Five multiparous Holstein cows in midlactation that were fitted with ruminal and duodenal cannulas were used in a 3 x 5 incomplete Latin square. The objective of this study was to examine the effects on nutrient digestion of wheat processing and method of tallow addition to the diets of lactating dairy cows. Diets consisted of 45% forage and 55% concentrate, and each diet contained 20% wheat and 2% tallow (as-fed basis). Treatments were dry-rolled wheat with tallow added to the concentrate, steam-rolled wheat with tallow added to the concentrate, and steam-rolled wheat with tallow added first to the wheat. The dry matter intake; digestion of starch, fiber, and fatty acids; ammonia N concentration; and molar proportions of volatile fatty acids in ruminal fluid were not affected by treatments. The apparent digestibility in the total tract of organic matter and nitrogenous compounds was significantly higher for the steam-rolled treatment with tallow added first to the wheat. Mean ruminal fluid pH was similar across treatments; however, cows fed the diet containing steam-rolled wheat with tallow added first to the wheat had the smallest pH change from 0 to 2 h postfeeding. Milk yield did not differ, regardless of cow diet. Method of tallow addition had marked effects on the apparent digestibility of organic matter and N in the total tract of lactating dairy cows.  相似文献

A glycoprotein B (gB) gene homologue was identified in a 5.4-kb BamHl genomic fragment of the phocid herpesvirus type-1 (PhHV-1) which represents a widespread and important pathogen of pinnipeds. Sequence analysis revealed a gB-specific open-reading frame comprising 881 amino acids. Phylogenetic analysis gave evidence for a close evolutionary relationship between PhHV-1 and members of the Varicellovirus genus of the alpha-Herpesvirinae and canid herpesvirus in particular. In PhHV-1-infected Crandell feline kidney cells gB is expressed as a 113-kDa glycosylated molecule which is proteolytically cleaved into at least two fragments of 67 and 53-59 kDa apparently forming disulfide-linked heterodimers of 140 kDa. Cell surface expression of PhHV-1 gB was confirmed by FACS analysis. Thus, synthesis and processing of the gB protein of PhHV-1 follows a pattern also observed in other Varicelloviruses. Since the gB protein of herpesviruses, expressed in the baculovirus system, has been shown to be a suitable target for vaccine design, we used this system for expression of PhHV-1 gB. Recombinant (rec) baculovirus-expressed gB was identified as a 105-kDa glycosylated molecule. Proteolytic cleavage into fragments of 62 and 52 kDa was markedly delayed compared to wild-type (wt) gB. Wt and rec gB harbored endoglycosidase H (precursor)- as well as N-glycosidase F-sensitive N-glycans (proteolytic fragments). Baculovirus-expressed gB appeared to be antigenically authentic, since it was recognized in radioimmunoprecipitation and immune peroxidase monolayer assays by PhHV-1-neutralizing seal sera and by gB-specific neutralizing murine monoclonal antibodies. Furthermore, PhHV-1-neutralizing antibodies were induced in mice following immunization with baculovirus-expressed gB, indicating its suitability for incorporation in a candidate vaccine for seals.  相似文献

As part of a total mixed ration, two rumen-fistulated dairy cows were fed meat and bone meal that had been artificially contaminated with Salmonella spp. Samples from the rumen, feces, and milk were taken 3 d/wk and cultured for salmonella. Rectal temperatures and rumen pH were also measured at the time of sample collection. Over the 2-mo study, salmonella were intermittently recovered from rumen contents, from feces, and from necropsy specimens of rumen contents, cecal contents, and mesenteric lymph nodes. No excretion of salmonella in milk was detected. An elevated rumen pH was associated with increased isolation of salmonella. No clinical illness was observed for either cow. Meat and bone meal that has been contaminated with low concentrations of salmonella is unlikely to result in clinical illness in healthy adult lactating cows. However, dairy producers should continue to be concerned about feed biosecurity and water contamination of animal by-products to prevent and control contamination by salmonella.  相似文献

Adequacy of amino acids in diets fed to lactating dairy cows   总被引:1,自引:0,他引:1  
The Cornell Net Carbohydrate and Protein System was used to evaluate absorbable limiting amino acids (AA) for milk yield. The system was utilized to characterize whether diets in five previous experiments met AA requirements for milk protein synthesis. Twenty-nine treatment means for milk yield from 367 cows constituted the database for the evaluation. Using the mechanistic relationships of nutrient metabolism described in the Cornell system, absorbed amounts were predicted of each essential AA from a diet and milk yields allowed by the most limiting AA. Regression of observed milk yield (Y) on predicted milk yield (X) using all treatment means (n = 29) was Y = 2.3 + 0.799X (r2 = 0.72). The linear relationship was stronger using 6 treatment means (n = 18) when protein supplements were fed rather than when ruminally protected AA were fed or when AA were postruminally infused (Y = -1.8 + 0.983X; r2 = 0.93). The Cornell system predicted that, for diets based on corn, Met or Lys was limiting when soybean meal was the protein source, but Lys was limiting when corn gluten meal or brewers grains were the source of protein. By AA limitation, the Cornell system explained differences in milk yield for diets that differed in supplemental protein sources in some of the experiments. As determined from milk protein yields in these studies, requirements for individual essential AA were expressed as a percentage of dietary dry matter or total essential AA, and values were relatively constant among dietary treatments and experiments.  相似文献

Although the effect of hyperoxia on antioxidant enzymes is well known, the effect of subtoxic levels of hyperoxia on gamma-glutamyltransferase (gamma-GT), involved in the degradation and uptake of extracellular GSH for intracellular GSH synthesis, is unknown. The aim of the study was to investigate (1) the effects of in vitro hyperoxia on gamma-GT activity of type II cells and (2) the effects of the lazaroid U-74389G and N-acetylcysteine (NAC) on the hyperoxia-induced changes in gamma-GT and antioxidant enzyme activities. At 48 h after isolation, rat type II cells were exposed for 2 days to air, 60% O2 or 85% O2 with or without 30 microM U-74389G or 100 microM NAC. After the exposure, the cells were harvested and assayed for superoxide dismutase (SOD), glutathione peroxidase (GPx), gamma-GT activity, and GSH levels. In another series of experiments 85% O2-exposed cells, with or without U-74389G, were used for Northern blotting of gamma-GT mRNA. Exposure to 60% O2 decreased gamma-GT and GSH by -47 and -34%, respectively, while SOD and GPx activities remained unchanged. After 85% O2-exposure gamma-GT decreased by -55%, SOD and GPx increased by +55 and +87%, respectively, while GSH decreased by -35%. NAC treatment decreased gamma-GT activity by -42% in the air-exposed cells. After 60% O2, U-74389G led to significantly higher gamma-GT (+117%) and GSH (+26%) while NAC only led to higher GSH (+28%) compared to the oxygen-exposed cells not treated with NAC or U-74389G. After 85% O2 U-74389G increased gamma-GT, SOD, and GSH by +72, +58, and +68%, respectively, while NAC only increased SOD (+49%) and GSH (+26%) compared to the oxygen-exposed cells not treated with NAC or U-74389G. The 85% O2 exposure, with or without U-74389G, had no effect on gamma-GT mRNA levels. The results show that hyperoxia decreases rat type II cell gamma-GT activity in vitro. This effect was not related to an altered regulation at mRNA level and it was not associated with the hyperoxia-induced decrease in intracellular GSH, since restoration of the GSH levels by NAC did not restore gamma-GT activity. The lazaroid U-74389G with vitamin E-like properties effectively prevented the decrease in gamma-GT and GSH, so that direct inactivation of the membrane-bound gamma-GT by hyperoxia is the most likely mechanism.  相似文献

An 18-wk lactation study was conducted to determine whether the effects of tallow on the lactation performance of dairy cows were influenced by particle size of hay in the ration. A total mixed ration containing 50% concentrate, 25% corn silage, and 25% alfalfa hay (dry matter basis) was fed to Holstein cows. Four total mixed rations were developed based on differences in the percentage of tallow in the concentrate and particle size of alfalfa hay: 1) 0% tallow, long-cut hay; 2) 0% tallow, short-cut hay; 3) 5% tallow, long-cut hay; and 4) 5% tallow, short-cut hay. Ration had no effect on dry matter intake, body weight gain or change in body condition score. Tallow increased milk and milk protein yields but reduced milk protein concentration. However, the effects of tallow on milk and milk protein yields were the same, regardless of hay length in the ration. A tendency for an interaction of tallow and hay particle size was detected for fat-corrected milk (FCM) because tallow increased FCM more when hay was short. Ration had no effect on volatile fatty acids in ruminal samples collected via a stomach tube. In this study, the effects of tallow on milk yield and composition from Holstein cows were the same, regardless of hay particle size in the ration. The tendency for tallow to increase FCM more when hay was short suggests at least a limited role of forage particle size in the determination of how fat supplements in dairy rations affect lactation performance.  相似文献

Twelve Holstein cows and 12 Jersey cows were used in six 4 x 4 Latin squares to investigate the effects of the degradability of dietary protein and supplemental dietary fat on milk components. Dietary dry matter contained 16% crude protein with two concentrations of ruminally undegradable protein (RUP) obtained by substituting blood meal for a portion of the soybean meal. Treatments were 1) 29% RUP, 0% added fat; 2) 29% RUP, 2.7% added fat (Ca soaps of fatty acids); 3) 41% RUP, 0% added fat; and 4) 41% RUP, 2.7% added fat. The dry matter of the total mixed ration fed at 1000 and 1400 h consisted of 30% corn silage, 29% alfalfa haylage, and 41% concentrate. Supplemental dietary fat depressed dry matter intake by 6.2%. Plasma urea N was greater at 0700 and 1600 h for Jerseys fed diets containing added fat and greater at 0700 h for Holsteins fed diets containing 41% RUP than for Holsteins fed 0% added fat and 29% RUP. When averaged across both breeds, milk production increased 7.1%, and production of 4% fat-corrected milk by Jerseys increased 8.4%, in response to added dietary fat. Milk protein was reduced when Holstein diets contained 41% RUP. Milk protein content was reduced 7.1 and 3.9%, and milk urea N was increased 4.9 and 8.5%, by added fat and 41% RUP in both breeds, respectively. Added fat reduced the concentration, but not the yield, of milk components. Substitution of blood meal decreased the concentration and yield of milk protein and casein N.  相似文献

Interactions of dietary energy and fat with recombinant bST (sometribove) injections were tested for their effects on ovarian function. Lactating dairy cows were fed one of three diets differing in energy concentration (NEL) and percentage of DM of calcium salts of long-chain fatty acids: diet 1, 1.68 Mcal/kg and 0%; diet 2, 1.68 Mcal/kg and 2.2%; and diet 3, 1.78 Mcal/kg and 2.2%. Cows were injected daily with bST or saline during one of two 19-d interestrous periods (crossover design) in which ovarian follicles were measured by ultrasonography. The bST-treated cows produced more FCM and were in lower energy balance than saline-treated cows. Before d 12 (first follicular wave, estrus = d 0), bST-treated cows had more ovarian follicles in classes 1 (3 to 5 mm; cows on diet 3) or 2 (6 to 9 mm; cows on diets 1 and 2) than saline-treated cows. After d 12 (preovulatory follicular wave), numbers of follicles in different size classes were similar for bST-treated and saline-treated cows, but cows fed diet 2 had larger preovulatory follicles. Lower dietary energy and bST treatment were both associated with larger subordinate follicles. Ovarian follicles less than 10 mm were stimulated by bST, and calcium salts of long-chain fatty acids increased diameters of preovulatory follicles.  相似文献

In an experiment with 405 Hisex white hens rations with 5-20% low glucosinolate rapeseed (RS) or rapeseed meal (RSM) of 00 quality were used. Feed intake, egg production, individual egg weight and live weight gain were reduced in hens fed 5-20% RS and 10-20% RSM. There was a positive correlation between the RS proportion in the ration and the thyroid weight as well as the iodine content of the thyroid. The relation between the iodine content related to one gram of thyroid and the RS proportion was not significant. There were not any significant differences between the RSM groups and the iodine content of the thyroid. The T3 and T4 concentrations in the blood plasma was independent of the RS- or RSM-proportion in the diet. The outward egg quality was not influenced by feeding. The Haugh units were lower when RS or RSM were offered. The fatty acid pattern of the yolk was changed by RS or RSM feeding, particularly the proportion of oleic acid increased. We concluded from the results, RS cannot be recommended for feeding white hens and only a maximum of 5% RSM should be administered.  相似文献

Three semipurified diets containing a low level of fat or 10 percent of either beef tallow or beef tallow free fatty acids were fed to young pigs. Jejunal digesta was sampled 1.5, 2.5, 3.5, and 4.5 h after feeding by aspiration through tubes leading from the jejunal lumen to the exterior. The samples were forced through Millipore filters (1 x 10(-7) m pore size) to separate aqueous phase and oil phase lipid. The total and aqueous phase lipid was separated into triglyceride, monoglyceride, and free fatty acid, and the fatty acid composition of each fraction determined. The concentration of aqueous phase lipid was not influenced by diet, although the concentration of the oil phase lipid was generally higher for the addition of fat to the diets; the increase was greater for the beef tallow free fatty acid diet than for the beef tallow diet. Free fatty acids were the predominant component of the aqueous phase lipid along with some monoglyceride and traces of triglyceride. The major component of the oil phastions of triglyceride and monoglyceride. These must have been derived from endogenously secreted lipid in the case of the tallow fatty acid diet. Thus, the lower digestibility of completely hydrolyzed beef tallow than of conventional beef tallow was not due to an absence of monoglyceride in the intestinal lumen. The proportion of stearic acid in the jejunal digesta was greater than in the dietary lipid, whereas there were lower proportions of palmitic and oleic acids in the jejunal digesta than in the diet; the effect being most pronounced for the tallow free fatty acid diet. The ratio of oleic to palmitic acid in the aqueous phase was less than in the lipid phase suggesting preferential uptake of oleic acid from the micelle by the intestinal mucosa.  相似文献

Four lactating dairy cows that were ruminally and duodenally cannulated were used in an experiment with a 4 x 4 Latin square design to determine the effects of the substitution of neutral detergent fiber (NDF) from forage with NDF from wheat middlings, corn gluten feed, or a blend of distillers dried grains and hominy. Dietary crude protein and NDF averaged 18 and 31%, respectively, for the diet with 71.2% of the NDF from forage (control diet) and for diets with 55% of the NDF from forage (by-product diets). The substitution of NDF from these by-products for forage NDF did not affect dry matter intake (20.1 kg/d) or digestibility of organic matter. Total tract digestibility of acid detergent fiber was lower for cows fed the diet containing a blend of distillers dried grains and hominy than for cows fed the diet containing corn gluten feed. Microbial crude protein synthesis, milk production (23.9 kg/d), and milk fat percentage were similar for all cows, regardless of diet. Cows fed the diets containing wheat middlings or a blend of distillers dried grains and hominy had reduced ruminal pH compared with that of cows fed the diet containing corn gluten feed or the control diet. Diets containing 55% of total NDF from forage with 31% of total NDF from corn gluten feed, wheat middlings, or a blend of distillers dried grains and hominy can supply sufficient effective fiber to maintain normal ruminal function.  相似文献

Thirty-two Holstein cows (8 primiparous) were assigned to negative control or to one of three treatments to assess three forage harvesting and conservation techniques. Forage was harvested as low moisture silage by either a cylinder-type forage harvester, a self-loading forage harvester, or a round baler. Treatment diets were fed from wk 4 to 15 of lactation and consisted of silages harvested by the respective methods supplemented with concentrate at 1.1% of BW. Unsupplemented silage harvested by a cylinder-type forage harvester was used as a control. Daily DMI was higher for cows fed supplemented heap silage cut with either a cylinder-type forage harvester (23.7 kg) or a self-loading forage harvester (22.6 kg) than for cows fed the control (20.0 kg) or supplemented round bale silage (20.1 kg). Milk yield was highest for cows fed supplemented heap silage cut with a cylinder-type forage harvester (26.6 kg/d) and lower for those fed supplemented heap silage cut with a self-loading forage harvester (22.7 kg/d) or the control (20.8 kg/d). Milk composition and digestibilities of DM, N, ADF, and energy were similar among treatments. Postfeeding NEFA concentration decreased more for control cows than for those fed supplemented silage, which was related to greater BW loss. The high milk yield for cows fed supplemented heap silage cut with a cylinder-type forage harvester could be related to a high DMI and low BW gain.  相似文献

Four experiments involving 225 pigs were conducted to assess the efficacy of a microbial phytase (FINASE, Alko Ltd. Biotechnology, Rajam?ki, Finland) produced by Aspergillus niger in corn-soybean meal or dextrose-cornstarch-soybean meal-based diets. In two experiments with growing-finishing pigs, fortified corn-soybean meal diets were formulated to be adequate (.50%) or inadequate (.40 or .30%) in P during the growing phase followed by adequate (.40%) or inadequate (.30%) P in the finishing phase. Mono-dicalcium phosphate was the source of supplemental P. Half the diets were supplemented with phytase (500 phytase units/g). Rate and efficiency of gain and bone breaking strength were decreased when P-deficient diets were fed. Phytase supplementation of the low-P diets restored growth rate and feed:gain to levels that approached those of pigs fed the adequate-P control diet. Bone strength was partially restored to that of the controls. In two additional experiments, pigs were fed low-P basal diets in which all the dietary P came from soybean meal or a corn-soybean meal blend. Both diets contained .05% available P. Graded levels of monosodium phosphate were added to these diets, up to .15% added P, to establish a standard curve. Phytase was added to the basal diet at 250, 500, or 1,000 units/g. Growth rate and bone strength improved linearly (P < .01) with added monosodium phosphate and with increasing levels of supplemental phytase. Based on estimates of total and available P intakes, the highest level of phytase (1,000 units/g) increased the bioavailability of the P from 25% in the soybean meal diet to 57% in the phytase-supplemented diet, and from 15% in the corn-soybean diet to 43% in the phytase-supplemented diet. Expressed on the basis of the improvement in phytate P availability, this level of phytase converted approximately one-third of the unavailable P to an available form. The results indicate that the phytase was efficacious in improving the bioavailability of phytate P for pigs.  相似文献

Our objective was to determine to what extent rate and efficiency of protein gain in finishing cattle can be enhanced by feeding an amino acid-balanced mixture of undegraded intake proteins. The Cornell Net Carbohydrate and Protein System (CNCPS) model was used to formulate a corn-based diet that would meet the rumen requirements for 410-kg large-framed steers with an estrogen implant and fed an ionophore. The CNCPS model was also used to formulate a highly undegradable intake protein (UIP) mixture from meat and bone meal, blood meal, fish meal, and hydrolyzed feather meal to provide the amino acids needed to supplement those derived from microbial protein to better meet amino acid requirements for growth. Four Holstein steers weighing 407 kg were offered a 90:10 concentrate-forage diet at hourly intervals at 95% of ad libitum intake. The steers were injected with 500 microg of estradiol-17beta at 12-h intervals to mimic the effects of an estrogenic implant. Treatments planned consisted of inclusion of the UIP mixture at 0, 2.5, 5, and 7.5% of the diet DM. Dry matter intake was fixed at 6.4 kg/d, and DM digestibility was not significantly affected by varying the amount of UIP addition. Apparent digestibility of N increased (P = .011) from 63.8 to 65.8, 70.7, and 71.5%, the amount of N absorbed increased (P = .001) from 73 to 84, 100, and 106 g/d, and N balance increased (P = .003) from 20 to 30, 33, and 39 g/d when UIP was fed at 0, 2.6, 5.2, and 7.8% of diet DM, respectively. The efficiency of N use increased 39.7%, and biological value increased 31.6% when the UIP mixture was added to the diet. Circulating concentrations of plasma urea N (PUN) were increased (P = .017) from 4.5 for the control diet to 5.7, 6.2, and 6.1 mg/dL when the UIP mixture was added at 2.6, 5.2, and 7.8%, respectively. Corresponding IGF-I concentrations were also increased from 491 to 558 and 624 ng/mL with 2.6 and 5.2% levels of UIP addition. Plasma glucose, NEFA, and insulin concentrations were not affected by feeding the UIP mix. The rate and efficiency of N use for growth improved with addition of an amino acid-balanced UIP mixture to the diet.  相似文献

Experiments were carried out with lactating cows and ewes consisting in the intramammary application of ampicillin-trihydrate with the use of syringe cannules, and with paraffin and vaselin as constituents. Cows were treated with 10 cm3 (500,000 IU) (in the respective quarter of the udder), and ewes--with one half of the syringe content. The milk of the treated animals was sampled prior to manipulation, at the 24th, the 48th, 72nd and 96th hour in order to follow up the time during which the antibiotic was retained in the milk as well as the changes in the cell content and the hydrogen ion concentration. It was found that ampicillin persisted in milk up to the 48th hour in therapeutic concentrations. At the 96th hour the milk proved free of the antibiotic in both cows and ewes and could be offered for consumption or technologic processing.  相似文献

Six semipurified canned diets ranging in composition from 0 to 62% of energy from carbohydrate and from 20% to 48% of energy from protein were fed to female beagle dogs for 8 months. Additionally, three commercial-type diets were also fed. The effects of these diets on growth, body composition and selected blood metabolite levels in the dogs were studied. The dogs readily consumed each of the nine diets fed. The level of carbohydrate, fat or protein in the diet did not influence body weight gain during the first 16 weeks nor was nitrogen balance affected by the diets. At the end of the 32-week study, dogs fed the high-carbohydrate (62% of energy) diet contained less body fat but an equal-free mass, than did dogs fed lower-carbohydrate (20--42% of energy) diets with a similar quantity of protein. Consumption of carbohydrate-free diets did not influence postprandial levels of circulating glucose or insulin in the dogs. Plasma cholesterol levels were elevated in dogs consuming in the diets high in fat but plasma triglyceride levels were not influenced by the diets fed. Consumption of high-protein (46--48% of energy) diets elevated plasma urea nitrogen levels but had minimal influence on plasma amino acid levels. The general response of these young dogs was not markedly influenced by consumption of diets ranging from carbohydrate-free to high-carbohydrate and from adequate-protein to high-protein.  相似文献

Cows were used in an experiment with a 3 x 3 Latin square design to study the effect of corn processing on nutrient flow from the rumen to the duodenum and on ruminal fermentation in lactating cows. Cows were fed diets consisting of 40% forage and 60% concentrate. The only difference in dietary ingredients was the type of processed corn included at 24% of the total diet (as-fed basis). Treatments were dry-rolled corn with a bulk density of 0.54 kg/L and steam-flaked corn with a bulk density of either 0.39 kg/L or 0.31 kg/L. Ruminal fluid pH was not affected by corn processing, but steam-flaking decreased the molar percentage of acetate and increased the molar percentage of propionate. Digestibility of starch in the rumen was not affected by processing. Digestibility of starch entering the duodenum as well as apparent digestibility of starch in the total tract were increased by steam-flaking. Flow of feed and microbial N from the rumen and microbial N efficiency were not affected by grain processing. Yields of milk and milk components as well as composition of milk were not affected by grain processing.  相似文献
