Chronic use of high oxytocin (OT) dosages can cause a reduced response to endogenous OT. In this study the OT dosages used in the milking practice of 82 dairy cow farms were recorded. The OT dosages per cow used were high, especially when injected i.m. (23 ± 2 IU) compared with i.v. (7 ± 1 IU). In addition, the minimum OT dosages needed to obtain normal milk removal in cows with disturbed milk ejection were investigated. Seventeen cows routinely treated with OT during milking (group T) and 17 cows without previous OT treatment were used (group C). After cessation of spontaneous milk flow, both T and C groups were injected i.v. with a low dosage of OT (0.2 or 0.5 IU/cow). The time from injection until cessation of the OT-induced milk flow was recorded (response phase). The response phase and the amounts of removed milk by effect of the OT injection increased with increasing OT dosage. Values for 0.2 and 0.5 IU/cow of OT injected i.v. were (response phase and amount of milk removed) 198 ± 27 and 302 ± 18 s and 3.4 ± 0.7 kg and 6.5 ± 1.3 kg, respectively, for the C group, and 157 ± 15 and 221 ± 16 s and 3.2 ± 0.5 and 5.5 ± 1.0 kg, respectively, for the T group. Within 20 min of the OT injection, plasma concentrations returned to basal levels. The threshold OT concentration at cessation of milk flow after injection of 0.2 or 0.5 IU/cow of OT was calculated based on the OT plasma half-life. The threshold increased with increasing dosages of OT and was higher in group T (8 ± 1 and 14 ± 1 pg/mL for 0.2 and 0.5 IU/cow, respectively) than in group C (7 ± 1 and 11 ± 1 pg/mL for 0.2 and 0.5 IU/cow, respectively). In conclusion, desensitization of the udder toward OT occurs when the udder is exposed to elevated OT plasma concentrations, both short-term during the actual milking and long-term due to chronic high-dosage OT treatment. Feed management is one of the principal levers by which the production and composition of milk by dairy cows can be modulated in the short term. The response of milk yield and milk composition to variations in either energy or protein supplies is well known. However, in practice, dietary supplies of energy and protein vary simultaneously, and their interaction is still not well understood. The objective of this trial was to determine whether energy and protein interacted in their effects on milk production and milk composition and whether the response to changes in the diets depended on the parity and potential production of cows. From the results, a model was built to predict the response of milk yield and milk composition to simultaneous variations in energy and protein supplies relative to requirements of cows. Nine treatments, defined by their energy and protein supplies, were applied to 48 cows divided into 4 homogeneous groups (primiparous or multiparous × high or low milk potential) over three 4-wk periods. The control treatment was calculated to cover the predicted requirements of the group of cows in the middle of the trial and was applied to each cow. The other 8 treatments corresponded to fixed supplies of energy and protein, higher or lower than those of the control treatment. The results highlighted a significant energy × protein interaction not only on milk yield but also on protein content and yield. The response of milk yield to energy supply was zero with a negative protein balance and increased with protein supply equal to or higher than requirements. The response of milk yield to changes in the diet was greater for cows with high production potential than for those with low production potential, and the response of milk protein content was higher for primiparous cows than for multiparous cows. The model for the response of milk yield, protein yield, and protein content obtained in this trial made it possible to predict more accurately the variations in production and composition of milk relative to the potential of the cow because of changes in diet composition. Milking stall dimensions have not been adapted to the increase in cow body size caused by selection for better milking performance over the past decades. Improper milking stall dimensions might limit cow comfort, could lead to stress responses during milking and thus could negatively affect cow welfare. A crossover study was conducted in an experimental milking parlor that was converted from a herringbone (HB) to a side-by-side (SBS) parlor. The milking stall dimensions were modified in length and width and for HB also in depth (perpendicular distance between rump rail and breast rail). The stall dimensions applied during the experiments ranged from much smaller than common in European dairy farming to much larger. Treatments were applied for 2 wk per milking parlor type. In each milking parlor type, a total of 30 cows, kept in 2 groups were observed during milking for behavioral and physiological stress responses and for milking performance. In addition, milk cortisol levels and somatic cell counts were measured at the end of the 2-wk period. Outcome variables were selected based on a principal component analysis and analyzed using mixed effects models reflecting the experimental design. The results showed that the first cow per milking batch required more time (on average >40 s) to enter very small HB stalls than to enter small, large and very large stalls (<30 s). Also, cows yielded more milk per milking in very small and very large HB stalls than in the small and large milking stall dimensions [very small: 15.8 kg; 95% confidence interval (CI), 14.2–17.4 kg); small: 14.3 kg (95% CI, 12.8–15.9 kg); large: 14.6 kg (95% CI, 13.1–16.1 kg); very large: 16.1 kg (95% CI, 14.6–17.6 kg)]. The other behavioral, physiological and milk flow parameters as well as udder health were not affected by stall dimensions. For the SBS parlor, effects of milking stall dimensions were not detectable in any of the parameters. Despite the strong avoidance behavior to enter the milking parlor (measured as latency), no acute stress responses were found during milking. However, the study cannot exclude long-term effects of narrow stall dimensions on stress levels and possibly udder health, which should be investigated in future studies. 相似文献

Two experiments were conducted to determine whether longer-term deficiencies in the supply of limiting amino acids would be accompanied by a decline in mammary function (total DNA, cell proliferation rate and activities of key enzymes), and whether this would adversely affect the cow's ability to respond to a return to a nutritionally adequate diet. The first experiment was performed in early/mid lactation, and the second, using the same cows, was carried out in mid/late lactation. A control group of six cows were given a grass silage-cereal diet containing fish meal as the sole protein supplement (amino acid adequate) throughout the experiments, whereas another group of six cows in treatment received the control diet for 2 wk (lactation wk 5 and 6) and then were changed to a diet in which the fish meal was replaced by an equivalent amount of protein as feather meal (amino acid deficient) for 6 wk before returning to the fish meal diet for 4 wk (Experiment 1). After a rest period of 5 wk, the experimental procedure was repeated (Experiment 2). Although there was a fall in milk yield as lactation advanced, leading to lower milk yields in Experiment 2, the marked difference in milk yield between treatments was similar for the two stages of lactation (21% vs 16% in Experiment 1 and 2, respectively). In both experiments, the marked fall of milk yield in cows given the feather meal diet was completely recovered by a return to the fish meal diet. Despite the markedly lower milk yield with the amino acid-deficient diet, however, there was no clear evidence of corresponding changes in measurements of mammary function. 相似文献

Two experiments were conducted to study changes induced by stage of lactation and milk ejection in the cisternal compartment of the udder in dairy cows. In experiment 1, 18 cows grouped according to stage of lactation were used 12 h after milking for measuring alveolar and cisternal milk volumes (by cannula) and cisternal area (by ultrasonography) in the front quarters. Cisternal milk and cisternal area were correlated (r = 0.74 to 0.82) for all stages of lactation. As lactation advanced, volumes of alveolar and cisternal milk and cisternal area decreased. Proportion of cisternal milk varied between stages (early, 33.2%; mid, 23.1%; and late, 42.6%). In experiment 2, 7 cows were used to show return of milk from cisternal to alveolar compartments when milk ejection was induced without milking. Cisternal area was measured before (0 min) and after (3, 15, 30, and 60 min) an i.v. oxytocin (OT) injection administered immediately before normal a.m. and p.m. milking times. Cisternal area increased dramatically from 0 to 3 min (98%) and decreased slowly thereafter. The 0- and 3-min data provide clear evidence of milk ejection, and their difference indicated cistern elasticity. Maximum cisternal area in each cow was similar for the 8- and 16-h milking intervals, indicating that in both cases the cistern was completely full of milk. Decrease in cisternal area after 3 min was significant at 15, 30, and 60 min. Decreased cisternal area was interpreted as the reflux of cisternal milk to the alveolar compartment. We termed this 'cisternal recoil.' In conclusion, ultrasonography was a useful method to evaluate dynamic changes in cisternal milk throughout lactation and after udder stimulation in dairy cows. Evidence exists that udder cisterns decrease when lactation advances and milk returns to the alveolar compartment when cows remain unmilked after milk ejection. 相似文献

This paper investigates the impact of lameness on milk yield. The dataset includes approximately 8000 test-day milk yields from 900 cows on five farms in Gloucester, UK, collected over 18 mo from 1997 to 1999. The data were structured to account for repeated measures of test-day yield (1 to 10 per cow) and analyzed to account for this autocorrelation. Factors affecting milk yield included: farm of origin, stage of lactation, parity, and whether a cow ever became lame. In clinically lame cows, milk yield was reduced from up to 4 mo before a case of lameness was diagnosed and treated and for the 5 mo after treatment. The total mean estimated reduction in milk yield per 305-d lactation was approximately 360 kg. We conclude that clinical lameness has a significant impact on milk production. This is important information for assessing the economic impact of clinical lameness and its impact on cow health. It adds weight to the importance of early identification of clinical lameness and the urgency of techniques to improve the definition of this highly subjective diagnosis. 相似文献

This study investigated the effect of lameness, measured by serial locomotion scoring over a 12-mo period, on the milk yield of UK dairy cows. The data set consisted of 11,735 records of test-day yield and locomotion scores collected monthly from 1,400 cows kept on 7 farms. The data were analyzed in a multilevel linear regression model to account for the correlation of repeated measures of milk yield within cow. Factors affecting milk yield included farm of origin, stage of lactation, parity, season, and whether cows were ever lame or ever severely lame during the study period. Cows that had been severely lame 4, 6, and 8 mo previously gave 0.51 kg/d, 0.66 kg/d, and 1.55 kg/d less milk, respectively. A severe case of lameness in the first month of lactation reduced 305-d milk yield by 350 kg; this loss may be avoidable by prompt, effective treatment. Larger reductions can be expected when cases persist or recur. Evidence-based control plans are needed to reduce the incidence and prevalence of lameness in high yielding cows to improve welfare and productivity. 相似文献

A difficult calving affects the welfare of the cow and has economic implications for the farm. The degree of calving difficulty can vary from no assistance needed through a slight pull required to surgery being needed. With respect to milk production, it is not clear at which degree of calving difficulty adverse effects occur or for how long they last. Studies usually only consider the milk produced by animals who completed full lactations but the saleable milk production of the whole herd, regardless of each cow having achieved a full lactation, might be a better indicator of the productivity of the cows and the underlying stresses they experience, as well as being more representative of the real losses that producers incur. The objective of this study was to investigate how various degrees of calving difficulty would alter both the cow's milk production and their production of saleable milk over different stages of their subsequent lactation. The calving difficulty scores and the subsequent milk production were retrieved from an experimental dairy farm (in the United Kingdom) for 2 herds that contained 2,430 and 1,413 lactations. To account for milk saleable by the farmer, individual cumulative saleable milk yields, referred to as saleable milk yields (SMY), were calculated at 30, 60, 90, and 300 d in milk unconditional on the animal having achieved the lactation stage of interest. Lactation SMY were obtained based on the real lactation length achieved by the animal. Mean daily milk yields were also calculated for the same lactation stages as an estimate of the cow's milk production (CMP). Calving difficulty impaired milk production of dairy cows in terms of CMP and SMY in both herds, highlighting impaired income for dairy producers as well as detrimental effects to the productivity of the cows and potentially impaired health and survival. The management of the herd affected the presence of an effect of each degree of difficulty on SMY and CMP as well as its magnitude and duration. The analysis of SMY, independently of each animal having achieved a full lactation, could be a more sensitive indicator of the subsequent long-lasting biological stresses than CMP alone. 相似文献

The objective was to quantify the effect of postpartum uterine diseases on milk production and culling. Data from 2,178 Holstein cows in 6 herds enrolled in a randomized clinical trial were used. Milk production data from the first 4 Dairy Herd Improvement Association (DHIA) test-days and culling data from farm records were collected. Retained placenta (RP; ≥24 h after parturition) and metritis [≤20 d in milk (DIM)] were diagnosed by farm managers using standardized definitions. Farms were visited weekly and cows were examined at 35 and 56 (±3) DIM using endometrial cytology (cytobrush device), vaginal discharge scoring (Metricheck device), and measurement of cervical diameter by transrectal palpation. Diagnostic criteria for cytological endometritis (CYTO) and purulent vaginal discharge (PVD) were established based on a detrimental effect on subsequent reproduction. Statistical analyses were performed using linear mixed models, logistic regression models, and Cox proportional hazard models, accounting for the effects of experimental treatments and herd clustering. Milk production and culling were the outcomes. Primiparous and multiparous cows were modeled separately for milk production. Milk production of primiparous cows was unaffected by uterine diseases. The effect of metritis on milk production was variable over time in multiparous cows: it decreased production per cow by 3.7 kg at the first DHIA test, but was not different at later tests. Retained placenta decreased milk production by 2.6 kg/d in multiparous cows through the first 4 DHIA tests. The projected effects of metritis and RP in multiparous cows were reductions of 259 kg and 753 kg over 305 DIM, respectively; these effects were additive. Neither CYTO nor PVD affected milk production. Culling risks at 30 and 63 DIM were unaffected by RP and metritis. Culling hazard up to 300 DIM was unaffected by RP, metritis, CYTO, or PVD, whether or not pregnancy status, milk production, and displaced abomasum were accounted for. Uterine disease decreased pregnancy rate, which was a substantial risk factor for culling; however, if affected cows became pregnant they were not at greater risk of culling. 相似文献

The effects of postpartum milking strategy on plasma mineral concentrations, blood β-hydroxybutyrate (BHB) concentration, and colostrum, transition milk, and first monthly test milk yield and composition were evaluated in 90 multiparous Jersey and Jersey × Holstein crossbreed cows from a commercial farm. Before first postpartum milking, cows were randomly assigned to the following milking strategies, implemented during the first 2 d postpartum: twice-a-day milking (M2, standard industry practice, milking every 12 h; n = 22), once-a-day milking (M1, milking every 24 h; n = 24), restricted milking (MR, 3-L milking every 12 h; n = 21), and delayed milking (MD, no milking for the first 24 h, and milking every 12 h afterward; n = 23). Blood samples for total plasma Ca, P, and Mg determination were collected from enrollment every 4 h up to 48 h, and at 3 d in milk. Blood BHB concentration was determined at 3 and 11 d in milk. Colostrum and transition milk yields were recorded, and samples were collected at each study milking for IgG and somatic cell count (SCC) determinations. Information for first monthly test milk yield and composition was obtained from the Dairy Herd Improvement Association. Statistical analyses were conducted using generalized multiple linear and Poisson regressions with Dunnett adjustment and M2 as reference group for mean comparisons. Overall, plasma Ca concentration within 48 h after enrollment was higher for MD (2.17 mmol/L), tended to be higher for MR (2.15 mmol/L), and was similar for M1 (2.09 mmol/L) compared with M2 cows (2.06 mmol/L). No statistically significant differences compared with M2 cows were observed for plasma P and Mg concentrations. Colostrum and transition milk and total Ca harvested within 48 h after enrollment were lower for M1, MR, and MD compared with M2 cows. The MD strategy prevented harvesting colostrum with >50 g of IgG/L. No statistically significant effects were detected on plasma mineral concentrations at 3 DIM, blood BHB concentration, colostrum and transition milk SCC within 48 h after enrollment, or milk yield, energy-corrected milk yield, and SCC at first monthly test. Our results suggest that postpartum plasma Ca concentration may be influenced by postpartum milking strategy, without interfering with future milk yield and udder health. Further studies should evaluate whether the proposed milking strategies in early postpartum affect production, reproduction, or health. 相似文献

Recent surveys have identified the presence of perchlorate, a natural compound and environmental contaminant, in forages and dairy milk. The ingestion of perchlorate is of concern because of its ability to competitively inhibit iodide uptake by the thyroid and to impair synthesis of thyroid hormones. A recent study established that milk perchlorate concentrations in cattle highly correlate with perchlorate intake. However, there is evidence that up to 80% of dietary perchlorate is metabolized in clinically healthy cows, thereby restricting the available transfer of ingested perchlorate into milk. The influence of mastitis on milk perchlorate levels, where there is an increase in mammary vascular permeability and an influx of blood-derived components into milk, remains unknown. The present study examined the effect of experimentally induced mastitis on milk perchlorate levels in cows receiving normal and perchlorate-supplemented diets. Over a 12-d period, cows were ruminally infused with 1 L/d of water or water containing 8 mg of perchlorate. Five days after the initiation of ruminal infusions, experimental mastitis was induced by the intramammary infusion of 100 μg of bacterial lipopolysaccharide (LPS). Contralateral quarters infused with phosphate-buffered saline served as controls. A significant reduction in milk perchlorate concentration was observed in the LPS-challenged glands of animals ruminally infused with either water or perchlorate. In control glands, milk perchlorate concentrations remained constant throughout the study. A strong negative correlation was identified between mammary vascular permeability and milk perchlorate concentrations in LPS-infused glands. These findings, in the context of a recently published study, suggest that an active transport process is operative in the establishment of a perchlorate concentration gradient across the blood-mammary gland interface, and that increases in mammary epithelial and vascular endothelial permeability lead to a net outflow of milk perchlorate. The overall finding that mastitis results in lower milk perchlorate concentrations suggests that changes in udder health do not necessitate increased screening of milk for perchlorate. 相似文献

In experiment I, the effect of i.m. oxytocin (OT) injection (50 IU) on OT blood pattern was tested. Blood samples from 6 cows were collected for 2 h after OT injection either with or without milking. To test the effect of i.m. OT injection (50 IU) on milk ejection efficiency, intramammary pressure (IMP) was measured in 13 cows (experiment II). Milk ejection was induced by manual teat stimulation. After IMP increased and reached a plateau, OT was injected. In experiment III, the effect of chronic OT treatment on mammary gland sensitivity and endogenous OT release was tested. For 19 d, cows were i.m. injected at each milking with 50 IU OT (n = 13) or 5 mL of NaCl 0.9% (n = 14) 1 min before the start of udder preparation. To test mammary gland sensitivity, IMP recording was performed after a long (11 h, 7 OT cows, and 7 NaCl cows) and a short (3 h, 6 OT cows, and 7 NaCl cows) milking interval at d -1 and d 18. To test the effect of withdrawal of chronic i.m. treatment on OT release, blood sample collection was performed during evening milking at d 0 and d 19. Intramuscular oxytocin injection (experiment I) caused elevated OT blood levels observed at least for 2 h and showed an even more pronounced effect when milking was also performed. Intramuscular OT injection after teat stimulation (experiment II) caused additional milk ejection but only in 6 out of 13 cows. Withdrawal of chronic OT treatment (experiment III) did not reduce OT release during milking. However, ejection time was prolonged during OT infusion after a long milking interval. Ejection pressure tended to be lower after a short milking interval. It seems that the reduction of spontaneous milk removal after chronic OT treatment was due to reduced contractibility of myoepithelial cells in the mammary gland at a physiological range of OT concentrations. 相似文献

Increased milking frequency (IMF) during early lactation has the potential for carryover responses following the return to normal herd milking frequency. The objective was to determine the consistency of response of cows in commercial dairy farms to IMF during early lactation. Cows (n = 398) were assigned randomly at calving within each of the 4 participating farms to 1 of 2 treatments. The control group was milked twice-daily (2×) during the entire lactation. The IMF group was milked 4-times daily (4×) starting on d 1 to 7, depending on farm, until d 21 postcalving and 2× thereafter. Cows in the IMF group were milked at the beginning and again at the end of the normal milking routine. Milking intervals differed across the farms for the 4× cows with a minimum interval of 3.5, 4.0, 5.0, and 6 h for each of the 4 farms, respectively. The milk yield of cows subjected to IMF increased by 2.2 ± 0.4 kg/d during the first 7 mo of lactation. Interactions of treatment with lactation group (primiparous vs. multiparous) were not significant. Although percentages of fat and protein in milk were decreased by early lactation IMF (3.69% ± 0.03 fat and 3.05% ± 0.02 true protein for control vs. 3.57% ± 0.03 fat and 2.99% ± 0.02 true protein for IMF), overall yields of protein were increased by IMF (1.02 ± 0.01 vs. 0.98 ± 0.01 kg/d). Early lactation IMF did not affect udder health as assessed by somatic cell count linear score. Cows subjected to IMF were 1.4 times more likely classified as subclinically ketotic than the control cows. Early lactation IMF has the potential to increase milk yield on commercial dairy farms. Although the direction of response was the same on all farms, the magnitude of the response was different among farms and appears influenced by management practices specific to each farm, which included, but were not limited to, housing system, stocking density, nutrition, genetics, and other covariates differing among farms. 相似文献

Previously observed strong relationships between dry matter (DM) intake and milk yield in dairy cows were the basis for this meta-analysis aimed to determine the influence of intake of specific dietary nutrients on milk yield and milk protein yield in Holstein dairy cows. Diets (563) from feeding trials published in the Journal of Dairy Science were evaluated for nutrient composition using 2 diet evaluation programs. Intake of nutrients was estimated based on DM intake and program-derived diet composition. Data were analyzed with and without the effect of stage of lactation. Models based on intake of nutrients improved prediction of milk yield and milk protein yield compared with DM intake alone. Intake of net energy of lactation was the dominant variable in milk yield prediction models derived from both diet evaluation models. Milk protein yield models also improved prediction over the DM intake model. These models were dominated by ruminally undegradable protein intake and included a number of energy-related intake variables. In most models, incorporating stage of lactation improved the model fit. 相似文献

Periparturient hypocalcemia is frequently observed and considered as a gateway disease that is associated with various health issues. The objective of this study was to evaluate the association of hypocalcemia with early-lactation milk yield, reproductive performance, and culling across a large number of different managerial systems. A prospective cohort study was conducted based on a convenience sample of 125 dairy herds from 8 federal states of Germany between February 2015 and August 2016. A blood sample was drawn from 1,709 animals within 48 h after parturition and analyzed for serum calcium concentration. After discarding cows (n = 283) with missing data, a total of 1,426 cows were considered for final analyses. The median time from calving to sampling was 14.0 h (interquartile range = 5.0–24.9 h). For each herd, a record of the herd management software was requested 150 d after the last cow was sampled. Serum calcium concentration of each cow was associated with early-lactation milk yield (Dairy Herd Improvement Association equivalent test 1 to 3), reproductive performance [days in milk (DIM) at first artificial insemination (AI), pregnancy at first AI, time to pregnancy within 150 DIM], and culling (until 60 DIM) data. Generalized linear mixed models were used to analyze continuous or categorical data. Shared frailty models were used for time to event data. Five different thresholds were used to define hypocalcemia. Thresholds ranged from 1.8 to 2.2 mmol/L using 0.1-mmol/L increments. Clinical hypocalcemia was defined as serum calcium concentration <2.0 mmol/L in combination with clinical signs (e.g., recumbency). The effect of hypocalcemia on milk yield was conditional on parity. In primiparous cows a serum calcium concentration <2.0 mmol/L (6.4% of cows were below this threshold) had no effect on milk production, whereas there was a tendency for multiparous cows with a serum calcium concentration <2.1 mmol/L (63.2% of cows were below this threshold) to produce 0.80 kg/d more milk compared with multiparous cows at or above the threshold. Multiparous cows suffering from clinical hypocalcemia produced 2.19 kg/d less milk compared with normocalcemic cows in early lactation. Calcium status was not associated with days to first insemination. Cows with a serum calcium concentration <1.9 mmol/L (34.6% of cows below this threshold) had decreased odds (odds ratio = 0.56) of pregnancy at first AI. A serum calcium concentration <1.8 mmol/L (24.1% of cows below this threshold) had a significant effect on time to pregnancy. Compared with animals with a serum calcium concentration ≥1.8 mmol/L, the hazard of becoming pregnant within 150 DIM was reduced when cows had a serum calcium concentration <1.8 mmol/L (hazard ratio = 0.68). Cows with a serum calcium concentration <2.0 mmol/L (44.3% of cows were below this threshold) had a 1.69 times greater hazard of being culled within the first 60 DIM compared with normocalcemic animals. The present study shows that the association of hypocalcemia with milk yield was conditional on parity and serum calcium concentration measured once within 48 h after calving. Diet composition defines the amount and type of nutrients absorbed by dairy cows. Endocrine-metabolic interactions can influence these parameters, and so nutrient availability for the mammary gland can significantly vary and affect milk yield and its composition. Six dairy cows in early and then late lactation received, for 28 d in a changeover design, 2 diets designed to provide, within the same stage of lactation, similar amounts of rumen fermentable material but either high starch plus sugar (HS) content or low starch plus sugar content (LS). All diets had similar dietary crude protein and calculated supply of essential amino acids. Dry matter intake within each stage of lactation was similar between groups. Milk yield was similar between groups in early lactation, whereas a higher milk yield was observed in late lactation when feeding HS. At the metabolic level, the main difference observed between the diets in both stages of lactation was lower blood glucose in cows fed LS. The lower glucose availability during consumption of LS caused substantial modifications in the circulating and postprandial pattern of metabolic hormones. Feeding LS versus HS resulted in an increase in the ratio of bovine somatotropin to insulin. This increased mobilization of lipid reserves resulted in higher blood concentrations of nonesterified fatty acids and β-hydroxybutyrate, which contributed to the higher milk fat content in both stages of lactation in the LS group. This greater recourse to body fat stores was confirmed by the greater loss of body weight during early lactation and the slower recovery of body weight in late lactation in cows fed LS. The lower insulin to glucagon ratio observed in cows fed LS in early and late lactation likely caused an increase in hepatic uptake and catabolism of amino acids, as confirmed by the higher blood urea concentrations. Despite the higher catabolism of amino acids in LS in early lactation, similar milk protein output was observed for both diets, suggesting similar availability of amino acids for peripheral tissue and mammary gland. The latter could be the result of sparing of amino acids at the gut level due to starch that escaped from the rumen, and to the balanced amino acid profile of digestible protein. This last aspect appears worthy of further research, with the aim to enhance the efficiency of protein metabolism of dairy cows, reducing environmental nitrogen pollution without affecting milk yield potential. 相似文献

研究发酵黄芪粉对奶牛产奶量及乳品质的影响.对照组奶牛饲喂基础日粮,试验Ⅰ、Ⅱ和Ⅲ组奶牛分别饲喂10、20和30 g/(头·d)的发酵黄芪粉基础日粮.结果表明:与对照组相比,试验组奶牛产奶量增加了0.95、1.45和1.58 kg/(头·d),但试验Ⅱ、Ⅲ组组间差异不显著;可显著提高奶牛乳蛋白率,但对奶牛乳脂率、乳糖及非脂固型物均无显著影响;可降低奶牛乳体细胞数,其中试验Ⅱ、Ⅲ组奶牛乳体细胞数显著低于对照组和试验Ⅰ组,但试验Ⅱ、Ⅲ组组间差异不显著;可提高奶牛经济效益,各试验组奶牛经济效益分别高于对照组2.6、3.4和2.72元/(头·d).该试验条件下,发酵黄芪粉以20 g/(头·d)的添加量较好. 相似文献

The present study was conducted to assess rumen bacteria in lactating cows with different milk protein yield, aiming to understand the role of rumen bacteria in this trait. Cows with high milk protein yield (high milk yield and high milk protein content, HH; n = 20) and low milk protein yield (low milk yield and low milk protein content, LL; n = 20) were selected from 374 mid-lactation Holstein dairy cows fed a high-grain diet. Measurement of the rumen fermentation products showed that the concentrations of ruminal total volatile fatty acids, propionate, butyrate, and valerate and the proportion of isobutyrate were higher in the HH cows than in the LL cows. Amplicon sequencing analysis of the rumen bacterial community revealed that the richness (Chao 1 index) of rumen microbiota was higher in the LL cows than in the HH cows. Among the 10 predominant bacterial phyla (relative abundance being >0.10%, present in >60% of animals within each group), the relative abundance of Proteobacteria was 1.36-fold higher in the HH cows than in the LL cows. At the genus level, the relative abundance of Succinivibrio was significantly higher and that of Clostridium tended to be higher in the LL cows than in the HH cows. Sharpea was 2.28-fold enriched in the HH cows compared with the LL cows. Different relationships between the relative abundances of rumen microbial taxa and volatile fatty acid concentrations were observed in the HH and the LL animals, respectively. Succinivibrio and Prevotella were positively correlated with acetate, propionate, and valerate in the LL cows, whereas Sharpea was positively correlated with propionate and valerate concentrations in the HH cows. Collectively, our results revealed that rumen bacterial richness and the relative abundances of several bacterial taxa significantly differed between dairy cows with high and low milk protein yields, suggesting the potential roles of rumen microbiota contributing to milk protein yield in dairy cows. 相似文献

A comparative study was performed to evaluate differences in milk yield between an automatic milking system (AMS) and a conventional herringbone milking parlor system. Two herds of Italian-Friesian cows were reared in the same barn, located in the Po Valley in northern Italy. Twenty-five primiparous cows and 10 multiparous cows were milked with an AMS, while at the same time 29 primiparous and 9 multiparous were milked twice daily in a milking parlor on the other side of the barn. A selection gate allowed cows to access the AMS only if the interval from last milking was >5 h. Multiparous cows in the AMS yielded more milk than multiparous cows in the milking parlor (34.2 ± 0.7 vs. 29.4 ± 0.6 kg/d). There was no difference in milk yield between primiparous cows in the AMS and in the milking parlor (28.9 ± 0.4 vs. 28.8 ± 0.3 kg/d). Milking frequency in the AMS was significantly higher in primiparous (2.8 ± 0.03) than in multiparous cows (2.5 ± 0.04). The hot season negatively affected milk yield; the milk yield reduction was higher for cows milked with the AMS (−4.5 ± 0.6 kg/d) than in the milking parlor (−3.0 ± 0.8 kg/d). In the AMS, milking frequency decreased during the hot season in primiparous cows (−0.3 ± 0.1). We concluded that a positive AMS effect on milk yield is possible, but that steps must be taken to alleviate the discomfort involved with attracting cows to the AMS. 相似文献

This study investigated the effect of 2 different types of long-acting insulin on milk production, milk composition, and metabolism in lactating dairy cows. Multiparous cows (n = 30) averaging 88 d in milk were assigned to one of 3 treatments in a completely randomized design. Treatments consisted of control (C), Humulin-N (H; Eli Lilly and Company, Indianapolis, IN), and insulin glargine (L). The H and L treatments were administered twice daily at 12-h intervals via subcutaneous injection for 10 d. Cows were milked twice daily, and milk composition was determined every other day. Mammary biopsies were conducted on d 11, and mammary proteins extracted from the biopsies were analyzed by Western blot for components of insulin and mammalian target of rapamycin signaling pathways. Treatment had no effect on dry matter intake or milk yield. Treatment with both forms of long-acting insulin increased milk protein content and tended to increase milk protein yield over the 10-d treatment period. Analysis of milk N fractions from samples collected on d 10 of treatment suggested that cows administered L tended to have higher yields of milk protein fractions than cows administered H. Milk fat content and yield tended to be increased for cows administered long-acting insulins. Lactose content and yields were decreased by treatment with long-acting insulins. Administration of long-acting insulins, particularly L, tended to shift milk fatty acid composition toward increased short- and medium-chain fatty acids and decreased long-chain fatty acids. Plasma concentrations of glucose and urea N were lower for cows administered long-acting insulins; interactions of treatment and sampling time were indicative of more pronounced effects of L than H on these metabolites. Concentrations of nonesterified fatty acids and insulin were increased in cows administered long-acting insulins. Decreased concentrations of urea N in both plasma and milk suggested more efficient use of N in cows administered long-acting insulins. Western blot analysis of mammary tissue collected by biopsy indicated that the ratios of phosphorylated protein kinase b (Akt) to total Akt and phosphorylated ribosomal protein S6 (rpS6) to total rpS6 were not affected by long-acting insulins. Modestly elevating insulin activity in lactating dairy cows using long-acting insulins altered milk composition and metabolism. Future research should explore mechanisms by which either insulin concentrations or insulin signaling pathways in the mammary gland can be altered to enhance milk fat and protein production. 相似文献
