在体外模型上,对人工养殖的葛仙米藻胆蛋白粗提物、藻蓝蛋白和藻红蛋白的抗氧化活性进行比较研究。通过考察羟基自由基清除能力、超氧阴离子自由基清除能力、脂质过氧化抑制能力,对葛仙米中藻胆蛋白粗提物、藻蓝蛋白和藻红蛋白的抗氧化活性进行评价。体外抗氧化作用结果显示藻胆蛋白、藻红蛋白和藻蓝蛋白都有明显的抗氧化作用,但作用方式和抗氧化程度也有不同,在羟基自由基清除试验中,抗氧化活性由强到弱依次为藻胆蛋白藻蓝蛋白藻红蛋白;在超氧阴离子清除试验中藻红蛋白≧藻蓝蛋白藻胆蛋白;在抑制脂质过氧化试验中藻红蛋白藻胆蛋白藻蓝蛋白。人工养殖的葛仙米中藻蓝蛋白、藻红蛋白及藻胆蛋白粗提物具有很好的体外抗氧化活性,但三者抗氧化方式略有差异。 相似文献

藻蓝蛋白是一种存在于蓝藻、螺旋藻细胞中的色素蛋白复合体,除了能够捕获光能为藻类细胞提供能量外,藻蓝蛋白还具有抗炎性、抗肿瘤、抗氧化等多种生理功能,全面了解和掌握藻蓝蛋白的生理调控功能以及作用机制对藻蓝蛋白的开发与利用有着重要的指导意义。作者以藻蓝蛋白的研究现状出发,分别从抗肿瘤、抗炎、抗氧化以及免疫调节活性几个方面对藻蓝蛋白的生理功能和作用机制进行了阐述,对藻蓝蛋白提取过程中关键瓶颈——藻毒素的去除方法进行了总结和分析,并进一步提出了研究藻蓝蛋白调控机制的新筛选方法。作者为深入研究藻蓝蛋白的调控机理以及进一步开发安全性药物提供参考。 相似文献

目的:对葛仙米藻蓝蛋白和藻红蛋白的抗炎和抗紫外活性进行体外比较研究。方法:通过Lipopolysaccharide(LPS)诱导RAW264.7细胞分泌NO和TNF-α,比较葛仙米中藻蓝蛋白和藻红蛋白的抗炎活性;通过Ultraviolet radiation B(UVB)辐照诱导NIH-3T3成纤维细胞损伤,考察藻蓝蛋白和藻红蛋白的抗紫外损伤活性。结果:体外抗炎结果显示LPS能极显著诱导RAW264.7细胞分泌NO和TNF-α(P<0.01),藻蓝蛋白和藻红蛋白均能极显著抑制LPS诱导RAW264.7分泌的NO和TNF-α的含量水平(P<0.01),其中藻红蛋白抑制NO分泌能力高于藻蓝蛋白,藻蓝蛋白对TNF-α的分泌抑制作用高于藻红蛋白。体外抗紫外损伤结果显示UVB 4000 mJ/cm2可以成功诱导NIH-3T3细胞的紫外损伤,但是藻蓝蛋白和藻红蛋白对NIH-3T3细胞的紫外损伤并无明显保护作用。结论:葛仙米中藻蓝蛋白和藻红蛋白具有很好的体外抗炎活性,但两者对UVB诱导的细胞损伤并无明显保护作用。 相似文献

本研究利用代谢工程技术原理在重组大肠杆菌表达合成了嗜热聚球藻(Thermosynechococcus elongatus BP-1)别藻蓝蛋白β亚基(holo-apcB),并利用亲和层析的方法对holo-apcB进行了分离纯化。SDS-PAGE电泳胶图显示,holo-apcB分子量与18.4 ku的蛋白条带相近,与理论分子量吻合。光谱学分析结果显示,其最大吸收峰为615 nm,最大荧光发射峰为640 nm,与天然别藻蓝蛋白β亚基具有相同的分子量和光谱学性质。在55℃条件下,holo-apcB的半衰期高达31.9 h。以嗜热聚球藻天然别藻蓝蛋白(APC)及脱辅基别藻蓝蛋白β亚基(apo-apcB)为参照,分析了holo-apcB的抗氧化活性。结果显示,holo-apcB具有最强的抗氧化能力,其清除羟自由基和氢过氧自由基半数抑制浓度(IC50)分别为224.9μg/mL和20.6μg/mL,显著低于apo-apcB和天然APC的IC50,这表明holo-apcB中的藻蓝胆素也参与了抗氧化过程。 相似文献

蔷薇藻(Rhodella reticulata)是属于红藻门的一种海洋单细胞微藻,在生长过程中会产生藻胆蛋白、胞外多糖等生物活性物质。文中研究了蔷薇藻的藻蓝蛋白的分离纯化及抗氧化性能,藻细胞经过破碎,通过硫酸铵沉淀和DEAE Sepharose FF柱层析后,可以分离出较纯的藻蓝蛋白,纯化的藻蓝蛋白在620 nm处有一个最高的吸收峰,纯度为3.92,经SDS-PAGE电泳后,测得藻蓝蛋白含有α和β两个亚基,分子质量分别为18 ku和22 ku。蔷薇藻藻蓝蛋白由17种氨基酸组成。蔷薇藻纯化后的藻蓝蛋白比粗藻蓝蛋白具有更高的抗氧化能力,且呈剂量依赖效应。 相似文献

选用Sephadex G-150(G-150)、Sephadex G-200(G-200)、Sephacryl S-200(S-200)和Sephacryl S-300(S-300)四种凝胶对富含藻红蛋白的多管藻藻胆蛋白提取液进行过滤,并对所得藻蓝、别藻蓝蛋白组分做进一步纯化,优化建立了有效分离去除藻红蛋白、纯化制备藻蓝、别藻蓝蛋白组分的技术方法,即依次使用G-150、S-300和S-200凝胶过滤,所得样品中藻蓝和别藻蓝蛋白相对含量显著提高。并且根据实验现象,对凝胶柱的选择性、柱效、内水体积以及样品的蛋白组成等主要因素与凝胶过滤实际效果的相互关系进行了论述。 相似文献

目的:研究藻蓝蛋白对环磷酰胺诱导小鼠肝肾组织损伤的保护作用,为评价肝肾治疗药物提供新的研究思路。方法:雌性BALB/c小鼠随机分为空白组、模型组、阳性对照组以及藻蓝蛋白低、中、高剂量组。采用腹腔注射环磷酰胺后建立小鼠肝肾损伤模型,然后空白组和模型组小鼠灌胃生理盐水,样品组给予不同剂量藻蓝蛋白溶液(50、100、200 mg/kg),阳性对照组灌胃盐酸左旋咪唑(40 mg/kg),灌胃时间5 d。采用试剂盒法分别测定各组小鼠血清中的白细胞介素-2(IL-2)、肿瘤因子-α(TNF-α)和免疫球蛋白(IgG)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平;肝组织的超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、天冬氨酸氨基转移酶(AST)、谷丙转氨酶(ALT);及肾组织的超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、尿酸(UA)、尿素氮(BUN)水平并观察小鼠肝脏、肾脏组织形态学变化。结果:与模型组对比,藻蓝蛋白剂量组小鼠血清中的IL-2、TNF-α、MDA水平极显著下降(P<0.01);藻蓝蛋白高剂量组可明显降低血清中的IgG水平(P<0.05)并提高血清中的SOD水平(P<0.05);同时可降低由环磷酰胺引起的小鼠血清中MDA含量升高的情况(P<0.01),升高SOD活性(P<0.05);此外,藻蓝蛋白剂量组小鼠的肝组织GSH-Px水平极显著高于模型组(P<0.01),而MDA、ALT、AST水平均极显著低于模型组小鼠(P<0.01);相较于肝组织,藻蓝蛋白剂量组小鼠肾组织的MDA水平极显著低于模型组(P<0.01),仅低剂量藻蓝蛋白组小鼠肾组织的BUN水平极显著低于模型组(P<0.01),高剂量藻蓝蛋白组小鼠肾组织的UA水平极显著低于模型组(P<0.01)。结论:综合各项指标结果,藻蓝蛋白对环磷酰胺导致的小鼠肝肾损伤具有明显改善作用。 相似文献

Abstract: The aim of this study is to investigate in vitro antioxidant, antimicrobial, and anticancer activity of the acetone extracts of the lichens Umbilicaria crustulosa, U. cylindrica, and U. polyphylla. Antioxidant activity was evaluated by 5 separate methods: free radical scavenging, superoxide anion radical scavenging, reducing power, determination of total phenolic compounds, and determination of total flavonoid content. Of the lichens tested, U. polyphylla had largest free radical scavenging activity (72.79% inhibition at a concentration of 1 mg/mL), which was similar as standard antioxidants in the same concentration. Moreover, the tested extracts had effective reducing power and superoxide anion radical scavenging. Total content of phenol and flavonoid in extracts was determined as pyrocatechol equivalent, and as rutin equivalent, respectively. The strong relationships between total phenolic and flavonoid contents and the antioxidant effect of tested extracts were observed. The antimicrobial activity was estimated by determination of the minimal inhibitory concentration by the broth microdilution method. The most active was extract of U. polyphylla with minimum inhibitory concentration values ranging from 1.56 to 12.5 mg/mL. Anticancer activity was tested against FemX (human melanoma) and LS174 (human colon carcinoma) cell lines using MTT method. All extracts were found to be strong anticancer activity toward both cell lines with IC50 values ranging from 28.45 to 97.82 μg/mL. The present study shows that tested lichen extracts demonstrated a strong antioxidant, antimicrobial, and anticancer effects. That suggests that lichens may be used as possible natural antioxidant, antimicrobial, and anticancer agents. 相似文献

探讨紫苏籽皮提取物(PSCE)的体外自由基清除能力及其抗癌活性。结果表明,以DPPH自由基和羟基自由基清除实验检测自由基清除能力。MTT法和平板克隆形成实验分别检测体外抗癌活性。PSCE具有较好的自由基清除能力,对DPPH自由基和羟基自由基的半清除剂量(IC50)分别为105.53μg/mL和269.65μg/mL。同时,PSCE对A375SM人黑色素瘤细胞的增殖抑制作用呈浓度和时间依赖关系。 相似文献

BACKGROUND: Lichens are symbiotic organisms consisting of algae and fungi. They are used for human and animal nutrition and in the production of colours, perfumes and alcohol. Lichens have also been used in traditional medicine to treat diseases such as jaundice, pulmonary, stomach and cranial diseases. In this study the acetone extracts of three lichens, Parmelia caperata, Parmelia sulcata and Parmelia saxatilis, were tested for their antioxidant, antimicrobial and anticancer potential. RESULTS: Of the lichens tested, P. saxatilis had the highest free radical‐scavenging activity (55.3% inhibition). Moreover, all tested extracts showed effective reducing power and superoxide anion radical scavenging. Strong relationships between total phenolic and flavonoid contents and antioxidant effects of the tested extracts were observed. The extract of P. sulcata was most active in terms of antimicrobial ability, with minimum inhibitory concentration values ranging from 0.78 to 12.5 mg L?1. All extracts were found to have strong anticancer activity, with IC50 values ranging from 9.55 to 22.95 µg mL?1. CONCLUSION: The present study showed that the tested lichen extracts exhibited strong antioxidant, antimicrobial and anticancer effects. This study aimed to investigate in vitro anticancer activity by antiproliferative activity, antihypertensive activity by angiotensin-converting enzyme inhibition, antidiabetic activity by α-amylase and α-glucosidase inhibitions, and antioxidant activities of camel milk fermented with camel milk probiotic compared with fermented bovine milk. The camel milk probiotic strain Lactococcus lactis KX881782 (Lc.K782) and control Lactobacillus acidophilus DSM9126 (La.DSM) were used to prepare fermented camel and bovine milks separately. The proteolytic activities of water-soluble extract (WSE) in all fermented camel milk were higher than those in fermented bovine milk. The α-glucosidase inhibitions in both milk types fermented by Lc.K782 ranged from 30 to 40%. Camel milk fermented by Lc.K782 had the highest antioxidant activity by 2,2'-azino-bis(3-ethylbenzo-thiazoline-6-sulphonic acid). The highest angiotensin-converting enzyme inhibition of WSE in camel milk fermented by Lc.K782 was >80%. The proliferations of Caco-2, MCF-7, and HELA cells were more inhibited when treated with WSE of fermented camel milk extracts. 相似文献
