Objectives were to determine the effects of an injectable formulation of calcitriol on Ca concentration, risk of clinical diseases, and performance in dairy cows. Cows were blocked by lactation number (1 vs. >1) and calving sequence and, within block, assigned randomly within 6 h of calving to receive subcutaneously vehicle only (CON, n = 450) or 200 (CAL200, n = 450) or 300 μg of 1α,25-dihydroxyvitamin D3 (CAL300, n = 450). Cows were fed the same acidogenic diet prepartum. Blood was sampled before treatment administration and again during the first 11 d postpartum and analyzed for concentrations of ionized Ca (iCa), total Ca (tCa), Mg (tMg), and P (tP), β-hydroxybutyrate, carboxylated osteocalcin (cOC), and undercarboxylated osteocalcin (uOC). Cows were evaluated for diseases in the first 60 d postpartum. Reproduction and survival were monitored for the first 300 d postpartum. Calcitriol increased concentration of blood iCa (CON = 1.12 vs. CAL200 = 1.23 vs. CAL300 = 1.27 mM), plasma tCa (CON = 2.29 vs. CAL200 = 2.44 vs. CAL300 = 2.46 mM), and plasma tP (CON = 1.72 vs. CAL200 = 2.21 vs. CAL300 = 2.28 mM), and differences were observed during the first 5 d postpartum for iCa and tCa, and the first 7 d postpartum for tP. Concentrations of tMg were lower in calcitriol-treated cows than in CON cows (CON = 0.81 vs. CAL200 = 0.78 vs. CAL300 = 0.75 mM), and differences were observed during the first 5 d postpartum. Calcitriol increased plasma concentrations of cOC (CON = 14.5 vs. CAL200 = 23.0 vs. CAL300 = 19.8 ng/mL) and uOC (CON = 1.6 vs. CAL200 = 3.4 vs. CAL300 = 2.6 ng/mL). Prevalence of subclinical hypocalcemia was less in calcitriol-treated cows (CON = 19.0 vs. CAL200 = 4.7 vs. CAL300 = 9.3%); however, benefits on health were only observed in overconditioned cows (n = 270/1,350). Calcitriol reduced incidence of retained placenta (CON = 14.3 vs. CAL200 = 5.1 vs. CAL300 = 5.9%), puerperal metritis (CON = 12.7 vs. CAL200 = 6.1 vs. CAL300 = 2.5%), and morbidity (CON = 72.1 vs. CAL200 = 57.4 vs. CAL300 = 56.9%) in cows with BCS greater than 3.50, but no benefit on health was observed in cows with BCS equal to or less than 3.50 at parturition. Milk yield did not differ among treatments. Pregnancy at first AI did not differ, but pregnancy rate after the first AI was slower for calcitriol-treated cows because of reduced insemination rate and pregnancy per AI. We found that CAL200 reduced death but increased culling in cows without calving problems. Collectively, results indicate that treatment with calcitriol at parturition was effective in improving concentrations of iCa, tCa, and tP, which reduced the risk of hypocalcemia. Pregnancy rate was reduced by calcitriol treatment, and benefits on health performance were limited to overconditioned cows. Thus, treatment of all cows is not supported, and proper identification of cohorts of cows that benefit from postpartum interventions that increase blood calcitriol or calcium is needed.  相似文献

The objective of this study was to evaluate the effect of 2 subcutaneous injections of a multimineral preparation, each containing 60 mg of zinc, 10 mg of manganese, 5 mg of selenium, and 15 mg of copper at 3 and 30 d after birth on immunity, health, and growth of dairy calves during the preweaning period. The study was conducted in upstate New York in 2 commercial dairy farms. A total of 790 Holstein heifer calves were randomly allocated at birth into 1 of 2 treatments: trace mineral supplement (TMS) treated or control. Blood samples were collected at 3, 14, and 35 d after birth to evaluate glutathione peroxidase (GPx) activity, superoxide dismutase (SOD) activity, haptoglobin, and neutrophil and monocyte function. Incidence of diseases and average daily gain was evaluated in the first 50 d of life. At 14 d of life, TMS-treated calves had increased neutrophil activity compared with control calves. Moreover, TMS-treated calves had greater GPx activity on d 14 after birth than control calves. The TMS treatment reduced the incidence of diarrhea (TMS = 41.7% vs. control = 49.7%) and combined incidence of pneumonia or otitis or both (TMS = 41.7% vs. control = 49.1%). Additionally, GPx was greater for calves diagnosed with otitis at d 35 after birth. However, calves diagnosed with pneumonia had decreased GPx activity at d 35 after birth. Serum SOD and haptoglobin concentrations were not affected by treatment or disease. Moreover, no effects were observed on average daily gain and survivability between TMS-treated and control calves during the preweaning period. Supplementation with trace minerals at 3 and 30 d of life increased neutrophil function and GPx activity and reduced the incidence of health disorders.  相似文献

Two hundred fifty multiparous and primiparous cows were assigned to a study at approximately 70 d prepartum to determine the effect of trace mineral source on lactation performance, claw integrity, and fertility. Cows received treatments from 3 wk prepartum through wk 35 postpartum. Treatments consisted of 1) all supplemental Zn, Mn, Cu, and Co provided in sulfate form (Sulfate) and 2) 360 mg of Zn, 200 mg of Mn, 125 mg of Cu, and 12 mg of Co supplied daily by Sulfate minerals replaced with similar amounts of minerals supplied by Availa-4 (CTM). Individuals involved with daily animal care or data recording, or both, were blinded to treatment assignments. Cows from all treatments were housed in common pens, and treatments were dispensed to cows via a computerized feeder. All claws of cows were examined before treatment administration and at 16 and 36 wk postpartum by personnel trained in identifying claw lesions. Cows fed the CTM diet tended to produce more milk and energy-corrected milk than cows fed the Sulfate diet. Cows fed the CTM diet also produced more milk protein and solids (fat + protein) than cows fed the Sulfate diet. Replacing Sulfate minerals with those supplied by CTM decreased incidence of sole ulcers at wk 36 postpartum and tended to decrease incidence of interdigital dermatitis at wk 16 and 36 postpartum. Severity of heel erosion tended to be less for cows fed CTM than cows receiving the Sulfate diet. Despite first service conception rates tending to be greater for cows fed the Sulfate diet, there was no effect of treatment on rate of conception. A greater percentage of cows fed the Sulfate diet tended to be culled from the herd before wk 36 postpartum than cows fed the CTM diet. Replacing Sulfate minerals with CTM resulted in improved lactation performance and claw integrity.  相似文献

A total of 825 dairy cows from a commercial dairy farm in central California were used to evaluate effects of one or 2 doses of an injectable trace mineral supplement containing 20 mg/mL of zinc, 20 mg/mL of manganese, 5 mg/mL of selenium, and 10 mg/mL of copper on first-service conception rate. Cows were randomly allocated into treatment or control group to either a single dose (experiment 1) or a double dose (experiment 2) of injected supplement. Allocation was based on days in lactation for experiment 1 and the length of gestation periods for experiment 2. In experiment 1, cows 38 to 45 d in lactation (n = 190) received a single dose of 5 mL of injected supplement. Similar cows were used as controls (n = 227). In experiment 2, cows and pregnant heifers received an initial injection of 5 mL of the mineral supplement from 2 to 3 wk precalving (n = 186). An equal dose was repeated 38 to 45 d in lactation. A similar group of cows and pregnant heifers served as controls for experiment 2 (n = 222). Health and reproductive events postcalving were recorded. In experiment 1, the odds of first-service conception were not significantly different for cows receiving a one-dose regimen of minerals compared with untreated control cows; conception rates were 26.8 and 27.5% for experiment 1 treatment and control groups, respectively. In experiment 1, the odds of first-service conception were significantly lower (odds ratio = 0.66) for cows and heifers given the 2-dose regimen of minerals compared with untreated controls; overall conception rates were 21.5 and 31.5% for experiment 2 treatment and control groups, respectively. In this intensively managed dairy herd, a single dose of injected trace minerals before breeding had no beneficial effects on first-service conception rate. However, dairy cows receiving a dose of trace minerals before calving and another dose before breeding had lower conception at first service.  相似文献

The objectives were to study the effects of induced subclinical hypocalcemia [SCH, blood ionized Ca (iCa2+) <1.0 mM, without recumbency] on physiological responses and function of immune cells in dairy cows. Ten nonpregnant, nonlactating Holstein cows were blocked by lactation and assigned randomly to a normocalcemic (NC; intravenous infusion of 0.9% NaCl i.v. plus 43 g of oral Ca, as Ca sulfate and Ca chloride, at −1 and 11 h) or an induced SCH [SCHI, 5% ethylene glycol tetraacetic acid (EGTA), a selective iCa2+ chelator, intravenous infusion] treatment for 24 h, using a crossover design. The sequence of treatments was either NC–SCHI or SCHI–NC, with a 6-d washout period. Ionized Ca was evaluated before, hourly during the infusion period, and at 48 and 72 h, to monitor concentrations and adjust the rate of infusion, maintaining blood iCa2+ <1.0 mM in SCHI throughout the 24-h infusion period. Additional measurements included heart and respiratory rates, rectal temperature, dry matter intake, rumen contractions, whole-blood pH, concentrations of glucose and K in whole blood, concentrations of total Ca, Mg, nonesterified fatty acids, β-hydroxybutyrate, and insulin in plasma, and urinary excretion of Ca. Total and differential leukocyte count in blood was also performed. The concentration of cytosolic iCa2+ in neutrophils and lymphocytes was quantified and neutrophil function was assayed in vitro. Infusion of a 5% EGTA solution successfully induced SCH in all SCHI cows, resulting in decreased blood iCa2+ concentrations throughout the 24-h treatment period (0.77 ± 0.01 vs. 1.26 ± 0.01 mM iCa2+). Induction of SCH reduced dry matter intake on the day of infusion (5.3 ± 0.8 vs. 9.1 ± 0.8 kg/d) and rumen contractions (1.9 ± 0.2 vs. 2.7 ± 0.2 contractions/2 min) for the last 12 h of infusion. Cows in SCHI had decreased plasma insulin concentration (1.44 ± 0.23 vs. 2.32 ± 0.23 ng/mL) evident between 6 and 18 h after the beginning of the infusion, accompanied by increased concentrations of glucose (4.40 ± 0.04 vs. 4.17 ± 0.04 mM). Plasma nonesterified fatty acids concentration was greater for SCHI than NC cows (0.110 ± 0.019 vs. 0.061 ± 0.014 mM). Neutrophils of cows in SCHI had a faster decrease in cytosolic iCa2+ after stimulation with ionomycin (9.9 ± 1.0 vs. 13.6 ± 1.4 Fluo-4:Fura Red post-end ratio) in vitro. Furthermore, induction of SCH reduced the percentage of neutrophils undergoing phagocytosis (22.1 ± 2.1 vs. 29.3 ± 2.1%) and reduced the oxidative burst response after incubation of pathogenic bacteria (16.1 ± 1.7 vs. 24.2 ± 1.7%). Subclinical hypocalcemia compromised appetite, altered metabolism, and impaired function of immune cells in dairy cows.  相似文献

Paraoxonase (PON) is a liver protein with hydrolase activity that is released into the blood stream. Paraoxonase may serve as an index of liver function because it is drastically reduced in chronic liver damage. Sixty-seven periparturient dairy cows were used to evaluate the relationship between plasma PON, health problems, inflammatory conditions, and liver function. Baseline plasma PON concentrations during the first 30 d in milk (DIM) were retrospectively used to group cows into quartiles. Metabolic profile, lipid metabolites (e.g., nonesterified fatty acids, β-hydroxybutyrate), inflammatory indices (haptoglobin, ceruloplasmin), low and high density lipoprotein cholesterol, vitamin A, vitamin E, reactive oxygen metabolites, total antioxidants, and PON in plasma were measured 2 wk before to 8 wk after calving. Weekly milk yield, body condition score, and all health problems were recorded. After parturition (7 DIM), cows in the lower PON group had the lowest plasma concentrations of negative acute phase proteins compared with the higher PON group for retinol binding protein (23.2 ± 2.86 vs. 36.0 ± 2.96 μg/dL of vitamin A), albumin (31.6 ± 0.73 vs. 33.9 ± 0.75 g/ L), total cholesterol (2.04 ± 0.30 vs. 2.45 ± 0.42 mmol/ L), and the highest concentrations of haptoglobin (0.67 vs. 0.24 ± 0.03 g/L; positive acute phase protein) and globulins (37.2 vs. 32.3 ± 1.4 g/L). Plasma bilirubin was highest in the cows (10.1 vs. 6.2 ± 0.6 μmol/L) in the lowest PON quartile. Plasma PON was negatively correlated with haptoglobin (r = −0.39) and bilirubin (r = −0.42) and positively correlated with retinol binding protein (r = 0.54), albumin (r = 0.38), and cholesterol (r = 0.55) fractions. A total of 82.3% of cows in the lower quartile and no cows in the upper quartile experienced serious inflammation. Lower quartile cows produced 28.1 ± 10.3 kg of milk/d; whereas upper quartile cows produced 38.3 ± 7.7 kg of milk/d during the first 30 DIM. A reduction in the ability of the liver to cope with the increased metabolic demand near parturition in dairy cows can be diagnosed using changes in baseline plasma PON.  相似文献

The objectives of this experiment were to determine the effects of supplementing 25-hydroxyvitamin D3 (calcidiol, CAL) compared with vitamin D3 (cholecalciferol, CHOL) at 1 or 3 mg/d in late gestation on production outcomes of dairy cows. One hundred thirty-three parous and 44 nulliparous pregnant Holstein cows were enrolled in the experiment. Cows were blocked by parity and previous lactation milk yield (parous) or genetic merit (nulliparous) and assigned randomly to receive 1 or 3 mg/d of CAL or CHOL in a 2 × 2 factorial arrangement of treatments (CAL1, CAL3, CHOL1, and CHOL3). Treatments were provided to individual cows as a top-dress to the prepartum diet from 250 d in gestation until parturition. The prepartum diet had a dietary cation-anion difference of ?128 mEq/kg of dry matter. Production and disease were evaluated for the first 42 d in milk, and reproduction was evaluated to 300 d in milk. Incidence of postpartum diseases did not differ among treatments. Feeding CAL compared with CHOL increased yields of colostrum and colostrum fat, protein, and total solids, resulting in an increased amount of net energy for lactation secreted as colostrum (CHOL = 7.0 vs. CAL = 9.0 ± 0.7 Mcal). An interaction between source and amount was observed for milk yield: CAL3 increased milk yield compared with CHOL3 (CHOL3 = 34.1 vs. CAL3 = 38.7 ± 1.4 kg/d) but milk yield did not differ between CAL1 and CHOL1 (CHOL1 = 36.9 vs. CAL1 = 36.4 ± 1.4 kg/d). Concentrations of serum calcidiol on day of calving and average serum Ca from d 2 to 11 postpartum were positively associated with milk yield in the first 42 d in milk. Interactions between source and amount of vitamin D were also observed for pregnancy after first AI: the percentage of cows receiving CHOL1 and CAL3 that became pregnant was smaller than that of cows receiving CHOL3 and CAL1. However, pregnancy per AI and pregnancy by 300 d in milk did not differ among treatments. Overall, CAL3 increased milk yield compared with CHOL3, whereas in cows fed 1 mg/d (CAL1 and CHOL1), the source of vitamin D generally had no effect. The effect of CAL3 may be explained in part by serum CAL concentrations and postpartum serum Ca, which were associated with milk yield.  相似文献

This observational study described first-milking colostrum mineral concentrations and total yields, and evaluated its associations with cow serum mineral concentrations, parity, and first postpartum milking yield in 100 multiparous Jersey cows from a single herd fed a negative dietary cation-anion difference diet prepartum. Additionally, first- and second-milking colostrum mineral concentrations and total yields were compared in a subset of 65 cows. Serum minerals (Ca, P, Mg, Na, K, Zn, Fe, Cu) were assessed before first milking. Cows were milked at 9 h and 4 min ± 3 h and 32 min and at 21 h and 11 min ± 3 h and 43 min postpartum (± standard deviation); yields were recorded and samples collected for mineral concentrations assessment (Ca, P, Mg, Na, K, Zn, Fe, Cu). Linear regression was used to evaluate the associations between first-milking colostrum mineral concentrations and total yields and cows' serum mineral concentrations, parity, first-milking colostrum yield, and calving-to-milking interval. The most abundant minerals in first-milking colostrum were (least squares mean ± standard error of the mean) Ca (55.71 ± 13.52 mmol/L; 8.75 ± 5.74 g) and P (41.91 ± 13.01 mmol/L; 5.26 ± 3.72 g), followed by Na (39.65 ± 13.23 mmol/L; 3.08 ± 1.77 g), K (36.47 ± 7.57 mmol/L; 5.79 ± 4.20 g), Mg (13.43 ± 3.09 mmol/L; 1.25 ± 0.78 g), Zn (272.12 ± 113.34 μmol/L; 71.98 ± 55.34 mg), Fe (12.51 ± 3.79 μmol/L; 2.56 ± 1.55 mg), and Cu (3.34 ± 1.22 μmol/L; 0.77 ± 0.56 mg). Higher concentrations of Ca, Mg, Na, Zn, Fe, and Cu, and total yields of Mg, Zn, Fe, and Cu were observed at first- compared with second-milking colostrum. Serum and first-milking colostrum Cu concentrations were positively associated, but no significant associations were observed between other minerals' serum and first-milking colostrum concentrations or total yields. Parity was associated with first-milking colostrum Ca, P, K, and Fe concentrations and yields; younger multiparous cows had higher concentrations and total yields of these minerals. Linear (Fe), quadratic (P, Na, and K), and cubic (Mg, Zn, and Cu) associations were observed between first-milking colostrum mineral concentrations and yield. In all cases, mineral total yields were linearly associated with first-milking colostrum yield. In conclusion, variation in first-milking colostrum mineral concentrations and total yields across cows could be partially explained by cow parity and colostrum yield. Further research including primiparous and cows under different management settings is needed to expand the knowledge and understanding of colostrum mineral concentrations and total yields in dairy cows.  相似文献

A double-blind, randomized clinical trial was conducted in 5 commercial dairy herds in southern Ontario with 1,362 cows enrolled to evaluate the effect of prepartum administration of recombinant bovine somatotropin (rbST) on health and performance. Cows were randomly assigned to receive either 325 mg of sometribove zinc suspension (n = 680) or a placebo injection (n = 682; control) subcutaneously every 14 d until calving. Treatments started 28 to 22 d before expected calving, with a maximum of 3 treatments per cow. Serum samples taken at the time of enrollment, 1 wk before calving, and weekly for 3 wk after calving were analyzed for nonesterified fatty acids (NEFA), β-hydroxybutyrate (BHBA), glucose, aspartate aminotransferase, calcium, and haptoglobin. Diseases were recorded by farm staff. Incidences of clinical ketosis, clinical mastitis, displaced abomasum, metritis, retained placenta, milk fever, and lameness were similar between groups. Body condition score was lower for treated than for control cows at 3 wk after calving (3.13 and 3.17, respectively). Serum NEFA tended to be higher for treated than for control cows by 0.01 mmol/L. Overall BHBA was not different between groups, but BHBA for treated cows was higher in wk 1 after calving (750 and 698 μmol/L, respectively) and tended to be higher in wk 2 after calving (779 and 735 μmol/L, respectively). Incidence of hyperketonemia was similar between groups. Treated cows had higher serum glucose compared with control cows (2.8 and 2.7 mmol/L, respectively). We detected no differences in serum aspartate aminotransferase, calcium, or haptoglobin between groups. Milk yield was recorded daily for each cow for 63 d, and did not differ between groups (37.1 ± 0.5 kg and 36.7 ± 0.5 kg, respectively) but we detected a tendency for treated cows to produce 0.8 kg/d more milk than control cows in wk 1 after calving. We observed no difference between groups in the time from calving to first insemination or the probability of pregnancy at the first insemination. Groups did not differ in the proportion of anovular cows at 53 ± 3 d in milk based on serum progesterone measured from a subset of cows (38.0 and 34.3%, respectively, for treated and control groups). We found no difference between groups in dry matter intake from 21 d before calving to 63 d after calving in a subset of cows (17.4 ± 0.4 and 17.5 ± 0.4 kg/d, respectively). Based on results of the current study, biweekly (every 14 d) administration of rbST before calving to prevent disease and enhance performance is not recommended.  相似文献

In the current study, we used heat stress (HS) as an oxidative stress model to examine the effects of hydroxy-selenomethionine (HMSeBA), an organic selenium source, on selenium's bioavailability, antioxidant status, and performance when fed to dairy cows. Eight mid-lactation Holstein dairy cows (141 ± 27 d in milk, 35.3 ± 2.8 kg of milk/d, parity 2 or 3) were individually housed in environmental chambers and randomly assigned to 1 of 2 treatments: inorganic Se supplementation (sodium selenite; SS; 0.3 mg of Se/kg of dry matter; n = 4) or HMSeBA supplementation (0.3 mg of Se/kg of dry matter; n = 4). The trial was divided into 3 continuous periods: a covariate period (9 d), a thermal neutral (TN) period (28 d), and a HS period (9 d). During the covariate and TN periods, all cows were housed in TN conditions (20°C, 55% humidity). During HS, all cows were exposed to cyclical HS conditions (32–36°C, 40% humidity). All cows were fed SS during the covariate period, and dietary treatments were implemented during the TN and HS periods. During HS, cows fed HMSeBA had increased Se concentrations in serum and milk, and total Se milk-to-serum concentration ratio compared with SS controls. Superoxide dismutase activity did not differ between Se sources, but we noted a treatment by day interaction in glutathione peroxidase activity as HS progressively reduced it in SS controls, whereas it was maintained in HMSeBA cows. Supplementation with HMSeBA increased total antioxidant capacity and decreased malondialdehyde, hydrogen peroxide, and nitric oxide serum concentrations compared with SS-fed controls. We found no treatment effects on rectal temperature, respiratory rate, or dry matter intake. Supplementing HMSeBA tended to increase milk yield and decrease milk fat percentage. No other milk composition parameters differed between treatments. We observed no treatment effects detected on blood biochemistry, except for a lower alanine aminotransferase activity in HMSeBA-fed cows. These results demonstrate that HMSeBA supplementation decreases some parameters of HS-induced oxidative stress.  相似文献

The objectives were to determine the effects of a rumen-protected blend of B vitamins and choline (RPBC) on the incidence of health disorders, milk yield, and reproduction in early lactation and the effects on gene expression and liver fat infiltration. A randomized controlled trial in 3 commercial dairy herds (n = 1,346 cows with group as the experimental unit; experiment 1) and a university research herd (n = 50 cows with cow as the experimental unit; experiment 2) evaluated the use of 100 g/cow per d of commercially available proprietary RPBC supplement (Transition VB, Jefo, St. Hyacinthe, Quebec, Canada), or a placebo, fed 3 wk before to 3 wk after calving. In experiment 2 liver biopsies were taken at 4 and 14 ± 1 d in milk to measure triacylglycerol concentrations and expression of 28 genes selected to represent relevant aspects of liver metabolism. Treatment effects were assessed using multivariable mixed logistic regression models for binary health and reproductive outcomes; linear regression models for milk yield, dry matter intake, and liver outcomes; and survival analysis for time insemination and pregnancy. In experiment 1, treatment did not have an effect on the incidence of hyperketonemia (blood β-hydroxybutyrate ≥ 1.2 mmol/L; cumulative incidence to 3 wk postpartum of 28 to 30%), clinical health disorders, or udder edema. The prevalence of anovulation at 8 wk postpartum was 11% in the treatment group and 23% in the control but did not differ statistically given group-level randomization. Pregnancy at first insemination (33 and 35%) and median time to pregnancy to 200 d in milk (96 and 97 d) were not different between treatment and control, respectively. No difference was observed between treatment groups in milk yield or components through the first 3 Dairy Herd Improvement Association test days (44 kg/d in both groups, accounting for parity and components). In experiment 2, there were no differences between treatment groups in feed intake. Mean blood β-hydroxybutyrate was lower at wk 3 in RPBC (0.6 vs. 0.9 ± 0.12 mmol/L) with no difference between treatments for mean blood concentrations of fatty acids (wk ?1 or 1) and β-hydroxybutyrate at wk 1 or 2. The gene for acyl-CoA oxidase 1 (ACOX1) had lower mRNA abundance in RPBC with no difference between treatments for the other genes, but the expression of half of the genes assessed differed with days in milk. Liver triacylglycerol was lower in primiparous cows at 4 d in milk in RPBC (2.0 vs. 4.4 ± 1.2%) but not at 14 d in milk (2.2 vs. 3.2 ± 0.97%) with no treatment effect in multiparous cows (4.6 ± 0.8%). Accounting for parity, days in milk, fat and protein percentages, repeated test days, and a random effect of cow, no significant difference was observed between treatments in milk yield across the first 3 Dairy Herd Improvement Association tests (41.2 ± 1.3 in RPBC vs. 38.0 ± 1.4 kg/d in control). Under the diet and management conditions of the field study including low prevalence of clinical health disorders, in experiment 1 we did not detect a benefit of RPBC, but in experiment 2 liver fat content decreased in primiparous cows.  相似文献

The aims of the present study were to evaluate the effects of live yeast supplementation (Vistacell MUCL 39855, AB Vista, Marlborough, UK) on performance, rumination time, and rumen pH on dairy cows in commercial farm environments. Three trials were carried out, the trials lasted 12 (trial 1), 15 (trial 2), and 19 wk (trial 3). In each trial, 14 multiparous Holstein dairy cows were allocated to 2 groups that received (trial 1) a standard diet plus yeast, (trial 2) an acidogenic diet plus yeast, and (trial 3) grazing pasture plus yeast. Milk production, milk chemical characteristics, body weight and body condition score, rumination time, and rumen pH were monitored for each group throughout the 3 trials. No statistically significant differences were observed in any of the 3 trials for any of the recorded variables. In contrast, an effect of time (period or days in milk) on rumen pH was observed in all 3 of the trials, as time spent under the acidotic thresholds increased across the experimental periods; however the differences were not associated with live yeast supplementation. No effect of live yeast supplementation was observed in any of the 3 trials reported. Further research should include studies on animals at different stages of lactation (with emphasis on transition period and early lactation), consuming more challenging diets (higher level of inclusion of concentrates or starch), or under different environments such as grazing of succulent forages. Such studies might be required to elucidate any possible effect of live yeast supplementation of dairy cows when the rumen environment is under challenge.  相似文献

The effect of peripartal supplementation with concentrate enriched at 10% propylene glycol (PG) on metabolism, animal health, fertility, and milk production was studied using 234 cows from 8 dairy farms with production averages of 8019 to 10,656 kg/yr. The feeding schedule for the PG group (n=117) was as follows: 13 d antepartum: 1.5 kg/d (= 150 mL PG); 12 d antepartum until parturition: 3 kg/d (= 300 mL PG); 1 to 12 d postpartum: 1 kg/d (= 100 mL PG). Control cows (n=117) received the same concentrate without PG. From a subset of cows (PG: n = 43; control: n = 40), blood samples were collected at 6, 3, and 1 wk antepartum, 3 d antepartum, on the day of parturition, and 1, 3, 5, 7, 9, and 11 wk postpartum for the determination of nonesterified fatty acids (NEFA), beta-hydroxybutyrate (BHBA), insulin-like growth factor (IGF)-I, and activities of aspartate-aminotransferase (AST) and glutamate-dehydrogenase (GLDH). From another subset of cows (PG: n=11; control: n=10), blood samples were collected 1 wk antepartum, on the day of parturition, and 1 wk postpartum to determine immunophenotypical and functional parameters of blood neutrophils. From 1 wk antepartum to 1 d postpartum, concentrations of NEFA were significantly lower in cows receiving PG compared with controls. Also, concentrations of BHBA in cows receiving PG were significantly lower from 1 wk antepartum until 7 wk postpartum. Concentrations of IGF-I were significantly higher in the PG group, from 1 wk antepartum until 1 wk postpartum than in the control group. Activities of AST and GLDH did not differ between groups. Immunophenotypical and functional characteristics of blood neutrophils were not influenced by treatment nor were animal health, reproduction, or milk production. Although indicators of metabolic status were improved by peripartal use of PG-enriched concentrate, economic benefits are questionable for dairy farms with good nutritional programs, as economically important factors such as milk production, animal health, and fertility were not influenced.  相似文献

The objective of this study was to measure cow response to feeding of two dietary concentrations of P, one of which was close to recent National Research Council requirements, and the other of which was well in excess of the requirement. Diets containing 0.37 or 0.57% P (dry basis) were fed to Holstein cows for the first 165 d of lactation, and occasionally longer until cows were confirmed pregnant approximately 60 d after insemination. At calving, cows were randomly assigned to experimental diets. The number of cows completing a minimum of 165 d of lactation was 123 for the 0.37 and 124 for the 0.57% P groups. Cows were housed in a stanchion barn and fed one of two transition diets, each formulated to contain one of the P treatments for the first 3 wk of lactation, and then cows were moved to a free-stall barn where the experimental diets were group fed. Milk production, milk fat, and milk protein averaged 35.1 kg/d, 3.92%, and 2.90% for the 0.37% P diet, and 34.9 kg/d, 3.98%, and 2.91% for the 0.57% P diet. None of these measures were different between treatments. Blood serum P concentrations on d 50 and 100 of lactation averaged 6.1 and 6.2 mg/dL for the 0.37% P diet, and 6.8 and 6.9 mg/dL for the 0.57% P diet. No treatment differences were detected in milk production, cow health, or body condition score.  相似文献

Treatment with granulocyte colony-stimulating factor (G-CSF) increases polymorphonuclear cell (neutrophil) count and enhances neutrophil function in the periparturient cow. Prepartum undernutrition was hypothesized to reduce the effect of a commercially available recombinant bovine G-CSF product (pegbovigrastim) on neutrophil count and function. Hence this study was undertaken to test the effect of undernutrition for approximately 1 mo before calving on the innate immune response to pegbovigrastim. Cows (n = 99) on pasture were blocked by expected calving date and body condition score and randomly assigned in a 2 × 2 factorial design. The first factor was that cows were fed to exceed energy requirements prepartum (full feeding) or restricted to approximately 85% of prepartum energy requirements (restricted feeding). The second factor was that at approximately 7 d before expected calving date, half the cows in each feed group were injected with pegbovigrastim and the remaining half were injected with saline. Treatments were repeated on the day of calving. Blood samples were collected pre- and postcalving for complete blood count, biochemistry, and in vitro assessment of neutrophil function including phagocytosis, myeloperoxidase release, and oxidative burst. Prepartum energy restriction resulted in lower body weight, a higher proportion of cows with elevated concentrations (i.e., >0.4 mmol/L) of fatty acids, and higher average β-hydroxybutyrate concentrations before calving relative to fully fed cows. Treatment with pegbovigrastim increased the total white cell, neutrophil, lymphocyte, and monocyte counts. Pegbovigrastim treatment resulted in increased release of myeloperoxidase by neutrophils. Prepartum feeding group did not have an effect, and no feeding group × treatment interaction was observed for any of the white cell counts or functional tests. We concluded that pegbovigrastim treatment results in significant increases in neutrophil count and enhances neutrophil function as indicated by increased myeloperoxidase release. The response to pegbovigrastim was not affected by restricted prepartum energy intake.  相似文献

A 9-wk trial was conducted to study the performance of 24 Holstein cows during the transition period (3 wk prepartum to 6 wk postpartum). Cows were assigned to either a control or liquid-flavored (0.52 mL/kg of feed) total mixed ration in a randomized complete block design. The diets contained corn silage, alfalfa haylage, cottonseed, and a grain mix based on ground corn and soybean meal. Cows were fed to ensure 10% orts, and the diet provided (on a dry matter basis) 13% crude protein, 32% acid detergent fiber, 44% neutral detergent fiber, and 1.54 Mcal/kg of NEL prepartum and 17.5% crude protein, 30% acid detergent fiber, 40% neutral detergent fiber, and 1.57 Mcal/kg of NEL postpartum. An additional 2.3 kg of alfalfa hay was fed during the first 5 d postpartum. Weekly means of dry matter intake (DMI), milk yield, milk protein, milk fat, SNF, somatic cell counts, and body weight (BW) were analyzed using a repeated measures procedure. There was no effect of treatment on these variables, and least squares means were 16.9 and 15.7 kg/d for DMI, 38 and 35.3 kg/d for milk yield, 3.10 and 3.11% for milk protein, 3.69 and 3.74% for milk fat, 8.37 and 8.16% for SNF, 1.99 x 10(5) and 4.33 x 10(5) for somatic cell count, and 631 and 651 kg for BW for cows fed control and flavored diets, respectively. Individual cow daily DMI data were fitted to an exponential model describing pre- and postpartum feed consumption [DMI = a - b x e(-c x t), where DMI was measured in kg, a = asymptotic DMI, b = potential fractional increase in DMI, c = fractional rate of increase in DMI, and t = days prior to calving or days in milk]. Fractional rates of increase in DMI were similar: 0.139 and 0.123/d for control and flavored diets, respectively. Data for both groups were separately analyzed using multiple regression with 3.5% fat-corrected milk as the dependent variable and BW and DMI as independent variables. More BW was mobilized per unit increase in 3.5% fat-corrected milk in cows fed the control than in cows fed the flavored diet. Cows fed the control diet tended to be in more negative energy balance during early lactation than cows fed the flavored diet. It was concluded that feeding flavor improved energy balance of cows in early lactation and may reduce the risk of health or reproductive problems.  相似文献

The objectives of this study were to evaluate the effect of stillbirth on survival and reproductive performance of lactating dairy cows. Data were collected from 2 different regions of the US calving-ease scores (CES) were recorded by farm personnel on a scale of 1 (no problem) to 5 (extreme difficulty). Stillbirths were recorded by farm personnel. The final analysis included 13,608 calvings of which 93.4% were live calves and 6.6% stillbirths. An increasing or decreasing trend in the incidence of stillbirth by parity and by CES was analyzed by Cochran-Armitage trend tests. A significant decreasing trend in the incidence of stillbirth by parity group was detected. The incidence of stillbirth increased as the CES increased. The incidence of stillbirths was 3.6, 11.2, 25.9, and 60.1% for CES score 1, 2, 3, and 4, respectively. Dam survival in the herd and reproductive performance were analyzed by the Cox proportional hazards model. Variables that decreased dam survival time were stillbirths, primiparity, and CES of 3 and 4. The variables that reduced reproductive performance were stillbirths, multiparity, male calves, and CES of 3 and 4. Cows that had stillbirths had significantly increased risk of culling/death throughout the lactation and increased median days open by 88 d compared with cows that had live calves. In conclusion, losses from stillbirths are far greater than just the value of the calf.  相似文献

The objective of this study was to determine the effect of feeding direct-fed microbial (DFM) products containing Lactobacillus acidophilus and Propionibacteria freudenreichii on the performance, nutrient digestibility, and rumen fermentation of Holstein dairy cows in midlactation. Experiments were conducted from February to May 2003. Cows were fed 1 of 3 dietary treatments: 1) 1 × 109 colony-forming units (cfu)/d of live L. acidophilus strain LA747 and 2 × 109 cfu/d of live P. freudenreichii strain PF24 (DFM1); 2) 1 × 109 cfu/d of live L. acidophilus strain LA747, 2 × 109 cfu/d of live P. freudenreichii strain PF24, and 5 × 108 cfu/d of L. acidophilus strain LA45 (DFM2); or 3) lactose (control). Treatments were administered by mixing 45 g of finely ground corn with 5 g of DFM products or lactose and top dressing on the total mixed rations once daily. All cows received the same total mixed ration: 12.7% alfalfa hay, 46.2% corn silage, and 41.1% concentrate on a dry matter (DM) basis. In study 1 (lactation study), 39 multiparous and 18 primiparous Holstein cows were blocked by parity and randomly assigned to treatments for 84 d. Starting on d 35, fecal grab samples were collected from each cow at 5- to 8-h intervals over 48 h for digestibility measurements. A rumen fermentation study (study 2) was conducted concurrently with the lactation study. Three rumenfistulated, multiparous Holstein cows were randomly assigned to dietary treatments DFM1, DFM2, and control in a 3 × 3 Latin square design with 28-d periods. In study 1, there was no difference in average DM intake (23.9, 23.6, and 24.2 kg/d) or 4% fat-corrected milk (36.8, 35.3, and 36.2 kg/d) for treatments DFM1, DFM2, and control. Percentage or yield of milk components also did not differ among treatments. Feed efficiency averaged 1.52 kg of 4% fat-corrected milk/kg of DM intake and did not differ among treatments. There were no differences in apparent DM, crude protein, neutral detergent fiber, or starch digestibility among treatments. In study 2, there was no difference in rumen pH and concentrations of ammonia or total volatile fatty acids measured at 0, 1, 3, and 6 h after feeding. Under the conditions of these studies, supplementing midlactation cows with DFM products containing L. acidophilus and P. freudenreichii did not affect cow performance, diet digestibility, or rumen fermentation.  相似文献

Phosphorus depletion and hypophosphatemia have been described to interfere with immune function in rats and humans. In dairy cows, hypophosphatemia has been associated with muscle weakness and recumbency as well as with intravascular hemolysis resulting from increased osmotic fragility of erythrocytes, but so far, the influence of P depletion and hypophosphatemia on immune function has not been studied. Therefore, the aim of this study was to investigate whether P depletion and ensuing hypophosphatemia are associated with impaired granulocyte and lymphocyte function. Eight mid-lactation dairy cows were fed a P-deficient ration (0.2% P/kg of DM) for a period of 4 wk. The depletion phase was preceded by a 2-wk acclimatization period and followed by a 2-wk repletion phase, during which the same ration was supplemented with P to meet or exceed daily requirements. Blood samples were collected at the end of the acclimatization period, after 2 and 4 wk of P depletion, and at the end of the repletion phase. Plasma phosphate concentrations ([Pi]) were determined and white blood cells were counted and isolated. General immune function was investigated by performing a phagocytosis assay with Staphylococcus aureus and a lymphocyte stimulation test (LST) with concanavalin A and pokeweed mitogen. The plasma [Pi] decreased significantly, with the lowest values (mean 0.7 ± 0.2 mmol/L) occurring after 2 wk of depletion, although depletion was continued for another 2 wk. During repletion, plasma [Pi] increased above baseline concentrations. Granulocyte counts changed in parallel with plasma [Pi] over time, decreasing significantly at 2 wk after P depletion and increasing again thereafter. Granulocyte survival after phagocytosis was lowest after 4 wk of P depletion. Phagocytosis activity of surviving granulocytes determined by mean fluorescence intensity was higher, indicating that phagocytosis was not negatively influenced by P depletion. Lymphocyte stimulation showed a similar trend, with a decreasing stimulation index at the end of P depletion, but differences were not statistically significant. Data presented in this study indicate that hypophosphatemia leads to a decrease in granulocyte counts. Chronic P depletion impairs granulocyte survival during phagocytosis but not phagocytosis activity. Lymphocyte function is not influenced by P depletion.  相似文献
