Airborne enteric coliphages and bacteria in sewage treatment plants   总被引:3,自引:0,他引:3  
The concentrations of airborne culturable microorganisms were determined in wastewater and sludge treatment processes of seven sewage treatment plants. Two types of coliphages, Salmonella and total viable bacteria were sampled by the BioSampler and the numbers of faecal coliforms and enterococci were obtained from the Andersen 6-stage impactor. The BioSampler recovered higher numbers of airborne coliphage viruses than has been measured with other liquid samplers in previous studies, suggesting that this sampler has improved efficiency for sampling airborne coliphages. Airborne coliphages were detected in many stages of the wastewater or sludge treatment process. The highest microbiological air contaminations were found in pre-treatment and aerated grit separation stages of the operation. This was attributed to aerosolisation of microorganisms by mechanical handling or forced aeration. Aeration and settling processes located outdoors caused low microbial concentrations, but the brush aerator released more microorganisms into the air. Our results emphasize the necessity for controlling the exposure of sewage workers to airborne microorganisms, especially in process areas that involve mechanical agitation or forced aeration of wastewater.  相似文献

Results of a study conducted to assess the degree of airborne microbial contamination generated by a wastewater treatment plant (WWTP) with bioreactor “BIO-PAK” closed treatment system and evaluation of the dispersion of potential pathogens are described. Over the year aerosol samples were collected simultaneously with sedimentation and impact methods from several plant sites and the surroundings. External upwind sites were used as control. Total colony-forming counts of heterotrophic psychrophilic, psychrotrophic, mesophilic, haemolytic bacteria, as well as members of the Enterobacteriaceae family, Staphylococcus, Enterococcus and Pseudomonas genera, actinomycetes and fungi (moulds, yeasts and yeast-like fungi) were determined. Their highest concentrations ranged up to 101-103 CFU/m3 (with the exception of moulds exceeding 2 × 104) were determined in air sampled inside the bioreactor, its vicinity, and near the grate chamber. Higher species diversity of the family Enterobacteriaceae (including Shigella spp., Yersinia enterocolitica, Escherichia coli, Klebsiella pneumoniae ozaenae) in the air sampled inside or near the bioreactor may imply a health risk for staff exposed for longer periods of time. Notwithstanding, no increased emission of the analysed groups of microorganisms, including faecal bacteria, were found outside the WWTP.  相似文献

The objective of this study was to determine the frequency and densities of yeasts and filamentous fungi in coastal water samples as well as their correlation with the indicator bacteria of faecal pollution. The prevalence of fungi was investigated in parallel with the standard pollution indicator microorganisms in 197 marine water samples from six northern Greek prefectures during the bathing season May–October 1999. Filamentous fungi were isolated from all the examined samples and yeasts from 29 (14.7%) of them; among the positive samples, their mean counts were 90.9 and 38.4 cfu/100 ml, respectively. A total of 23 genera of filamentous fungi and four genera of yeasts were identified. Prevailing genera of filamentous fungi were Penicillium, Aspergillus and Alternaria spp., whereas Candida spp. was the most frequently isolated yeast. Counts of yeasts were significantly (p<0.01) correlated with those of total and faecal coliforms, whereas no correlation was found between filamentous fungi and the indicator bacteria of faecal pollution. Significantly higher counts of total and faecal coliforms (p<0.05), and enterococci (p<0.001), were found during the months with the higher water temperatures and bather numbers. In the six prefectures, significant differences were observed in the counts of filamentous fungi and yeasts as well as in the counts of all the faecal pollution indicators. The results of this study indicate that coastal water can be a path for contamination of swimmers with yeasts and filamentous fungi and that the pollution indicator microorganisms cannot always predict their presence.  相似文献

In this study, bacterial removal efficiencies of planted and unplanted subsurface vertical flow constructed wetlands are compared. Indicator organisms such as faecal coliforms (Escherichia coli, total coliforms) and enterococci, and a number of heterotrophic bacteria (heterotrophic plate counts) have been analysed from the influent and effluent of the constructed wetlands as well as at different depths (water and substrate samples). Furthermore dry matter content and total organic carbon (TOC) have been analysed and correlated. The investigated systems show a high removal rate for indicator organisms (a log removal rate of 2.85 for HPC, 4.35 for E. coli, 4.31 for total coliforms and 4.80 for enterococci was observed). In general no significant difference in the removal efficiency of planted and unplanted vertical flow beds could be measured. Only enterococci measured in the substrate samples of the main layer of the filter could a statistically significant difference be observed.  相似文献

A detailed knowledge on decay or inactivation kinetics of faecal indicator microorganism in rivers is essential for control of bathing water quality. Both reliable inactivation rate coefficients for such kinetics and the knowledge on pathogen accumulation in benthic biofilms are needed for the assessment of river self purification capability and for being able to make decisions for an optimized water management. Therefore, the inactivation kinetics for main indicator microorganisms like faecal coliforms and intestinal Enterococci in water and on stones of the river Isar (Germany) were measured at artificial sunlight radiation. A flume was built, containing Isar water and sediment. By adding a pulse of pre-treated sewage water, the subsequently change of indicator bacteria levels in the bulk and biofilm phase is measured. Bacterial inactivation was mainly dependent on sunlight intensity. Mean inactivation rate coefficients in the experimental set up with river water obtained for a radiation intensity comparable to average midday sunlight in June in 50 degrees north latitude (I((290-390nm))=40.0W/m(2)) were found to be 21.4d(-1) for faecal coliforms and 20.0d(-1) for intestinal Enterococci. For a radiation intensity conform with the annual mean radiation in Germany (I((290-390nm))=8.0W/m(2)) inactivation rate coefficients were 12.7d(-1) for faecal coliforms and 9.3d(-1) for intestinal Enterococci. A measurement without direct artificial sunlight (I((290-390nm))=0.08W/m(2)) yields inactivation rate coefficients of 3.4d(-1) for faecal coliforms and 1.7d(-1) for intestinal Enterococci, which were similar to those found in wastewater treatment ponds. Due to re-growth and better environmental conditions the concentrations of faecal coliforms and intestinal Enterococci within the biofilm were 10(2) to 10(4) fold higher compared to the bulk water depending on the exposure time.  相似文献

Raw wastewater is a potential carrier of pathogenic microorganisms and may pose a health risk when pathogenic microorganisms become aerosolized during aeration. Two different types of wastewater treatment plants were investigated, and the amounts of cultivable bacteria and fungi were measured in the emitted aerosols. Average concentrations of 17,000 CFU m(-3) of mesophilic, 2,100 CFU m(-3) of TSA-SB bacteria (bacteria associated with certain waterborne virulence factors), 1700 CFU m(-3) of mesophilic and 45 CFU m(-3) of thermotolerant fungi, were found in the aerosol emitted by the aeration tank of the activated sludge plant. In the aerosol of the fixed-film reactor 3000 CFU m(-3) mesophilic and 730CFUm(-3) TSA-SB bacteria, and 180 CFUm(-3) mesophilic and 14 CFU m(-3) thermotolerant fungi were measured. The specific emissions per population equivalent between the two types of treatment plants differed by two orders of magnitude. The microbial flux based on the open water surface area of the two treatment plants was similar. The aerosolization ratios of cultivable bacteria (expressed as CFU m(-3) aerosol/m(-3) wastewater) ranged between 8.4 x 10(-11) and 4.9 x 10(-9). The aerosolization ratio of fungi was one to three orders of magnitude higher and a significant difference between the two types of treatment plants could be observed.  相似文献

It has been suggested that bacteriophages can provide useful information about the pathogenic microorganisms, particularly enteric viruses, present in water. This information is complementary to that obtained from bacterial indicators of faecal contamination, which would be of great value for evaluating the risks associated with the use of certain types of water. Before bacteriophages can be used as indicators of faecal contamination, we need to confirm that bacteriophages give a different response to that given by the well-known bacteria indicators and to determine what happens to bacteriophages in river water. Indeed, drinking water is often produced from river water, either by natural filtration through the soil or after undergoing various treatments. We collected 96 river water samples from six different sites between February and November 2000. The samples were analysed for three faecal indicator bacteria (thermotolerant coliforms, enterococci and spores of sulphite-reducing anaerobes) and three types of bacteriophages (somatic coliphages, F-specific phages and Bacteroides fragilis phages). The densities of thermotolerant coliforms and enterococci depended mainly on physical factors such as flow rate and water temperature. High temperature and low flow rate led to a decrease in the density of these microorganisms, especially in the absence of a major input of faecal pollution. Conversely, the densities of somatic coliphages, F-specific phages and spores of sulphite-reducing anaerobes remained constant regardless of the flow rate and temperature. The density of Bacteroides fragilis phages was too low for unambiguous determination of their fate in river water.  相似文献

This study was aimed at assessing the prevalence of pathogenic Escherichia coli strains, relative to the total count of E. coli, faecal coliforms and other heterotrophic mesophilic aerobic bacteria (HMAB) isolated in groundwater in the equatorial region of Cameroon (Central Africa). Bacteria were isolated using standard methods. Pathogenic E. coli strains were then identified using haemagglutination and antisera tests. The maximum abundance of HMAB, faecal coliforms and E. coli strains were 4.9 × 106, 5.6 × 103 and 1 × 103 colony‐forming units (CFU)/100 mL, respectively. The count of pathogenic E. coli strains reached 3 CFU/100 mL. The counts of commensal and pathogenic E. coli strains underwent temporal and spatial fluctuations. In 21% of sampling sites, the abundance of faecal coliforms was significantly correlated to that of E. coli (P < 0.05). However, the isolated bacterial count was not significantly correlated to that of the pathogenic E. coli strains (P > 0.05). The bacteria abundance dynamics may be impacted by many interacting factors.  相似文献

In this study, for better understanding the practical removal effect of air handling unit (AHU) system on airborne microorganisms (including bacteria and fungus) and particles and microbial growth, the samples of microorganisms and particles in 10 air handling unit (AHU) systems including fan coils and indoor air were collected and analyzed in air and component surfaces of such systems in two large public buildings. It is found that the removal efficiency is of the highest for bacteria 73.9% and the lowest for particles (0.5–2 μm) 24.4%. The surface concentration of equipment bacteria is 29 CFU/cm2 and fungi 137 CFU/cm2. Five of 10 systems have higher fungi concentrations on air intake than that on diffuser. The results also show that the central air supply system with common components (e.g., pre-filter and bag filter) has difficulty to achieve/maintain good performance once microorganisms and particles exist, especially for particle size D ≤ 3.3 μm. The size distribution has large influence on removal efficiency. The microbial growth on surfaces of duct and equipment was noticed and may be transferred into indoor air. This will decrease the indoor air quality and lead to adverse health effect.  相似文献

It has been found that during heterotrophic denitrification, biocenosis of the upflow anaerobic sludge blanket (UASB) reactor is composed of bacteria, fungi, protozoa and rotifers. Among these groups of organisms close ecological relationships exist. The scheme of these relationships is proposed. Also described are quantity, taxonomy and biochemical features of denitrifying bacteria and fungi occurring in biocenoses during the processes carried out in the presence of methanol, glycerol, acetic and lactic acids.

Quantitative microbial risk assessment (QMRA) was used to evaluate the relative contribution of faecal indicators and pathogens when a mixture of human sources impacts a recreational waterbody. The waterbody was assumed to be impacted with a mixture of secondary-treated disinfected municipal wastewater and untreated (or poorly treated) sewage, using Norovirus as the reference pathogen and enterococci as the reference faecal indicator. The contribution made by each source to the total waterbody volume, indicator density, pathogen density, and illness risk was estimated for a number of scenarios that accounted for pathogen and indicator inactivation based on the age of the effluent (source-to-receptor), possible sedimentation of microorganisms, and the addition of a non-pathogenic source of faecal indicators (such as old sediments or an animal population with low occurrence of human-infectious pathogens). The waterbody indicator density was held constant at 35 CFU 100 mL−1 enterococci to compare results across scenarios. For the combinations evaluated, either the untreated sewage or the non-pathogenic source of faecal indicators dominated the recreational waterbody enterococci density assuming a culture method. In contrast, indicator density assayed by qPCR, pathogen density, and bather gastrointestinal illness risks were largely dominated by secondary disinfected municipal wastewater, with untreated sewage being increasingly less important as the faecal indicator load increased from a non-pathogenic source. The results support the use of a calibrated qPCR total enterococci indicator, compared to a culture-based assay, to index infectious human enteric viruses released in treated human wastewater, and illustrate that the source contributing the majority of risk in a mixture may be overlooked when only assessing faecal indicators by a culture-based method.  相似文献

Details of a method to isolate culturable bacteria and fungi from carpet dust were evaluated to isolate the greatest numbers of these agents. Four broad groups were evaluated: mesophilic and thermophilic bacteria and moderately hydrophilic and xerophilic fungi. Features studied included: 1) mixing time; 2) addition of glass beads; 3) length of time dust settled before suspensions were plated; 4) relative concentrations of microorganisms in the fibrous and fine dust fractions; and 5) storage temperature and period. The findings are preliminary because of the small number of samples, but a votexing time of at least 2 min with glass beads gave the best results in terms of the highest estimate of the concentration of culturable bacteria or fungi. Microorganisms were retrievable-from the upper portion of a suspending liquid for at least 30 min with little detectable change in concentration. Both bacteria and fungi were more abundant in the fine than the fibrous dust fraction. No significant losses were observed for samples stored up to 25 days at 5 degrees C or 25 degrees C. This procedure optimized measurement of total concentration, but may not identify the range of genera and species in dust if microorganisms present in small numbers and as single units are underrepresented relative to those in clusters and aggregates.  相似文献

Water pollution by microorganisms of fecal origin is a current world-wide public health concern. Total coliforms, fecal coliforms (Escherichia coli) and enterococci are indicators commonly used to assess the microbiological safety of water resources. In this study, influent water samples and treated water were collected seasonally from a water treatment plant and two major water wells in a Black Belt county of Alabama and evaluated for water quality indicator bacteria. Influent river water samples serving the treatment plant were positive for total coliforms, fecal coliforms (E. coli), and enterococci. The highest number of total coliform most probable number (MPN) was observed in the winter (847.5 MPN/100 mL) and the lowest number in the summer (385.6 MPN/100 mL). Similarly E. coli MPN was substantially higher in the winter (62.25 MPN/100 mL). Seasonal variation of E. coli MPN in influent river water samples was strongly correlated with color (R2 = 0.998) and turbidity (R2 = 0.992). Neither E. coli nor other coliform type bacteria were detected in effluent potable water from the treatment plant. The MPN of enterococci was the highest in the fall and the lowest in the winter. Approximately 99.7 and 51.5 enterococci MPN/100 mL were recorded in fall and winter seasons respectively. One-way ANOVA tests revealed significant differences in seasonal variation of total coliforms (P < 0.05), fecal coliforms (P < 0.01) and enterococci (P < 0.01). Treated effluent river water samples and well water samples revealed no enterococci contamination. Representative coliform bacteria selected by differential screening on Coliscan Easygel were identified by 16S ribosomal RNA gene sequence analysis. E. coli isolates were sensitive to gentamicin, trimethoprim/sulfamethazole, ciprofloxacin, vancomycin, tetracycline, ampicillin, cefixime, and nitrofurantoin. Nonetheless, isolate BO-54 displayed decreased sensitivity compared to other E. coli isolates. Antibiotic sensitivity pattern can be employed in microbial source tracking.  相似文献

The specificity of indicators that depend on elevated incubation temperature as a selective factor for their enumeration was questioned because of the possibility of interference from autochthonous microorganisms adapted to high ambient temperatures. Lactose-fermenting cultures isolated from fecal coliform tests of tropical marine surface waters were identified as consisting of about 70% Escherichia coli, and most of the remaining cultures being Klebsiella, Enterobacter or Citrobacter species. This confirmed the taxonomic specificity of fecal coliform tests for these waters. Fecal and total coliforms, fecal streptococcus, heterotrophic bacteria and yeast counts had correlations of above 99% confidence levels with most other microbial and chemical parameters studied. Waters with fecal coliform counts above 1000 per 100 ml had increased incidence of presumptive pathogenic yeasts, Pseudomonas aeruginosa and Salmonella. Our data support the use of coliforms or fecal streptococci as indicators of recent fecal pollution in tropical marine waters and yeast or heterotrophic bacteria counts as complementary indicator methods for these waters.  相似文献

Sewage-contaminated rivers are ecosystems deeply disturbed by human activity due to the release of heavy metals, organic pollutants and pharmaceuticals as well as faecal and pathogenic micro-organisms, which coexist with the autochthonous microbial population. In this study, we compared the percentage of resistance in faecal and heterotrophic bacteria in rivers with different degrees of sewage pollution. As a matter of fact, no correlation was found neither between the degree of sewage pollution and the percentage of antimicrobial resistant heterotrophic bacteria nor between the number of resistant faecal bacteria and that of resistant heterotrophic bacteria. Most of the resistant isolates from the Zenne river downstream Brussels were multi-resistant and the resistance patterns were similar among the strains of each phylogenetic group. The total microbial community in this polluted river (as evaluated through a 16S rRNA gene clone library analysis) appeared to be dominated by the phyla Proteobacteria and Bacteroidetes while the phylum TM7 was the third most represented.  相似文献

Coprostanol is a faecal sterol that has been proposed as an alternative measure of faecal pollution. While the technique has been used successfully to trace sewage-derived organic matter in a range of environments, it has not been embraced for use as a water quality indicator. This is mostly because of a lack of epidemiological evidence relating coprostanol abundance to any health risk. However, there is a valuable reason why the concentration of coprostanol should be related as quantitatively as possible to the abundance of bacterial indicators currently used to measure faecal pollution. The measurement of coprostanol (and concurrently other faecal sterols) offers many diagnostic and quantitative advantages over traditional techniques for detecting human sewage pollution versus faecal contamination from animal sources. Knowing the amount of coprostanol expected given a certain amount of human sewage pollution would provide a measure against which water managers could quantitatively assess faecal pollution as a whole and relate that assessment to variables with which they are more familiar. This study determines the relationships between coprostanol concentrations and indicator bacterial counts and synthesises the results from several environments to propose coprostanol concentrations broadly equivalent to existing bacterial standards. Our data suggest that 60 and 400 ng L−1 of coprostanol correspond to currently defined primary and secondary contact limits for bacteria measured as thermotolerant coliforms (commonly referred to as faecal coliforms) or enterococci.  相似文献

A study was undertaken to determine the effect of variations in temperature, relative humidity, occupancy density and location (indoor/outdoor) on the concentrations of viable airborne bacterial and fungal spores at an air-conditioned and a non air-conditioned food stall in Singapore. Typically, bioaerosols consisted of 50.5% bacteria and 49.5% fungi in the indoor environment. In contrast, for the outdoor environment, bacteria on an average only accounted for 20.6% of culturable airborne microorganisms whereas fungal concentrations were 79.4%. Results on bioaerosol size distributions revealed that 67% of indoor bacteria and 68% of outdoor bacteria, 85% of indoor fungi and 68% of outdoor fungi were associated with fine mode particulates (<3.3 μm). Occupant density was the key factor that affected indoor airborne bacteria concentrations while concentrations of outdoor airborne bacteria depended strongly on ambient temperature. Indoor fungal concentration was positively correlated to relative humidity whereas outdoor fungal concentration was positively correlated to relative humidity and negatively correlated to temperature. The study also compared the biological air quality between a non air-conditioned food stall (Stall A) and an air-conditioned food stall (Stall B). The dining area of the former had lower bacterial concentrations as compared to the latter, while fungal spore’s concentrations showed a reverse trend. The dominant airborne bacteria genera were Staphylococcus, Pseudomonas, Alcaligens, and Corynebacterium whereas Penicillium, Aspergillus and Cladosporium were the most common fungal genera and groups in both food stalls.  相似文献

The efficiency of solar disinfection (SODIS), recommended by the World Health Organization, has been determined for rainwater disinfection, and potential benefits and limitations discussed. The limitations of SODIS have now been overcome by the use of solar collector disinfection (SOCO-DIS), for potential use of rainwater as a small-scale potable water supply, especially in developing countries. Rainwater samples collected from the underground storage tanks of a rooftop rainwater harvesting (RWH) system were exposed to different conditions of sunlight radiation in 2-L polyethylene terephthalate bottles in a solar collector with rectangular base and reflective open wings. Total and fecal coliforms were used, together with Escherichia coli and heterotrophic plate counts, as basic microbial and indicator organisms of water quality for disinfection efficiency evaluation. In the SOCO-DIS system, disinfection improved by 20–30% compared with the SODIS system, and rainwater was fully disinfected even under moderate weather conditions, due to the effects of concentrated sunlight radiation and the synergistic effects of thermal and optical inactivation. The SOCO-DIS system was optimized based on the collector configuration and the reflective base: an inclined position led to an increased disinfection efficiency of 10–15%. Microbial inactivation increased by 10–20% simply by reducing the initial pH value of the rainwater to 5. High turbidities also affected the SOCO-DIS system; the disinfection efficiency decreased by 10–15%, which indicated that rainwater needed to be filtered before treatment. The problem of microbial regrowth was significantly reduced in the SOCO-DIS system compared with the SODIS system because of residual sunlight effects. Only total coliform regrowth was detected at higher turbidities. The SOCO-DIS system was ineffective only under poor weather conditions, when longer exposure times or other practical means of reducing the pH were required for the treatment of stored rainwater for potable purposes.  相似文献
