Regulation of calcium and phosphorus homeostasis in the dairy cow   总被引:2,自引:0,他引:2  
This paper describes the mechanism of calcium and phosphorus homeostasis in dairy cows in an effort to provide a clearer understanding of the rationale behind current management and supplementation practices. Specifically addressed is the need to keep prepartum dietary calcium intake at less than or equal to 50 g/d to minimize the incidence of milk fever. Also discussed is the need to increase National Research Council recommendations for postpartum dietary calcium from 2.7 to 3.4 g/kg milk. This is particularly important during the first 1 to 2 mo of lactation to maintain calcium balance.  相似文献

Sodium propionate (2.5 mmol/kg) was infused rapidly via a jugular vein into each of 13 multiparous Holstein cows at 7 wk postpartum for observation of clearance of propionate. Associated concentration changes of acetate and glucose in blood plasma and glucagon and insulin in blood serum were quantified. This dose elevated concentrations of propionate, which declined subsequently at an exponential rate (.108 min-1). Concentrations of glucagon and insulin were increased in the sampling immediately following infusion, yet subsequent decline of insulin concentrations acted to decrease the molar ratio of insulin:glucagon as propionate returned to preinfusion concentrations. Analysis of sample means disclosed a negative correlation -.82 between glucose and molar ratio of insulin:glucagon. These experimental observations suggest that a supraphysiological dose of propionate has an immediate effect on the pancreas to alter endocrine secretion in the lactating cow.  相似文献

Eleven nonlactating Holstein cows in late gestation were used to study the effect of dietary calcium concentration on apparent selenium absorption. Digestion trials with total collection helped to estimate apparent absorption of specific nutrients. Mean daily selenium intake ranged from 900 to 1700 micrograms per day. Regression analysis indicated apparent selenium absorption was maximum when dietary calcium was .8% of dry matter intake. Amounts of dietary calcium less or greater than .8% of dry matter intake reduced apparent selenium absorption. Dietary calcium quantitatively affected apparent selenium absorption in amounts of nutritional significance when selenium was provided from natural feedstuffs.  相似文献

Dairy cows suffer blood Ca losses as lactation begins and might be affected by hypocalcemia in its clinical (total serum Ca concentration <1.50 mM) or subclinical form (total serum Ca concentration ≤2.14 mM). Several studies have suggested that hypocalcemia is associated with different health problems of the cow but results from different studies are not consistent. The objective of this study was to assess potential associations between subclinical hypocalcemia (SCHC) and displaced abomasum, intramammary infections, metritis, retained placenta, and ketosis. Also, the associations between SCHC and milk yield and reproductive function were evaluated. After discarding cows (32) with clinical hypocalcemia, a total of 764 cows from 6 different commercial farms were enrolled in this study. Blood samples were collected at 24 to 48 h postcalving and analyzed for total Ca concentration. Odds ratios of the different afflictions potentially associated with SCHC were calculated. Seventy-eight percent of the analyzed cows incurred SCHC. The occurrence of displaced abomasum, ketosis, retained placenta, and metritis was 3.7, 5.5, 3.4, and 4.3 times more likely, respectively, in cows that had SCHC than in cows with normocalcemia. Furthermore, the risk of incurring retained placenta or metritis increased in multiparous cows as serum Ca concentrations decreased compared with that in primiparous cows. Normocalcemic cows, independent of parity, were more likely to show their first estrus sooner after calving than SCHC cows, but no correlation was found between SCHC and other reproductive parameters. Different serum Ca concentration cutoffs were identified for several postpartum afflictions (≤1.93, ≤2.05, ≤2.05, and ≤2.10 mM for ketosis, retained placenta, metritis, and displaced abomasum, respectively). In conclusion, SCHC, defined as serum Ca ≤2.14 mM, is a frequent illness affecting the majority of the dairy cows with important repercussions on health. However, if SCHC were to be used to predict postpartum disease, different serum Ca cutoff points are likely to be needed because best predictive cutoff values varied among postpartum ketosis, displaced abomasum, retained placenta, and metritis.  相似文献

A study was conducted to evaluate the potential association between Ca status at calving and postpartum energy balance, liver lipid infiltration, disease occurrence, milk yield and quality parameters, and fertility in Holstein cows. One hundred cows were assigned to 1 of 2 groups based on whole-blood ionized Ca concentration ([iCa]) on the day of calving [d 0; hypocalcemic [iCa] <1.0 mmol/L (n = 51); normocalcemic [iCa] ≥1.0 mmol/L (n = 49)]. Cows were blocked based on calving date and parity. Blood samples were collected approximately 14 d from expected calving date (d −14), the day of calving (d 0), and on d 3, 7, 14, 21, and 35 postpartum for measurement of plasma nonesterified fatty acid, iCa, total Ca, glucose, and total and direct bilirubin concentrations, and plasma aspartate aminotransferase and gamma glutamyl transferase activities. Liver biopsies were obtained from a subset of cows on d 0, 7, and 35 for quantification of lipid content. Milk samples were collected on d 3, 7, 14, 21, and 35 postpartum for measurement of somatic cell count and percentages of protein, fat, and solids-not-fat. Data for peak test-day milk yield, services per conception, and days open were obtained from Dairy Herd Improvement Association herd records. Disease occurrence was determined based on herd treatment records. Hypocalcemic cows had significantly higher nonesterified fatty acids on d 0. Hypocalcemic cows also had significantly more lipid in hepatocytes on d 7 and 35 postpartum. However, no statistically significant differences were observed between groups for plasma aspartate aminotransferase and gamma glutamyl transferase activities or total and direct bilirubin concentrations. Milk protein percentage was lower in hypocalcemic cows on d 21 and 35. However other milk quality variables (somatic cell count, milk fat percentage, and solids-not-fat) and milk yield variables (peak test-day milk yield and 305-d mature-equivalent 4% fat-corrected milk yield) did not differ between groups. No differences were observed between groups in the occurrence of clinical mastitis, ketosis, displaced abomasum, dystocia, retained placenta, metritis, or fertility measures (percentage cycling at 50–60 d postpartum, services per conception, or days open). These data suggest that early lactation fatty acid metabolism differs between cows with subclinical hypocalcemia and their normocalcemic counterparts.  相似文献

The objective of this study was to investigate whether administration of l-Gln would affect mediators of acute phase response in postparturient dairy cows. Twenty-four multiparous Holstein cows were blocked by the expected day of calving and randomly assigned to 1 of the 3 treatment groups (n = 8/group): 1) i.v. infusion of 10 L of 0.85% NaCl (control), 2) i.v. infusion of 106, or 3) 212 g/d of l-Gln mixed with 10 L of 0.85% NaCl solution; each treatment was given 8 h/d for each of 7 consecutive days starting on d 1 after calving. Blood samples were collected 1 wk before the expected day of parturition as well as on d 0, 7, 14, and 21 after parturition; plasma concentrations of serum amyloid A (SAA), haptoglobin, and lipopolysaccharide-binding protein were measured by ELISA, and α1-acid glycoprotein was assessed by radial immunodiffusion. Concentrations of SAA, haptoglobin, and α1-acid glycoprotein increased in control cows after parturition, reaching peak values on d 0 or 7 postpartum (60, 1,093, and 963 μg/mL, respectively). Cows infused with 106 g/d of l-Gln had greater concentrations of SAA in plasma on d 14 and 21 compared with controls (62.8 vs. 30.2 and 71.1 vs. 34.5 μg/mL, respectively). Cows infused with 212 g/d of l-Gln had greater concentrations of SAA on d 7 (82.5 vs. 53.9 μg/mL) and lower concentrations of haptoglobin on d 14 and 21 postpartum compared with controls (264 vs. 621 and 175 vs. 587 μg/mL, respectively). Cows treated with 106 and 212 g/d of l-Gln had greater plasma lipopolysaccharide-binding protein concentrations on d 7 compared with control group (50.0 and 35.6 vs. 10.8 μg/mL, respectively). There were no treatment differences with respect to milk yield and DM intake during the experimental period. In conclusion, our data indicate that i.v. administration of l-Gln modulated acute phase mediators in dairy cows after parturition and warrants further research into the mechanisms behind these effects.  相似文献

Two Holstein cows were used to study the metabolism and excretion of phenobarbital in the lactating cow. Each cow received 6 g of sodium phenobarbital, intrarumenally, daily for 10 days. On the 10th day carbon 14 phenobarbital was mixed with the unlabeled material prior to dosing. Total milk, urine, and feces were collected for 6 days, and about 93% of the recovered radioactivity was in urine, 5% in milk, and 2% in feces. The biological half-lives of phenobarbital for milk, urine, and feces were 40.5, 29.5, and 46.5 h. From these values we calculated that 7 days after the last 6-g dose of phenobarbital the milk produced would contain less than .5 ppm of this compound.A second study determined how long after withdrawal from phenobarbital feeding the drug could be detected in milk. Three Holstein cows received 6 g of sodium phenobarbital, intrarumenally, daily for 10 days. Starting on the 11th day milk samples were collected at each milking and analyzed for phenobarbital by gas liquid chromatography and spectrophotometrically. After 7 days withdrawal, phenobarbital could not be detected in milk by either method.  相似文献

Reproductive disorders in the periparturient dairy cow   总被引:2,自引:0,他引:2  
Incidence, predisposing factors, and implications of various reproductive disorders (dystocia, twinning, stillbirth, retained placenta, cystic ovaries, anovulation, infections of the reproductive tract, metritis, and abnormal health status) are reviewed as to their inter-relationships and collective impact on reproductive performance, milk yield and predisposition to other diseases or disorders in the periparturient dairy cow. All reproductive disorders reviewed reviewed reduce reproductive performance either directly or indirectly. Concurrent milk yield was reduced marginally in a few studies as a consequence of twinning, retained placenta, cystic ovaries, metritis, or other uterine disorders, and in cows with an abnormal health status. There is strong evidence for associated losses in milk yield following surgical delivery of a stillborn calf. We conclude that most periparturient disorders occur as a complex, rather than as a single abnormality. Cows with one disorders are at increased risk for other disorders, including metabolic ones. In contrast, actual milk yield or potential for high production generally does not predispose cows to increased risk for any of the reproductive disorders. The literature suggests that prophylactic measures to prevent occurrence of the one disorder might decrease the risk and incidence of other related disorders, either directly or indirectly.  相似文献

Study objectives were to determine whether a nonsteroidal antiinflammatory drug would reduce parturition-induced inflammation and fever and consequently improve appetite, bioenergetic parameters, and production variables in transitioning dairy cows. Multiparous cows (n = 26) were randomly assigned to 1 of 2 treatments beginning at parturition: 1) flunixin meglumine (FM; 2.2 mg/kg of BW; Banamine, 50 mg/mL, Schering-Plough Animal Health, Kenilworth, NJ), or 2) saline (control) at 2.0 mL/45.5 kg of BW. All treatments were administrated i.v. daily for the first 3 d in milk (DIM). Individual milk yield and dry matter intake (DMI) were recorded daily for the first 35 DIM. Rectal temperature was measured daily at 0700 and 1600 h for the first 7 DIM. Milk composition was determined on 2, 7, 14, 21, 28, and 35 DIM and blood plasma was collected on 1, 2, 3, 4, 7, 14, 21, 28, and 35 DIM. Body weight and body condition score were determined on −7, 1, 7, 14, 21, 28, and 35 DIM. Flunixin meglumine treatment slightly increased rectal temperature (38.99 vs. 38.76°C) during the first 7 DIM and reduced overall DMI (22.04 vs. 19.48 kg/d), but there were no treatment differences in overall milk yield (35.2 kg/d), 3.5% fat-corrected milk (37.6 kg/d), energy-corrected milk (37.7 kg/d), DMI (2.97% of BW), or overall energy balance (−2.32 Mcal/d). There were no treatment differences in milk fat (3.91%), protein (3.32%), or lactose (4.57%). Treatment had no effect on plasma glucose (66.5 mg/dL) or nonesterified fatty acids (553 μEq/L), but plasma urea nitrogen tended to be less in FM-treated cows (16.4 vs. 14.5 mg/dL). Daily FM administration to cows for the first 3 d after parturition slightly increased rectal temperatures by 0.23°C, reduced feed intake, and did not improve production or energetic variables during the first 35 DIM in transition dairy cows.  相似文献

The purpose of the present study was to determine changes of plasma osteocalcin levels in periparturient cows and to examine the correlation of plasma osteocalcin level with that of 1,25-dihydroxyvitamin D, Ca, and inorganic P. Five Holstein-Friesian cows, aged 3 to 5 yr, were used from 5 d before to 15 d after calving. Concentration (mean +/- SE) of plasma osteocalcin decreased rapidly from d 1 prepartum, reached a low of 8.0 +/- 3.0 ng/ml at d 1 postpartum, and then recovered gradually to 17.8 +/- 3.8 ng/ml at 15 d after calving. In contrast, the concentration of plasma 1,25-dihydroxyvitamin D increased from calving to 3 d postpartum. Plasma concentrations of osteocalcin did not correlate significantly with those of 1,25-dihydroxyvitamin D, but it was significantly proportional to that of Ca and inorganic P. It is suggested that osteoblast function is depressed by a number of factors around the time of parturition.  相似文献

The tensile strength of 576 pieces of white line horn collected over 6 mo from 14 dairy cows restricted to parity 1 or 2 was tested. None of the cows had ever been lame. Seven cows were randomly assigned to receive 20 mg/d biotin supplementation, and 7 were not supplemented. Hoof horn samples were taken from zones 2 and 3 (the more proximal and distal sites of the abaxial white line) of the medial and lateral claws of both hind feet on d 1 and on 5 further occasions over 6 mo. The samples were analyzed at 100% water saturation. Hoof slivers were notched to ensure that tensile strength was measured specifically across the white line region. The tensile stress at failure was measured in MPa and was adjusted for the cross-sectional area of the notch site. Data were analyzed in a multilevel model, which accounted for the repeated measures within cows. All other variables were entered as fixed effects. In the final model, there was considerable variation in strength over time. Tensile strength was significantly higher in medial compared with lateral claws, and zone 2 was significantly stronger than zone 3. Where the white line was visibly damaged the tensile strength was low. Biotin supplementation did not affect the tensile strength of the white line. Results of this study indicate that damage to the white line impairs its tensile strength and that in horn with no visible abnormality the white line is weaker in the lateral hind claw than the medial and in zone 3 compared with zone 2. The biomechanical strength was lowest at zone 3 of the lateral hind claw, which is the most common site of white line disease lameness in cattle.  相似文献

Using an activity monitoring system (AMS) equipped with an accelerometer, 2 experiments were conducted to test the hypotheses that (1) enhancing progesterone before inducing luteolysis or (2) exposing cows to estradiol cypionate (ECP) or testosterone propionate (TP) after luteolysis would increase occurrence and intensity of estrus. Our goal was to determine if more cows could be detected in estrus by an AMS compared with other estrus-detection aids. In experiment 1, cows (n = 154) were fitted with both an AMS collar and a pressure-sensitive, rump-mounted device (HeatWatch; HW) and assigned to 3 treatments: (1) no CL + progesterone insert (CIDR) for 5 d, (2) CL only, or (3) CL + 2 CIDR inserts for 5 d to achieve a range in concentrations of progesterone. Prostaglandin F was administered to all cows upon CIDR insert removal or its equivalent. Progesterone concentration up to 72 h posttreatment was greatest in CL + 2 CIDR, followed by CL only, and no CL + CIDR cows. Estrus occurred 14 to 28 h earlier in no CL + CIDR compared with CL-bearing cows. Estrus intensity was greater for CL + 2 CIDR than for CL-only cows. The AMS and HW detected 70 and 59% of cows defined to be in estrus, respectively. In experiment 2, cows (n = 203) were equipped with both an AMS and a friction-activated, rump-mounted patch (Estrotect patch) and assigned to receive 1 mg of ECP, 2 mg of TP, or control 24 h after PGF. Concentrations of estradiol 24 and 48 h after treatment were greater in ECP cows compared with controls. Estrus expression detected by AMS or patches in cows defined to be in estrus tended to be greater or was greater for ECP compared with controls, respectively. Compared with controls and in response to TP or ECP, estrus occurred 8 to 18 h earlier and was of greater intensity for ECP cows, respectively. The AMS and patches determined 73 and 76% of cows defined to be in estrus, respectively. Of cows exposed to the AMS, HW, or patches, 70, 61, and 75%, respectively, were detected in estrus and more than 93% of these subsequently ovulated. In contrast, of the residual cows not detected in estrus, 62 to 77% ovulated in the absence of detected estrus. Only ECP was successful in inducing more expression and intensity of estrus, and proportions of cows detected in estrus exceeded 80%. Given the large proportion of cows equipped with AMS collars ovulating in the absence of estrus, further research is warranted to determine if more pregnancies can be achieved by inseminating those cows not detected in estrus at an appropriate time when PGF is administered to induce luteolysis.  相似文献

Multiparous Holstein cows (n = 40) were used in a randomized complete block design to determine the effects of feeding Ca and Na salts (1:1, wt/wt) of propionate and Ca salts of long-chain fatty acids (LCFA) on transition cow performance. All cows were fed the same basal diet once daily for ad libitum intake. Treatments (g/d) were 320 cornstarch (CS) as a control, 120 propionate (PRO), 120 propionate and 93 LCFA (PF1), and 178 propionate and 154 LCFA (PF2). Treatments were hand-mixed into the upper one-third of the TMR from 2 wk pre- through 3 wk postpartum. Intakes were recorded from 21 d pre- through 21 d postpartum. Energy density and crude protein were 1.54 and 1.65 Mcal/kg and 14.4 and 18.8% for pre- and postpartum diets, respectively. All cows received a common diet from 22 to 70 days in milk (DIM). Milk composition was analyzed on d 7, 14, and 21. Blood was sampled at 14, 7, and 2 d prepartum and 2, 7, 14, and 21 DIM. Pre- and postpartal dry matter intake (DMI) averaged 11.9 and 16.4 kg/d, respectively, and did not differ among treatments. A diet x week interaction for postpartal DMI was observed as cows fed PF2 consumed 2 kg/d less DM during wk 2 relative to other treatments. Milk yields from 22 to 70 DIM were 48.8, 48.5, 47.8, and 51.3 kg/d for CS, PRO, PF1, and PF2, respectively, and were not significantly affected by treatments. Milk true protein (3.32 vs. 3.16%) was increased and MUN (12.5 vs. 14.4 mg/dL) was decreased for CS relative to other treatments. Milk fat yield from cows fed PRO tended to be greater than those fed PF1 (1.58 vs. 1.29 kg/d). Plasma glucose, insulin, and beta-hydroxybutyrate were not affected by treatments. The PF2 treatment tended to decrease NEFA in plasma relative to PF1 over all times measured (492 and 670 muEq/L) and significantly decreased plasma NEFA relative to those fed PF1 postpartum (623 and 875 muEq/L). Relative to PF1, feeding propionate and LCFA at the higher level in this experiment improved energy balance postpartum as evidenced by decreased concentrations of plasma NEFA.  相似文献

Relationships between calcium of blood plasma and measurements of the electrocardiogram QT interval corrected for heart rate and interval from Q to the apex of T corrected for heart rate were investigated in six dairy cows. Calcium was varied by infusing 4.7% solution of ethylenediaminetetra-acetic acid disodium salt to induce hypocalcemia, followed by treatment with calcium borogluconate. The within-cow regressions of the intervals on calcium in plasma were significant in all four electrocardiogram leads (leads I, aVF, SIII, and SaVF). These regressions differed between cows in these same leads. Correlation coefficients between calcium and the intervals were consistently larger than --.94 in lead SIII in all cows. When the results from four of the cows similar in age and breed were analyzed separately, regression coefficients between individual cows were not significantly different. However, intervals adjusted for calcium in plasma differed between cows.  相似文献

The objective of this study was to examine the effects of dietary starch source and content in the immediate postpartum period on plasma metabolites and hormones and ovarian follicular development. One of 3 diets was fed in a randomized block design to 40 cows from calving until 70 d in milk. The diets contained 45% alfalfa silage (AS), 45% barley silage (BS), or 41% barley silage and 4% supplemental starch (SS) on a dry matter basis. All diets contained 45% barley-based concentrate and 10% alfalfa hay. Resulting starch levels were 25.2, 23.3, and 26.7% for AS, BS, and SS, respectively. Body condition was scored every other week and dry matter intake and milk yield were recorded daily. Milk samples were obtained weekly and blood samples were taken at calving and then every other week to determine concentrations of glucose, insulin, insulin-like growth factor-1, β-hydroxybutyrate, and nonesterified fatty acids. Transrectal ultrasonography was performed twice per week from 7 d after calving until first ovulation or 62 d in milk in all cows. For a subset of 7 AS-, 8 BS-, and 9 SS-fed cows, a complete estrous cycle was monitored for ovarian dynamics, and blood samples were collected every second day for progesterone and estradiol. Luteinizing hormone pulsatility was also determined (5 cows/treatment) approximately 15 d postcalving. Treatment had no effect on body condition score, dry matter intake, blood metabolites, milk yield, or milk fat and protein contents, but BS-fed cows had significantly higher levels of milk urea nitrogen compared with SS cows. Cows fed SS (31 d) tended to have a shorter interval from calving to first ovulation than cows fed AS (43 d) or BS (38 d). The incidence of double first ovulations was higher in cows fed SS (46%) compared with those fed BS (0%). Treatment had no effect on LH pulse frequency or amplitude, ovarian dynamics, or progesterone and estradiol concentrations during the observed estrous cycle. Energy balance did not differ among cows fed the 3 diets. Overall, dietary starch source and concentration had little effect on productivity or metabolic status of postpartum cows.  相似文献
