The main purpose of this paper is to analyze the effects of low-molecular-weight silk fibroin peptides (LMW-SF) on cell growth characteristics. Firstly, LMW-SF was achieved from silk fibroin enzymolysis by neutral protease. Secondly, LMW-SF was sterilized by filtration and high-pressure steam, respectively. Finally, the effects of LMW-SF on the growth of human normal liver cell (L02) and human liver cancer cell (HepG2) were tested under the volume concentration of 0. 1 ∼5.0 mg/mL, and the relative growth rate (RGR) was calculated. The results show that LMW-SF have slight effect on cell growth at low concentration (0. 1 mg/mL and 0. 5 mg/mL); when the volume concentration is high (2. 5 mg/mL and 5. 0 mg/mL), the lowest value of RGR is 8. 3%, indicating a strong inhibition effect on cell growth. RGR is up to 227. 4% after high-pressure steam sterilization, showing a strong promotion effect on cell growth. It is obvious that LMW-SF have a strong effect on promoting cell growth after highpressure steam sterilization, and high-pressure steam sterilization could be a necessary process when protein is applied in tissue engineering scaffolds. © 2017 China Silk Association. In this study, the target bioactive compounds (e.g., alkaloids, flavonoids, saponins and other polar compounds) in Vietnamese Caricapapaya leaves were extracted. The cytotoxic activities of the papaya leaves extracts on the selected tumor cell lines, such as lung cancer cell line LU-1, carcinoma cell line KB, breast cancer cells MCF7 and leukemia cell line HL-60, were examined. Preliminary findings showed a high inhibitive activity of papaya leave extracts against the four tested tumor cell lines at the concentration of 100 μg/mL. Out of the bioactive compounds in papaya leaves extract, alkaloids showed the highest inhibitive activity (105.95% on MCF7 and 91.86% on LU-1), followed by polar compounds (62.88% on LU-1 and 21.80% on KB), and saponins (59.74% on MCF7 and 25.25% on LU-1). Flavonoids has the lowest inhibitive activity on cell lines (e.g., 45.51% on MCF7 and 20.32% on LU-1). Taken together, the results suggest that alkaloids have a relatively high inhibitive activity on the selected tumor cell lines and their stimulated concentration at 50% (IC50) values for on MCF7 and KB were 24.67 μg/mL and 33.56 μg/mL, respectively. However, the result pointed out the immunostimulatory ability of only polar compounds and saponins which could stimulate the growth of in vitro lymphocytes but not flavonoids and alkaloids. The SC50 (stimulated concentration at 50%) values of polar compounds and saponins were 287.87μg/mL and 192.99 μg/mL, respectively.  相似文献

Antioxidants play an important role in inhibiting and scavenging free radicals in human, providing protection against cellular damage in relation to cancer initiation. Seaweeds have been proved to have high antioxidant activity. Thus, this research was carried out to determine the antioxidant and anticancer properties of edible red seaweed, Gracilaria manilaensis (Gracilariales, Rhodophyta). The extracts were prepared by Soxhlet extraction using organic solvents with different polarities. The antioxidant activities of extracts were determined in terms of their flee radical scavenging activity (RSA IC50) and total phenolic content (TPC). The cytotoxic activity of extracts were tested against human ovarian cancer cell line (Caov-3), human breast cancer cell line (MDA-MB-231 and MCF-7), human cervical cell line (HeLa), mouse fibroblast cell line (L929) and Madin-Darby canine kidney (MDCK) and the cell viability after 72 h incubation was determined by methylene blue assay. The findings showed that acetone extract has the lowest DPPH IC50 value followed by ethyl acetate extract. Both extracts also showed high values of TPC. Dichloromethane extract had the strongest cytotoxic on MDA-MB-231 (53.90 μg/mL ± 5.59 μg/mL) and HeLa (95.50 μg/mL). While, acetone and ethyl acetate extracts were cytotoxic on MCF-7 (66.07 μg/mL) and Caov-3 (69.67 μg/mL ± 13.94 μg/mL). It could be concluded that the antioxidant and cytotoxic activities of G. manilaensis were influenced by the types of solvents used and thus had a potential to develop as a cancer chemoprevention or anticancer agent against selected cancer.  相似文献

The growth conditions of La0.7Ca0.3MnO3(LCMO) epitaxial thin film are investigated.The results indicate that the deposition temperature plays a crucial role on the growth mode of LCMO thin film.There is an optimal deposition temperature,To,for the epitaxial growth of LCMO thin film.Just only at the To,the real epitaxial LCMO thin film can be obtained.When the deposition temperature deviates from the To,the thin film tends to be polycrystalline,although it orientes growth along a certain direction(a or c axis).Moreover,the experimental results show that just only for the epitaxial thin film the ferromagnetic-paramagnetic transition temperature(i.e.the Courier temperature Tc) equals to the metal-semiconductor transition temperature(Tp).The ferromagnetic state shows metallicity and the paramagnetic state exhibits semiconducting character.  相似文献

Pulp and papermaking industries generate high volumes of carbohydrate-rich effluents. Microbial fuel cell (MFC) technology is based on organic materials’ consumption?and efficient power production. Using a classical two-chamber lab-scale MFC design with an external resistance of 2000 W, we investigated the effects of anode chamber biofilm adaptation (ACBA) and cathode chamber redox solutions (CCRS) on the operation efficiency of MFC when treating wastewater. In ACBA studies, biofilm growth activation showed an increase in the power density to 20.48, 35.18, and 36.98 mW/m2 when the acetate feeding concentrations were 3, 6, and 12 g/L, respectively. Improvement by biofilm adhesion on granular activated carbon (GAC) was examined by scanning electron microscopy (SEM). The obtained power density increased to 25.47, 33.42, and 40.39 mW/m2 when the GAC particles concentrations were 0, 50, and 100 g/L, respectively. The generated power densities were 51.26 and 40.39 mW/m2 as well as the obtained voltages were 0.41 and 0.72 V when the electrode area increased from 16 to 64 cm22, current density of 0.094 A/m2, and voltage of 1.20 V with a successful organic removal efficiency of 86.0% after 264 h of operation.  相似文献

In this study,natural bamboo fiber was prepared combining chemical pretreatment with mechanical disc refining,opening,and carding.An orthogonal experiment was designed based on four factors and three levels;thereafter,the manufacturing process was optimized.The length,diameter,tensile strength,and elastic modulus of the bamboo fiber were determined,and the crystallinity and morphology of the fiber were analyzed using X-ray diffraction(XRD)and scanning electron microscopy(SEM).The results showed that the optimum parameters for the chemical pretreatment were a cooking temperature of 130℃,heating time of 2 h,NaOH dosage of 2%,and Na2SO3 dosage of 10%.The cooking yield of bamboo chips was 89.5%,and the carding yield of natural bamboo fiber was 43.0% under the optimum conditions.The length,diameter,tensile strength,and elastic modulus of the obtained fiber were 36.71 mm,0.285 mm,407 MPa,and 27.7 GPa,respectively.XRD analysis and SEM observations showed that the technology used in this study can produce bright and compact natural bamboo fibers with high crystallinity.  相似文献

为解决土豆烧牛肉方便菜肴产品的胀袋问题,对胀袋微生物进行分离、纯化、鉴定和产气特性研究。分离、纯化得到7株优势微生物。结合形态学观察、16S rRNA序列分析及基质辅助激光解析/电离飞行时间质谱(MALDI-TOF MS)技术鉴定分离微生物,结果显示2株为枯草芽孢杆菌(Bacillus subtilis),2株为表皮葡萄球菌(Staphylococcus epidermidis),2株为地衣芽孢杆菌(Bacillus licheniformis),1株为凝结芽孢杆菌(Bacillus coagulans)。将分离菌株接种到无菌土豆烧牛肉样品中,发现枯草芽孢杆菌(Bacillus subtilis)和地衣芽孢杆菌(Bacillus licheniformis)有明显产气现象,是引起产品胀袋的主要原因。  相似文献

为提高传统米酒的香气品质、实现其标准化生产,对从中国几种地方特色甜酒曲中分离筛选出的具有优良发酵和产香特性的酵母菌株进行26S rDNA序列分析及系统发育树构建,并分析了其发酵米曲汁的香气成分及特征。结果表明:所筛选的6 株酵母菌中,CMY001、CMY003和NBY003为酿酒酵母(Saccharomyces cerevisiae),CMY002和NBY002为异常威克汉姆酵母(Wickerhamomyces anomalus),YCY001为光滑假丝酵母(Candida glabrata)。乙醇体积分数小于10%时YCY001耐受性较好,大于10%时CMY002耐受性较好;NBY002对温度耐受性良好;各菌株在pH 3~11条件下均能生长;发酵能力最强的是NBY003,菌株NBY002的产酯量最高,为5.125 g/L。在米曲汁培养基中,NBY002产β-苯乙醇能力明显高于其他菌株,发酵产物呈现较强的花香味;YCY001发酵产物中酯类的多样性及相对含量显著高于其他菌株,具有较丰富的酯香味;CMY001发酵产物具有脂肪香、酒香和草香特征,CMY002和CMY003的发酵产物果香浓郁;6 种酵母菌发酵产物的整体香气有明显差异,其中丙烯酸辛酯、乙酸苯乙酯、乙酸乙酯、甲酸异戊酯、β-苯乙醇、辛酸和苯甲酸乙酯是不同酵母发酵产物呈现不同香气特征的关键物质。  相似文献

实验以白色念珠菌为对象,对从渤海湾不同站位分离获得的55株海洋丝状真菌进行了抗真菌活性菌株的筛选。从中筛选得到了BH431、BH515、BH531和BH09721 4株抑菌活性较高的菌株,并进一步对不同浓度发酵液的抑菌活性及代谢物性质进行了检测。结果显示,4株海洋真菌对野生型白色念珠菌SC5314均具有较强抑菌活性,其最小抑菌浓度分别为 65 g/mL、19 g/mL、54 g/mL、23 g/mL;其中BH431和BH515菌株还显示出对基因敲除型菌株RM1000的抑菌活性,最小抑菌浓度分别为 32 g/mL、39 g/mL。4株海洋真菌所产生的活性代谢物具有较好的酸碱适应性,活性物质分子质量<6 000 u;其中BH431菌株的活性代谢物还具有良好的热稳定性。  相似文献

该研究对坛装贮存黄酒关键污染微生物进行分离纯化和鉴定。利用梯度稀释法和划线纯化法得到编号为B001、B002、B003的3株细菌和编号为F004、F005、F006的3株真菌。通过形态学特征观察、生理生化鉴定和分子生物学鉴定方法对分离菌株进行鉴定。结果表明,细菌B001、B002、B003分别为解淀粉芽孢杆菌(Bacillus amyloliquefaciens)、铜绿假单胞菌(Pseudomonas aeruginosa)和假肠膜明串珠菌(Leuconostoc pseudomesenteroides),真菌F004、F005、F006分别为头状螺旋地霉(Saprochaete capitate)、爪哇正青霉(Eupenicillium javanicum)及红曲菌(Monascus sp.)。  相似文献

目的 对小米乳酸菌饮料胀罐微生物进行分离和菌株鉴定,并对所分离菌株的产气特性进行研究。方法 使用孟加拉红琼脂培养基、营养肉汤琼脂培养基和改良MRS琼脂培养基对小米乳酸菌饮料胀罐微生物进行分离和纯化,根据菌落形态观察、菌体形态观察、16S rRNA 序列分析并结合基质辅助激光解析/电离飞行时间质谱(ALDI-TOF MS)技术对分离菌株进行鉴定。最后将分离菌株接种到无菌小米乳酸菌饮料产品中,培养观察产气现象。结果 共分离出6株细菌,编号为B001、B002、B003、B003、B005、B006,菌种鉴定结果均为副干酪乳杆菌(Lactobacillus paracasei),6株细菌均具有产气现象。结论 副干酪乳杆菌是引起本研究产品胀罐的主要原因。  相似文献

目的旨在明确污染速冻猕猴桃产品加工环节中主要的高渗酵母种类,并且研究污染菌种的耐渗特性,从而在加工中能够预防性地识别和控制高渗酵母污染。方法对酵母基因组5.8S rDNA-ITS区或26S rDNA的D1/D2区两个保守区域进行扩增和测序,确定分离筛选的高渗酵母的种属信息和分布。测定部分菌株在2%、50%、60%(m:V)葡萄糖浓度下的OD600,确定耐糖特性。同时,测定部分菌株在2%和50%(m:V)糖浓度下的产气量。结果在速冻猕猴桃加工环境中共分离筛选到33株高渗酵母,分属于10个酵母种。其中,Hanseniaspora uvarum、Hanseniaspora opuntiae、Candida intermedia和Candida tropicalis是发生频次高的耐高渗酵母种。Lodderomyces elongisporus和Debaryomyces hansenii耐渗性强,能够耐受60%(m:V)葡萄糖;Hanseniaspora opuntiae和Metschnikowia pulcherrima在高糖培养基中产气量大。结论速冻猕猴桃加工车间中分离到潜在污染性的耐高渗酵母,尤其是车间空气中存在多种耐高渗酵母,因此需要加强加工过程中的卫生管理,增加预防性的杀菌措施,保证速冻猕猴桃加工过程的质量安全。  相似文献

为分离筛选具有产区特色的菌种,本实验采用梯度稀释和划线分离技术,从沙城地区3 个龙眼葡萄园土壤中共分离得到210 株酵母菌。按照形态表型分为9 类,又利用核糖体5.8S rDNA-ITS 区域RFLP 方法将这9 类酵母在分子水平上进行了区分,其中两类酵母为常见酵母,分别为东方伊萨酵母(Issatchenkia orientalis)和热带假丝酵母(Candida tropicalis)。实验证明了该地区土壤中酵母菌的多样性。  相似文献

使用3M PetrifilmTM快速测试片法(3M测试法)和滤膜法对饮料中的菌落总数进行定量测定,分析两种方法的一致性程度。选用大肠埃希氏菌、金黄色葡萄球菌、爱媛类芽孢杆菌、枯草芽孢杆菌、Asia Siamensis、白色念珠菌和黑曲霉七种菌,对饮料样本进行人工接种并测试。实验结果显示,对于不同菌株及不同污染水平的测试,3M测试法和滤膜法的线性拟合度较好(决定系数R2均>0.95)。且3M测试片法结果回收率均>70%,RSD值(2.9%~34.3%)也均在2020《中国药典》的规定范围内,说明3M测试片法的准确度及精密度较好。对于细菌、真菌分别污染的专属性检验和不同批次的3M测试片的重复性测试中,3M测试片法的回收率>70%,且RSD值(3.6%~34.3%)均在可接受范围之内,表现出良好的专属性和重现性。使用相对正确度和精度剖面分析方法对3M测试片法和滤膜法的结果进行分析,结果显示两种方法具有较好的一致性和等效性。综合以上分析结果,3M测试片法可等效于滤膜法,进行饮料中的菌落总数测定。  相似文献

Polyamines (putrescine, spermidine and spermine) participate in many biochemical processes that are indispensable for cell differentiation and proliferation. In the present study, polyamine types were determined in 13 Debaryomyces hansenii strains isolated from different sources by high pressure liquid chromatography. Very low putrescine levels were observed in the strains DhhBCS007, DhfBCS001 and DhfBCS002. Spermidine and spermine were the most prevalent amines detected in all the yeasts, ranging from 7.71 to 18.42 mg/kg of spermidine in the strain L2 and 9.39–18.96 mg/kg for spermine in strain DhhCBS004. D. hansenii strain L2 and CBS004 isolated from citrus fruit and marine water, respectively, were the main polyamine-secreting yeasts. The results will contribute to our ability to select the most appropriate strains for use, as dietary complement (probiotic) in the animal food industry, including aquaculture.  相似文献
