This study reports the development of a new, modified delayed matching to sample (DMS) visual recognition memory task that controls the relative novelty of test stimuli and can be used in human and nonhuman primates. We report findings from normal humans and unoperated monkeys, as well as three groups of operated monkeys. In the study phase of this modified paradigm, subjects studied lists of two-dimensional visual object stimuli. In the test phase each studied object was presented again, now paired with a new stimulus (a foil), and the subject had to choose the studied item. In some lists one study item (the novel or isolate item) and its associated foil differed from the others (the homogenous items) along one stimulus dimension (color). The critical experimental measure was the comparison of the visual object recognition error rates for isolate and homogenous test items. This task was initially administered to human subjects and unoperated monkeys. Error rates for both groups were reliably lower for isolate than for homogenous stimuli in the same list position (the von Restorff effect). The task was then administered to three groups of monkeys who had selective brain lesions. Monkeys with bilateral lesions of the amygdata and fornix, two structures that have been proposed to play a role in novelty and memory encoding, were similar to normal monkeys in their performance on this task. Two further groups--with disconnection lesions of the perirhinal cortex and either the prefrontal cortex or the magnocellular mediodorsal thalamus--showed no evidence of a von Restorff effect. These findings are not consistent with previous proposals that the hippocampus and amygdala constitute a general novelty processing network. Instead, the results support an interaction between the perirhinal and frontal cortices in the processing of certain kinds of novel information that support visual object recognition memory.  相似文献

In steroid-synthesizing cells, like the MA-10 mouse tumor Leydig cells, the peripheral-type benzodiazepine receptor (PBR) is an outer mitochondrial membrane protein involved in the regulation of cholesterol transport from the outer to the inner mitochondrial membrane, the rate-determining step in steroid biosynthesis. Expression of PBR in Escherichia coli DE3 cells, which have no PBR, no cholesterol, and do not make steroids, induced the ability to take up cholesterol in a time-dependent, temperature-sensitive, and energy-independent manner. These cells took up no other steroids tested. Addition of the high affinity PBR ligand PK 11195 to cholesterol-loaded membranes, obtained from cells transfected with PBR, resulted in the release of the uptaken cholesterol. Expression in DE3 cells of mutant PBRs demonstrated that deletions in the cytoplasmic carboxy-terminus dramatically reduced the cholesterol uptake function of PBR, although it retained full capacity to bind PK 11195. Site-directed mutagenesis in the carboxy-terminal region of PBR demonstrated that bacteria expressing the mutant PBR proteins PBR(Y153S) and PBR(R156L) do not accumulate cholesterol, suggesting that amino acids Y153 and R156 are involved in the interaction of the receptor with cholesterol. Considering these results, we postulate the existence of a common cholesterol recognition/interaction amino acid consensus pattern (-L/V-(X)(1-5)-Y-(X)(1-5)-R/K-). Indeed, we found this amino acid consensus pattern in all proteins shown to interact with cholesterol. In conclusion, these data suggest that the expression of PBR confers the ability to take up and release, upon ligand activation, cholesterol. Considering the widespread occurrence of this protein and its tissue and cell specific subcellular localization, these results suggest a more general role of PBR in intracellular cholesterol transport and compartmentalization.  相似文献

The Ni-Al-X (X is an interstitial element or phase) phase diagrams are analyzed to reveal systems that can be used as the basis for designing promising alloys and natural composites based on nickel aluminides reinforced by interstitial phases (natural composites I). The most thermally stable materials are shown to be heterophase alloys and composite materials (CMs) located in the eutectic-type (including degenerate eutectic) pseudobinary sections of ternary or multicomponent phase diagrams. They exhibit insignificant (or zero) dissolution of interstitial phases at operating temperatures and the absence of an intense interaction between CM components (natural composites II). Natural composites I based on the NiAl-or Ni3Al-interstitial phase alloys produced upon cooling from a melt can be reinforced by the refractory thermally stable rigid interstitial phases, namely, borides and carbides, that are present in pseudobinary sections in equilibrium with these nickel aluminides, since the elements forming these phases dissolve completely in matrix melts and the mutual solubility of these phases in the solid state is low. Such borides are TiB2 and HfB2 in equilibrium with β-NiAl, and such carbides are, e.g., TiC and HfC in equilibrium with β-NiAl and La2C3, NbC, and TaC in equilibrium with γ′-Ni3Al. Natural composites II should be produced using solid-phase methods (NiAl with AlN, Y2O3, Al2O3) or a combination of methods, where a refractory interstitial phase of the Al2O3 or Y2O3 type is solid and the intermetallic NiAl or Ni3Al matrix is liquid. NiAl-TiB2 (HfB2), NiAl-Al2O3 (Y2O3), and Ni3Al-La2C3 (NbC, TaC) composites are considered as examples.  相似文献

Readers construct at least 2 interrelated mental representations when they comprehend a text: a textbase and a situation model. Two experiments were conducted with recognition memory to examine how domain knowledge and text coherence influence readers' textbase and situation-model representations. In Experiment 1, participants made remember-know judgments to text ideas. Knowledge and coherence interacted to influence remember judgments differently than know judgments. In Experiment 2, the authors used the process-dissociation procedure to obtain recollection and familiarity estimates. Knowledge and coherence interacted to influence recollection estimates but not familiarity estimates. The authors claim that recollection and familiarity can be used as markers of the different processes involved in constructing a textbase and a situation model. Translocation of alkaline extracellular protease (AEP) into the endoplasmic reticulum of Yarrowia lipolytica is cotranslational and signal recognition particle (SRP)-dependent, whereas translocation of P17M AEP (proline to methionine at position 17, second amino acid in the pro-region) is posttranslational and SRP-independent. P17M signal peptide mutations that resulted in more rapid SRP-dependent translocation of AEP precursor were isolated. Most of these mutations significantly increased hydrophobicity, but the A12P/P17M mutation did not. The switch from SRP-dependent to SRP-independent translocation without a decrease in hydrophobicity (wild type to P17M) and restoration of SRP-dependent translocation without an increase in hydrophobicity (P17M to A12P/P17M) indicate that some factor(s) in addition to hydrophobicity determines selection of targeting pathway. Models of extended forms of wild type and A12P/P17M signal peptides are kinked, whereas the P17M signal peptide is relatively straight. Possibly the conformation/orientation of signal peptides at the ribosomal surface affects SRP binding and consequently the targeting route to the endoplasmic reticulum. Kinked signal peptides might approach SRP more closely more often. Most likely, these effects were only detectable because of the short length and low average hydrophobicity of the AEP signal peptide.  相似文献

利用热力学平衡相计算方法及相应的高温合金数据库对René95,René88DT和an741нп三种合金在平衡相析出温度范围和析出量以及之间的关联性进行了系统的热力学计算对比分析.结果表明:三种粉末高温合金的平衡相种类基本相同,主要平衡相为γ,γ'和碳化物M23C6,MC,M6C.René95合金中的碳化物含量要高于其他两种合金.зп741нп有着较高的γ'相析出温度和较大的析出量.随温度降低,Co和Nb从γ'相中析出,在其周围富集,其中зп741нп合金中尤为明显.合金元素的偏聚度也和合金的初熔点与终熔点之差的大小有关,其熔解区间越大即初熔点和终熔点之差越大,合金元素的偏聚度越大.  相似文献
