Solid–liquid extraction and response surface methodology were used to optimise conditions for the antioxidant activity of grape cane extracts. The independent processing variables were ethanol concentration, temperature and solvent to solid ratio. Ethanol concentration and temperature significantly affected antioxidant activity measured by the Trolox equivalent antioxidant capacity (TEAC) assay and the oxygen radical absorbance capacity using fluorescein (ORACFL) method (p  0.01), whereas the solvent to solid ratio did not significantly affect the activity (p > 0.05). Antioxidant activity of the extracts, determined by the TEAC assay, varied from 85.6 to 238.6 μmol Trolox equivalents/g of dry sample. ORACFL values ranged from 308.4 to 1302.7 μmol Trolox equivalents/g of dry sample. Ethanol concentrations of 40.4% and 55.4% were optimal for the highest antioxidant activities measured by the TEAC assay and the ORACFL method, respectively. The optimal temperature was 83.6 °C. Antioxidant activity correlates with total phenolic content of extracts.  相似文献

Total phenolic content (TPC) and total antioxidant capacity (TAC) of four onion varieties (red, white, yellow and sweet) and shallot from selected locations (Washington, Idaho, Oregon, Texas and Georgia) were determined using Fourier transform infrared (FT-IR) spectroscopy (4000–400 cm−1). The Folin–Ciocalteu (F–C) assay was used to quantify TPC and three assays were used to determine TAC, including 2,2-diphenyl-picrylhydrazyl (DPPH) assay, Trolox equivalent antioxidant capacity (TEAC) assay and ferric reducing antioxidant power (FRAP) assay. Partial least squares regression (PLSR) with cross-validation (leave-one-out) was conducted on onion and shallot extracts (n = 200) and their corresponding F–C, DPPH, TEAC and FRAP values were employed to obtain four independent calibration models for predicting TPC and TAC for the extracts. Spectra from an extra 19 independent extracts were used as an external validation set for prediction. A correlation of r > 0.95 was obtained between FT-IR predicted and reference values (by F–C, DPPH, TEAC and FRAP assay) with standard errors of calibration (SEC) and standard errors of cross-validation (SECV) less than 2.85, 0.35 and 0.45 μmol Trolox/g FW of extracts for TEAC, FRAP and DPPH assay, respectively; and 0.36 mg gallic acid/g FW of extracts for the F–C assay. In addition, cluster analysis (principal component analysis (PCA)) and discriminant function analysis (DFA) could differentiate varieties of onions and shallot based upon infrared spectral features. Loading plots for the various chemometrics models indicated that hydroxyl and phenolic functional groups were most closely correlated with antioxidant capacity. The use of mid-infrared spectroscopy to predict the total antioxidant capacity of vegetables provides a rapid and precise alternative to traditional wet chemistry analysis.  相似文献

Rosehip extracts of 18 samples representing six taxa in the genus Rosa were evaluated for antioxidant activities by use of different test systems. The ferric‐reducing antioxidant power (FRAP) and Trolox‐equivalent antioxidant capacity (TEAC) of the crude extracts ranged from 983.4 to 2187.1 µmol FRAP g−1 dry matter and from 457.2 to 626.2 µmol TEAC g−1 dry matter. The high antioxidant capacity was related to high contents of phytonutrients. The overall mean of antioxidants was 23.23 mg g−1 total carotenoids and 76.26 mg g−1 total phenolics. The phenolic component made a major contribution to the total antioxidant activities in both assays (overall mean was 90.5% and 75.7%), whereas the ascorbate made a minor contribution (8.6% and 16.9%) and the lipophilic component made an even smaller one (0.9% and 7.3%). However, the lipophilic component was the most effective when the comparison was based on the ratio of antioxidant activity to content of antioxidants. The crude extracts exhibited 50.9% (46.6–60.3%) inhibitory effect against the lipid peroxidation induced by 2,2′‐azobis(2,4‐dimethylvaleronitrile) (AMVN) and 85.0% (80.1–90.2%) inhibition in 2,2′‐azobis(2‐amidinopropane)hydrochloride (AAPH) assay at a concentration of 250 µg ml −1. Ascorbate acted as an antioxidant in both peroxy radical‐induced lipid peroxidations, but as a pro‐oxidant in the metal ion‐induced lipid peroxidation. The crude extracts showed a large inhibitory effect in the ferric ion‐induced lipid peroxidation and caused 83.7% inhibition at a concentration of 25 µg ml −1 dried rosehip powder. © 2000 Society of Chemical Industry  相似文献

Antioxidant activity of ethyl acetate extracts from rice koji (EAERK) fermented with Aspergillus candidus CCRC 31543 was evaluated, and the antioxidant components in koji were isolated and identified. EAERK (200 μg/ml) showed a 90% inhibitory effect on peroxidation of linoleic as determined by the thiocyanate method. EAERK also showed a marked antioxidant activity on the oxidation of lard determined by using the Rancimat method. EAERK had a strong scavenging effect on the DPPH radical. Silica gel column chromatography was used to separate EAERK into eight fractions (A–H). Fractions C–F possessed significant antioxidant activity and showed more than 88% inhibition of peroxidation of linoleic acid. Fraction C was crystallized and purified to obtain compound 1. Fractions D–F were combined and re-separated by means of column chromatography and the subfraction that exhibited strong antioxidant activity was crystallized to yield purified compound 2. Compounds 1 and 2 were shown to be ergosterol and terphenyllin, respectively, on the basis of UV-vis spectral, MS and NMR analyses. Ergosterol showed no antioxidant activity on the peroxidation of linoleic acid. However, the antioxidant activity of terphenyllin was equal to that of BHA at 200 μg/ml.  相似文献

This study investigated the antioxidant and anti-inflammatory properties of the fractionated tea extract, a lower-polymerized polyphenol-rich extract ultrafiltrated from oolong tea. The amounts of total polyphenols, flavonoids and condensed tannin were enriched in fractionated tea extract compared to oolong tea. Furthermore, fractionated tea extract was a stronger scavenger of nitric oxide and 2,2-diphenyl-1-picryhydrazyl radicals than oolong tea. When lipopolysaccharide-treated RAW264.7 macrophages were co-incubated with both tea extracts (500 μg/mL) or (-)-epigallocatechin gallate (100 μg/mL) for 16 h, all samples (oolong tea, fractionated tea extract, and (-)-epigallocatechin gallate) suppressed nitric oxide production by 60, 48, and 46%, respectively, and the suppression was due to nitric oxide-scavenging. However, only fractionated tea extract lowered the proportion of phospholipid arachidonic acid and type II-cyclooxygenase expression, thereby decreasing PGE2 synthesis by 29%. In conclusion, fractionated tea extract was rich in potent antioxidant substances capable of inhibiting the production of pro-inflammatory mediators.  相似文献

A total of 28 different plants from different regions of Çanakkale, Turkey, were investigated for their antioxidant capacity and total phenol contents to find new potential sources of natural antioxidants. The antioxidant capacity of methanolic extracts prepared from various parts of plants was evaluated by both trolox equivalent antioxidant capacity (TEAC) and 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) assays, while the total phenolics were determined using the Folin‐Ciocalteu method. The TEAC values of plant extracts ranged in a large scale from 1472.36 to 17.61 μmol of trolox equivalents per g dry weight (dw), and EC50 values (concentration at which 50% radical scavenging occurred) varied from 0.174 to 42.475 mg dw of plant, while the total phenol content of plant extracts ranged between 117.20 and 1.27 mg of gallic acid equivalents per g dw. There was a positive linear correlation between the TEAC and total phenols of plant materials (r = 0.916). The extracts of Hypericum perforatum, Arbutus andrachne and Paliurus spina‐christii showed higher antioxidant activities (both TEAC and DPPH assays). However, there was no clear relationship between TEAC and EC50 values (r = 0.477) of plant extracts.  相似文献

Fermented marine blue mussel (Mytilus edulis) derived peptides were purified using ion exchange, gel filtration and high performance liquid chromatographic techniques to identify a potent radical scavenging activity. The hepta-peptide sequence, HFGBPFH (MW 962 kDa) was found to be highly effective for radical scavenging and was named as mussel derived radical scavenging peptide (MRSP). Under the assay conditions, MRSP could scavenge superoxide (98%), hydroxyl (96%), carbon-centered (91%) and DPPH radicals (72%) at 200 μg/ml concentration and the respective IC50 values were found to be 21, 34, 52 and 96 μM. In addition, MRSP exhibited a strong lipid peroxidation inhibition at 54 μM concentration and it was higher than α-tocopherol. Further, it exhibited a 95% higher Fe2+ chelating effect than citrate. Moreover, MRSP could enhance the viability of oxidation induced cultured human cells by 76% following 75 μg/ml of treatment. Therefore, these results identified MRSP as a potent natural antioxidant, which performs its activity via different mechanism of actions.  相似文献

测定茴香体外抗氧化活性,并分析茴香中总多酚与总黄酮含量与抗氧化活性的关系。结果表明,茴香乙酸乙酯部位和正丁醇部位具有体外抗氧化活性。其中,乙酸乙酯部位清除DPPH自由基和ABTS+自由基的能力(IC50分别为(37.67±0.21)、(24.88±0.07)μg/mL)比正丁醇部位的清除能力(IC50分别为(47.48±0.18)、(34.19±0.19)μg/mL)强,且具有显著性差异(p<0.05),而正丁醇部位对铁离子的还原能力(TEAC值为(569.9±0.69)μmol/g)比乙酸乙酯部位的还原能力(TEAC值为(281.23±4.73)μmol/g)强,且具有显著性差异(p<0.05)。乙酸乙酯部位清除DPPH和ABTS自由基的能力强可能与其总黄酮含量高有关,正丁醇部位还原铁离子的能力好与其总多酚含量高有关。   相似文献

The antioxidant activity of water and ethanol extracts of clove (Eugenia carophyllata) buds and lavender (Lavandula stoechas L.) was studied. The antioxidant properties of both extracts of clove and lavender were evaluated using different antioxidant tests; reductive potential, free radical scavenging, superoxide anion radical scavenging and metal chelating activities. The both extracts of clove and lavender exhibited strong total antioxidant activity. At the concentrations of 20, 40, and 60 μg/ml, water extract of clove and lavender showed 93.3%, 95.5%, 97.9%, 86.9%, 92.3%, and 94.8% inhibition on lipid peroxidation of linoleic acid emulsion, respectively. At the same concentrations, ethanol extract of clove and lavender exhibited 94.9%, 95.5%, 98.2%, 92.5%, 93.8%, and 96.5%, respectively. Comparable, 60 μg/ml of standard antioxidant such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), and α-tocopherol exhibited 96.5%, 99.2%, and 61.1% inhibition on peroxidation of linoleic acid emulsion, respectively. The both extracts of clove and lavender had effective reductive potential, free radical scavenging, superoxide anion radical scavenging, and metal chelating activities at all tested concentrations (20, 40, and 60 μg/ml). Those various antioxidant activities were compared to standard antioxidants such as BHA, BHT, and α-tocopherol. In addition, total phenolic compounds in the both extracts of clove and lavender were determined as gallic acid equivalent.  相似文献

Different solvent extracts of endemic Sideritis (Labiatae) species, Sideritis congesta Davis et Huber-Morath and Sideritis arguta Boiss et Heldr, were analyzed for free flavonoids (quercetin, apigenin, myricetin and kaempferol) and cinnamic acid derivatives (rosmarinic acid, ferulic acid, caffeic acid, p-coumaric acid and chlorogenic acid) using HPLC-DAD. All the phenolics were quantified in acid-hydrolyzed extracts, except rosmarinic acid, chlorogenic acid and myricetin which were quantified in raw samples. Antioxidant activities of extracts of these two plants and many of their components in pure form were evaluated based on DPPH. and ABTS.+ assays. In general, S. arguta extracts displayed higher antioxidant activity than S. congesta extracts possibly due to their richness in antioxidant components of strong activity. Acetone extract of S. arguta, with its strikingly high TEAC value of 3.2 mM trolox and low IC50 value of 38.3 ??g/mL showed the highest antioxidant potency among all extracts. ??-tocopherol, the positive control, displayed IC50 and TEAC values of 33.8 ??g/mL and 2.9 mM trolox, respectively. No direct correlation was found between antioxidant activities and total phenolic contents of the plant extracts studied.  相似文献

This work was undertaken to explore the potential of fruit waste materials as sources of powerful natural antioxidants. The peels of eight kinds of fruits commonly consumed and grown in Thailand were used. The ethanolic fruit peel extracts were subjected to the scavenging tests of DPPH and ABTS radicals. Results from both assays were in good agreement that the top three markedly high free radical-scavenging power was from the peel extracts of Punica granatum (pomegranate), Nephelium lappaceum (rambutan), and Garcinia mangostana (mangosteen). The IC50 values to quench the DPPH free radicals of these three extracts were 0.003, 0.006, and 0.023 mg/ml and the trolox equivalent antioxidant capacity (TEAC) values from ABTS assay were 4.066, 3.074, and 3.001 mM/mg, respectively. The extract of mangosteen peel showed moderate toxicity to Caco-2 cells and high toxicity to peripheral blood mononuclear cells (PBMC) with the IC50 values of 32.0 and 4.9 μg/ml, respectively. Pomegranate peel extract stimulated Caco-2 cell and PBMC proliferation with the ED50 of 4.7 and 44.4 μg/ml, respectively. Peel extract of rambutan exhibited extremely high value of IC50 (>100 μg/ml) against both cell types indicating non-toxic activity to the cells. It was concluded that the peel of rambutan may be considered potentially useful as a source of natural antioxidants for food or drug product because of its high antioxidant activity and non-toxic property to normal cells.  相似文献

Antioxidant activity of aqueous hull extracts of commonly consumed legumes in India namely Vigna radiata (mung bean), Cicer arietinum (Bengal gram) and Cajanus cajan (pigeon pea) was studied by several in vitro assays. The extracts showed high phenolic content and excellent DPPH scavenging activity (75-85%) at very low concentration. The IC50 values for superoxide radical scavenging activity was also low (85-150 ??g/ml). The antioxidant activity coefficient as determined by ??-carotene bleaching assay was very high (828-1086). The hull extracts also exhibited high reducing power indicating good antioxidant potential. Of the extracts studied pigeon pea hull extract had the highest antioxidant activity. It also exhibited good antibacterial activity against Bacillus cereus. This study indicated that legume hulls have good antioxidant potential comparable to the synthetic antioxidant butylated hydroxy toluene and could therefore have application in food, cosmetic and pharmaceutical industry.  相似文献

The ethanol extracts from 24 samples plant species commonly found in Thailand were investigated and compared on their antioxidant activity by ABTS assay. The ethanol extract from the leaves of guava (Psidium guajava) showed the highest antioxidant capacity with the TEAC value of 4.908 ± 0.050 mM/mg, followed by the fruit peels of rambutan (Nephelium lappaceum) and mangosteen (Garcinia mangostana) with the TEAC values of 3.074 ± 0.003 and 3.001 ± 0.016 mM/mg, respectively. The further investigation of guava leaf extracts from different solvents; n-hexane, ethyl acetate, n-butanol, and methanol, was examined using ABTS and FRAP assays. The total phenolic content was done by Folin–Ciocalteu reaction. The results indicated that the methanol fraction possessed the highest antioxidant activity, followed by the butanol and ethyl acetate fractions, respectively. The hexane fraction showed the lowest antioxidant activity. The results demonstrated that the mechanism of antioxidant action of guava leaf extracts was free radical scavenging and reducing of oxidized intermediates. The phenolic content in guava leaf fraction played a significant role on the antioxidant activity via reducing mechanisms.  相似文献

Sphallerocarpus gracilis roots harvested at two different times (T1: 12-10-2009 and T2: 15-04-2010) were analysed for their main nutrients, phenolics, antioxidant activity, DNA damage protective effect and microstructure. Results revealed that higher amounts of starch were detected in T1 S. gracilis root, whereas higher levels of reducing sugar, protein, amino acids and phenolic compounds were determined in T2 S. gracilis root. The antioxidant activity was assessed by DPPH, FRAP and TEAC assay and T2 S. gracilis root was found to possess higher antioxidant activity. Also, results of the correlation analysis indicated that caffeic, p-coumaric and gallic acids contributed substantially to the antioxidant property and there was significant correlation between DPPH radicals scavenging capacity and FRAP and TEAC. The highest and lowest DNA damage protective effects were found of free phenolic extracts from T2 and T1 S. gracilis roots, respectively. The microstructure of S. gracilis root varied greatly with the harvest time.  相似文献

The antioxidant properties of aqueous extracts of 6 medicinal plants, Phyllanthus emblica, Terminalia chebula (black and yellow), Terminalia arjuna, Balsamodendron Mukul, and Alium sativum against lipid peroxidation in mouse tissues were investigated. Extracts showed inhibition against thiobarbituric acid reactive species (TBARS) induced by pro-oxidant (5 μM sodium nitroprusside) in the mouse brain and liver. Extracts displayed high free radical scavenging activities against DPPH (IC50, 23.23±1.2 μg/mL, P. emblica), 20.24±0.9 μg/mL (T. chebula yellow), 17.33±1.1 μg/mL (T. chebula black), 19.44±0.45 μg/mL (T. arjuna), 56.59±2.1 μg/mL (Balsamo-dendron Mukul), and higher than 200 μg/mL (A. sativum). Higher antioxidant and inhibitory effects of T. chebula black are attributed to a higher phenolic content, Fe(II) chelating ability, reducing ability, nitric oxide radical scavenging, and free radical scavenging activity. Oxidative stress in the brain and liver could potentially be managed/prevented by dietary intake of these plants.  相似文献

This study aimed to evaluate the in vitro antioxidant and enzyme inhibitory activities of ethyl acetate, methanol, and water extracts of Bituminaria bituminosa. In phosphomolybdenum assay, the methanol extract showed the highest activity (166.78 μmol TEs/g dry plant). The water extract exhibited the highest scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH?) and 2,2-azino-bis (3-ethylbenzothiazloine-6-sulphonic acid) (ABTS?+). In addition, it exhibited the highest activity in cupric ion reducing (CUPRAC) and ferric reducing antioxidant power (FRAP) assays (41.26 and 46.82 μmol TEs/g dry plant). The extracts did not show cholinesterase and tyrosinase inhibitory activity. However, α-glucosidase inhibition assay resulted in the superiority of water extract (1233.86 μmol ACEs/g dry plant). In the case of α-amylase inhibitory assay, the ethyl acetate extract showed the highest activity (53.65 μmol ACEs/g dry plant). The water extract exhibited the highest phenolic content (31.70 μmol GAEs/g dry plant). In contrast, the methanol extract was found rich in flavonoid compounds (5.29 μmol REs/g dry plant). The water extract contained considerable amounts of rosmarinic acid, luteolin, quercetin, and rutin. Therefore, it can be used as a source of new and alternative antioxidant and enzyme inhibitory agents.  相似文献

In this study, antioxidant and antimicrobial activities of water and ethanol extracts of anise (Pimpinella anisum L.) seed (PAS) were investigated. The antioxidant properties of both extracts of PAS were evaluated using different antioxidant tests, including reducing power, free radical scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, and metal chelating activities. Twenty μg/ml of water and ethanol extracts exhibited 99.1 and 77.5% inhibition of peroxidation in linoleic acid system, which was greater than the same concentration of α-tocopherol (36.1%). These various antioxidant activities were compared with synthetic antioxidants such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), and α-tocopherol. The water extract of PAS exhibited greater antioxidant capacity than that of ethanol. Antimicrobial activity tests were carried out using disc diffusion methods with 10 microbial species.  相似文献

Five extracts from five plants, of which four are endemic to Turkish flora, were screened for their possible in vitro antioxidant activities by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid. In the first case, Pelargonium endlicherianum extract exerted greater antioxidant activity than those of other plant extracts studied with an IC50 value of 7.43 ± 0.47 μg/ml, followed by Hieracium cappadocicum (30.0 ± 0.14 μg/ml). When compared to the synthetic antioxidant BHT (18.0 ± 0.40 μg/ml), the methanolic extract of P. endlicherianum exhibited more than two fold greater antioxidant activity. In the β-carotene/linoleic acid test system, the most active plant was P. endlicherianum with 72.6% ± 2.96 inhibition rate, followed by H. cappadocicum (55.1% ± 2.33) and Verbascum wiedemannianum (52.5% ± 3.11). Antioxidant activities of curcumin and ascorbic acid were also determined as positive controls in parallel experiments.  相似文献

Changes in β-glycosidase activity, total phenolic and isoflavone contents, and antioxidant activities during the fermentation of brown soybeans (Galmi) fermented food cheonggukjang by the potential probiotic Bacillus subtilis CSY191 were investigated. The total phenolic and isoflavone-glycoside and -aglycone contents as well as ABTS radical scavenging activity and FRAP assays of extracts increased, but malonylglycoside content decreased after a steaming process. It was found that fermentation enhanced the total phenolic and isoflavone-aglycone and -malonylglycoside contents, corresponding to antioxidant activities increased, but isoflavone-glycoside content decreased. The highest levels of daidzein, glycitein, and gensitein were present at concentrations of 156.5 μg/g, 10.2 μg/g, and 2.5 μg/g after 48 h of fermentation. In addition, ABTS radical scavenging activity and FRAP assay increased significantly during fermentation. We suggest that the high antioxidant activity of cheonggukjang of brown soybeans might be related to the markedly higher levels of total phenolic content and isoflavone-aglycones achieved during fermentation.  相似文献
