为研究天然黏土为载体固定化脂肪酶的可行性,采用羟基化、硅烷化处理,对黏土进行改性,并以此为载体吸附固定化脂肪酶,探讨黏土固定化脂肪酶的条件对酶活及蛋白吸附量的影响,优化固定化脂肪酶条件。研究结果表明:黏土经羟基化、硅烷化改性处理后能显著提高固定化酶活和蛋白固定量,其中硅烷化改性最优;载体固定脂肪酶最优条件为:加酶量50 mg/g,载体粒径180—250μm,pH值为4.0,固定化温度25℃,固定化时间2.0 h;与游离酶相比,固定化酶显示出更广的pH值适应性。黏土固定化脂肪酶重复使用10批次后,仍能保留76.85%的初始活力。以天然黏土为载体固定化脂肪酶,具有较好的实际可应用性及操作稳定性,在较低pH值条件下应用具有一定优势。 相似文献

为了提高扩展青霉TS414脂肪酶(PEL)对布洛芬的拆分效率,建立了适于非水相中选择性拆分(R,S)-布洛芬的固定化方法. 结果表明,固定化介质的类型、冻干pH和外加水量等因素对固定化PEL酶促拆分(R,S)-布洛芬有较大影响. 在冻干pH为9.0、外加水量为0、以大孔吸附树脂AB-8为固定化载体的体系中,40℃反应30 h后,拆分反应的转化率可达47%,对映体过量值eeP可达98.75%. AB-8固定化后,PEL在有机相反应体系中的分散性得到了明显改善,大幅度提高了酶促拆分反应的效率;大孔吸附树脂AB-8固定化PEL具有较高的操作稳定性,连续10批拆分反应的平均转化率在47%以上,eeP值均稳定在98%以上. 相似文献

利用海藻酸钙-戊二醛交联法对本实验室筛选到的一株能选择性拆分(R,S)-2-羟基-4-苯基丁酸乙酯〔(R,S)-HPBE〕的沙雷氏菌脂肪酶进行固定化,并对固定化脂肪酶的酶活和拆分条件进行了研究。结果表明,最适反应温度为50℃,pH=7.0,底物浓度40 mmol/L,ρ(酶液)=200 g/L,在该条件下,反应10 h后,(R)-2-羟基-4-苯基丁酸乙酯〔(R)-HPBE〕产率达93.4%,光学纯度(e.e.)为96.2%。连续反应12批次,固定化脂肪酶仍可使(R)-HPBE产率维持在80%以上。 相似文献

Lipase triacylglycerol acylhydrolase, (E.C. 3.1.1.3) is an enzyme that is fully active on aggregated substrates and practically
inactive on monodisperse systems. A lipase immobilized on polymeric membranes has been applied for sunflower oil hydrolysis.
The influence of membrane properties on enzyme activity is studied. Membranes made of poly(vinyl chloride), collagen, cellulose
acetate and polytetrafluoroethylene were used for adsorption of lipase. The porosity and hydrophobicity of membranes did not
influence the lipase activity. The difference of the work of adhesion for the water/membrane system and oil/membrane system
reflected the activity data, while work of adhesion for water or oil (done separately) did not. For oil hydrolysis to occur
on the membrane surface, accessibility of two liquid phases is important, and the lower the difference of work of adhesion
between water and oil, the greater the activity of immobilized lipase. 相似文献

PVA/ Chitosan (CS) composite membrane was studied in this paper, which could be used for enzyme processing of fat and oils. The parameters such as concentration of lipase, pH, and cross-linking agent as well as metal ions, which influence the immobilization of lipase in membrane, were optimized. The immobilized lipase was 0.66 u/cm2 and the recovery of immobilized lipase activity was 24%. The membrane reactor could be used to synthesize monoglyceride (MG) with many batches. 相似文献

PVA/Chitosan(CS) composite membrane was used for enzyme processing of fats and oils. The concentration of lipase and cross-linking agent which influence the immobilization of lipase in membrane were determined. Epichlorohydrin is used as the cross-linking agent. The immobilized lipase is 0.66 u·cm -2 and the recovery of immobilized lipase is 24%. The membrane reactor was tested to synthesis monoglyceride(MG), which could be used many times without loss conversion yield of MG. The PVA/CS lipase membrane reactor is a new reactor for lipase catalytic biphase systems. 相似文献

Pure silica zeolites have been prepared in powder and pellets form with different surface chemical properties in order to
investigate how the binding forces in the lipase enzyme adsorption influence the final conformation of the immobilized enzyme
and its catalytic activity. The catalytic activity of the adsorbed lipase have been tested in the alkyl esters hydrolysis
reaction. The higher alkyl-esters conversion is observed for the lipase immobilized on the supports prepared in OH− media compared to the fluoride media supports. The obtained results can be explained by the two different non-covalent interactions

Immobilization of lipase on bamboo charcoal was confirmed by spectra of FTIR, UV‐Vis, and XPS. Under optimal conditions, the immobilized lipase retains 87 % of the hydrolytic activity of the free lipase. Structural changes of the immobilized lipase were analyzed by circular dichroism and FTIR spectra, indicating that the secondary structure of the lipase was preserved well after immobilization. For catalyzing the enantioselective reaction in heptane, the catalysis efficiency of the immobilized lipase is 1.4 times that of free lipase. Immobilized lipase maintains the selectivity of free lipase. 相似文献
