The seasonal variation of the essential oil composition, the antioxidant activity (DPPH, FRAP assays) and the total phenolic content (Folin-Ciocalteu assay) of two aromatic wild plants, Pistacia lentiscus L. (Anacardiaceae) and Myrtus communis L. (Myrtaceae), grown in Zakynthos, a Greek island, was investigated. Hydroalcoholic extracts obtained from myrtle berries of five different selections were studied to obtain unique clones. During the typical 40 day maceration period, dry matter, pH, colour, and anthocyanins, were analyzed. Variability among myrtle selections was observed both in fresh berries (yield berries/plant, weight of the berries, number and weight of seeds per berry) and in the hydroalcoholic extract physical–chemical characteristics (dry matter, colour intensity, tint and anthocyanins). Reversed-phase high performance liquid chromatography (RP-HPLC) analysis with photodiode array detection (PDA) was used for anthocyanins characterization in myrtle hydroalcoholic extracts. Anthocyanin qualitative composition was the same in all selections, but varied strongly in its total amount. D was the selection with the characteristics most suitable for industrial use.  相似文献

Myrtle (Myrtus communis L.) berries extracts were prepared with solvents at different polarity (water, ethanol, and ethyl acetate) and analysed using different in vitro tests in order to evaluate their antioxidant properties. Antiradical and total antioxidant activities were measured with DPPH and FRAP tests, respectively. Their ability to protect biological molecules was assessed using the cholesterol and LDL oxidation assays. In addition, phenolic compounds and unsaturated fatty acids composition was analysed by HPLC–DAD and HPLC–MS/MS. Ethanol and water extracts showed the highest amount of extracted compounds, but the highest antiradical and antioxidant activities were found in ethanol and ethyl acetate extracts. These extracts were also the ones with the highest content of phenolic compounds. In addition, our results showed a highly significant correlation between the amount of total phenols and antiradical (R2 = 0.9993) or antioxidant activities (R2 = 0.9985) in these extracts. HPLC–DAD and HPLC–MS analyses showed significant quantitative and qualitative differences among these three extracts. The ethyl acetate extract had the highest protective effect in assays of thermal (140 °C) cholesterol degradation and Cu2+-mediated LDL oxidation, inhibiting the reduction of polyunsaturated fatty acids and cholesterol, and the increase of their oxidative products. These results suggest that because of these properties, myrtle berries could be used in dietary supplements preparations or as food additives.  相似文献

This study is designed to examine the fruit essential oil composition, the total phenolic amounts and the antioxidant activities in methanolic extracts of Coriandrum sativum under saline conditions. Increasing NaCl levels to 75 mM reduced significantly the fruit yield by 36%. The essential oil yield was 0.30%, based on the dry weight; it increased by 77% and 84% at 50 and 75 mM NaCl, respectively, in comparison to the control. The major constituents were linalool and camphor, whose amounts increased with increasing NaCl concentrations. Antioxidant activities of the methanol extracts were determined by three different test systems, namely DPPH, β-carotene/linoleic acid and reducing power assays. In these three test systems, the highest activity was exhibited in control plants and was reduced significantly with increasing NaCl levels. In control plants, the total phenolic amount was 1.04 mg GAE/g DW which decreased by 43% and 66% at 50 and 75 mM NaCl, respectively.  相似文献

Fatty acids and essential oils from hydroponically cultivated Salvia officinalis leaves were analyzed by GC and GC–MS. Four different levels of NaCl (25, 50, 75 and 100 mM) were applied. The first results showed that salt treatment reduced significantly the plant growth by 61% and the total fatty acids (TFA) content by 32% at 100 mM NaCl. Alpha-linolenic, gadoleic, palmitic and oleic acids were the major fatty acids. Moreover, the polyunsaturated fatty acids decreased, while the monounsaturated ones increased with respect to increasing salinity. Regarding the essential oil composition, the main compounds were α-and β-thujone, 1,8-cineole, camphor, α-humulene, viridiflorol and manool at all salt treatments. The yield had a maximum increase at 75 mM NaCl. Hence, sage can be considered as moderately salt sensitive.  相似文献

The antimicrobial activity of the hydro-alcohol extract of Myrtus communis L. (ME) berries was investigated against six Listeria monocytogenes strains (2 type strains and 4 isolates). Sub-lethal ME concentrations reduced L. monocytogenes counts by at least 2 log cycles. A Central Composite Design was used to investigate the combined effects of sub-lethal concentrations of ME (0.039–0.195 mL/100 mL), NaCl (0–2.0 g/100 mL) and pH (5.0–7.0) on strains growth. ME affected growth parameters, generally extending lag phase length and reducing maximum growth, sometimes with interactive effects with pH. The highest ME concentrations (0.117–0.195 mL/100 mL) combined with the lowest pH values (5.0–6.0) strongly reduced or even inhibited strains growth.  相似文献

Essential oils of Myrtus communis L. leaves were obtained using supercritical fluid extraction (SFE) and hydrodistillation methods. The experimental parameters of SFE such as pressure, temperature, modifier volume, static and dynamic extraction time were optimised using a central composite design after a 2n−1 fractional factorial design. The chemical compositions of the SFE extract were identified by GC–MS and determined by GC–FID. The major components of essential oils obtained by hydrodistillation were α-pinene (31.8%), 1,8-cineole (24.6%), limonene (14.8%), linalool (8.3%) and α-terpinolene (4.8%). However, by using the supercritical carbon dioxide in optimum conditions, only three components represented more than 85% of the extract. Therefore, by using the proper SFE conditions, the supercritical extraction is more selective than the conventional hydrodistillation methods. The oil yields based on the hydrodistillation was 0.47% (v/w). Extraction yields based on the SFE varied in the range of 0.5–6.3% (w/w) under different conditions.  相似文献

The aim of this work is to examine the chemical composition and antioxidant activity of separated essential oils and different solvent extracts of Thymus praecox subsp. skorpilii var. skorpilii (TPS). The ethanol, acetone, methanol, hexane, aqueous extracts and separated essential oils of TPS were assessed for their antioxidant activities. Antioxidant activities were evaluated by reduction of Mo(VI) to Mo(V), reducing power, superoxide scavenging activity, free radical-scavenging activity, metal chelating activity, linoleic acid peroxidation, hydrogen peroxide scavenging activity and peroxide scavenging activity. Essential oils were characterized in total to be 41 components, whereas 9 components were isolated by column chromatography for antioxidant activity. TPS essential oil was found to contain thymol (40.31%) and o-cymene (13.66%) as the major components. The ethanol, methanol and water extracts exerted significant free radical-scavenging activity. The methanol and water extracts displayed highest superoxide scavenging activity. The water extract has the highest total phenolics (6.211 mg gallic acid (GAE)/g DW) and flavonoids (0.809 mg quercetin/g DW).  相似文献

In the current study, neuroprotective effects of the ethyl acetate and methanol extracts of four terebinth coffee brands and the fruits of Pistacia terebinthus L. were investigated through enzyme inhibition tests against acetylcholinesterase, butyrylcholinesterase, and tyrosinase as well as antioxidant test systems. Antioxidant activity was measured using radical scavenging activity tests and metal-related tests including metal-chelation capacity, ferric-reducing antioxidant power (FRAP), and phosphomolybdenum reducing power (PRAP). The fatty oils of the coffee brands and the fruits and the fruit essential oil were examined by GC–MS. Total phenol and flavonoid contents were calculated spectrophotometrically. The extracts had moderate inhibition against butyrylcholinesterase (9.78–45.74% at 200 μg mL−1) and potent scavenging activity against DPPH. They exerted strong activity in FRAP and metal-chelation tests and modest activity in PRAP test. Oleic acid was identified as the major fatty acid in the fatty oils, while α-pinene (26.31%) was dominant in the essential oil.  相似文献

The variations in the content and the composition of dormant bud essential oil in six blackcurrant (Ribes nigrum L.) cultivars collected at various vegetation phases (from December 15, 2004 until April 19, 2005) were studied. Essential oil yield varied from 0.6% to 1.8%, except for the buds harvested in April, when the yield was considerably lower, 0.19–0.27%. Fifty volatile compounds were identified in the bud oils, hydrocarbon (38–55%) and oxygenated (on average 30%) terpenes being the major chemical constituents. Sabinene, δ-3-carene, terpinolene were dominant components, while cis- and trans-β-ocimene, α-thujene, α- and β-pinene, myrcene, α- and β-phellandrene, α- and γ-terpinene, p-cymene, cis- and trans-sabinene hydrate, terpinen-4-ol, α-terpineol, trans-piperitol, bornyl acetate, terpinyl acetate, citronellyl acetate, germacrenes D, β-caryophyllene, α-humulene, α-selinene, δ-cadinene and α-cadinol were found in reasonable amounts. Almiai may be considered as a superior cultivar, as possessing the most even content of oils ant the main constituents at all harvesting periods, except for April; however some other cultivars (Gagatai, Joniniai) accumulated higher amounts of oil at particular vegetation phases. January may be considered as a preferable harvesting time of buds; the amount of major terpenes at this phase was the highest in the all cultivars except for Joniniai. The concentration of the main oil compounds in buds harvested in April was 2–50 times lower than at other periods in the all six cultivars.  相似文献

The leaf essential oil of Photinia serrulata was obtained by hydrodistillation and analyzed by GC and GC-MS. Seventy-one components were identified in the essential oil and the main components of the oil were 10-epi-γ-eudesmol (12.72%), pinene (6.85%), sabinene (5.93%), α-humulene (5.87%) and α-thujene (5.47%). The in vitro cytotoxicity of the oil on human cancer cell lines Hela, A-549 and Bel-7402 was examined. The oil was found to be very active against all the three human tumor cell lines tested with low IC50 of 0.0427 μl/ml (Hela), 0.0219 μl/ml (A-549) and 0.0301 μl/ml (Bel-7402). The oil was also found to possess antioxidant activity as demonstrated by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical method.  相似文献

The essential oils composition of coriander (Coriandrum sativum L.) fruits obtained by hydrodistillation was studied at three stages of maturity by GC–FID and GC–MS. Essential oil yields showed marked increase during maturation process and forty one compounds were identified. Geranyl acetate (46.27%), linalool (10.96%), nerol (1.53%) and neral (1.42%) were the main compounds at the first stage of maturity (immature fruits). At the middle stage, linalool (76.33%), cis-dihydrocarvone (3.21%) and geranyl acetate (2.85%) were reported as the main constituents. Essential oils at the final stage of maturity (mature fruits) consist mainly on linalool (87.54%) and cis-dihydrocarvone (2.36%). Additionally, accumulation of monoterpene alcohols and ketones was observed during maturation process of coriander fruit.  相似文献

The essential oils obtained from leaves and fruits of Daucus gingidium ssp. gingidium have been studied. The main constituents of the essential oil from the leaves were sabinene (26.8%), α-pinene (10.8%), germacrene D (6.9%) and limonene (5.7%). Sabinene (60.6%) was the main compound identified in the essential oil of the fruits, followed by α-pinene (12.2%) and 4-terpineol (5.4%). Furthermore, qualitative and quantitative considerations about differences with literature data on Daucus carota have been made in order to confirm the species status of D. gingidium.  相似文献

This study was designed to examine the chemical composition and in vitro antioxidant activity of the essential oil of Clinopodium vulgare. GC–MS analysis of the oil resulted in the identification of 40 compounds, representing 99.4% of the oil; thymol (38.9%), γ-terpinene (29.6%) and p-cymene (9.1%) were the main components. The samples were subjected to a screening for their possible antioxidant activity by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and β-carotene-linoleic acid assays. In the first case, IC50 value of the C. vulgare essential oil was determined as 63.0 ± 2.71 μg/ml. IC50 value of thymol and γ-terpinene, the major compounds of the oil, was determined as 161 ± 1.3 μg/ml and 122 ± 2.5 μg/ml, respectively, whereas p-cymene did not show antioxidant activity. In β-carotene-linoleic acid system, C. vulgare essential oil exhibited 52.3 ± 1.19% inhibition against linoleic acid oxidation. In both systems, antioxidant capacities of BHT, curcumine and ascorbic acid were also determined in parallel experiments.  相似文献

Guided isolation through bioautography on TLC using 1,1-diphenyl-2-picryl-hydrazyl radical (DPPH) as a detection reagent led to the isolation of four antioxidant compounds from fruit of Perilla frutescens var. acuta. These compounds were identified as rosmarinic acid (1), luteolin (2), apigenin (3), and chrysoeriol (4), by means of UV, NMR, and ESI MS. All the compounds were isolated for the first time from the fruit of the plant. Compounds 1 and 2 showed significant DPPH scavenging capacities, with IC50 values of 8.61 and 7.50 μM, respectively. Further quantitative HPLC analysis confirmed that compounds 1-4 are the predominant contributors to the free radical scavenging activity of the extract of P. frutescens var. acuta.  相似文献

In the present work, the essential oil compositions of Myrtus communis berries from ten Corsican localities were studied and no chemical variability was observed. HS–SPME, GC and GC/MS analysis were carried out for the characterisation of volatile fractions of M. communis berries and two derived commercial alcoholic beverages (liqueur and eau-de-vie). Quantitative variations due to the distillation process were observed between the two beverages. The volatile compositions of Corsican myrtle alcoholic products were characterised by high amounts of monoterpene hydrocarbons and oxygenated monoterpenes with α-pinene and 1,8-cineole as major components. The polyphenolic compositions of the berry extract and the liqueur were also established using HPLC–DAD and LC–MS–MS; high concentrations of flavonol glycosides, flavonols and flavanols were reported.  相似文献

In this study, total contents of phenolic, flavanol and flavonol, antioxidant activities and antimicrobial activities of the Turkish endemic Salvia pisidica Boiss. & Heldr. ex Bentham (Lamiaceae) extract and essential oil were assessed in vitro. Total phenolic, flavanol and flavonol contents in the extract were 54.57 mg gallic acid equivalents (GAE)/g, 16.70 mg catechine equivalents (CE)/g and 18.19 mg rutin equivalents (RE)/g, respectively. Antioxidant activities (IC50 value) of the extract and essential oil were determined as 4.88 and 6.41 mg/mL by DPPH assay, respectively. 31 compounds were determined in the essential oil using GC-MS and the major compounds (%) were camphor (23.76), sabinol (19.2), α-thujone (14.2) and eucalyptol (1.8-cineole) (5.8).The antimicrobial activity of the methanolic extract and the essential oil against 13 bacterial and two yeast strains was determined. The extract (concentration 5 g/100 ml or 10 g/100 ml) was effective against most of the strains tested, yet not against Bacillus cereus, Staphylococcus aureus, Aeromonas hydrophila and the two yeast strains tested. The essential oil (2 g/100 ml) showed an antimicrobial effect against all the gram (+) bacteria tested, against Saccharomyces cerevisiae, but was not effective against all gram (−) bacteria and Candida albicans. These results show that S.piscidica essential oil and extract could be considered as a natural alternative to traditional food preservatives and be used to enhance food safety and shelf life.  相似文献

The composition of the essential oil from Dictamnus dasycarpus was analysed by GC–MS; 47 constituents (88.9% of the total oil) were identified. The main compounds were syn-7-hydroxy-7-anisylnorbornene (29.4%), pregeijerene (15.5%) and geijerene (11.4%). The antimicrobial activity of the essential oil was evaluated against nine microorganisms using disc diffusion and broth microdilution methods. The oil showed the strongest bactericidal activity against Staphylococcus aureus ATCC 25923 and methicillin-resistant S. aureus. The in vitro cytotoxicity of the oil on six human cancer cell lines was also examined. The cytotoxicity of the oil on three human breast cancer cell lines was significantly stronger than on other cell lines.  相似文献

The objective of this study was to compare the composition of fatty acids stored in the various parts of swordfish. Muscle and organ sections of a series of swordfish samples were collected. Their chemical analysis allowed the classification of swordfish as a semi-fatty fish, with its byproducts totalling 35.6% of the total fatty acids (TFA) and its white and red muscles (MR) 7.2%. The highest contents were found in the liver (26%), the gonads (4.7%) and the red muscle (RM) (4.5%). The UFA/SFA ratio as recommended by nutritionists is 3; in the liver, skin and RM samples this ratio was, respectively, 3.5, 2.8 and 2.7. There is a high level of unsaturated fatty acids (UFA) in swordfish. More than 40% of the muscular tissues are made up of polyunsaturated fatty acids (PUFA) of the n-3 series (EPA + DHA). The byproducts contain more than 30% of monounsaturated acids characterised by the fatty acids of the n-9 series and particularly by oleic acid.  相似文献
