川西高原黑唇鼠兔乳酸脱氢酶C基因的原核表达

[目的]研究黑唇鼠兔乳酸脱氢酶C(LDH-C)基因的原核表达及重组蛋白的纯化.[方法]采用RT-PCR方法克隆鼠兔LDH-C基因,并将其连接到表达载体pET-32a上,构建鼠兔LDH-C基因的大肠杆菌BL21(DE3)工程菌,通过IPTG诱导表达,表达产物用SDS-PAGE进行分析,使用亲和层析方法进行蛋白纯化.[结果]RT-PCR扩增出1条约1.0 kbp的条带,与预期结果相符;重组表达载体经PCR和双酶切鉴定,产生1个约1.0 kbp的目的片段,表明表达载体构建成功;表达产物经SDS-PAGE分析,得到了分子量略大于45.0kDa以包涵体形式存在的融合蛋白;通过镍亲和层析柱进行纯化,得到了较纯的融合蛋白.[结论]黑唇鼠兔LDH-C基因被成功克隆和表达.     

肝素黄杆菌2-O-sulfatase基因的克隆与表达

[目的]研究肝素黄杆菌2-O-sulfatase基因的克隆与表达.[方法]以肝素黄肝菌为模板,通过PCR从肝素黄杆菌基因组DNA中扩增2-O-sulfatase的基因,测序正确后插入到表达质粒PET-28a(+)上构建表达载体,重组质粒转化E.Coll BL21(DE3)进行蛋白质表达.[结果]成功地将PCR扩增得到的测序正确的2-O-sulfatase基因构建入PET-28a(+)载体上,重组质粒转入E.Coil BL21(DE3)后,表达产物经SDS-PAGE凝胶电泳鉴定得到目的蛋白.[结论]克隆并成功表达了黄杆菌2-O-sulfatase基因,为分析肝素多糖分子及其降解产物结构奠定了基础.     

犬IL-2基因克隆及原核表达

[目的]研究犬IL-2基因的克隆和表达,为开发新型免疫增强荆和基因工程疫苗提供理论支持.[方法]从犬全血中分离出白细胞,经刀豆素刺激培养20 h后,提取总RNA;根据GenBank 中犬IL-2基因序列,设计1对引物,进行PCR扩增,并构建原核表达载体pET-28a,将其转化到宿主菌B121中,经IPTG 诱导表达.表达产物用SDS-PAGE进行分析.[结果]RT-PCR 扩增出一条约500 bp的条带,与预期结果相符;重组质粒pMD18-T-IL2 经双酶切后,产生一个约500 bp基因片段,说明目的基因被成功克隆;表达载体pET-28a-IL2 经双酶切和PCR鉴定后,分别产生一个约500 bp目的片段,表明表达载体构建成功;表达产物经SDS-PAGE 分析,分子量约为20 kDa,与预期蛋白大小基本相同.[结论]IL-2基因被成功克隆和表达.     

牛整合素β6基因的原核表达及其抗血清的制备研究

[目的]为深入研究ITGB6亚基的基因功能奠定基础.[方法]利用基因克隆技术获得整合素β6基因(ITGB6)胞外区,然后将其重组到pET-32a(+)质粒中,经鉴定及序列分析确定获得了重组阳性克隆;转化大肠杆菌BL21感受态细胞,IFTG诱导及蛋白纯化.[结果]SDS-PAGE结果显示,在94 kD处出现特异性条带;对该融合蛋白进行分离纯化后免疫新西兰兔,制备ITGB6蛋白抗血清,Western-blot分析表明抗血清可与原核表达的ITGB6蛋白特异性结合,ELISA方法检测抗血清的效价可达到1:2560.[结论]成功表达了牛的重组整合素β6基因.     

传染性法氏囊病病毒VP5蛋白跨膜区的敲除及可溶性原核表达研究

[目的]构建敲除传染性法氏囊病病毒(IBDV)VP5蛋白跨膜区序列的原核表达载体,并进行目的蛋白的表达、分离和纯化.[方法]利用PCR技术分别扩增IBDV VP5基因的胞外片段和胞内片段,然后将2个片段及pET-28b+)同时相接,即载体-胞内片段-胞外片段-载体,构建了敲除跨膜区基因片段的VP5重组表达质粒pET-VP5-FC及改进后的pET-VP5-SC.将表达质粒转化B121(DE3),IPTI;诱导后经Ni亲和层析及凝胶过滤层析纯化重组蛋白.[结果]得到可溶性表达的IBDV VPS.[结论]为进一步研究VP5蛋白的结构与功能奠定了基础.     

穿山龙薯蓣皂苷糖苷酶基因的亚克隆及表达

[目的]克隆并表达穿山龙薯蓣皂苷糖苷酶基因.[方法]将穿山龙薯蓣皂苷糖苷酶基因亚克隆到毕赤酵母表达载体 pPIC9K,再将构建后的表达载体电转化到毕赤酵母GS115中,并诱导表达该基因.[结果]重组酵母菌在0.5%甲醇中培养144 h后,所提取的酶蛋白具有穿山龙薯蓣皂苷糖苷酶的活性,经SDS-PAGE 分析,确定其相对分子质量为58 kD.[结论]穿山龙薯蓣皂苷糖苷酶基因得到了表达.     

公农2号紫花苜蓿抗寒基因CAS19在烟草中的表达

[目的]研究公农2号紫花苜蓿抗寒基因CAS19在烟草中的表达.[方法]根据苜蓿抗寒基因(CAS19 )核酸序列设计1 对引物,用RT-PCR 扩增出分离CAS19 的蛋白基因,并克隆到pMD18-T Vector载体中,再亚克隆到表达载体PBI121,成功构建重组表达质粒 PBCAS.经农杆菌介导转基因入烟草,并通过Sounthern-blotting杂交分析检测转基因苗.[结果]CAS19抗寒基因可以在烟草中得以高效表达.[结论]为进一步探明CAS19抗寒基因在烟草中的表达机制提供了理论依据.     

黄鳝抗菌肽hepcidin基因大肠杆菌表达载体的构建

[目的]构建黄鳝抗菌肽hepcidin基因大肠杆菌表达栽体.[方法]根据Genbank中黄鳝hepcidin基因序列设计引物进行PCR扩增,将测序正确的基因片段连接入栽体pET-28a中.[结果]以黄鳝cDNA为模板,扩增出hepcidin基因片段,长度为291 bp,连接到pET-28a上.经PCR扩增、双酶切验证,证实成功构建原核表达栽体.[结论]成功构建了黄鳝hepcidin基因大肠杆菌表达载体.     

纹枯病菌诱导水稻表达的WRKY基因克隆及分析

[目的]克隆纹枯病菌诱导的水稻上调表达基因.[方法]对应用抑制差减杂交枝术(SSH)获得的纹枯病菌诱导水稻表达的EST序列K16进行电子克隆,根据电子克隆产物设计引物进行RT-PCR、克隆测序,利用生物信息学技术对克隆产物进行功能预测.[结果]克隆出一个长度为1079bp的序列,结构功能分析显示该基因编码236个氨基酸的蛋白,存在多个功能位点,含有2个典型的WRKY结构域和1个C2H2锌指型结构,该基因与水稻WRKY8、WRKY24和WRKY30基因有较高的同源性.[结论]经电子克隆获得的纹枯病菌诱导表达的基因为典型的WRKY家族基因,该基因可能在水稻抗纹枯病中起重要作用.     

微针注射甜玉米胚芽gus基因转化瞬时表达研究

[目的]研究微针注射甜玉米(Zea mays L.)胚芽gus基因转化瞬时表达,为玉米遗传转化的进一步研究奠定基础.[方法]以农杆菌为介导,采用gus基因瞬时表达技术,建立了胚芽注射开放性基因转化体系.[结果]3 mm长的胚芽为最适转化受体;农杆菌EHA105菌液浓度最适OD值为0.558～0.630;gus基因转化最适共培养天数为3 d.在以上最适转化条件下,微针注射胚芽生长点基因转化率为1%.[结论]该研究可为改良甜玉米遗传性状,培育基因工程新品种提供参考.     

The effect of amount of possession of the trait of hostility on accuracy of perception of hostility in others.

Some theories of personality hold that an individual's perception of others is influenced by his own personality characteristics; more specifically, that a trait of low social value will be projected more into others, and that insight into the possession of these characteristics reduces this distortion. These 2 hypotheses were tested by having a group of college students rate each other (including themselves) on degree of friendliness or hostility. Agreement with group rating about self yielded the insight score. In general, the hypotheses were supported; the "friendly" Ss were most accurate in their perception of others, and insight was positively associated with accurate perception. Comparison of rates of infection of two methods of emergency ventricular drainage

The rates of infection of two methods of external ventricular drainage in use at Atkinson Morley's Hospital--namely, (a) percutaneous drainage with Rickham reservoirs and (b) tunnelled ventriculostomies--were compared in this retrospective review. Percutaneous drainage of CSF with Rickham reservoirs was associated with a 27% rate of infection as identified by positive microbiological cultures; tunnelled ventriculostomy catheters had a 10% infection rate. The difference in the infection rate between the two methods was statistically significant (P < 0.015). Other variables examined, including the age and sex of the patients and the reasons for ventricular drainage, were not associated with an increased rate of infection. Most infections from either method were caused by a coagulase negative staphylococcus. The average duration of ventricular drainage before identification of positive cultures was 5.7 days for Rickham reservoirs and 6.0 days for ventriculostomies.

转炉炉壳热应力分析

根据实际转炉建立实体模型。以有限元为手段,考虑了炉衬和炉壳材料的物性参数随温度变化的特点和炉衬与炉壳之间膨胀间隙,计算了转炉炉壳在温度载荷和炉衬膨胀压力同时作用下的热应力。所得结果可为转炉炉壳设计提供依据。     

Comparison of the results of surgical and conservative treatment of fractures of the diaphysis of the tibia

In this paper is treated the fundamental problematic of the fracture of the diaphysis of the tibia and the therapeutic results after the conservative and operative treatment are confronted. Among 518 patients with 433 open fractures and 35 with closed fractures of the diaphysis of the tibia, successful treatment of more 2/3 was obtained in the period from 1968 to 1972. Following our casuistics and literature, whose results are similar, we find out that the average total treatment of the osteosynthesis of the fracture of the diaphysis was shortened for 2,5 months depending upon the type and the localisation of the fracture. Apolied methods of the operative and conservative and conservative treatment in relation to the velocity of the medical cure as well as to the extreme anatomical-functional results are analised. We noticed that osteosynthesis, made by the intermedullary nail and the extension of the canals or by a small AO table, by the achieved stability makes conditions of immediate motions and an early rehabilitation. Because of that it gives the best functional results.

Results of treatment of cancer of the cardia

Cancer of the cardia is traditionally discussed with cardiac cancer of the lower portion of the esophagus and upper gastric cancer invading the esophagus, and the specific characteristics of cancer of the cardia have never been clearly defined. We reviewed the outcome of 172 patients with adenocarcinoma of the cardia who had undergone radical surgery between 1949 and 1994 in the Division of Gastrointestinal Surgery of the Cancer Institute Hospital. The centers of the tumors were located within 2 cm above and below the boundary between the esophagus and the stomach, and their longitudinal diameter was less than 8 cm. We divided the patients into an early period (1949-1979; n = 79) and a late period (1980-1994; n = 93), and focused on the historical transition. There were no differences in patient gender or histology between the two periods. However, the late period was associated with fewer cases of esophageal invasion and shorter longitudinal diameter, as the age of the population advanced. The number of advanced cancers such as the localized and infiltrative type had decreased, and early cancer and early clinical stage had become more common, but despite the fact that the number of early cancers had increased, extended dissection, such as thoracic and paraaortic lymph node dissection, was performed more frequently. By clinical stage, the long-term outcome markedly improved in Stage I patient in the late period, and tended to improve in Stage II and III patients. This appears to have been attributable to the prevention of micrometastasis by extended dissection, although the number of early cancers is another major potential cause. There were no differences in the outcome of Stage IV patients between the two periods, and further advances in multimodality therapy must be awaited. The range of resection is basically proximal gastrectomy, and if there are adequate indications, the prognosis is favorable. In view of the status of lymphatic flow and lymph node metastasis, and long-term results, lateroaortic lymph node dissection is important. Since the number of early cancer patients has been increasing, if intraabdominal recurrence is prevented, intrathoracic lymph node dissection will contribute greatly to the outcome of such patients.
