In preparation for studies using natural products to probe interactions between bacterial consortia and settlement stage barnacles, we isolated 16 strains of bacteria associated with barnacles and examined: (1) effects of films of bacterial isolates on barnacle settlement, and (2) bacteriostatic effects of juncellins and standard antibiotics. Bacteria were isolated from the biofilm associated withBalanus amphitrite. On the basis of morphological and biochemical characteristics, bacteria were classified into five major groups:Aeromonas, Alcaligenes, Flavobacterium, Pseudomonas, andVibrio. Barnacle settlement was inhibited by allVibrio films and 64% of the other isolates. No film stimulated barnacle settlement. Juncellins were approximately as potent as standard antibiotics for all bacterial species tested.Vibrio spp. were most resistant to juncellins.  相似文献

Linear alkylbenzene sulfonate (LAS) and polyoxyethylene lauryl ether (POLE) are the major surfactants in washing detergents. In the present study, we isolated surfactant-resistant bacteria from soil samples collected from a sports ground and a farm field. The samples were treated with 2.0% LAS or POLE at 25°C for 30 min and cultivated on agar plates at 25°C for several days, after which manifold bacterial colonies were isolated. Thereafter, we tested the ability of each bacterial isolate to resist the antibacterial activity of the surfactant. Ten LAS-resistant strains were isolated, and all were found to be Gram-negative bacteria such as Enterobacter and Pseudomonas. On the other hand, 18 POLE-resistant strains were isolated, of which 14 were Gram-positive bacteria including Bacillus and Microbacterium. Notably, one POLE-resistant strain was identified as Bacillus cereus, a potential causative agent for foodborne illness. The genera of LAS- and POLE-resistant bacteria did not overlap. Therefore, the combination of LAS and POLE could be more effective to eliminate soil bacteria from clothes and/or daily necessities.  相似文献

Photocatalytic inactivation of algae,Anabaena, Microcystis, andMelosira, was performed with TiO2-coated pyrex glass beads under the illumination of UV light (370 nm wavelength). After being irradiated with UV light in the presence of the TiO2-coated pyrex glass beads,Anabaena andMicrocystis, known as typical cyanobacteria, lost their photosynthetic activity, and the string ofAnabaena cells and the colonies ofMicrocystis cells were completely separated into individual spherical ones. In the case ofMelosira, which is a typical diatom, however, somewhat lower photocatalytic inactivation efficiency was obtained, which was believed to be due to the presence of the inorganic siliceous wall surrounding the cells ofMelosira.  相似文献

Experiments on the growth of the natural populations of algae and of other microorganisms as a function of the supply of various nutrients was carried out for Lake Kinneret water sampled at about 2 week intervals during January-June 1972. The addition of orthophosphate caused a rapid rise in biomass, which, after several days, increased to 2-5 times that in untreated water, as measured by the increase in turbidity and protein content. On the other hand, the addition of sodium or potassium nitrate, ammonium chloride, ferrous ethylenediamine sulfate, sodium silicate or vitamin B12 had no noticeable effect. The plot of biomass produced versus the concentration of added phosphate is a steeply rising function in the range of 0.1 to 1.0 μ molar orthophosphate, which levels off at higher phosphate concentrations, reaching saturation above about 2 μ molar phosphate. Thus, in the range of its natural concentration in Lake Kinneret, 0.09-0.5 μ molar, orthophosphate is the limiting factor. Above 2 μ molar phosphate, nitrate becomes the limiting factor, as was shown by experiments in which both phosphate (>2 μ molar) and nitrate were added, resulting in extremely abundant algal growth. A model of the reduction of nutrient input was achieved by laboratory experiments, in which the lake water was diluted with distilled water (by 1/4), thus resulting in an approximately proportional (to about 1/4) decrease in biomass. The effect of restoring the natural concentration of either nitrate or orthophosphate was then measured. Added nitrate had no effect, while added phosphate caused a rapid increase in biomass, which after 2-4 weeks came close to that in the natural lake water. These results indicate that during the spring of 1972, phosphorus has been the only element limiting algal growth in the lake.  相似文献

Chloroquine is commonly used in the treatment and prevention of malaria, but Plasmodium falciparum, the main species responsible for malaria‐related deaths, has developed resistance against this drug. Twenty‐seven novel chloroquine (CQ) analogues characterized by a side chain terminated with a bulky basic head group, i.e., octahydro‐2H‐quinolizine and 1,2,3,4,5,6‐hexahydro‐1,5‐methano‐8H‐pyrido[1,2‐a][1,5]diazocin‐8‐one, were synthesized and tested for activity against D‐10 (CQ‐susceptible) and W‐2 (CQ‐resistant) strains of P. falciparum. Most compounds were found to be active against both strains with nanomolar or sub‐micromolar IC50 values. Eleven compounds were found to be 2.7‐ to 13.4‐fold more potent than CQ against the W‐2 strain; among them, four cytisine derivatives appear to be of particular interest, as they combine high potency with low cytotoxicity against two human cell lines (HMEC‐1 and HepG2) along with easier synthetic accessibility. Replacement of the 4‐NH group with a sulfur bridge maintained antiplasmodial activity at a lower level, but produced an improvement in the resistance factor. These compounds warrant further investigation as potential drugs for use in the fight against malaria.  相似文献

Serratia marcescens MTCC 86 when grown on sucrose produced a novel sucrose lipid that was different from the amino lipid produced from n-alkane by other strains of S. marcescens. The sucrose lipid isolated by acetone precipitation technique was found to be predominantly a mixture of 3-(3′-hydroxytetradecanoyloxy) decanoate and 3-(3′-hydroxyhexadecenoyloxy) decanoate. This sucrose lipid exhibited excellent emulsification activity with a wide variety of hydrocarbons. It was effective in recovering up to 90% residual oil from the oilsaturated sandpack column suggesting its use in enhanced oil recovery. It also showed a strong ability to remove oil from the walls of the containers, indicating its potential use in cleaning operations.  相似文献

Twenty‐four strains of marine Roseobacter clade bacteria were isolated from macroalgae and investigated for the production of quorum‐sensing autoinducers, N‐acylhomoserine lactones (AHLs). GC/MS analysis of the extracellular metabolites allowed us to evaluate the release of other small molecules as well. Nineteen strains produced AHLs, ranging from 3‐OH‐C10:0‐HSL (homoserine lactone) to (2E,11Z)‐C18:2‐HSL, but no specific phylogenetic or ecological pattern of individual AHL occurrence was observed when cluster analysis was performed. Other identified compounds included indole, tropone, methyl esters of oligomers of 3‐hydroxybutyric acid, and various amides, such as N‐9‐hexadecenoylalanine methyl ester (9‐C16:1‐NAME), a structural analogue of AHLs. Several compounds were tested for their antibacterial and antialgal activity on marine isolates likely to occur in the habitat of the macroalgae. Both AHLs and 9‐C16:1‐NAME showed high antialgal activity against Skeletonema costatum, whereas their antibacterial activity was low.  相似文献

Poly(3-hydroxybutyrate)-producing Enterococcus faecium KT722772 was isolated from cattle rumen fluid by Sudan black staining. It was found to be gram-positive cocci. Poly(3-hydroxybutyrate) production using E. faecium KT722772 was optimized at different pH ranging from 2 to 7 and with different substrates such as glucose, butyric acid, propionic acid, sodium acetate, and glycerol as a sole carbon source at a concentration ranging from 0 to 4%. Maximum poly 3-hydroxy butyrate production (1.8?g/L) was achieved with 1% butyric acid at pH 7. Poly 3-hydroxy butyrate produced by E. faecium was characterized by Fourier transform infrared spectroscopy, thermogravimetric analysis, and X-ray diffraction analysis.  相似文献

Dichloromethane extracts of culture broth from three strains of the entomopathogenic fungusHirsutella thompsonii var.synnematosa were toxic to two species of tephritid fruit fly and inhibited conidial germination in vitro in several other entomopathogenic fungi includingBeauveria bassiana, Tolypocladium spp., andMetarhizium anisopliae. A major metabolite, toxic to apple maggot,Rhagoletis pomonella, and inhibitory to conidial germination inB. bassiana, was isolated and identified as the antibiotic (+)-phomalactone, 6-(1-propenyl)-5,-6-dihydro-5-hydroxypyran-2-one. In the search for effective and sustainable drugs for human African trypanosomiasis (HAT), we developed hybrid compounds by merging the structural features of quinone 4 (2-phenoxynaphthalene-1,4-dione) with those of phenolic constituents from cashew nut shell liquid (CNSL). CNSL is a waste product from cashew nut processing factories, with great potential as a source of drug precursors. The synthesized compounds were tested against Trypanosoma brucei brucei, including three multidrug-resistant strains, T. congolense, and a human cell line. The most potent activity was found against T. b. brucei, the causative agent of HAT. Shorter-chain derivatives 20 (2-(3-(8-hydroxyoctyl)phenoxy)-5-methoxynaphthalene-1,4-dione) and 22 (5-hydroxy-2-(3-(8-hydroxyoctyl)phenoxy)naphthalene-1,4-dione) were more active than 4 , displaying rapid micromolar trypanocidal activity, and no human cytotoxicity. Preliminary studies probing their mode of action on trypanosomes showed ATP depletion, followed by mitochondrial membrane depolarization and mitochondrion ultrastructural damage. This was accompanied by reactive oxygen species production. We envisage that such compounds, obtained from a renewable and inexpensive material, might be promising bio-based sustainable hits for anti-trypanosomatid drug discovery.  相似文献

Tetrapedia diversipes and other Apidae (Anthophoridae) may be deceived by floral similarities between Malpighiaceae and Orchidaceae of the Oncidiinae subtribe. The latter do not usually exudate floral oils. Thus, visitors may pollinate the flowers in a deceit/food/pollination syndrome. We studied the chemical compositions of Byrsonima intermedia (Malpighiaceae) floral oil and T. diversipes (Anthophoridae) cell provisions. From B. intermedia floral oil, we isolated a novel fatty acid (3R, 7R)-3,7-diacetoxy-docosanoic acid, here named byrsonic acid, and from T. diversipes cell provisions we isolated two novel fatty acid derivatives 3,7-dihydroxy-eicosanoic acid and 3,7-dihydroxy-docosanoic acid, here named tetrapedic acids A and B, respectively. The three fatty acid derivatives have common features: possess long chains (20 or 22 carbon atoms) with no double bond and either hydroxy or acetoxy groups at carbons 3 and 7. This characteristic was also encountered in the fatty acid moiety of oncidinol (2S, 3′R, 7′R)-1-acetyl-2-[3′, 7′-diacetoxyeicosanyl)-glycerol, a major floral oil constituent of several Oncidiinae species (Orchidaceae). Thus, both tetrapedic A (C20) and B (C22) could be the biotransformation products of oncidinol and byrsonic acid by T. diversipes hydrolases. These are the chemical clues for bee visitation and oil collecting from both plant species. The results indicate that the deceit/pollination syndrome should not be applied to all Oncidiinae flowers.  相似文献

A novel phenolic rigid organic filler (KT) was used to modify isotactic polypropylene (iPP). The influence of KT particles on the tensile properties of PP/KT microcomposites was studied by uniaxial tensile test and the morphological structures of the stretched specimens were observed by scanning electron microscopy (SEM) and polarized optical microscopy (POM). We found that the Young’s modulus of PP/KT specimens increased with filler content, while the yield and break of the specimens are related to the filler particles size. The yield stress, the breaking stress and the ultimate elongation of PP/KT specimens were close to those of unfilled iPP specimens when the maximal filler particles size is less than a critical value, which is 7 μm at a crosshead speed of 10 mm/min and 3 μm at 200 mm/min, close to that of glass bead but far more than those of other rigid inorganic filler particles. The interfacial interaction was further estimated from yield stress, indicating that KT particles have a moderate interfacial interaction with iPP matrix. Thus, the incorporation of small KT particles can reinforce iPP matrix and simultaneously cause few detrimental effects on the other excellent tensile properties of iPP matrix, due to their organic nature, higher specific area, solid true-spherical shape and the homogenous dispersion of the ROF particles in microcomposites.  相似文献

The diversity and secondary metabolite potential of culturable actinomycetes associated with eight different marine sponges collected from the South China Sea and the Yellow sea were investigated. A total of 327 strains were isolated and 108 representative isolates were selected for phylogenetic analysis. Ten families and 13 genera of Actinomycetales were detected, among which five genera represent first records isolated from marine sponges. Oligotrophic medium M5 (water agar) proved to be efficient for selective isolation, and "Micromonospora-Streptomyces" was proposed as the major distribution group of sponge-associated actinomycetes from the China Seas. Ten isolates are likely to represent novel species. Sponge Hymeniacidon perleve was found to contain the highest genus diversity (seven genera) of actinomycetes. Housekeeping gene phylogenetic analyses of the isolates indicated one ubiquitous Micromonospora species, one unique Streptomyces species and one unique Verrucosispora phylogroup. Of the isolates, 27.5% displayed antimicrobial activity, and 91% contained polyketide synthase and/or nonribosomal peptide synthetase genes, indicating that these isolates had a high potential to produce secondary metabolites. The isolates from sponge Axinella sp. contained the highest presence of both antimicrobial activity and NRPS genes, while those from isolation medium DNBA showed the highest presence of antimicrobial activity and PKS I genes.  相似文献

We previously reported thatPolygonum aviculare has a strong allelopathic action againstCynodon dactylon (L.) Pers. and other test species. Moreover, we found that several phenolic compounds appeared to be important allelochemics in this activity. We have now isolated other potential inhibitors fromP. aviculare residues and soil underPolygonum stands, and none of these occurred in soil underC. dactylon stands. GC-MS analysis revealed that these additional inhibitors are long-chain fatty acids with 14–22 carbons. Nine were identified inP. aviculare residues and seven in soil underP. aviculare. Sodium salts of all the identified fatty acids inhibited seedling growth ofC. dactylon and at least some test strains of the nitrogen-fixing bacteria,Azotobacter andRhizobium.  相似文献

Abstract

Ophiostoma species have been demonstrated to metabolize wood extractives and be useful to the pulp and paper industry. In order to have new isolates for the Asian market, Eucalyptus camaldulensis logs were harvested from forest sites in central Taiwan and 28 strains of the Ophiostoma genus were isolated from them. These strains were subsequently inoculated onto Eucalyptus wood chips to evaluate their effects on weight losses of wood and the amounts of acetone extractives degraded. At the same time, Gas Chromatography-Mass Spectroscopy (GC-MS) analysis was conducted and by using calibration curves and a database of GC-MS mass spectra, changes in lipophilic compounds were analyzed. Fatty acids, hydrocarbons, sterol compounds, sterol esters, and triglycerides were significantly reduced after two weeks’ inoculation by the fungal strains. The results show that six of the strains were capable of reducing the lipophilic fractions by more than 60% in a two-week treatment. DNA of the most effective strains were analyzed and found to be a variant of Ophiostoma querci.  相似文献

This is the first report of the chemical and biological properties of the lipooligosaccharide (LOS) endotoxin isolated from Burkholderia dolosa IST4208, an isolate recovered from a cystic fibrosis (CF) patient in a Portuguese CF center. B. dolosa is a member of the Burkholderia cepacia complex, a group of closely related species that are highly problematic and opportunistic pathogens in CF. B. dolosa infection leads to accelerated loss of lung function and decreased survival. The structural determination of its endotoxin was achieved using a combination of chemistry and spectroscopy, and has revealed a novel endotoxin structure. The purified LOS was tested for its immunostimulatory activity on human HEK 293 cells expressing TLR‐4, MD‐2, and CD‐14. In these assays, the LOS showed strong proinflammatory activity.  相似文献

The conjugation of organometallic complexes to known bioactive organic frameworks is a proven strategy revered for devising new drug molecules with novel modes of action. This approach holds great promise for the generation of potent drug leads in the quest for therapeutic chemotypes with the potential to overcome the development of clinical resistance. Herein, we present the in vitro antiplasmodial and antiproliferative investigation of ferrocenyl α-aminocresol conjugates assembled by amalgamation of the organometallic ferrocene unit and an α-aminocresol scaffold possessing antimalarial activity. The compounds pursued in the study exhibited higher toxicity towards the chemosensitive (3D7) and -resistant (Dd2) strains of the Plasmodium falciparum parasite than to the human HCC70 triple-negative breast cancer cell line. Indication of cross-resistance was absent for the compounds evaluated against the multi-resistant Dd2 strain. Structure-activity analysis revealed that the phenolic hydroxy group and rotatable σ bond between the α-carbon and NH group of the α-amino-o-cresol skeleton are crucial for the biological activity of the compounds. Spectrophotometric techniques and in silico docking simulations performed on selected derivatives suggest that the compounds show a dual mode of action involving hemozoin inhibition and DNA interaction via minor-groove binding. Lastly, compound 9 a , identified as a possible lead, exhibited preferential binding for the plasmodial DNA isolated from 3D7 P. falciparum trophozoites over the mammalian calf thymus DNA, thereby substantiating the enhanced antiplasmodial activity of the compounds. The presented research demonstrates the strategy of incorporating organometallic complexes into known biologically active organic scaffolds as a viable avenue to fashion novel multimodal compounds with potential to counter the development drug resistance.  相似文献

Insecticidal and growth-reducing properties of extracts of 14 species of American neotropical Piperaceae were investigated by inclusion in diets of a polyphagous lepidopteran, the European corn borer,Ostrinia nubilalis. Nutritional indices suggested most extracts acted by postdigestive toxicity.Piper aduncum, P. tuberculatum, andP. decurrens were among the most active species and were subjected to bioassay-guided isolation of the active components. Dillapiol was isolated from the active fraction ofP. aduncum, piperlonguminine was isolated fromP. tuberculatum, and a novel neolignan fromP. decurrens. The results support other studies on Asian and AfricanPiper species, which suggest that lignans and isobutyl amides are the active defence compounds in this family.  相似文献

SAR studies on a set of novel hydrophilic C-2 aminopyridinyl riminophenazines bearing variously functionalized basic side chains at C-3 were conducted. The novel compounds were evaluated for in vitro activity against two different species of Leishmania promastigotes, intramacrophage Leishmania amastigotes, chloroquine-sensitive and chloroquine-resistant strains of P. falciparum, and also against mature-stage P. falciparum gametocytes. Their cytotoxicity was evaluated as well on BMDM cell lines. Most of the new compounds potently inhibited the growth of both genera of protozoa with IC50 values in the high nanomolar range and good selectivities versus mammalian cells. Besides their potent activity against asexual intraerythrocytic stages of P. falciparum, three compounds showed potential as transmission-blocking agents. The key role of the hydrophilic C-2 aminopyridinyl substituent to improve the leishmanicidal activity and the influence of the length and the nature of the basic side chain on the antiprotozoal activity and cytotoxicity were underlined.  相似文献
