In this study the antioxidant activity of absolute ethanol, 50 % ethanol and water extracts of two species of seaweeds, namely Fucus serratus and Polysiphonia fucoides, were evaluated both in in vitro assays and in 5 % fish oil‐in‐water (o/w) emulsions. The 50 % ethanolic extracts of P. fucoides showed higher antioxidant activity both in in vitro assays and in 5 % oil‐in‐water emulsion in the presence or absence of iron. In spite of the higher phenolic content and very good antioxidant activity in some of the in vitro assays, the absolute ethanol extracts of both the species showed a pro‐oxidative tendency in 5 % fish oil‐in‐water emulsion in the presence or absence of iron. In order to investigate the reason for the higher antioxidant activity of 50 % ethanolic extracts of P. fucoides, these extracts were further fractionated into polyphenol‐rich, protein‐rich, polysaccharide‐rich and low‐molecular‐weight fractions. These fractions were tested both in in vitro and in 5 % oil‐in‐water emulsions. The results of the present study showed that the main effect was due to the phenolic compounds. In conclusion, the 50 % ethanolic extracts of P. fucoides can be a potential source of natural antioxidants as these extracts have antioxidant activities similar to those of synthetic antioxidants such as BHT.  相似文献

Crude ethanol/ethyl acetate extracts of industrial evening primrose (Oenothera biennis L.) seed meal were separated into six fractions using the Sephadex LH‐20 column chromatography and 96% aqueous ethanol as a mobile phase. Their antioxidant activities were tested in sunflower and rapeseed oils by using an Oxidograph apparatus at a temperature of 110 °C. Only the fractions III and IV displayed a pronounced antioxidant activity while the other fractions were either inactive or even pro‐oxidative. The active fractions contained phenolic acids and their esters; gallic acid, methyl and ethyl gallates, protocatechuic acid and its methyl ester were identified by GC/MS. Catechin was present, too, but exhibited only moderate antioxidant activity in sunflower oil.  相似文献

Majorana syriaca is an important food‐flavouring ingredient in the Middle Eastern culture known commonly as Za'atar. The components of the plant were extracted in a Soxhlet apparatus sequentially with solvents of increasing polarity, specifically with petroleum ether (P), diethyl ether (D), ethyl acetate (EAc), and ethanol (E). The activity of the extracts to scavenge the 2,2‐diphenyl‐1‐picrylhydrazyl radical (DPPH?) followed the order D > E > EAc > P. Moreover, the antioxidant activity in corn oil at concentrations of 200, 500 and 1000 ppm was monitored through peroxide value and conjugated hydroperoxide measurements and revealed that D and E extracts resulted in the highest protection followed by EAc and P extracts. Increasing the concentration from 200 to 500 ppm depressed peroxide formation, while a further increase had no effect. The main components of the P extract according to GS‐MS analysis were thymol (42.89%) and carvacrol (42.75%). The remaining extracts comprise flavonoids and phenolic acids that provide the antiradical and antioxidant activity.  相似文献

The capacity of the natural antioxidant from barley husks to retard oxidation of PUFA in cod liver oil (Gadus morhua) was investigated and compared to synthetic antioxidants. The results confirm the efficacy of a natural antioxidant derived from barley husks to slow down the progress of lipid hydrolysis and increase oxidative stability in cod liver oil. The rates of lipid hydrolysis and lipid oxidation were slowed down with increasing concentration of natural antioxidant used. Using 100 mg of the natural antioxidant was more effective than some synthetic antioxidants (BHA 200 mg and BHT 200 mg) against primary and secondary oxidation. The use of propyl gallate (PG) as an antioxidant (200 mg/kg in cod liver oil) was the most effective antioxidant employed in reducing the production of primary and delaying secondary oxidation products. The formation of free fatty acids (FFA) was significantly lower in samples with natural antioxidant (BE200 and BE100) than in the control samples. BHA and BHT were the most effective antioxidants employed to delay the lipid hydrolysis. Practical applications: The use of barley husks, which are residues of the brewing process, was optimized to obtain a crude antioxidant extract. Natural extracts of phenolic compounds with high antioxidant activity were obtained after prehydrolysis and delignification of barley husks. The raw extracts show more than two‐fold antioxidant capacity compared to BHT in terms of EC50. The results demonstrate the efficacy of a natural antioxidant derived from barley husks. The extract could be used in fatty foods (such as butter, oil, etc.) to prevent rancidity.  相似文献

The food industry is seeking natural antioxidants for edible oils that have comparable activity to synthetic counterparts. In this study, Osage orange extract (OOE) rich in osajin (42.9%) and pomiferin (30.0%) was obtained after hexane extraction of the fruit, and its antioxidant activity was examined in stripped soybean oil (SBO) and fish oil (FO), in which antioxidants and polar compounds were removed. The antioxidant activity of OOE was compared with commercial natural antioxidants (i.e., rosemary extract and mixed tocopherols) and a synthetic antioxidant, butylated hydroxytoluene (BHT), during storage at 25 and 40 °C. The 0.1% OOE had stronger antioxidant activity than 0.1% rosemary extract and 0.1% mixed tocopherols in both oils at 25 and 40 °C. Its activity was similar to 0.02% BHT in SBO and was similar or slightly stronger than 0.02% BHT in FO. When OOE was studied at 0.05, 0.1, and 0.2%, there was a weak dose–response in SBO but a stronger dose–response in FO. Headspace volatile analysis using solid phase micro-extraction (SPME) combined with GC–MS indicated that 0.1% OOE was very effective in preventing the formation of volatile oxidation products in both oils. Although it should be further tested for safety before the actual use, this study shows that OOE can be developed as an antioxidant for edible oils.  相似文献

We assessed the polyphenolic contents and antioxidant potential of the aqueous, ethanol and methanol stem bark extracts of Jatropha curcas. The total phenol, flavonoids, flavonols and proanthocyanidin contents of the extracts were evaluated to determine their effect on the antioxidant property of this plant, using standard phytochemical methods. The antioxidant and free radical scavenging activity of ethanol, methanol and aqueous extracts of the plant were also assessed against 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), ferric reducing, nitric oxide (NO), superoxide anion, (O(2) (-)) and hydrogen peroxide (H(2)O(2)) using spectroscopic methods and results were compared with that of butylated hydroxyl toluene (BHT) and ascorbic acid as standards. The concentrations of different classes of phenolic compounds were higher in methanol and ethanol extracts compared to aqueous extracts. There was correlation between total phenol, total flavonoids, total flavonol and total proanthocyanidins (r = 0.996, 0.978, 0.908, and 0.985) respectively. There was correlations between the amount of phenolic compounds and percentage inhibition of DPPH radicals scavenging activity of the extract (r = 0.98). Findings from the present study indicated that J. curcas is a potential source of natural antioxidants and may be a good candidate for pharmaceutical plant based products.  相似文献

采用清除二苯代苦味肼基(DPPH)自由基、清除[2,2′-连氨-(3-乙基苯并噻唑啉-6-磺酸)二氨盐](ABTS)自由基及铁离子还原/抗氧化能力(FRAP)测定法,对开封产的3种白色菊花(兼六香白、国华万胜及白玉带)不同溶剂提取物的体外抗氧化活性进行了评价,将所测定结果与水溶性维生素E(6-hydroxy-2,5,7,8-tetramethyl-chroman-2-carboxylic acid,Trolox)及阳性对照二丁基羟基甲苯(BHT)进行了比较。结果表明,3种菊花的不同溶剂提取物抗氧化活性不同。同一种菊花的甲醇提取物具有很好清除DPPH自由基和还原铁离子的能力,而石油醚提取物几乎无活性。菊花的3个品种中,兼六香白和国华万胜的抗氧化活性较好,其活性远远超过白玉带的抗氧化活性。9个提取物中,兼六香白的甲醇提取物总的抗氧化活性最好。它对DPPH自由基的清除能力(IC50值为20.49 mg/L)比BHT(IC50值为18.92 mg/L)作用略低;其还原Fe3+的能力(FRAP值为731.73±1.77μmol TE/g)比BHT(1 581.68±97.41μmol TE/g)作用低1/2。3种方法测定结果基本一致,其中以DPPH法和FRAP法相关性最好(R=0.982 0)。  相似文献

The extraction of biologically active compounds from eggplant pulp by modified supercritical CO2 extraction was investigated. Response surface methodology was used to optimize the extraction of phenolic compounds and antioxidant capacity by assessing the pressure, temperature, and cosolvent percentage. The results demonstrated that the maximum phenolic content (3,530.79 mg CAE/100 g extract) and antioxidant capacity (4,593.22 μmol TE/g extract) were observed at 56.8°C, 280 bar, and 1.22% of ethanol and were higher than those obtained by conventional solvent extraction. Four phenolic acids were identified in the supercritical extracts and not in the conventional extracts using HPLC-DAD analysis, suggesting that modified supercritical CO2 extraction is more efficient and selective.  相似文献

In the present study, the effect of enzymatic hydrolysis on the antioxidant activity of three red seaweeds (Chondria cornuta, Chondria dasyphylla, and Murrayella periclados) and two green seaweeds (Cladophora sp. and Ulva lactuca) from the Kuwait coast were evaluated. The seaweeds were hydrolyzed with five carbohydrases and three proteases, and the resulting extracts were tested for antioxidant activity. The total phenolic content and yield of the extracts varied greatly depending upon the species and enzyme used for hydrolysis. Of the 40 enzymatic extracts screened for antioxidant activity, the Viscozyme and Alcalase extracts of M. periclados, Neutrase and Ultraflo extracts of Cladophora sp., and Neutrase extracts of C. cornuta had high antioxidant activity compared to other enzymatic extracts in various in vitro assays. Fractionation of the extracts revealed that the radical scavenging and reducing power of the extracts were mainly due to phenolic fractions. In contrast, the iron-chelating ability was mainly due to protein fractions. The level of prevention of lipid oxidation in the liposome model system varied for different fractions of extracts and did not correlate with total phenolic content and other antioxidant properties. The results of the study show that, by hydrolyzing seaweeds with specific enzymes, customized seaweed extracts with specific bioactivity could be obtained.  相似文献

The pepino (Solanum muricatum) is an edible and juicy fruit native to the Andean region which is becoming increasingly important. However, little information is available on its phenolic composition and bioactive properties. Four pepino varieties (37-A, El Camino, Puzol, and Valencia) and one accession (E-7) of its close wild relative S. caripense were characterized by HPLC-DAD-MSn/ESI. Twenty-four hydroxycinnamic acid derivatives were detected (5 to 16 compounds per variety or accession), with differences of more than two-fold for their total content among the materials studied. The major phenolics in the pepino varieties were chlorogenic acids and derivatives, while in S. caripense a caffeoyl-synapoyl-quinic acid was the major compound. The in vitro antioxidant capacity (DPPH (2,2-diphenyl-1-picrylhydrazyl hydrate), ORAC (oxygen radical absorbance capacity), and TRC (total reducing capacity) tests) was higher in S. caripense. Pepino and S. caripense extracts were not toxic for RAW 264.7 macrophage cells, and the raw extracts inhibited NO production of the lipopolysaccharide (LPS)-stimulated macrophages by 36% (El Camino) to 67% (37-A). No single variety ranked high simultaneously for hydroxycinnamic acids content, antioxidant activity and biological activity. We suggest the screening of large collections of germplasm or the use of complementary crosses between Puzol (high for hydroxycinnamic acids and biological activity) and S. caripense E-7 (high for antioxidant activity) to select and breed pepino varieties with enhanced properties.  相似文献

Micronutrients (tocols, sterols, and total phenolic) and antioxidant activities of 15 varieties of common vegetable oil samples obtained from different countries are investigated. All methanol extracts are assayed for total antioxidant ability and cellular antioxidant activity (CAA) using oxygen radical absorbance capacity (ORAC) method and CAA assay. CAA has been widely used in the evaluation of food antioxidants recently. It quantifies antioxidant capacity utilizing a HepG2 cell model, which is more biologically representative. Linseed and sesame oils show much higher CAA values than the others tested; however, levels of walnut, sunflower, and coconut oils are extremely low, which are hard to be quantified. A significant correlation between the ORAC and CAA values and total phenolic content (p < 0.05) is observed. High‐phenolic olive oil has the highest level of phenolics and the highest ORAC, while linseed oil has the highest CAA value. Based on this, choosing proper edible oil consumption may reduce oxidative damage of human body and promote the precision processing of edible oil such as retaining beneficial ingredients moderately. Practical Application: This study demonstrates the evaluation of the universality of vegetable oils by the cellular antioxidant model and provides a data reference for the selection of edible oils with excellent antioxidant properties.  相似文献

Sour cherry (Prunus cerasus) is rich in biologically active phenolic compounds. These compounds are concentrated in fruit skin and most of them remain in the leftovers during the production of juice. Supercritical carbon dioxide extraction was used to separate phenolic compounds from sour cherry pomace. The effects of temperature, pressure and the addition of ethanol on anthocyanin and the total phenolic content and radical scavenging activity of the extracts were investigated. The best results were acquired for 35°C, 10 MPa and 80% ethanol addition. A strong correlation was found between the phenolic content and other features of the extracts.  相似文献

The total phenolic content, total flavonoid content, vitamin C content, and antioxidant activities of ethanol extracts from different kiwifruit varieties (Actinidia kolomikta, Actinidia arguta, Actinidia chinensis) were determined in this study. Multiple scavenging activity assays including the hydroxyl radical, O(2) (-)·radical, DPPH, and the ABTS(+) radical scavenging activity assays were used to identify the antioxidant activities of Actinidia extracts. The cell viability of HepG2 and HT-29 cells was also examined in this study. The results demonstrated that the Actinidia kolomikta extract had a higher antioxidant activity than the other two Actinidia extracts. There is a positive correlation between antioxidant activity and the polyphenols and vitamin C content in all three extracts (R(2) ≥ 0.712, p < 0.05). The Actinidia arguta extract had the highest inhibitory effect on HepG2 and HT-29 cell growth. These results provide new insight into the health functions of fruit and demonstrate that Actinidia extracts can potentially have health benefits.  相似文献

作为棕榈科植物Euterpe属中分布最为广泛、研究与商业化程度最高的品种,Euterpe oleraceaE. oleracea)的果实(为亚马逊莓的一种)因其卓越的抗氧化性能而跻身“超级水果”之列,同时也成为国际上新的研究热点。本文综述了E. oleracea的相关成分和作用研究现状,指出其富含不饱和脂肪酸、氨基酸、维生素等营养成分,并含有大量的以酚类化合物为主的生物活性物质,例如黄酮类化合物、酚酸、花青素与原花青素等。其中,总脂肪酸、亚油酸、阿魏酸、槲皮素、原儿茶酸、原儿茶酸甲酯等成分被证实具有皮肤美白作用。对E. oleracea进行的细胞实验和动物实验发现,其果肉提取物具有抑制酪氨酸酶活性以及使细胞免受紫外线引起的氧化应激伤害等潜在的皮肤美白功效。文章建议在未来的研究中,对E. oleracea的美白成分进行进一步挖掘,并通过动物实验与临床试验对其美白功效进行验证。  相似文献

Natural antioxidants to inhibit oxidation in edible oils are in high demand. Grape pomace is an abundant, inexpensive source of polyphenolic antioxidants, which are responsible for numerous health benefits. We examined pomace from eight varieties of Midwestern hybrid grapes for phenolic content and antioxidant activity. Ethanolic extracts produced from the pomace of each grape variety were added to two model systems, bulk soybean oil and oil‐in‐water emulsions, to determine antioxidant activity. Oxidation was monitored in each model system at a temperature appropriate to that particular system. While the extracts had relatively little effect in bulk oil, we observed dose‐dependent antioxidant effects of some extracts in oil‐in‐water emulsions. Oxidation in bulk oils was assessed via total polar compounds and polymerized triacylglycerols. Oxidation in emulsions was assessed by peroxide value, headspace oxygen measurements, gas chromatography of headspace volatiles, and fatty acid analysis. Pomace extracts derived from red grapes generally outperformed those from white grapes, with the Marechal Foch variety showing high antioxidant activity at intermediate concentrations. At higher concentrations, Marechal Foch, Corot Noir, Frontenac, and Norton extracts showed promising antioxidant activity. This is the first report on antioxidant activity in an oil and emulsion setting for many of these grape varieties.  相似文献

The antioxidant activity of different Satureja montana L. subsp. kitaibelii extracts was tested by measuring their ability to scavenge reactive hydroxyl radical during the Fenton reaction, using ESR spectroscopy. Also, the influence of these extracts on lipid peroxyl radicals obtained during lipid peroxidation of: (I) sunflower oil (37 ºC, 3h) induced by 4,4′-azobis(4-cyanovaleric acid) (ACVA) and (II) liposomes induced by 2,2′-azobis(2- amidino-propane)dihydrochloride (AAPH) was studied. n-Butanol extract had the best antioxidant activity (100% at 0.5 mg/mL in Fenton reaction system; 89.21% at 5 mg/mL in system I; 83.38% at 5 mg/mL in system II). The antioxidant activities of the extracts significantly correlated with total phenolic content. The antimicrobial activity of Satureja montana L. subsp. kitaibelii extracts was investigated. Petroleum ether, chloroform and ethyl acetate extracts expressed a wide range of inhibiting activity against both grampositive and gram-negative bacteria.  相似文献

A high-performance liquid chromatography method for analyzing the phenolic diterpenes present in rosemary (Rosmarinus officinalis L.) and commercial rosemary extracts is reported. Carnosic acid was the major phenolic diterpene present in rosemary leaves, with lesser amounts of 12-methoxycarnosic acid and carnosol. Several commercial rosemary extracts also were analyzed by this method, and in addition to these three compounds other phenolic diterpenes, such as 7-methoxyrosmanol, 7-methoxy-epirosmanol, and rosmanol, were found in some samples. These latter three compounds seem to be artifacts, produced from carnosic acid by oxidation and cyclization. The major phenolic diterpenes were isolated, and their relative antioxidatn activities in soybean oil were measured by the Rancimat. The potency of carnosic acid was more than twice that of any other compound. The antioxidant activity of pure carnosic acid was compared to butylated hydroxytoluene (BHT), butylated hydroxyanisole (BHA), and tertiary butylhydroquinone (TBHQ) and was several times greater than BHT and BHA but less than TBHQ. Nuclear magnetic resonance data for several of the compounds that were incompletely characterized in previous literature are reported.  相似文献

Extraction and identification of antioxidants in oats   总被引:3,自引:0,他引:3  
Eight separate solvent systems were used with groats and hulls of several lines of oats to determine which system resulted in the most effective, rapid extraction of antioxidants. Antioxidant activity at room temperature was estimated by using thin-layer chromatography (TLC) along with a β-carotene spray. The greatest antioxidant activities were obtained with methanolic antioxidant extracts derived from Noble and Ogle oats and hulls. These extracts were added to soybean oil (SBO) and their effectiveness was compared with that of butylated hydroxytoluene (BHT), tertiary butyl hydroquinone (TBHQ) and a control (no additives) at 32°C, 60°C and 180°C. A petroleum ether extract of Noble oats also was tested in SBO at 180°C. Peroxide values (PV) for oils with added antioxidants during storage at 32°C and 60°C showed that the Ogle oat extract was more effective than the other oat and hull extracts or the control. There were no significant differences in effectiveness among the extracts and the control at 60°C. At 180°C, the stability of each oil was determined by measuring conjugated dienoic acid values (CD) and the relative amounts of the unoxidized fatty acid methyl esters (FAME) 18:2, 18:3 and 18:2/16:0. All oils with added oat and hull extracts had significantly lower CD and significantly higher 18:2/16:0 than oils with added BHT, TBHQ or the control during 14 days at frying temperature. Phenolic and hydroxy-phenolic antioxidant compounds with acids, alcohols, sugars or glycerides attached were tentatively identified in the oat and hull extracts by using TLC and Chromatographic sprays. Peanut skin, which is removed in the peanut blanching process, is rich in bioactive compounds with antioxidant properties. The aims of this study were to measure bioactive compounds in peanut skins and evaluate the effect of gamma radiation on their antioxidant activity. Peanut skin samples were treated with 0.0, 5.0, 7.5, or 10.0 kGy gamma rays. Total phenolics, condensed tannins, total flavonoids, and antioxidant activity were evaluated. Extracts obtained from the peanut skins were added to refined-bleached-deodorized (RBD) soybean oil. The oxidative stability of the oil samples was determined using the Oil Stability Index method and compared to a control and synthetic antioxidants (100 mg/kg BHT and 200 mg/kg TBHQ). Gamma radiation changed total phenolic content, total condensed tannins, total flavonoid content, and the antioxidant activity. All extracts, gamma irradiated or not, presented increasing induction period (h), measured by the Oil Stability Index method, when compared with the control. Antioxidant activity of the peanut skins was higher than BHT. The present study confirmed that gamma radiation did not affect the peanut skin extracts' antioxidative properties when added to soybean oil.  相似文献
