Coliform bacteria have long been used to indicate fecal contamination of water and thus a health hazard. In this study, the in-mill water and external effluent treatment systems of seven typical Canadian pulp and paper mills were all shown to support the growth of numerous coliforms, especially Klebsiella Spp., Escherichia coli. Enterobacter spp., and Citrobacter spp. In all mills and most sampled locations, klebsiellas were the predominant coliforms. Although all but one of the mills had no sewage input and most disinfected their feed (input) water, all contained the most typical fecal indicator bacterium, E. coli. Many of the mill coliforms were classified as fecal coliforms by standard "MPN" and metabolic tests, but this was shown to be due to their thermotolerance, not their origin. Mill coliforms were shown not to be just simple transients from feedwater or furnish (wood), but to be continuously growing, especially in some of the primary clarifiers. Isolated mill coliforms grew very well on a sterilized raw combined mill effluent. The fecal streptococci (enterococci), alternative indicators of fecal health hazards, were common in all mills in the absence of sewage. Ten strains of E. coli isolated from four mills were all shown to be non-toxigenic strains of harmless serotypes. No salmonellas were found. Therefore, the use of total coliform, fecal coliform, enterococci, or E. coli counts as indicators of fecal contamination, and thus of health hazard in pulp and paper mill effluents or biosolids (sludges) known to be free of fecal input is invalid.  相似文献

The effect of autoheated aerobic thermophilic digestion on the pathogen content of sewage sludges was studied and compared to that of conventional mesophilic anaerobic digestion. Both systems were full scale, continuously-fed facilities operated in parallel and utilized a feed sludge of thickened primary and waste-activated sludge.The relative populations of viruses, Salmonella sp., total and fecal coliforms, fecal streptococci and parasites found before and after digestion were compared. The full scale mesophilic anaerobic digesters were operated at relatively constant conditions, i.e. digester temperature constant at 35°C, and loading rates constant, etc., while the full scale autoheated aerobic digester was operated under a wide range of loading conditions. At all of the conditions studied, the autoheated digester temperature exceeded 45 C. Virus and Salmonella sp. concentrations in the effluent from the aerobic unit were below detectable limits in 10 of 11 samples and 6 of 6 samples, respectively, whereas the anaerobic digester effluent contained detectable numbers of viruses and Salmonella sp. Bacterial indicator counts and parasite concentrations were less in the autoheated digester effluent than in the effluent from the anaerobic digester. It was concluded that the simple autoheated aerobic digestion process could be used to produce a virtually pathogen-free sludge at a cost comparable to that of conventional, mesophilic anaerobic digestion.  相似文献

The specificity of indicators that depend on elevated incubation temperature as a selective factor for their enumeration was questioned because of the possibility of interference from autochthonous microorganisms adapted to high ambient temperatures. Lactose-fermenting cultures isolated from fecal coliform tests of tropical marine surface waters were identified as consisting of about 70% Escherichia coli, and most of the remaining cultures being Klebsiella, Enterobacter or Citrobacter species. This confirmed the taxonomic specificity of fecal coliform tests for these waters. Fecal and total coliforms, fecal streptococcus, heterotrophic bacteria and yeast counts had correlations of above 99% confidence levels with most other microbial and chemical parameters studied. Waters with fecal coliform counts above 1000 per 100 ml had increased incidence of presumptive pathogenic yeasts, Pseudomonas aeruginosa and Salmonella. Our data support the use of coliforms or fecal streptococci as indicators of recent fecal pollution in tropical marine waters and yeast or heterotrophic bacteria counts as complementary indicator methods for these waters.  相似文献

Persistence of indicator organisms (total coliforms, fecal coliforms, and fecal streptococci) associated with natural samples of raw sewage was studied following in situ exposure to five aquatic environments. Three of these streams contained significant amounts of acid mine water (AMW) while the other two were relatively uncontaminated. Indicator organisms were rapidly killed upon exposure to the acid mine systems, whereas little reduction in numbers was observed in the uncontaminated streams. Seasonal changes affected survival of indicators as reflected by prolonged persistence at colder in situ water temperatures. The fecal coliform group was most susceptible to the AMW stress, while the fecal streptococci were most persistent. An enrichment technique resulted in substantially enhanced recovery of certain species of sublethally injured survivors of acid stress. Enrichment was particularly beneficial for recovery of AMW-injured fecal coliforms. Relatively little improvement in recovery of fecal streptococci was afforded by the enrichment technique.  相似文献

The City of Montreal Wastewater Treatment Plant uses enhanced physicochemical processes (ferric and/or alum coagulation) for suspended solids and phosphorus removal. The objective of this study was to assess the ability of peracetic acid (PAA), UV, or ozone to inactivate the indicator organisms fecal coliforms, Enterococci, MS-2 coliphage, or Clostridium perfringens in the effluent from this plant. PAA doses to reach the target fecal coliform level of 9000 CFU/100mL exceeded 6 mg/L; similar results were obtained for enterococci, and no inactivation of Clostridium perfringens was observed. However a 1-log reduction of MS-2 occurred at PAA doses of 1.5 mg/L and higher. It was expected that this effluent would have a high ozone demand, and would require relatively high UV fluences, because of relatively high effluent COD, iron and suspended solids concentrations, and low UV transmittance. This was confirmed herein. For UV, the inactivation curve for fecal coliforms showed the typical two-stage shape, with the target of 1000 CFU/100 mL (to account for photoreactivation) occurring in the asymptote zone at fluences >20 mJ/cm(2). In contrast, inactivation curves for MS-2 and Clostridium perfringens were linear. Clostridium perfringens was the most resistant organism. For ozone, inactivation was already observed before any residuals could be measured. The transferred ozone doses to reach target fecal coliform levels ( approximately 2-log reduction) were 30-50 mg/L. MS-2 was less resistant, but Clostridium perfringens was more resistant than fecal coliforms. The different behaviour of the four indicator organisms studied, depending on the disinfectant, suggests that a single indicator organism might not be appropriate. The required dose of any of the disinfectants is unlikely to be economically viable, and upstream changes to the plant will be needed.  相似文献

Removal of fecal coliforms and fecal streptococci was monitored over a period of 13 months in 14 buried pilot scale filters, treating septic tank effluent. The effects of grain size, hydraulic dosing rate and distribution method were investigated. Two different natural sands (sorted sand and unsorted sand) and three different types of light weight aggregates (LWA 0-4 mm, LWA 2-4 mm and crushed LWA 0-3 mm) were used. Intermittent dosing rates from 20 to 80 mm/day in 12 doses per day were applied to the filters by uniform pressure distribution or point application by gravity dosing. Removal of fecal coliforms was more than three orders of magnitude higher in the media with the finest grain sizes (unsorted sand) as compared to the coarsest media (LWA 0-4 mm and LWA 2-4 mm) operated under same conditions. Fecal streptococci were determined only in effluent from filters with LWA 0-4 mm and LWA 2-4 mm. Higher removal of fecal coliforms was observed in pressure dosed filters compared to gravity dosed filters. A lower removal was observed by increasing the hydraulic dosing rate. Minimum retention time was found to be a key parameter for predicting removal of bacteria in unsaturated, aerobic filters. At minimum retention times lower than about 50 h, there was a correlation of 0.96 between retention time and removal of fecal coliforms. Retention times longer than 50 h gave almost complete removal of fecal coliforms.  相似文献

Observations made both in the field in chlorinated effluent, and in laboratory experiments show that coliforms and fecal coliforms are capable of regrowth in chlorinated wastewater. Under field conditions regrowth of coliforms in chlorinated effluent held in a storage reservoir for about 3 days appeared inversely correlated to: (1) The residual chlorine in the storage reservoir and (2) The number of coliforms surviving chlorination. In the laboratory experiments regrowth occurred after initial doses as high as 11 ppm total chlorine even when there was no chemical inactivation of the chlorine. Fecal coliforms did not generally show regrowth to the same extent as coliforms. Regrowth occurred even when coliforms were not detectible in 10-ml of samples after chlorination.Since coliforms and fecal coliforms are capable of regrowth in chlorinated sewage effluent and admixtures of it, the sanitary significance of the number of coliforms after storage or in receiving bodies of water is difficult to interpret. Thus standards might be based on the number of coliforms, or fecal coliforms detected in effluents immediately after chlorination. However, this would not be justified if in addition to coliforms, pathogenic bacteria can regrow in chlorinated effluents.  相似文献

Field and laboratory studies were initiated on the die-away kinetics of coliforms and enteric viruses in coastal waters off Tel-Aviv. Samples were taken from the sewage boil located approx. 880 m out to sea and from three main Tel-Aviv beaches designated as sampling stations, ranging 3 km to the north and 2 km to the south. Routine bacteriological tests were made from these stations; in addition, approx. 50% of the samples were also examined for enteroviral content and occasionally, samples were taken from most of the other Tel-Aviv beaches for comparison of bacterial and viral content.The concentrations of total coliforms, fecal coliforms and fecal streptococci were correlated with the concentrations of enteroviruses. In general, the data suggests that the number of enteric bacteria in the sea was reduced relatively more rapidly than that of the enteroviruses, while fecal streptococci displayed a die-away rate similar to that of enteroviruses. The percentage of samples positive for enteroviruses was found to be lower during the summer months of the year than in the remaining months. The concentration of coliforms was also significantly lower during the summer months. This suggests that the decreased microbial level is associated with increased summertime daylight and solar radiation which affect the micro-organisms' die-away rate. About 76% of the positive enteroviral samples were found at beaches with a fecal coliform bacterial level considered safe for bathing beaches.  相似文献

Limited information is available on the ability of subsurface flow wetlands to remove enteric pathogens. Two multi-species wetlands, one receiving secondary sewage effluent and the other potable (disinfected) groundwater were studied from February 1995 to August 1996, at the Pima County Constructed Ecosystems Research Facility in Tucson, Arizona. Each wetland had a retention time of approximately 4 days. The objectives of this study were (1) to evaluate the ability of multi-species subsurface wetlands to physically remove Giardia cysts; Cryptosporidium oocysts, total and fecal coliforms, and coliphages; and (2) to determine the likely impact of local wildlife on the occurrence of these indicators and pathogens. In the wetland receiving secondary sewage effluent, total coliforms were reduced by an average of 98.8% and fecal coliforms by 98.2%. Coliphage were reduced by an average of 95.2%. Both Giardia cysts and Cryptosporidium oocysts were reduced by an average of 87.8 and 64.2%, respectively. In the wetland receiving disinfected groundwater, an average of 1.3 x 10(2) total coliforms/100 mL and 22.3 fecal coliforms/100 mL were most likely contributed by both flora and fauna. No parasites or coliphages were detected.  相似文献

The fecal coliform populations of raw sewage, sewage lagoon effluent, and river water were determined using the most probable number technique. The total coliphage populations of the three water sources were determined using Escherichia coli B (ATCC 11303-1) host cells. The ratios of fecal coliforms to coliphage in the three water samples were 87:1, 4.2:1, and 0.15:1, respectively. The ratio of fecal coliforms to coliphage in stored raw sewage decreased from 87:1 to about 1:1 within 7 days at 20°C and within 28 days at 4°C. These changing ratios resulted from the greater longevity of the coliphage compared with that of the coliform bacteria. The use of the ratio of fecal coliforms to coliphage is not considered reliable as an index of when a fecal pollution event occurred because the ratio is influenced by prior contamination, presence of sediment, chlorination, and temperature.  相似文献

The application of poultry litter to agricultural fields can provide plant nutrients for crops and forage production, but fecal bacteria and the sex hormones estradiol and testosterone are components of litter that can be detrimental to the environment. Our objective was to determine if applications of poultry litter to small watersheds would contribute to the load of fecal bacteria and sex hormones to soil and runoff. We, therefore, investigated the fate and transport of fecal bacteria, estradiol and testosterone from surface applied poultry litter to four small cropped watersheds. Poultry litter was applied to meet the nitrogen requirements of pearl millet (Pennisetum glaucum L.) in 2000 and grain sorghum [Sorghum bicolor (L.) Moench] in 2001. Neither Salmonella nor Campylobacter were detected in the litter but the fecal indicator bacteria were. The average load of total coliforms, Escherichia coli, and fecal enterococci applied with the litter was 12.2, 11.9, and 12.7 log(10) cells ha(-1), respectively. The average load of estradiol and testosterone was 3.1 and 0.09 mg ha(-1), respectively. Runoff events first occurred 7 months after the first litter application in 2000, and 3 weeks after the second application in 2001. Only for the 25 July 2001 runoff event 3 weeks after the second litter application were the concentrations of total coliforms, E. coli, and fecal enterococci in runoff greater than background concentrations which were on average 5.2, 2.9, and 1.1 log(10) MPN 100 ml(-1), respectively. Average background levels of total coliforms, fecal enterococci, and E. coli in surface soil were 8.2, 7.9, and 3.5 log(10) cells kg(-1) soil. At the rate of litter application the concentrations of estradiol and testosterone in the litter did not appear to impact the background levels in the soil and runoff. Because concentrations of sex hormones in litter from other broiler operations are known to be greater than in the litter we applied, further study on the connection between concentrations of sex hormones in poultry litter and operational practices is recommended.  相似文献

Hospital effluent with its high content of multidrug resistant (MDR) enterobacteria and the presence of enteric pathogens could pose a grave problem for the community. It was planned at our tertiary care hospital in central India to study the population changes at various steps of effluent treatment plant (ETP) like collection, aeration, clarification, liquid sludge, dried sludge, high-pressure filter and treated wastewater. The study included viable bacterial counts, coliform counts, staphylococcal, enterococcal, Pseudomonas and multiple drug resistant (MDR) gram negative bacterial counts in the different stages of ETP. In order to study the distribution of bacteria as free floating in liquid and adherent to suspended particles, enumeration of the bacteria in the filterate and the sediment was also carried out. The effluent input showed 55% of the 8.6 x 10(6)/ml bacteria as coliforms and E. coli which was a typical of fecal flora. The prevalence of MDR coliforms was 0.26%. The substantial reduction (> 3log) was seen for the effluent coming from the clarifier. The bulk of the bacteria in the hospital effluent remains firmly adhered to solid particles; aeration and clarification removes bulk of the bacteria by physical processes like flocculation. The treated liquid effluent still contains sizeable loads of MDR bacteria and inactivation by procedure such as chlorination is required. The bacteria get concentrated in sludge and a greater concentration of chlorine is required for decontamination.  相似文献

The microbial quality of raw greywater was found to be much better than that of municipal wastewater, with 1.6 x 10(7)cfu ml(-1) heterotrophic plate count (HPC), and 3.8 x 10(4), 9.9 x 10(3), 3.3 x 10(3) and 4.6 x 10(0)cfu 100 ml(-1) faecal coliforms (FC), Staphylococcus aureus sp., Pseudomonas aeruginosa sp. and Clostridium perfringes sp., respectively. Further, three viral indicators monitored (somatic phage, host: Escherichia coli CN(13) and F-RNA phages, hosts: E. coli F+(amp), E. coli K12) were not present in raw greywater. The greywater was treated by an RBC followed by sedimentation. The treatment removed two orders of magnitude of all bacteria. UV disinfection kinetics, survival and regrowth of HPC, FC, P. aeruginosa sp. and S. aureus sp. were examined. At doses up to 69 mW s cm(-2) FC were found to be the most resistant bacteria, followed by HPC, P. aeruginosa sp. and S. aureus sp. (inactivation rate coefficients: 0.0687, 0.113, 0.129 and 0.201 cm2 mW(-1)s(-1), respectively). At higher doses (69-439 mW s cm(-2)) all but HPC (which exhibited a tailing curve) were completely eliminated. Microscopic examination showed that FC self-aggregate in the greywater effluent. This provides FC an advantage at low doses, since the concentration of suspended matter (that can provide shelter from UV radiation) in the effluent was very low. FC, P. aeruginosa sp. and S. aureus sp. did not exhibit regrowth up to 6h after exposure to increasing UV doses (19-439 mW s cm(-2)). HPC regrowth was proven to be statistically significant in un-disinfected effluent and after irradiation with high UV doses (147 and 439 mW s cm(-2)). At these doses regrowth resulted from growth of UV-resistant bacteria due to decreased competition with other bacteria eliminated by the irradiation.  相似文献

Microbiological research was conducted on a dual, secondary wastewater treatment system which was part of The Pennsylvania State University wastewater treatment plant. Each aeration basin received identical wastewater which was the effluent from a high rate trickling filter. One of the aeration basins was dosed with aluminum sulfate for the purpose of phosphorus removal. The other aeration basin (control) was operated in the conventional manner without alum addition. Plate counts performed on combined chemical-biological sludge and control activated sludge revealed that a higher number of viable micro-organisms was contained in the chemical-biological sludge, but the magnitude of difference between the two sludges was significant depending on the culture medium employed. Results suggest the aluminum flocs formed in the chemical-biological treatment enmesh dispersed wastewater micro-organisms, some of which are qualitatively unlike those indigenous to natural activated sludge. The combined chemical-biological sludge contained significantly higher numbers of lipolytic, gelatinolytic, and thiosulfate oxidizing micro-organisms and, possibly, fewer nitrite oxidizing micro-organisms than did control activated sludge. Alum did not appear to affect flagellated protozoa in mixed liquor; however, amoeboid and ciliated protozoa were found less frequently in alum dosed than in control mixed liquor. The settled effluent from the combined chemical-biological aeration basin generally contained fewer total coliforms, fecal coliforms, and fecal streptococci than did counterpart control effluents.  相似文献

In order to evaluate the efficacy of constructed wetlands for treatment of domestic wastewater for small communities located in rural areas, small-scale wetland mesocosms (400 L each) containing two treatment designs (a mixture of Typha, Scirpus, and Juncus species; control without vegetation) were planted into two depths (45 or 60 cm) with pea gravel. Each mesocosm received 19 L/day of primary-treated domestic sewage. Mesocosms were monitored (inflow and outflow samples) on a monthly basis over a 2-year period for pH, total suspended solids (TSS), 5-day biochemical oxygen demand (BOD(5)), total Kjeldahl nitrogen (TKN), dissolved oxygen (DO), and conductivity. Microbiological analyses included enumeration of fecal coliforms, enterococci, Salmonella, Shigella, Yersinia, and coliphage. Significant differences between influent and effluent water quality for the vegetated wetlands (p<0.05) were observed in TSS, BOD(5), and TKN. Increased DO and reduction in fecal coliform, enterococcus, Salmonella, Shigella, Yersinia, and coliphage populations also were observed in vegetated wetlands. Greatest microbial reductions were observed in the planted mesocosms compared to those lacking vegetation. Despite marked reduction of several contaminants, wetland-treated effluents did not consistently meet final discharge limits for receiving bodies of water. Removal efficiencies for bacteria and several chemical parameters were more apparent during the initial year compared to the second year of operation, suggesting concern for long-term efficiency and stability of such wetlands.  相似文献

The efficiency of peracetic acid (PAA) disinfection against enteric bacteria and viruses in municipal wastewaters was studied in pilot-scale. Disinfection pilot-plant was fed with the primary or secondary effluent of Kuopio municipal wastewater treatment plant or tertiary effluent from the pilot-scale dissolved air flotation (DAF) unit. Disinfectant doses ranged from 2 to 7 mg/l PAA in the secondary and tertiary effluents, and from 5 to 15 mg/l PAA in the primary effluents. Disinfection contact times were 4-27 min. Disinfection of secondary and tertiary effluents with 2-7 mg/l PAA and 27 min contact time achieved around 3 log reductions of total coliforms (TC) and enterococci (EC). PAA disinfection also significantly improved the hygienic quality of the primary effluents: 10-15 mg/l PAA achieved 3-4 log reductions of TC and EC, 5 mg/l PAA resulting in below 2 log reductions. F-RNA coliphages were more resistant against the PAA disinfection and around 1 log reductions of these enteric viruses were typically achieved in the disinfection treatments of the primary, secondary and tertiary effluents. Most of the microbial reductions occurred during the first 4-18 min of contact time, depending on the PAA dose and microorganism. The PAA disinfection efficiency remained relatively constant in the secondary and tertiary effluents, despite of small changes of wastewater quality (COD, SS, turbidity, 253.7 nm transmittance) or temperature. The disinfection efficiency clearly decreased in the primary effluents with substantially higher microbial, organic matter and suspended solids concentrations. The results demonstrated that PAA could be a good alternative disinfection method for elimination of enteric microbes from different wastewaters.  相似文献

This study was undertaken to determine the comparative incidence of drug-resistant coliforms and salmonellae in the water and bottom sediments of six coastal canal communities which are used heavily for recreational activities. Of a total of 423 coliforms, 300 fecal coliforms and 100 salmonella isolates examined, 74.9% coliforms, 61.3% fecal coliforms and 71.0% salmonellae were resistant to one or more of the 12 antibiotics tested. Multiple resistance occurred in 48.9% coliforms, 32.7% fecal coliforms and 68.0% salmonella strains. A total of 178 coliforms, 137 fecal coliforms and 51 salmonella isolates were tested for the presence of resistance transfer factors, and 53.9%, 53.3% and 56.9% isolates, respectively, were capable of transferring their resistance patterns to Escherichia coli and/or Salmonella choleraesuis recipient strains. A significantly higher number of drug-resistant bacteria carrying R-factors (R+) was found to occur in sediment than in surface water.  相似文献

Bacterial contamination of tile drainage water in three manured and one non-manured cropped fields was studied over a 4-year period at a large, mixed farm near Ottawa, Ontario, Canada. Concentrations of total coliforms, fecal coliforms (FC), fecal streptococci (FS) and Standard Plate Counts at 20 and 35°C, in both the drainage water and the applied dairy cattle liquid manure varied over a wide range. Bacterial concentrations in long-term winter-stored manure were much lower than in relatively fresh manure. Concentrations of FC and FS in the drainage water from the manured fields were lower than concentrations in the applied manure by 3–5 orders of magnitude. Bacterial concentrations in drainage water from all fields had low geometric mean and median values for all parameters tested. Water quality was satisfactory most of the time for recreational use or for use as a raw water source for drinking water supplies. Factors such as precipitation appeared to have a greater influence on drainage water quality than manure application. Influence of soil type on bacterial concentrations in drainage water from the manured fields was not evident. However, a lower unit-area tile outflow in coarse-textured soil compared to fine-textured soil indicated a greater potential in the former for fecal bacteria reaching groundwater beneath the tile drains.  相似文献

The objective of this study was to evaluate chemical compositions of first flush and composite storm water runoff in four small, mixed land use watersheds in north-central Texas. The watersheds range from urban to rural, all discharging to a local lake providing aquatic habitat, recreation, flood control and drinking water. Automated devices near watershed outlets collected samples from seven storm events, from April 2001 to September 2003. Samples were analyzed for suspended solids, pesticides (diazinon, triazine and chlorpyrifos), nutrients (nitrogen and phosphorus), fecal coliforms and metals. Observed concentrations of most parameters were low relative to drinking water maximum contaminant levels (MCLs) and aquatic life criteria. Fecal coliforms and lead were detected in most samples, with highest lead concentrations in the most developed watershed, likely influenced by building material and automobile traffic. Pesticide concentrations were higher in an initial spring storm event following applications to control weeds and insects. Typical of urban runoff, most ammonia and phosphorus observations exceeded freshwater criteria for streams draining to lakes. Treated wastewater effluent accounted for the highest nitrogen and phosphorus observations. For most constituents, composite concentrations exceeded first flush concentrations, though differences were significant only for fecal coliforms.  相似文献
