The optical rotator is an unbiased, local stereological principle for estimation of cell volume and cell surface area in thick, transparent slabs. The underlying principle was first described in 1993 by Kiêu &38; Jensen ( J. Microsc170, 45–51) who also derived an estimator of length. In this study we further discuss the methods derived from this principle and present two new local volume estimators.
The optical rotator benefits from information obtained in all three dimensions in thick sections but avoids over-/underprojection problems at the extremes of the cell. Using computer-assisted microscopes the extra measurements demand minimal extra effort and make this estimator even more efficient when it comes to estimation of individual cell size than many of the previous local estimators. We demonstrate the principle of the optical rotator in an example (the cells in the dorsal root ganglion of the rat), evaluate its efficiency and compare it with other unbiased, local stereological principles available for estimation of cell volume and surface area.  相似文献

The rotator     
The mean particle volume can be stereologically estimated using the nucleator principle. In the present paper, we discuss another principle for estimating mean particle volume, namely the rotator. The vertical rotator has already been previously described and is supplemented in the present paper by the isotropic rotator. For a collection of particle profiles, simulations show that the variance of the rotator is smaller than that of the nucleator.  相似文献

This paper presents a new local volume estimator, the spatial rotator, which is based on measurements on a virtual 3D probe, using computer assisted microscopy. The basic design of the probe builds upon the rotator principle which requires only a few manual intersection markings, thus making the spatial rotator fast to use. Since a 3D probe is involved, it is expected that the spatial rotator will be more efficient than the the nucleator and the planar rotator, which are based on measurements in a single plane. An extensive simulation study shows that the spatial rotator may be more efficient than the traditional local volume estimators. Furthermore, the spatial rotator can be seen as a further development of the Cavalieri estimator, which does not require randomization of sectioning or viewing direction. The tissue may thus be sectioned in any arbitrary direction, making it easy to identify the specific tissue region under study. In order to use the spatial rotator in practice, however, it is necessary to be able to identify intersection points between cell boundaries and test rays in a series of parallel focal planes, also at the peripheral parts of the cell boundaries. In cases where over‐ and underprojection phenomena are not negligible, they should therefore be corrected for if the spatial rotator is to be applied. If such a correction is not possible, it is needed to avoid these phenomena by using microscopy with increased resolution in the focal plane.  相似文献

Turbulent flow and heat transfer characteristics downstream of a sudden circular pipe expansion are predicted by using the full Reynolds stress model. The uniform wall temperature condition is imposed to the downstream wall. The governing differential equations are discretized by finite volume method with power-law scheme. The results show that the precise inlet conditions taken from the experimental data improve the results, but that the overall magnitude of Nusselt number is still under-predicted due to the use of conventional wall functions.  相似文献

Stereological estimation using vertical sections in a complex tissue   总被引:1,自引:0,他引:1  
A method designed for stereological estimation in a very complex tissue using vertical sections is presented. In some tissues, the random rotation of the tissue for vertical sections may obscure recognition of the anatomical structures of interest. The present method overcomes this problem by generating sections with both a particular orientation, 'mapping sections', and ordinary random vertical sections usable for the required observations. A map describing the positions of the vertical sections is produced to make the complex reference space recognizable. The method is illustrated by estimating the number and size of neurones in the dorsal raphe nucleus of the human brainstem with its dense packing of roughly 100 nuclei within a volume less than 50 cm3.  相似文献

The issue of quench is related to safety operation of large-scale superconducting magnet system fabricated by cable-in-conduit conductor. A numerical method is presented to simulate the thermal hydraulic quench characteristics in the superconducting Tokamak magnet system. One-dimensional fluid dynamic equations for supercritical helium and the equation of heat conduction for the conduit are used to describe the thermal hydraulic characteristics in the cable-in-conduit conductor, The high heat transfer approximation between supercritical helium and superconducting strands is taken into account due to strong heating induced flow of supercritical helium, The fully implicit time integration of upwind scheme for finite volume method is utilized to discretize the equations on the staggered mesh, The scheme of a new adaptive mesh is proposed for the moving boundary problem and the time term is discretized by the-implicit scheme, It remarkably reduces the CPU time by local linearization of coefficient and the compressible storage of the large sparse matrix of discretized equations. The discretized equations are solved by the IMSL The numerical implement is discussed in detail, The validation of this method is demonstrated by comparison of the numerical results with those of the SARUMAN and the QUENCHER and experimental measurements.  相似文献

Previous morphometric methods for estimation of the volume of components, surface area and thickness of the diffusion barrier in fish gills have taken advantage of the highly ordered structure of these organs for sampling and surface area estimations, whereas the thickness of the diffusion barrier has been measured orthogonally on perpendicularly sectioned material at subjectively selected sites. Although intuitively logical, these procedures do not have a demonstrated mathematical basis, do not involve random sampling and measurement techniques, and are not applicable to the gills of all fish. The present stereological methods apply the principles of surface area estimation in vertical uniform random sections to the gills of the Brazilian teleost Arapaima gigas. The tissue was taken from the entire gill apparatus of the right‐hand or left‐hand side (selected at random) of the fish by systematic random sampling and embedded in glycol methacrylate for light microscopy. Arches from the other side were embedded in Epoxy resin. Reference volume was estimated by the Cavalieri method in the same vertical sections that were used for surface density and volume density measurements. The harmonic mean barrier thickness of the water‐blood diffusion barrier was calculated from measurements taken along randomly selected orientation lines that were sine‐weighted relative to the vertical axis. The values thus obtained for the anatomical diffusion factor (surface area divided by barrier thickness) compare favourably with those obtained for other sluggish fish using existing methods.  相似文献

The analysis for the three-dimensional fluid flow past tube banks arranged in equilateral-triangular form at Remax=4,000 is carried out using a large eddy simulation technique. The governing equations for the mass and momentum conservation are discretized using the finite volume method. Parallel computational techniques using MPI (Message Passing Interface) are implemented in the present computer code. The computation time decreases linearly proportional to the number of used CPUs in the present parallel computation. We obtained the time-averaged streamwise and cross-streamwise velocities and turbulent intensities. The present numerical results are compared with the PIV experimental data and agree generally well with the experimental data.  相似文献

针对喷射点胶阀存在点胶一致性差的问题,提出了一种基于体积估计模型的批次PI控制策略。通过对胶体流变特性进行分析,基于胶体剪切应力和剪切速率关系建立了喷射点胶的流量模型,依此推导出体积估计模型。基于建立的模型,运用批次PI算法来实现喷射点胶阀的一致性控制。仿真和实验结果验证了体积估计模型的可靠性,并表明该控制方法能有效地改善喷射点胶的一致性。  相似文献

为了实现对内置式永磁同步电机的高效准确控制,解决电机运行过程中参数变化对控制性能的影响,提出了一种基于在线参数辨识的内置式永磁同步电机最大转矩电流比电流预测控制方法。根据内置式永磁同步电机的转矩特性对最大转矩电流比的d-q电流最优关系进行化简以便于工程计算;并针对电机参数变化对最大转矩电流比工作点偏移的影响进行了分析。同时,对MTPA算法影响较大的关键参数q轴定子电感,转子永磁体磁链进行基于参考模型的自适应辨识,以确保实时的最优d,q轴电流分配。在得到准确辨识的参数和最优电流指令的基础上进行电流的鲁棒预测控制,使实际电流更快地跟踪指令,提高系统的动态性能。实验结果表明q轴定子电感,转子永磁体磁链在线辨识的误差分别小于3%,3.5%,收敛时间小于20 ms;电机能有效跟踪最大转矩电流比工作点,电流响应时间小于30 ms,能够满足内置式永磁同步电机系统稳定可靠、高效快速等运行要求。  相似文献

A general formulation of the instrumental variable filter (IVF) method for parameter identification of a n-DOF (Degrees Of Freedom) mechanical linear system is presented. The IVF is a frequency domain method and an iterative variation of the least-squares approximation to the system flexibilities. Weight functions constructed with the estimated flexibilities are introduced to reduce the effect of noise in the measurements, thus improving the estimation of dynamic force coefficients. The IVF method is applied in conjunction to impact force excitations to estimate the mass, stiffness, and damping coefficients of a test rotor supported on a squeeze film damper (SFD) operating with a bubbly lubricant. The amount of air in the lubricant is varied from nil to 100 percent to simulate increasing degrees of severity of air entrainment into the damper film lands. The experimental results and parameter estimation technique show that the SFD damping force coefficients increase as the air volume fraction in the mixture increases to about 50 percent in volume content. The damping coefficients decrease rapidly for mixtures with larger air concentrations. The unexpected increase in direct damping coefficients indicates the complexity of the SFD bubbly flow field and warrants further experimental verification.  相似文献

The application of design-based stereological methods for estimating nuclear features quantitatively in invasive ductal breast cancer is described. Nuclear number, size and size variability are explored in relation to the tumour grade and patient prognosis. The study includes an examination of the efficiency in estimating different nuclear volumes, and two different estimators of the nuclear size variability are contrasted. Forty-two invasive ductal breast carcinomas diagnosed and graded by two pathologists were used. Both 5-μm and 25-μm-thick sections were obtained from paraffin blocks for stereological study. More undifferentiated tumours show significantly larger nuclei than low-grade tumours. The estimates based on the disector method demonstrate a decrease in the number of tumour cell nuclei per unit volume of tissue from grades 1 to 2 and especially from grades 2 to 3. The univariate survival analysis shows a high prognostic value of the nuclear volume estimates. The study shows that an efficient sampling procedure was performed, particularly when estimating volume-weighted mean nuclear volume using the point-sampled intercepts method. This method is more efficient than estimation of the number-weighted mean nuclear volume using the selector method; however, the latter provides paired estimates of volume- and number-weighted mean nuclear volume, as well as an estimate of the coefficient of variation of nuclear volume in the number distribution of the same cells.  相似文献

Cavitation behavior is very important in pumps for long time operation. However, there is difficulty in predicting the cavitation phenomena of pumps by Computational fluid dynamics (CFD). In order to accurately ascertain cavitation behavior, a comparison between CFD and experimental data is a significant and essential process. The purpose of this study is to analyze cavitating behavior in multistage centrifugal pumps numerically and experimentally. For this investigation an experimental set up was used to obtain cavitation performance results. The CFD method was used to investigate the multistage centrifugal pump performance under developed cavitating conditions. The Reynolds-averaged Navier-Stokes (RANS) equations were discretized by the finite volume method. The two-equation SST turbulence model was adopted to account for turbulent flows. Numerical data were validated with experimental data and a good comparison of results was achieved. Numerically, cavitation performances were obtained for different pump stages and the effects on cavitation were described according to different NPSH (Net positive suction head). The occurrence of cavitation was also described according to NPSH3% in the head drop lines and water vapor volume fraction on the impeller blade. The rapid drop in head at low NPSH was captured for different flow conditions. It was found that for stage to stage performance, the head drop changes could be related to losses inside the pump. It was also shown that the simulation results can truly represent the development of the attached sheet cavitation in the impeller.  相似文献

比较目前在板料成形一步模拟法中常用到的初始域计算方法的特点,针对这些方法的缺点提出采用基于能量理论的空间网格映射法与反向变形法相结合的初始域计算方法.该算法可克服垂直投影法因零投映面积而不能迭代计算的问题,并能解决几何映射法需要寻找对称线、断面线展开法无法解决局部网格畸变的问题.复杂拉延件的实例中,通过对各种结合方案的比较得知:尽管它们获得的最终初始域是一致的,但Desbrun二次能量法及一致参数化法与反向变形法相结合的方案具有较高的稳定性和较好的计算效率,并能解决无法获取通用并高效的一步模拟法初始域求解问题.  相似文献

Evaluating cylindricity is a very important application in metrology. In this paper, we focus on cylindricity evaluation based on radial form measurements. The standard characterization of cylindricity is the notion of zone cylinder, i.e. the cylindrical crown contained between two coaxial cylinders with minimum radial separation and containing all the data points. Unfortunately, the construction of the zone cylinder is a very complex geometric problem, which can be formulated as a nonlinear optimization. Recently a new method (referred to here as the hyperboloid method) has been discussed, which avoids the direct construction of the zone cylinder of a point set, but approximates it with guaranteed accuracy through a computationally very efficient iterative process based on a linearization of the underlying problem. The iterations can be viewed as the construction of a sequence of “zone hyperboloids” tending to the desired “zone cylinder.” An important requirement of the method, however, is that the initial position of the cylindrical specimen axis be nearly vertical, since significant deviations from this condition essentially invalidate the process. It is the purpose of this paper to remove this shortcoming of the hyperboloid technique by providing a simple procedure for appropriately initializing the data (axis estimation). Axis estimation and the hyperboloid technique constitute an integrated methodology for cylindricity evaluation, which is currently the most effective. The theoretical foundations of the method are reviewed from a viewpoint that highlights its essential features and intuitively explains its effectiveness. The analytical discussion is complemented by experimental data concerning a few significant samples.  相似文献

Practical, unbiased stereological methods are described to estimate lung volume and external surface area, and total volume and surface area of relatively large and anisotropic structures (bronchi and arteries) inside the lung. The volume of each of five lung strata was estimated first by fluid displacement and then by computed tomography (CT) using Cavalieri's method; the reliability of CT was assessed through a calibration procedure, and image thresholding criteria for an accurate volume estimation using CT were established. The parallel, perfectly registered CT section images were also used to estimate the external surface area of each stratum by the spatial grid method. Unbiased estimation of internal surface areas in lung is a long-standing problem: since the structures are large and essentially void, large sections are needed; to facilitate identification, thin sections have to be used for light microscopy, and since such structures are anisotropic, the sections should be vertical. A practical stereological design is demonstrated here on an infant lung, which fulfils all these requirements. This study illustrates the potential of using unbiased stereology to characterize infant pulmonary hypoplasia.  相似文献

The 3D spatial arrangement of particles or cells, for example glial cells, with respect to other particles or cells, for example neurons, can be characterized by the radial number density function, which expresses the number density of so-called 'secondary' particles as a function of their distance to a 'primary' particle. The present paper introduces a new stereological method, the saucor, for estimating the radial number density using thick isotropic uniform random or vertical uniform random sections. In the first estimation step, primary particles are registered in a disector. Subsequently, smaller counting windows are drawn with random orientation around every primary particle, and the positions of all secondary particles within the windows are recorded. The shape of the counting windows is designed such that a large portion of the volume close to the primary particle is examined and a smaller portion of the volume as the distance to the primary object increases. The experimenter can determine the relation between these volumina as a function of the distance by adjusting the parameters of the window graph, and thus reach a good balance between workload and obtained information. Estimation formulae based on the Horvitz-Thompson theorem are derived for both isotropic uniform random and vertical uniform random designs. The method is illustrated with an example where the radial number density of neurons and glial cells around neurons in the human neocortex is estimated using thick vertical sections for light microscopy. The results indicate that the glial cells are clustered around the neurons and the neurons have a tendency towards repulsion from each other.  相似文献

对配有ABS系统的汽车在湿滑路面上的制动距离进行了仿真研究。基于有限元商业软件ABAQUS,采用制动过程离散分析方法,考虑ABS功能和路面积水与轮胎间的流固耦合作用,仿真研究了某纵向沟槽花纹子午线轮胎在10mm积水路面各离散速度下的制动情况。根据有限元仿真结果,使用相关分析方法,获得了该轮胎的制动时间和制动距离。对干、湿路面轮胎制动性能仿真结果进行了比较,验证了轮胎湿滑路面制动性能仿真评价方法的有效性。  相似文献
