Separation of adult rat bone marrow cells by the method of thin layer countercurrent distribution permits the analyses of 59Fe-tagged erythroid cells for the various multiple hemoglobins and the assignment of such hemoglobins to erythroid cells at different stages of their development. Of the six adult red cell hemoglobins, hemoglobin 5 is synthesized most actively in the earliest erythroid cell whereas hemoglobin 4 (the major hemoglobin of the red cell) is synthesized most actively in the latest erythroid cells, e.g. the reticulocyte. Experimental evidence also indicates that maturation of the erythroid cell is accompanied by a decreased rate of synthesis of hemoglobin 5. The earliest erythroid cells of the marrow contain two hemoglobins, 7 and 8, which are absent in the adult red cell. Similar studies with the guinea pig confirm the nonuniform biosynthesis of its two hemoglobins and suggest that the phenomenon may be a general one among mammalian multiple hemoglobins. 相似文献

Species differences in behavior and responses of guinea pig (GPT) and rat (RT) tracheal smooth muscle to electrical field stimulation (EFS) and chemicals were investigated under semi-isometric conditions. Indomethacin augmented and papaverine reduced the contractions elicited by EFS in both tissues. They relaxed the GPT and did not affect the tone decline in the RT. Experiments with different load, cartilage content, thread and rubber strips suggest the role of elastic elements in passive elongation of the RT. This was independent of muscarinic, adrenergic, histaminergic and serotoninergic receptors, nitric oxide, eicosanoids and metabolic processes. The second response on repeated administration of acetylcholine was augmented and that of serotonin attenuated in both GPT and RT. Upon repeated elevation of [K+]o the tissues responded in an opposite manner. Further data are provided on similarities (modulation of cholinergic transmission by prostaglandins, sensitization to acetylcholine and desensitization to serotonin) and differences (innervation, responses to repeated elevation of [K+]o, presence/absence of prostaglandin-regulated basal tone and spontaneous activity) in reactivity of GPT and RT. 相似文献

This study describes differences between guinea pig and rat in the immunoreactivities for calbindin (CB-IR) and parvalbumin (PV-IR) in cartwheel (CWC) and Purkinje-like (PLC) cells of the dorsal cochlear nucleus (DCN). CWCs are the most important inhibitory interneurons of the DCN. Their soma and dendrites stain intensely for CB-IR in guinea pigs but only weakly and incompletely in rats. In both species, the CWCs do not show PV-IR. PLCs, a rare type of DCN cells often interpreted as displaced cerebellar Purkinje cells misrouted during migration, are known from rat and mouse and are here described for guinea pig DCN. PLCs are intensely and completely stained for CB-IR and PV-IR in guinea pigs. In rats, they stain with similar completeness only for CB-IR, PV-IR being weak and restricted to the cell's soma. Similar staining differences between the two species are seen with the cerebellar Purkinje cells, i.e., PLCs resemble the cerebellar Purkinje cells more than do the CWCs. Based on the present material (and preliminary findings in a primate (marmoset), we speculate that the PLCs have their place in the circuitry of the DCN receiving input via parallel fibers, like the CWCs, and possibly projecting their axon onto the cerebellum. 相似文献

The tooth interface with resin-modified glass-ionomer cements (RM GICs) is poorly understood. This study examined the interface, especially with dentin. Cervical cavities in extracted teeth were restored with Fuji II LC, Vitremer, Photac-Fil, or a conventional GIC, Fuji Cap II. Fluorescent dye was placed in the pulp chambers for 3 hrs before the specimens were sectioned. Examination of the tooth/material interface with a confocal microscope showed that dye uptake by the restoration varied among materials. A "structureless", non-particulate, highly-stained layer of GIC was observed next to dentin in Fuji II LC. This layer varied in width, was prominent where the dentin tubules were cut "end-on" and in areas closer to the pulp, and was not seen adjacent to enamel. Vitremer showed minimal dye uptake, and the "structureless" layer was barely discernible. Photac-Fil showed more uniform uptake and absence of this layer. Cracking of enamel was also noted with these materials. The conventional GIC did not show any dye uptake, presence of a "structureless" layer, or enamel cracking. We elucidated the potential mechanisms involved in the formation of a "structureless" interfacial layer in Fuji II LC by studying the variables of cavity design, surface pre-treatment, water content of the tooth, time for it to develop, early finishing, and coating of the restoration. This layer, the "absorption layer", is probably related to water flux within the maturing cement, depending on environmental moisture changes and communication with the pulp in a wet tooth. Inhibition by taurine of the inwardly rectifying K+ current in guinea pig ventricular cardiomyocytes

Effects of taurine on the inwardly rectifying K+ current (IK1) in isolated guinea pig ventricular cardiomyocytes were examined using patch voltage-clamp methods. All experiments were performed at 36 degrees C. Taurine (10-20 mM) increased the action potential duration, but failed to affect the resting potential. Holding potential was maintained at -30 mV. The current was activated with an inwardly going rectification, and was completely blocked by Ba2+ (2 mM). Taurine inhibited IK1 at - 120 mV by 28.3+/-1.1% (n=6, P < 0.05) at 10 mM and by 36.0+/-2.1% (n=6, P < 0.01) at 20 mM. The reversal potential was shifted in the hyperpolarizing direction by 3.7+/-0.6 mV (n=6) at 20 mM. In inside-out patch-clamp experiments, the amplitude of unitary channels was -2.7+/-0.3 pA (n=21) at -90 mV. Symmetrical high-K+ (150 mM) solutions in both bath and pipette were used. The channel conductance was 32+/-2 pS (n=9). Taurine did not affect channel conductance, but markedly decreased the open probability at - 120 mV of channel by 21.5+/-2.4% (n=8, P < 0.01) at 10 mM, and by 56.7+/-3.8% (n=8, P < 0.001) at 20 mM. These responses were almost reversible. These results suggest that taurine directly modulates the open probability of the inwardly rectifying K+ current, resulting in regulation of the functions of heart cells. 相似文献

In this study the function of transforming growth factor-beta (TGF-beta) in preimplantation mouse embryos was examined. By RT-PCR, mRNA for the signalling type I (T beta R-I) and type II (T beta R-II) receptors for TGF-beta was shown to be present in two distinct time windows: in fertilized oocytes and at the blastocyst stage. The function of TGF-beta at these times was analysed in two ways. Firstly, the TGF-beta signalling pathway was blocked by injecting a DNA construct encoding a truncated T beta R-II, that acts as a dominant-negative receptor, in fertilized oocytes, and the effect on development was determined. Secondly, inner cell masses isolated at the blastocyst stage were cultured in vitro with and without TGF-beta under conditions that favour the outgrowth of parietal endoderm. The results show that TGF-beta signalling mediated by maternally expressed receptors is important for development of preimplantation embryos beyond the two-cell stage, and suggest a regulatory role for TGF-beta in the outgrowth of parietal endoderm. 相似文献

This article presents a new method for analyzing the spreading of skin erythemas. These occur as a result of the cutaneous vascular axon reflex which can be evoked by a noxious stimulation of the skin. Series of true-color images of the observed skin patch were recorded using a video camera. The images were digitized and stored on computer disk. The delineation of the reddening was segmented for every image of the sequence by a newly developed image processing method. Each image taken after the noxious stimulation was compared with the baseline before the stimulation and each image point was classified as: "unchanged" or "changed skin color." To improve the classification the CIE L*a*b* color space was used. The boundaries of the erythema were extracted from the resulting binary images. Every image of a sequence was analyzed in the same way in order to follow the time course of the flare response. The erythema reaction could be determined in an objective way using this methods. The automatically detected flare sizes were independent of human observers and had a high spatial and temporal resolution. It was used for a crossover study to assess the power of new drugs which modify the blood flow of the skin induced by an intradermal histamine application. 相似文献

Under scanning electron microscopy (SEM) the surface cells of the guinea pig bladder have pentagonal or hexagonal outlines. Their borders are clearly defined since they are elevated. They possess a large reserve of surface membrane which is markedly folded and wrinkled when the bladder is empty. The folds disappear and the cells become flat during distension. The luminal surface is characterized by numerous reticular ridges which are a remarkably constant feature and persist even under acute artificial distension. A small proportion of the surface cells are small and have less than five sides. Since they show only sparse microvilli as a surface feature, they have a smooth appearance. These are believed to be young surface cells which have just emerged from the intermediate layer, and have not yet acquired the ridged pattern of mature cells. 相似文献

This study examined the healing of articular defects, with and without carbon fiber implants, and the response of repair tissue to its interim removal in guinea pigs of different ages. These were investigated after the induction of full thickness articular cartilage defects in the patellar groove of skeletally mature and immature guinea pigs. To indicate its capacity for replacement after attrition, repair tissue occurring in untreated (control) and carbon fiber treated (experimental) defects was ablated after 8 weeks, and the animals were sacrificed after an additional 8 weeks. The repair tissue was studied quantitatively at gross and microscopic levels and qualitatively using scanning and transmission electron microscopic study. The principal findings were as follows. The initial formation of repair tissue was variable, but it occurred in most cases. Secondary repair tissue formation consistently occurred after excision. Age did not influence the degree of repair. Carbon fiber implants of the type used impaired healing of small full thickness articular cartilage defects, compared with no treatment. Microscopically, repair tissue contains five main cell types, each with a characteristic surrounding matrix. Intermediate forms also are found. These, together with four of the five main types comprise a morphologic continuum and fit readily into a proposed developmental sequence that may stem from the fibroblast. 相似文献

Adult chicken hepatocytes were obtained by an adaptation of the two step in situ collagenase perfusion. Usually 0.5 to 1 x 10(9) cells were obtained, with 75 to 95% viability. Hepatocytes attached within 2 h when plated on plastic cell culture dishes and spread in 4 h, surviving for several months in a specific serum-free medium. These cells retained a typical parenchymal cell morphology and the ability to produce a specific protein (albumin) throughout the culture period. We hereby provide a suitable model for studying hepatic metabolism in birds. 相似文献

Electrophysiological properties of guinea pig ambiguus (AMB) neurons were studied in a brainstem slice preparation. During subthreshold depolarization AMB neurons displayed an early slow depolarization and a late outward rectification both of which were blocked by replacing Ca2+ with Co2+ in the extracellular solution. AMB neurons showed hyperpolarizing inward rectification which was blocked by extracellular Cs+ and is likely caused by the activation of Ih: In 58% (n = 49) of AMB neurons spike firing was restricted to the early phase of a long-lasting depolarizing current injection (phasic firing). The remaining AMB neurons showed repetitive firing throughout the depolarization (tonic firing). A Ca(2+)-mediated K+ current (IK(Ca)) caused an afterhyperpolarization that followed both single and repetitive spike firing. IK(Ca) also controlled the firing pattern in both types of firing, especially in the phasic firing. Norepinephrine (NE) blocked both the hyperpolarizing inward rectification and the Ca(2+)-dependent AHP. These effects of NE were antagonized by propranolol. It is proposed that the blockade of IK(Ca) and Ih contribute to the improvement of the 'signal-to-noise ratio' by NE in AMB neurons. 相似文献

While there is agreement that unilateral vestibular deafferentation (UVD) invariably produces an immediate severe horizontal vestibulo-ocular reflex (HVOR) deficit, there is disagreement about whether or not this deficit recovers and, if so, whether it recovers fully or only partly. We suspected that this disagreement might mainly be due to experimental factors, such as the species studied, the means chosen to carry out the UVD, or the nature of the test stimulus used. Our aim was to sort out some of these factors. To do this, we studied the HVOR of alert guinea pigs in response to low and high acceleration sinusoidal and high acceleration impulses after UVD by either labyrinthectomy or by vestibular neurectomy. The HVOR in response to high acceleration impulsive yaw rotations was measured before, and at various times after, either unilateral labyrinthectomy or superior vestibular neurectomy. Following UVD, there was a severe impairment of the HVOR for ipsilesional rotations and a slight impairment for contralesional rotations, after either operation. This asymmetrical HVOR deficit in the guinea pig parallels the deficit observed in humans. Between the first measurement, which was made 1 week after UVD, and the last, which was made 3 months after UVD, there was no change in the HVOR. This lack of recovery was the same after labyrinthectomy as after vestibular neurectomy. The HVOR to low and high acceleration sinusoidal yaw rotations were measured after UVD, and the results were compared with those in response to impulsive rotations. For low acceleration sinusoidal rotations (250 degrees/s2), the gain was symmetrical, although reduced bilaterally. As the peak head acceleration increased, the HVOR became increasingly asymmetric. The HVOR asymmetry for sinusoidal rotations was significantly less than for impulsive rotations that had the same high peak head acceleration (2500 degrees/s2). Our results show that the HVOR deficit after UVD is the same in guinea pigs as in humans; that it is the same after vestibular neurectomy as after labyrinthectomy; that it is lasting and severe in response to high acceleration rotations; and, that it is more obvious in response to impulses than to sinusoids. 相似文献

Osteoclasts are multinuclear bone-resorbing cells which contain abundant mitochondria. Morphological studies have suggested that a correlation may exist between mitochondrial concentration and bone resorption by osteoclasts. However, investigation of mitochondrial transmembrane potential (delta psi) and volume has been hampered by the difficulty in obtaining a sufficient number of osteoclasts for assessing these characteristics by flow cytometric analysis. In this study, we have used confocal laser scanning microscopy after loading the cells with Rhodamine 123 and 10-nonyl Acridine Orange to record mitochondrial delta psi and volume, respectively, in isolated rat osteoclasts cultured on bovine bone slices. Optimal staining conditions were found to be 10 micrograms ml-1 for 40 min for Rhodamine, and 1 microM for 10 min for the 10-nonyl Acridine Orange derivative. Two osteoclast populations, whose shape seemed to reflect bone resorption and migratory functions, were identified depending on their shape and on the distribution of the two dye probes. 'Round-shaped' osteoclasts had significantly higher mitochondrial delta psi and volume in the apical regions than in the basolateral portions (p < 0.00001). In contrast, mitochondrial delta psi and volume in 'irregular-shaped' osteoclasts were rather evenly distributed in both these regions (p > 0.05). Our results indicate that there is an apical polarization of mitochondria in osteoclasts corresponding to the energy demands associated with bone resorption. 相似文献

Angiotensin II (AII) tachyphylaxis occurs in the guinea pig ileum, but is not induced by analogs lacking the N-terminal amino group or the Arg2 guanidino group. Both AII and Lys2AII increased cell inositol trisphoshate content in cultured intestinal smooth muscle cells. Protein kinase C inhibition by staurosporine or downregulation by prolonged incubation with phorbol reverted tachyphylaxis of the inositol trisphoshate response, but not that of the Na+ uptake response, indicating that the uncoupling of the phosphoinositide signal system by protein kinase C did not involve all processes distal to receptor activation. Tachyphylaxis of the Na+ uptake response was prevented when receptor internalization was blocked by reduction of the temperature (4 degrees C) or by pretreatment of the cells with phenylarsine oxide. Acid washings, which prevented tachyphylaxis of the 24Na+ influx response, also prevented tachyphylaxis of the contractile response of the guinea pig ileum to AII. Although these findings suggest that sequestration or internalization of the AII receptor might be involved in AII tachyphylaxis, binding of [125I]AII and of [125I]Lys2AII to the cells was equally unaffected by repeated administrations of the peptides. The results suggest that conformational change of the AII-receptor complex within the plasma membrane, but not internalization, is the most important factor responsible for tachyphylaxis. 相似文献

Following administration of 6-n-propylchromone-2-carboxylic acid (6-n-PCCA) (500 mumol/kg) to male rats, three metabolic products were detected and isolated from the 0-24 h urine. All were identified as resulting from oxidation exclusively along the 6-n-propyl moiety. Some 66% of the dose was excreted in the 0-24 h urine, 55% of which was 6-PCCA, with 15% as (6-1'-hydroxypropyl)chromone-2-carboxylic acid (6-1'-HPCCA), 22% as 6-(2'-hydroxypropyl)chromone-2-carboxylic acid (6-2'-HPCCA), and 4% as 6-3'-carboxypropyl)chromone-2-carboxylic acid (6-3'-CPCCA). Derivatization of the methyl esters of the hydroxylated metabolites with S-alpha-methoxy-alpha-(trifuloromethyl)-phenylacetyl chloride (Mosher's reagent) allowed the evaluation of urinary enantiomeric composition by HPLC and assignment of their absolute configurations by NMR. This was found to be 90:10 (R/S) for 6-2'-HPCCA, and 7:93 (R/S) for 6-1'-HPCCA. When rats were dosed with the racemic 1'- and 2-hydroxy metabolites; no stereoselective metabolism or excretion was observed. Administration of 6-n-PCCA to male guinea pigs revealed that this species was unable to metabolise this compound. 相似文献
