The effect of two thermal treatments (fast freezing at −40 °C and vapor cooking at 100 °C) on connective tissue extract (CTE) from jumbo squid (Dosidicus gigas) was investigated. Samples of CTE frozen at −40 °C were taken at 0, 3, 5 and 12 min. Also CTE was cooked at 100 °C and samples were taken at 0, 1, 2.5 and 5 min. Light microscopic observations of CTE after 12 min of freezing showed rupture of fibres. The CTE fibres showed agglutination during cooking time. The CTE insoluble fraction increased with freezing and cooking time. Maximum zeta potential value of untreated CTE was detected at pH 5.0 at +30 mV, meanwhile in the frozen CTE it was detected at pH 7.0 at +30 mV and two peaks (at pH 5.5 and 9.0) were observed at +20 mV in the cooked CTE. One endothermic peak was found at 105.9 °C in the untreated CTE, while in the frozen and cooked CTE the endothermic peaks were found at lower temperatures and enthalpies. Electrophoresis analysis of untreated CTE showed three bands. In the frozen CTE two bands appeared above 200 kDa, and in the cooked CTE, a 45 kDa band disappeared. These results suggest that during freezing and cooking processes there were modifications to molecular bonds that hold the integrity of the structure of the connective tissue of the jumbo squid mantle.  相似文献

Giant squid (Dosidicus gigas) inner and outer tunics were subjected to hydrolysis with pepsin prior to gelatin extraction (G1 gelatin) by a mild-acid procedure. Furthermore, a second gelatin extraction (G2 gelatin) was done using the collagenous residues that remained from the first extraction. Pepsin allows the collagen solubilisation and the extraction yield to increase by yielding extracts high in α-chains. G1 exhibited good gel forming ability but G2 showed poor viscoelastic behaviour and low gel strength, in agreement with the results for the molecular weight distribution, which showed a considerably higher content of low molecular weight components. In spite of these differences, both G1 and G2 showed good filmogenic ability and similar properties were found including the absence of colour, opacity, low water vapour permeability and high puncture deformation. Nevertheless, films made from G1 had a higher puncture force than films made from G2 as a result of the different molecular weight distribution.  相似文献

Physicochemical and functional changes of jumbo squid (Dosidicus gigas) mantle muscle stored in ice for 15 days were evaluated. Proximate analysis, pH, protein solubility (PS) and SDS-PAGE profile changes were monitored for muscle proteins, while texture profile analysis (TPA) (gel strength, elasticity and cohesiveness), water holding capacity (WHC) and colour changes were monitored for gels produced from this muscle. Fresh muscle chemical composition varied according to fishing season and/or physiological status presented by specimens. pH and PS were stable (p ? 0.05) throughout the study with a slight tendency to decrease towards the end of study. SDS-PAGE revealed no drastic changes in proteins influencing gelation (i.e., myosin). TPA and WHC of gels decreased towards the end of experiment; however, due to sampling variability, no significant differences (p ? 0.05) were found over storage time. Changes in gel colour were observed with hue angle decreasing towards yellow hues. Improved post-capture management of mantle muscle preserved its integrity and functionality for up to two weeks.  相似文献

The effects of ferrous iron, heating temperature and different additives on the decomposition of trimethylamine oxide (TMAO) to formaldehyde (FA) and dimethylamine (DMA) and generation of free radicals in jumbo squid (Dosidicus gigas) extract during heating were evaluated by electron spin resonance (ESR). The thermal decomposition of TMAO to TMA, DMA and FA and free radical signals was observed in squid extract, whereas no DMA, FA and free radical signals were detected in cod extract or in aqueous TMAO solution in vitro at high temperatures. Significant increase in levels of DMA, FA and radicals intensity were observed in squid extract and TMAO solution in the presence of ferrous iron with increasing temperature. Hydrogen peroxide stimulated the production of DMA, FA and ESR signals in squid extract, while citric acid, trisodium citrate, calcium chloride, tea polyphenols and resveratrol had the opposite effect. Similar ESR spectra of six peaks regarded as amminium radical were detected in the squid extract and TMAO–iron(II) solution, suggesting that the amminium radical was involved in the decomposition of TMAO.  相似文献

The enzyme 5′-nucleotidase of jumbo squid (Dosidicus gigas) mantle was purified and its SDS–PAGE showed a single band of 33 kDa, whereas a protein with a molecular mass of 107 kDa was detected by gel filtration suggesting a homotrimeric nature of this enzyme. Subunits of the named enzyme were not linked by covalent bonds. Isoelectric focusing of this enzyme showed a pI of 3.6–3.8 and presented a hyperbolic kinetics with Vmax of 1.16 μM/min/mg of protein, Km of 1.49 mM, Kcat of 3.48 μM of Pι s−1 and Kcat/Km relation of 356.52 ((mol/L)−1 s−1). Purified enzyme preferred AMP as substrate (by 6.7-folds) than IMP, showing a Km of 6.34 mM, Vmax of 0.19 μM/min/mg of protein a Kcat of 0.3388 mol of Pι s−1 and Kcat/Km relation of 53.44 ((mol/L)−1 s−1). The low Km in relation to purified AMP deaminase of the same organism suggested a high contribution of 5′-nucleotidase in AMP degradation in jumbo squid mantle.  相似文献

Adenosine monophosphate (AMP) deaminase was purified from jumbo squid mantle muscle by chromatography in cellulose phosphate, Q-Fast and 5′-AMP sepharose. Specific activity of 2.5 U/mg protein, 4.5% recovery and 133.68 purification fold were obtained at the end of the experiment. SDS–PAGE showed a single band with 87 kDa molecular mass, native PAGE proved a band of 178 kDa, whereas gel filtration detected a 180 kDa protein, suggesting the homodimeric nature of this enzyme, in which subunits are not linked by covalent forces. Isoelectric focusing of this enzyme showed a pI of 5.76, which agrees with pI values of AMP deaminase from other invertebrate organisms. AMP deaminase presented a kinetic sigmoidal plot with Vmax of 1.16 μM/min/mg, Km of 13 mM, Kcat of 3.48 μM.s−1 and a Kcat/Km of 267 (mol/L)−1.s−1. The apparent relative low catalytic activity of jumbo squid muscle AMP deaminase in the absence of positive effectors is similar to that reported for homologous enzymes in other invertebrate organisms.  相似文献

Differential scanning calorimetry (DSC) was used to follow the thermal denaturation of two differently processed surimis from giant squid (Dosidicus gigas), A (isoelectric precipitation) and B (acid washing). Both surimis were added with several cryoprotectants (sucrose, sorbitol and trehalose), and kept in frozen storage at −15 °C for 6 months. DSC profiles and transition enthalpies showed that protein quality was better in B than in A surimis whether fresh or frozen. It was also demonstrated that the sucrose-plus-sorbitol mixture and trehalose were the most efficient for preserving B and A type formulations respectively after frozen storage. Mechanical (puncture), small amplitude oscillatory (SAOS) and dynamic-mechanical thermoanalyses were performed on both gels from A and B series surimis throughout frozen storage. Gels from surimi A (GA) were considerably brittler than gels from surimi B (GB) and presented significantly lower strain amplitudes. Several correlations among different experimental data have been developed.  相似文献

A cysteine proteinase from Jumbo squid (Dosidicus gigas) hepatopancreas was partially purified by a two step procedure involving ammonium sulfate precipitation and gel filtration chromatography and further by SDS–PAGE. The molecular weight of the proteinase was 24 kDa determined by SDS–PAGE and 23.7 kDa with mass spectrometry. The activity had an optimum pH of 4.5 and optimum temperature of 55 °C under the assay for cathepsin L specific synthetic substrate Z-PAAFC. The cathepsin B and H specific synthetic substrates Z-AAAFC and H-AMC did not show any hydrolysis with the partially purified enzyme. Peptide mapping of trypsin digests of the 24 kDa band from SDS–PAGE showed the squid cysteine proteinase was homologous to cathepsin L from different animal sources. The activity of the partially purified fraction with the cathepsin L specific substrate Z-PAAFC was inhibited 75–89% by enzyme inhibitors specific for cysteine proteinases but was also significantly inhibited by serine and aspartate proteinase inhibitors.  相似文献

Properties and storage stability of two different jumbo squid myofibrillar protein-based films were investigated. Myofibrillar proteins were extracted by isoelectric precipitation after acidic and alkaline solubilization, obtaining the same extraction yield. During extraction the edible fraction, which was discarded during mantle skinning, was recovered, and the fishy flavour produced by nitrogen and other undesirable compounds was removed, although some sarcoplasmic proteins were lost simultaneously. In alkaline-Concentrate (C), myosin unfolding led to water resistant films, less water vapour permeable and more mechanically resistant than acidic–Films (F); whereas acidic-C protein hydrolysis resulted in more transparent and soluble films, with higher protein release (∼80 g/L). During 4 months of storage some structure reorganization occurred, and both films incremented their yellowish tendency, especially the acidic-F, being this attributed to a Maillard reaction with plasticizers. After storage time, water solubility increased in C-films. While acidic-F aggregation led to a protein release reduction and tensile strength improvement, alkali-F became weak and brittle, loosing transparency. C-films offered different filmogenic properties, proving to be promising biodegradable packaging materials.  相似文献

The aim of this work was to study the effect of temperature and air velocity on the drying kinetics and quality attributes of apple (var. Granny Smith) slices during drying. Experiments were conducted at 40, 60 and 80 °C, as well as at air velocities of 0.5, 1.0 and 1.5 m s−1. Effective moisture diffusivity increased with temperature and air velocity, reaching a value of 15.30 × 10−9 m2 s−1 at maximum temperature and air velocity under study. The rehydration ratio changed with varying both air velocity and temperature indicating tissue damage due to processing. The colour difference, ΔE, showed the best results at 80 °C. The DPPH-radical scavenging activity at 40 °C and 0.5 m s−1 showed the highest antioxidant activity, closest to that of the fresh sample. Although ΔE decreased with temperature, antioxidant activity barely varied and even increased at high air velocities, revealing an antioxidant capacity of the browning products. The total phenolics decreased with temperature, but at high air velocity retardation of thermal degradation was observed. Firmness was also determined and explained using glass transition concept and microstructure analysis.  相似文献

The giant squid or dosidicus (Dosidicus gigas) is normally shipped frozen from the coasts of America. During the period prior to freezing-when conditions are not always optimum-and during frozen storage, the functional capacity of the muscle proteins declines, rendering the material useless for certain processes such as conversion to gel. This paper examines the reasons for the lack of a good gel-forming capacity as measured both rheologically (breaking force, breaking deformation and gel strength) and in terms of ultrastructure (scanning microscopy). The study was carried out at four different temperatures known to be critical for fish muscle protein gel formation, and at two salt concentrations. At 35°C the structure was spongier in gels made with 1.5% NaCl, although a true gel was still not formed, as shown by rheological measurements. Above 30°C, the ultrastructure became more cellular, particularly in samples made with 2.5% NaCl, at which concentration gel strength values were higher. However, at neither salt concentration nor at any of the experimental temperatures was the mesh as spongy as in other fish gels reported in the literature. The values of rheological analysis and folding test were correspondingly low.  相似文献

The effect of water content and temperature on glucosinolate thermal degradation in broccoli (Brassica oleracea var. italica) was investigated. Broccoli was freeze dried obtaining batches with water content between 13% and 82% (aw from 0.32 to 0.99). These samples were heated at different temperatures (from 60 to 120 °C) and glucosinolate levels were monitored. To rule out enzymatic breakdown, myrosinase was inactivated prior to heating. Degradation could be described by first-order kinetics for all glucosinolates and all water contents. In the temperature range 60–100 °C the sample with 13% showed the lowest degradation rate, whereas at 120 °C the degradation rate increased with the water content. This particular behavior was reflected by the high activation energy value of the driest sample. Several hypotheses to explain the observed behavior are discussed.  相似文献

Pink discoloration and quality changes of squid (6-10 squids/kg) with and without deskinning during iced storage at different squid/ice ratios (1:1 and 1:2, w/w) for 16 days were investigated. The increases in a* and b*-values of squid mantle were observed with increasing storage time (p < 0.05), indicating the formation of pink color on the mantle. The increase was more pronounced in squid without deskinning with a squid/ice ratio of 1:1 (p < 0.05). No changes in a*-value were observed in deskinned squid throughout the storage, regardless of squid/ice ratio (p > 0.05). However, the slight increase in b*-value was found in the squid with deskinning during the storage. Psychrophilic bacteria counts of squid increased continuously as the storage time increased. Coincidental increases in total volatile base (TVB), trimethylamine (TMA) and ammonia contents were observed during the storage. The rates of increase were greater in the samples with a squid/ice ratio of 1:1 than those found in the samples kept in ice with a squid/ice ratio of 1:2. Pink discoloration, psychrophilic bacteria count, TVB and TMA contents were much lowered when the squid without deskinning was treated with 0.1 g/100 mL sodium azide (NaN3) prior to storage, suggesting that microbial growth was associated with those changes. Therefore, deskinning together with icing using a sufficient amount of ice as well as the use of safe antimicrobial agent could be a means to lower the pink discoloration and retard the losses in quality of squid stored in ice.  相似文献

This paper seeks to compare the ultrastructure of gels made from frozen muscle of giant squid (Dosidicus gigas) at various temperatures with a number of different rheological parameters, with reference to a variety of added ingredients (non-muscle proteins and hydrocolloids) and to NaCl concentration. Interesting data on gel rheological properties were found where formulae containedl-carrageenan, starch and egg white, with a low salt concentration (1.5%). This seems to be because carrageenan forms an independent network which supports the principal structure formed by the fish protein; starch is incorporated into the network and retains water; and egg white forms a supplementary network which helps to improve rheological properties.  相似文献

目的:研究金银花的红外鼓风干燥特性及其动力学模型。方法:金银花在不同温度(35、45、55℃)条件下进行红外鼓风干燥,计算水分比、干燥速率等参数,拟合建立干燥动力学数学模型,并测定干制品的色泽、主要成分等干燥特性。结果:金银花在红外鼓风干燥过程中,干燥温度越高,用时越短,降速阶段为其干燥的主要阶段。干燥的动力学模型拟合结果表明Page模型的预测值与实验值吻合性好,可以用来预测和描述金银花红外鼓风干燥的失水过程。费克第二定律求得干燥过程中的水分有效扩散系数(Deff)在5.06606×10-97.59909×10-8m2/s内,且随温度的升高而增大;阿伦尼乌斯方程计算得出金银花红外鼓风干燥平均活化能为113.569 k J/mol。35℃时金银花的色差值ΔE最小,有效成分含量最高,与阴干相比可提高效率并保证产品质量。结论:金银花红外鼓风干燥的主要阶段为降速阶段,Page模型适于预测和描述其失水过程,35℃红外干燥与阴干相比可提高效率,且产品品质更优。   相似文献

The influence of high pressure processing (HPP) on the inhibition of trimethylamine-N-oxide demethylase (TMAOase) activity and off-odour production in squid treated at 300 MPa for 20 min was investigated during 12 days of refrigerated storage. TMAOase activity of raw squid (21.5 nkat/g) was significantly decreased to approximately 5 nkat/g after 20 min of HPP. The production of dimethylamine (DMA) in HPP-treated squid for 20 min was significantly decreased to 0.31 μmol/g after 12 days of storage. The decrease in DMA was correlated with the decrease in TMAOase activity. At 300 MPa, the number of total aerobic bacteria in squid was reduced by 1.26 log units after 20 min of HPP. The HPP-treated samples effectively reduced the amount of trimethylamine (TMA). Therefore, the HPP could be used as a promising alternative technology to retard the quality deterioration of squid by inhibiting TMAOase activity and microbial growth.  相似文献

A protein concentrate from giant squid (Dosidicus gigas) was produced under acidic conditions and its functional–technological capability evaluated in terms of its gel-forming ability, water holding capacity and colour attributes. Technological functionality of the concentrate was compared with that of squid muscle and a neutral concentrate. Protein–protein aggregates insoluble at high ionic strength (I = 0.5 M), were detected in the acidic concentrate as result of processing with no preclusion of its gel-forming ability during the sol-to-gel thermal transition. Even though washing under acidic condition promoted autolysis of the myosin heavy chain, the acidic concentrate displayed an outstanding ability to gel giving samples with a gel strength of 455 and 1160 g cm at 75% and 90% compression respectively, and an AA folding test grade indicative of high gel strength, elasticity, and cohesiveness. The process proved to be a good alternative for obtaining a functional protein concentrate from giant squid muscle.  相似文献

Nopal (Opuntia ficus-indica) is an endemic plant of the arid and semiarid lands of America. Preservation problems are associated with nopal utilization and processing, so a practical alternative is dehydration. This work deals with the drying of nopal correlated with the textural properties of the dried product. Convective drying studies were performed and the most representative experimental results at two air flow rates (3 and 5 m/s) and two air temperatures (45 and 65 °C) are displayed here. Experiments of osmotic drying with glucose at 40 and 60 °Brix, and drying temperatures of 25 and 45 °C were tested. A combination of both drying processes was also performed. Results have shown forced convection drying with a mostly decreasing trend at the constant rate period. In the osmotic drying, moisture loss was associated to glucose concentration. Texture profile analysis of dehydrated nopal suggested that samples undergoing convective drying have more cohesiveness than samples treated solely by osmotic drying. On the other hand, samples treated by osmotic drying became more elastic. Between the two drying methods studied, the convective drying has shown the best results at the experimental conditions assayed.  相似文献

The effect of High Hydrostatic Pressure (HHP) and other pretreatments on the drying kinetics, antioxidant activity, firmness and microstructure of Aloe vera gel was investigated during convective drying at 70 °C. The pretreatments analyzed were high hydrostatic pressure, blanching, enzymatic and microwaves. Simulation of drying curves was studied through the application of several mathematical models such as Newton, Henderson and Pabis, Page, Modified Page, Wang and Singh, and Weibull. Among them, the Weibull model provided the best fit for the experimental data. All pretreatments increased the water diffusion coefficient compared to the control sample. Microwaves followed by HHP presented the fastest drying rates. All pretreatments modified the microstructure and hence the texture of the product. HHP and microwaves increased firmness while blanching and enzymatic treatments produced a softer final product. Blanching, microwaves and HHP enhanced A. vera antioxidant activity. However, the HHP pretreated samples showed the highest antioxidant activity compared to the rest. Based on these results, HHP together with convective drying offers the chance of producing dried aloe with high antioxidant attributes.  相似文献
