The effect of two slaughter methods (immersion in ice-water slurry and electrical stunning followed by ice slurry asphyxiation) on chemical and microbiological parameters of grass carp (Ctenopharyngodon idella) stored in ice for 20 days was evaluated. No differences in total volatile basic nitrogen (TVB-N), pH, carbohydrate or protein content of mucus were observed between the slaughter methods. Ice-slaughtered fish had lower bacteria counts at the beginning of storage, but higher counts than fish slaughtered by electricity at the end of storage (p < 0.05). However, no significant differences in the shelf life were observed between the slaughter methods evaluated (limit of acceptability – counts > 3 × 106 CFU g−1 – attained after 13–16 days). Results indicated that the chemical parameters evaluated have a limited applicability to assess the shelf life of grass carp stored in ice, since pH limit (6.8) was exceeded after 4 days, while TVB-N limit (30 mg%) was not attained after 20 days of storage.  相似文献

Commercially collected records of Atlantic salmon (Salmo salar L.) muscle texture hardness were used to evaluate the effect of slaughter procedures and seasonality on texture quality. A database collected by Marine Harvest® contained flesh hardness records of Atlantic salmon slaughtered at processing plants in Norway from summer 2010 to summer 2011. The fish were slaughtered either by (1) percussion followed by automated bleeding (“Percussive”) or (2) live chilling with exposure to carbon dioxide (CO2) followed by manual severing gill arches and bleeding (“CO2”) or (3) live chilling with exposure to CO2 followed by percussive stunning and at the end automated bleeding (“CO2·percussive”). Hardness in salmon muscle cutlets was measured in Newtons (N) by Materials Testing Machine Zwick 500N. The hardness in salmon varied significantly over the study period (P < 0.05, mixed effect model) and showed the softest value of 21.2 (± 0.7) Newton (N) in summer 2011 and hardest 24.1 (± 0.2) N in autumn 2010. Slaughter procedures had a significant effect on salmon muscle hardness (P < 0.05, mixed effect model), where percussion followed by automated bleeding resulted in the hardest value (24.0 ± 0.4 N) as compared with CO2 stunning (21.8 ± 0.2 N) and combination of CO2 and percussive stunning (23.1 ± 0.15 N). CO2 is suspected as a causal factor in accelerated postmortem softening of the salmon muscle.  相似文献

CO2 stunning gave rise to the lowest pH values and water-holding capacities. Rigor mortis in carp and eel advanced the most. Testing of raw and prepared fish was performed by a panel assessing organoleptic properties. In many cases, fish anaesthetized manually were ranked to be better than those in the other groups. The use of a combined ozonised-slurry ice system was investigated as a new refrigeration system for the on-board storage of megrim (Lepidorhombus whiffiagonis), a fish species that is usually stored aboard fishing vessels for 1 to 2 weeks. The time elapsed between the catch and unloading at the harbour affects its quality and commercial value directly. Microbiological, chemical and sensory analyses were carried out in megrim after 2 weeks of on-board storage in ozonised slurry ice, slurry ice or flake ice, and for an additional period of 6 days. Sensory analyses revealed that megrim specimens stored in ozonised slurry ice (oSI600 batch) maintained A quality even after 20 days of storage, while counterpart batches stored in flake ice showed B quality at unloading, after 14 days of on-board storage. Storage in ozonised slurry ice (oSI600 batch) also led to significantly (p<0.05) lower counts of aerobic mesophiles, psychrotrophic bacteria, Enterobacteriaceae and proteolytic microorganisms in megrim muscle as compared with flake ice. Biochemical analyses revealed that the use of ozonised-slurry ice or slurry ice alone slowed down the formation of total volatile base-nitrogen (TVB-N) and trimethylamine-nitrogen (TMA-N) in comparison with storage in flake ice, also allowing a better control of pH. Lipid hydrolysis and oxidation events also occurred at a lower rate in the ozonised-slurry ice and slurry ice batches than in the flake ice batch. The present study demonstrates that the combination of slurry ice and ozone for the on-board storage of megrim is advisable, thus improving the quality and extending the shelf life of this fish species.  相似文献

After elaborating a suitable scheme to assess consciousness/unconsciousness of turbot, the effects on fish welfare of commercial and experimental slaughtering techniques were evaluated. Strong adverse reactions and slow loss of clinical reflexes were observed when fish were slaughtered by bleeding without prior anaesthesia. The efficiency of electricity (150 V for 2 s followed by 25 V for 5 min) as a stunning and killing technique was dependent on the current frequency and mode of administration, ie whether the electrical discharge was applied to the head only or throughout the whole body. Finally, percussion of the head with a mechanical device resulted in immediate and permanent loss of all responses and reflexes in all fish tested. Three methods were selected as a result of the preliminary tests (bleeding in ice slurry, whole‐body electrical treatment and percussion), and their influence on flesh quality was studied over a 9 day period. Fish killed by percussion were characterised by a higher pH and higher water content in the very early stage of post mortem storage, but also by a much delayed rigor mortis. In contrast, fish killed by electricity entered most rapidly into rigor mortis; their flesh was significantly softer (p < 0.05) throughout the entire storage time and was also redder and darker, as indicated by higher a* values (p < 0.001) and lower L* values (p < 0.01) respectively. The present paper demonstrates that a nonstntggling slaughter method can delay degradation of type V collagen in meat of chub mackerel Scomber japonicus and softening of the meat during postharvest chilled storage. The fish were slaughtered by piercing a knife into nape (nonstruggling method) or by leaving on ground (struggling method) and then stored in an ice box. Sensory study revealed that the postharvest softening of the meat was moderated at 4 and 8 h by the non‐struggling slaughter method in comparison with the struggling method. On the basis of the specific solubilization of type V collagen and reduced tyrosine content in it, a cleavage of the nonhelical regions (telopeptides) of the type V collagen occurred during the chilled storage in the fish slaughtered by the struggling method. The degradation of type V collagen was also slower in the meat of the fish slaughtered by the nonstruggling method, which can be directly linked to the moderation of the postharvest softening.  相似文献

ABSTRACT: The effect of 2 slaughter methods (immersion in ice-water slurry and electrical stunning followed by ice slurry asphyxiation) on the quality of grass carp ( Ctenopharyngodon idella ) stored in ice for 20 d was evaluated using sensory and chemical analysis. Electricity immediately stunned the fish and did not induce blood spots in the flesh. Fish killed by electricity showed a faster initial rate of ATP degradation and entered into rigor mortis earlier, but did not show significant differences in the sensory score when compared with fish killed by immersion in ice-water slurry. Thus, no differences were observed in the shelf life of carps between the 2 slaughter methods evaluated. The limit for acceptability of grass carp stored in ice was around 13 to 16 d. Grass carp accumulated more inosine than hypoxanthine. K, Ki, P , Fr, and H values were highly correlated with storage time and with the TFRU sensory scores in both groups; these could be used to assess the freshness quality of grass carp.  相似文献

Five test diets were formulated with decreasing levels of fish meal (up to 50%) replaced by alternative protein sources. Rainbow trout were fed the experimental diets for 12 weeks.The effects of feed ingredients on spoilage of Oncorhynchus mykiss in ice and under MAP/ice (40% CO2, 30% N2 and 30% O2) were investigated in terms of sensory, chemical and microbiological analyses. The results showed that the trout in MAP/ice was rejected at 14 days, after sensory analysis, due to excessive drip, whereas trout in ice were found to be acceptable even after 14 days of storage. However, cooked trout fillets, under both storage conditions, were rejected at 17 days. Fish in ice produced higher K values and higher concentrations of biogenic amines during the storage period of 17 days than the fish in MAP/ice. Bacteria grew more quickly in rainbow trout kept in ice than in MAP/ice. MAP/ice storage extended the shelf life of rainbow trout by approximately 2 days compared to ice storage alone in terms of microbiological analyses.  相似文献

The effect of modified atmosphere packaging (MAP 1: 70% CO2/30% N2 and MAP 2: 50% CO2/30% N2/20% O2) and vacuum packaging (VP), on the shelf-life of chub mackerel (Scomber japonicus) fillets stored under refrigeration was studied. Quality assessment was based on sensory analysis and biochemical indices determination. Increase in total volatile basic nitrogen (TVBN) and trimethylamine nitrogen (TMAN) followed the order: MAP 1 < MAP 2 < VP < air (control samples). The presence of oxygen into the fish package (air or MAP) resulted in an increase in the 2-thiobarbituric acid (TBA) values in comparison with samples packaged in the absence of oxygen. The most effective MAP used was MAP 1 which contributed to a considerably slower rate of fish spoilage. Based primarily on odour scores it was observed that raw chub mackerel fillets stored in the presence of air remained acceptable up to ca. 11 days, VP and MAP 2 samples up to ca. 15–16 days, while MAP 1 samples up to ca. 20–21 days of storage. On the other hand, flesh texture and flesh colour of all packaged samples received scores above or equal than the acceptability limit up to ca. 13–14 days of storage.  相似文献

A trial was undertaken to evaluate Differential Scanning Calorimetry (DSC) as a fast analytical tool to differentiate gilthead seabream subjected to variable conditions of slaughter stress and post-mortem storage. Fish were subjected to different harvesting stress conditions: profound anaesthesia (PA, low stress) and net crowding (NC, high stress). Fish were slaughtered in an ice-salt water slurry, and subsequently stored on ice (7 days). Additional NC fish were frozen (−20 °C) and subjected to a freeze–thaw cycle. Dorsal muscle was assessed for cathepsins activity, liquid loss and DSC analysis. It is demonstrated that DSC analysis is capable of differentiating fresh, frozen and thawed-re-frozen fish, while liquid loss and cathepsin B activity are good markers to distinguish fresh from frozen fish. Harvesting stress had little effect on myosin and actin enthalpy transitions, as observed by DSC at 49 and 74 °C, respectively, but a lower ΔH actin/myosin ratio was found in PA fish, suggesting that intense exercise prior to slaughter promoted partial denaturation of muscle myosin.  相似文献

The use of slurry ice was evaluated as a technological treatment prior to cooking processing of fish. Thus, sardine (Sardina pilchardus) was stored in slurry ice for 2, 5 and 8 days. At such times, sardine specimens were taken and subjected to steam cooking, and the results were compared with those from a parallel control batch previously stored in flake ice. Quality assessment of lipid damage in cooked fish was performed by measuring the formation of free fatty acids, peroxides, thiobarbituric acid-reactive substances and interaction compounds. The volatile amines–total and trimethylamine–assessment was also carried out. A significant (p<0.05) inhibition of lipid damage–peroxides and fluorescent compounds assessment–and trimethylamine formation was observed in cooked sardine as a consequence of the preliminary treatment in slurry ice. This work opens the way to the use of slurry ice as a preliminary treatment of fish material prior to its thermal processing.  相似文献

Shear force and rigor mortis were used to evaluate the post‐mortem changes of muscle texture in Atlantic salmon (Salmo salar). The fish were either stunned with carbon dioxide (CO2), electricity, or percussion prior to slaughter. The pre‐mortem stress during CO2 stunning resulted in an earlier onset and resolution of rigor mortis, and accelerated post‐mortem softening of the muscle tissue as compared to the other stunning methods. No significant differences, either in development of rigor mortis or shear force, were seen between fish that were stunned with electricity or by percussion. Consequently electricity can be used for stunning fish prior to slaughter.  相似文献

The effects of electrical and percussive captive bolt stunning both on initial and up to 14 days post mortem meat quality of Friesian young bulls were studied. The first group (NS; n=10) were slaughtered without stunning under Turkish slaughter procedure (these animals were slaughtered under conditions acceptable to the appropriate ethics committee) in a modern abattoir. In the second group (ES; n=10) animals were electrically stunned. In the third group (PS; n=10) animals were stunned with percussive captive bolt. Meat quality was assessed by examining muscle glycogen concentration, pH and WHC. Also cooking loss, texture parameters and consumer sensory properties were assessed at 24 h, 4, 7 and 14 days post mortem. Meat colour as L*, a*, b*, C*, h* values were evaluated at 0, 48 h, 3, 5, 7, 9 and 14 days post mortem. Pre-slaughter handling affected muscle glycogen concentration (P<0.05). Animals in electrical and percussive captive bolt stunned groups had higher muscle glycogen concentrations compared to animals in the non-stunned group (P<0.05). Although pre-slaughter handling did not affect significantly pH24 h and WHC, differences were significant for cooking loss, colour coordinates and texture parameters at some storage times. For all sensory attributes (odour, flavour, tenderness and overall acceptability) at all ageing periods (24 h, 4, 7 and 14 days) the percussive stunned (PS) group was significantly superior to the non-stunned (NS) group. The electrically-stunned (ES) group also tended to be superior to the NS group although not all differences were statistically significant. These results indicate that percussive captive bolt stunning of cattle improved meat quality compared with cattle electrically stunned using head only tongs and those non stunned under approved Turkish slaughter procedure.  相似文献

The slurry ice technology has shown profitable advantages when employed instead of traditional flake ice for the manufacture of chilled aquatic species. The present work is aimed at evaluating the effect of slurry ice as a preliminary treatment prior to frozen storage. For it, specimens of a small pelagic fatty fish species (sardine; Sardina pilchardus) were stored in slurry ice for 2, 5 and 9 days, then subjected to freezing (-80 °C; 24 h) and finally kept frozen (−20 °C) during 1, 2 and 4 months. At such times, rancidity development in frozen sardine was measured by sensory (odour, skin, colour and flesh appearance) and biochemical (lipid hydrolysis and oxidation) analyses and compared to a control batch previously chilled in flake ice. Sensory analysis indicated an extended shelf-life time for frozen sardine that was preliminary stored under slurry ice for 2, 5 or 9 days, as compared to their counterparts subjected to flake icing. Sensory results were corroborated (P<0.05) by biochemical lipid oxidation indices (thiobarbituric acid reactive substances and the fluorescence formation). The present work opens the way to the use of slurry ice instead of flake ice as preliminary treatment of fish material prior to the frozen storage.  相似文献

The application of slurry ice, a biphasic system formed by small spherical ice crystals surrounded by seawater at sub-zero temperature, was evaluated as a new storage method for ray (Raja clavata), the elasmobranch fish species that exhibits highest commercial value in the European food markets. This advanced technique was used to compare with a control batch stored 10 days in flake ice. The results obtained in the sensory analysis indicated a significant extension of the overall quality (A class fish) from 3 days (flake ice) to 6 days (slurry ice). The development of ammonia external odour was the limiting parameter in both batches and was correlated with the activity of the endogenous mechanisms involved in the degradation of proteins and non-protein-nitrogen (NPN) rather than with the activity of proteolytic microorganisms. Storage of ray in slurry ice significantly (P<0.05) slowed down both biochemical (as estimated by the follow-up of the pH, TVB-N and K-value evolution) and microbial degradation mechanisms (estimated by the development of psychrotrophes and mesophiles counts) in chilled ray muscle. According to the parameters evaluated, storage of ray in slurry ice extends the shelf-life of this elasmobranch fish species due to a better maintenance of sensory, biochemical and microbiological quality, thus facilitating its commercialization.  相似文献

The effects of aluminium foil and cling film on microbiological, chemical and sensory changes in wild sea bass (Dicentrarchus labrax) stored at chill temperature (4 °C) were studied. A quality assessment of wild sea bass stored in ice, in boxes without ice, wrapped in aluminium foil (WAF) and wrapped in cling film (WCF) at 4 °C was performed by monitoring sensory quality, nucleotide breakdown products, total volatile base nitrogen (TVB-N), and total viable counts (TVCs). The observed organoleptic shelf-life of sea bass was found to be 16 days in ice, 4 days in boxes without ice, 8 days in aluminium foil and 8 days in cling film. Demerit points did not differ significantly (P>0.05) between WCF fish and WAF fish. The nucleotide degradation pattern was found to be similar for all storage conditions except for inosine and hypoxanthine contents, which decreased after 12 days of storage for WAF and WCF. The content of TVB-N for all storage conditions showed similar tendencies until 12 days storage but reached the highest level (41.6 mg TVB-N 100 g–1 flesh) for fish stored in WAF and WCF. No significant differences (P>0.05) were found in TVB-N concentrations within the treatments during the early stages of the storage period. Bacteria grew most quickly in the sea bass kept in boxes without ice, followed by those kept WAF, WCF and in ice. The aim of this work was to evaluate a slurry ice (SI) packaging system on three bivalve species: clams (Venerupis rhomboideus), oysters (Ostrea edulis) and mussels (Mytilus galloprovincialis). Comparative analyses were performed on specimens caught in autumn and spring and compared to batches stored in conventional flake ice. Microbiological analysis of autumn specimens stored in SI showed significantly (P < 0.05) reduced numbers of mesophiles and psychrotrophs in all three species, as well as fewer Enterobacteriaceae in clams and oysters. Spring specimens also exhibited significantly (P < 0.05) lower microbial loads, with SI storage significantly (P < 0.05) improving the control of mesophiles, psychrotrophs and proteolytic bacteria in clams and oysters. Sensory analysis correlated well with microbial analyses, with a significantly (P < 0.05) better quality in terms of odour, taste, juiciness, appearance and shelf life. The SI packaging system evaluated in this work proved to be a valuable method for maintaining quality of these bivalve species.  相似文献

The effect of modified atmosphere packaging (MAP) on the quality changes of Pacific white shrimp (Litopenaeus vannamei) treated with or without green tea extract (1 g/L; GTE) in combination with or without ascorbic acid (0.05 g/L; AA) during refrigerated storage of 10 days was investigated. Shrimp without treatment stored under MAP had lowered psychrotrophic bacteria, enterobacteriaceae and H2S-producing bacteria count (P < 0.05) but similar lactic acid bacteria count (P > 0.05), in comparison with shrimp stored in air (control). The coincidental lowered rate of increase in pH, total volatile base (TVB) content and thiobarbituric acid reactive substances (TBARS) were obtained in shrimp stored under MAP (P < 0.05). However, MAP slightly lowered melanosis formation and improved likeness score to some extent. When shrimp were treated with GTE and stored under MAP, the lower microbiological and chemical changes as well as the lowest melanosis formation were observed, compared to shrimp kept under MAP without treatment and the control (P < 0.05). GTE treatment in combination with MAP could retard chemical changes and melanosis formation, regardless of AA incorporation (P > 0.05). Nevertheless, GTE in combination with AA had higher inhibition on microbial growth and yielded the shrimp with higher likeness, compared with the other treatments (P < 0.05). Therefore, shrimp treated with GTE in combination with AA prior to MAP had the lowest losses in quality during refrigerated storage.  相似文献

This study examined the use of nitrogen gas for stunning of Rainbow Trout. It was found that nitrogen could be used as an effective stunning method and that the strong aversive reaction reported for carbon dioxide stunning was not observed. Measurement of ATP, its metabolites and pH were carried out to evaluate the effects of nitrogen stunning on post mortem muscle biochemistry. This treatment was compared to percussive stunning and air asphyxiation. Post mortem ATP levels in nitrogen stunned fish (1.96 ± 0.36 μmol g?1) were significantly higher than for asphyxiated fish (0.2 ± 0.11 μmol g?1) but lower than values for percussively stunned fish (5.9 ± 0.89 μmol g?1). This work indicates that the use of gasses other than carbon dioxide for stunning of fish deserves further study.  相似文献
