The effect on shear force of skeletal restraint and removing muscles from lamb m. longissimus thoracis et lumborum (LT) immediately after slaughter and electrical stimulation was undertaken at a rigor temperature of 18°C (n=15). The temperature of 18°C was achieved through chilling of electrically stimulated sheep carcasses in air at 12°C, air flow 1-1.5 ms(-2). In other groups, the muscle was removed at 2.5 h post-mortem and either wrapped or left non-wrapped before being placed back on the carcass to follow carcass cooling regimes. Following rigor mortis, the meat was aged for 0, 16, 40 and 65 h at 15°C and frozen. For the non-stimulated samples, the meat was aged for 0, 12, 36 and 60 h before being frozen. The frozen meat was cooked to 75°C in an 85°C water bath and shear force values obtained from a 1 × 1 cm cross-section. Commencement of ageing was considered to take place at rigor mortis and this was taken as zero aged meat. There were no significant differences in the rate of tenderisation and initial shear force for all treatments. The 23% cook loss was similar for all wrapped and non-wrapped situations and the values decreased slightly with longer ageing durations. Wrapping was shown to mimic meat left intact on the carcass, as it prevented significant prerigor shortening. Such techniques allows muscles to be removed and placed in a controlled temperature environment to enable precise studies of ageing processes. 相似文献

A study was undertaken to obtain information on survival of Escherichia coli O157:H7 in ground beef subjected to heat treatment, refrigeration and freezing and on survival of E. coli O157:H7 in fermented sausage kept at 7°C and 22°C. For the challenge test, a mixture of E. coli O157:H7 strains (EH 321, EH 385, EH 302) was used and enumeration was performed on an isolation medium suitable for recovery of stressed organisms: modified Levine's eosin methylene blue agar (mEMB). Heat resistance of E. coli O157:H7 decreased after pre-incubation at a reduced temperature.Escherichia coli O157:H7 was more susceptible to heat inactivation after storage at 7°C and die-off was even more enhanced if cultures were frozen prior to heat inactivation. The enhanced reduction of the pathogen at 56°C after prior storage under refrigeration was confirmed in a test with inoculated ground beef.Escherichia coli O157:H7 was able to survive in ground beef at 7°C for 11 days and at −18°C for 35 days showing maximal one log reduction during the storage period. Thus, ground beef contaminated with E. coli O157:H7 will remain a hazard even if the ground beef is held at low or freezing temperatures. At both 7°C and 22°C, a gradual reduction of E. coli O157:H7 was noticed in fermented sausage over the 35 days storage period resulting in a 2 log decrease of the high inoculum (106cfu 25 g−1). For the low inoculum (103cfu 25 g−1) a 2·5 log reduction was obtained in 7 and 28 days storage at respectively 22 and 7°C. Application of good hygienic practices and implementation of HACCP in the beef industry are important tools in the control of E. coli O157:H7. 相似文献

It was found that the optimal cooling rate depends mainly on cell properties (size, membrane structure). Biogenic amines, putrescine, cadaverine, and histamine were determined in unpacked and vacuum-packed beef; during storage at 4°C for 12 and 35 days, respectively, under the same conditions. Analysis by High Performance Liquid Chromatography in both kind of samples, showed variation of cadaverine and histamine with an increasing tendency to a definite increase of putrescine. In unpacked samples, maximum mean contents for each amine reached 10.4, 5.2 μg/g for putrescine and cadaverine, respectively (12th day) and 2.2 μg/g for histamine (5th day). The total amine content reached 16.5 μg/g (12th day). After 8 days of storage, organoleptic fall-off began and, on the 12th day, all samples were disqualified. In vacuum packs, amine levels increased after 19 days. Maximum levels were determined for putrescine on the 35th day (36.3 μg/g), for cadaverine on the 26th day (28.9 μg/g) and for histamine on the 35th day (19.0 μg/g). Although the vacuum-packed beef was organoleptically acceptable up to 35 days, the total amine levels were greater than 5 μg/g after the 12th day, indicating a leveling off of quality. 相似文献

The objective of this study was to investigate differences in adipose tissue fatty acid composition among stress genotypes as well as to determine relationships of those fatty acids with performance, carcass and meat quality traits. Haplotypic analysis of parents and offspring was employed to assign 48 pigs to three stress genotypes of swine. Individual fatty acid profiles were determined on subcutaneous backfat. Stress genotype had a significant effect on the proportions of C14:0 (P<0.05), C16:0 (P<0.10), C16:1 (P<0.05) and C18:0 (P<0.05) fatty acids. After adjusting for stress and gender, there was a negative correlation between backfat thickness and essential fatty acids (linoleic and linolenic) (P<0.001). In contrast, the correlation of loin muscle area with C18:2 (P<0.01) and C18:3 (P<0.001) was positive. The degree of fat firmness was negatively correlated to the proportion of total unsaturated fatty acids (P<0.01). No significant correlations were observed between fatty acid contents and lean meat quality traits. 相似文献

Our objective was to assess the effects of lactate enhancement in combination with different packaging systems on beef longissimus lumborum and psoas major steak color. Strip loins and tenderloins (n = 16) were assigned to one of four injection treatments (non-injected control, water-injected control, 1.25%, and 2.5% lactate in the finished product). Steaks were individually packaged in either vacuum, high-oxygen (80% O2/20% CO2), or 0.4% CO (30% CO2/69.6% N2) and stored for either 0, 5, or 9 days at 1 °C. The L∗ and a∗ values of both the longissimus and psoas responded similarly to lactate, which at 2.5% darkened steaks (P < 0.05) packaged in all atmospheres and improved (P < 0.05) the redness of steaks packaged in high-oxygen. Packaging steaks in CO did not counteract the darkening effects of lactate. Nevertheless, CO improved (P < 0.05) color stability compared with high-oxygen packaging. 相似文献

Growth and virulence of pathogenicYersinia enterocoliticawere investigated on high (pH>6.0) and normal (pH<5.8) pH pork packaged in modified atmospheres and stored at 4°C. Modified atmospheres used in the study were vacuum packaging and saturated CO2. Pork was packaged in a high gas barrier packaging film and examined over a 30-day period. Phenotypic characteristics were used to detect the presence of the virulence plasmid ofY. enterocoliticaafter exposure to the pork packaging and storage regimen. Phenotypic characteristics ofY. enterocoliticaisolates from pork loin stored at 4°C for 30 days that were studied included Congo red uptake, calcium dependence and autoagglutination in methyl red Voges–Proskauer broth and tissue culture medium. Numbers ofY. enterocoliticaon the lean surface of high pH pork slices increased approximately 2.7logcfucm−2when vacuum packaged and stored at 4°C for 30 days. Storage of inoculated normal pH pork in 100% CO2resulted inY. enterocoliticaremaining in the lag phase over the storage period. Honeys from different regions of the province of Buenos Aires were stored at ?20°C, and factors that affect crystallization were analyzed. Crystals were observed by light microscopy. Firmness, adhesivity and viscosity of the samples were measured. Honey was characterized by determining the water activity, turbidity, moisture, fructose, and glucose contents. Results show that the viscous characteristics of the samples depend on the number, size, and disposition of crystals. Various honey samples exhibited Newtonian, pseudoplastic, and thixotropic behaviors. Crystallization was favored at higher moisture contents, suggesting that the parameters that affect honey crystallization at room temperature have a different effect at freezing temperatures. Honey that presented higher values of firmness had a moisture content lower than 17%, and a linear inverse relationship was observed between the adhesivity and firmness of honey samples. 相似文献

The aim of this study was to investigate the effect of a 10-day ageing at 30 ± 2 °C on the texture of dry-cured hams processed at temperatures up to 18 ± 2 °C for 12 months in relation with raw ham pH and salting time. Three pH groups (semimembranosus muscle at 24 h post-mortem: Low pH < 5.7, Medium pH = 5.7 pH 5.9, and High pH > 5.9), three salting times (6 d, 10 d and 14 d) and two ageing temperatures (18 °C and 30 °C) were investigated. Physicochemical characteristics, instrumental and sensory texture and product sliceability were evaluated on biceps femoris and semimembranosus muscles. Hams with pHSM24 < 5.7 should be avoided in order to reduce the incidence of texture problems in dry-cured ham elaboration. Texture problems are especially important in hams with a reduced salt content that are mechanically sliced (not frozen). A 10-day ageing at 30 °C could be useful for reducing the soft texture problems in dry-cured hams processed at temperatures up to 18 °C for 12 months without affecting the product flavour. 相似文献

The rates of extraction of caffeine from sieved Kapchorua PF (600–710 μm) have been measured at 80°C with a range of aqueous salt and buffer solutions of ionic strength 0·11 mol dm−3. The first-order rate constants and the half-times of infusion showed no trend with pH when buffers from pH 3·0 to pH 8·3 were employed. The rate constants decreased on the addition of common salts like NaCl, KCl and CaCl2 but increased in the presence of electrolytes such as Bu4NCl that contain large ions. The results cannot be interpreted by changes in osmotic pressure although Donnan effects may be involved. Close parallels were found between the rate constants and the solubilities of caffeine in electrolyte solutions at 25°C. In particular, the values of both properties rise appreciably in the presence of species containing aromatic or other organic rings with which caffeine molecules associate. 相似文献

The aim of this study was to determine the effects of tumbling at different post-mortem times on the proteolytic features and quality attributes of beef loins (M. longissimus lumborum). Loins (n = 12) were cut into 4 sections and assigned to tumbling at 1, 6 or 11 days post-mortem or non-tumbled control. Upon tumbling, additional ageing was applied to a common post-mortem time of 16 days. In general, tumbling had no considerable impacts on water-holding ability of samples. Tumbling resulted in an immediate decrease in shear force values (WBSF) of beef samples. Tumbling at 1 day post-mortem with no ageing had similar WBSF compared to the non-tumbled controls at 16 days (P > 0.05). With ageing, tumbling increased amounts of protein degradation, myofibrillar fragmentation and calpain-1 autolysis of samples. These results suggest that early post-mortem tumbling coupled with ageing can synergistically impact the tenderness development of beef loins and shorten the necessary ageing period. 相似文献

The effect of oregano essential oil and film permeability on the behaviour of Salmonella typhimurium in sterile and naturally contaminated beef fillets stored under aerobic, modified atmosphere consisting of 40% CO2 /30% O2 /30% N2 (MAP) and a vacuum packaged (VP) environment was studied during storage at 5°C. In samples without oregano essential oil, the pathogen survived under all storage conditions. Addition of oregano essential oil at a concentration of 0·8% v/w resulted in an initial reduction of 1–2 log10 cfu g−1 of the majority of the microbial population of meat with lactic acid bacteria and S. typhimurium showing the greatest reductions in all gaseous environments regardless of film permeability. The use of VP/MAP at chill temperatures in conjunction with oregano essential oil as a means of controlling spoilage and safety of meat is discussed. 相似文献

The textural properties of beef over the temperature range −18 to +5 °C were measured using Warner Bratzler (WB) and tensile techniques. In addition, the effects of rapid radio frequency (RF) tempering and slower conventional air tempering on texture were compared. Temperature showed a significant effect (P < 0.05) on WB and tensile shear force, with higher values obtained at temperatures on or below −5 °C. Work to fracture values showed two peaks at −15 and −3 °C. Sample thickness and muscle fibre direction were also important factors affecting shear force, with samples cut across fibres showing higher values. Tempering method showed no effect (P ? 0.05) on the textural properties measured. In light of the rapid nature of RF tempering, these findings will be of interest to the meat industry. 相似文献

The effect of oxygen level (20, 40, 60 and 80%) in modified atmospheres on the oxymyoglobin content of intact and minced beef (M. semimembranosus, SM) was evaluated. There was no significant difference in the oxymyoglobin content of minced SM stored for up to 4 days in modified atmosphere packs containing 20, 40, 60 or 80% O(2). After 7 days, oxymyoglobin in minced SM decreased significantly (P?0.05) with decreasing oxygen level but by day 10 all samples had similarly low oxymyoglobin contents. Lipid oxidation increased significantly (P?0.05) between day 7 and 10 of storage in minced SM stored in modified atmospheres containing 40, 60 or 80% O(2). Oxymyoglobin and lipid oxidation occurred in intact SM but the extent of oxidation was lower than for minced SM. Exogenous α-tocopherol, dispersed in olive oil and added to minced SM (300 and 3000 mg α-tocopherol/kg lipid), had no significant effect on Hunter 'a' values when the samples were stored in low (20%) or high (80%) oxygen atmospheres. Exogenous α-tocopherol addition led to a significant reduction in lipid oxidation (P?0.05) in minced SM stored in high but not in low oxygen atmospheres. 相似文献

The advantages of storing raw milk, which is to be used for Cheddar cheese manufacture, at 2°C rather than at 6°C was examined. Storage of milk at the lower temperature effectively reduced the level of psychrotroph growth, and after 4 days the psychrotroph counts in samples stored at 2°C were 100-fold lower than those found in samples stored at 6°C. There was no advantage in terms of cheese yield in storing milks at the lower temperature, but an overall improvement in cheese quality was noted in samples produced from milk stored at 2°C. 相似文献

Combined effects of high voltage cold atmospheric plasma (HV-CAP) generated with the mixture of argon and oxygen (90%Ar/10%O2) for 5 min and chitooligosaccharide (COS) from squid (Loligo formosana) pen (internalized shell) at different concentrations (0.05, 0.1 and 0.2 g/100 g fish slices) on the shelf-life of Asian sea bass (ASB) slices were investigated during storage at 4 °C for 18 days. Total viable count (TVC) of fish slices treated with 0.1 or 0.2 g/100 g COS in conjunction with HV-CAP was lower than the acceptable limit (log 106 CFU/g sample) within 18 days. The growth of various pathogenic and spoilage bacteria including psychrophilic bacteria, Clostridium perfringens (not detected), lactic acid bacteria (3.77–4.37 log CFU/g), Enterobacteriaceae (4.03–4.50 log CFU/g), Pseudomonas (6.62–6.82 log CFU/g) and hydrogen sulphide (H2S)-producing (4.04–5.05 log CFU/g) bacteria of HV-CAP-treated slices containing COS was more inhibited than other treatments, especially during the first 15 days of storage. The thiobarbituric acid reactive substances (TBARS) and peroxide values (PV) of slices treated with combined HV-CAP and COS (0.1 or 0.2 g/100 g) were reduced by 28‐–64 and 40–46%, respectively than those treated with only HV-CAP throughout the storage (p <.05). Similarly, for HV-CAP treated slices, addition of COS decreased the total carbonyl content by 15–24% than those added without COS. COS was able to lower the oxidation of polyunsaturated fatty acids of slices caused by HV-CAP. Therefore, HV-CAP together with COS effectively prolonged the shelf-life of ASB slices with sensory acceptability at 4 °C for at least 12 days. 相似文献

The influences of nanoemulsions based on citrus peel (orange, lemon, mandarin, and grapefruit) essential oils on the biogenic amine (BA) formation in rainbow trout (Oncorhynchus mykiss) fillets stored 4 ± 2°C were investigated. The results demonstrated that main BAs in the trout fillets were putrescine, cadaverine, spermidine, spermine, serotonin, tyramine, dopamine, and agmatine. It was determined that nanoemulsion application had generally suppressive effect on the formation of BAs during storage. It was also found that especially orange and lemon nanoemulsion groups were effective on histamine formation. Histamine values ranged from 0.00 to 4.08 in all groups during storage and the levels were below the limit values of FDA and EU throughout the storage period for all treatment groups. Similarly, Quality Index (QI) values of BAs were found to be below legal limits in all groups. Consequently, it was found that nanoemulsions prepared from essential oils kept the BA formation at a lower level during storage. 相似文献

The viabilities of cells in cultures of Escherichia coli growing with formation of filaments at 6 °C
Cold adapted, exponential phase cells of three strains of Escherichia coli were incubated at 6 °C for 10 days. Cells in all cultures grew by elongation, with formation of filaments to different extents, but with generally only small changes in the numbers of colony forming units (cfu). At later incubation times some cells of all lengths were identified as dead by LIVE/DEAD staining, with cell wall damage being apparent in some cells stained as dead but not in cells stained as live. When samples of cultures were incubated at 37 °C for 2h, the numbers of cfu of two strains (ATCC 11775 and ATCC 23739) increased similarly and were not affected by the time of prior exposure to 6 °C. When incubated at 37 °C, the numbers of cfu of the other strain (8WT) increased less after incubation at 6 °C for 1, 2 or 3 days than when transferred to 37 °C from 15 °C. After>3 days at 6 °C numbers of strain 8WT were reduced after incubation at 37 °C for 2h. The findings show that in cultures of E. coli elongating at 6 °C, elongating cells and filaments do not divide; and cells of all lengths lose viability at similar rates. Also, the findings indicate that substantial fractions of cells in cultures elongating at refrigerator temperatures are inactivated by an abrupt change of temperature to 37°C. 相似文献
