The hydroxycinnamic acid (HA) content of starting barley for brewers’ spent grains (BSG), whole BSG and phenolic extracts from BSG was measured using high performance liquid chromatography (HPLC) and correlated with antioxidant potential. The effect of BSG phenolic extracts on antioxidant activity of fruit beverages was also assessed (using the total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assays). The concentration of HA present in barley extract and BSG was in the order of ferulic acid (FA), p-coumaric acid (p-CA) derivatives, FA derivatives, p-CA, caffeic acid (CA) and CA derivatives. Results suggested that brewing and roasting decreased the HA content. Antioxidant activity was significantly (P < 0.05) correlated with caffeic acid (R2 = 0.8309) and total HA (R2 = 0.3942) concentrations. Addition of extracts to fruit beverages resulted in a significant (P < 0.05) increase in antioxidant activity of cranberry juice, measured by the FRAP assay. In vitro digestion significantly (P < 0.05) reduced TPC, DPPH and FRAP activity of the fruit beverages.  相似文献

The purpose of the study was to determine polyphenols, anthocyanins and ascorbic acid in the extracts of black raspberry fruits and wine, along with their anti-oxidant, anti-proliferative and anti-inflammatory activities. Black raspberry fruits without or with seeds crushed were blended in 60% ethanol (FE and FES, respectively) or in water (FW and FWS, respectively). Black raspberry wine without or with seeds crushed (W and WS, respectively) were prepared. Polyphenol content was the highest in the FES (8.25 mg/g fruit). Generally the ethanol extracts with seeds crushed showed higher anti-oxidant activities with the lowest DPPH IC50 (130 μg/ml (freeze-dried extract/reaction solution)) for the FES and the lowest ABTS IC50 (198 μg/ml) for the WS. Cell viabilities were reduced by 13–70% when treated with 100 μg/ml (freeze-dried extract/medium) for HT-29 cells and 1000 μg/ml for LNCaP cells. The FES most actively suppressed nitric oxide production in LPS-stimulated RAW264.7 cells (p < 0.05). Superoxide dismutase and glutathione peroxidase activities treated with the extracts were higher than the control (p < 0.05).  相似文献

The purpose of this study was to evaluate the effect of adding condensed tannins in the form of non-purified (Liposterine®) or purified (Exxenterol®) extracts obtained from Carob fruit to prevent lipid cooked pork meat systems from oxidising during chilling and frozen storage. The antioxidant activity of these extracts was compared with that of α-tocopherol. Meat lipid alteration was evaluated as thiobarbituric acid reactive substances content (TBARS) and polar material-related triglyceride compounds followed by high-performance size-exclusion chromatography (HPSEC). TBARS levels were lower (P < 0.05) in samples containing Liposterine (LM), Exxenterol (EM), and α-tocopherol (TM) than in control sample (CM) under chilled storage. TBARS formation was similar (P > 0.05) for LM and EM but lower (P < 0.05) than for TM. Polar material increased several times in all samples, but significantly less in TM and EM than in LM. Thermal oxidation compounds determined by HPSEC were lower (P < 0.05) in EM than in LM or TM. The changes in polar material were proportionally smaller after six months frozen storage than after chilled storage, with Exxenterol displaying the highest antioxidant protection. Therefore Carob fruit extracts can be successfully used to reduce fat alteration in cooked pork meat at chilled and frozen temperatures.  相似文献

The antioxidant and anti-diabetic properties of the sequential extracts of Vernonia amygdalina based on the chemical composition of the most effective anti-diabetic extract were studied. Using DPPH and ABTS radical scavenging as well as FRAP assays, the extracts showed a consistent dose-dependent trend of potent antioxidant activity in the following solvents: water extract > methanol extract > chloroform extract > and petroleum ether extracts. In the oral glucose tolerance test, the chloroform extract exerted the highest response (33.3%), similar to metformin (27.2%), after 2 h compared to the control (50.8%, P < 0.05). After a 14-day administration in diabetic rats, the chloroform extract recorded the highest blood (23.5%) and serum (21.4%) glucose-lowering effects (P < 0.05). GC–MS analysis of the chloroform extract revealed high levels of linoleic acid (4.72%), α-linolenic acid (10.8%) and phytols (12.0%), as well as other compounds.  相似文献

The antioxidant activities of different polarity solvents extracts from barley were evaluated by various antioxidant assays, including reducing power, free radical scavenging and lipid oxidation inhibition. Those various antioxidant activities were compared to standard antioxidants, butylated hydroxyluene (BHT) and ascorbic acid (Vit. C). The properties of the extracting solvents significantly affected the total phenolics, proanthocyanidins and antioxidant activities of barley extract. The highest contents of total phenolics and proanthocyanidins were obtained from extraction with 70% acetone. For three different solvent extracts, the antioxidant activities were in this order: 70% acetone extract > 70% ethanol extract ? 70% methanol extract. Reducing powers of three extracts and their scavenging effects on 2, 2-diphenyl-1-picrylhydrazyl radicals were effective at amount of 200 μg. The 70% acetone extract of barley exhibited high antioxidant activity in linoleic acid system, which was not significantly (P < 0.05) different from BHT during the incubation time.  相似文献

The effects of brined onion extracts on selected aspects of quality in cooked encased turkey breast rolls were evaluated during refrigerated storage over 7 days. Using the thiobarbituric acid (TBA) assay, a 50% strength juice gave a significant reduction (P < 0.0001) in lipid oxidation in both 100 g and 1 kg rolls relative to controls. Quercetin, the main onion juice antioxidant, was reduced by 65% in freshly cooked 1 kg rolls compared with raw rolls but changed little thereafter in storage. Cook yields were significantly higher for rolls containing added onion extracts than for controls (P < 0.05) but no colour differences were detected using Hunter Lab values (P ? 0.05). A 30-member untrained sensory panel expressed no preference for freshly opened 4 day-stored control or onion extract-supplemented rolls packed in plastic casings but there was a significant preference (P < 0.05) for the onion extract product when 4 mm thick meat slices were stored in air for an additional 24 h.  相似文献

Efficacy of varying concentrations of dried Holy basil powder and its ethanolic extracts in retarding oxidative rancidity was tested with cooked ground pork. The development of lipid oxidation products during 14 days refrigerated storage (5 °C) was evaluated by means of thiobarbituric acid-reactive substances (TBARS) value, peroxide value (POV), conjugated diene and hexanal content. In the amounts studied, dried Holy basil powder and its ethanolic extract significantly (P < 0.05) inhibited the formation of TBARS, peroxide, conjugated dienes and hexanal in a dose-dependent manner. The oxidation process was significantly (P < 0.05) influenced by the type of antioxidant and its concentration. Addition of dried Holy basil powder to cooked ground pork was more effective in inhibiting lipid oxidation than its ethanolic extracts. Additionally, TBARS values and hexanal contents in cooked ground pork were highly correlated (r2 = 0.95; P < 0.05) with each other.  相似文献

Six different types of yogurt were manufactured from Damascus goat milk, Awassi ewe milk and a mixture of equal portions of the 2 species of milk using 2 types of commercial yogurt cultures (CH-1 and YF-3331). Yogurts were chemically analysed at 1, 7, 14 and 21 days of storage. Results showed that cultures significantly affected acetaldehyde (P < 0.05), acetone (P < 0.05) and diacetyl (P < 0.001) contents. Type of milk significantly influenced acetaldehyde (P < 0.05), diacetyl (P < 0.001), acetoin (P < 0.001) and ethanol (P < 0.05) levels. Significant variations occurred in acetaldehyde (P < 0.001) and acetoin (P < 0.05) contents during the storage. Short-chain free fatty acids were the highest in ewes’ milk yogurt made with culture YF-3331, and increased during storage, while the levels of medium-chain free fatty acids, except for decanoic acid, were unchanged and the amount of long-chain free fatty acids decreased during storage. Cultures used and types of milk had no effect on long-chain free fatty acids in yogurts.  相似文献

Barley β-glucan is a functional ingredient implicated to lower glycemic response, plasma cholesterol and food intake. The objective of the present study was to analyse the polyphenol content and antioxidant activities of four different solvent extracts of three barley β-glucan samples – Glucagel, Barley Balance (BB) and barley fibre rich fraction (BFRF). The polyphenol content and antioxidant activities of the β-glucans were in the order BFRF > BB > Glucagel (P < 0.05). Of the four different solvents used (70% acetone, 70% methanol, 70% ethanol and acidified methanol), 70% acetone gave the highest phenolic content (P < 0.05). The free radical scavenging capacities of 200 mg of BB and BFRF in 70% acetone were very similar to 40 μg of standard gallic acid, ferulic acid, ascorbic acid and trolox. We discuss the possibility that health benefits associated with β-glucan samples might depend on their polyphenol and antioxidant contents that vary with method of preparation and purity.  相似文献

The total phenol content, antioxidant and pro-oxidant activities of deodourised, water-soluble aniseed, basil, caraway, cardamon, fennel, ginger, juniper, laurel and parsley extracts were estimated using a number of in vitro assays. The laurel and basil extracts contained the highest phenol content of 107.3 ± 1.3 GAE [mg gallic acid equivalents/g (dry wt.) extract] and 98.5 ± 1.4 GAE, respectively, whilst the ginger extract contained the lowest content at 14.9 ± 0.9 GAE. Juniper, laurel and basil extracts were consistently better than the other extracts in terms of iron(III) reducing activity, inhibition of β-carotene-linoleate thermal co-oxidation and N,N-dimethyl-p-phenylenediamine and hydroxyl radical scavenging assays. Potential pro-oxidant activities of the extracts were assessed using both DNA and bovine serum albumin (BSA) as substrates. None of the extracts were capable of stimulating hydroxyl-mediated DNA fragmentation; however, the extracts could be categorised in the protein oxidation assay as extracts with (i) no significant (p > 0.05) effect, (ii) a significant (p < 0.05) protective effect or (iii) a significant (p < 0.05) pro-oxidant effect. The extracts from juniper, laurel and basil had a pro-oxidative effect upon BSA at a dose of 2 mg/ml, as estimated from the degree of carbonylation measured.  相似文献

The objective of this study was to characterise the antioxidant properties of both water and ethanol extracts from the fruit of Gardenia jasminoides Ellis (GJE). The IC50 values for DPPH (1,1-diphenyl-2-picryl-hydrazyl), ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)diammonium salt], hydroxyl and superoxide radical-scavenging activities were 0.14, 0.21, 1.08 and 1.43 mg/ml for the water-based extract, and 0.36, 0.39, 1.56 and 1.99 mg/ml for the ethanol-based extract, respectively. The extracts also showed strong reducing power, nitrite-scavenging activity, inhibition of linoleic acid oxidation, superoxide dismutase-like (SOD-like) activity and catalase activity. However, the water extract had a higher antioxidant activity than the ethanol extract. In addition, the antioxidant activities were highly correlated with the observed phenolic and flavonoid contents. Therefore, our study strongly suggests that extracts derived from the fruit of Gardenia jasminoides could be an excellent source of antioxidants as dietary supplements.  相似文献

Epidemiological and experimental studies have suggested that high dietary fat intake of mice is associated with many physically degenerative diseases. Since oxidative stress and abnormal lipid metabolism have been speculated to be critical mechanisms underlying degenerative diseases, we hypothesized that a high-fat (HF) diet might induce oxidative stress or lipid oxidation and subsequently contribute to the high risk of some diseases such as cardiovascular and cerebrovascular ones. To test this hypothesis, male kunming mice were placed on either a HF diet or a normal laboratory diet for 30 consecutive days. This investigation demonstrated that blood fat [low density lipoprotein (LDL), total cholesterol (TC), triacylglycerols (TAG), high density lipoprotein (HDL)], blood sugar (blood glucose and liver glycogen) and oxidative stress (activities of antioxidant enzymes and levels of non-enzymic antioxidants) of mice fed high-fat diet (group II) were significantly increased or decreased (P < 0.05, P < 0.01) when compared with the control group (I). The present study revealed that HF diet induced oxidative stress and provided novel evidence regarding the link between high dietary fat and increased risk of degenerative diseases. The administration of Lycium barbarum polysaccharides did not show any effect on the body weight of the experimental mice, but significantly decreased the levels of LDL, TC, TAG, blood glucose and thiobarbituric acid reactive substances (TBARS) or increased the activities of antioxidant enzymes (P < 0.05, P < 0.01) when compared with mice in HF group (II). These findings were further supported by significantly increased non-enzymic antioxidants levels (P < 0.05, P < 0.01), suggesting that L. barbarum polysaccharides showed a noticeable inhibition against lipid oxidation induced by free radicals caused by HF diet intake (groups III, IV, V) on the basis of their antioxidant activities.  相似文献

Eight male hamsters per group were assigned randomly to one of the following diets: chow diet (Control); high-fat/cholesterol diet (HFCD); HFCD supplemented with 1% Tau (HFCD/1% Tau); HFCD supplemented with 2% Tau (HFCD/2% Tau). Tau supplementation improved (P < 0.05) serum lipids and cholesterol profile in the high-fat/cholesterol-dietary hamsters. Although hepatic cholesterol/triacylglycerol in the high-fat/cholesterol-dietary hamsters were not (P > 0.05) changed by Tau supplementation, faecal cholesterol and bile acid outputs were increased (P < 0.05). Two percent Tau supplementation unregulated (P < 0.05) HMG-CoA reductase and cholesterol 7-α hydroxylase (CYP7A1) expressions in the high-fat/cholesterol-dietary hamsters. Besides, Tau supplementation also increased (P < 0.05) LDL receptor mRNA expressions in high-fat/cholesterol-dietary hamsters. Tau supplementation also reduced serum GPT and GOT values and C-reactive protein (CRP) levels in the high-fat/cholesterol-dietary hamsters. Results clearly indicated that Tau could alleviate blood lipids and hepatic damage induced by a high-fat/cholesterol-dietary diet.  相似文献

The objectives of this study were to determine phenolic content and antioxidant activity of methanolic extracts from different parts of cantaloupe (leaf, stem, skin, seed and flesh). The flesh extract afforded the highest yield (89.6 ± 0.3%) whilst the lowest yield was obtained from the seed (13.7 ± 0.5%) (p < 0.05). The leaf extract showed the highest total phenolic content (26.4 ± 0.3 mg GAE/g extract) and total flavonoid content (69.7 ± 3.37 μg RE/g extract) accompanied with best antioxidant activity through all antioxidant assays (p < 0.05). In addition, the stem extract also exhibited good antioxidant activity. Thus, these results suggest that methanolic extracts of cantaloupe leaf and stem may serve as a potential source of natural antioxidant for food and nutraceutical application.  相似文献

The effect of modified atmosphere packaging (MAP) on the quality changes of Pacific white shrimp (Litopenaeus vannamei) treated with or without green tea extract (1 g/L; GTE) in combination with or without ascorbic acid (0.05 g/L; AA) during refrigerated storage of 10 days was investigated. Shrimp without treatment stored under MAP had lowered psychrotrophic bacteria, enterobacteriaceae and H2S-producing bacteria count (P < 0.05) but similar lactic acid bacteria count (P > 0.05), in comparison with shrimp stored in air (control). The coincidental lowered rate of increase in pH, total volatile base (TVB) content and thiobarbituric acid reactive substances (TBARS) were obtained in shrimp stored under MAP (P < 0.05). However, MAP slightly lowered melanosis formation and improved likeness score to some extent. When shrimp were treated with GTE and stored under MAP, the lower microbiological and chemical changes as well as the lowest melanosis formation were observed, compared to shrimp kept under MAP without treatment and the control (P < 0.05). GTE treatment in combination with MAP could retard chemical changes and melanosis formation, regardless of AA incorporation (P > 0.05). Nevertheless, GTE in combination with AA had higher inhibition on microbial growth and yielded the shrimp with higher likeness, compared with the other treatments (P < 0.05). Therefore, shrimp treated with GTE in combination with AA prior to MAP had the lowest losses in quality during refrigerated storage.  相似文献

To overcome the disadvantages of using synthetic anti-oxidants in meat products, an investigation was carried out to evaluate the anti-oxidant effect of extracts of fruit by-products viz., kinnow rind powder (KRP), pomegranate rind powder (PRP) and pomegranate seed powder (PSP) in goat meat patties. Total phenolics content, DPPH radical scavenging activity and effect of these extracts on instrumental color, sensory attributes and TBARS values during storage (4 ± 1 °C) of goat meat patties were evaluated. Results showed that these extracts are rich sources of phenolic compounds having free radical scavenging activity. Hunter Lab L value significantly (P < 0.05) lowered in PRP followed by PSP and KRP patties. Sensory evaluation indicated no significant differences among patties. Further, a significant (P < 0.5) reduction in TBARS values (lipid oxidation) during storage of goat meat patties was observed in PRP, PSP and KRP as compared to control patties. Average TBARS values (mg/kg meat) during refrigerated storage (4 ± 1 °C) were significantly lower in PRP, followed by PSP and KRP as compared to control. The overall anti-oxidant effect was in the order of PRP > PSP > KRP. It was concluded that extracts of above fruits by-product powders have potential to be used as natural anti-oxidants in meat products.  相似文献

The effect of bioaccumulated Cu on the antioxidant properties of medicinal plants grown in Cu rich soil is studied. The total phenolic and flavonoid content, 2,2′-Diphenyl-1-picrylhydrazyl (DPPH•) scavenging activity, reducing power, metal chelating activity and inhibition of lipid peroxidation were assayed in methanol (MeOH) extracts of eight medicinal plants grown in the vicinity of copper mining impact site and compared with control samples. Corresponding IC50 values of DPPH• scavenging ability and metal chelating ability were found to be significantly (P < 0.05) higher in mining impact samples e.g., Withania somnifera, Azadirachta indica, Andrographics peniculata and Ocimum sanctum. The IC50 of inhibition of lipid peroxidation of all the mining impact samples were significantly (P < 0.05) higher than the control samples, indicating lower inhibition capacity of lipid peroxidation by the mining impact samples. This was attributed to higher Cu contents in the MeOH extracts of mining impact samples measured by inductively coupled plasma mass spectrometry (ICP-MS).  相似文献

This study investigated the optimum polyphenol extraction conditions for two Nigerian green leafy vegetables, Vernonia amygdalina (VA) and Gongronema latifolium (GL) that are widely used for food and medicinal purposes. Seven different solvents were used to extract the total phenolic contents of the leaves; 100% acetone, 80% acetone, 50% acetone, 100% ethanol, 70% ethanol, 100% methanol and 70% methanol and their total phenolic contents were determined. The results showed that the 80% acetone was significantly (p < 0.05) the most effective for polyphenol extraction from the leaves and generally the acetone-containing solvents were more effective than alcohol-containing solvents. The 80% acetone extract was fractionated on a silicic acid-packed column to give two main fractions: acetone eluted chlorophyll-enriched (flow-through) and ethanol eluted chlorophyll-depleted (column-bound). The ethanol fractions of VA and GL were significantly better (p < 0.05) DPPH radical scavengers than the acetone fractions. In contrast, the acetone fraction was a significantly better (p < 0.05) scavenger of hydroxyl radicals when compared to the ethanol fraction. Both acetone and ethanol fractions significantly (p < 0.05) and dose-dependently inhibited linoleic acid oxidation, though duration of inhibition was less at lower concentrations (10 and 25 ??g/ml) than at the highest concentration (50 ??g/ml).  相似文献

The efficacy of varying concentration of mustard leaf kimchi ethanolic extracts (MK) in retarding oxidative rancidity was tested with raw ground pork. Freshly ground pork meat was assigned to one of the following five treatments: control (no antioxidants); AC-0.02 (0.02% ascorbic acid); MK-0.05, 0.1, and 0.2 (0.05%, 0.1% and 0.2% MK, respectively). The pH of the samples decreased and the TBARS values and free fatty acids (%) increased considerably (P < 0.05) during storage. The total bacterial count was lower in MK-0.1 and MK-0.2 than the control during storage. The internal L∗ value and a∗ value decreased (P < 0.05) with the addition of MK. The internal b∗ value of MK treatments were higher (P < 0.05) than that for the control and increased incrementally with MK concentration. The TBARS values and free fatty acids (%) of MK-0.02 was lowest among the treatments. The peroxide value of the control increased until 7 days and reached the maximum value at a certain storage time and decreased thereafter. In the other treatments it increased. All treatments had lower concentration of conjugated dienes (P < 0.05) compared to the control sample, after the first day. Mustard leaf kimchi ethanolic extracts exhibited a protective effect against lipid oxidation in raw ground pork.  相似文献
