Effect of tuberculosis on milk production in dairy cows   总被引:1,自引:0,他引:1  
A survey of Isospora suis performed in 177 faecal samples from 30 swine farms detected thin wall type I. suis oocysts in seven samples. This type of oocyst measuring 23.9 by 20.7 microns had a retracted thin wall similar to that of the genus Sarcocystis. This type of oocysts, isolated from four different faecal samples, was inoculated in four-five-days-old piglets free of contamination in order to verify the life cycle and pathogenicity of the species. The pigs were kept in individual metal cages and fed with cow milk. Daily faecal collections and examinations were performed until the 21st day after infection. MacMaster and Sheather's methods were used for oocyst counting and identification. Infected piglets produced yellowish-pasty diarrhoea with slight dehydration. The prepatent and patent periods were respectively from 6 to 9 and 3 to 10 days after infection. Oocyst elimination was interrupted on the 10th and 11th days after infection with biphasic cycles. Thin and thick wall oocysts were detected in the same faecal samples. Thin walls were not observed in unsporulated oocytes. The observations suggest that this type of oocysts could appear in specific strains which occur in the later stages of their development. These oocysts seem to be responsible for clinical and pathogenic signs of neonatal isosporosis in pigs.  相似文献

A randomized clinical trial including 1109 cows from 12 Australian dairy herds was used to evaluate the effects of monensin on the health (n = > 686 cows), production (n = 915 cows), and reproduction (n = > 908 cows) of dairy cows. Cows were allocated to a treatment group receiving a slow-release intraruminal bolus containing 32 g of sodium monensin that was administered 40 d before and 50 d following the anticipated calving date or to a control group. Treatment did not significantly alter any reproductive outcome; 54.5% of cows treated with monensin and 58.2% of control cows were pregnant at first service, and days to conception were lower for cows treated with monensin. The hazard rate (0.95) was not significant for these cows. The percentage of cows pregnant was 83.8 for control cows, and days to first estrus (hazard rate = 1.04) and first service (hazard rate = 1.04) were not significantly higher for treated cows. Treatment with monensin did not significantly alter the risk of any disease. The incidence of retained fetal membranes, pyometra, lameness, abortion, and infectious diseases was not significantly lower for cows in the treatment group, and the incidence of mastitis was not significantly higher for cows in the treatment group. Monensin significantly increased milk production by 0.75 L/d per cow and tended to increase milk fat and protein yields but had no significant effect on milk fat or milk protein percentages. Changes in the production of milk and milk constituents were consistent throughout lactation.  相似文献

Adequacy of amino acids in diets fed to lactating dairy cows   总被引:1,自引:0,他引:1  
The Cornell Net Carbohydrate and Protein System was used to evaluate absorbable limiting amino acids (AA) for milk yield. The system was utilized to characterize whether diets in five previous experiments met AA requirements for milk protein synthesis. Twenty-nine treatment means for milk yield from 367 cows constituted the database for the evaluation. Using the mechanistic relationships of nutrient metabolism described in the Cornell system, absorbed amounts were predicted of each essential AA from a diet and milk yields allowed by the most limiting AA. Regression of observed milk yield (Y) on predicted milk yield (X) using all treatment means (n = 29) was Y = 2.3 + 0.799X (r2 = 0.72). The linear relationship was stronger using 6 treatment means (n = 18) when protein supplements were fed rather than when ruminally protected AA were fed or when AA were postruminally infused (Y = -1.8 + 0.983X; r2 = 0.93). The Cornell system predicted that, for diets based on corn, Met or Lys was limiting when soybean meal was the protein source, but Lys was limiting when corn gluten meal or brewers grains were the source of protein. By AA limitation, the Cornell system explained differences in milk yield for diets that differed in supplemental protein sources in some of the experiments. As determined from milk protein yields in these studies, requirements for individual essential AA were expressed as a percentage of dietary dry matter or total essential AA, and values were relatively constant among dietary treatments and experiments.  相似文献

Although the effect of hyperoxia on antioxidant enzymes is well known, the effect of subtoxic levels of hyperoxia on gamma-glutamyltransferase (gamma-GT), involved in the degradation and uptake of extracellular GSH for intracellular GSH synthesis, is unknown. The aim of the study was to investigate (1) the effects of in vitro hyperoxia on gamma-GT activity of type II cells and (2) the effects of the lazaroid U-74389G and N-acetylcysteine (NAC) on the hyperoxia-induced changes in gamma-GT and antioxidant enzyme activities. At 48 h after isolation, rat type II cells were exposed for 2 days to air, 60% O2 or 85% O2 with or without 30 microM U-74389G or 100 microM NAC. After the exposure, the cells were harvested and assayed for superoxide dismutase (SOD), glutathione peroxidase (GPx), gamma-GT activity, and GSH levels. In another series of experiments 85% O2-exposed cells, with or without U-74389G, were used for Northern blotting of gamma-GT mRNA. Exposure to 60% O2 decreased gamma-GT and GSH by -47 and -34%, respectively, while SOD and GPx activities remained unchanged. After 85% O2-exposure gamma-GT decreased by -55%, SOD and GPx increased by +55 and +87%, respectively, while GSH decreased by -35%. NAC treatment decreased gamma-GT activity by -42% in the air-exposed cells. After 60% O2, U-74389G led to significantly higher gamma-GT (+117%) and GSH (+26%) while NAC only led to higher GSH (+28%) compared to the oxygen-exposed cells not treated with NAC or U-74389G. After 85% O2 U-74389G increased gamma-GT, SOD, and GSH by +72, +58, and +68%, respectively, while NAC only increased SOD (+49%) and GSH (+26%) compared to the oxygen-exposed cells not treated with NAC or U-74389G. The 85% O2 exposure, with or without U-74389G, had no effect on gamma-GT mRNA levels. The results show that hyperoxia decreases rat type II cell gamma-GT activity in vitro. This effect was not related to an altered regulation at mRNA level and it was not associated with the hyperoxia-induced decrease in intracellular GSH, since restoration of the GSH levels by NAC did not restore gamma-GT activity. The lazaroid U-74389G with vitamin E-like properties effectively prevented the decrease in gamma-GT and GSH, so that direct inactivation of the membrane-bound gamma-GT by hyperoxia is the most likely mechanism.  相似文献

OBJECTIVE: To determine whether cows seropositive to Neospora caninum produced less milk during their first lactation than seronegative cows. DESIGN: Repeated-measures, prospective study. ANIMALS: 372 Holstein cows in their first lactation. PROCEDURE: Cows were tested repeatedly before and during their first lactation for antibodies to N caninum. One-way and repeated-measures ANOVA were used to determine whether mean daily milk weights and milk weights from Dairy Herd Improvement Association testing were less for seropositive cows than for seronegative cows. RESULTS: Weekly mean daily milk weights for the 118 seropositive cows were significantly less than those for the 254 seronegative cows, and milk production for seropositive cows (mean, 55.2 lb/cow/d) was 2.5 lb/cow/d less than that for seronegative cows (mean, 57.7 lb/cow/d). Analysis of results from Dairy Herd Improvement Association testing revealed that production of seropositive cows was less for milk (3.1 lb/cow/d), fat-corrected milk (3.6 lb/cow/d), and fat (0.14 lb/cow/d) than production of seronegative cows. CLINICAL IMPLICATIONS: The economic impact of N caninum infection in dairy cows can include reduced revenues from decreased milk production, which may warrant culling of young, seropositive replacement stock.  相似文献

This experiment examined the effect of feed quality on the relationship between intake and stage of lactation in dairy cows. Two total mixed diets composed of grass silage and concentrate were formulated. The high concentrate total mixed diet was designed to meet energy requirements, and the low concentrate total mixed diet was designed to limit intake. Twenty-four Holstein-Friesian cows were offered the total mixed diets in a full 2 x 2 change-over design with control treatments. The changeover was at 153 d in milk (DIM). For the statistical analyses, two periods of 13 wk, one period before and one period after the changeover, were used. Dry matter intake (DMI), milk yield, body weight, and body condition score were significantly greater for cows fed the high concentrate total mixed diet than for cows fed the low concentrate total mixed diet. Significant interactions between total mixed diet and period were observed for DMI and milk yield. However, no significant residual effects of changing from one total mixed diet to the other were observed. The interactions were due to substantially different slopes of DMI and milk yield relative to DIM for cows fed the two different total mixed diets. For cows fed the low concentrate total mixed diet, there was no effect of stage of lactation on DMI; the slope was 0. For cows fed the high concentrate total mixed diet, there was a significant decline in DMI as lactation progressed.  相似文献

Four cannulated cows in a 4 x 4 Latin square design were used to study the effects of supplemental dietary fatty acids from roasted soybeans combined with tallow or partially hydrogenated fats, varying in esterification and fatty acid chain length, on nutrient digestion and lactation performance. Diets were formulated to contain (DM basis) 48% forage in addition to 1.5% fatty acids from roasted soybeans and 2.5% fatty acids from tallow, partially hydrogenated tallow triglycerides, partially hydrogenated tallow fatty acids, or a blend of 30% tallow and 70% hydrogenated fatty acids that were rich in palmitic acid. Apparent total tract digestibilities of OM, N, and NDF were similar among diets. Supplemental fat as fatty acids, compared with triglycerides, increased digestibilities of total fatty acids and C18:1 in the small intestine, perhaps indicating that lipolysis was rate-limiting. Fatty acids also increased milk fat percentage and efficiency of 4% FCM production. Although intake of C16 fatty acids was higher for cows fed the fat rich in palmitic acid than for those fed the tallow fatty acids, total duodenal flows of C16 fatty acids were similar, and digestibility in the small intestine was unaffected. Dry matter intake, 4% FCM production, and milk protein percentage were similar among treatments.  相似文献

The objective of the study was to determine the effects of dietary fat source on duodenal flow, apparent absorption, and milk fat incorporation of trans-C18:1 fatty acids. Four ruminally and duodenally cannulated multiparous Holsteins cows in mid to late lactation were fed a basal diet containing 36% corn silage, 24% alfalfa haylage, and 40% concentrate (dry matter basis). Diets contained 0% supplemental fat (control diet), 3.7% high oleic sunflower oil, 3.7% high linoleic sunflower oil, or 3.7% partially hydrogenated vegetable shortening; treatments were administered in a 4 x 4 Latin square design with 3-wk experimental periods. The flow of trans-C18:1 to the duodenum was higher for cows fed diets supplemented with fat than for cows fed the control diet (283 vs. 64 g/d). Incomplete biohydrogenation accounted for the increased flow of trans-C18:1 to the duodenum in cows fed diets containing high oleic and high linoleic sunflower oil. Increased flow of trans-C18:1 in cows fed the diet containing partially hydrogenated vegetable shortening most likely originated from the trans-C18:1 in the diet. Milk fat percentages were 3.48, 3.07, 3.18, and 3.38% for cows fed the control diet and diets containing high oleic sunflower oil, high linoleic sunflower oil, and vegetable shortening, respectively. Milk trans-C18:1 increased from 2.9 to 11.2% of the total fatty acids for cows fed the control diet and the diets supplemented with fat, respectively. Milk trans-C18:1 were equal across all diets supplemented with fat. Ruminal and total tract digestion of organic matter, neutral detergent fiber, and N did not change with fat supplementation. Results showed that trans-C18:1 is increased in the milk of cows with reduced milk fat; however, excessive amounts of trans-C18:1 in milk do not necessarily correspond directly to milk fat depression.  相似文献

A low level of energy (110 MJ ME) and a low concentrate/forage ratio (10/90) at calving resulted in low basal concentrations of glucose and insulin, but the cows had the capacity to increase the glucose level after glucagon injections. No signs of disturbances in the metabolic adaptation were observed. High intensity feeding (200 MJ ME and 50% concentrates) resulted in high basal serum insulin levels. The increase in the insulin concentrations after glucagon injections and the changes in insulin levels around calving varied widely between individual cows. The metabolic adaptation period was longer than in cows in the former group. An energy level of 170 MJ ME and variations of concentrate/forage ratios (5/95, 30/70 and 60/40) resulted in small differences in basal glucose and insulin concentrations and in response to glucagon injections. But the cows fed 60% concentrates showed signs of prolonged metabolic adaptation. Increased lipid concentrations in diets containing equal levels of energy and protein resulted in a fall in basal glucose and insulin levels and the metabolism seemed to be directed towards catabolism. Because of these metabolic effects, more needs to be known on fat supplementation if it is used in practical feeding. It is also necessary to take more interest in the effects of protein feeding on the periparturient metabolism.  相似文献

Individual milk production curves of 542 cows with 722 cases of mastitis were compared with control curves drawn from healthy lactating cows. First, differences were classified into patterns of milk loss, and their distribution was analyzed among the patterns with regard to breed, season, lactation number, stage of lactation, and milk production. Then, individual milk losses were estimated and analyzed according to the same factors. In early lactation, almost 7% of mastitis cases necessitated culling or drying off. For 36% of the cases of mastitis occurring in early lactation, milk production was affected for an extended period, and the milk loss induced was 911 kg on average over the entire lactation. Fifty-two percent of the mastitis cases occurring after lactation peak was not accompanied by marked modifications of the lactation curves. For 38% of cases of mastitis occurring from mid to late lactation, milk production was affected for an extended period. When milk production was the most affected (26% of total cases), the average loss was 850 kg over the entire lactation. For cases in early or mid to late lactation, the production at mastitis onset was a determining factor of the amount and pattern of milk production loss induced. Results were applied in a simulation over two simulated groups of cows.  相似文献

Lactation records (n = 2362) of 1402 crossbred cows in 22 cooperating dairy herds in southeastern Brazil were evaluated. Cows were mixtures of Zebu (Gir, Guzera, and unknown) and European breeding (mostly Holstein). Lactation milk yields were expressed as total, 3050-d, or deviated 305-d yields, either adjusted or unadjusted for days in milk (DIM). Mean DIM was 280. Arithmetic means unadjusted for DIM were 1942, 1666, and 5 kg per record. Milk yields of daughters from sires of 6/8 and 7/8 European breeds were higher than yields of daughters from sires of 5/8 European breeds when data were either adjusted or unadjusted for DIM. The differences associated with breed group of sire were only slightly reduced when records were adjusted for DIM. There was no evidence of a decline in milk yield as the fraction of European breeding of the sire increased from 6/8 to 7/8. For a given breed group of sire, whether the grandsire was purebred or crossbred had no detectable effect. These results should be useful in determining strategies for crossbreeding of dairy cows in tropical areas, particularly when crossbred sires are used.  相似文献

Lactating dairy cows were offered diets containing increasing quantities of Tifton 85 bermudagrass hay or silage and increasing dietary neutral detergent fiber (NDF) to determine effects of method of bermudagrass storage and dietary fiber content on intake, milk yield, and nutrient digestion. Bermudagrass was added as hay or silage at the rate of 8.5, 15.9, or 23.3% of dietary dry matter and was substituted for an equal quantity of corn silage. The method of bermudagrass storage did not affect intake [20.8 vs. 20.3 (+/- 0.3) kg/d for bermudagrass hay and silage diets; respectively], but an increase in the amount of bermudagrass and NDF in the diet did reduce intake. Dietary NDF concentration was well above minimum recommendations of the National Research Council. Milk yield was not altered by method of bermudagrass storage but declined as dietary NDF increased. Digestion of NDF tended to be lower for bermudagrass silage than for bermudagrass hay. An increase in the amount of bermudagrass in the diet improved digestion of dietary dry matter, acid detergent fiber, and NDF; however, milk yield declined because total intake declined. Improved NDF digestion caused by added bermudagrass hay or silage was confirmed by in vitro analyses. High quality bermudagrass hybrids could potentially be used in diets of lactating dairy cows because of the good digestion characteristics and relatively small effects on intake despite the high dietary fiber content. Tifton 85 bermudagrass may serve as a forage alternative for lactating dairy cows.  相似文献

Forty-four multiparous and 43 primiparous Holstein cows were used to study the effect of dietary alfalfa silage: grain ratio on digestion, passage of digesta, and energy balance in a complete lactation experiment. Cows were placed on one of five treatments with forage contents from 38.2 to 98.2% (DM basis) during the first 12 wk of lactation. Forage content was increased during wk 13 to 26 of lactation to give diets containing 48.2 to 98.2% forage and again during wk 27 to 44 to give diets containing 68.2 to 98.2% forage. Both DMI and DM digestibilities decreased as proportion of dietary forage increased. The DM digestibilities for multiparous and primiparous cows ranged between 69.3 and 57.3% during early lactation and 64.9 and 55.5% during late lactation. Increased percentage of forage in the diet decreased and then increased (quadratic relationship) the ruminal retention time of La, a marker applied to the alfalfa silage in early and late lactation. Time cows spent eating and ruminating per kilogram of DMI increased as proportion of forage in the diet increased. Cows fed diets with a high proportion of alfalfa silage remained in negative energy balance longer than cows fed high grain diets. Intake of NEL (calculated by either of two methods) minus NEL output (milk, maintenance, and BW change) resulted in net balances of NEL after 36 wk of lactation within 5% of NEL intake and indicated that estimates of the NEL value of feedstuffs used in this experiment were reasonably accurate.  相似文献

Five primiparous Holstein cows (55 d in milk) that were fitted with ruminal and duodenal cannulas were used in a 4 x 5 incomplete Latin square to determine the effects of blends of steam-flaked and dry-rolled corn on site and extent of nutrient digestion and milk yield and composition. Diets were fed as total mixed rations and consisted of 45% forage and 55% concentrate; each diet contained 27% corn grain. Dietary treatments were composed of blends of dry-rolled and steam-flaked corn in ratios of 100:0, 67:33, 33:67, and 0:100. Intake of dry matter; digestibilities of dry matter, organic matter, acid detergent fiber, cellulose, neutral detergent fiber, fatty acids, and N; and microbial efficiency were unaffected by diet. Ruminal, postruminal, and total tract digestion of starch increased linearly, and starch passage to the duodenum decreased linearly, as the proportion of dry-rolled corn in the diet decreased. Ruminal propionate and valerate increased linearly, and acetate, butyrate, isovalerate, and the acetate to propionate ratio decreased linearly, as proportions of dry-rolled corn in the diet decreased; however, no changes in total volatile fatty acid concentrations in ruminal fluid were observed. Ruminal fluid pH was similar across diets. A decrease in dry-rolled corn decreased ruminal ammonia N and plasma urea N linearly. Milk yield and composition, as well as milk N fractions, were similar across diets. Although changes in fatty acid composition of milk fat were small, linear decreases in percentages of trans-C16:1 and cis-9- and cis-10-C18:1, as well as a linear increase in the percentage of C18:2 occurred as the proportion of dry-rolled corn in the diet decreased. An increased proportion of dry-rolled corn in the diet decreased digestion of starch in the rumen, and patterns of volatile fatty acid concentrations shifted accordingly. However, no effects on lactational parameters were observed.  相似文献

Interactions of dietary energy and fat with recombinant bST (sometribove) injections were tested for their effects on ovarian function. Lactating dairy cows were fed one of three diets differing in energy concentration (NEL) and percentage of DM of calcium salts of long-chain fatty acids: diet 1, 1.68 Mcal/kg and 0%; diet 2, 1.68 Mcal/kg and 2.2%; and diet 3, 1.78 Mcal/kg and 2.2%. Cows were injected daily with bST or saline during one of two 19-d interestrous periods (crossover design) in which ovarian follicles were measured by ultrasonography. The bST-treated cows produced more FCM and were in lower energy balance than saline-treated cows. Before d 12 (first follicular wave, estrus = d 0), bST-treated cows had more ovarian follicles in classes 1 (3 to 5 mm; cows on diet 3) or 2 (6 to 9 mm; cows on diets 1 and 2) than saline-treated cows. After d 12 (preovulatory follicular wave), numbers of follicles in different size classes were similar for bST-treated and saline-treated cows, but cows fed diet 2 had larger preovulatory follicles. Lower dietary energy and bST treatment were both associated with larger subordinate follicles. Ovarian follicles less than 10 mm were stimulated by bST, and calcium salts of long-chain fatty acids increased diameters of preovulatory follicles.  相似文献

The effects of monensin on the energy metabolism of dairy cows in early lactation were investigated in a large clinical trial that was randomized and double-blinded. A total of 1010 Holstein cows and first lactation heifers were allocated to receive a controlled-release capsule of monensin or a placebo at 3 wk prior to expected calving date. Treatments were randomized across 25 dairy farms located near Guelph, Ontario, Canada. Serum samples obtained at the time of treatment administration and at wk 1, 2, 3, 6, and 9 postcalving were analyzed for beta-hydroxybutyrate, glucose, aspartate aminotransferase, urea, total protein, calcium, and phosphorus. Cows were also assigned a body condition score at the time each sample was obtained. Monensin treatment significantly reduced serum beta-hydroxy-butyrate concentrations at wk 1, 2, and 3 postpartum and significantly raised serum glucose concentrations during wk 1 and 2 of lactation. In addition, monensin treatment significantly reduced the loss of body condition score and decreased serum activity of aspartate aminotransferase during the postpartum period. Concentrations of serum urea were significantly higher during wk 2 and 3 postpartum for the cows that were treated with monensin. Monensin treatment had no effect on the concentrations of calcium, phosphorus, or total protein.  相似文献

An experiment was designed to determine the response of milk protein production and N utilization in dairy cows to supplementation of a predominantly rumen-undegradable protein (RUP) mixture with a fixed amino acid (AA) pattern and the response to the amount of feed intake. The experiment was designed as a 6 x 6 Latin square with a 3 x 2 factorial arrangement of treatments. The factors were three concentrations of RUP supplement (4.5, 14.9, and 29.1% of dry matter intake) and two levels of feed intake restriction (10 and 20%) of the basal diet. The supplement was designed to approximate a postruminal AA pattern that was similar to bovine caseins for Met, Lys, Phe, His, and Thr. Measurements were made during the last 5 d of each 21-d period. Milk protein production responded linearly as the concentration of RUP supplement in the treatment diet increased within the given range. The difference in feed intake restriction did not affect milk protein production. Efficiency of N utilization for milk production exceeded 30% for cows fed the lowest RUP supplement. Results indicated that there is an opportunity to increase milk protein production by using RUP formulations that are balanced for AA while minimizing waste N excretion.  相似文献

Isolated rat adipocytes were incubated with serum lipoproteins or lymphchylomicrons which contained 14c-labeled cholesterol. The specific activity of lipoprotein free cholesterol decreased and that of cellular free cholesterol increased linearly up to 7 h. At this time the cell cholesterol specific activity was only 11% of that of medium cholesterol indicating that the rate of exchange was slow. The specific activity of lipoprotein esterified cholesterol remained unchanged while that of cells showed a slight increase suggesting esterification of incorporated free cholesterol. No detectable change of the lipoprotein or cellular cholesterol concentration occurred indicating that the uptake of radioactive free cholesterol was due to exchange without net movement of sterol. The radioactive cholesterol was incorporated into both membrane fraction and the fat droplet of the adipocytes. The rate of cholesterol exchange was temperature-dependent but it was not influenced by the metabolic state of the cells and not by addition of metabolic inhibitors. Trypsin or pronase treatment of the cells were without influence on the rate of the exchange and denaturation of the plasma lipoproteins with formalin increased the rate of exchange. These results indicate that the exchange of cholesterol is a physical chemical process, which is not linked to energy metabolism of the cells, and which is not mediated by either specific lipoprotein receptors on fat cell membranes or pinocytic uptake of lipoproteins. The rate of free cholesterol exchange showed a linear correlation with the concentration of lipoprotein particles in the medium. The relative transfer rate was highest for chylomicrons and decreased in order chylomicron remnants greater than very low density lipoprotein greater than low density lipoprotein greater than high density lipoprotein. A saturation of the system could be obtained only with high density lipoprotein.  相似文献

Milk protein secretion is changed by increasing the proportion of energy, mainly as propionic acid, or the availability of AA. Whether associative effects exist between energy nature and protein amounts is unknown. Therefore, ruminal isoenergetic infusions of low or high propionate mixtures were combined factorially with duodenal infusion of sodium caseinate or control. Four ruminally and duodenally fistulated Holstein cows were used. The diet was limited and consisted of 70% forage and 30% concentrate. Caseinate infusion increased milk yield and protein and casein contents and decreased milk fat content; curd yields and coagulation properties of milk were improved. The infusion of propionic acid caused a large increase in rumen propionate. Milk yield tended to decrease, and milk fat decreased, but protein, casein, and curd yields were unchanged; milk-coagulating properties were improved.
