The effect of anatomical retail cut (knuckle, topside, and silverside) on physicochemical properties, fatty acid profile, and sensory characteristics of dry-cured foal “cecina” was studied. Almost all physicochemical properties were affected by anatomical retail cut, since the silverside presented significantly (p < 0.05) less moisture and protein content and more NaCl content than the topside and the knuckle cuts. Instrumental color parameters were also affected by piece type, having the knuckle pieces a more intense red, higher yellowness, and higher lightness compared to silverside and topside cuts. The results from the Warner-Bratzler test showed a marked difference (p < 0.001) among anatomical retail cuts, since the highest values of shear force were observed in silverside samples (9.5 versus 6.3 versus 2.3 kg/cm2; for silverside, knuckle, and topside cuts, respectively). Finally, sensorial analysis showed that dry-cured foal “cecina” manufactured from silverside was saltier, harder, and less juicy than those from the knuckle and topside cuts.  相似文献

An innovative edible wrapping with potential use for designing functional foods with antimicrobial capacity was developed by complexation of ε-polylysine with peptide-loaded liposomes. Unmarketable long-term frozen cooked shrimp (Litopenaeus vannamei) muscle was used as a source of both bioactive peptides and complex liposomal suspension carrier, producing a sustainable value-added protein wrapping material with desirable sensory properties. A <10-kDa peptide fraction (SH) with antioxidant and angiotensin-converting enzyme inhibitory capacity was encapsulated in partially purified phosphatidylcholine (PC) liposomes (LSH) with an entrapment efficiency of 85 %. The average size and zeta potential of LSH were 164?±?2 nm and –37.0?±?1.7 mV, respectively. The LSH surface changed from electronegative to electropositive upon adsorption of ε-polylysine (PL) with an optimal concentration of 0.5 %. The average diameter and zeta potential of the resulting complex ε-polylysine-adsorbed liposomes containing the peptide hydrolysate (PL-LSH) were 216?±?5 nm and +51.1?±?1.1 mV, respectively. The ε-PL proved to be effective as liposome stabilizing and antimicrobial agent. The PL-LSH suspension was incorporated in the formulation of the protein wrapping to provide it with both bioactive and antimicrobial properties. The wrapping showed low water solubility (≈30 %) and low mechanical resistance (tensile strength?=?0.23?±?0.06 MPa; elongation at break?=?0.91?±?0.19 %) properties that allowed it to be very versatile for varied food design and was effective against Listeria monocytogenes, Escherichia coli, Staphylococcus aureus and Yersinia enterocolitica.  相似文献

Phytosterols are used to lower the blood cholesterol. β-Sitosterol is one of the most important phytosterols with very poor water solubility and bioaccessibility. The main goal of the current research was to investigate the effects of different emulsification variables (i.e. shear, time, pressure and cycle) on the physicochemical properties, release behaviour and encapsulation efficiency of β-sitosterol in water-in-oil-in water (W/O/W) emulsion. Initially, 19 β-sitosterol emulsions were prepared under different experimental conditions (shear 3000 and 6000 rpm; time 2.5 and 5 min; pressure 100, 200 and 300 bar; cycle 1–3). In this study, the β-sitosterol emulsion prepared at 6000 rpm, 5 min, 300 bar and 3 cycles had the smallest average droplet size (139 nm) among all prepared samples. The interaction between shear and time showed the most significant (P < 0.05) effect on the polydispersity index (PDI) and turbidity of the β-sitosterol emulsion. Increasing homogenization pressure and cycle led to improving emulsion stability and encapsulation efficiency. The β-sitosterol emulsion homogenised at 100 bar for 1 cycle had the highest encapsulation efficiency (81.7%) among all samples. The optimization study showed that the emulsification at 300 bar for 3 cycles resulted in the most desirable β-sitosterol emulsion.  相似文献

Physicochemical properties of sap from Deglet Nour date palm (Phoenix dactylifera L.) were studied. Composition analysis revealed (on a dry-weight basis) a high content of carbohydrates (94.98 g/100 g of dry matter basis) mainly sucrose, 2.72 g/100g (dry matter basis) of proteins and 2.29 g/100 g (dry matter basis) of ash. Date palm sap also contains 7.64 mg gallic acid equivalent/100 ml of total polyphenol. Thus, date palm sap showed antioxidant activity with a percentage inhibition of the DPPH radical value of 47.64%. Surface and foaming properties were also performed by drop volume and bubbling method, respectively. Equilibrium surface tension of fresh sap was 63.51 mN/m. Freeze-drying method preserved surface activity. Native sap showed better foam power (1.03) and foam stability (1150 s) than solutions prepared from lyophilised sap (5–30 g /100g of solution). Results demonstrated that this natural juice could be regarded as functional food due to its high nutritional value, antioxidant activity, surface activity, and foam power.  相似文献

This study evaluates the antimicrobial effectiveness of ε-polylysine against Escherichia coli O157:H7, Salmonella typhimurium and Listeria monocytogenes in laboratory media and roast beef slurry. ε-Polylysine supplemented laboratory media and roast beef slurry were inoculated with three-strain cocktails of each pathogen and survival was periodically monitored using conventional spread plating. Inoculated laboratory media was stored at room temperature (22 °C) for 48 h, and inoculated roast beef slurry was stored at 4 °C for up to 7 days. Maximum log reductions in laboratory media/roast beef slurry were 6.01 ± 1.43/3.81 ± 0.37, > 7.82 ± 0.05/5.23 ± 0.08, and 4.58 ± 0.86/5.83 ± 0.48 for E. coli O157:H7, S. tyhimurium, and L. monocytogenes, respectively. Injured cells were produced as a result of exposure to polylysine. This study confirms the effectiveness of polylysine against pathogens in laboratory media, and demonstrates its potential as a novel antimicrobial agent in complex food matrix such as roast beef.  相似文献

The effects of dietary α-lipoic acid (α-LA) on the pH value, AMP-activated protein kinase (AMPK) activation, and the activities of glycogen phosphorylase and pyruvate kinase in postmortem muscle were investigated. Eighteen C57BL/6J mice were fed diets containing 0%, 0.5%, and 1.0% α-LA. At the end of 2-week feeding trial, the mice were killed and longissimus muscles were sampled at 0-, 1-, and 24-h postmortem for pH determination and enzyme assay. The results showed that dietary α-LA treatment significantly slowed down the decrease of pH values in postmortem muscle. The ultimate pH values in postmortem muscle of mice receiving 0.5% and 1.0% α-LA treatments were 6.40 and 6.63, respectively, significantly higher than that (6.21) of no α-LA treatment (p<0.05). AMPK was activated at the early postmortem stage. Dietary α-LA can suppress the activation of AMPK in postmortem muscle. At 1- and 24-h postmortem, activities of AMPK were much lower in postmortem muscle of mice receiving 0.5% and 1.0% α-LA treatments than that with no α-LA treatment. Between these two dietary α-LA treatments, however, no difference in AMPK activity was observed, indicating that 0.5% dietary α-LA is enough to suppress AMPK in postmortem muscle. Similar to AMPK, glycogen phosphorylase activity was higher in the treatment without dietary α-LA than those with 0.5% and 1.0% dietary α-LA supplements. No difference in the activity of glycogen phosphorylase was observed between the 0.5% and 1.0% dietary α-LA treatments. Dietary α-LA had no significant influence on the activity of pyruvate kinase in postmortem muscle. All these results indicate that AMPK plays a role in glycolysis in postmortem muscle. Dietary α-LA supplementation can suppress the activation of AMPK in postmortem muscle, down-regulate the activity of glycogen phosphorylase, resulting in a higher ultimate pH values in postmortem muscle. Therefore, dietary α-LA supplementation is a potential way to reduce the incidence of pale, soft, exudative (PSE) meat.  相似文献

Little is known about transfer of dietary β-carotene into colostrum, its absorption by the calf, and its effects on retinol and α-tocopherol in the calf when the dam's dietary vitamin A is adequate. Our objective was to assess the effect of β-carotene supplementation during the close-up dry period on the colostrum and calf. The study was conducted on a large commercial dairy farm in Indiana during early summer of 2015. Ninety-four multiparous Holstein cows were blocked by calving data, parity, and previous production, and then randomly assigned to either control or β-carotene (BC) treatments. While locked in headgates each morning, each cow received a topdress of β-carotene (Rovimix, DSM Nutritional Products, 8 g/d; provided 800 mg β-carotene) or carrier from 21 d before expected calving until calving. Colostrum was collected within 2 h of parturition. Calf blood samples were obtained within 2 h of birth before receiving the dam's colostrum, at 24 h after birth, and at 7 d and 60 d of age. Blood serum was analyzed for β-carotene, retinol, α-tocopherol, and other metabolites and enzymes. Colostrum was analyzed for β-carotene, retinol, α-tocopherol, colorimetry profile, and milk components. Data were analyzed using mixed-effects models in SAS (SAS Institute Inc.). Calf serum β-carotene data were analyzed using the FREQ procedure. Colostrum β-carotene was higher for BC cows. Colostrum from BC cows had increased a* [measures red (positive) to green (negative)] and b* [measures yellow (positive) to blue (negative)] colorimeter values, indicating that β-carotene altered colostrum color toward red and yellow. Supplementation did not affect colostral or calf IgG concentrations. Colostrum color indices were correlated with IgG concentrations as well as concentrations of β-carotene, retinol, and α-tocopherol. Before receiving colostrum, the concentration of β-carotene in calf serum was below the detectable threshold of 0.05 μg/mL. At 24 h of age, the number of calves with detectable β-carotene concentrations increased, with more calves from BC cows (52.1%) having detectable concentrations than calves from cows in the control group (6.1%). No differences in concentrations of retinol or α-tocopherol were observed in calf serum. Supplementation of β-carotene to cows decreased activities of gamma-glutamyl transpeptidase and glutamate dehydrogenase in calf serum. In pregnant cows already receiving adequate vitamin A, supplementation of β-carotene increased concentration of β-carotene in colostrum, altered colostrum color, and increased serum β-carotene in calves at birth.  相似文献

A solvent method was used to prepare the inclusion compound of neohesperidin (NH) and hydroxypropyl-β-cyclodextrin (HP-β-CD) to improve the water solubility and thermal stability of NH. The physical and chemical properties and supramolecular structure of the inclusion complex were investigated by UV–vis spectroscopy, FTIR, SEM, TG-DTG, PXRD, NMR and molecular docking techniques. The results showed that the C-ring of NH was embedded in the cavity of HP-β-CD to form an inclusion complex. The phase has changed significantly, NH is completely dispersed in HP-β-CD in an amorphous state, and the two are combined in the form of non-covalent bonds such as hydrogen bonds or van der Waals forces. Through inclusion of HP-β-CD, the solubility of NH in water at 37 °C increased from 161.81 μg ml−1 to 1927.12 μg ml−1, and its stability was also significantly improved.  相似文献

In wool dyeing and finishing processes, fabric is often treated under conditions of different pHs and is subjected to a variety of physical and chemical environments. This work investigates fabric tensile properties at three different fabric pHs. Wool fabric extensibility under a 5 N/cm load was observed to be greatest at the wool isoelectric point of pH 4.8 and lower at both pH 2.1 and pH 7.2. The impact of pH on fabric extensibility was found to be similar to the variation in fabric hygral expansion previously observed. Fabric stress–strain curves at different pHs showed that for a given fabric extension level, the work required to stretch a fabric was less at pH 2.1 than at pH 4.8. These results suggest that the strength of wool fabric is at maximum when the pH of the fibres is close to the wool isoelectric point and that for consistency, the pH of fabric should be adjusted before standard strength tests are carried out.  相似文献

A total of 168 carcasses of crossbred (Korean native black pig × Landrace) F2 pigs were divided into four carcass weight groups (A, 70–79 kg; B, 80–89 kg; C, 90–99 kg; D, 100–109 kg). The influence of carcass weight on muscle fiber characteristics and meat quality traits of pork, from the longissimus dorsi, were investigated. Carcass traits such as loin-eye area and back-fat thickness were found to increase with increasing carcass weight. Fat content, sarcomere length, drip loss, and meat color values (L*, a*, and b*) also increased with increasing carcass weight. However, moisture content and Warner–Bratzler shear force decreased with increasing carcass weight. A decrease in the compositions of type I and IIA fibers and an increase in the composition of type IIB fibers were observed with increasing carcass weight. The heavier carcasses showed type I and IIB fibers of bigger size and a smaller size of type IIA fibers. Fiber number percentage and the diameter of type IIB showed a significant positive correlation with carcass weight, fat content, and lightness (p?<?0.05). Fiber diameter of type IIB was positively correlated with loin-eye area, but was negatively correlated with moisture content. However, muscle fiber characteristics had no correlation with back-fat thickness, ultimate pH, Warner–Bratzler shear force, and b* value. In conclusion, crossbred pig which has of high composition and size of type IIB has high lightness (L* value) and water-holding capacity (drip loss). Therefore, excessively high weight of carcass has influence on pale and exudative properties in pig because of muscle fiber characteristics, especially type IIB fiber.  相似文献

The effects of high hydrostatic pressure (HHP) processing (at 200 to 600 MPa, 25 to 55°C, and from 5 to 15 min) on some structural properties of α-lactalbumin was studied in a pH range of 3.0 to 9.0. The range of HHP processes produced a variety of molten globules with differences in their surface hydrophobicity and secondary and tertiary structures. At pH values of 3 and 5, there was a decrease in the α-helix content concomitant with an increase in β-strand content as the pressure increased. No changes in molecular size due to HHP-induced aggregation were detected by sodium dodecyl sulfate-PAGE. All samples showed higher thermostability as the severity of the treatment increased, indicating the formation of a less labile structure related to the HHP treatment.  相似文献

Synergistic effects of mesenchymal stem cells (MSCs) isolated from bone marrow (BM), umbilical cord blood (UCB) and periosteum, and fibroblasts as mixed feeder cells (MFCs) on the expansion of hematopoietic progenitor cells (HPCs) were investigated in serum- and exogenous cytokine-free conditions. Enriched CD34(+) cells were cultured for 2weeks over the cell lines alone, individually, or selected combinations of them. When the cells were cultured over MFCs, the maximum increase in expansion of total nucleated cells and CD34(+)/CD38(-) cells was 157.3- and 128.6-fold, respectively. Furthermore, hematopoietic cytokine such as IL-6 and chemokines (e.g., IL-8, growth related oncogene (GRO), GRO-alpha, matrix metalloproteinase (MMP)-1, and MMP-3) were significantly increased in mixed feeder cells. Based on these results, MFCs can be more efficient for the ex vivo expansion of HPCs. These results strongly suggest that MFCs are more suitable for HPCs mass production.  相似文献

A nanoclay composite film was produced using warm water fish gelatin as a base material and its physical, mechanical, and molecular weight change properties were observed after treatment with microbial transglutaminase. The viscosity of the MTGase-treated gelatin solution (2% w/w) increased from 86.25 ± 1.77 (0 min) to 243 ± 12.37 cp (80 min). SDS–PAGE results indicated that the molecular weight of fish gelatin solutions increased after treatment with microbial transglutaminase. Tensile strength decreased from 61.60 ± 1.77 (0 min) to 56.42 ± 2.40 MPa (30 min), while E% increased from 13.94 ± 5.09 (0 min) to 15.78 ± 5.97% (30 min) at 2% (w/w) MTGase concentration. The oxygen permeability and water vapour permeability did not change as a function of treatment time at 2% (w/w) MTGase concentration. The incorporation of nanoclay inhibited the increase of oxygen permeability. Film colour values (L, a, and b) did not change, but haze values increased from 5.24 ± 0.40 (0 min) to 6.44 ± 0.94 (50 min). XRD and TEM results suggested that the nanoclay was exfoliated in fish gelatin film.  相似文献
