Biogenic amines, total volatile base nitrogen (TVB‐N) and trimethylamine nitrogen (TMA‐N) concentrations in the muscle of postmortem Chinese mitten crab stored at 4 and 20 °C were studied. Among five biogenic amines, histamine, tyramine, tryptamine, putrescine and cadaverine, histamine was the main biogenic amine formed during the storage time and reached highest levels of 91.22 mg kg?1 after 72 h at 4 °C and 181.23 mg kg?1 after 24 h at 20 °C. Histamine could be used a safety index for crab. The levels of other biogenic amines were less than 10 mg kg?1 and fluctuated with storage time. TVB‐N and TMA‐N levels did not change greatly throughout 72 h storage at refrigerated temperature. TVB‐N and TMA‐N were not considered to be reliable indicators of the freshness of crab.  相似文献

The simultaneous effects of processing factors such as ripening time (25–75 days), ripening temperature (4–14 °C) and brine concentration (10–13%) on biogenic amines content, proteolysis and sensory score of Iranian white brine cheese were studied, in 12 cheeses. Response surface methodology (RSM) was used to minimise biogenic amines content. At low level of ripening time, biogenic amines content decreased with increasing levels of brine concentration but at high level of ripening time, brine concentration had inverse effect. Ripening time showed quadratic effect on biogenic amines content. Based on biogenic amines content and sensory score, the optimum conditions were 13% brine and ripening at 9–14 °C for 43–65 days.  相似文献

为探究鲣鱼中优势腐败产胺菌的种类,从冷藏的鲣鱼中筛选出3种优势腐败产胺菌,通过16S rDNA分子鉴定技术对菌株进行鉴定为荧光假单胞菌(Pseudomonas fluorescens)、弗氏柠檬酸杆菌(Citrobacter freundii)和嗜水气单胞菌(Aeromonas hydrophila),将鉴定出的优势腐败产胺菌接种至无菌鱼肉中4 ℃条件下贮藏,通过测定菌落总数和挥发性盐基氮(TVB-N)变化,以腐败代谢产物产量因子(YTVB-N/CFU)分析3种优势腐败产胺菌对鲣鱼的致腐能力,并通过样品中生物胺的含量比较3种优势腐败产胺菌的产胺能力。结果表明:在贮藏8 d后接种荧光假单胞菌、弗氏柠檬酸杆菌和嗜水气单胞菌组的菌落总数分别达到8.36、8.27和8.13 lg CFU/g,TVB-N值分别为28.21、30.30和31.29 mg/100 g,3种优势腐败产胺菌对鲣鱼的致腐能力大小为嗜水气单胞菌>弗氏柠檬酸杆菌>荧光假单胞菌。接种荧光假单胞菌组、弗式柠檬酸杆菌组和嗜水气单胞菌组的组胺含量分别为196.23、83.43和261.22 mg/kg,3组样品中总生物胺含量为嗜水气单胞菌>荧光假单胞菌>弗氏柠檬酸杆菌。综合比较得出,嗜水气单胞菌对4 ℃冷藏鲣鱼致腐产胺能力最强,本研究增加对冷藏鲣鱼中优势腐败产胺菌种类的了解并提供了部分理论基础。  相似文献

Biogenic amines are identified as toxicological substances in foods and may have detrimental effects on consumers’ health. In recent years, the application of microorganisms that can degrade biogenic amines has become an emerging method for their reduction. The degradation characteristics and application potential of a salt-tolerant bacterium Halomonas shantousis SWA25 were investigated in this study. H. shantousis SWA25 exhibited degradation activity against eight biogenic amines at 10–40°C (optimum, 30–40°C) and pH 3.0–9.0 (optimum, 6.0–7.0) in the presence of 0–20% (w/v) NaCl (optimum, 0%). Specifically, H. shantousis SWA25 degraded all tryptamine (TRY) and tyramine (TYR) in 6 h, all phenethylamine (PHE) in 9 h, 66.7% of histamine (HIM), 52.4% of cadaverine (CAD), 48.0% of spermidine (SPD), 42.9% of putrescine (PUT) and 42.0% of spermine (SPM) in 20 h at 30°C and pH 7.0 with shaking at 120 r min?1. The enzymes from H. shantousis SWA25 responsible for degradation of biogenic amines were mainly amine oxidases located on the cell membrane. Further studies showed that H. shantousis SWA25 effectively degraded TRY, PHE, PUT, CAD, HIM and TYR in commercial fish sauce and soy sauce samples. Nevertheless, significant SPD and SPM degradation were not observed due to low initial concentrations. Therefore, H. shantousis SWA25 can be applied as a potential biogenic amines degradation bacterium in foods.  相似文献

The physicochemical and microbiological parameters of postmortem crayfish stored at 4 °C and 25 °C were evaluated in order to reduce safety risks of crayfish from temperature abuse during transportation and storage. Results showed that hepatopancreas of postmortem crayfish had significantly higher contents of total volatile basic nitrogen, biogenic amines (BAs) and higher microbial loads than tail meat, regardless of storage temperature. Enterobacteriaceae and Pseudomonas counts reached log 6.41 log10 CFU g−1 and 6.31 log10 CFU g−1 in hepatopancreas of crayfish at 25 ℃ for 6 h. Putrescine and cadaverine were the main BAs in tail meat with levels of 28.18 ± 0.73 mg kg−1 and 187.32 ± 3.57 mg kg−1, respectively, whilst cadaverine, spermidine and spermine were the major BAs in hepatopancreas, reaching 283.45 ± 3.95 mg kg−1, 62.87 ± 9.02 mg kg−1 and 155.31 ± 4.55 mg kg−1, respectively, after postmortem storage at 25 °C for 12 h. With time increasing, Acinetobacter, Flavobacterium, Aeromonas and Chryseobacterium at 25 °C and Acinetobacter, Flavobacterium, Psychrobacter at 4 °C in tail meat as well as Bacteroides and Muribaculaceae at 25 °C, and Acinetobacter, Psychrobacter at 4 °C in hepatopancreas, gradually became the major genus at the end of storage. Based on the results of spoilage microorganisms and biogenic amines, crayfish stored at 4 °C and 25 °C could be edible within 24 h and 6 h, respectively.  相似文献

Pseudomonas marinoglutinosa, Aeromonas hydrophila isolated from unirradiated spoiling Indian mackerel (Rastrelliger kanagurta) and Bacillus megaterium, Micrococcus colpogenes isolated from gamma-irradiated mackerel were examined for their spoilage potential in fish homogenate and fish muscle sarcoplasmic and myofibrillar protein fractions. Pseudomonas marinoglutinosa (0° -2°C), Aeromonas hydrophila (0° - 2°C) and Bacillus megaterium (26°C) proliferated in the fish homogenate and sacroplasmic protein fraction which was associated with losses in sensory attributes and increase in non-protein-nitrogen (NPN) compounds, total volatile acids (TVA) and total volatile basic nitrogen (TVBN) compounds. On the other hand, the organisms exhibited diminished growth in MF fraction which was also accompanied by less changes in sensory and chemical quality parameters. Growth of M. colpogenes was not accompanied by putrefaction of the fish media as assessed by sensory and chemical criteria, suggesting contribution of varying spoilage potentials of the bacterial isolates in the overall spoilage of the fish.  相似文献

Abstract

The use of a controlled lactic fermentation has been studied by several authors as a means to extend meat shelf‐life without notably altering its sensory quality. Growth and metabolite production by spoilage microorganisms as well as by bioprotective strains are influenced by storage conditions. The objective of this work was to study the effect of temperature and storage time upon the growth and metabolite production of four spoilage bacteria and three bioprotective lactic acid strains in vacuum packaged, finely cut beef. Lipolysis was mainly due to the presence of Brochothrix thermosphacta at 4°C and at 20°C, whereas proteolysis was mainly caused by pseudomonads. Presence of Lactobacillus minor, a heterofermentative strain, did not promote proteolysis nor lipolysis at either temperature. No considerable lipolysis was observed until day 8 of storage in samples inoculated with bioprotective strains, but increased at day 12 when stored at 20°C; no proteolysis was observed at any storage temperature.  相似文献

The shelf life and freshness changes in pond-grown common carp (Cyprinus carpio L) during storage at 0–2°C, 5–6°C and room temperature (26–29°C) were investigated by sensory, microbiological, physical and chemical analyses. The effect of gutting on the shelf life during storage at 0–2°C was examined. Iodine/starch and potassium sorbate were examined for their effects on shelf life of whole fish stored at 0–2°C and 5–6°C. Sensory results indicated that the whole fish had a maximum shelf life of 24 to 25 days at 0– 2°C. The life of the fish to the point beyond which it would be unsuitable for sale (commercial shelf life) was 17 days at 0–2°C. Storage at 5–6°C shortened shelf life 2- to 2.5-fold. At room temperature (26–29°C), spoilage was evident after 13 h. Gutting the carp shortened its storage potential at 0–2°C. Iodine treatment of this species stored at 0–2°C and at 5–6°C did not extend shelf life. The maximum shelf life of sorbate-treated fish at 0–2°C and 5–6°C was extended by 1–2 days, commercial shelf life by 3–4 days. Total volatile basic nitrogen, pH and penetrometer analyses were not reliable indicators of changes in freshness during shelf life. Thiobarbituric acid values were not useful as rancid odours or flavours were not detected during storage.  相似文献

The possibility of using visible and short‐wavelength near‐infrared (SW‐NIR: 600 to 1100 nm) spectroscopy to detect the onset of spoilage and to quantify microbial loads in rainbow trout (Oncorhynchus mykiss) was investigated. Spectra were acquired on the skin and flesh side of intact trout fillet portions and on minced trout muscle samples stored at 4 °C for up to 8 d or at room temperature (21 °C) for 24 h. Principal component analysis (PCA) and partial least squares (PLS) chemometric models were developed to predict the onset and degree of spoilage. PCA results showed clear segregation between the control (day 1) and the samples held 4 d or longer at 4 °C. Clear segregation was observed for samples stored 10 h or longer at 21 °C compared with the control (0 h), indicating that onset of spoilage could be detected with this method. Quantitative PLS prediction models for microbial loads were established. For trout fillets, 4 °C: R= 0.97, standard error of prediction (SEP) = 0.38 log colony‐forming units (CFU)/g (flesh side); R= 0.94, SEP = 0.53 log CFU/g (skin side); R= 0.82, SEP = 0.82 log CFU/g for minced fish held at 21 °C. These results indicate that SW‐NIR in combination with multivariate statistical methods can be used to detect and monitor the spoilage process in rainbow trout and quantify microbial loads rapidly and accurately.  相似文献

以冷藏金枪鱼货架期终点筛选、纯化、鉴定所得优势腐败菌假单胞菌(Pseudomonas spp.)、不动杆菌 (Acinetobacter spp.)和热死环丝菌(Brochothrix thermosphacta)作为研究对象,通过分析接种优势腐败菌的冷藏 金枪鱼中各腐败微生物生长动态以及感官品质、挥发性盐基氮(total volatile basic nitrogen,TVB-N)含量、生物 胺含量的变化,以生长动力学参数和腐败物质的产量因子YTVB-N/CFU为评价标准,分析评价冷藏金枪鱼优势腐败菌假 单胞、不动杆菌和热死环丝菌的致腐败能力。结果表明:4 ℃冷藏金枪鱼中假单胞菌、不动杆菌和热死环丝菌的生 长曲线拟合效果最好的方程分别为修正Gompertz模型、Baranyi-Roberts模型和Logistic模型,分析所得生长动力学 参数发现,冷藏金枪鱼中假单胞菌、不动杆菌和热死环丝菌生长延滞时间和最大比生长速率分别为10.85、83.93、 79.11 h和0.045 7、0.182 9、0.111 1 h-1;接种假单胞菌、不动杆菌和热死环丝菌的冷藏金枪鱼感官货架期分别为 96、108 h和105 h,此时TVB-N含量分别为27.32、22.80 mg/100 g和24.85 mg/100 g;分析冷藏过程中金枪鱼生物胺 变化情况发现,假单胞菌产生物胺能力强于不动杆菌和热死环丝菌,贮藏96 h时接种假单胞菌组的生物胺指数已达 155.84,超过规定阈值100;冷藏金枪鱼致腐败能力分析结果显示,假单胞菌、不动杆菌和热死环丝菌的产量因子 YTVB-N/CFU分别为7.36×10-8、1.85×10-8 mg TVB-N/CFU和5.07×10-8 mg TVB-N/CFU,研究表明4 ℃冷藏金枪鱼贮 藏过程中假单胞菌致腐败能力较强,其次为热死环丝菌和不动杆菌。  相似文献

This study investigated the characteristics of spoilage bacteria isolated from fresh produce including growth at various temperatures, biofilm formation, cell hydrophobicity, and colony spreading. The number of spoilage bacteria present when stored at 35 °C was significantly greater than when stored at lower temperatures, and maximum population size was achieved after 10 h. However, Bacillus pumilus, Dickeya zeae, Pectobacterium carotovorum subsp. Carotovorum Pcc21, and Bacillus pumilus (RDA‐R) did not grow at the storage temperature of 5 °C. The biofilm formation by Clavibacter michiganensis, Acinetobacter calcoaceticus, and A. calcoaceticus (RDA‐R) are higher than other spoilage bacteria. Biofilm formation showed low correlation between hydrophobicity, and no significant correlation with colony spreading. These results might be used for developing safe storage guidelines for fresh produce at various storage temperatures, and could be basic information on the growth characteristics and biofilm formation properties of spoilage bacteria from fresh produce.  相似文献

BACKGROUND: Biogenic amines have received considerable attention owing to their undesirable effects in humans. There are few studies of changes in biogenic amine contents related to freshwater fish. Silver carp is an important freshwater fish species in China. This study aimed to investigate the changes in biogenic amines and their relation to total volatile base nitrogen (TVB‐N), microbiological and sensory score of silver carp fillets stored at 0, 3 and 15 °C. RESULTS: The total biogenic amine contents of all silver carp fillets (regardless of storage time and temperature) ranged from 13.05 to 318.10 mg kg?1. Putrescine and histamine were the main biogenic amines in silver carp fillets during storage. Cadaverine was only detected after 12 days at 3 °C and after 2 days at 15 °C. Spermidine and spermine contents increased during the early storage period and then slightly decreased. CONCLUSION: Low temperature could control the quality of silver carp fillets by inhibiting the contents of biogenic amines. Putrescine showed significant correlation with TVB‐N, total aerobic content, sensory score, tryptamine and phenylethylamine. A study on the evolution of free amino acids and biogenic amines in Azeitão cheese during 4 weeks at different temperatures of storage (4 and 25°C) was performed. Free amino acids and biogenic amines were determined by RP-HPLC with visible detection, following extraction from the cheese and derivatization with dabsyl chloride. The method presented a linear relation between peak area and concentration from 2–200 mg/l. The detection limit value was less than 1.5 mg/l. The average repeatability was less than 4%. The major free amino acids were proline, valine, isoleucine and leucine and the major amines were tyramine, cadaverine and histamine. Room temperature (25°C) promoted a significant increase of the contents of valine, leucine, tyramine and putrescine, expressed as g/kg of dry matter. These two free amino acids and two biogenic amines may serve as indicators of temperatures changes in ripened cheese.  相似文献

Shelf life (onset of sensory spoilage) and potential for toxin production by Clostridium botulinum type E in retail type packages of fresh catfish fillets in high barrier film were investigated under selected atmospheres when stored under refrigeration and temperature-abuse conditions. Shelf life of fillets in all atmospheres decreased with increase of storage temperature from 4°C to 16°C. Trimethylamine content associated with onset of spoilage was different for each storage temperature and atmosphere. Surface pH and K-values were not good indicators of onset of sensory spoilage. Toxin development coincided with sensory spoilage at 16°C storage for fillets packaged in either atmosphere. At 4°C, none of the MA-packaged fillets became toxic, even after 37 days of sensory spoilage.  相似文献

This study was conducted to determine the initial and spoilage microbiota of Pacific white shrimp during ambient and cold storage using next‐generation sequencing (NGS) and a culture‐dependent method. The quality changes were also evaluated by sensory analysis and total volatile basic nitrogen (TVB‐N) values. After 1 d of storage, the psychrotrophic bacteria were only 5.97 log CFU/g, accounting for 1.1% of the mesophilic bacteria counts (7.94 log CFU/g). The psychrotrophic bacteria counts exceeded the counts of mesophilic bacteria for shrimp stored at 4 °C after 6 d of storage, indicating that psychrotrophic bacteria became predominant. The NGS was used to identify the bacterial species in samples stored at 25 and 4 °C. The results showed that the dominant microorganisms were Vibrio at 25 °C, and Acinetobacter, Psychrobacter, and Shewanella at 4 °C. By the culture‐dependent method based on 16S rRNA gene and VITEK®2 CompactA system, it showed that the dominant microorganisms were Proteus spp. at 25 °C, and Shewanella putrefaciens, Acinetobacter johnsonii, and Aeromonas sobria at 4 °C. In conclusion, differences in results of microbiota analyzed by culture dependent and independent approaches were observed. The combination of both methodologies may provide more comprehensive information about the dominant spoilage microbiota in Pacific white shrimp during ambient and cold storage.  相似文献

The present work aimed to investigate the influence of freezing (−20 °C for 6 months) on the susceptibility of hake to chemical, microbiological and sensory spoilage once thawed and stored in ice. Thus, volatile and biogenic amines, counts of psychrotrophic and mesophilic bacteria, enterobacteria, pseudomonads and Shewanella together with sensory evaluation (of raw and cooked samples) were monitored during 14 days of iced storage in both fresh and frozen-thawed hake (Merluccius merluccius). At the beginning of ice storage, freshness parameters were equivalent in both batches (fresh and frozen-thawed hake), but different behaviour was observed during the iced storage. Overall, levels of volatile and biogenic amines increased much later and then decreased to lower values in frozen-thawed hake in accordance with the delay of microbial development in comparison with fresh hake. On the contrary, sensory spoilage occurred earlier in frozen-thawed hake. Therefore, the usual accepted or regulated limits of acceptability of chemical and microbiological parameters would not be suitable for freshness assessment of frozen-thawed hake.  相似文献

Gastrodia elata tuber is vulnerable to spoilage after harvested. A fungal strain Geotrichum pseudocandidum causing the spoilage was isolated from Gastrodia elata tuber and identified in this study. The log phase of the strain growth during incubation at 28 °C was from 24 to 48 h. The pH for highest spore production and largest colony formation were 5–6 and 7–9, respectively, while the temperature for them was 20–30 °C. Antifungal natamycin and tea polyphenols extract inhibited the stain at minimum inhibitory concentration of 0.25 and 0.5 mg mL−1, respectively. A short-time heating treatment at temperature at 65 °C for 10 min also effectively prevented the strain growth. As tea extract is a GRAS (generally recognised as safe) food ingredient, the tea extract treatment could be a safe and non-thermal treatment to prevent the spoilage of Gastrodia elata tuber with less loss of heat-sensitive bioactives and other nutrients.  相似文献

BACKGROUND: Changes in the biogenic amine content of tilapia (Oreochromis mossambicus) with time during iced and ambient temperature (28 °C) storage were quantified by high‐performance liquid chromatography in a gradient elution system using acetonitrile and water as solvents. RESULTS: The levels of amines in tilapia were significantly low even after a long storage period (18 h under ambient temperature storage and 15 days under iced storage). All amines except spermidine and spermine increased with time during both iced and ambient temperature storage. Spermidine and spermine showed a decrease with increasing storage time. CONCLUSION: Being euryhaline (tolerant of a wide range of salinity) in nature, tilapia shows significantly low quantities of biogenic amines even after a long period of ambient temperature or iced storage, unlike most other marine fish. ABSTRACT: The formation of biogenic amines in fish sauce made from fresh and temperature-abused (left at 35°C for 8 and 16 h) Indian anchovy ( Stolephorus indicus ) was investigated. Histamine, cadaverine, putrescine, and tyramine were the predominant biogenic amines found in anchovy left at 35°C for 16 h and its fish sauce product. Changes of biogenic amines were subtle during the course of fermentation at room temperature (RT) and at 40°C, suggesting that the main source of biogenic amines was associated with raw material, rather than with the fermentation process. Soluble peptide and total nitrogen of fish sauce prepared from temperature-abused anchovy were higher at the initial stage of fermentation at RT and 40°C and became comparable to those prepared from fresh anchovy at the end of fermentation. Total free amino acid contents of samples fermented at RT for 52 wk (7208.3 to 8473.6 mg/100 mL) were higher than those fermented at 40°C for 13 wk (4560.9 to 5730.9 mg/100 mL). Fish sauce prepared from temperature-abused anchovy contained less free histidine and arginine. Fish sauce of a good quality was obtained using fresh anchovy fermented at RT. Besides total nitrogen content, biogenic amines should be considered as quality indicators of fish sauce.  相似文献

Quality and safety parameters of Atlantic bonito gravad during 42 days of storage at 7 ± 1 °C were analysed using monitoring sensory quality, microbial contamination, nucleotide degradation products, biogenic amines (BA), trimethylamine (TMA) and thiobarbituric acid reactive substances (TBARS). The shelf‐life of vacuum packed Atlantic bonito gravad was found to be 28 days. Total viable count (TVC) did not exceed 6 log CFU g?1. Malonodialdehyde (MDA) content did not surpass 10 mg kg?1. Nine biogenic amines were detected. Histamine level at the end of the storage was 355.3 mg kg?1. TMA increased from 53.9 to 619.1 mg kg?1. Among indicators based on the products of nucleotide degradation, the H value is best correlated with the sensory assessment. It can be concluded that the storage temperature of bonito gravad must be below 7 °C to prevent the threat to safety and quality posed by biogenic amines, especially histamine.  相似文献
