in North Carolina and Colorado. After a normal calving, the heifer received either 4 or 5.6 L of colostrum within 4 h of birth, an aliquot of the fed colostrum was saved, and a blood sample was collected between 24 and 36 h after birth. Birth weights were measured using the same weight tape on each farm. Radial immunodiffusion assay was performed to obtain IgG concentrations in the colostrum and serum samples. From this data, the AEA was calculated. The AEA ranged from 7.7 to 59.9% with mean of 28.1 ± 9.5% and median of 27.5%. The AEA of 69% of the calves fell between 21 and 40%. The AEA varied widely between calves, even when feeding was standardized. Results suggest that serum IgG concentration may potentially be increased by feeding increased volumes of colostrum or genetic selection, given the wide range of AEA values obtained.  相似文献

Acquisition of high quality colostrum is an important factor influencing neonatal calf health. Many methods have been used to assess the Ig concentration of colostrum; however, improved, validated evaluation tools are needed. The aims of this study were to evaluate both optical and digital Brix refractometer instruments for the measurement of Ig concentration of colostrum as compared with the gold standard radial immunodiffusion assay laboratory assessment and to determine the correlation between Ig measurements taken from fresh and frozen colostrum samples for both Brix refractometer instruments. This research was completed using 288 colostrum samples from 3 different farms. It was concluded that the optical and digital Brix refractometers were highly correlated for both fresh and frozen samples (r = 0.98 and r = 0.97, respectively). Correlation between both refractometer instruments for fresh and frozen samples and the gold standard radial immunodiffusion assay were determined to be very similar, with a correlation coefficient between 0.71 and 0.74. Both instruments exhibited excellent test characteristics, indicating an appropriate cut-off point of 22% Brix score for the identification of good quality colostrum.  相似文献

Ingestion and absorption of greater quantities of IgG are required to increase serum IgG levels in newborn calves. This could be achieved by adding colostrum replacer (CR) to maternal colostrum (MC). The objective of this study was to investigate whether low and high-quality MC can be enriched with bovine dried CR to achieve adequate serum IgG levels. Male Holstein calves (n = 80; 16/treatment) with birth body weights (BW) of 40 to 52 kg were randomly enrolled to be fed 3.8 L of the following combinations: 30 g/L IgG MC (C1), 60 g/L IgG MC (C2), 90 g/L IgG MC (C3), C1 enriched with 551 g of CR (60 g/L; 30-60CR), or C2 enriched with 620 g of CR (90 g/L: 60-90CR). A subset of 40 calves (8/treatment) had a jugular catheter placed and were fed colostrum containing acetaminophen at a dose of 150 mg/kg of metabolic body weight, to estimate abomasal emptying rate per hour (kABh). Baseline blood samples were taken (0 h), followed by sequential samples at 1, 2, 3, 4, 5, 6, 8, 10, 12, 24, 36, and 48 h relative to initial colostrum feeding. Results for all measurements are presented in the following order, unless otherwise stated: C1, C2, C3, 30-60CR, and 60-90CR. Serum IgG levels at 24 h were different among calves fed C1, C2, C3, 30-60CR, and 60-90CR: 11.8, 24.3, 35.7, 19.9, and 26.9 mg/mL ± 1.02 (mean ± SEM), respectively. Serum IgG at 24 h increased when enriching C1 to 30-60CR, but not from C2 to 60-90CR. Similarly, apparent efficiency of absorption (AEA) values for calves fed C1, C2, C3, 30-60CR, and 60-90CR were different: 42.4, 45.1, 43.2, 36.3, and 33.4% ± 1.93, respectively. Enriching C2 to 60-90CR reduced AEA, and enriching C1 to 30-60CR tended to decrease AEA. The kABh values for C1, C2, C3, 30-60CR, and 60-90CR were also different: 0.16, 0.13, 0.11, 0.09, and 0.09 ± 0.005, respectively. Enriching C1 to 30-60CR or C2 to 60-90CR reduced kABh. However, 30-60CR and 60-90CR have similar kABh compared with a reference colostrum meal (90 g/L IgG, C3). Even though kABh was reduced for 30-60CR, results indicate that C1 has the potential to be enriched and achieve acceptable serum IgG levels at 24 h without affecting AEA.  相似文献

Transfer of sufficient IgG to the newborn calf via colostrum is vital to provide it with adequate immunological protection and resistance to disease. The objectives of the present study were to compare serum IgG concentration and health parameters of calves (1) fed different volumes of colostrum [7, 8.5, or 10% of body weight (BW)] within 2 h of birth and (2) given 0, 2, or 4 subsequent feedings of transition milk (i.e., milkings 2 to 6 postcalving). Ninety-nine dairy calves were fed 7, 8.5, or 10% of BW in colostrum within 2 h of birth and given 0, 2, or 4 subsequent feedings of transition milk. The concentration of IgG in the serum of calves was measured at 24, 48, 72, and 642 h of age by an ELISA. The apparent efficiency of absorption for IgG was determined. Health scores were assigned to calves twice per week and all episodes of disease were recorded. The effect of experimental treatment on calf serum IgG concentration differed by the age of the calf. Calves fed 8.5% of BW in colostrum had a greater mean serum IgG concentration than calves fed 7 or 10% of BW at 24, 48, and 72 h of age. At 642 h of age, serum IgG concentrations of calves fed 8.5% of BW (24.2 g/L) and calves fed 10% of BW (21.6 g/L) did not differ, although the serum IgG concentration of calves fed 8.5% of BW was still greater than that of calves fed 7% of BW (20.7 g/L). No difference in serum IgG concentration existed between calves fed 7% of BW and those fed 10% of BW at any age. No significant effect of number of subsequent feedings of transition milk on calf serum IgG concentration was detected. The apparent efficiency of absorption of calves fed 8.5% of BW in colostrum (38%) was greater than calves fed 7% of BW in colostrum (26%) and tended to be greater than in calves fed 10% of BW (29%). Calves fed further feedings of transition milk after the initial feeding of colostrum had a lower odds (0.62; 95% confidence interval: 0.41 to 0.93) of being assigned a worse eye/ear score (i.e., a more copious ocular discharge or pronounced ear droop) and a lower odds (0.5; 95% confidence interval: 0.32 to 0.79) of being assigned a worse nasal score (i.e., a more copious and purulent nasal discharge) during the study period relative to calves that received no further feedings of transition milk. In conclusion, calves fed 8.5% of BW in colostrum within 2 h of birth achieved a greater concentration of IgG in serum in the first 3 d of life than calves fed either 7 or 10% of BW. Feeding calves transition milk subsequently reduced their odds of being assigned a worse eye/ear and nasal score.  相似文献

The objective of this study was to determine the effect of different durations of heat treatment on passive transfer of IgG from high-, medium- and low-quality colostrum. Colostrum was collected from The Pennsylvania State University dairy herd and divided by quality (high, medium, or low) based on colostrometer measurements. Colostrum was pooled by quality to create 3 unique batches. Each batch was further divided in thirds as follows: frozen to be fed without heat treatment, heated at 60°C for 30 min, or heated at 60°C for 60 min. Colostrum samples from each treatment were collected and analyzed for standard plate count, gram-negative noncoliforms, coliforms, and total IgG concentration. Serum samples were collected from 108 Holstein calves before feeding colostrum and 24 h after birth. Blood samples were analyzed for total protein, total IgG, and hematocrit. Colostrum quality (high, medium, or low), heat treatment (unheated, 60°C for 30 min or, 60°C for 60 min), and their interaction were analyzed as fixed effects, with calf sex included as a random block effect. Colostrum IgG was different between quality groups (92.5, 59.4, and 48.1 mg/mL of IgG). Heating colostrum reduced IgG concentration compared with the control by 9% when heated for 30 min and by 12% when heated for 60 min. Colostrum heated for 60 min had a lower standard plate count than colostrum heated for 30 min or not heated (1.8, 2.0, and 3.6 log cfu/mL, respectively). Serum IgG concentration at 24 h increased as colostrum quality increased (18.0, 22.2, and 24.8 mg/mL) and tended to increase as heat treatment time increased (19.7, 20.3, and 25.0 mg/mL of IgG). Apparent efficiency of IgG absorption was greater in calves that received medium-quality colostrum compared with calves fed high-quality colostrum (38.1 and 25.0%, respectively). These results suggest an upper limit may exist to the amount of IgG absorption in a given time period and that medium- or high-quality colostrum yields similar blood IgG concentration given the same volume of intake.  相似文献
