The genetic correlations (ra) of milk lactose percentage (LP), lactose yield (LY), and ratios of LP to other milk solids with udder, metabolic, and fertility disorders have not been assessed in dairy cattle so far. To evaluate the potential role of milk lactose as indicator of cow health, 142,285 lactation records of 84,289 Austrian Fleckvieh cows were analyzed with univariate and bivariate animal models. Milk traits were on a 150-d basis and health traits were coded as binary (0/1). Other than LP and LY, 3 new phenotypes were defined and included in the present study, namely the lactose-to-fat, lactose-to-protein, and lactose-to-solids ratios. The most heritable trait was LP (0.566 ± 0.008) and heritability of LY was much lower (0.145 ± 0.005). Heritability estimates close to 0.50 were assessed for the ratios. The frequency of health disorders was higher in multiparous cows yielding milk with low LP (≤4.553%) compared with cows yielding milk with high LP (≥5.045%). Heritabilities of health traits were in the expected ranges, with the highest estimate for ovarian cysts (CYS; 0.037 ± 0.004) and the lowest for retained placenta (0.005 ± 0.001). Mastitis (MAS) genetically correlated with LY (0.518 ± 0.057); considering that the amount of synthesized lactose is the key regulator of milk volume, this result confirmed that high-producing cows are more genetically susceptible to MAS than low-producing animals. Similar to MAS, ketosis (KET) was also positively genetically associated with LY (0.420 ± 0.077) and a weak and unfavorable ra between KET and lactose-to-protein ratio was estimated (0.159 ± 0.077). The ra of LY with milk fever (MFV) and CYS were approximately 0.20. The ra of LP with MAS, KET, and MFV were negative (?0.142 on average), supporting the idea that LP is a potential health indicator. Genetic correlations between health traits ranged from zero (retained placenta with MAS and CYS) to 0.463 ± 0.090 (MAS and MFV). Results of the present study suggest that LP has potentiality to be used as indicator trait to improve udder health in Austrian Fleckvieh population.  相似文献

Societal pressure to limit the use of antibiotics in livestock production systems, including dairy cattle systems, is consistently increasing. To motivate farmers to reduce antibiotic usage, it is important to understand the factors that determine whether a cow will be treated with antibiotics or not. If farmers' usual practices regarding antibiotic treatments are taken into account, they may be motivated to adopt control measures that can facilitate prudent use of antibiotics and are at the same time cost-effective. In this study, we analyzed database recordings of milk yield and somatic cell count from the routine milk recording scheme, clinical registrations of mastitis and PCR results, and cow factors such as days in milk and parity in relation to antibiotic treatments for 518 dairy herds in Denmark. Farm-wise logistic regressions were used to predict antimicrobial treatment based on these factors. The resulting regression coefficients of 422 herds were further analyzed by principal component analysis and clustering to determine the driving predictors for treatment in different groups of farms. The results showed that determinants that were most important for predicting antibiotic treatments vary from one farm to another. Health indicators such as PCR or somatic cell count were most indicative for treatment on some farms, whereas other groups seemed to depend more on production factors (milk yield) or later culling of the cows. This shows that farmers behave differently and differences can be identified in register data. This information can be considered when developing cost-effective herd-specific control measures of mastitis to promote prudent use of antibiotics in Danish dairy cattle farms.  相似文献

The aim of this study was to estimate genetic parameters for milk urea (MU) content in 3 main Danish dairy breeds. As a part of the Danish milk recording system, milk samples from cows on commercial farms were analyzed for MU concentration (mmol/L) and the percentages of fat and protein. There were 323,800 Danish Holstein, 70,634 Danish Jersey, and 27,870 Danish Red cows sampled with a total of 1,436,580, 368,251, and 133,922 test-day records per breed, respectively, included in the data set. Heritabilities for MU were low to moderate (0.22, 0.18, and 0.24 for the Holstein, Jersey, and Red breeds, respectively). The genetic correlation was close to zero between MU and milk yield in Jersey and Red, and −0.14 for Holstein. The genetic correlations between MU and fat and protein percentages, respectively, were positive for all 3 dairy breeds. Herd-test-day explained 51%, 54%, and 49% of the variation in MU in Holstein, Jersey, and Red, respectively. This indicates that MU levels in milk can be reduced by farm management. The current study shows that there are possibilities to influence MU by genetic selection as well as by farm management.  相似文献

The objective of this study was to estimate heritabilities and repeatabilities for milk coagulation traits [milk coagulation time (RCT) and curd firmness (E30)] and genetic and phenotypic correlations between milk yield and composition traits (milk fat percentage and protein percentage, urea, somatic cell count, pH) in first-lactation Estonian Holstein dairy cattle. A total of 17,577 test-day records from 4,191 Estonian Holstein cows in 73 herds across the country were collected during routine milk recordings. Measurements of RCT and E30 determined with the Optigraph (Ysebaert, Frepillon, France) are based on an optical signal in the near-infrared region. The cows had at least 3 measurements taken during the period from April 2005 to January 2009. Data were analyzed using a repeatability animal model. There was substantial variation in milk coagulation traits with a coefficient of variation of 27% for E30 and 9% for the log-transformed RCT. The percentage of variation explained by herd was 3% for E30 and 4% for RCT, suggesting that milk coagulation traits are not strongly affected by herd conditions (e.g., feeding). Heritability was 0.28 for RCT and 0.41 for E30, and repeatability estimates were 0.45 and 0.50, respectively. Genetic correlation between both milk coagulation traits was negligible, suggesting that RCT and E30 have genetically different foundations. Milk coagulation time had a moderately high positive genetic (0.69) and phenotypic (0.61) correlation with milk pH indicating that a high pH is related to a less favorable RCT. Curd firmness had a moderate positive genetic (0.48) and phenotypic (0.45) correlation with the protein percentage. Therefore, a high protein percentage is associated with favorable curd firmness. All reported genetic parameters were statistically significantly different from zero. Additional univariate random regression analysis for milk coagulation traits yielded slightly higher average heritabilities of 0.38 and 0.47 for RCT and E30 compared with the heritabilities of the repeatability model.  相似文献

The aim of this study was to investigate the association between udder health (UH) status and blood serum proteins (i.e., total protein, albumin, globulin, and albumin-to-globulin ratio) in dairy cows. Blood and milk samples were collected from 1,508 cows of 6 different breeds (Holstein Friesian, Brown Swiss, Jersey, Simmental, Rendena, and Alpine Grey) that were housed in 41 multibreed herds. Bacteriological analysis was performed on milk samples with somatic cell count (SCC) >100,000 cells/mL and bacteria identification was confirmed by multiplex-PCR assays. Milk samples were grouped into 7 clusters of UH status: healthy (cows with milk SCC <100,000 cells/mL and not cultured); culture-negative samples with low, medium, or high SCC; and culture-positive samples with contagious, environmental, and opportunistic intramammary infections. Data of blood serum proteins were analyzed using a linear mixed model that included the fixed effects of stage of lactation, parity, breed, herd productivity (high or low production) and UH status, and the random effect of herd-date within herd productivity. Culture-negative samples with high milk SCC, which were most likely undergoing a strong inflammatory response and whose pathogens could not be isolated because they were engulfed by macrophages or because they had already cleared, and milk samples infected by contagious and environmental bacteria were associated with greater globulin concentrations (and lower albumin-to-globulin ratio) in blood. Variation in blood serum proteins seems to be associated with inflammatory status rather than infection, as serum globulin significantly increased in UH status groups with the highest milk SCC and no differences were observed among intramammary infections pathogens. Blood serum proteins can be a mammary gland inflammation indicator, but cannot be used to differentiate among different UH status groups.  相似文献

The objective of this study was to investigate the ability of a milk line sampling device to obtain a representative sample by comparing SCC and bacterial culture results between milk line and bulk tank samples for milk harvested from the same group of cows at the same milking. A total of 42 paired milk line and bulk tank samples were collected at separate milking events from 21 different herds. Concordance correlation coefficients showed a high level of agreement between the two sample types, with values ranging between 0.74 and 0.99 for all parameters and bacterial species measured. ANOVA showed that SCC and bacterial culture results for Streptococcus agalactiae, Staphylococcus aureus, Streptococcus non-agalactiae, Coliforms, and coagulase-negative staphylococci were neither numerically or statistically different between milk line and bulk tank samples. KAPPA analysis showed that overall agreement beyond chance between milk line and bulk tank samples in determining whether a herd was positive or negative for either Strep. agalactiae or Staph. aureus were 100 and 75%, respectively. While further research is needed to fully assess the utility of this tool for the purpose of bacterial culture, the results of this study suggest that the strategy of milk line sampling is a very promising monitoring tool. This sampling strategy should provide producers with inexpensive and timely information that will help to improve programs for monitoring milk quality and udder health in commercial dairy herds.  相似文献

The performance of different models for genetic analyses of clinical mastitis in Austrian Fleckvieh dual-purpose cows was evaluated. The main objective was to compare threshold sire models (probit and logit) with linear sire and linear animal models using REML algorithm. For comparison, data were also analyzed using a Bayesian threshold sire model. The models were evaluated with respect to ranking of sires and their predictive ability in cross-validation. Only minor differences were observed in estimated variance components and heritability from Bayesian and REML probit models. Heritabilities for probit and logit models were 0.06 and 0.08, respectively, whereas heritabilities for linear sire and linear animal models were lower (0.02). Correlations among ranking of sires from threshold and linear sire models were high (>0.99), whereas correlations between any sire model (threshold or linear) and the linear animal model were slightly lower (0.96). The worst sires were ranked very similar across all models, whereas for the best sires some reranking occurred. Further, models were evaluated based on their ability to predict future data, which is one of the main concerns of animal breeders. The predictive ability of each model was determined by using 2 criteria: mean squared error and Pearson correlation between predicted and observed value. Overall, the 5 models did not differ in predictive ability. In contrast to expectations, sire models had the same predictive ability as animal models. Linear models were found to be robust toward departures from normality and performed equally well as threshold models.  相似文献

The objective of this study was to examine the characteristics of milk samples submitted for microbiological examination at the Wisconsin Veterinary Diagnostic Laboratory between 1994 and 2001. Results (n = 83,650) of microbiological testing of milk samples (n = 77,172) submitted to the Wisconsin Veterinary Diagnostic Laboratory from January 1994 until June 2001 were analyzed. Submissions included milk samples obtained from cases of clinical and subclinical mastitis as well as samples obtained for mastitis surveillance programs. Results were recorded as no growth, contaminated, or identified as specific bacterial pathogens. Statistical analysis was performed to determine trends in the isolation of mastitis pathogens. The proportion of samples identified as contaminated decreased from 20.6 (1997) to 9.5% (2001). The proportion of samples coded as no growth increased from 22.6 (1994) to 49.7% (2001). Isolation of Staphylococcus aureus decreased from 17.7% (1994) of isolates to 9.7% (2001), while isolation of Streptococcus agalactiae decreased from 8.1 (1994) to 3.0% (2001). Coagulase-negative Staphylococcus spp. were isolated from 12.7 to 17.5%, environmental Streptococcus spp. were isolated from 11.6 to 20.1%, and Escherichia coli were isolated from 3.1 to 6.7% of all isolates. No growth and contaminated samples comprised almost 50% of total submissions, and it is important that producers have proper expectations when submitting milk samples. The proportion of isolates identified as Staph. aureus and Strep. agalactiae decreased, suggesting the proportion of contagious bacteria causing mastitis has decreased. Environmental and contagious pathogens demonstrated characteristic differences by season.  相似文献

Electrical conductivity (EC) of milk is an indicator of mastitis. If EC shows genetic variation and is genetically correlated to mastitis, it could be used in a breeding program that includes selection for improved mastitis resistance. In this study, daily records of EC and mastitis from about 1,500 Holstein cows were analyzed. A bivariate animal model was used for estimation of (co)variance components, including fixed effects of age of calving, herd-test-day, and days in milk, in addition to random additive genetic effects and permanent environmental effects. For EC, the estimated heritability was moderate (0.22 to 0.39), whereas for mastitis, the heritability was low (0.013). The genetic correlation between EC and mastitis was estimated to be 0.75, and genetic improvement of mastitis resistance should be feasible through selection for reduced EC.  相似文献

Recent surveys have identified the presence of perchlorate, a natural compound and environmental contaminant, in forages and dairy milk. The ingestion of perchlorate is of concern because of its ability to competitively inhibit iodide uptake by the thyroid and to impair synthesis of thyroid hormones. A recent study established that milk perchlorate concentrations in cattle highly correlate with perchlorate intake. However, there is evidence that up to 80% of dietary perchlorate is metabolized in clinically healthy cows, thereby restricting the available transfer of ingested perchlorate into milk. The influence of mastitis on milk perchlorate levels, where there is an increase in mammary vascular permeability and an influx of blood-derived components into milk, remains unknown. The present study examined the effect of experimentally induced mastitis on milk perchlorate levels in cows receiving normal and perchlorate-supplemented diets. Over a 12-d period, cows were ruminally infused with 1 L/d of water or water containing 8 mg of perchlorate. Five days after the initiation of ruminal infusions, experimental mastitis was induced by the intramammary infusion of 100 μg of bacterial lipopolysaccharide (LPS). Contralateral quarters infused with phosphate-buffered saline served as controls. A significant reduction in milk perchlorate concentration was observed in the LPS-challenged glands of animals ruminally infused with either water or perchlorate. In control glands, milk perchlorate concentrations remained constant throughout the study. A strong negative correlation was identified between mammary vascular permeability and milk perchlorate concentrations in LPS-infused glands. These findings, in the context of a recently published study, suggest that an active transport process is operative in the establishment of a perchlorate concentration gradient across the blood-mammary gland interface, and that increases in mammary epithelial and vascular endothelial permeability lead to a net outflow of milk perchlorate. The overall finding that mastitis results in lower milk perchlorate concentrations suggests that changes in udder health do not necessitate increased screening of milk for perchlorate.  相似文献

The aim of this study was to estimate heritabilities of rennet coagulation time (RCT) and curd firmness (a30) and their genetic correlations with test-day milk yield, composition (fat, protein, and casein content), somatic cell score, and acidity (pH and titratable acidity) using coagulating and noncoagulating (NC) milk information. Data were from 1,025 Holstein-Friesian (HF) and 1,234 Brown Swiss (BS) cows, which were progeny of 54 HF and 58 BS artificial insemination sires, respectively. Milk coagulation properties (MCP) of each cow were measured once using a computerized renneting meter and samples not exhibiting coagulation within 31 min after rennet addition were classified as NC milk. For NC samples, RCT was unobserved. Multivariate analyses, using Bayesian methodology, were performed to estimate the genetic relationships of RCT or a30 with the other traits and statistical inference was based on the marginal posterior distributions of parameters of concern. For analyses involving RCT, a right-censored Gaussian linear model was used and records of NC milk samples, being censored records, were included as unknown parameters in the model implementing a data augmentation procedure. Rennet coagulation time was more heritable [heritability (h2) = 0.240 and h2 = 0.210 for HF and BS, respectively] than a30 (h2 = 0.148 and h2 = 0.168 for HF and BS, respectively). Milk coagulation properties were more heritable than a single test-day milk yield (h2 = 0.103 and h2 = 0.097 for HF and BS, respectively) and less heritable than milk composition traits whose heritability ranged from 0.275 to 0.275, with the only exception of fat content of BS milk (h2 = 0.108). A negative genetic correlation, lower than −0.85, was estimated between RCT and a30 for both breeds. Genetic relationships of MCP with yield and composition were low or moderate and favorable. The genetic correlation of somatic cell score with RCT in BS cows was large and positive and even more positive were those of RCT with pH and titratable acidity in both breeds, ranging from 0.80 to 0.94. Including NC milk information in the data affected the estimated correlations and decreased the uncertainty associated with the estimation process. On the basis of the estimated heritabilities and genetic correlations, enhancement of MCP through selective breeding with no detrimental effects on yield and composition seems feasible in both breeds. Milk acidity may play a role as an indicator trait for indirect enhancement of MCP.  相似文献

Automatic milking (AM) is increasing in modern dairy farming, and over 8,000 farms worldwide currently use this technology. Automatic milking system is designed to replace conventional milking managed by a milker in a milking parlor or in tie stalls. Cows are generally milked more frequently in AM than in conventional milking, and milking is quarter-based instead of udder-based. Despite improvements in the milking process and often building of a new barn before the introduction of AM, udder health of the cows has not improved; on the contrary, problems may appear following conversion from conventional milking to AM. This review focuses on udder health of dairy cows in AM, and we discuss several aspects of cow and milking management in AM associated with udder health. Finally, adequate management methods in AM are suggested. According to several studies comparing udder health between automatic and conventional milking or comparing udder health before and after the introduction of automatic milking in the same herds, udder health has deteriorated during the first year or more after the introduction of AM. Automatic detection of subclinical and clinical mastitis and cleaning the teats before milking are challenges of AM. Failures in mastitis detection and milking hygiene pose a risk for udder health. These risk factors can partly be controlled by management actions taken by the farmer, but AM also needs further technical development. To maintain good udder health in AM, it is imperative that the barn is properly designed to keep the cows clean and the cow traffic flowing. Milking frequency must be maintained for every cow according to its stage of lactation and milk production. Careful observation of the cows and knowledge of how to use all data gathered from the system are also important. “Automatic” does not mean that the role of a competent herdsman is in any way diminished.  相似文献

Milk production per cow has increased significantly as a result of breeding, feeding, and other management factors. This study aims to address concerns about udder health risks for low- and high-producing dairy cows. In a 2 × 2 × 2 factorial design, Holstein-Friesian heifers (n = 100) of low or high genetic merit for milk production, milked 2 or 3 times a day, and fed a mixed ration with low or high energy content, were compared during the first 14 wk of lactation. Milk composition and cell counts were determined weekly; quarter milk samples for bacteriology were taken in wk 2, 8, and 14; and teat condition was scored in wk 2, 6, 10, and 14 during the experiment. The experimental factors resulted in substantial differences in milk production between treatment groups (24.1 for low vs. 25.6 kg/d for high genetic merit; 23.3 for 2 times vs. 26.5 kg/d for 3 times daily milking; and 20.9 for low-energy ration vs. 29.0 kg/d for high-energy ration). Ration composition was the most important determining factor for milk production, but did not affect cell counts or intramammary bacterial infections, although cows that received low-energy rations had rougher teat ends than cows receiving high-energy rations. This indicates that high production itself is not a major risk factor for udder health in the first lactation. A higher milking frequency impaired teat condition and improved cell counts in general, but did not clearly influence bacteriological status. High genetic merit was related to higher cell count, more Staphylococcus, and less Bacillus and other environmental pathogens in cultures and did not affect teat condition. The effects of milking frequency and feeding on udder health were similar for cows with high and low genetic merit. Genetic selection on milk production, without taking udder health into account, reduces udder health. As a result, maintaining udder health will require increasing the skills and time of dairy farmers who have to divide their attention to more cows when farm sizes increase, or selection should put more emphasis on udder health traits.  相似文献

Control (CL) and select line (SL) dairy cows (n = 22) managed identically but differing in milk yield (>4100 kg/305 d) were used to determine differences in milk fatty acid profile as lactation progressed. Milk yield was recorded daily and milk samples were collected during wk 1, 4, 8, 12, and 16 postpartum for milk composition analysis. Milk samples from wk 1, 8, and 16 were also analyzed for fatty acid composition. Select-line cows produced more milk (44.4 vs. 31.2 kg/d) and milk components than CL cows during the 16-wk period. There was no difference in rate of milk yield increase, but peak milk yield for SL cows was greater and occurred later in lactation. There were no differences in milk SCC or milk fat, protein, or lactose content. Selection for milk yield did not affect the content of most individual milk fatty acids; however, compared with CL, SL cows had a reduced Delta(9)-desaturase system and tended to produce milk with lower monounsaturated fatty acid content. Selection for milk yield did not affect milk fatty acid origin but the percentage of de novo fatty acids increased and preformed fatty acids decreased as lactation progressed. Milk fat trans-11 18:1 and cis-9,trans-11 conjugated linoleic acid increased with progressing lactation (10.7 vs. 14.1 and 3.1 vs. 5.4 mg/g, or 31 and 76%, respectively) and were correlated strongly among wk 1, 8, and 16 of lactation. Temporal changes in the Delta(9)-desaturase system occurred during lactation but these changes were not correlated with milk fat cis-9,trans-11 conjugated linoleic acid content. Results indicate prolonged genetic selection for milk yield had little effect on milk fatty acid composition, but milk fatty acid profiles varied markedly by week of lactation.  相似文献

The aim of this study was to estimate genetic parameters for test-day milk urea nitrogen (MUN) and its relationships with milk production traits. Three test-day morning milk samples were collected from 1,953 Holstein-Friesian heifers located on 398 commercial herds in the Netherlands. Each sample was analyzed for somatic cell count, net energy concentration, MUN, and the percentage of fat, protein, and lactose. Genetic parameters were estimated using an animal model with covariates for days in milk and age at first calving, fixed effects for season of calving and effect of test or proven bull, and random effects for herd-test day, animal, permanent environment, and error. Coefficient of variation for MUN was 33%. Estimated heritability for MUN was 0.14. Phenotypic correlation of MUN with each of the milk production traits was low. The genetic correlation was close to zero for MUN and lactose percentage (−0.09); was moderately positive for MUN and net energy concentration of milk (0.19), fat yield (0.41), protein yield (0.38), lactose yield (0.22), and milk yield (0.24), and percentage of fat (0.18), and percentage of protein (0.27); and was high for MUN and somatic cell score (0.85). Herd-test day explained 58% of the variation in MUN, which suggests that management adjustments at herd-level can reduce MUN. This study shows that it is possible to influence MUN by herd practice and by genetic selection.  相似文献

In this study, we investigated associations among subclinical intra-mammary infection (IMI) and quarter-level milk composition, udder health indicators, and cheesemaking traits. The dataset included records from 450 Holstein cows belonging to three dairy herds. After an initial screening (T0) to identify animals infected by Streptococcus agalactiae, Streptococcus uberis, Staphylococcus aureus, and Prototheca spp., 613 quarter milk samples for 2 different sampling times (T1 and T2, 1 mo after T1) were used for analysis. Milk traits were analyzed using a hierarchical linear mixed model including the effects of days in milk, parity and herd, and bacteriological and inflammatory category [culture negative with somatic cell count (SCC) <200,000 cells/mL; culture negative with SCC ≥200,000 cells/mL; or culture positive]. All udder health indicators were associated with increased SCC and IMI at both sampling times. The largest effects were detected at T2 for milk lactose (?7% and ?5%) and milk conductivity (+9% and +8%). In contrast, the increase in differential SCC (DSCC) in samples with elevated SCC was larger at T1 (+17%). Culture-negative samples with SCC ≥200,000 cells/mL had the highest SCC and greatest numbers of polymorphonuclear-neutrophils-lymphocytes and macrophages at both T1 and T2. Regarding milk cheesemaking ability, samples with elevated SCC showed the worst pattern of curd firmness at T1 and T2. At T2, increased SCC and IMI induced large decreases in recoveries of nutrients into the curd, in particular recovered protein (?14% and ?16%) and recovered fat (?12% and ?14%). Different behaviors were observed between Strep. agalactiae and Prototheca spp., especially at T2. In particular, samples that were positive for Strep. agalactiae had higher proportions of DSCC (+19%) compared with negative samples with low SCC, whereas samples that were positive for Prototheca spp. had lower DSCC (?11%). Intramammary infection with Prototheca spp. increased milk pH compared with culture-negative samples (+3%) and negative samples that had increased SCC (+2%). The greatest impairment in curd firmness at 30 min from rennet addition was observed for samples that were positive for Prototheca spp. (?99% compared with negative samples, and ?98% compared with negative samples with high SCC). These results suggest that IMI caused by Prototheca spp. have detrimental effects on milk technological traits that deserve further investigation of the mechanisms underlying animals' responses to infection.  相似文献

The aim of this study was to propose the survival analysis technique as a statistical approach for the analysis of rennet coagulation time (RCT) able to make use of coagulating and noncoagulating (NC) milk information in order to estimate potential sources of variations that affects RCT. A total of 1,025 Italian Holstein-Friesian cows (HF; progeny of 54 sires) and 1,234 Brown Swiss cows (BS; progeny of 58 sires) reared in 34 and 38 herds, respectively, were milk-sampled once. Rennet coagulation time was analyzed with a semiparametric proportional hazard model (i.e., a Cox model), with the NC samples considered as censored records. Furthermore, a different censoring scenario, with a new end point at 18 min, was considered after the rearrangement of the time space originally used for the observation of RCT. The percentage of NC samples was almost 10% for HF and 3.5% for BS cows in in the 31 min set, whereas it increased to 44 and 24.9%, respectively, in the 18 min set. Estimated hazard ratios indicated that the most important factors affecting the coagulation process were herd, days in milk, casein number, and milk acidity (expressed in terms of titratable acidity) for both HF and BS, whereas the SCS was relevant only for BS. The survival model seems to be particularly suitable for this analysis, as it can properly account for censored and uncensored records and appropriately use all available information. Moreover, this methodology allows us to rearrange the time space used for the observation of RCT and to define alternative traits (i.e., RCT with an end point at 18 min). Our restriction of the time space and the increased percentage of censored records did not highlight any substantial differences in terms of the risk of coagulating with respect to the traditional 31 min testing time. Although further research is needed to investigate the effect of these sources of variation on cheese yield, our results indicate that casein number, acidity, and SCS may be used as indicator traits for enhancing the technological properties of bovine milk.  相似文献

International genetic bull evaluations of somatic cell counts (SCC) from 8 different Holstein populations and clinical mastitis from 3 of these populations were inferred simultaneously using a multiple-trait-multiple-country evaluation (MT-MACE) model. This model considered effective independent weighting factors and multivariately deregressed national genetic evaluations for countries with multiple-trait national models. Predictions of genetic merit from MT-MACE and their reliabilities were compared with the corresponding results from 2 separate single-trait-multiple-country evaluations (ST-MACE) for different groups of bulls. The assumed heritabilities for clinical mastitis (h2 = 0.02 to 0.05) were substantially lower than the heritabilities for SCC (h2 = 0.08 to 0.27). The predictive ability of MT-MACE was essentially equal to or better than the predictive ability of ST-MACE for all country-trait combinations, but both methods yielded effectively unbiased and consistent consecutive predictions (correlation > 0.93). Both sets of predictions also agreed well with future national genetic evaluations for bulls receiving additional daughter information (correlation > 0.96), except for evaluations for which within-country correlations were utilized internationally, but not nationally (correlation = 0.86 to 0.97). The reliabilities for MT-MACE were essentially equal to or higher than reliabilities for ST-MACE, depending on the trait and group of bulls in question. Reliabilities increased most for young bulls, and for clinical mastitis in countries that did not use the within-country correlations with SCC in the national evaluation (up to a 23% increase in average reliability).  相似文献

Mastitis affects a high proportion of dairy cows and is still one of the greatest challenges faced by the dairy industry. Staphylococcal bacteria remain the most important cause of mastitis worldwide. We investigated how distinct staphylococcal species evade some critical host defense mechanisms, which may dictate the establishment, severity, and persistence of infection and the outcome of possible therapeutic and prevention interventions. Thus, the present study investigated variations among distinct bovine-associated staphylococci in their capability to resist phagocytosis and to trigger respiratory burst activity of blood and milk polymorphonuclear neutrophil leukocytes (PMNL) in dairy cows. To do so, PMNL of 6 primiparous and 6 multiparous dairy cows were used. A collection of 38 non-aureus staphylococci (NAS) and 12 Staphylococcus aureus were included. The phagocytosis and intracellular reactive oxygen species (ROS) production by blood and milk PMNL were analyzed by flow cytometry. Phagocytosis, by both blood and milk PMNL, did not differ between S. aureus and NAS as a group, although within-NAS species differences were observed. Staphylococcus chromogenes (a so-called milk-adapted NAS species) better resisted phagocytosis by blood PMNL than the so-called environmental (i.e., Staphylococcus fleurettii) and opportunistic (i.e., Staphylococcus haemolyticus) NAS species. Otherwise, S. haemolyticus was better phagocytosed by blood PMNL than S. aureus, S. fleurettii, and S. chromogenes. No influence of the origin of the isolates within the staphylococci species in the resistance to phagocytosis by blood and milk PMNL was found. Overall, both S. aureus and NAS did not inhibit intracellular ROS production in blood and milk PMNL. Non-aureus staphylococci induced fewer ROS by milk PMNL than S. aureus, which was not true for blood PMNL, although species-specific differences in the intensity of ROS production were observed. Staphylococcus chromogenes induced more blood PMNL ROS than S. fleurettii and S. haemolyticus, and as much as S. aureus. Conversely, S. chromogenes induced fewer milk PMNL ROS than S. aureus. The origin of the isolates within the staphylococci species did not affect the ROS production by blood and milk PMNL. In conclusion, our study showed differences in staphylococci species in evading phagocytosis and triggering ROS production, which may explain the ability of some staphylococci species (i.e., S. aureus and S. chromogenes) to cause persistent infection and induce inflammation.  相似文献

The aim of the present study was to assess genetic variation and heritability of a novel indicator of udder health, milk differential somatic cell count (DSCC), which represents the percentage of neutrophils plus lymphocytes in the total somatic cell count (SCC). Furthermore, we estimated genetic and phenotypic correlations of DSCC with other milk traits routinely measured in Italian Holstein cows. Besides DSCC, test-day data included milk yield, composition traits (i.e., fat, protein, casein, and lactose percentages), pH, milk urea nitrogen, and SCC. After editing, the final data set included 10,709 test-day records of 5,142 cows in 299 herds. Mean of DSCC was 62.07%, which means that macrophages were approximately 38% of total SCC. Comparing our results with the literature offered compelling evidence of the importance of acquiring information about the proportion of the different cell types in milk to better define the udder health status. In addition, our analysis revealed, for the first time, that DSCC is a heritable trait, and heritability (0.08 ± 0.02) was higher than that of traditional somatic cell score (0.04 ± 0.02). Nevertheless, heritability of DSCC is still low compared with milk yield and quality traits. Single-trait analysis within parity showed that DSCC was less heritable in primiparous than in multiparous cows, whereas bivariate analysis confirmed that DSCC and somatic cell score were 2 different traits, as their genetic and phenotypic correlations differed from unity. From a genetic point of view, the DSCC was positively weakly associated with milk yield, lactose percentage, and milk urea nitrogen, and negatively associated with pH. Our findings contributed to the understanding of the genetic background of DSCC and are a precursor to the potential use of DSCC in breeding programs to enhance cow resistance to mastitis. However, further research is needed to determine the weight this novel trait should receive in a selection program aimed to reduce udder health problems.  相似文献
