ABCD1 is a gene responsible for X‐linked adrenoleukodystrophy (X‐ALD), and is critical for the transport of very long‐chain fatty acids (VLCFA) into peroxisomes and subsequent β‐oxidation. VLCFA‐containing lipids accumulate in X‐ALD patients, although the effect of ABCD1‐deficiency on each lipid species in the central nervous system has not been fully characterized. In this study, each phospholipid and lysophospholipid species in Abcd1‐deficient mice brains were profiled by liquid chromatography‐mass spectrometry. Among the phospholipid and lysophospholipid species that are significantly more enriched in Abcd1‐deficient mice brains, VLCFA were present in 75, 15, 5, 4, and 1 species of phosphatidylcholine, phosphatidylethanolamine, sphingomyelin, lysophosphatidylcholine, and lysophosphatidylethanolamine, respectively. Most VLCFA were incorporated at the sn‐1 position of phosphatidylcholine and phosphatidylethanolamine. Among the phospholipid species that are significantly less enriched in Abcd1‐deficient mice brains, odd‐numbered saturated or mono‐unsaturated fatty acyl moieties are contained in all phosphatidylcholine species. In addition, a number of phosphatidylglycerol, phosphatidylinositol, and phosphatidylserine species contained highly unsaturated fatty acyl moieties. Intriguingly, 44:1 phosphatidylcholine with VLCFA was mainly distributed in the gray matter, such as the cortex, but not in the white matter in the cerebrum and cerebellum. These results show that ABCD1‐deficiency causes metabolic alternation of long‐chain fatty acids and VLCFA. Moreover, our results imply a molecular mechanism for the incorporation of saturated or monounsaturated VLCFA into the sn‐1 position of phospholipids, and also indicate that the distribution of phospholipids with VLCFA may correlate with the development of X‐ALD.  相似文献

The benzoxazine scaffolds are of much interest as they are found in a large array of natural products and pharmaceutical drugs with diverse activities. We have developed a palladium‐catalyzed decarboxylative selective mono‐ and bis‐acylation of 4H‐benzo[d][1,3]oxazin‐4‐one derivatives with α‐oxo carboxylic acids via preferential cyclic imine‐N‐directed C−H activation. 2‐Aryl‐4H‐benzo[d][1,3]oxazin‐4‐one was acylated with a variety of substituted phenylglyoxylic acids to produce the corresponding products. It was observed that electron‐donating groups (CH3, OCH3) at any position of the aromatic ring of phenylglyoxylic acid provided good to excellent yields, whereas phenylglyoxylic acids containing electron‐withdrawing groups (COCH3, CN, NO2) gave the products in moderate yields. Interestingly when the reaction was performed with silver triflate (AgOTf) in place of silver nitrate (AgNO3) in the presence of 4 equivalents of glyoxylic acid, the bis‐acylated product was obtained together with a small amount of mono‐acylated product. This is the first report of acylation of 2‐aryl‐4H‐benzo[d][1,3]oxazin‐4‐ones via C−H activation. The notable features of this reaction are acylation with more challenging heteroarene‐oxo carboxylic acids and alkyl oxo carboxylic acids. This new protocol provides an easy and efficient access to a variety of o‐acyl‐4H‐benzo[d][1,3]oxazin‐4‐one derivatives which are of pharmaceutical importance.

A vinyl‐modified anthraquinone (AQ) derivative (Vinyl‐AQ) is synthesized through a palladium‐mediated Suzuki coupling reaction between vinylphenylboronic acid and 2‐chloromethylanthraquinone and, subsequently, copolymerized with N,N‐dimethylacrylamide (DMAM) through free radical copolymerization in organic solvent. The chemical structure of the resulting water‐soluble copolymer, P(DMAM‐co‐AQ), is verified using techniques such as proton nuclear magnetic resonance, attenuated total reflection‐infrared spectroscopy, thermogravimetric analysis, and UV–vis spectroscopy. The evolution of the oxygen scavenging abilities of aqueous P(DMAM‐co‐AQ) solutions after UV irradiation is monitored as a function of UV irradiation time, concentration of AQ moieties, and pH. The copolymer is proved an effective UV‐triggered oxygen scavenger, leading to dissolved oxygen contents below 1 ppm for the optimized experimental conditions. This behavior is related with the appearance of novel chemical species with interesting optical properties, as suggested by the respective evolution of the UV–vis absorption and photoluminescence spectra after UV irradiation.  相似文献

The first copper‐catalyzed enantioselective conjugate addition of indoles to β‐substituted unsaturated acyl phosphonates was successfully realized by using a heteroarylidene‐tethered bis(oxazoline) ligand. The reaction features high efficiency, cheap catalyst and broad generality. In the case of either β‐alkyl‐ or β‐aryl‐substituted unsaturated acyl phosphonates, the 3‐indolyl adducts were achieved in high yields with good to excellent enantioselectivities (up to 97% ee). The 3‐indolyl adducts can serve as important intermediates in the synthesis of indole alkaloids.

The highly regioselective cobalt‐catalysed 1,4‐hydrovinylation of terminal alkenes with 2‐trimethylsilyloxy‐1,3‐butadiene generates in a stereospecific fashion unsaturated E‐configured silyl enol ether intermediates that are suitable for diastereoselective Mukaiyama‐aldol reactions with bulky aliphatic aldehydes. The acidic hydrolysis of the enol ethers to γ,δ‐unsaturated ketones followed by ozonolysis can be used for the synthesis of various 1,4‐diketones and polycarbonyl derivatives. The 1,4‐diketones and polycarbonyl derivatives were successfully tested for the synthesis of some mono‐ and bis‐pyrrole derivatives. The γ,δ‐unsaturated ketones are useful building blocks (e.g., in natural product synthesis) and can be generated in a one‐pot procedure.  相似文献

The enantioselective 1,4‐addition of arylboronic acids to β‐arylenones to give β‐diaryl ketones was carried out at 0–25 °C in the presence of a dicationic palladium(II) catalyst, [Pd(S,S‐chiraphos)(PhCN)2](SbF6)2. Addition of a silver salt such as silver tetrafluoroborate [AgBF4] or silver hexafluoroantimonate [AgSbF6] (5–10 mol %) was effective to achieve high enantioselectivities at low temperatures (92–99 % ee) and to reduce the catalyst loading to 0.05 mol %. The protocol provided a simple access to 4‐aryl‐4H‐chromenes. Optically active chromenes were synthesized with up to 99 % ee via dehydration of the 1,4‐adducts between arylboronic acids and β‐(2‐hydroxyaryl)‐α,β‐unsaturated ketones.  相似文献

Forty‐three oxime derivatives were synthesized by allowing O‐benzylhydroxylamines to react with primary benzaldehydes or salicylaldehydes; these products were gauged as potential inhibitors of β‐ketoacyl‐(acyl‐carrier‐protein) synthase III (FabH). Among the 43 compounds, 38 are reported herein for the first time. These compounds were assayed for antimicrobial activities against Escherichia coli, Pseudomonas aeruginosa, Pseudomonas fluorescens, Bacillus subtilis, Staphylococcus aureus, and Enterococcus faecalis. Compounds with prominent antibacterial activities were tested for their E. coli FabH inhibitory activities. 3‐((2,4‐Dichlorobenzyloxyimino)methyl)benzaldehyde O‐2,4‐dichlorobenzyl oxime ( 44 ) showed the best antibacterial activity, with minimum inhibitory concentrations of 3.13–6.25 μg mL?1 against the tested bacterial strains, exhibiting the best E. coli FabH inhibitory activity, with an IC50 value of 1.7 mM . Docking simulations were performed to position compound 44 into the E. coli FabH active site in order to determine the most probable binding conformation.  相似文献

Palladium‐catalyzed direct mono‐ and diarylations involving C(sp3) H cleavage at the γ‐position of acrylamides are described. The monoarylation products can be obtained with ortho‐substituted aryl halides. Single crystal X‐ray diffraction has shown that the double bond has shifted towards the introduced aryl group to afford the γ‐arylated β,γ‐unsaturated amide products. A second arylation occurs when less sterically hindered aryl halides are employed. This chemistry offers a novel disconnection for the synthesis of γ‐arylated compounds.  相似文献

The first example of the synthesis of 4,5‐disubstiuted furan‐2(5H)‐ones by base‐mediated tandem acyl shift/cyclization/decarbonylation reactions of aroylmethyl 2‐alkynoates has been developed. This new and inexpensive tandem route allows both a C O bond and a Csp3 Csp2 bond forming in one step involving an unprecedented acyl rearrangement process.  相似文献

The purple photosynthetic bacterium Rhodospirillum centenum has a putative type III polyketide synthase gene (rpsA). Although rpsA was known to be transcribed during the formation of dormant cells, the reaction catalyzed by RpsA was unknown. Thus we examined the RpsA reaction in vitro, using various fatty acyl‐CoAs with even numbers of carbons as starter substrates. RpsA produced tetraketide pyranones as major compounds from one C10–14 fatty acyl‐CoA unit, one malonyl‐CoA unit and two methylmalonyl‐CoA units. We identified these products as 4‐hydroxy‐3‐methyl‐6‐(1‐methyl‐2‐oxoalkyl)pyran‐2‐ones by NMR analysis. RpsA is the first bacterial type III PKS that prefers to incorporate two molecules of methylmalonyl‐CoA as the extender substrate. In addition, in vitro reactions with 13C‐labeled malonyl‐CoA revealed that RpsA produced tetraketide 6‐alkyl‐4‐hydroxy‐1,5‐dimethyl‐2‐oxocyclohexa‐3,5‐diene‐1‐carboxylic acids from C14–20 fatty acyl‐CoAs. This class of compounds is likely synthesized through aldol condensation induced by methine proton abstraction. No type III polyketide synthase that catalyzes this reaction has been reported so far. These two unusual features of RpsA extend the catalytic functions of the type III polyketide synthase family.  相似文献

(Z)‐3‐((3‐hydroxybenzylidene)amino)pyridin‐1‐ium4‐dodecylbenzenesulfonate anionic surfactant and its cobalt, copper and zinc complexes were synthesized (I, I‐Co, I‐Cu and I‐Zn). The chemical structures of it were characterized by elemental analysis, FTIR, 1H‐NMR, UV–Vis spectroscopy and atomic adsorption. The effects of the chemical structures of the synthesized anionic surfactant and the type of transition metals on the surface activity are presented in this paper. The thermodynamic parameters show that adsorption and micellization processes are spontaneous. The results of biological activity measurements showed that the synthesized compounds have a great efficiency against gram positive (Bacillus subtilis and Staphylococcus aureus), gram‐ negative bacteria (Pseudomonas aeruginosa and Escherichia coli) and fungi (Candida albicans and Aspergillus niger) as well as the sulfate reducing bacteria (Desulfomonas pigra). The complexation of the anionic surfactant with Co2+, Cu2+ and Zn2+ increase the antimicrobial activity values.  相似文献

A one‐pot, three‐component method for preparation of 1,4,5‐trisubstituted 5‐acyl‐1,2,3‐triazoles has been developed based on a highly selective acylation of the copper(I)‐carbon(sp2) bond with acyl chlorides in the presence of the copper(I)‐carbon(sp) bond. The procedure is characterized by high efficiency and simple conditions.

Four series of noble networks were synthesized with acrylic acid (AAc) copolymerized with varying amount of 2‐hydroxy propyl methacrylate or dodecyl methacrylate (AAc/HPMA or AAc/DMA; 5:1 to 5:5, w/w) in the presence of ethylene glycol dimethacrylate (EGDMA; 1, 5, 10, 15, and 20%, w/w) as a crosslinker and ammonium per sulfate (APS) as an initiator. Each of the networks was used to immobilize a purified lipase from Pseudomonas aeruginosa MTCC‐4713. The lipase was purified by successive salting out with (NH4)2SO4, dialysis, and DEAE anion exchange chromatography. Two of the matrices, E15a, i.e. [poly (AAc5co‐DMA1cl‐EGDMA15)] and I15c, i.e. [poly (AAc5co‐HPMA3cl‐EGDMA15)], that showed relatively higher binding efficiency for lipase were selected for further studies. I15c‐hydrogel retained 58.3% of its initial activity after 10th cycle of repetitive hydrolysis of p‐NPP, and I15c was thus catalytically more stable and efficient than the other matrix. The I15c‐hydrogel‐immobilized enzyme showed maximum activity at 65°C and pH 9.5. The hydrolytic activity of free and I15c‐hydrogel‐immobilized enzyme increased profoundly in the presence of 5 mM chloride salts of Hg2+, NH4+, Al3+, K+, and Fe3+. The immobilized lipase was preferentially active on medium chain length p‐nitrophenyl acyl ester (C:8, p‐nitrophenyl caprylate). © 2006 Wiley Periodicals, Inc. Highly efficient syntheses of indolo[2,1‐a]isoquinolines, indolo[2,1‐a][2]benzazepines, pyrrolo[2,1‐a]isoquinolines and pyrrolo[1,2‐a]benzazepines in excellent yields have been achieved by the intramolecular photochemical cross‐coupling reactions of 3‐acyl‐2‐halo‐N‐(ω‐arylalkyl)indoles and 2‐chloro‐N‐(ω‐arylalkyl)pyrrole‐3‐carbaldehydes in acetone. A new heterocyclic ring system – pyrrolo[1,2‐d][1,4]benzoxazepine – has also been constructed for the first time in this work by the photocyclization of 2‐chloro‐N‐(2‐phenoxyethyl)pyrrole‐3‐carbaldehyde.  相似文献

P‐glycoprotein (P‐gp)‐mediated multidrug resistance (MDR) is a major obstacle for successful cancer chemotherapy. Based on our previous study, 17 novel compounds with the 6,7‐dimethoxy‐2‐{2‐[4‐(1H‐1,2,3‐triazol‐1‐yl)phenyl]ethyl}‐1,2,3,4‐tetrahydroisoquinoline scaffold were designed and synthesized. Among them, 2‐[(1‐{4‐[2‐(6,7‐dimethoxy‐3,4‐dihydroisoquinolin‐2(1H)‐yl)ethyl]phenyl}‐1H‐1,2,3‐triazol‐4‐yl)methoxy]‐N‐(p‐tolyl)benzamide (compound 7 h ) was identified as a potent modulator of P‐gp‐mediated MDR, with high potency (EC50=127.5±9.1 nM ), low cytotoxicity (TI>784.3), and long duration (>24 h) in reversing doxorubicin (DOX) resistance in K562/A02 cells. Compound 7 h also enhanced the effects of other MDR‐related cytotoxic agents (paclitaxel, vinblastine, and daunorubicin), increased the accumulation of DOX and blocked P‐gp‐mediated rhodamine 123 efflux function in K562/A02 MDR cells. Moreover, 7 h did not have any effect on cytochrome (CYP3A4) activity. These results indicate that 7 h is a relatively safe modulator of P‐gp‐mediated MDR that has good potential for further development.  相似文献

Two new highly stable energetic salts were synthesized in reasonable yield by using the high nitrogen‐content heterocycle 3,4,5‐triamino‐1,2,4‐triazole and resulting in its picrate and azotetrazolate salts. 3,4,5‐Triamino‐1,2,4‐triazolium picrate (1) and bis(3,4,5‐triamino‐1,2,4‐triazolium) 5,5′‐azotetrazolate (2) were characterized analytically and spectroscopically. X‐ray diffraction studies revealed that protonation takes place on the nitrogen N1 (crystallographically labelled as N2). The sensitivity of the compounds to shock and friction was also determined by standard BAM tests revealing a low sensitivity for both. B3LYP/6–31G(d, p) density functional (DFT) calculations were carried out to determine the enthalpy of combustion (ΔcH (1) =−3737.8 kJ mol−1, ΔcH (2) =−4577.8 kJ mol−1) and the standard enthalpy of formation (ΔfH° (1) =−498.3 kJ mol−1, (ΔfH° (2) =+524.2 kJ mol−1). The detonation pressures (P (1) =189×108 Pa, P (2) =199×108 Pa) and detonation velocities (D (1) =7015 m s−1, D (2) =7683 m s−1) were calculated using the program EXPLO5.  相似文献

Novel compounds, based on the 2(1H)‐quinolone skeleton, were synthesised and characterised using 1H‐nuclear magnetic resonance (NMR) spectroscopy and chemical ionisation mass spectrometry (CI MS). Their spectroscopic properties, such as absorption and emission spectra and fluorescence quantum yield, were also examined. 7‐maleimido‐4‐methyl‐2(1H)‐quinolone, 7‐maleimido‐3,4‐dimethyl‐2(1H)‐quinolone, 7‐maleimido‐4‐propyl‐2(1H)‐quinolone and 7‐maleimido‐4‐phenyl‐2(1H)‐quinolone were evaluated as potential sensors for the detection of sulphydryl amino acids groups. These new compounds demonstrate a high ‘turn‐on’ fluorescence response and selectivity towards l ‐cysteine in the presence of other amino acids and metal cations.  相似文献

The new ruthenium‐sulfonate catalyst Ru(Cp*)(η3‐C3H5) (p‐CH3C6H4SO3)2, (Cp*=pentamethylcyclopentadienyl), rapidly and regioselectively mono‐allylates dimedone to the branched products using substituted allyl alcohols as substrates, without acid, base or other additives, under relatively mild conditions. We consider the ruthenium sulfonate to be a “green” alternative in that it uses allyl alcohols as substrate, (rather than carbonates, acetates, etc.) and therefore does not waste the leaving group. The catalyst induces rapid double allylation of various 1,3‐diketones in high yield using allylic alcohol.  相似文献
