High‐fat diets (HFD) are commonly used in rodents to induce obesity, increase serum fatty acids and induce lipotoxicity in various organs. Invitro studies commonly utilize individual free fatty acids (FFA) to study lipid exposure in an effort to model what is occurring in vivo; however, these approaches are not physiological as tissues are exposed to multiple fatty acids in vivo. Here we characterize circulating lipids in obesity‐prone rats fed an HFD in both fasted and fed states with the goal of developing physiologically relevant fatty acid mixtures for subsequent in vitro studies. Rats were fed an HFD (60 % kcal fat) or a control diet (10 % kcal fat) for 3 weeks; liver tissue and both portal and systemic blood were collected. Fatty acid profiles and absolute concentrations of triglycerides (TAG) and FFA in the serum and TAG, diacylglycerol (DAG) and phospholipids in the liver were measured. Surprisingly, both systemic and portal serum TAG were ~40 % lower in HFD‐fed compared to controls. Overall, compared to the control diet, HFD feeding consistently induced an increase in the proportion of circulating polyunsaturated fatty acids (PUFA) with a concomitant decline in monounsaturated fatty acids (MUFA) and saturated fatty acids (SFA) in both serum TAG and FFA. The elevations of PUFA were mostly attributed to increases in n‐6 PUFA, linoleic acid and arachidonic acid. In conclusion, fatty acid mixtures enriched with linoleic and arachidonic acid in addition to SFA and MUFA should be utilized for in vitro studies attempting to model lipid exposures that occur during in vivo HFD conditions.  相似文献

Structured TAG (STAG) containing medium-chain FA (MCFA) in the sn-1,3 positions and essential FA in the sn-2 position were synthesized by lipase-catalyzed acidolysis. In our previous studies we found that part of the MCFA from STAG could be absorbed in the small intestine; however, it was unclear how they were absorbed. In order to get a better understanding of the metabolism of STAG to improve future design and application of STAG, in the present study lymph lipids collected after feeding STAG were fractionated into different classes and the FA composition of each lipid class was studied by GC after methylation to FAME. Caprylic acid was detected in the fraction of TAG only after administration of 1,3-dioctanoyl-2-linoleyl-sn-glycerol (8∶0/18∶2/8∶0), whereas lauric acid was detected in TAG, DAG, and FFA as well as phospholipids after administration of 1,3-didodecanoyl-2-linoleyl-sn-glycerol (12∶0/18∶2/12∶0). We conclude that the enterocyte has the ability to reacylate the MCFA into TAG and that the intestinal absorption of MCFA from STAG mainly occurs by resynthesis of TAG. Caprylic acid from STAG is not incorporated into phospholipids, whereas lauric acid from STAG can be incorporated into phospholipids.  相似文献

Male Sprague-Dawley rats were fed for one week diets containing 20% by weight fat/oil mixtures with different levels of erucic acid (22∶1n−9) (∼2.5 or 9%) and total saturated fatty acids (∼8 or 35%). Corn oil and high erucic acid rapeseed (HEAR) oil were fed as controls. The same hearts were evaluated histologically using oil red O staining and chemically for cardiac triacylglycerol (TAG) and 22∶1n−9 content in cardiac TAG to compare the three methods for assessing lipid accumulation in rat hearts. Rats fed corn oil showed trace myocardial lipidosis by staining, and a cardiac TAG content of 3.6 mg/g wet weight in the absence of dietary 22∶1n−9. An increase in dietary 22∶1n−9 resulted in significantly increased myocardial lipidosis as assessed histologically and by an accumulation of 22∶1n−9 in heart lipids; there was no increase in cardiac TAG except when HEAR oil was fed. An increase in saturated fatty acids showed no changes in myocardial lipid content assessed histologically, the content of cardiac TAG or the 22∶1n−9 content of TAG at either 2.5 or 9% dietary 22∶1n−9. The histological staining method was more significantly correlated to 22∶1n−9 in cardiac TAG (r=0.49;P<0.001) than to total cardiac TAG (r=0.40;P<0.05). The 22∶1n−9 content was highest in cardiac TAG and free fatty acids. Among the cardiac phospholipids, the highest incorporation was observed into phosphatidylserine, followed by sphingomyelin. With the addition of saturated fat, the fatty acid composition showed decreased accumulation of 22∶1n−9 and increased levels of arachidonic and docosahexaenoic acids in most cardiac phospholipids, despite decreased dietary concentrations of their precursor fatty acids, linoleic and linolenic acids.  相似文献

Seeds from different collections of cultivatedSesamum indicum Linn. and three related wild species [specifically,S. alatum Thonn.,S. radiatum Schum and Thonn. andS. angustifolium (Oliv.) Engl.] were studied for their oil content and fatty acid composition of the total lipids. The wild seeds contained less oil (ca. 30%) than the cultivated seeds (ca. 50%). Lipids from all four species were comparable in their total fatty acid composition, with palmitic (8.2–12.7%), stearic (5.6–9.1%), oleic (33.4–46.9%) and linoleic acid (33.2–48.4%) as the major acids. The total lipids from selected samples were fractionated by thin-layer chromatography into five fractions: triacylglycerols (TAG; 80.3–88.9%), diacylglycerols (DAG; 6.5–10.4%), free fatty acids (FFA; 1.2–5.1%), polar lipids (PL; 2.3–3.5%) and steryl esters (SE; 0.3–0.6%). Compared to the TAG, the four other fractions (viz, DAG, FFA, PL and SE) were generally characterized by higher percentages of saturated acids, notably palmitic and stearic acids, and lower percentages of linoleic and oleic acids in all species. Slightly higher percentages of long-chain fatty acids (20∶0, 20∶1, 22∶0 and 24∶0) were observed for lipid classes other than TAG in all four species. Based on the fatty acid composition of the total lipids and of the different acyl lipid classes, it seems thatS. radiatum andS. angustifolium are more related to each other than they are to the other two species.  相似文献

The lipid and FA compositions of various organs (light muscle, dark muscle, liver, pyloric cecum, and the orbital region) and of the stomach contents of a highly migratory fish species Thunnus tonggol (Bleeker) were analyzed. TAG and phospholipids (PE and PC) were the major lipid classes in the total lipids of T. tonggol. DHA was characteristically the major FA of all the major classes of all its organs except for only one case of liver TAG. The mean DHA contents of the various organs accounted for more than 20% of the total FA (TFA), even though it is a neutral depot lipid. However, DHA in the stomach contents, originating from their prey, fluctuated and was generally low. DHA levels were generally higher in a year (2000) when water temperatures were colder than in one when it was warmer (1998). Furthermore, DHA levels in muscle TAG were consistently high in spite of the fluctuation of those in the visceral TAG, which might be directly influenced by the prey lipids. This phenomenon suggests the physiological selective accumulation of DHA in the muscle, after the migration of the digested FA in the vascular system and absorption of the prey lipids in the intestine. In contrast, the FA composition of other species is generally variable and their DHA contents of TAG are usually less than 20% of TFA.  相似文献

The mdx mouse is a model for Duchenne muscular dystrophy (DMD), a debilitating disease affecting striated muscle. It is established that the fatty acid (FA) composition of skeletal muscle phospholipid (PL) is altered in mdx mice, but it is not known if cardiac muscle is similarly affected by dystrophin-deficiency. We tested FA profiles in PL and triacylglycerol (TAG) in cardiac muscle of 12-week old mdx and control (con) mice. Of 22 different FA, similar to our previous finding for skeletal muscle, the most abundant FA in heart PL were palmitic, stearic, cis-vaccenic, linoleic, and docosahexaenoic acid, while for TAG the most abundant FA were palmitic, oleic, cis-vaccenic, and linoleic acid. In comparing mdx and con, no significant group differences were detected for any FA in PL or TAG. Thus, unlike skeletal muscle, FA composition in cardiac muscle PL is not different between mdx and con at the age studied. The results can be understood in the context of tissue-specific disease severity in mdx mice, as pathology is quite modest in cardiac compared with skeletal muscle.  相似文献

The blubber, liver, and muscle of the Antarctic sei whale were analyzed for total lipid content, composition of lipid by classes and positional distribution of fatty acids in individual lipids. The major glycerolipids (triglycerides, phosphatidylcholine, and phosphatidylethanolamine) were fractionated by silver nitrate thin layer chromatography. The phospholipid fractions were analyzed for fatty acid positional distribution. The whale stomach contained almost exclusively the amphipodParathemisto gaudichaudi. Its lipids were also studied and compared with the lipids of the body tissues. The results indicate that the stomach content lipids are subjected to modifications before being deposited in the blubber, liver, and muscle. According to the silver nitrate thin layer chromatographic studies, liver and blubber triglycerides resemble each other in their patterns of positional distribution of fatty acids and in molecular species composition. The phospholipids of liver and blubber also exhibited closely related fatty acid distribution patterns. In general, while the proportions of lipid classes and their predominant fatty acids varied from tissue to tissue, the patterns according to which the lipids had been synthesized seemed to be common. “...And beneath the effulgent Antarctic skies I have boarded the Argo-Navis, and joined the chase against the starry Cetus far beyond the utmost stretch of Hydrus and the Flying Fish.” (1).  相似文献

Lipid peroxidation was induced in freshly isolated rat hepatocytes by incubation in the presence of Fe3+, resulting in accumulation of thiobarbituric acid reactive substances. Analysis of lipid classes revealed that the levels and fatty acid compositions of the two major phospholipids, phosphatidylcholine (PC) and phosphatidylethanolamine (PE), remained unchanged but the levels of triacylglycerols (TAG) were significantly reduced, and some of their polyunsaturated fatty acids were selectively lost as the result of oxidant treatment. Acyl turnover in PC and PE as determined by 18O incorporation from H2 18O-containing media remained largely unchanged during oxidant treatment, while some increased turnover of the saturated fatty acids in TAG was observed. We hypothesize that constitutive recycling of membrane phospholipids rather than selective in situ repair eliminates peroxidized species of PC and PE, TAG could serve as an expendable fatty acid reserve, providing a limited but very dynamic pool of polyunsaturated fatty acids for the resynthesis of phospholipids.  相似文献

Hyperphagia was achieved by continuous intracerebroventricular infusion of a melanocortin receptor antagonist (HS024; Neosystem, Strasbourg, France) in rats. The effects of hyperphagia on FA composition and concentration of plasma phospholipids (PL), plasma FFA, and adipose tissue TAG were studied in rats for 8 d [short-term hyperphagia (STH); n=8], or 28 d [longterm hyperphagia (LTH); n=9]. The control rats were treated with artificial cerebrospinal fluid for 8 d (n=8) or 28 d (n=10). The rats were fed the same regular diet. In STH rats the plasma PL and fasting plasma FFA contained higher concentrations of saturated FA (SFA) and monounsaturated FA (MUFA), and plasma FFA contained lower n−6 PUFA than in the control rats. In LTH rats the plasma PL contained higher concentrations of SFA, MUFA, and n−3 PUFA and higher proportions of 16∶1n−7 and 18∶1n−9 at the expense of 18∶2n−6 than in the control rats. In LTH rats the abundant dietary intake of 18∶2n−6 did not enrich 18∶2n−6 of the plasma PL or adipose tissue TAG. In LTH rats the fasting plasma FFA contained more than twofold higher concentrations of SFA and MUFA, and higher proportions of 16∶1n−7 and 18∶1n−9 at the expense of 18∶2n−6 than in the control rats. This animal obesity model shows that LTH affects the FA composition and concentration of plasma PL, plasma FFA, and adipose tissue TAG, a result consistent with changes associated with increased risk of various diseases in humans. These results also demonstrate that LTH alters the FA composition of plasma PL and adipose tissue TAG in a way that does not reflect the FA composition of dietary fat.  相似文献

This study was undertaken to investigate the effects of long term betaine intake on the cholesterol and lipid profile of Alentejano (AL) pigs. At ?36 kg body weight (BW), castrated male and female pigs fed a commercial (C) diet, were divided into two groups: i) Group C, consuming the C diet; and ii) Group CB, consuming the C diet supplemented with 1 g/kg betaine. Pigs were slaughtered at ?100 kg BW. Fasting plasma concentrations of protein, urea, glucose, TAG, phospholipids, homocysteine, total and LDL‐ and HDL‐cholesterol were determined. Liver TAG, phospholipids, and total and free cholesterol were analyzed, as well as total lipids, cholesterol contents, and fatty acid (FA) composition of M. semimembranosus and dorsal subcutaneous fat. Betaine supplemented pigs presented significantly higher plasma concentrations of TAG, phospholipids, cholesterol, and lipoprotein cholesterol. Dorsal subcutaneous fat cholesterol concentration was also significantly higher in CB than in C pigs. No differences were detected in the most abundant FA profile (including the unsaturated to saturated FA ratio) of muscle and subcutaneous fat tissues among treatments. These data suggest that betaine induces dyslipidemia, and increases cholesterol concentration in dorsal subcutaneous fat, without affecting the FA profile of M. semimembranosus and dorsal subcutaneous fat.  相似文献

The sea urchin Diadema setosum is edible and desirable as food by locals in central Vietnam and a promising target for potential fishing. The lipid profiles of the gonads of the sea urchin inhabiting the coastal area in Nha Trang Bay are studied for the first time. The determination of the content of the total lipids (TL), total phospholipids (PL), monoacylglycerols (MAG), diacylglycerols (DAG), triacylglycerols (TAG), cholesterol (Chol), sterol esters, and free fatty acids (FFA) is analyzed by high-performance thin layer chromatography (HPTLC); the phosphatidylserine (PS), phosphatidylethanolamine (PE), phosphatidylinositol (PI), phosphatidylcholine (PC), lysophosphatidylcholine (LPC), and sphingomyelin (SM) are determined by high-pressure liquid chromatography (HPLC); and the fatty acid levels of TLs are identified using gas chromatography (GC). Non-polar TAG, FFA, Chol, and DAG dominated. The content of total PL is significant. PC is abundant among PL fractions, followed by PE, PI, and PS. The TLs contain a high proportion of PUFA, mainly due to arachidonic fatty acid and eicosapentanoic fatty acid. The (n−6)/(n−3) ratio is 1.68, and the atherogenic and thrombogenic indexes of D. setosum are 1.13 and 0.73. The results can be used in the development of marine bioprospecting and methodological approaches for the creation of functional substances.  相似文献

To ensure complete lipid extraction of algal samples collected on glass fiber filters, one must facilitate the access of extracting solvent to the lipids by using ultrasonication, grinding, or a combination of these two methods. This study examines the effect of these three treatments, in combination with storage time and sampling volumes, on the determination of lipid class composition of the diatom Chaetoceros gracilis. The TAG level was significantly influenced by treatment in relation to either storage time or sampling volume. FFA and other degradation products increased markedly with storage time at the expense of TAG and phospholipids (PL). Finally, sampling volume and storage time interacted in their effects on TAG and PL contents in an inverse manner.  相似文献

Compared with triacylglycerol (TAG), dietary 1,3‐diacylglycerol (1,3‐DAG) is associated with reduced serum lipid and glucose levels. We investigated the metabolism of 1,3‐DAG by assaying its intermediate metabolites during digestion and absorption in the rat small intestine. After gavage with TAG emulsion, TAG was digested mainly to 2‐monoacylglycerol (2‐MAG) and unesterified fatty acid (FFA) in the rat small intestinal lumen. 2‐MAG was directly absorbed into the small intestinal epithelial cells and esterified to 1,2(2,3)‐DAG, and further esterified to TAG. After gavage with 1,3‐DAG emulsion, 1,3‐DAG was digested mainly to 1(3)‐MAG and FFA in the rat small intestinal lumen with subsequent significant increase of 1‐MAG and 1,3‐DAG concentrations in small intestinal mucosal epithelial cells, and the 2‐MAG, 1,2(2,3)‐DAG, and TAG concentrations in mucosal epithelial cells were not significantly different after 1,3‐DAG than after TAG gavage, suggesting that the metabolic pathway of 1,3‐DAG is different from that of TAG. In intestinal mucosal epithelial cells, we further assayed enzyme levels and gene expression of proteins in the phosphatidic acid (PtdOH) pathway. The glycerol kinase, phosphatidate phosphatase, and diacylglycerol acyltransferase‐2 expression and the relative expression of mRNA of enzymes were significantly increased in the 1,3‐DAG group compared with the TAG group, suggesting that TAG synthesis from dietary 1,3‐DAG was mainly via PtdOH pathways, which may partially account for the effect of dietary DAG on postprandial serum TAG.  相似文献

Seed oils from four legume cultivars of Vicia faba, grown in Japan, were extracted and classified by thin-layer chromatography (TLC) into eight fractions. The major lipid components were triacylglycerols (TAG: 48.8–50.1%) and phospholipids (PL: 47.5–50.5%), while hydrocarbons (HC), steryl esters (SE), free fatty acids (FFA), diacylglycerols (1,3- and 1,2-DAG) and monoacylglycerols (MAG) were present in minor proportions (1.8–2.4%). All lipid samples had high amounts of total unsaturated FA, representing 79.7–82.8% and 77.6–79.7% for TAG and PL, respectively. Molecular species and FA distributions of TAG, isolated from the total lipids in the broad beans, were analyzed by a combination of argentation-TLC and GC. Fourteen different molecular species were detected. With a few exceptions, the main TAG components were S2D (6.1–8.9%), SD2 (7.8–10.5%), SMT (6.3–8.5%), M2D (4.5–6.2%), MD2 (18.9–21.8%), D3 (21.0–23.9%) and MDT (8.1–10.2%) (where S, M, D, and T denote a saturated fatty acid, a monoene, a diene, and a triene, respectively). These results suggest that the lipid classes, FA distributions and TAG molecular species of broad beans are not dependent on the cultivation areas during the growing season.  相似文献

Circulating fatty acids (FA) derived largely from dairy consumption have most commonly been measured in total human serum or phospholipid (PL) fractions, and have been used as validated biomarkers of dairy intake in a growing number of epidemiological studies. Nevertheless, measurement and characterization of a wider spectrum of FA biomarkers of dairy across the four major serum lipid fractions is lacking. This study aimed to (1) quantify FA biomarkers of dairy in PL, triacylglycerol (TAG), cholesteryl ester (CE), and unesterified fatty acid (FFA) serum lipid fractions; and (2) identify potential demographic and metabolic factors that may modify the proportions of these FA across serum fractions. Baseline data from 444 adults in the PROMISE cohort were analyzed. FA biomarkers, 15:0, t16:1n-7, 18:2-c9,t11, and t18:1n-7 were quantified from serum. Dairy intake was estimated using the validated Canadian Diet History Questionnaire. Our results show that t18:1n-7 was the most abundant FA biomarker in all fractions except CE, where 18:2-c9,t11 was the most abundant. Positive correlations within fractions, and across FA in the PL, CE, and FFA fractions were found, however, TAG FA were negatively correlated with the other fractions. PL and CE FA were positively associated with dairy intake, and negatively associated with markers of dysmetabolism while, in contrast, these markers were predictors of higher TAG dairy FA. This study is the first to demonstrate distinct proportions of dairy FA in different serum lipid fractions. PL and CE FA marked dairy intake in this cohort, while TAG FA appeared to be markers of dysmetabolism.  相似文献

In this paper, we report the full-length coding sequence of bovine ATGL cDNA and analyze its expression in bovine tissues. Similar to human, mouse, and pig ATGL sequences, bovine ATGL has a highly conserved patatin domain that is necessary for lipolytic function in mice and humans. This suggests that ATGL is functionally intact as a triglyceride lipase in cattle. Tissue distribution of ATGL gene expression was highest in fat and muscle (skeletal and cardiac) tissue, while protein expression was solely detectible in the adipose tissue. The effect of 109 days of flaxseed supplementation on ATGL and adipocyte fatty acid-binding protein (FABP4 or A-FABP, E-FABP or FABP5) expression was examined in Angus steers. Supplemented steers had greater triacylglycerol (TAG) content in the muscle compared with unsupplemented ones. Additionally, supplementation increased A-FABP expression and decreased stearoyl-CoA desaturase 1 (SCD-1) expression in muscle, while total ATGL expression was unaffected. In summary, supplementation of cattle rations with flaxseed increased muscle TAG concentrations attributed in part to increased expression of key enzymes involved in lipid trafficking (A-FABP) and metabolism (SCD-1).  相似文献

Pigs were fed a commercial conjugated linoleic acid (CLA) mixture, prepared by alkali isomerization of sunflower oil, at 2% of the basal diet, from 61.5 to 106 kg live weight, and were compared to pigs fed the same basal diet with 2% added sunflower oil. The total lipids from liver, heart, inner back fat, and omental fat of pigs fed the CLA diet were analyzed for the incorporation of CLA isomers into all the tissue lipid classes. A total of 10 lipid classes were isolated by three-directional thin-layer chromatography and analyzed by gas chromatography (GC) on long capillary columns and by silver-ion high-performance liquid chromatography (Ag+-HPLC); cholesterol was determined spectrophotometrically. Only trace amounts (<0.1%; by GC) of the 9,11–18∶2 cis/trans and trans, trans isomers were observed in pigs fed the control diet. Ten and twelve CLA isomers in the diet and in pig tissue lipids were sepatated by GC and Ag+-HPLC, respectively. The relative concentration of all the CLA isomers in the different lipid classes ranged from 1 to 6% of the total fatty acids. The four major cis/trans isomers (18.9% 11 cis, 13 trans-18∶2; 26.3% 10 trans, 12 cis-18∶2; 20.4% 9 cis, 11 trans-18∶2; and 16.1% 8 trans, 10 cis-18∶2) constituted 82% of the total CLA isomers in the dietary CLA mixture, and smaller amounts of the corresponding cis,cis (7.4%) and trans,trans (10.1%) isomers were present. The distribution of CLA isomers in inner back fat and in omental fat of the pigs was similar to that found in the diet. The liver triacylglycerols (TAG), free fatty acids (FFA), and cholesteryl esters showed a similar patterns to that found in the diet. The major liver phospholipids showed a marked increase of 9 cis,11 trans-18∶2, ranging from 36 to 54%, compared to that present in the diet. However, liver diphosphatidylglycerol (DPG) showed a high incorporation of the 11 cis,13 trans-18∶2 isomer (43%). All heart lipid classes, except TAG, showed a high content of 11 cis,13 trans-18∶2, which was in marked contrast to results in the liver. The relative proportion of 11 cis,13 trans-18∶2 ranged from 30% in the FFA to 77% in DPG. The second major isomer in all heart lipids was 9 cis,11 trans-18∶2. In both liver and heart lipids the relative proportions of both 10 trans,12 cis-18∶2 and 8 trans,10 cis-18∶2 were significantly lower compared to that found in the diet. The FFA in liver and heart showed the highest content of trans,trans isomers (31 to 36%) among all the lipid classes. The preferential accumulation of the 11 cis,13 trans-18∶2 into cardiac lipids, and in particular the major phospholipid in the inner mitochondrial membrane, DPG, in both heart and liver, appears unique and may be of concern. The levels of 11 cis,13 trans-18∶2 naturally found in foods have not been established.  相似文献

The changes in composition of total and neutral lipids (NL) as well as glycolipids (GL) and phospholipids (PL) of borage (Borago officinalis L.) seeds, germinated in the dark at 25°C for 10 d, were studied. Total lipids constituted 34.0% of the dry matter of borage seeds. During germination, the content of total lipids was decreased by 95%. NL accounted for 95.7% of total lipids prior to germination and were composed of triacylglycerols (TAG; 99.1%), diacylglycerols (DAG; 0.06%), monoacylglycerols (MAG;0.02%), free fatty acids (FFA;0.91%), and sterols (0.02%). The content of TAG was significantly (P≤0.05) decreased, while that of other components, such as MAG and FFA, significantly (P≤0.05) increased during germination. However, the content of DAG did not change. GL and PL accounted for 2.0 and 2.3% of total lipids, respectively, and their contents significantly (P≤0.05) increased as germination proceeded. The thin layer chromatography-flame-ionization detection studies showed that phosphatidylcholine (PC; 69.7%) was the major PL present. The total content of phosphatidylserine (PS) and phosphatidylethanolamine (PE), which were coeluted, was 18.2%; phosphatidic acid (PA) was present at 11.2% of the total PL fraction. Lysophosphatidylcholine was detected at 0.9%. The proportion of PC, PS, and PE significantly (P≤0.05) decreased during germination, but that of PA increased (P≤0.05) markedly. The fatty acid composition of lipid fractions changed as germination proceeded. The predominant fatty acids of total lipids, NL, and GL were linoleic and linolenic acids, while those of PL were linoleic and palmitic acids. The present study demonstrated that the overall changes of lipids seen in borage seeds during germination agree well with results for other oilseeds. Changes in lipid compositions during germination result from the formation of tissues and metabolic interconversion of lipid classes. Rapid changes in lipid composition during seed germination may enhance the nutritional value of the sprouts.  相似文献

To assess the suitability of microalgal strains for biodiesel production the lipid content and composition, especially individual triacylglycerols (TAG) and free fatty acids (FFA) must be determined. In this study, the compositions and concentrations of TAG and FFA were analysed in four halophytic algal species, Dunaliella salina, D. tertiolecta, D. bardawil, and D. granulata. These species were selected as part of a larger screen to identify species suitable for biofuel feedstocks. An accelerated solvent extraction instrument was used for lipids and fatty acid extraction using a dichloromethane–hexane solvent system. Ultra-performance liquid chromatography coupled with mass spectrometry (MS) detection was optimized and applied to the quantitative analysis of TAG and FFA in the different algal extracts. Individual TAG were characterized structurally using direct electrospray ionization (ESI) MS and MS/MS techniques. Cationic adducts (NH4 +) of TAG were detected and quantified in the positive ESI MS and MS/MS modes, while the negative ESI mode was used for FFA analysis. Over 20 TAG were identified and quantified in the four Dunaliella strains. Analysis of FFA compositions demonstrated that the most abundant FFA in these four algal species were palmitic, linolenic, linoleic, and oleic acids.  相似文献
