Leaves, bark and catkins of Salix aegyptiaca L. were extracted into solvents of increasing polarity from cyclohexane (non-polar), butanol, ethanol and water (polar) and analysed for their antioxidant capacity, total phenol and flavonoids. The highest antioxidant activity (19 μg/ml IC50 for inhibition of DPPH radical activity), total phenolic content (212 mg gallic acid equivalents/g of dried extract) and total flavonoid (479 mg catechin equivalents/g of dried extract) was observed in the ethanolic extract of bark. HPLC identification of phenolic compounds from the extracts indicated the presence of gallic acid, caffeic acid, vanillin and p-coumaric acid, myricetin, catechin, epigallocatechin gallate, rutin, quercetin as well as salicin. Our data indicates the presence of high amounts of phenols and flavonoids in different parts of S. aegyptiaca species and propose that extracts from this plant may be utilised as a source of health promoting antioxidants.  相似文献

The antioxidant and antimicrobial activities as well as the quantity of phenolic substances of Impatiens balsamina L. stem extracts obtained with various solvent were determined in this study. All of the extracts possessed moderate antioxidant potential in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and reducing power assays. Antimicrobial activity was estimated using the cylinder-plate and agar dilution methods against four bacterial and six fungal strains. The extracts showed good antimicrobial activity especially antifungal activity against all of the tested microorganisms. The total phenolic and flavonoid contents ranged from 2.88 to 13.63 mg gallic acid equivalents/g dried extract and 0.98 to 7.87 mg quercetin equivalents/g dried extract, respectively. The results presented here indicate that the I. balsamina stem extracts have antioxidant and antimicrobial properties and are therefore a potential source of antioxidant and antimicrobial agents for the food and pharmaceutical industries. PRACTICAL APPLICATION: Our work indicates that the I. balsamina stem may be a good candidate as natural antioxidant and antimicrobial agents. In this study, aqueous extraction of phenolic compounds from jamun (Syzygium cumini L.) seed was undertaken. The effects of various parameters such as extraction temperature (34.8–85.2°C), extraction time (49.8–100.2 min), and liquid-to-solid ratio (9.8–60.2 mL/g) on the extraction yield, extract purity (i.e., total polyphenol content), and its antioxidant activities (1,1-diphenyl-2-picrlthydrazyl free radical scavenging assay and ferric reducing antioxidant power) were investigated. Response surface methodology was used to optimize the extraction conditions. The optimum extraction conditions (49.2°C, 89.4 min, and 51.6:1 mL/g) produced an extract with 17.3% extraction yield, high total polyphenol content (415 mg gallic acid equivalent/g dried extract) and significant antioxidant activity (IC50: 35.4 ± 0.7 µg/mL). The high-performance liquid chromatography analysis of seed extract revealed the presence of gallic acid (90.8 mg/g dried extract), ellagic acid (36 mg/g dried extract), caffeic acid (26.07 mg/g dried extract), p-coumaric acid (0.26 mg/g dried extract), catechin (9.05 mg/g dried extract), epicatechin (0.42 mg/g dried extract), and quercetin (1.54 mg/g dried extract). Tannic acid (188.5 mg/g dried extract) was also identified as a major phenolic compound. The extraction kinetics was also studied and experimental data were fitted to four kinetic models such as first-order model, second-order model, Peleg’s model, and Minchev and Minkov model, to evaluate their applicability.  相似文献

The objective of this study was to assess antioxidant and antiproliferative activities of four different Turkish pomegranate varieties (Hatay, Hicaz, Adana and Antalya) using an in vitro HepG2 cancer cell model. All the pomegranate extracts employed in this study significantly diminish the proliferation of HepG2 cells in a dose‐dependent manner. The total phenolic acid, anthocyanin and flavonoid contents for each of the four varieties were determined. The Hatay pomegranate variety had the highest total phenolic acid (337.4 ± 2.34 mg/100 g) and flavonoid (58.42 ± 2.25 mg/100 g) contents of the pomegranates examined. Antioxidant activities of the pomegranates were determined using DPPH and ABTS radical scavenging assays. The lack of correlation between colour index value and antioxidant–antiproliferative activities suggested that phenolic acids and flavonoids are predominant compounds influencing pomegranate's bioactivity rather than anthocyanins. Individual phenolic acids found in Hatay pomegranates were determined, using an HPLC system, as gallic acid being the most predominant phenolic compound.  相似文献

The response surface methodology was used to study the combined effect of temperature (60 to 80°C), time (10 to 15 min), and pH (3 to 6) on the antioxidant activity (1,1-diphenyl-2-picrylhydrazyl-radical scavenging activity, total phenolic content, and total flavonoid content) of apple honey. Statistical analysis revealed that all the responses were significantly (p < 0.05) affected by independent process variables. 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, total phenolic content, and total flavonoid content increased with the increase in time and temperature due to the formation of browning pigments. The antioxidant properties of apple honey significantly (p < 0.05) decreased with increase in pH from 3 to 6. The thermal treatment of apple honey at 80°C was found to be more effective than at 70 and 60°C. The average increase in 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, total phenolic content, and total flavonoid content of raw apple honey due to combined effect of three process variables was 9.6% ± 1.9, 13.7 ± 2.5 mg gallic acid equivalent/100 g and 49.3 ± 3.3 mg quercetin/100 g, respectively. The results showed that the most desirable optimum conditions for temperature, time and pH for 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity (92.39%), total phenolic content (133.55 mg gallic acid equivalent/100 g of honey), and total flavonoid content (25.82 mg quercetin/100 g of honey) were 80°C, 15 min and 3.01, respectively. The results demonstrated that thermal treatment significantly increased the antioxidant activity of apple honey.  相似文献

为探索酚类提取物及其复配物的协同增效作用,以桑葚和甘蔗糖蜜多酚作为研究对象,分别测定其总酚、黄酮含量及酚类组成,基于单一提取物的总酚含量按一定配比制得复配物,评价抗氧化活性。结果表明,桑葚游离多酚(MFP)占桑葚总酚含量的90.3%,为677.62 mg没食子酸当量(Gallic Acid Equivalent,GAE)/100 g,黄酮含量为736.65 mg芦丁当量(Rutinum Equivalent,RE)/100 g,共检测出原儿茶酸、3,4-二羟基苯丙酸、咖啡酸、丁香酸、对香豆酸、阿魏酸、矢车菊素3-O-葡萄糖苷和芦丁8种多酚物质,其中矢车菊素3-O-葡萄糖苷含量占92.6%。甘蔗糖蜜多酚(SMP)总酚含量为339.69 mg GAE/100 g,总黄酮含量为314.61 mg RE/100 g,主要单体包括没食子酸、原儿茶酸、绿原酸、咖啡酸、丁香酸、对香豆酸、阿魏酸、芦丁和表儿茶素9种多酚物质。在0~10 mg/mL的浓度范围内SMP、MFP及其复配物具有良好的抗氧化活性且呈现量效关系。同时,两种复配物的抗氧化活性优于SMP和MFP,其中中效原理计算复配物DPPH自由基清除能力的0.25 IC50、0.5 IC50、0.75 IC50、1 IC50、1.25 IC50、1.5 IC50的联合作用指数CI分别为0.73、0.85、0.94、0.91、0.89和0.90,均小于1,表明桑葚和甘蔗糖蜜多酚复配物具有协同抗氧化作用。  相似文献

In this study, antioxidant and antihemolytic activities of ethanolic extract of flowers, leaves, and stems of Hyssopus officinalis L. Var. angustifolius were investigated employing different in vitro assay systems. Extracts showed good antioxidant activity. IC50 for 1,1-diphenyl-2-picryl hydrazyl radical-scavenging activity were 148.8 ± 4.31 μg mL?1 for flowers, 79.9 ± 2.63 μg mL?1 for stems, and 208.2 ± 6.45 μg mL?1 for leaves. All extracts showed moderate iron (II) chelating ability. Extracts exhibited good antioxidant activity in the hemoglobin-induced linoleic acid model and also they were capable of scavenging hydrogen peroxide in a concentration dependent manner. Extracts showed good antihemolytic activity againts hydrogen peroxide-induced hemolysis (IC50 were 48.51 ± 2.27 μg mL?1 for flowers, 19.47 ± 0.73 μg mL?1 for leaves, and 63.1 ± 2.65 μg mL?1 for stems). The total amount of phenolic compounds in the extracts was determined as gallic acid equivalents and total flavonoid content was calculated as quercetin equivalents from a calibration curve.  相似文献

The present work investigated the phenolic profiles (including nonanthocyanin and anthocyanin phenolics), antioxidant activities, and neuroprotective potential of mulberry fruit (MF) (Morus atropurpurea Roxb.) grown in China at different ripening stages. High‐performance liquid chromatography‐tandem mass spectrometry method (HPLC‐MS/MS) was used to identify and quantify the phenolic compounds. The antioxidant capacity, total phenolic content (TPC), total flavonoid content (TFC), and total monomeric anthocyanin content (TAC) were determined using spectrophotometric methods. The neuroprotective effects of MFs at different ripening stages were investigated using Aβ25‐35‐treated PC12 cells as the cellular model of Alzheimer's disease. Of the 19 phenolic compounds characterized from the MF extracts, the contents of rutin and anthocyanins increased and that of chlorogenic acid decreased significantly with maturity. At the fully ripened stage, MF extracts showed the highest amounts of TPC (11.23 mg gallic acid equivalents/g fresh weight), TFC (15.1 mg rutin equivalents/g fresh weight), and TAC (1177 mg cyanidin 3‐O‐glucoside equivalents/100 g fresh weight). Meanwhile, antioxidant activity of MF extracts at this stage was highest according to ABTS (an IC50 value of 4.11 μg/mL) and DPPH (an IC50 value of 10.08 μg/mL) assays. Cellular assays revealed increased cell viability in cells treated with the ripe MF extracts; compared with the control groups, the ripening fruits also increased the antioxidant enzyme levels in PC12 cells. Together, these results suggest that the antioxidant activities and neuroprotective properties of ripening MFs are related to the contents and types of phenolic compounds that are present in the fruits.  相似文献

Anti-oxidant activities of ethanol extracts from whole and hulled mung bean starches (WMS and HMS) isolated using distilled water, and gels (WMSG and HMSG) were investigated. Total chlorophyll contents of WMS and WMSG were 4.61 and 2.14 μg/mL, respectively. WMS at 0.29 mg of gallic acid/g of MS and 0.54 mg of quercetin/g of MS had higher total phenolic compounds and flavonoid contents than HMS at 0.11 mg of gallic acid/g of MS and 0.28 mg of quercetin/g of MS. Total phenolic contents of WMSG and HMSG increased by 2.14x and 1.64x, respectively. Total flavonoid contents decreased by 0.98x and 0.57x. The 1,1-diphenyl-2-picrylhydrazyl and 2,2’-azino-bis(3-ethyl-benzothiazoline-6-sulfonic acid) diammonium salt radical scavenging activities and reducing powers of WMS and WMSG were higher than for HMS and HMSG. Cheongpomuk made using WMS exhibits increased anti-oxidant activities of gels and is a good food for improvement in health functionality.  相似文献

为阐明覆盆子不同形态多酚抗氧化和抗糖尿病活性差异,采用不同溶剂和方法提取覆盆子中的水溶、醇溶、游离和结合多酚,比较不同提取物的总酚、总黄酮和可水解单宁含量差异,测定其自由基清除能力、α-葡萄糖苷酶抑制和抗糖基化能力,并采用质谱技术测定活性最佳样品的主要化学成分。结果表明:覆盆子醇溶性多酚提取物的总酚和可水解单宁含量最高,分别为414.95、182.47 μg/mg(以每毫克提取物中所含没食子酸当量表示),同时具有最高的1,1-二苯基-2-三硝基苯肼自由基清除能力、2,2’-联氮-双(3-乙基苯并噻唑啉-6-磺酸)阳离子自由基清除能力、α-葡萄糖苷酶活性抑制能力,半抑制浓度分别为40.68、23.90 μg/mL和1.65 μg/mL。结合多酚提取物的总黄酮含量和抗糖基化能力最强。共从醇溶性多酚中初步鉴定出31 个化合物,单宁类和黄酮类化合物为其主要活性成分。因此,提取游离多酚和水溶性成分后的覆盆子残渣具有进一步利用、开发高活性抗氧化和抗糖尿病活性成分的应用价值。  相似文献

Artemisia rutifolia (Asteraceae) had been used in traditional medicines for the treatment of different ailments. In the current study, an effort was made to explore the phenolic composition, antioxidant, and antimicrobial activities of different solvent extracts obtained from A. rutifolia leaves. The reverse-phase high-performance liquid chromatographic (RP-HPLC) analysis revealed the higher extent of polyphenolic compounds (i.e., gallic acid, caffeic acid, chlorogenic acid, syringic acid, sinapic acid, p-coumaric acid, m-coumaric acid, ferulic acid, vanillic acid, myricetin, and quercetin) in methanol extract. Methanol extract consistently showed the highest total phenolic contents (98 ± 2 µg GAE/mg of plant extract), total flavonoid contents (28 ± 0.0 µg QE/mg of plant extract), antimicrobial activity, free radical (DPPH) scavenging (IC50 = 39 µg/mL) activity, and reducing power (18.3 ± 0.2 mg GAE/g of plant extract) followed by those of chloroform and hexane extracts, respectively. The current study concluded that extracts of A. rutifolia are novel natural source of antioxidative and antimicrobial agents for the treatment of oxidative stress-related disorders and microbial infections.  相似文献

Our aim was to assess the antioxidant capacities and phenolic constituents of methanol and aqueous extracts of Rhodiola imbricata Edgew. root from Trans‐Himalayan cold desert of Ladakh. The 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) and 2,2′‐azinobis‐(3‐ethylbenzothiazoline‐6‐sulfonic acid (ABTS) radical scavenging capacity of the root extracts increased in a dose‐dependent manner (up to 0.1 mg/mL) and root extract concentrations required for 50% inhibition of radical scavenging effect (IC50) were recorded as 0.013 and 0.014 mg/mL (for DPPH) and 0.016 and 0.017 mg/mL (for ABTS) for methanol and aqueous extracts, respectively. The total antioxidant power of the extract was determined by ferric reducing antioxidant power (FRAP) assay. Total polyphenol and phenolic acid content of methanol and aqueous extracts were 112.24, 59.06, 39.02, and 16.95 mg gallic acid equivalent/g of extract, respectively. Total flavonoid and flavonol contents were estimated to be 30.2, 17.67, 20.68, and 7.38 mg quercetin equivalent/g of extract, respectively. In all antioxidant capacity assays, the methanol extract exhibited significantly higher antioxidant capacity than that of aqueous extract due to the presence of significantly higher amount of vital phytoconstitiuents, viz. polyphenol, phenolic acid, and flavonol. GC/MS analysis showed that phytosterols, alkyl halide, phenols, and fatty acid esters were major phytochemical clusters. On the other hand, monoterpenes, fatty acids, tocopherols, aliphatic hydrocarbons, and ethers were found to be present in comparatively less amount in the methanol extract. Hence, our study signifies that this high‐altitude medicinal herb could be used as the natural source of antioxidants and supports its use in traditional system of medicine to ameliorate oxidative stress and high‐altitude maladies.  相似文献

BACKGROUND: Two varieties of Origanum majorana (Canadian and Tunisian) were evaluated for their phenolic, flavonoid and tannin contents, individual phenolic compounds and antioxidant activities under NaCl constraint. RESULTS: The results showed a significant variability in phenolic composition and antioxidant behavior between the two varieties under salt stress. The phenolic composition of methanolic extracts was determined by reversed‐phase high‐performance liquid chromatography. Amentoflavone was the predominant flavonoid compound; in addition, trans‐2‐hydrocinnamic acid became the major phenolic acid with salt treatment of the Tunisian variety. In the control, Canadian variety extract was characterized by high levels of gallic acid and amentoflavone. However, under 75 mmol L?1 NaCl, gallic acid content doubled, whereas amentoflavone content was maintained in the Canadian variety. Stimulation of phenolic acid biosynthesis was observed in these two varieties under salt treatment despite the fact that shoots of the Tunisian variety showed higher antioxidant activities compared to those from the Canadian variety. Tunisian O. majorana might have developed tolerance to salinity and avoided tissue damage by activating enzymes involved in the galactosylation of quercetin into quercetin‐3‐galactoside and quercetin‐3‐rhamnoside. The antioxidant capacity and the phenolic content of 3 dried red currant samples from different geographical regions were studied. Three solvent systems were used (methanol, ethanol, and acetone) at the same concentrations (70%) and with 100% deionized water in presence 0.1% HCl. The antioxidant capacity of the dried fruit extracts was evaluated using DPPH radical scavenging assays. The efficiency of the solvents used to extract phenols from the 3 dried red currant samples varied considerably. The polyphenol content of the dried red currant samples was 3.26 to 12.68 mg gallic acid equivalent/g dried fruit. The high phenolic content was significantly correlated with the high antioxidant capacity. The following compounds were identified and quantified using HPLC with photodiode-array detection: 3 anthocyanins, 2 flavonols, 2 flavan-3-ols, and 3 hydroxycinnamic acids in all samples. Generally, red currant fruits are a rich source of phenolics, which shows an evident antioxidant capacity.  相似文献

ABSTRACT: The methanolic extract of Cnidoscolus chayamansa leaves from Mexico was screened for antioxidant and antimutagenic properties by the DPPH, ABTS, iron chelating, and Kado microsuspension assays, respectively. The hypoglycemic effect was also studied. Total phenolic and flavonoid contents as well as HPLC identification and quantification of protocatechuic acid and rutin were also carried out. The C. chayamansa leaves extract contained 71.3 ± 1.7 mg gallic acid equivalent/g extract and 42.6 ± 3.7 mg (+)-catechin equivalent/g extract of total phenols and flavonoids, respectively. Concentrations of 0.242 ± 0.001 mg/g and 2.00 ± 0.097 mg/g were found for protocatechuic acid and rutine, respectively. The extract was capable of scavenging DPPH and ABTS•+ radicals in a concentration dependent manner. The extract was not toxic to TA100 and TA98 strains at the concentrations tested; moreover, the extract at a concentration of 1000 μg inhibited 24% and 39% the mutagenicity induced by 4-nitro-O-phenylenediamine and sodium azide, respectively. An acute hypoglycemic effect in diabetic rats was observed. Practical Application: C. chayamansa has been proposed as an herbal medicine to treat diabetes; however, the reported results are not conclusive and further studies need to be performed. Despite this fact, chaya leaves can be commercialized as tea in a dried presentation since the dried leaves conserve high polyphenol contents.  相似文献

Concentrated muscadine pomace extract was chromatographically analyzed for its individual phenolic, flavonoid, and anthocyanidin compounds. This extract was also characterized regarding its total phenolic content (TPC), total flavonoid content (TFC), antioxidative activities in terms of scavenging DPPH free radicals, reducing ferric, and chelating Fe2+. The TPC of this product was 34.1 ± 1.8 mg of gallic acid equivalents(GAE)/g of extract, and TFC was 3.0 ± 0.3 mg of quercetin equivalents/g of extract. Some phenolic compounds including ellagic acid, gallic acid, (−)-epicatechin, (−)-epigallocatechin, catechin, myricetin, quercetin, and kaempferol were identified. Some anthocyanidins including delphinidin, cyanidin, petunidin, peonidin, and malvidin were also identified in the extract by using a combination of retention and spectral properties on a reverse-phase HPLC–PDA. In addition, 3,3′,4,4′5,5′-hexahydroxystilbene-a resveratrol analogue present in the extract was identified for the first time by LC–MS. The results from this study demonstrate that the muscadine pomace extract is rich of natural antioxidants such as phenolics, flavonoids and anthocyanins, and possesses strong antioxidant properties. Besides, the developed methods can be used for routine quality control of the muscadine products for manufacturing efficiency and consistency.  相似文献

The anti-inflammatory activity of 4 plant extracts [guava (Psidium guajava) leaf, capillary wormwood (Artemisia capillaris Thunb.), Chinese goldthread (Coptis chinensis), and dandelion (Taraxacum platycarpum)] was investigated in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Six phenolic compounds (gallic acid, caffeic acid, chlorogenic acid, catechin, quercetin, and baicalin) were analyzed using LC-MS/MS. Guava leaf extracts showed the highest inhibitory effects on LPS-induced nitric oxide (NO, 52.58%) and prostaglandin E2 (PGE2, 43.45%) production. The total phenolic contents (TPC) in guava leaf, capillary wormwood, Chinese goldthread, and dandelion were 426.84, 154.42, 41.73, and 122.04 mg of gallic acid equivalent (GAE)/g of extract, respectively. TPC was positively correlated with the NO-inhibitory effect (r= 0.963, p<0.05) and the PGE2-inhibitory effect (r=0.971, p<0.05) at 30 μg/mL of treatment. The guava leaf extracts contained the highest levels of gallic acid and catechin, while the capillary wormwood extracts contained the highest levels of chlorogenic acid and quercetin.

A large number of compounds have been reported in peanut plants. Many of these compounds are phytoalexins, which are produced by plants experiencing environmental stress and often exhibit antioxidant activity. It is difficult to determine which of the many compounds has the greatest impact on total antioxidant capacity in a mixture. The objectives of this research were to examine the oxygen-radical absorbing capacity (ORAC) value and total phenolic contents of peanut root extracts and peanut root extract fractions collected via HPLC. Peanut roots were extracted from four different cultivars (Brantley, NC-12, Phillips, and Wilson) with 70% aqueous ethanol with ultrasonic assistance. Each cultivar was sampled in duplicate. The extracts were fractionated into 18 3-min fractions by HPLC using a C-18 column. Fractions and crude extracts were freeze dried. ORAC values and total phenolic content were then determined for all fractions and crude extracts. Fractions had a significant effect on the μM TE/mg gallic acid equivalents (GAE). ORAC values ranged from -46.89 μM TE to 185 μM TE in HPLC fractions. ORAChromatography can be used to focus on antioxidants in complex samples.  相似文献

以5种中国单花原蜜,不同产地的42个样本通过Amberlite XAD-2吸附树脂得到的多酚提取物为原料,采用分光光度法,测定总酚酸和总黄酮含量、还原能力、抗氧化活性(清除DPPH自由基、O2- ·能力),评估蜂蜜多酚类物质抗氧化活性及蜂蜜品种、颜色和地域差异对清除自由基的影响。结果表明:样本总黄酮含量为(9.41±0.18)~(92.76±0.13)mg/kg;总酚酸含量为(9.10±0.05)~(149.57±0.14)mg/100g;深色枣花蜜抗氧化能力最强,对DPPH自由基、O2- ·清除作用分别为EC50=(0.042±0.014)g/mL和EC50=(0.038±0.009)g/mL。提示蜂蜜的多酚类物质影响抗氧化活性,酚酸、黄酮含量与抗氧化能力呈现正相关系;色深原蜜的酚酸、黄酮含量高于色浅原蜜,抗氧化能力也强于浅色原蜜。  相似文献
