The prevalence and development of adult worms in the lungs of mice and gerbils infected with Schistosoma mansoni was investigated. All infected BALB/c mice harbored the schistosomes in their lungs at 10-12 weeks post-infection, showing the distinct relocation of adult worms to the lungs, from the hepatic portal system. The male and female flukes from lungs of BALB/c mice were significantly smaller than those from livers. The percentage of gravid females in lungs was considerably lower than that in the livers. The number of eggs recovered from lungs of BALB/c mice and gerbils having lung female worms, however, was higher than that from animals without lung females, indicating egg deposition of lung females. The number of eggs detected in the brains correlated well with the number of eggs from the lungs in BALB/c and ICR mice. Out of 119 infected gerbils at 8 weeks post-infection, only two animals had egg-emboli in the brain vessels, although many eggs embolized in the lungs of those animals. These data suggest that transfer of worms to the lungs from livers involves reduction of worm recovery from the portal circulation, and also pulmonary pathology of the disease.  相似文献

Helminth-free donor pigs were inoculated with infective larvae of Oesophagostomum dentatum. Five weeks later, the donor pigs were killed to recover the adult worms. By nonsurgical rectal transplantation, 6 groups of helminth-free recipient pigs were then given different female-to-male ratios (FMR) of O. dentatum worms (10% females [F], 50% F, 90% F) at either high (300 worms) or low (30 worms) doses. Fecal egg excretions were measured once weekly following transplantation until the pigs were killed 4 wk posttransplantation to assess the worm burdens and their location. There was a strong and significant correlation between numbers of male and female worms located in the different sections of the large intestine, suggesting that the worm sex as well as physiological factors in the pig intestine may govern the location of the worms. There were no significant differences in fecal egg counts or female worm fecundity between any of the groups. However at the low dose level, the female worm fecundity was markedly lower in the L10% F group than in the 2 other groups, thus suggesting for low worm densities an upper threshold for the FMR above which O. dentatum females produce fewer eggs.  相似文献

Schistosoma mansoni and S. japonicum infections in nude mice (nu/nu) were compared with infections in nu/+ heterozygotes or intact mice. Seven to 12 weeks after exposure to S. mansoni, the responses of Swiss NCR, C3H, BALB/c and C57B1/6 nude mice did not differ substantially. Nude mice of all these strains showed minute granulomas around eggs in the liver and minimal hepatic fibrosis. Microvesicular and necrotizing changes in hepatocytes were similar in all mouse strains, and S. mansoni infections were frequently lethal to nude, but not to intact mice between the seventh and ninth weeks of infection. Nude mice that survived the ninth week of infection generally lived until the 12th week. The number of eggs per mature worm pair in the tissues of S. mansoni-infected nude mice was similar to the number in intact mice, but nude mice passed fewer eggs in the feces. Nude mice that received serum from infected intact mice excreted eggs in the stool in numbers equivalent to intact mice, but continued to form minute granulomas around S. mansoni eggs. Reconstitution with fetal thymus or with splenocytes from normal or S. mansoni-infected mice partially or completely restored hepatic granuloma size, granuloma eosinophils, hepatic fibrosis, and excretion of eggs in the feces. In contrast to S. mansoni infection, S. japonicum infections in nude mice did not cause necrosis of hepatocytes or excessive mortality, and S. japonicum eggs were passed in the feces in numbers equivalent to those passed by infected intact mice.  相似文献

Humoral and cellular responses to Schistosoma bovis antigens have been evaluated over a period of 11 weeks in mice exposed to S. bovis cercariae and data analysed in the context of the parasitic parameters (worm and egg loads) recorded at days 30, 60 and 80 of the ongoing infection. Results revealed a decrease of worm burden, particularly marked for female worms, between day 60 and day 80 of infection suggesting a higher susceptibility of female schistosomes to attrition mechanisms. The B-cell response, studied by measuring the production of different isotypes, was directed against different stage specific antigens, with a predominance of IgG1 antibodies associated with a significant increase of IgA and IgE antibodies after egg deposition. The T-cell response, assessed after in vitro stimulation of splenocytes, showed a predominant production of Th-2 cytokines (IL-4, IL-5 and IL-10) occurring after egg laying. Interestingly in contrast to S. mansoni infection the Th-2 polarization did not seem to be exclusively triggered by egg-associated antigens since significant amounts of IL-10 were produced after stimulation with adult worm antigen preparation (SWAP) before the beginning of egg deposition.  相似文献

The present investigation was undertaken to determine some morphological characteristics of Japanese strain of Schistosoma japonicum, the results are summarized as follows: (1) The pattern of testes in male worms can be divided into two types: the tandem (37.6%) and non-tandem (62.4%). (2) The number of testes varied from 1 to 13. The percentage of specimens with 7 testes was much higher than of others with both in tandem (70% against 30%) and non-tandem (86% against 14%) arrangement. (3) The unusual/irregular sets of testes including 2-3 additional sets were found in 158 (or 4.5%) of 3,498 male worms. Specimens with 7 testes in the unusual/irregular location numbered the highest (69.6%); the next with 8 (14.6%) and 6 (12%); and the lowest with 5 (3.8%). (4) Forty or 2.8% of 1,417 male worms were found to have 2-4 cecal loops. The cecal union located in 71-80% of worm bodys from the anterior end of male worm was found in highest number; the next in 61-70%; and the lowest in 51-60% and 81-90%. The location of united ceca in female worms was found all in the posterior part of body. (5) The ovary in all 125 females was located in the posterior portion of worm body except in 4 specimens. (6) The number of eggs in the uterus of female worms varied from 50 to 300. The mean number was 103.  相似文献

Uterine egg counts were determined by means of an electronic particle counter in individual female sealworm, Pseudoterranova decipiens (Nematoda: Ascaridoidea: Anisakidae) collected from grey seals (Halichoerus grypus) from Anticosti Island, northern Gulf of St. Lawrence, in August and September 1992. Mean number of eggs per female was 54,916.9 (+/- 51,866.4), with a maximum of 300,000. Egg counts were strongly correlated with worm length, and less so with host sex and age, sealworm sex ratio, and densities of Anisakis simplex and Phocascaris sp. No effect of sealworm density on worm egg counts was determined and competitive effects by Contracaecum osculatum on sealworm fecundity were not observed. Analyses demonstrated that variation in sealworm fecundity was greater among seal hosts within groups than between groups for host age and sex, density of sealworm, and density of C. osculatum. No evidence for density-dependent regulation of sealworm fecundity could be found. Variations in worm egg counts are attributed to individual host effects on worm growth and fecundity.  相似文献

In order to study the role of eosinophils in the host defense against Trichinella spiralis infection, worm recovery after infection with T. spiralis was compared between interleukin-5 transgenic (IL-5 Tg) mice with a constant high level of peripheral eosinophils and nontransgenic C3H/HeN mice. No significant difference in the recovery of muscle larvae or adult worms in the small intestine, fecundity of female adult worms, or infectivity of newborn larvae was observed between nonimmunized C3H/HeN and IL-5 Tg mice or C3H/HeN and IL-5 Tg mice immunized with somatic antigen of T. spiralis. However, a significant difference was observed in the fecundity of female adult worms and recovery of muscle larvae between nonimmunized and immunized IL-5 Tg mice or C3H/HeN mice. These results demonstrate that having more eosinophils does not improve immunity against the various aspects of T. spiralis infection.  相似文献

The inhibition of acetylcholinesterase secretion by male and female Heligmosomoides polygyrus was tested on worms taken from experimentally infected mice and maintained for 3 days in vitro in levamisole. The dose inhibiting 50% of enzyme secretion (ID 50) of male worms was twice the ID 50 for female worms. A similar difference was observed in vivo between the dose of levamisole removing 50% (LD50) of male and female worms from the mouse. Acetylcholinesterase secretion by worms and ID 50 were tested in vitro at 3-weekly intervals from 3 to 21 weeks post infection (WPI). Acetylcholinesterase secretion was always significantly higher for male than for female worms. A decrease of ID 50, correlated with the age of the worms was observed: from 1.5 to 0.5 micrograms/ml for males and from 0.7 to 0.1 micrograms/ml for females. These results were confirmed in vivo by a higher efficacy of the anthelminthic at 21 than at 4 WPI.  相似文献

The regulation of the worm population and of its pathological effects was studied after a single or trickle infection with T. circumcincta. One hundred and twenty lambs, 60 Romanov and 60 Mérinos d'Arles, 3 months old, were distributed in four balanced groups: non-infected (G0), infected with 7000 L3 per animal once and slaughtered after 4 weeks (G14) or 8 weeks (G18), and infected 8 times and slaughtered after 8 weeks (G88). Parasitological, histological, haematological parameters and weight gains were recorded on each animal. Female and artificially nursed lambs had lower worm burdens and egg counts (epg) than males and naturally suckled lambs. No difference in parasitological parameters was seen between the two breeds, but Mérinos lambs infected once, had a higher increase in pepsinogen concentrations than Romanov lambs. In the infected animals, a significant proliferation of mast and eosinophil cells was observed in the abomasum wall. Serum pepsinogen concentrations were significantly higher 3 weeks p.i. and the weight gain was depressed during the first month p.i. The worm population was more numerous and younger in group G14 compared with G18 in which 24% of the worms had been expelled during the second month p.i. The female worms in G18 were longer and had more eggs in utero and higher egg output. After the trickle infection (G88) the take was reduced, female worms were longer with more eggs in vagina (pars ejectrix) and there was a higher variability in the number of eggs (compared with G18 data). A Schistosoma mansoni adult worm anionic fraction (PIII) has previously been shown to protect mice against challenge infection and to reduce pulmonary and hepatic granulomatous hypersensitivity. Serum from PIII-immunized rabbit was used to screen a lambda gt11 cDNA library from S. mansoni adult worm in order to identify antigens capable of modulating granulomatous hypersensitivity. We obtained four clones with 400 (Sm-III.11), 900 (Sm-III.16), 1100 (Sm-III.10) and 1300 (Sm-III.12) bp of length. All clone-specific antibodies were able to recognize most of the PIII components. The sequence analysis showed that these clones presented high homology with S. mansoni paramyosin (Sm-97). These findings ascribe a new function to this antigen with an important role in modulation of granulomatous hypersensitivity to S. mansoni eggs.  相似文献

Schistosome parasites utilize hemoglobin as a major protein source for their metabolism. Degradation of hemoglobin has been hypothesized to be mediated by both cysteine and aspartyl proteases secreted into the lumen of the parasite intestine. We now show that two distinct types of irreversible cysteine protease-specific inhibitors both arrest schistosome hemoglobin degradation in vitro. Arrest of hemoglobin degradation is followed by death of developing schistosomula 1 week later. Schistosome infected mice treated by a dose of 2 mg inhibitor per day for 1 week early in infection, and 2 weeks at the time of egg production, showed a significant reduction in worm burden, hepatomegaly, and the number of eggs produced per female worm. Histopathology showed a minimal immune response to those eggs which were produced, consistent with a delay in egg production relative to untreated infections. By tagging the inhibitor with biotin, specific cysteine protease targets were identified in extracts of schistosome worms.  相似文献

In the present study, each of 43 cats was fed with a single metacercaria of Paragonimus pulmonalis and 2 cats (controls) with 2 metacercariae. They were killed 22-372 days postinfection. In the test group, 18 (41.9%) cats were infected, as well as the 2 cats in the control group. Among the 18 worms recovered from the test group, 9 found in the lungs were mature, whereas 5 mature and 4 immature worms were in the pleural cavity. Most eggs from worms encased in a cyst were passed in the feces. However, eggs from worms in the pleural cavity were only found on the lung surface and pleural cavity. Mature worms in cysts seemed to exhibit higher egg production capacity than those in the pleural cavity. Paired mature worms from a cyst laid more eggs than a single encysted mature worm in vitro at room temperature.  相似文献

The development of five schistosome species was compared in mice by the recovery of schistosomula from chopped lung tissue and of adult worms by portal perfusion. Three developmental patterns appeared. (1) Schistosoma japonicum was unique in showing an early establishment of schistosomula in and a rapid departure from the lungs together with the highest worm recovery; (2) S. haematobium contrasted by establishing later and persisting in the lungs for at least 2 weeks while yielding the lowest adult worm recovery; and (3) S. intercalatum, S. mansoni, and S. rodhaini had an intermediate pattern--they resided in the lungs for several days, then disappeared and produced intermediate numbers of adults. Lung petechiae, known to accompany the migration of S. japonicum, were never detected after infection with the other species. We speculate that the three migration patterns of schistosomes are related to the size of the relative spectra of naturally infected definitive hosts.  相似文献

C57BL/6 mice genetically deficient in interleukin-5 (IL-5-/-) and normal C57BL/6 (IL-5+/+) mice were infected with larvae of a homogonic strain of the nematode Strongyloides ratti. In primary infections both male and female IL-5-/- mice released two to four times more eggs and larvae than IL-5+/+ mice. IL-5-/- mice harboured about 60% more intestinal worms, which were more fecund, than IL-5+/+ mice. The duration of the infection was similar in normal and IL-5-deficient mice. Both IL-5-/- and IL-5+/+ mice resisted a secondary infection. IL5-/- mice lost more weight during the infection than normal mice and took longer to regain their initial weight after expelling the worms. The number of eosinophils increased in the bone marrow, peritoneal cavity and small intestine of IL-5+/+ mice, but not IL-5-/- mice, following infection. No significant differences between infected IL-5+/+ and IL-5-/- mice in mast cells or other leucocytes were observed in the peritoneal cavity. Thus, IL-5 functions to protect the host in a primary infection of S. ratti by limiting the number and fecundity of worms establishing in the small intestine. This protection is correlated with elevated blood and tissue eosinophilia which occurs in normal mice but not in IL-5-/- mice.  相似文献

Between June and November 1993, a parasitological, clinical and ultrasonographical survey was carried out in the mid west of Madagascar, among the whole population of a village where Schistosoma mansoni was endemic. S. mansoni eggs were found in the stools of 61.8% of the 544 individuals examined; the highest prevalence of infection (85.7%) was observed in the 15-19 years old age group. The most intense infections were recorded in the 10-14 years age group, in which the geometric mean egg count was 265 eggs/g of faeces and 34.3% of the individuals excreted more than 400 eggs/g. The egg count decreased steadily from the age of 15 years. Ultrasonographical examination was performed in 482 inhabitants older than 2 years: 29% of them presented definite schistosome associated liver alterations according to the proposals for staging S. mansoni infection made by the World Health Organization Cairo working group. Stage 3 was never observed in subjects under 30 years old. This is consistent with the need for a long-standing infection before serious lesions occur. A significant relationship between egg count and degree of liver alterations was found only in the 20-29 years age group.  相似文献

The capacity of a recombinant glutathione S-transferase from Schistosoma mansoni (rSm28GST) to vaccinate primates (Erythrocebus patas) against a heterologous infection with Schistosoma haematobium has been tested. Two injections of the purified molecule with Muramyl-Di-Peptide (MDP) as adjuvant resulted in a high level antibody response in the five immunized animals and in a significant reduction in worm fecundity compared to the controls which received adjuvant alone. Mean levels of daily egg excretion in urine an faeces were reduced by respectively 55% and 74% although perfusion revealed that worm burdens were similar in both groups. The protective effect was long lasting since it was maintained up to the end of the experiment, 42 weeks after infection. Hatching rates and the numbers of intra-uterine eggs were also significantly affected by the vaccination. Tissue eggs were also drastically diminished in the urogenital system (-80%) but the reduction was not statistically significant. One animal was not protected by the immunization. There was a good correlation between parasitological data and the intensity of bladder lesions assessed by microscopic examination. Polypoid formations together with an intense exudation of the lamina propria were frequently seen in the controls but rarely in the vaccinated group where formation of scar tissue was predominant. These results underline the vaccine potential of the recombinant Sm28GST as a possible valuable prophylactic tool for the control of egg-induced pathology and transmission of African schistosomes.  相似文献

We infected interleukin-5 (IL-5)-transgenic (IL-5-Tg) and IL-5 receptor alpha knockout (IL-5R alpha -/-) mice with Angiostrongylus cantonensis to determine the possible roles of IL-5 and eosinophils in A. cantonensis infection in mice. IL-5-Tg mice demonstrated significantly higher eosinophilia in bone marrow, blood and cerebrospinal fluid (CSF), lower intracranial worm recovery and smaller female worms than naive C3H/HeN mice. Both IL-5-Tg and C3H/HeN mice evoked antigen-specific serum and CSF IgA antibody responses as early as days 5 and 7 postinfection, respectively. Prominent eosinophil infiltration was noted around intracranial worms in the subarachnoid spaces of the mouse brains; eosinophils adhering to the worm surface were degranulated. In contrast, IL-5R alpha -/- mice yielded a higher worm recovery than wild-type or heterozygous mice at day 20 postinfection and failed to provoke CSF eosinophilia. These findings indicate that A. cantonensis infection in the mouse causes IL-5 production and subsequent CSF eosinophilia, the latter probably being involved in the killing of intracranial worms.  相似文献

Swiss Albino mice have been infected with S. haematobium and challenged 4 weeks later with S. mansoni. Parasitological, pathological and ultrastructural studies were done. The results revealed cross mating between the two species. A reduction in S. mansoni worm load, egg count, hepatic granuloma number and size was noticed. A study on the seasonal variations in the population structure of Haemonchus contortus and Trichostronglyus colubriformis was conducted for a period of 12 months in a typical large scale sheep farm on improved pasture in Peninsular Malaysia which has a wet tropical climate. Successive groups of helminth-free tracer lambs were grazed for 4 weeks together with naturally infected sheep and were necropised for worm counts 2 weeks after their removal from the pasture. The monthly populations of H. contortus fluctuated slightly except in May and August during which more worms were found in the tracer animals. The numbers of T. colubriformis were comparatively high from October to December 1992 and again in March 1993, low during April and June 1992. Small numbers of hypobiotic larvae of H. contortus were detected in the tracer animals. Development and survival of infective larvae of H. contortus and T. colubriformis on pasture were investigated by spreading faeces containing eggs on grass plots in October 1993, February and May 1994. Development of the eggs to the infective larvae occurred within one week and their survival times were 7 weeks in the 3 experiments. The potential for control by rotational grazing is discussed.  相似文献
