Ceratocystis fagacearum (Bretz) Hunt is the causative agent of oak wilt disease, which is transmitted primarily by nitidulid beetles. This fungus was compared with four non-insect-dependent fungi for their volatile profiles using gas chromatography and mass spectrometry (GC-MS) and for their attractiveness to nitidulids using a wind-tunnel bioassay. The four additional fungi includedXerula radicata Sing,Pluteus atricapillus Kumm,Tyromyces chioneus Karst, andBotrytis cinerea. Nitidulids have been reported in association with each of these fungi, but unlikeC. fagacearum, they are dispersed primarily by wind or rain. Significant attraction of three nitidulid species,Carpophilus hemipterus (Linne),C. lugubris Murray, andStelidota geminata (Say) was elicited byC. fagacearum and to a lesser extent byX. radicata, but not by the others. A comparison of headspace volatile profiles showed that the odor ofC. fagacearum was the strongest, both with regard to the number of components and in their rates of production. Chemical characterization of the headspace profile ofC. fagacearum revealed 16 components: one aldehyde, one ketone, five alcohols, and nine esters. These components were all common fruit-odor constituents and many of them were previously shown to be attractive to nitidulid beetles. The results of this study suggest that, by mimicking food odors,C. fagacearum odor is an adaptation for attracting nitidulid and possibly other insect vectors.  相似文献

The chemical mediation of host-finding was investigated for the dusky sap beetle,Carpophilus lugubris Murray. GC-MS analysis of the headspace volatiles above whole-wheat bread dough inoculated with baker's yeast, a substrate previously determined to be an effective attractant, revealed seven major components in the following order of decreasing concentration: ethanol, acetaldehyde, 2-methylpropanol, 3-methylbutanol, propanol, 2-methylbutanol, and ethyl acetate. Solutions of these seven compounds blended so as to mimic the odor of whole-wheat bread dough elicited upwind orientation from 1.8 m in a wind tunnel byC. lugubris at a level comparable to that elicited by the bread dough. A series of bioassays investigating the role of individual components from the synthetic blend determined that all seven compounds contributed to behavioral activity; however, the simplest blend evoking attraction comparable to bread dough included acetaldehyde, ethyl acetate, 2-methylpropanol, and 3-methylbutanol. Of these compounds, acetaldehyde was essential, ethyl acetate was interchangeable with ethanol or partially replaceable with propanol, and 2-methylpropanol and 3-methylbutanol were partially replaceable with a combination of the other alcohols. Headspace volatiles above aseptic or fungus-inoculated tomato, banana, sweet corn, and strawberry were also qualitatively and quantitatively analyzed. This study suggests thatC. lugubris locates its food sources by response to variable blends of common volatile constituents of plants and fungi.  相似文献

A male-produced aggregation pheromone was identified for the Australian sap beetle,Carpophilus davidsoni Dobson (Coleoptera: Nitidulidae), by bioassay-guided fractionation of volatiles collected from feeding beetles. The most abundant components were: (2E,4E,6E)-5-ethyl-3-methyl-2,4,6-nonatriene, (3E,5E,7E)-6-ethyl-4-methyl-3,5,7-decatriene, (2E,4E,6E,8E)-3,5,7-trimethyl-2,4,6,8-undecatetraene, and (2E,4E,6E,8E)-7-ethyl-3,5-dimethyl-2,4,6,8-undecatetraene. The relative abundance of these components in collections from individual males feeding on artificial diet was 100:7:9:31, respectively. Pheromone production began within several days after males were placed onto diet medium and continued for at least 20 weeks. Peak production was >3 µg total pheromone per male per day. Males in groups of 50–60 emitted less pheromone (the peak level was 0.09 µg per beetle per day), and the emissions from groups contained relatively little tetraene (proportions of the components listed above were 100:7:2:7, respectively). Three additional trienes and one additional tetraene were identified in minor amounts; the entire eight-component male-specific blend is qualitatively identical and quantitatively similar to that of the North American sibling species,C. freemani Dobson. A synthetic blend of the four major components on rubber septa, prepared to emit in the same proportions as from individual males, was highly attractive in the field when synergized with fermenting whole-wheat bread dough. Cross-attraction was observed in the field involving the pheromones ofC. davidsoni, C. hemipterus (L.), andC. mutilatus Erichson. Potential uses of the pheromones in pest management are discussed.Names are necessary to report factually on available data; however, the USDA neither guarantees nor warrants the standards of the products, and the use of the names by USDA implies no approval of the products to the exclusion of others that may also be suitable.  相似文献

Males ofCarpophilus obsoletus Erichson produce an aggregation pheromone to which both sexes respond. The pheromone was identified by GC-MS as (2E,4E,6E,8E)-3,5,7-trimethyl-2,4,6,8-undecatetraene (1), which is also a minor constituent of the pheromone blends ofC. hemipterus (L.),C. freemani Dobson, andC. lugubris Murray. The pheromone was synergized in wind-tunnel bioassays by propyl acetate, a host-type coattractant. In a dose-response study, 50 pg of1, plus propyl acetate, was significantly more attractive than just propyl acetate. Pheromone emission from groups of 65 males, feeding on artificial diet, averaged 2.2 ng/male/day. Emissions from individual males were larger, averaging 72 ng/day and ranging as high as 388 ng/day. Synthetic1 was tested in a date garden in southern California (500 µg/rubber septum), using fermenting whole-wheat bread dough as the coattractant. The pheromone plus dough attracted significantly more beetles than dough alone (means were 4.2 and 0.0 beetles per week per trap). Captured beetles were 54% females. Field trap catches were highest during the months of July and August.All programs and services of the U.S. Department of Agriculture are offered on a nondiscriminatory basis without regard to race, color, national origin, religion, sex, age, marital status, or handicap. Males ofCarpophilus mutilatus Erichson produce an aggregation pheromone to which both sexes respond. The pheromone includes two hydrocarbon components, (3E,5E,7E)-5-ethyl-7-methyl-3,5,7-undecatriene (1) and (3E,5E,7E)-6-ethyl-4-methyl-3,5,7-decatriene (2). These were emitted in a 101 ratio and in a total amount of ca. 5 ng per feeding male per day. All tested doses of1 and2, from 0.03 to 30 ng, were more attractive than controls in wind-tunnel tests, but there was no evidence of synergism between these trienes. Dramatic synergism between the pheromone and a food-type coattractant occurred in the field, however. In a date garden in southern California, traps with a combination of synthetic1 and fermenting whole-wheat bread dough attracted 22 times more beetles than dough by itself and 295 times more than1 by itself. Volatile collections from males also contained three oxygenated compounds that were absent from females. One of these was tetradecanal (ca. 5 ng per male per day), but the structures of the other two are presently undetermined (0.8 and 1.1 ng per male per day). No function for these was demonstrated. One compound originating in the artificial diet, 2-phenylethanol, was particularly attractive in the wind-tunnel bioassay, as was the chromatographic solvent, methanol.  相似文献

A male-produced aggregation pheromone was demonstrated in Colopterus truncatus Randall (Coleoptera: Nitidulidae) by gas chromatographic comparisons of male and female volatile emissions. Male-specific compounds were identified with coupled gas chromatographic–mass spectrometric (GC-MS) analysis and GC and MS comparison of authentic standards. Physiological activity was evaluated by coupled gas chromatographic–electroantennographic (GC-EAG) recordings, and electroantennographic (EAG) assays of standards. The male-produced volatiles eliciting responses from male and female antennae (and relative abundance) were (2E,4E,6E)-3,5-dimethyl2,4,6-octatriene (1) (1.8), (2E,4E,6E)-4,6-dimethyl-2,4,6-nonatriene (2) (100), and (2E,4E,6E,8E)-3,5,7-trimethyl-2,4,6,8-decatetraene (3) (3.3). A fourth male-specific compound, (2E,4E,6E,8E)-4,6,8-trimethyl-2,4,6,8-undecatetraene (4) (0.6) was not EAG-active. EAG dose–response studies showed that the antennae were most sensitive to 2 followed by 3 and 1. Synthetic 2, binary blends of 1 and 3, and tertiary blends of 1, 2, and 3 were highly attractive in the field when synergized with fermenting whole-wheat bread dough. In the field, cross-attraction to the C. truncatus pheromone components was observed for Carpophilus lugubris Murray, C. antiquus Melsheimer, and C. brachypterus Say.  相似文献

New sex pheromone component of femaleStegobium paniceum L. was isolated and identified as 2,3-dihydro-2,3,5-trimethyI-6-(1-methyl-2-hydroxybutyl)-4H-pyran-4-one, stegobiol, by spectral data and chemical conversion from stegobinone. Relative configuration at C-2, C-3, and C-1 was determined as 2S,3R,1S by the conversion from (2S,3R,1R)-stegobinone. A slow release formulation of the bark beetle pheromones,endo- andexo-brevicomin, significantly reduced landing of southern pine beetles on host trees. Beetle flight activity within the infestation was not affected by the pheromones. The pheromones were released at approximately 600 mg/ha/day via Conrel® hollow fiber controlled-release dispensers. The major component of the male-produced aggregation pheromone ofCarpophilus dimidiatus (F.) is (3E, 5E, 7E, 9E)-6,8-diethyl-4-methyl-3,5,7,9-dodecatetraene. It attracts beetles of both sexes in the field and is synergized by odors from fermenting bread dough; mean trap catches for the tetraene alone, tetraene plus dough, dough alone, and control were 24.5, 48.3, 0.02, and 0.00, respectively. In the laboratory, individual males produced 0.58 µg±0.35 µg (SD) of the tetraene per day, but males in groups of 10–50 produced <2% as much per beetle. A second male-specific compound, (3E, 5E, 7E, 9E)-5,7-diethyl-9-methyl-3,5,7,9-tridecatetraene, was also identified fromC. dimidiatus and is about 5% as abundant as the major pheromone component.Carpophilus flight activity was monitored for one year in South Carolina corn fields with the pheromones forC. dimidiatus, C. freemani Dobson,C. mutilatus Erichson,C. hemipterus (L.),C. lugubris Murray, andC. obsoletus Erichson, all in combination with bread dough. The first four of these species accounted for 18, 70, 5.7, and 0.03%, respectively, of the totalCarpophilus trapped, but noC. lugubris orC. obsoletus were captured. Captures ofC. freemani were as high as 11,400/trap/week. Species specificity for the first four pheromones was high, except that a synthetic impurity in theC. dimidiatus pheromone was somewhat attractive toC. freemani andC. mutilatus. Three other species captured.C. antiques Melsheimer,C. marginellus Motschulsky, andC. humeralis (F.), accounted for 0.005, 5.0, and 1.3% of the total catch, respectively.C. antiquus was attracted primarily to the pheromone ofC. dimidiatus, butC. marginellus andC. humeralis responded to most of the test pheromones. The chemical basis underlying orientation to fruit and fungal odors was investigated for the dried-fruit beetle,Carpophilus hemipterus (L.). In wind-tunnel bioassays of walking and flight response from 1.8 m, beetles were attracted to odors of the yeastSaccharomyces cerevisiae on agar, aseptic banana, or banana inoculated withS. cerevisiae, although both banana substrates elicited greater response than the yeast alone. When presented in a two-choice bioassay, the yeast-inoculated banana attracted approximately twice as many beetles as did the aseptic banana. GC-MS analysis of the headspace volatiles above these odor sources revealed a somewhat more complex and concentrated volatile profile for yeast-inoculated banana than for aseptic banana. The odor from yeast on agar had fewer components, and these were present at lower concentrations than the odors of either banana substrate. By blending mineral-oil or aqueous solutions of the 18 components of inoculated-banana odor in varying concentrations, it was possible to mimic closely the headspace profile of the natural odor. This synthetic odor also elicited beetle attraction in the wind tunnel at levels comparable to the inoculated banana. Through a series of bioassays in which individual components were subtracted from or added to a synthetic odor blend, it was determined that ethyl acetate, acetaldehyde, 2-pentanol, and 3-methylbutanol comprised the simplest blend of compounds evoking full behavioral response. However, 2-methylpropanol or butanol were apparently interchangeable with 3-methylbutanol in this blend, and comparable response could also be elicited by replacing acetaldehyde with a combination of both 2-pentanone and 3-hydroxy-2-butanone. Thus, our results suggest that this generalist insect herbivore locates its hosts by a long-range response to a variety of blends of common fruit volatiles, whose concentrations are enhanced by fungi.  相似文献

A chemical study of the sex pheromone of the cigarette beetle was carried out. Seven components were isolated from active fractions of column chromatography of the female extract, and their structures were elucidated by spectroscopic evidence and confirmed by synthesis to be (4S,6S,7S)-4,6-di-methyl-7-hydroxynonan-3-one (serricornin) (I), 2,6-diethyl-3,5-dimethyl-3,4-dihydro-2H-pyran (anhydroserricornin) (II), 4,6-dimethylnonan-3,7-dione (III), 4,6-dimethylnonan-3,7-diol (IV), 4,6-dimethyl-7-hydroxy-4-nonen-3-one (V), (2S,3R)-2,3-dihydro-3,5-dimethyl-2-ethyl-6-(l-methyl-2-oxobutyl)-4H-pyran-4-one (serricorone) (VI) and (2S,3R)-2,3-dihydro-3,5-dimethyl-2-ethyl-6-(1-methyl-2-hydroxybutyl)-4H-pyran-4-one (serricorole) (VII).These structural features suggested that the occurrence of these components might be related to the polyketide biosynthesis. The behavioral bioassay and BAG experiments revealed the biological role of each component in the copulatory behavior of this insect.  相似文献

Males ofCarpophilus hemipterus (L.), the dried-fruit beetle, (Coleoptera: Nitidulidae) were found to emit nine all-E tetraene and one all-E triene hydrocarbons in addition to two pheromonally active tetraenes that had been reported previously. The previously known compounds are (2E,4E,6E,8E)-3,5,7-trimethyl-2,4,6,8-decatetraene(1) and (2E,4E,6E,8E)-3, 5,7-trimethyl-2,4,6,8-undecatetraene(2). The new tetraenes were all related to structure1 by having one additional carbon at either one or two of the following four locations: at carbon 1 of the chain, at carbon 10 of the chain, at the 5-alkyl branch, or at the 7-alkyl branch. (Structure 2 also fits within this pattern.) The triene inC. hemipterus is (2E,4E,6E)-5-ethyl-3-methyl-2, 4,6-nonatriene. Also identified from volatile collections from the beetles were the 2Z and 4Z isomers of1. All structures were proven by synthesis, with NMR and mass spectral data for the compounds provided. Two of the newly discovered compounds, (2E,4E,6E,8E)-7-ethyl-3,5-dimethyl-2,4,6,8-decatetraene and (2E,4E,6E,8E)-7-ethyl-3,5-dimethyl-2,4,6,8-undecatetraene, were quite active in the wind-tunnel bioassay, but others, such as (2E,4E,6E,8E)-5-ethyl-3,7-dimethyl-2,4,6,8-decatetraene and (2E,4E,6E,8E)-4,6,8-trimethyl-2,4,6,8-undecatetraene were not. Structureactivity relationships are explored among the natural compounds and additional, synthetic analogs, which were never detected from the beetles. Some of these analogs, such as (2E,4E,6E,8E)-3,5-dimethyl-7-propyl-2,4,6,8-undecatetraene, were quite active in the bioassay. The biosynthesis of the beetle-derived compounds is discussed. A single biosynthetic scheme that lacks complete enzyme specificity at four specific steps could account for the entire series of compounds found in the beetles and their relative proportions. The definition of pheromone is discussed in relation to these hydrocarbons.  相似文献

Antennal and behavioral responses of the rust-red grain beetle,Cryptolestes ferrugineus, and the flat grain beetle,C. pusillus, to synthetic samples of the macrocyclic lactones reported to comprise their aggregation pheromones were investigated. Electroantennogram (EAG) recordings were obtained successfully from both species for the first time. Females of both species showed larger EAGs than males. The EAGs ofC. ferrugineus showed a high degree of specificity for conspecific aggregation pheromone components;C. pusillus showed much less specificity. Behavioral tests were conducted using two-choice pitfall bioassays. Separation of the results into the two effects of activity stimulation and direction finding showed that both effects contributed to the overall response, although sometimes to different extents. The strain ofC. pusillus studied responded equally well to both components of its pheromone, whereas it had been reported previously that only one was active, the other acting as a Synergist and eliciting no response when tested alone. With both species, behavioral response was elicited with a single lactone, suggesting that it might not be necessary to use both components for field use. Particularly surprising was thatC. pusillus showed a greater response to the pheromone components ofC. ferrugineus than to its own. Aeration of the two species and thermal desorption of the collected volatiles confirmed production of the expected lactones, and aeration of authentic lactones showed that the response was not due to the C.ferrugineus pheromone components being markedly more volatile. This response, which seems to be an actual preference, is the first to be discovered among the cucujid beetles and encourages optimism that a practical lure for various species may not need to be as complex as originally feared.  相似文献

Antennal olfactory (electroantennogram) and laboratory and field behavioral tests were carried out on the response ofDendroctonus frontalis to its aggregation pheromone frontalin and analogs. The analogs were compounds modified by altering the position and methyl groups and/or by their deletion. Any modification to the frontalin structure significantly reduced both the antennal olfactory and behavioral response byD. frontalis. Beetle response, although significantly reduced, was elicited at the receptor level and in a laboratory bioassay by all analogs. Male coconut rhinoceros beetles,Oryctes rhinoceros (L.), produce three sex-specific compounds, ethyl 4-methyloctanoate, ethyl 4-methylheptanoate, and 4-methyloctanoic acid, the first of which is an aggregation pheromone. Synthesis of these compounds involving conjugate addition of organocuprates to ethyl acrylate is reported. In field trapping experiments, (4S)-ethyl 4-methyloctanoate and the racemic mixture were equally attractive and 10 times more effective in attracting beetles than ethyl chrysanthemumate, a previously recommended attractant. Ethyl 4-methylheptanoate was as attractive as ethyl chrysanthemumate and more attractive than 4-methyloctanoic acid, but further studies are required before it can be classed as an aggregation pheromone. Compared to ethyl 4-methyloctanoate alone, combinations of the three male-produced compounds did not increase attraction, whereas addition of freshly rotting oil palm fruit bunches to pheromone-baited traps significantly enhanced attraction. With increasing dose, captures ofO. rhinoceros increased, but doses of 6, 9, and 18 mg/day were competitive with 30 mg/day lures. Newly designed vane traps were more effective in capturing beetles than were barrier or pitfall traps. Results of this study indicate that there is potential for using ethyl 4-methyloctanoate in operational programs to controlO. rhinoceros in oil palm plantations.  相似文献

The attractiveness of representative host materials, host extracts, and individual host volatiles (primarily carboxylic acids, alcohols, and esters) toCarpophilus hemipterus (L.) (Coleoptera: Nitidulidae) adults in wind-tunnel bioassays was examined. Attractiveness of the materials was examined alone and in combination with the aggregation pheromone. Host materials and extracts were often attractive on their own, and the attractancy was synergized when they were combined with the pheromone. Propanoic and butanoic acids, methanol, 2-propanol, 1-heptanol, methyl butanoate, and propanal were among the most effective attractants relative to the pheromone, but many other compounds significantly synergized the pheromone (typically three- to four fold). Attractiveness and synergism were influenced by the carbon chain length and branching of the substitutents. Straight-chain compounds that had at least three carbon atoms were generally effective as synergists. Many branched-chain compounds were also effective synergists. In general, the degree of attractiveness and synergism could be predicted fairly well with the physicochemical steric (Es) parameter, although the lipophilicity (Pi) parameter also appeared to be useful in explaining the lower activity of short-chain substituents. Thus, many compounds that had only limited attractiveness on their own may nevertheless play and important role in synergizing the pheromone. Structure-activity studies appear to be appropriate not only for determining optimal attractants for these insects, but also for determining effective synergists for the pheromone.The mention of firms or trade names does not imply that they are endorsed or recommended by the U.S. Department of Agriculture over other firms or similar products not mentioned.  相似文献

Electroantennograms (EAGs) were recorded from male and female black turpentine beetles,Dendroctonus terebrans (Olivier), exposed to bark beetle pheromones and host terpenes. The dose-response curves indicated similarities in the receptor mechanisms for both sexes for each compound. Antennal sensitivity was greatest toendo-brevicomin, which correlates with the importance of the compound in the behavior of the beetles. At above-threshold concentrations, EAGs were greatest toendo-brevicomin and frontalin, suggesting a large population of antennal receptors for these compounds. A large population of receptors would be expected for compounds that play such a significant role in this beetle's behavior. Evidence for the existence of a male-produced aggregation pheromone secreted from the prothoracic femoral setiferous sex patch ofTribolium castaneum is reported. Both sexes were attracted toca. 60 ng of crude secretion. Males and females perceive the pheromone on the day of emergence while perception differs between the sexes: male response reaches a maximum on day 1 posteclosion, when tested at <1, 1, and 30 days; females show a maximum response at 30 days posteclosion. Porapak Q-captured volatiles of malePolygraphus rufipennis (Kirby) boring in black spruce and white spruce from Newfoundland and British Columbia, respectively, contained 3-methyl-3-buten-1-ol. Volatiles from logs in which the males had been joined by females contained the compound in reduced quantities. Hindgut extracts from male and femaleP. rufipennis disclosed no male-specific volatiles, but 3-methyl-3-buten-1-ol was detected in extracts of male-produced frass. The compound was not present in extracts from fresh phloem tissue.P. rufipennis of both sexes responded strongly in the field to traps baited with 3-methyl-3-buten-1-ol released at 4390 g/day. There was little response to unbaited traps, fresh uninfested black spruce logs, or to 3-methyl-3-buten-1-ol released at lower rates. Combination of 3-methyl-3-buten-1-ol with either one of five terpenes prevalent in black spruce did not enhance beetle catch. Approximately half of 20 black spruce trees baited with 3-methyl-3-buten-1-ol were attacked, compared to 20.0% of 10 unbaited control trees. Loblolly and shortleaf pine growing on a single site in the North Carolina piedmont were examined to determine similarities and differences in their defensive mechanisms against the southern pine beetle,Dendroctonus frontalis Zimmermann, and its fungal associate,Ceratocystis minor (Hedgecock) Hunt. Both species responded to wounding and fungal inoculation by forming a hypersensitive lesion around the wound site. There were significantly less soluble sugars and more monoterpenes in the lesion tissue than in unwounded inner bark. The two species were similar in resin flow rate and inner bark soluble sugar content, but the loblolly pines had thicker bark, longer hypersensitive lesions, and a higher concentration of inner bark monoterpenes. Inner bark monoterpene composition was also significantly different between the two pine species. It is hypothesized that two different defensive strategies against southern pine beetle attack may be utilized.  相似文献
