A heterotrophic acidothermophilic bacterial strain, YNTC-1, was isolated from an acidic hot spring in Tengchong, Yunan, China. YNTC-1 grows at pH value of 1.5-8.0 and temperature of 40-70 ℃, with optimal pH and temperature at 3.0 and 55 ℃, respectively. The cells of the strain are in shape of short rod, with 1.0-1.2 μm in length and 0.7-0.8 μm in diameter, and with distinct spores at both poles of each cell. The predominant fatty acids in cellular membrane of the strain are C18:1 ω7c. 16s rRNA gene analysis reveals that this strain is closely related to Alicyclobacillus sendaiensis, with over 99% sequence similarity. Based on phenotypic and genotypic analyses, YNTC-1 is identified as a member ofA. sendaiensis. Considering some important morphological and biochemical differences between strain YNTC-1 and A. sendaiensis ATCC 27009T, YNTC-1 may be proposed to be a novel subspecies of A. sendaiensis. However, this viewpoint has to be confirmed by further studies. Co-bioleaching of pyrite and chalcopyrite with strain YN22, Sulfobacillus thermosulfidooxidans, shows that strain YNTC-1 has no evident influence on bioleaching rates of these two sulphide minerals.  相似文献

Twelve samples derived from different locations in south central area of China are treated by enrichment and spread-plate technique for initial screening. Seven chitinase-producing strains are isolated. The chitinase present in the culture supernatant of strain CS-01 possesses the maximum activity of 0.118 U/mL. Analysis of the morphological feature and the ITS rDNA sequence reveals that strain CS-01 belongs to Aspergillus fumigatus. Production of the chitinase is regulated by a inducible way and the maximum activity appears at 36 h in colloidal chitin culture. Purification of the chitinase is carried out by salting out, gel filtrate chromatography and anion exchange chromatography sequentially. Native-PAGE and SDS-PAGE indicate that the chitinase from A. fumigatus CS-01 is a monomer with the relative molecular mass estimated to be 4.50×104. Its maximum activity appears at pH 5 and 55 °C. In order to establish the quantitative relationship between equivalent strain and the performance index of the deformed material within the range of certain passes for equal channel angular processing (ECAP), a new approach to characterize the equivalent strain was proposed. The results show that there exists better accordance between mechanical property (such as hardness or strength) and equivalent strain after rolling and ECAP in a certain range of deformation amount, and Gauss equation can be satisfied among the equivalent strain and the mechanical properties for ECAP. Through regression analysis on the data of hardness and strength after the deformation, a more generalized expression of equivalent strain for ECAP is proposed as: ɛ=k 0exp[−(k 1 Mk 2)2], where M is the strength or hardness of the material, k 1 is the modified coefficient (k 1∈(0, 1)), k 0 and k 2 are two parameters dependent on the critical strain and mechanical property that reaches saturation state for the material, respectively. In this expression the equivalent strain for ECAP is characterized novelly through the mechanical parameter relating to material property rather than the classical geometry equation. According to physiological and biochemical characteristics of Leptospirillum ferriphilum,a strain of object bacteria was isolated successfully.Bacteria were enriched by selective liquid medium and plated on designed single-layered agar solid medium.Colony was cultured and bacteria were collected.The morphologies of the object bacteria were observed using crystal violet staining,scanning electron microscope(SEM)and transmission electron microscope(TEM).The result of 16S rDNA identification shows that this bacterium belongs to Leptospirillum ferriphilum and it is named as Leptospirillum ferriphilum strain D1.These results indicate that this new single-layered agar solid medium is efficient and simple for isolation of Leptospirillum ferriphilum.Additionally,physiological-biochemical characteristics show that the optimum initial pH value and its growth temperature are 1.68 and 40 ℃,respectively.The culture of it is used to leach a complex concentrate chalcopyrite,the leaching efficiencies of copper and iron are 1.93 % and 13.74 %,respectively,and it is more effective than the A.ferrooxidans culture in the leaching of the complex concentrate chalcopyrite.  相似文献

An advanced sludge reduction process, i.e. sludge reduction and phosphorous removal process, was developed. The results show that excellent sludge reduction and biological phosphorous removal can be achieved perfectly in this system. When chemical oxygen demand ρ(COD) is 332 - 420 mg/L, concentration of ammonia p(NH3-N) is 30 - 40 mg/L and concentration of total phosphorous p(TP) is 6.0 -9.0 mg/L in influent, the system still ensures ρ(COD)〈23 mg/L, ρ(NH3-N)〈3.2 mg/L and ρ(TP)〈0. 72 mg/L in effluent. Besides, when the concentration of dissolved oxygen ρ(DO) is around 1.0 mg/L, sludge production is less than 0. 140 g with the consumption of 1 g COD, and the phosphorous removal exceeds 91 %. Also, 48.4% of total nitrogen is removed by simultaneous nitrification and denitrification.  相似文献

Strain E1 with resistance to 18 mmol/L cadmium (Cd), isolated from Cd-contaminated soil was identified by morphological observation, biochemical and physiological characterization and 16S rDNA sequence analysis. The resistance to heavy metals Cd, Cu, Co, Mn, Pb, Zn and 12 antibiotics was examined. The ability of removing Cd from solution was studied. The characterizations show that strain E1 is affiliated to Pseudomonas aeruginosa (P. aeruginosa). Strain E1 has high resistance to heavy metals and the order is found to be Cd>Mn>Zn>Cu>Pb>Co in solid media. Strain E1 also exhibits the resistance to 12 antibiotics. Both living and non-living cells of strain E1 can remove Cd from solution, and living cell has better biosorption than non-living cell. The effect of tin on synthesis of Ti3AlC2 by spark plasma sintering (SPS) from TiC/Ti/Al powders was investigated. X-ray diffraction (XRD) and scanning electron microscopy (SEM) were used for phase identification and microstructure evaluation. The experimental results show that addition of tin can considerably accelerate the synthesis reaction of Ti3AlC2 and fully dense, essentially single-phase polycrystalline Ti3AlC2 could be successfully obtained by sintering 2TiC/1Ti/1Al/0.2Sn powders at 1200–1250 °C under a pressure of 30 MPa. SEM images show that Ti3AlC2 samples in about 2–5 μm thick and 10–25 μm long platelets can be obtained. The fracture toughness and flexural strength of Ti3AlC2 were 6.5±0.2 MPa·m1/2 and 560±10 MPa, respectively. Boron-substituted carbon powder, B x C1−x with x up to 0.17, has been successfully synthesized by hot filament chemical vapor deposition. The boron concentration in prepared B x C1−x samples can be controlled by varying the relative proportions of methane and diborane. X-ray diffraction, transmission electron microscopy, and electron energy loss spectrum confirm the successful synthesis of an amorphous BC5 compound, which consists of 10–20 nm particles with disk-like morphology. Thermogravimetry measurement shows that BC5 compound starts to oxidize approximately at 620°C and has a higher oxidation resistance than carbon. The deformation behavior of a new Al-Zn-Cu-Mg-Sc-Zr alloy was investigated with compression tests in temperature range of 380–470 °C and strain rate range of 0.001–10 s−1 using Gleeble 1500 system, and the associated microstructural evolutions were studied by metallographic microscopy and transmission electron microscopy. The results show that true stress—strain curves exhibit a peak stress, followed by a dynamic flow softening at low strains (ɛ<0.05). The stress decreases with increasing deformation temperature and decreasing strain rate, which can be represented by a Zener-Hollomon exponential equation with the activation energy for deformation of 157.9 kJ/mol. The substructure in the deformed specimens consists of few fine precipitates with equaixed polygonized subgrains in the elongated grains and developed serrations at the grain boundaries. The dynamic flow softening is attributed mainly to dynamic recovery and dynamic recrystallization. A novel environmental friendly catalyst, H4SiW12O40/PAn was prepared and identified by FT-IR, XRD and TG/DTA. The optimal synthetic protocol was a PAn to H4SiW12O40 mass ratio of 1: 1, a 20 mL of methanol and a reaction time of 2 h at reflux. It was used in catalytic synthesis of ten important ketals and acetals with a high catalytic activity. With an aldehyde/ketone to glycol molar ratio of 1: 1.5 and a 1 wt% catalyst loading, the yields of ketals and acetals could reach 60.0%–93.8 % after 1 h. The viscoelastic properties of the suspension of monodisperse spherical silica produced by hydrolysis of tetraethoxysilane in alcohol solvent with ammonia as a catalyst in polyethylene glycol (PEG) were studied. The results show that the SiO2/PEG suspension possesses the reversible shear thinning and shear thickening behaviors. In the shear thinning region, the loss modulus (G″) almost remains unchanged, whereas the storage modulus (G′) decreases. In the shear thickening region, G″ and G′ increase for the formation of the "clusters". The larger G″ over G′ in all the stress studied shows that the system mainly possesses the viscous property, and that the energy dissipated(Ed) is larger than that stored. Ed of this suspension is proportional to the maximum strain (γmax) rising with the exponent of 1.92 under low shear stress; however, in the shear thickening region, Ed is proportional to γmax rising with the exponent of 5.00.  相似文献

Six strains of moderately thermophilic sulfur-oxidizing bacteria were isolated from several different typical environments in China. The identities of the isolates were confirmed by analyses of their 16S rRNA genes, and some key physiological traits. The isolates are Gram negative, rod-shaped bacteria, their optimal temperature and pH value for growth are 45-50 ℃ and 2.5-3.5 respectively. They are autotrophic and used'elemental sulfur, sodium thiosulfate and potassium tetrathionate as electron donor, while a little glucose stimulated their growth. 16S rDNA sequences analysis reveals that the strains are phylogenetically clustered to Acidithiobacillus caldus.  相似文献

The investigation results of the bonding structure of CaO−SiO2 slag by means of molecular dynamics simulation are presented. The characteristics of partial radial distribution functiong ij(r) are in good agreement with the measurement of X-ray diffraction, and the variation ofQ n with different SiO4 tetrahedra following the change ofX CaO is consistent with the results of Raman spectroscopy. The partial vibrational density of states ΓSi(ω) shows that two bands appear in the range of 636–737 cm−1 and 800–1200 cm−1 respectively which are also consistent with Raman spectroscopy. A novel scale-free network model based on clique (complete subgraph of random size) growth and preferential attachment was proposed. The simulations of this model were carried out. And the necessity of two evolving mechanisms of the model was verified. According to the mean-field theory, the degree distribution of this model was analyzed and computed. The degree distribution function of vertices of the generating network P(d) is 2m 2 m 1−3 (dm 1 + 1)−3, where m and m 1 denote the number of the new adding edges and the vertex number of the cliques respectively, d is the degree of the vertex, while one of cliques P(k) is 2m 2 k −3, where k is the degree of the clique. The simulated and analytical results show that both the degree distributions of vertices and cliques follow the scale-free power-law distribution. The scale-free property of this model disappears in the absence of any one of the evolving mechanisms. Moreover, the randomicity of this model increases with the increment of the vertex number of the cliques. ZrB2 ceramics were prepared by self-propagating high-temperature synthesis(SHS) and were sintered by hot pressing(HP).The effects of the granularities and doses of raw materials in Zr-B2O3-Mgon SHS process and product were investigated.XRD and combustion temperature curves prove that the ideal SHS reactants of Zr-B2O3-Mg are 50μm Zr powder,75μm B2O3 powder and 400μm Mg powder with 45% excessive.The particle sizes of SHS product,acid-leached product,sintered product are 2-5μm,0.5-2μm,2-10μm respectively.Chemical analysis indicates that the acid-leached product consists of ZrB2(94.59%),ZrO2(3.87%),and H3BO3(1.54%),The sintered product has a relative density of 95.4%.  相似文献

The structural and optical properties of amorphous semiconductor mercury cadmium telluride (a-MCT) are obtained by the first principles calculations. The total pair distribution functions and the density of states show that the a-MCT has the semiconductor characteristic. The calculated results of dielectric function show that E 2 peak of the imaginary of dielectric function for the crystal mercury cadmium telluride abruptly disappears in the amorphous case due to the long-range disorders. And the imaginary of dielectric function of a-MCT shows a large broad peak, which is in agreement with the available results of other amorphous semiconductors. From the linear extrapolation of the curve ħωɛ 2(ω)1/2 versus ħω, it can be obtained that the optical energy gap of amorphous semiconductor Hg0.5Cd0.5Te is 0.51±0.05 eV.
