In Mediterranean regions where the population is rapidly growing, the risk of water resource contamination by wastewater is likely to increase. This is the case of the Hérault watershed (south of France), where the presence of treated wastewater in surface and ground waters has been shown in a previous study. To assess the consequence of these wastewater contaminations as regards pharmaceuticals and other organic compounds, 16 common pharmaceuticals (amitryptilin, acetylsalicylic acid, carbamazepine, clenbuterol, diazepam, diclofenac, doxepin, gemfibrozil, ibuprofen, imipramine, ketoprofen, naproxen, nordiazepam, paracetamol, salbutamol, and terbutalin) as well as wastewater related pollutants (caffeine, gadolinium anomaly, and boron) were analyzed in wells pumped for potable water supply and in two wastewater treatment plant (WWTP) effluents. In addition, a monitoring along the Lergue River (the main tributary of the Hérault River) was achieved to assess pharmaceutical behavior in surface waters. Pharmaceuticals and other wastewater-related contaminants are present in several reservoirs tapped for drinking water, confirming wastewater contamination; paracetamol, caffeine, and diclofenac are the most frequently detected. Paracetamol is present at rather high concentrations (up to 11 microg/L and 211 ng/L, respectively, in a wastewater effluent and in a drinking water sample). Though degradable in WWTP, caffeine is commonly encountered in surface waters and detected in highly polluted groundwater. On the contrary, acetylsalicylic acid concentrations are generally low despite a large consumption in France; this is related to its metabolism in humans and rapid degradation in the aquatic environment. The monitoring of pharmaceuticals along the Lergue River shows that dilution is sufficient to decrease pharmaceutical values.  相似文献

Survival of Listeria monocytogenes in ground beef   总被引:2,自引:0,他引:2  
Listeria monocytogenes, due to its association with animals and animal products and its proven pathogenicity, is an organism of potential importance to the meat industry. The survival of L. monocytogenes in ground beef held at 4 degrees C for 2 weeks was investigated. The ground beef was inoculated with L. monocytogenes type 1 or type 4 at a level of 5 x 10(5) to 7 x 10(6) CFU/g and then packaged in either oxygen-permeable or oxygen-impermeable bags. Packages were sampled at random at 0, 2, 3, 5, 7, 11, and 14 days post-inoculation, and assayed for L. monocytogenes counts and pH. The number of L. monocytogenes in ground beef remained constant throughout the sampling period, and survival was not affected by package permeability to oxygen. Listeriae were not isolated from the control ground beef. The pH of the meat increased slightly during storage, but was always in the range of pH 5.6 to 5.9. It appears that L. monocytogenes in ground beef can survive without any substantial increase or decrease in viable cell population during refrigerated storage for 14 days.  相似文献

The viability of 5 probiotic lactobacilli strains (Lactobacillus acidophilus LA-5, Lactobacillus casei L01, Lactobacillus casei LAFTI L26, Lactobacillus paracasei Lcp37, and Lactobacillus rhamnosus HN001) was assessed in 2 types of probiotic flavored drink based on fermented milk during 21 days of refrigerated storage (5°C). Also, changes in biochemical parameters (pH, titrable acidity, and redox potential) during fermentation as well as the sensory attributes of final product were determined. Among the probiotic strains, L. casei LAFTI L26 exhibited the highest retention of viability during refrigerated storage period, while L. acidophilus LA-5 showed the highest loss of viability during this period. The decline in cell count of probiotic bacteria in strawberry fermented milk was significantly greater compared to peach fermented milk. In an overall approach, peach fermented milk containing L. casei LAFTI L26 was selected as the optimal treatment in this study in both aspects of viability and sensory accpeptibility.  相似文献

Tissue culture adapted rotavirus strains were propagated in MA104 and CaCo2 cells using standard cell culture procedures. The progress of infection was monitored by examining for a cytopathic effect, and for the presence of viral RNA in the tissue culture supernatant as determined by a guanidinium-based method. Subsequently, an effective plaque assay for rotavirus was developed using MA104 cells by optimising the adsorption time (2 h) and the levels of fetal calf serum (2.5%) in the overlay medium. Tragacanth gum was used in the overlay medium to immobilize the virus, and plaques were subsequently stained with 1% crystal violet. Using this optimised plaque assay, the survival of rotavirus following exposure to heat and UV irradiation was evaluated by enumerating the clear plaques. It was shown that 60 degrees C for 10 min was sufficient to reduce the viral titer by at least 7 logs, and 50 mJ of UV irradiation was sufficient to reduce the initial viral titer by > 2.5 logs. This optimised plaque assay was also used to determine the survival and stability of rotavirus from a range of experimentally contaminated foods including fruit juice, formula milk and lettuce.  相似文献

The purpose of this study was to evaluate the performance of artificial intelligence tools for the prediction of Salmonella presence and absence in agricultural surface waters based on the population of microbiological indicators (total coliform, generic Escherichia coli, and enterococci) and physicochemical attributes of water (air and water temperature, conductivity, ORP, pH, and turbidity). Previously collected data set from six agricultural ponds monitored for two growing seasons were used for analysis. Classification algorithms including artificial neural networks (ANNs), the nearest neighborhood algorithm (kNN), and support vector machines (SVM) were trained and tested with a 539-point data set for optimum prediction accuracy. Classification accuracy performances were validated with data set (400 samples) collected from different agricultural surface water sources. All tested algorithms yielded the highest accuracy around 75 ± 1% for generic E. coli followed by enterococci (65 ± 5%) and total coliform (60 ± 10%). Classifiers calculated 6–15% higher accuracy ranging from 62 to 66% for turbidity than all other tested physicochemical attributes. Based on E. coli populations measured in other water sources, trained algorithms predicted the presence and absence of Salmonella with an accuracy between 58.15 and 59.23%. The classification performance of ANN, kNN, and SVM algorithms are encouraging for the prediction of Salmonella in agricultural surface waters.  相似文献

Rangeland grazing as a source of steroid hormones to surface waters   总被引:2,自引:0,他引:2  
Cattle and other livestock excrete endogenous steroid hormones, including estrogens, androgens, and progestins; therefore, allowing grazing livestock direct access to surface waters can result in the release of steroids in agricultural watersheds. Elevated concentrations of steroids are problematic because low concentrations of certain steroids can affect fish reproduction. To assess the occurrence and transport of steroids arising from grazing cattle, gas chromatography-tandem mass spectrometry (GC/MS/MS) was used to quantify a suite of estrogens, androgens, and progestins in small creeks impacted by rangeland grazing. Steroids were detected in 86% of samples from rangeland creeks where cattle had direct access to the water, with concentrations as high as 44 ng/L observed shortly after rain events at the beginning of the winter wet season. Estrogens were present at concentrations above the predicted no-effect concentrations for fish in 10-20% of the samples, and androstenedione was detected at concentrations higher than response thresholds for pheromonal communication in fish. The results suggest that, in certain cases, measures such as stream fencing in rangeland areas to limit direct discharge of animal wastes to surface waters or better manure management practices might be merited to protect ecosystem health.  相似文献

Azole fungicides: occurrence and fate in wastewater and surface waters   总被引:2,自引:0,他引:2  
The mode of action of azole compounds implies a potential to affect endocrine systems of different organisms and is reason for environmental concern. The occurrence and fate of nine agricultural azole fungicides, some of them also used as biocides, and four azole pharmaceuticals were studied in wastewater treatment plants (WWTPs) and lakes in Switzerland. Two pharmaceuticals (fluconazole, clotrimazole, 10-110 ng L(-1)) and two biocides (propiconazole, tebuconazole, 1-30 ng L(-1)) were consistently observed in WWTP influents. Loads determined in untreated and treated wastewater indicated thatfluconazole, propiconazole, and tebuconazole were largely unaffected by wastewater treatment, but clotrimazole was effectively eliminated (> 80%). Incubation studies with activated sludge showed no degradation for fluconazole and clotrimazole within 24 h, but strong sorption of clotrimazole to activated sludge. Slow degradation and some sorption were observed for tebuconazole and propiconazole (degradation half-lives, 2-3 d). In lakes, fluconazole, propiconazole, and tebuconazole were detected at low nanogram-per-liter levels. Concentrations of the pharmaceutical fluconazole correlated with the expected contamination by domestic wastewater, but not those of the biocides. Per capita loads of propiconazole and tebuconazole in lakes suggested additional inputs; for example, from agricultural use or urban runoff rainwater.  相似文献

The presence of psychoactive stimulatory drugs in raw waters used for drinking water production and in finished drinking water was evaluated in a Spanish drinking water treatment plant (DWTP). Contamination of the river basin which provides raw water to this DWTP was also studied. In surface waters, illicit drugs such as cocaine, benzoylecgonine (cocaine metabolite), amphetamine, methamphetamine, MDMA (ecstasy), and MDA were detected at mean concentrations ranging from 4 to 350 ng/L. Nicotine, caffeine, and their metabolites were also found at the microg/L level. The elimination of these compounds during drinking water treatment was investigated in a real waterworks. Amphetamine-type stimulants (except MDMA) were completely removed during prechlorination, flocculation, and sand filtration steps, yielding concentrations lowerthan their limits of detection (LODs). Further, ozone treatment was shown to be effective in partially eliminating caffeine (76%), while subsequent granulated activated carbon (GAC) filtration removed cocaine (100%), MDMA(88%), benzoylecgonine (72%), and cotinine (63%). Postchlorination achieved the complete elimination of cocaine and nicotine and only one parent compound (caffeine) and two metabolites (cotinine and benzoylecgonine) persisted throughout treatment although reductions of 90% for caffeine and benzoylecgonine and 74% for cotinine were obtained.  相似文献

A major question regarding the global distribution of perfluorochemicals (PFCs) is one of transport. It has been suggested that atmospheric transport of volatile precursor compounds to remote areas and subsequent degradation to the nonvolatile PFCs is responsible for contamination of biota. This paper presents surface water PFC concentrations aimed at identifying tracers of atmospheric sources. Concentrations of PFCs including perfluorocarboxylates from C6 to C10 and perfluorooctane sulfonate (PFOS) are presented here from urban surface waters with presumably both atmospheric and nonatmospheric sources of PFCs, remote waters with only atmospheric sources of PFCs, and Lake Michigan. Perfluoroheptanoic acid (PFHpA) and perfluorooctanoic acid (PFOA) were detected in all surface water samples, and PFOS was detected in all but two samples. PFOS concentrations ranged from nondetect to 1.2 ng/L and from 2.4 to 47 ng/L in remote and urban surface waters, respectively. PFOA concentrations ranged from 0.14 to 0.66 ng/L and from 0.45 to 19 ng/L in remote and urban surface waters, respectively. The ratio of PFHpA to PFOA increased with increasing distance from nonatmospheric sources suggesting that it can be used as a tracer of atmospheric deposition of PFCs to surface waters. The ratio ranged from 0.5 to 0.9 in urban areas and from 6 to 16 in remote areas. Applying this tracer to measurements from Lake Michigan indicates that the primary source of PFCs to Lake Michigan is nonatmospheric, most likely inputs from wastewater treatment effluent.  相似文献

Three competing ligand methods were compared to determine characteristics of Ag(I) complexation by dissolved and colloidal ligands present in three rivers and one sewage treatment plant effluent. Iminodiacetate groups on Chelex-100 resin (used in batch and column experiments) and diethyldithiocarbamate (DDC) were used as competing ligands. Results of batch Chelex and DDC competition experiments show good agreement with regard to relative extent of Ag binding by natural ligands among the three river systems. Results of both methods also show a possible correlation between extent of Ag(I) complexation and organic matter concentration and/or Fe concentration. Fraction of Ag(I) associated with Chelex in both batch and column Chelex experiments was similar in each of the four systems tested, indicating that lability of Ag complexes does not change significantly on time scales ranging from seconds to 24 h. Results of Chelex and DDC competition were compared using a model based on a hypothetical single natural ligand. Under the experimental conditions used, this model quantified Ag(I) complexes with log Kcond values from approximately 12 to 14. For the three rivers studied, ligands with silver-association characteristics similar to those of reduced sulfur groups (log K = 14-16) present at subnanomolar concentrations likely dominate Ag(I) speciation in these systems. A weaker ligand (e.g., log Kcond < 12) at concentrations > 0.7 nM dominated Ag(I) speciation in the treatment plant effluent. This may result from elevated concentration of metals that compete for reduced sulfur groups rather than from a lower total concentration of these groups.  相似文献

The effect of heating rate on the heat resistance, germination, and outgrowth of Clostridium perfringens spores during cooking of cured ground pork was investigated. Inoculated cured ground pork portions were heated from 20 to 75°C at a rate of 4, 8, or 12°C/h and then held at 75°C for 48 h. No significant differences (P > 0.05) in the heat resistance of C. perfringens spores were observed in cured ground pork heated at 4, 8, or 12°C/h. At heating rates of 8 and 12°C/h, no significant differences in the germination and outgrowth of spores were observed (P > 0.05). However, when pork was heated at 4°C/h, growth of C. perfringens occurred when the temperature of the product was between 44 and 56°C. In another set of experiments, the behavior of C. perfringens spores under temperature abuse conditions was studied in cured and noncured ground pork heated at 4°C/h and then cooled from 54.4 to 7.2°C within 20 h. Temperature abuse during cooling of noncured ground pork resulted in a 2.8-log CFU/g increase in C. perfringens. In cured ground pork, C. perfringens decreased by 1.1 log CFU/g during cooling from 54.4 to 36.3°C and then increased by 0.9 log CFU/g until the product reached 7.2°C. Even when the initial level of C. perfringens spores in cured ground pork was 5 log CFU/g, the final counts after abusive cooling did not exceed 3.4 log CFU/g. These results suggest that there is no risk associated with C. perfringens in cured pork products under the tested conditions.  相似文献

Using boron as a conservative tracer of municipal wastewater effluents, a mass balance was developed to determine river flowrates that requires only wastewater discharge flowrates and boron concentrations in wastewater effluents and in the river upstream and downstream of these discharges. Furthermore, this method permits calculation of the percentage of the river deriving from wastewater. This method could be useful within river sections featuring no independent data regarding river discharge. We assessed the decay of nitrate and N-nitrosodimethylamine (NDMA) precursors within an engineered treatment wetland and, using our boron analysis technique to account for dilution, within the Quinnipiac River (CT). Although both decayed with several day half-lives, their slow decay indicates they can persist to impact downstream drinking water supplies. Concentrations of NDMA and N-nitrosomorpholine (NMOR) were measurable within the river, but concentrations of four other nitrosamines, their precursors, and NMOR precursors were not detectable.  相似文献

Field runoff is an important transport mechanism by which agricultural pesticides, including atrazine, move into the hydrologic environment. Atrazine is chosen because it is widely used, is transported in runoff relatively easily, is widely observed in surface waters, and has relatively little loss in the stream network. Data on runoff of atrazine from experimental plot and field studies is combined with annual estimates of load in numerous streams and rivers, resulting in a data set with 408 observations that span 14 orders of magnitude in area. The load as a percent of use (LAPU) on an annual basis is the parameter that is compared among the studies. There is no difference in the mean or range of LAPU values for areas from the size of experimental field plots (> or = 0.000023 ha) and small watersheds (< 100,000 ha). The relatively invariant LAPU value observed across a large range of watershed areas implies that the characteristics of atrazine itself (application method and chemical properties) are important in determining the extent of runoff. The variable influences on the extent of runoff from individual watershed characteristics and weather events are superimposed on the relatively invariant LAPU value observed across the range of watershed areas. The results from this study establish the direct relevance for agricultural field plot studies to watershed studies across the full range of scale.  相似文献
