The triglyceride (TG) composition of goat's milk fat (from the milk of five herds collected monthly, from November to May) was studied using AgNO3-TLC and GC–MS. Two of the four fractions obtained by TLC contained trisaturated TGs (SSS) and represented 55% of the total TGs; they were separated by the chain length of short-chain fatty acid (FA). The TGs of the remaining fractions were identified as mono- (SSM) and polyunsaturated, representing 29 and 16% of the total TGs, respectively. The distribution of two SSS fractions by carbon number (CN) was unimodal, with maxima at CN40 and CN36 respectively. The proportions of SSM and polyunsaturated TG were high between CN38–CN44 and CN46–CN54, respectively (mean values, 35 and 52%). One hundred and twenty-four peaks (some containing more that one molecular species of TGs) were detected in the chromatogram of total fat; the sum of the 30 peaks representing >1 mol% was 70% of the total TGs. One hundred and thirty-seven molecular species were identified in the total goat's milk fat: 50% SSS, 30% SSM and 20% polyunsaturated. The most important in quantitative terms were medium-chain TGs containing C8, C10 or C12, and C18 : 1 as unsaturated FA.  相似文献

The objectives of the present study were to evaluate the transfer efficiency of α-linolenic acid (ALA) from the abomasum into milk fat, its interaction with milk fat content and yield, and the relationship between ALA and C16:0 in milk fat. Three rumen-fistulated multiparous Holstein cows at midlactation were used in a 3×3 Latin square design. Treatments consisted of abomasal infusion of (1) 110mL of water/d (control), (2) 110mL of flaxseed oil/d (low flaxseed oil, LFO), and (3) 220mL of flaxseed oil/d (high flaxseed oil, HFO). Experimental periods were continued for 2 wk and fat supplements were infused abomasally during the last 7 d of each period. Average dry matter intake and milk yield were not affected by oil infusion. Milk fat and lactose content tended to be greater with flaxseed infusion compared with the control. Plasma ALA was 2.9- and 4.0-fold greater with LFO and HFO, respectively. The apparent transfer efficiency of ALA to milk was 44.8 and 45.7% with LFO and HFO, respectively. The C16:0 content in milk fat was decreased by 3.59 and 5.25 percentage units, whereas the ALA content was increased by 1.68 and 3.09 percentage units with LFO and HFO, respectively. Similarly, C18:2n-6 was increased by 0.95 and 1.31 percentage units with LFA and HFO, respectively, without changes in other fatty acids (FA). Total polyunsaturated FA was 4.4 and 2.7% lower in the HFO and LFO, respectively, than in the control. Furthermore, C16:0 content in the milk fat was reduced to a greater extent than the increase in ALA content, as a 1.68 and 3.09 percentage unit increase occurred in ALA compared with a 3.6 and 5.25 percentage unit decrease in C16:0 for LFO and HFO, respectively, such that a negative correlation existed between ALA and C16:0 (r=-0.72). In conclusion, abomasal infusion of flaxseed oil dramatically increased the ALA content in plasma and milk fat. Because the replacement of C16:0 with ALA and C18:2n-6 occurred without changes in other FA presumed to be synthesized de novo in the mammary gland, this suggests that the preformed C16:0 was replaced, rather than being caused, by an overall suppression of de novo FA synthesis in the mammary gland.  相似文献

The basis for marine lipid-induced milk fat depression (MFD) has not been established yet, but recent reports suggest the putative contribution of shifts in the ruminal metabolism of α-linolenic acid (ALA). To test this hypothesis, an isotopic tracer approach was used in batch cultures of rumen microorganisms with inoculum collected from cannulated ewes fed either a total mixed ration without lipid supplementation (control inoculum) or the same diet supplemented with 2% of fish oil, which is known to cause MFD in lactating sheep (FO-MFD inoculum). The [1-13C]ALA was added at a dose of 1% of incubated dry matter and the proportions of 13C-labeled fatty acids (FA) were examined after 24 h of incubation, using complementary gas chromatography and gas chromatography-combustion isotope ratio mass spectrometry (GC-C-IRMS) analyses. Expected differences in FA profiles were confirmed between control and FO-MFD inocula (e.g., large decreases in 18:0 and increases in most 18:1 and 18:2 intermediates, particularly trans isomers, to fish oil supply). The biohydrogenation of 13ALA was extensive and yielded multiple metabolites, with a total of 48 chromatographic peaks showing 13C enrichment, regardless of the inoculum type. However, although ALA was biohydrogenated through common pathways under standard or MFD conditions, large changes in the accumulation of 13C-labeled FA suggest important differences in the relative contribution of each specific route. First, increased accumulation of trans-11-containing FA in FO-MFD incubations was accompanied by a general repression of the trans-13/14 pathway (supported by lower trans-13+14 18:1 or trans-11,trans-13 18:2 proportions), together with a lower production of cis FA (e.g., cis-9, -12, and -15 18:1 and some cis,cis 18:2). Results also downplayed the relevance of the trans-11 to trans-10 shift as an effective marker of diet-induced MFD in sheep, and challenged the involvement of some trans-10 intermediates (e.g., trans-10 18:1 and trans-10,cis-15 18:2) in the low-fat milk syndrome in this species. Conversely, increased abundance of most 18:3 intermediates (including some unidentified isomers) might be representative of ruminal alterations related to fish oil supplementation in ewes. Further research is necessary to examine the potential association between these findings and MFD in lactating animals.  相似文献

Metabolism in most organisms can show variations between the day and night. These variations may also affect the composition of products derived from livestock. The aim of the present study was to investigate the difference in composition between the day milk and night milk of dairy cows. Ten multiparous Holstein cows (milk yield = 25.2 ± 5.00 kg/d) were randomly selected during mid lactation. Milk samples were collected at 0500 h (“night milk”) and 1500 h (“day milk”) and analyzed to determine their composition. Mid-infrared spectroscopy was used to analyze macronutrient content of milk. Metabolomics and lipidomics were used to detect and analyze small molecules and fatty acids, respectively. An automatic biochemical analyzer and ELISA kits were used to determine biochemical indicators, as well as antioxidant and immune parameters in the milk. Though milk fat, protein, lactose, and total milk solids were not different between day milk and night milk, small molecules, metabolites and lipids, and hormones and cytokines differed between day milk and night milk. Regarding biochemical and immune-related indicators, the concentrations of malondialdehyde, HSP70, and HSP90 in night milk were lower than that in day milk. However, interferon-γ levels were higher in night milk. Additionally, night milk was naturally rich in melatonin. Lipidomics analyses showed that the levels of some lipids in night milk were higher than those in day milk. Metabolomics analyses identified 36 different metabolites between day milk and night milk. Higher concentrations of N-acetyl-d-glucosamine, cis-aconitate, and d-sorbitol were observed in day milk. However, the other 33 metabolites analyzed, including carbohydrates, lipids, AA, and aromatic compounds, showed lower concentrations in day milk than in night milk. The present findings show that the composition of night milk differs considerably from that of day milk. Notable changes in the circadian rhythm also altered milk composition. These results provide evidence to support the strategic use and classification of day milk and night milk.  相似文献

The intense aroma of goat cheese is mainly due to volatile polar compounds, particularly short- and medium-chain length free fatty acids, contained in its water-soluble extract (WSE). This fraction also contains the taste compounds of the cheese and has been fractionated in order to assess its chemical composition. The aim of the present work was to evaluate the behaviour of the most abundant volatile compounds and fat components in the WSE during multiple successive fractionation steps: i.e. fractionation by 10 and 1 kDa ultrafiltrations and 0.5 kDa nanofiltration. Lipid and volatile components were analysed by TLC-FID and GC–MS methods, respectively, at each filtration step. In the WSE, 43 volatile components were identified and short-chain free fatty acids were the most abundant. Volatile and fat compounds were largely retained during the 10 and 0.5 kDa filtration steps and to a lesser extent during the 1 kDa filtration step. The retention of various volatile compounds could be explained, beside possible interactions with the membranes used, by adsorption or interaction of these compounds with fat or protein material.  相似文献

The phytanic acid content in milk correlates with the amount of green items in the cows’ feed. For this reason, the four-fold methyl-branched fatty acid has been suggested as a potential marker for the authentication of organic milk. In this study, we attempted to provide further support for this idea by studying the progression of the phytanic acid level and diastereomer ratio in milk fat by transition of the diets from high proportions of concentrate (typical “conventional” feed) to hay (typical “organic” feed in winter) and back to “conventional” feed. Milk samples taken from three cows were analyzed on both the phytanic acid concentration and diastereomer distribution. The cows were initially fed with “conventional” feed (ground feed with high portions (30–45?%) of concentrate), then the feed was changed within 1?week to 100?% hay (“organic” feed), and after ~6?weeks, the feed of two cows was changed back to the initial feed with concentrate (phase Cb). During the “conventional” feeding at the beginning of the experiment, the phytanic acid concentration was low (100–130?mg/100?g milk fat). When the feed was changed to hay (“organic” feed), the phytanic acid concentration immediately increased to a stable level of about 160?mg/100?g lipids. Changing back the feed to “conventional” feed, the phytanic acid concentration dropped immediately back below the value measured in the initial phase. Likewise, the SRR/RRR-diastereomer distribution of phytanic acid in the milk was an excellent indicator for the changes in the cows’ feed. While the SRR/RRR-diastereomer ratio was >1.5 during “conventional” feeding, it immediately decreased to equal amounts of both diastereomers when hay was supplied as feed. Abandonment of concentrate in conventional feeding increased the phytanic acid content but the SRR-diastereomer was still dominant and thus the SRR/RRR-diastereomer ratio was different to organic milk. Our results indicate that both parameters, i.e., the phytanic acid content and SRR/RRR-diastereomer ratio need to be measured for authentication of organic milk.  相似文献

The objective of this study was to evaluate the effect of addition of CO(2) to raw milk on UHT milk quality during storage. Control milk (without CO(2) addition) and treated milk (with CO(2) addition up to pH 6.2) were stored in bulk tanks at 4°C for 6d. After storage, both samples were UHT processed using indirect heating (140°C for 5s). Samples were aseptically packed in low-density polyethylene pouches and stored in the dark at room temperature. Raw milk was evaluated upon receipt for physicochemical composition, proteolysis, lipolysis, standard plate count, psychrotrophic bacteria, and Pseudomonas spp. counts, and after 6d of storage for proteolysis, lipolysis, and microbial counts. After processing, UHT milk samples were evaluated for physicochemical composition, proteolysis, and lipolysis. Samples were evaluated for proteolysis and lipolysis twice a month until 120d. Peptides from pH 4.6-soluble N filtrates were performed by reversed-phase HPLC after 1 and 120d of storage. A split-plot design was used and the complete experiment was carried out in triplicate. The results were evaluated by ANOVA and Tukey's test. After 6d of storage, CO(2)-treated raw milk kept its physicochemical and microbiological quality, whereas the untreated milk showed significant quality losses. A significant increase in proteolysis occurred during 120d of storage in both treatments, but the increase occurred 1.4 times faster in untreated UHT milk than in CO(2)-treated UHT milk. In both UHT milks, the proteolysis was a consequence of the action of plasmin and microbial proteases. However, the untreated UHT milk showed higher microbial protease activity than the treated UHT milk. The addition of CO(2) to the raw milk maintained the quality during storage, resulting in UHT milk with less proteolysis and possibly longer shelf life, which is usually limited by age gelation of UHT milk.  相似文献

The viscosity of concentrates (50–55% total solids) prepared from skim milk heated (5 min at 80 or 90 °C) at pH 6.5 and 6.7 was examined. The extent of heat-induced whey protein denaturation increased with increasing temperature and pH. More denatured whey protein and κ-casein were found in the serum phase of milk heated at higher pH. The viscosity of milk concentrates increased considerably with increasing pH at concentration and increasing heating temperature, whereas the distribution of denatured whey proteins and κ-casein between the serum and micellar phase only marginally influenced concentrate viscosity. Skim milk concentrate viscosity thus appears to be governed primarily by volume fraction and interactions of particles, which are governed primarily by concentration factor, the extent of whey protein denaturation and pH. Control and optimization of these factors can facilitate control over skim milk concentrate viscosity and energy efficiency in spray-drying.  相似文献

Milk quality is assessed using bulk milk analysis and by farm audits in the Netherlands. However, the extent of the effect that dairy farm audits have on milk quality is unknown. Data from over 13,000 audits performed on 12,855 dairy farms from February 2006 to April 2008 were merged with laboratory test results of 325,150 bulk milk samples collected 6 mo before and after the audit. A linear mixed model with the method of restricted maximum likelihood was conducted to study whether the total bacterial counts (TBC) of bulk milk were lower during the periods before and after the dairy farm audit. Results showed that TBC values were 2 to 6% lower (i.e., 0.010 to 0.026 log cfu/mL) for a period from 1.5 to at least 6 mo after an audit. Additionally, several variables were significantly associated with bulk milk TBC values: seasonality, total number of attention points (given if some checklist points were not appropriate), audit type, audit result, and the categories milking equipment maintenance, and utility room-tank maintenance. The TBC values increased with a higher level of attention points. Furthermore, the farms rejected based on the audit results had the highest average TBC values and the approved farms had the lowest values. If dairy farms had an overall negative audit assessment and consequently needed a re-audit in the following year, the TBC values of bulk milk were more likely to be higher. Auditing may provide dairy farmers the opportunity to receive advice about factors that influence bulk milk TBC values, for a period of at least 6 mo following the audit.  相似文献

Current knowledge of the genetic polymorphism in ovine milk proteins including heterogeneity detected and their relationships with the technological properties of milk is reviewed. In the casein fraction a great heterogeneity has been determined either by the presence of genetic variants or other factors such as a discrete phosphorylation level and the coexistence of protein forms of different chain length. In the whey fractions, three genetic variants (A, B, and C) are described for β-lactoglobulin (β-LG), and two for α-lactalbumin (α-LA). The topics discussed include the rennetability of ovine milk, cheese yield and susceptibility of β-LG genetic variants to heat denaturation as well as the findings concerning the relationships between ovine milk protein polymorphism and milk production and composition. However, further investigation is needed in order to better outline the relevant features of polymorphism and technological properties of ovine milk.  相似文献

The texture, microstructure and composition of a milk coagulum obtained by coagulating milk with calcium chloride and heat were evaluated. A direct relationship was established between the microstructure and texture of the calcium–milk coagulum. The microstructure was comprised of casein micelles that were interlinked by appendages or fused with each other depending on the nature of the pre-heat treatment employed. The casein micellar network also entrapped fat globules. The pre-heat treatment of milk and the pH of milk during coagulation affected the hardness, adhesiveness, protein content and microstructure of the calcium–milk coagulum significantly. This study characterises a calcium–milk coagulum as a novel dairy product which differs from an acid coagulum in texture, microstructure and composition.  相似文献

The percentages of total, soluble and dialysable calcium of human milk, cow milk and milk and soy based infant formulas were determined in order to detect possible differences in the calcium bioavailability of the samples. For this purpose an in vitro method was applied to these four calcium sources. The ranking of the analysed samples in terms of calcium bioavailability depends on the criteria applied. Calcium ranked dialysis percentage was: cow milk>human milk>soy based formula>milk based formulas. Calcium ranked solubility percentage was: human milk>cow milk>soy-based formula>milk-based formulas. Comparison of the results of the in vitro assay with the information available on in vivo calcium absorption showed that the total soluble calcium contents agree with the in vivo absorption values better than with calcium dialysis percentages.  相似文献

Fluid milk consumption has declined for decades while consumption of nondairy alternatives has increased. A better understanding of why consumers purchase fluid milk or nondairy alternatives is needed to assist increased sales of milk or maintain sales without further decline. The objective of this study was to determine the extrinsic attributes that drive purchase within each product category. The second objective was to determine the personal values behind the purchase of each beverage type to give further understanding why particular attributes are important. An online conjoint survey was launched with 702 dairy consumers, 172 nondairy consumers, and 125 consumers of both beverages. Individual means-end chain interviews were conducted with fluid milk consumers (n = 75), plant-based alternative consumers (n = 68), and consumers of both beverages (n = 78). Fat content was the most important attribute for dairy milk followed by package size and label claims. Consumers of fluid milk preferred 1 or 2% fat content, gallon, or half-gallon packaging, conventionally pasteurized store-brand milk. Sugar level was the most important attribute for plant-based beverages, followed by plant source and package size. Almond milk was the most desirable plant source, and half-gallon packaging was the most preferred packaging. Means-end chain interviews results suggested that maintaining a balanced diet and healthy lifestyle was important to all consumer groups. Lactose free was an important attribute for plant-based alternative consumers and consumers of both dairy and nondairy. A distinguishing characteristic of those who only drank nondairy plant-based alternatives was that plant-based beverages contributed to a goal to consume less animal products, beliefs about animal mistreatment, and perceived lesser effect on the environment than fluid milk. Unique to fluid milk consumers was that fluid milk was perceived as a staple food item. These results suggest that the dairy industry should focus on the nutrition value of milk and educating consumers about misconceptions regarding dairy milk. Future beverage innovation should include the development of lactose-free milk that is also appealing to consumers in flavor.  相似文献

The immune effect of 2′-fucosyllactose (2′FL) in mice fed an infant formula (IF) milk diet was studied. Forty-five BALB/C mice were randomly divided into 3 groups: control, and low and high dose of 2′FL. 2′FL was administered once daily for 30 days and all the mice were fed IF milk diet. On day 30, the spleen and thymus were isolated and weighed for immune organ index calculation. Natural killer (NK) cell activity, T-lymphocyte proliferation capacity and delayed-type hypersensitivity reactivity (DTH) were measured. There was a significant increase in thymus index in high dose 2′FL group (p < 0.05). T-lymphocytes in high dose 2′FL group also tended to proliferate more compared with the control group (p < 0.05). NK cell activity and the degree of DTH were improved in both 2′FL groups (p < 0.05). These data demonstrated that 2′FL can improve immunity in mice fed an IF milk diet.  相似文献

This study was aimed at developing a new functional fermented beverage manufactured with semi-skimmed sheep milk and strawberry pulp (Fragaria × ananassa Duch.) and commercial prebiotic ingredients. We also compared the performance of the yogurt starter cultures and a Lactobacillus plantarum strain (CECT_8328) with potential probiotic properties. We assessed the nutritional profile, bioactivity compounds, viability of lactic acid bacteria during storage, and survival of L. plantarum after in vitro simulated digestion during the storage period. The lactic acid bacteria were viable throughout the storage period, but only L. plantarum maintained good viability after simulated digestion. Nevertheless, neither inulin nor potato starch increased bacterial viability. The fermented semi-skimmed sheep milk strawberry beverages we developed are good sources of minerals and proteins.  相似文献

Replacing fat and saturated fat in baked goods without affecting their quality characteristics is a challenging task. This study evaluated complete and partial substitution of saturated fats (butter and palm oil) by structured emulsions [SE, oil (sunflower) – in – water (fibre-water) emulsion] in cookies by investigating its impact on product quality. Nutritional labelling underlined a drastic reduction in saturated fatty acids [−35% (50% substitution) and −73% (100% substitution)] compared to their conventional counterparts (butter and palm oil). Partial substitution did not markedly affect physicochemical properties, while complete substitution resulted in thinner, harder and darker cookies compared to the controls. Particularly, cookies made with (50:50) structured emulsion – palm oil had the highest sensory scores, and they were perceived as soft, buttery and crunchy. Thus, the use of structured emulsion might be a valuable alternative to develop a potentially healthier product with acceptable sensory properties.  相似文献
