Risk factors suggestive of relatively late exposure to EBV have been consistently associated with Hodgkin's disease (HD) in younger adults. In addition, evidence of EBV infection has been found in the Reed-Sternberg cells themselves in about one-third to one-half of all HD cases. However, no study yet published has correlated these childhood social environment risk factors with the presence of EBV in Hodgkin's tumor cells. We examined whether EBV-positive HD occurs in those patients whose childhood environment would predispose them to relatively late exposure to EBV. The study population consisted of 102 cases of mixed cellularity (MC; n = 25) or nodular sclerosing (n = 77) HD. Samples that tested positive for either EBV-encoded RNA or latent membrane protein or both were considered EBV-positive. Of the 102 cases, 83 completed a questionnaire regarding childhood social environment. The association with EBV-positivity was estimated by the odds ratio (OR) with 95% confidence intervals (CI). Twenty-two percent of the cases were EBV-positive. These cases were more likely to be MC (OR, 6.2; CI, 2.3-16.3) and male (OR, 3.4; CI, 1.3-9.0). History of infectious mononucleosis (IM) was not predictive of EBV-positivity, with only 3 of 14 such patients being EBV-positive (P = 0.82). Contrary to our hypothesis, no association between EBV and childhood environment risk factors was identified. The association of EBV with MC histology and male gender agrees with previous reports. The most intriguing finding was the dissociation between IM history and EBV-positivity, in that almost all of the cases with a history of IM were EBV-negative.  相似文献

To define mechanisms involved in the transport of immunoglobulins into intestinal fluids, we localized IgM, IgA, IgG, and secretory component (SC) in human intestinal mucosa by the peroxidase-labeled antibody technique. At the light microscopic level, immunocytes containing IgA, IgM, or IgG were found in the lamina propria. IgA, IgM, and SC were prominent in the epithelium of gland crypts; IgG was limited to a few cells at tips of villi. At the electron-microscopic level, SC was localized to perinuclear spaces, endoplasmic reticulum, saccules associated with Golgi complexes, cytoplasmic vesicles, and lateral and basal plasma membranes of columnar epithelial cells. IgA and IgM, but not IgG, also were localized to plasma membranes and cytoplasmic vesicles of these cells. Neither the immunoglobulins nor SC was found within other types of epithelial cells (Paneth, goblet, endocrine). The findings provide evidence that (1) the site of SC synthesis in intestinal epithelium is secretory columnar cells, principally those in gland crypts; (2) the polymeric immunoglobulins IgM and IgA are translocated through such SC-containing cells by a process that involves formation of cytoplasmic vesicles; (3) IgM and IgA could combine with SC during transcellular transport (likely sites are lateral or basal plasma membranes or supranuclear cytoplasm); (4) the monomeric immunoglobulin IgG does not share the transepithelial cell route involved in IgM and IgA transport.  相似文献

It was shown that polyreactive properties of natural serum immunoglobulins or some monoclonal antibodies (i.e. their ability of binding to various antigenically dissimilar antigens) can be induced by the treatment with chaotropic ions (3.5 M KSCN, 5-30 min at 20 (C). Such transformation of monoclonal antibodies or serum immunoglobulins into polyreactive immunoglobulins (PRIG) seems to result of the changes in immunoglobulin conformation but is not from their unblocking. Interaction of PRIG with some antigens immobilized on the plate is strongly dependent on temperature unlike that of monospecific antibodies.  相似文献

Immunoglobulin molecules are localized in the dentinal tubules of non-carious and carious teeth, but their possible role in caries invasion is not understood. This study sought to examine the effects of immunoglobulin molecules on dentine permeability using a fluid-filtration method. Crown segments cut from impacted human third molars were treated by filtration with 100 micrograms/ml IgG, 100 micrograms/ml IgA or 30 micrograms/ml IgM under a constant pressure. Flow rates were recorded and percent changes in flow rate analysed over time. Filtrates collected at various times were tested for changes in immunoglobulin concentrations by an enzyme-linked immunosorbent assay, and the percent retention of immunoglobulins to dentine was calculated. There was a decreasing non-linear exponential relation between the percent changes in flow rate and filtration time for all three immunoglobulins. The percentage of retained immunoglobulins was significantly related to the filtration time for all three classes of immunoglobulins. Immunoglobulin retention contributed to significant changes in flow rate with time. These in vitro results indicate the potential mechanism of immunoglobulins in decreasing tabular permeability.  相似文献

Dreams provide access to underlying personality structure, defensive and adaptive functions, and they elucidate the psychological forces that lead to overt symptomatic behavior. Two hundred three dreams of 39 personality disordered patients were factor analyzed and compared with Hall and Van de Castle's normative data (Hall C, Van de Castle RI [1966] The content analysis of dreams. New York: Appleton-Century-Crofts). Results included a five-factor solution that sheds light on some core issues of the dreamers. Comparisons between the groups resulted in the personality-disordered group demonstrating more estrangement in their dreams, fewer interactions, and more emotionality. In their interactions, they demonstrated a lower ratio of aggressive interactions yet a higher tendency to view themselves as the aggressor. Results are related to theoretical literature on personality and defensive styles, mostly from a psychodynamic perspective.  相似文献

Serum, Na2SO4-precipitated serum immunoglobulins and bile from 12-week-old fowls, and serum from day-old chicks, were fractionated by Sephadex G-200 gel filtration, DEAE Sephadex A-50 ion exchange chromatography and ultracentrifugation through 10-40 per cent sucrose gradients. Elution of IgM, IgG, IgA and albumin was monitored by examination of fractions in agar gel diffusion against antisera specific to these proteins. Serum and bile from 12-week-old fowls contained IgM and IgA in two molecular sizes and a single molecular size of IgG. Day-old chick serum contained IgM estimated to be 7S, a polymerised form of IgG in addition to the normal 7S component, and a small molecular weight protein antigenically related to IgA. Most of the albumin in bile was of lower molecular weight than serum albumin, while heavy forms of albumin were detected in ultracentrifugation of bile and day-old chick serum.  相似文献

Mortality among adolescents resident in the municipal district of Botucatu, State of S. Paulo, Brazil, according to age group (10 to 14 and 15 to 19 years), sex, occupation and cause of death, during the period from 1984 to 1993, was analysed. The mortality data were obtained from death certificates and the population estimates were based upon census figures. There was great variation in the mortality rates of the years studied. The mortality was greater among the males of the older age group (15 to 19 years), including both students and workers. There was predominance of external causes of mortality which suggests the need for further studies its determining factors. The data indicate that health programs targeted at regarding adolescents, their families and community should be developed since these are avoidable causes of death.  相似文献

1. The phosphonium analogues of choline, phosphorylcholine, CDPcholine and phosphatidylcholine were synthesized chemically and characterized by 1H-NMR and 31P-NMR; in 1,2-distearoyl-DL-glycero-3-phosphorylphosphocholine, the 31P-NMR chemical shift of phosphonium relative to phosphate was--28.2 ppm. 2. A comparison was made of the rates of reaction of choline kinase, cholinephosphate cytidyltransferase, cholinephosphotransferase and phospholipase C on natural and phosphonium substrates. Enzyme reaction rates were similar for all but the cytidyltransferase, which exhibited a 3-fold preference for the normal substrate. 3. Weanling rats were maintained for 6 weeks on a diet in which choline was fully replaced by phospho[1,2-14C2]choline mixed with a trace of [Me-3H] choline. Incorporation of phosphocholine into liver lipids was detectable by 31P-NMR even in crude tissue homogenates. Choline-based phospholipids of liver, kidney, lung and brain were extracted, and phosphocholine incorporation calculated from 31P-NMR peak area ratios. The phosphatidylcholine analogues were separated by preparative thin-layer chromatography. Incorporation of phosphocholine ranged from 33% in lung phosphatidylcholine to 6% in kidney sphingomyelin. Variations in 14C/3H ratio between feed and phospholipid extracts indicated preferences for exogenous choline over phosphocholine varying from 1.3: 1 in brain to 3.2: 1 in liver. The results indicated that phosphocholine is a potentially useful 31P-NMR probe for the study of membrane lipids.  相似文献

It is essential to know how the immune system acts in different neurological diseases, some of them non very well known or of unknown etiology at all. It was applied Reiber and Felgenhauer's formula in 56 patients with different diseases. IgA, IgM, IgG and albumin were quantified in sera and cerebrospinal fluid by simple immunodiffusion. Serum immunoglobulin levels in Black children were estimated and the results analysed in 13 age ranges, from birth up to 15 years of age. Samples from children whose serum immunoglobulins were unlikely to be affected by their clinical condition were chosen for the analysis. The values measured were much higher when compared with conventionally accepted reference ranges in White children within the same age groups. Therefore, it is most important to interpret the results of immunoglobulin measurement within the range for the individual concerned.  相似文献

In order to elucidate the mechanism of induction of serum immunoglobulins polyreactive properties by chaotropic ions, gel-chromatography of immunoglobulins in the medium containing 3.3 M KSCN, has been done. It was found that the transformation of serum immunoglobulins to polyreactive antibodies, which could react with different serological unrelated antigens, was not followed by dissociation of any hypothetical blocking factors. Instead, it was demonstrated that the appearance of polyreactive properties of immunoglobulins was likely to depend on conformational changes of immunoglobulin molecules.  相似文献
